Cocksfoot (Dactylis glomerata L.). perennial ryegrass (Loliumperenne L.) and Italian ryegrass (L. multiflorum Lam.) plantswere grown on deep (75–95 cm) columns of soil in glasshousesand growth rooms with and without irrigation. The species inwhich growth declined least rapidly after water had been withheldwere those which transpired most slowly. During early establishmentin the glasshouse cocksfoot transpired least because of slowroot growth. In the growth room, when root systems were deeperand denser, perennial ryegrass transpired least because of lowleaf water conductance. Results are discussed in relation to(a) drought resistance in the three species; (b) breeding forincreased drought resistance through modifying root distributionand leaf water conductance; and (c) the use of isolated soilcolumns in water relations studies. Summary Tall fescue (Festuca arundinacea Schreb.) protoplasts, inactivated by iodoacetamide, and non-morphogenic Italian ryegrass (Lolium multiflorum Lam.) protoplasts, both derived from suspension cultures, were electrofused and putative somatic hybrid plants were recovered. Two different genotypic fusion combinations were carried out and several green plants were regenerated in one of them. With respect to plant habitus, leaf and inflorescence morphology, the regenerants had phenotypes intermediate between those of the parents. Southern hybridization analysis using a rice ribosomal DNA probe revealed that the regenerants contained both tall fescue- and Italian ryegrass-specific-DNA fragments. A cloned Italian ryegrass-specific interspersed DNA probe hybridized to total genomic DNA from Italian ryegrass and from the green regenerated somatic hybrid plants but not to tall fescue. Chromosome counts and zymograms of leaf esterases suggested nuclear genome instability of the somatic hybrid plants analyzed. Four mitochondrial probes and one chloroplast DNA probe were used in Southern hybridization experiments to analyze the organellar composition of the somatic hybrids obtained. The somatic hybrid plants analyzed showed tall fescue, additive or novel mtDNA patterns when hybridized with different mitochondrial gene-specific probes, while corresponding analysis using a chloroplast gene-specific probe revealed in all cases the tall fescue hybridization profile. Independently regenerated F. arundinacea (+) L. multiflorum somatic hybrid plants were successfully transferred to soil and grown to maturity, representing the first flowering intergeneric somatic hybrids recovered in Gramineae.  相似文献

Selected quality parameters were measured for forage leaf tissuefrom a spaced-plant nursery. The genotypes used were Ky 31 tallfescue and hybrids of Italian ryegrass (Lolium multiflorum Lam.)x tall fescue (Festuca arundinacea Schreb.) and tall fescuex giant fescue [Fescue gigantea (L ) Vill.]. Hybrid ploidy rangedfrom 2n = 28 to 84 chromosomes. Forage quality was characterizedby neutral detergent fibre (NDF), acid detergent fibre (ADF),total soluble carbohydrates (TSC) nutritive value index (NVI),hemicellulose, and in vitro dry matter disappearance (DMD). Quality of tall fescue, as measured by increased DMD, was improvedby hybridization with giant fescue. Improved DMD and NVI correlatedwith lower NDF and ADF in the hybrids. A few hybrids of Italianryegrass x tall fescue (2n = 28) were higher in some qualityparameters than Ky 31. Tall fescue x giant fescue hybrids (2n= 80 to 84), as a group, had significant quality improvementover Ky 31 in higher DMD and NVI and lower NDF and ADF. Whilesome individual hybrids within each group were significantlyhigher in quality, only the 2n = 80 to 84 chromosome group wasconsistently higher than Ky 31. Prediction equations for DMD,NDF, and ADF were established based on solvent extraction withnear-infrared reflectance spectroscopy (NIRS). Linear correlationcoefficients between chemical measurement and NIRS for eachquality parameter were 0–95 or higher. Effects of time of initial cut and subsequent cutting interval(1, 2 or 4 weeks) were examined in a glasshouse during summer1988 for reproductive plants of three prairie grass cultivars(Bromus willdenowii Kunth), Westerwolds ryegrass (Lolium multiflorumLam.) and perennial ryegrass (Lolium perenne L.). Measurementswere made of tiller and leaf numbers, sites of tillering, reproductivedevelopment, and herbage quality and yields. Effect of timeof initial cutting on regrowth appeared to be independent ofstage of reproductive development and unrelated to any of themeasured plant parameters. Characteristics for each cultivaras identified during undisturbed growth prior to the initialcuts, were modified by the subsequent cutting frequencies. Perennial ryegrass had the highest yields under frequent cuttingwith high herbage quality. Westenwolds ryegrass and the prairiegrass cultivars ‘Grasslands Matua’ and ‘Primabel’had the highest yields with infrequent cutting, but lower herbagequality than in perennial ryegrass. At each cut, tiller deathin prairie grass was determined by the number of reproductivetillers, and in the ryegrasses also by the numbers of elongatedvegetative tillers. Recovery of tiller numbers was rapid andprimarily from inhibited tiller buds at the base of reproductivetillers. In prairie grass, tiller numbers were relatively unaffectedby cutting frequency, but at frequent cutting, many axillarytillers originated from vegetative tillers rather than frominhibited tiller buds at the base of reproductive tillers. Little is known about interactions between seeds within theinflorescence of perennial ryegrass (Lolium perenneL.) and theireffect on final seed weight. These relations were investigatedin two glasshouse experiments using two genotypes, by reducingthe number of seeds in an inflorescence at anthesis. In thefirst experiment, entire spikelets were removed to study therelations between seeds in different spikelets. The effectson seed dry weight were not proportional to the number of spikeletsremoved. In one genotype, removal of two-thirds of the spikeletsincreased seed dry weight of the remaining seeds by 15%. Inthe other genotype, such treatment did not increase seed dryweight. In the second experiment, investigating seed interactionswithin a spikelet, either two proximal seeds or two centralseeds were maintained in a spikelet by removing the other ovulesin combination with no or 75% shading. Shading by 75% reducedseed dry weight by about 10%. In the unshaded treatment, seeddry weight was not affected by ovule removal. Under shading,the central seeds in a spikelet were about 12% heavier if theygrew alone, in contrast to the proximal seeds. The effects ofa reduction in seed number on seed dry weight were not relatedto the final nitrogen concentration of seeds. These experimentsshow that assimilate partitioning and the relationship betweenseeds in the inflorescence of perennial ryegrass are alreadylargely determined at anthesis; increasing seed yield by manipulationsafter anthesis is not feasible.Copyright 1998 Annals of BotanyCompany. The grass breeding industry is interested in a fast and cheap method of identifying contamination in seeds of Italian and perennial ryegrass (Lolium perenne L. and L. multiflorum Lam., respectively). This study shows that high-resolution melting curve analysis in combination with an unlabelled probe assay is an effective method of detecting single nucleotide polymorphisms (SNPs) in diverse Italian and perennial ryegrass backgrounds. This method proved efficient in differentiating ryegrass species and reducing the effect of additional DNA sequence polymorphisms close to the target SNP on the melting curve profiles. For the identification of contamination in Italian and perennial ryegrass seed production, high-resolution melting curve analysis shows great potential, as it is a single closed-tube PCR reaction with an easy workflow, providing results in <2 h after DNA extraction.  相似文献

Effects of cutting to 3 or 6 cm stubble height at frequenciesof 1, 2 or 4 weeks were investigated in young, vegetative, spacedplants of prairie grass (Bromus willdenowii Kunth), a tetraploidWesterwolds ryegrass (Lolium multiflorum Lam.) and perennialryegrass (Lolium perenne L.). Plants were grown for 8 weekson nutrient solution in a growth cabinet at day/night temperaturesof 15/10°C and in a short photoperiod. Leaf appearance,site filling and tiller numbers were all reduced by both increasedcutting frequency and a lower cutting height. The extent ofthese reductions was least in prairie grass and greatest inWesterwolds ryegrass. Relative growth rates in all species respondedin a similar manner with more frequent cutting and a lower cuttingheight. Cutting frequency had the greatest effect on growthrates, although the effect of cutting height increased withtime. Results confirm general recommendations of long intervalsbetween defoliations to achieve high yields for vegetative prairiegrass, but the relative amounts of dry matter harvested underthe different cutting regimes may be no different to that obtainedin ryegrass. Stubble height was of lesser importance in determiningyields. A typical U-shaped curve for depletion and recovery in water-solublecarbohydrates occurred after defoliation in all species, butlevels remained low at frequent cutting. Water-soluble carbohydratelevels in stubble and roots were higher in prairie grass. Regrowthat the end of the experiment was highly correlated with totalstubble and root weights (r = 0.84), while regrowth per tillershowed a good correlation with water-soluble carbohydrate content,although the response varied between species. Single plants of S24 perennial ryegrass (Lolium perenne L.) and S215 meadow fescue (Festuca pratensis Huds.) were transferred between, or exposed continuously to, contrasting light intensities obtained by decreasing the natural light in a glasshouse with Tygan shades. Inflorescence development in main shoots was studied by dissections of shoot apices, and by counts of branches and florets when ears emerged. Apical growth was slower, and spikelet initiation and inflorescence development were delayed or inhibited, in decreased light intensities. The number of main branches in the ear depended on the rate of apical growth before and after spikelet initiation, and on the time of spikelet initiation. In meadow fescue these processes were influenced by light intensity. Floret numbers per inflorescence branch were generally decreased by decreased light intensity. Most of the effects of light intensity on inflorescence development were smaller in ryegrass than in meadow fescue.  相似文献

In a field experiment the chlorophyll (a+b) (SPAD readings) and nitrogen contents of three grass species (Festuca arundinacea Schreb., Lolium perenne L., and Lolium multiflorum Lam.) and three intergeneric hybrids of Festuca pratensis Huds. × Lolium multiflorum Lam. and Festuca arundinacea Schreb. × Lolium multiflorum Lam. were measured. Close relationships were found between SPAD readings and nitrogen leaf content (r 2 = 0.873** 0.491** and 0.938**) for the 1st, 2nd, and 3rd cut, respectively. SPAD readings and N contents were closely correlated (r = 0.836**) confirming that SPAD measurements could be used in grass selection and/or breeding for high N concentration in herbage. Summary Many grasses are infected by endophytic fungi that grow intercellularly in leaves, stems, and flowers and are transmitted maternally by hyphal growth into ovules and seeds. The seed biology and seedling growth of endophyte-infected and uninfected perennial ryegrass (Lolium perenne) and tall fescue (Festuca arundinacea) were investigated under controlled environmental conditions. The percentage of filled seeds produced by infected tall fescue was over twice of uninfected tall fescue; infected and uninfected perennial reegrass had similar percentages. Weights of seeds from infected and uninfected plants were similar in both species. Seeds from infected plants of both species exhibited a higher rate of germination than seeds from uninfected plants. Shoot growth in the greenhouse was compared by making three sequential harvests of above-ground plant parts from infected and uninfected plants of both species. Infected perennial ryegrass plants produced significantly more biomass and tillers than uninfected plants after 6 and 10 weeks of growth and significantly more biomass after 14 weeks of growth. Infected tall fescue plants produced significantly more biomass and tillers than uninfected plants after 10 and 14 weeks of growth. The physiological mechanism of enhancement of growth is not known. The results of this study suggest that infected plants may have a selective advantage in populations with uninfected members.  相似文献

A detailed morphological study of three prairie grass cultivars(Bromus willdenowii Kunth) was conducted under ‘vegetative’and ‘reproductive’ growth conditions (short andlong photoperiods) and at different temperatures. Perennialryegrass (Lolium perenne L.) and Westerwolds ryegrass (Loliummuhiflorum Lam.) were compared during vegetative growth. Prairie grass had higher leaf appearance rates (leaves per tillerper day) and lower site filling (tillers per tiller per leafappearance interval) than the ryegrass species. Tillering rates(tillers per tiller per day) were also lower, except under vegetativeconditions at 4C. Low tiller number in prairie grass was notdue to lack of tiller sites but a result of poor filling ofthese sites. Lower site filling occurred because of increaseddelays in appearance of the youngest axillary tiller and lackof axillary tillers emerging from basal tiller buds. In prairiegrass, no tillers came from coleoptile buds while only occasionallydid prophyll buds develop tillers. Low tiller number in prairiegrass was compensated for by greater tiller weight. Prairiegrass had more live leaves per tiller, greater area per leafand a high leaf area per plant. Considerable variation between cultivars was found in prairiegrass. Perennial ryegrass (Lolium perenne L.) and Italian ryegrass (Lolium multiflorum Lam.) are important temperate forage grasses which are closely related, generating fertile interspecific hybrids. All groups are represented by multiple cultivars in the commercial pasture seeds market. Due to the close taxonomic relationship between the two species, differentiation based on morphophysiological criteria is not always readily achievable. In addition, an obligate outbreeding reproductive habit produces high levels of individual heterozygosity and intrapopulation diversity, which presents problems for discrimination between cultivars. Molecular genetic marker polymorphism provides an effective means of addressing these challenges. An iterative process of resequencing from loci distributed across the perennial ryegrass genome was used to identify single nucleotide polymorphism (SNP) markers, which were then validated and formatted in a highly multiplexed (384-plex) assay system. SNP genotyping was then performed across samples of 48–192 individuals from a total of 27 ryegrass cultivars (19 of perennial ryegrass, seven of Italian ryegrass and one hybrid cultivar). SNP markers from perennial ryegrass exhibited a high level of transfer to Italian ryegrass. Data analysis permitted quantification of intra- and inter-species diversity, as well as discrimination between cultivars within each species, including diploid and autotetraploid cultivars of perennial ryegrass. Lower levels of SNP-based diversity were detected in Italian ryegrass than in perennial ryegrass. A neighbour-joining tree based on genetic distance analysis located a hybrid cultivar to an intermediate position between the two species-specific cultivar groups. The resulting catalogue of ryegrass cultivars will provide support for the processes of cultivar accreditation and quality assurance.  相似文献

Airborne pollen and spore levels were monitored at seven sites in New Zealand using the Intermittent Cycling Rotorod sampler during the summer of 1988/1989. Grasses formed the major component of atmospheric pollen levels during spring and summer at every locality. Peak levels of grass and total pollen occurred during December or late November, with a slight latitudinal lag apparent at the more southern sites. Highest levels were recorded at the smaller rural centres of Gore and Kaikohe and the lowest at the larger urban centres of Auckland, Christchurch and Wellington. We make a first approximation of the likely risk to hayfever and allergic asthma patients at each of the seven centres. For example, significantly higher grass pollen levels were experienced at Kaikohe on 44% and 65% of days during November and December, compared with just 15% and 8% at Auckland. By recording the flowering seasons of the principal allergenic grass species at each locality, we determined the potentially allergenic grasses contributing to peak pollen levels, the most ubiquitous being tall fescue (Festuca arundinacea Schreb.), ryegrass (Lolium perenne L.,L. multiflorum Lam.), cocksfoot (Dactylis glomerata L.), Yorkshire fog (Holcus lanatus L.) and sweet vernal (Anthoxanthum odoratum L.). Perennial ryegrass (Lolium perenne L.) is a preferred choice for the turf grass industry due to its ability to provide a durable turf cover. Genetic or physical contamination of annual (L. multiflorum Lam.) or intermediate (L. hybridum) ryegrass species in perennial ryegrass is one of the major problems affecting the grass seed industry. At present, seedling root fluorescence (SRF), a biochemical marker, is used for the detection of annual ryegrass contamination. Due to the unreliability of the SRF test, the seed industry is seeking an alternative, more reliable and accurate detection method. Currently, there are no DNA tests available in ryegrass for detecting contamination with annual and intermediate ryegrass types. We developed a novel quantitative polymerase chain reaction (Q-PCR)-based DNA test for the detection of annual and/or intermediate ryegrass types in perennial ryegrass. This DNA test was designed using an insertion/deletion (InDel) site in the LpVRN2_2 (Vernalization 2) gene, which is one of the several genes controlling vernalization in ryegrass. The new DNA test is more reliable, accurate and cost-effective in detecting contamination, with a high sensitivity of 0.04% in a sample size of 5,000 seeds. Use of larger sample sizes (12.5-fold higher compared to SRF test) provided additional accuracy in detecting the level of contamination. The method has produced consistent results in 68 perennial, 26 annual and 14 intermediate ryegrass lines.  相似文献

Inter-specific relationships between mean seed weight, coleoptile + mesocotyl (sub-coleoptile internode) length and width, shoot strength and emergence from different sowing depths were examined for timothy (Phleum pratense L., mean seed weight 0.33-0.48 mg depending on seed line), cocksfoot (Dactylis glomerata L., 0.65-0.78 mg), perennial ryegrass (Lolium perenne L., 1.71-2.19 mg), tall fescue (Festuca arundinucea Schreb., 1.73–2.60 mg), annual ryegrass (Lolium multiflorum Lam., 5.10-5.20 mg) and prairie grass (Bromus willdenowü Kunth., 10.5–12.2 mg). Across species at 10,30 and 60 mm sowing depths in the field and 10, 15 and 30 mm sowing depths under controlled environment conditions, there was a significant (P < 0.05) positive correlation between emergence % and mean seed weight. Across species at 10–30 mm sowing depth under controlled environment conditions, emergence % was not significantly correlated with coleoptile + mesocotyl length but there were significant positive correlations between emergence % and coleoptile and mesocotyl width: shoot strength increased with increased coleoptile width across species. For seed lines of timothy of different mean seed weight (0.21-0.81 mg), emergence %, coleoptile + mesocotyl length and coleoptile and mesocotyl width increased with increased seed weight at 10 and 15 mm sowing depth. Shoot strength increased with increased coleoptile width for timothy. For emerged and non-emerged cocksfoot and timothy seedlings regardless of seed weight, mean coleoptile + mesocotyl length was > 10 mm at 10 mm sowing depth. It is concluded that at 10–30 mm sowing depth, increased emergence % with increased seed weight across species is not due to increased coleoptile + mesocotyl length. It is proposed that increased emergence % with increased seed weight across species at 10–30 mm sowing depths and across seed lines of timothy at 10 mm sowing depth is primarily due to increased coleoptile and mesocotyl width resulting in increased shoot strength and hence an increased ability to penetrate the substrate.  相似文献

Changes in light quality occur naturally within a canopy when a plant grows from unshaded to shaded conditions, and the reverse occurs after a cut that reduces shading. These changes in light quality could be responsible for the variation in leaf elongation and appearance rates of grasses. The role of blue light in leaf growth was investigated in tall fescue (Festuca arundinacea Schreb.) and perennial ryegrass (Lolium perenne L.). Leaf length was measured daily following a decrease or an increase in blue light to evaluate effects on duration of leaf growth, leaf elongation and the rate of leaf appearance rate. A reduction in blue light increased sheath length by 8 to 14% and lamina length by 6 to 12% for both species. These increases could be reversed by enrichment of blue light. With low blue light treatment, final leaf length was increased due to a greater leaf elongation rate. In tall fescue, but not in perennial ryegrass, this effect was coupled with a greater phyllochron and a longer duration of leaf elongation. Development of successive leaves on a tall fescue tiller were co-ordinated. A decrease in blue light increased the duration of elongation in the oldest growing leaf and also delayed the appearance of a new leaf, maintaining this co-ordination. We conclude that final leaf size and phyllochron for tall fescue can be significantly modified by blue light. Perennial ryegrass appeared less responsive, except for displaying longer sheaths and laminae in low blue light, as also occurred for tall fescue. Italian ryegrass (Lolium multiflorum Lam.) is the most widely cultivated annual forage grass in Japan. Lodging damage reduces both harvested yield and forage quality. To identify the chromosomal regions controlling lodging resistance in Italian ryegrass, we analyzed seven quantitative characters—heading date, plant height, culm weight, culm diameter, culm strength, tiller number, and culm pushing resistance—and evaluated lodging scores in the field in a two-way pseudo-testcross F1 population. Significant correlations between most combinations of the traits examined were found. Seventeen QTLs for all traits except culm weight were detected on six of seven linkage groups by simple interval mapping using cross-pollination (CP) algorithm, and 33 independent QTLs were also detected by composite interval mapping from both male and female parental linkage maps. In addition, up to 18 QTLs for lodging scores evaluated at nine different times were detected on all linkage groups. The flanking markers of those QTLs will serve as a useful tool for marker-assisted selection of lodging resistance in Italian ryegrass.  相似文献
