Experiments have been carried out with field-grown pear trees to investigate the effect of iron chlorosis on the composition of the leaf apoplast. Iron deficiency was associated with an increase in the leaf apoplastic pH from the control values of 5.5-5.9 to 6.5-6.6, as judged from direct pH measurements in apoplastic fluid obtained by centrifugation and fluorescence of leaves incubated with 5-CF. The major organic acids found in leaf apoplastic fluid of iron-deficient and iron-sufficient pear leaves were malate, citrate and ascorbate. The total concentration of organic acids was 2.9 mM in the controls and increased to 5.5 mM in Fe-deficient leaves. The total apoplastic concentration of inorganic cations (Ca, K and Mg) increased with Fe deficiency from 15 to 20 mM. The total apoplastic concentration of inorganic anions (Cl-, NO3-, SO4(2-) and HPO4(2-)) did not change with Fe deficiency. Iron concentrations decreased from 4 to 1.6 microM with Fe deficiency. The major Fe species predicted to exist in the apoplast was [FeCitOH](-1) in both Fe-sufficient and deficient leaves. Organic acids in whole leaf homogenates increased from 20 to 40 nmol x m(-2) with Fe deficiency. The accumulation of organic anions in the Fe-deficient leaves does not appear to be associated to an increased C fixation in leaves, but rather it seems to be a consequence of C transport via xylem.  相似文献

Capillary electromigration methods, zone electrophoresis (CZE), micellar electrokinetic chromatography (CMEKC) and isotachophoresis (CITP), have been used for analysis of water and water-buffer extracts from tree-common birch (Betula verrucosa) and grass-orchardgrass (Dactylis glomerata) pollen samples. Water extracts were analyzed by CZE using acetic acid as background electrolyte (BGE), by CMEKC in tris-phosphate BGE with anionic detergent sodium dodecyl sulfate (SDS) micellar pseudophase (TP-SDS) and by CITP in cationic mode with leading/terminating cations K+/BALA+ (beta-alanine (BALA)) and in anionic mode with leading/terminating anions Cl-/MES- (2-(N-morpholino)ethanesulphonic acid (MES)). Moreover, acetic acid extracts were analyzed by CZE using acetic acid as BGE, and alkaline water-SDS-buffer extracts were analyzed by CMEKC using TP-SDS as BGE. Extracted amounts of pollen allergens and other UV-absorbing compounds and the number of resolved components were evaluated from CZE, CMEKC and CITP analyses of the liquid extracts. Larger amounts of UV-absorbing material were found in the water-buffer pollen extracts than in the water extracts. More UV-absorbing material was found in all extracts from D. glomerata pollen than in relevant extracts from B. verrucosa pollen. It was found by CITP that the extracted amounts of anionic components and their number were much higher than those of cationic components. Concentrations of some inorganic ions (e.g. Cl-, K+, Na+, Ca2+) in pollen samples were also determined by CITP.  相似文献

Analyses have been made of the inorganic constituents of the juices expressed from the leaves of Rheum, Rumex, and Oxalis. It has been shown that in all cases there is a large excess of inorganic cations over anions in the sap, the average ratio of cations to anions being 3.8 (Part 1, p. 239). The ash analyses of plant tissues (chiefly leaves) reported in the literature have been examined critically, and it has been shown that the preponderance of inorganic cations over inorganic anions in the ash and in the sap is general. It has been concluded that the excess of inorganic cations is consistent with the view that cations pass into the protoplasm chiefly in the form of hydroxides, and are accumulated either in the form of organic salts (such as the oxalates) or in non-polar linkage. It has been concluded that practically all the potassium and sodium found in plant ash must have been present originally in the form of soluble ionogenic compounds, but that a considerable part of the calcium and magnesium may have been present originally in the form of insoluble salts or as components of non-polar compounds. The methods whereby the cations, particularly potassium, may have been accumulated have been discussed, and it has been concluded that as it does not seem very probable that they enter chiefly as nitrates or bicarbonates we may suppose that they go in to a large extent as hydrates: this is highly probable in the case which has been most carefully investigated (Valonia).  相似文献

The differences in chemical composition of leaves and stems of Ranunculus fluitans Lam. were investigated. Typical distribution of organic and inorganic compounds were generally found not to be influenced by factors as day-time, eutrophication, or age of the weed bed. Starch, sugars, amino acids, and organic acids were at a higher level in the stem, but inorganic cations and anions were accumulated in the leaf. The distribution pattern and the relative contents of individual sugars, acids, and inorganic ions are discussed in detail.  相似文献

1. Mustard plants have been grown under conditions in which the length of artificial day could be controlled. 2. Leaf samples were analysed for malic acid and citric acid, and for a number of inorganic anions and cations. A simple method is described by which sap was obtained from 0.5g. samples of leaves. 3. In days of 16hr. or more, malic acid was accumulated; the chief cation accumulated was calcium. 4. When the day-length was reduced the malic acid content decreased considerably but the calcium content remained the same. There was little change in the pH value of the sap, the balance of anions and cations having been maintained mainly by increases in citrate and nitrate contents. Analyses of the whole leaf still showed some deficiency in anion after sodium, potassium, calcium, magnesium, nitrate, sulphate, inorganic phosphate, chloride, malate and citrate had been accounted for. 5. Analyses at shorter intervals revealed a large diurnal variation in malic acid content, which increased during the first 5-6hr. of the light period, and fell during darkness. 6. The significance of these findings is discussed, and it is suggested that malic acid accumulation is a by-product of photosynthesis, calcium being taken up irreversibly to maintain anion/cation balance, and hence creating a continuing need for anions to balance it.  相似文献

The AMP deaminase isoenzymes from trout gill were activated by sodium and potassium, sodium being the most efficient. The optimal concentration for activation was 30-50 mM. The enzyme was sensitive to ionic strength, and imidazole was an inhibitor at concentrations higher than 25 mM. A possible regulation of gill AMP deaminase by intracellular imidazole buffers is discussed. AMP deaminase activity was tested in the presence of physiological concentrations of sodium and potassium. When the concentration of one of these cations was varied around its physiological concentration, the enzyme activity was relatively stable, indicating that the intracellular AMP deaminase activity would be insensitive to changes in the concentrations of monovalent cations. The effects of the sodium salts of different inorganic and organic anions were tested. Except chloride and gluconate, all were inhibitors of gill AMP deaminase.  相似文献

Ion chromatographic methods determined organic acids and mainnutrient minerals in the apoplastic solution from leaves ofseveral Fagaceae (Quercus ilex L., Quercus cerris L., Quercusvirgiliana (Ten.) Ten, and Fagus sylvatica L.). The anions oforganic acids found in high amounts (250 to 650 µM) werequinate, malate, and oxalate. Lactate, pyruvate, formate andacetate were detected in relatively low amounts with concentrationsbetween 20 and 200 µM. The total concentration of organicacids in the apoplastic sap ranged between 1.5 and 2 mM. Thetotal concentration of inorganic cations (K+, Mg2+, NH4+, Ca2+,Na+) and anions (C1, NO3, SO2–4 and PO3–4)in the apoplastic sap varied between 5 and 10 mM, and 0.35 and1.8 mM, respectively. We conclude that the concentration oforganic acid ions in the leaf apoplast depends mainly on theexchange with the leaf cells and is influenced by the electrochemicalgradient between the symplast and the apoplast in relation tothe water potential of the leaf. The determination of formateand acetate in the apoplastic compartment of leaves lend weightto the argument that the production of these acids by treesis a important emission source to the atmosphere. A procedure is described in which animal cells grown in culture on a dish are rapidly rinsed in situ with 0.25 M sucrose solutions for subsequent measurement of total, intracellular and rapidly exchangeab le Na+, K+, Mg2+ and Ca2+ by atomic absorption spectrophotometry. Repeated rinses with CO2-free (pH similar to 7) 0.25 M sucrose solution produced essentially no loss of cellular protein or cations. One 10-second rinse with CO2-saturated (pH 4) 0.25 M sucrose solution removed a rapidly proton exchangeable cellular cation fraction which is interpreted as being externally (membrane) bound. Rinses with physiological electrolyte solutions are shown to produce loss of cellular protein as well as displacement of surface exchangeable cations. Thus, isotonic sucrose solution is more satisfactory than electrolytic media for rinsing cultured cells prior to measurement of cellular cations. The technique employing sucrose rinse media is very rapid and reproducible and permits measurement of total, intracellular or surface bound Na+, K+, Mg2+ and Ca2+ in the same sample.  相似文献

A simple, reliable and rapid reversed-phase HPLC-PAD procedure for the characterisation and quantitative determination of the anti-diabetic sesquiterpene lactone enhydrin (1) from Smallanthus sonchifolius (yacón) has been evaluated and validated. The approach focused on the analysis of various leaf rinse extracts, as well as the glandular trichomes of intact leaves, in which 1 was the major compound detected. The best sample preparation of a rinse extract yielded 0.67 mg/mL of 1, whilst a rapid rinse of a small piece of one dried leaf gave 0.09 mg/mL of 1; the highest concentration obtained from a glandular extract was 0.07 mg/mL. The dried leaves of S. sonchifolius were found to contain a total of 0.97% of 1.  相似文献

Spatial compression among the longer DNA fragments occurs during DNA electrophoresis in agarose and non-agarose gels when using certain ions in the conductive buffer, impairing the range of fragment sizes resolved well in a single gel. Substitutions using various polyhydroxyl anions supported the underlying phenomenon as the complexation of Lewis acids to DNA. We saw significant improvements using conditions (lithium borate 10 mM cations, pH 6.5) favoring the formation of borate polyanions and having lower conductance and Joule heating, delayed electrolyte exhaustion, faster electrophoretic run-speed, and sharper separation of DNA bands from 100bp to 12 kb in a single run.  相似文献

This study evaluated the ability of gamma-aminobutyric acid (GABA), baclofen, monovalent anions, divalent cations, and various combinations thereof to protect solubilized benzodiazepine (BZ) receptors of types 1 and 2, when contained together on the complex, against heat inactivation. Neither anions, cations, nor GABA alone provided significant protection of solubilized BZ receptors against heat, but inclusion of monovalent anions or divalent cations together with 500 microM GABA did afford protection. Monovalent anions combined with GABA (500 microM) provided 50% to full protection. Divalent cations, such as CaCl2 (2.5 mM) or MgCl2 (2.5 mM) in the presence of GABA (500 microM) yielded 45% and 24% protection, respectively. Other divalent cations tested (Zn2+, Hg2+, Co2+, and Ni2+) were poor protectors, even when combined with GABA. Monovalent anions (200 mM NaCl) and divalent cations (5 mM CaCl2) when tested together provided no protection. Similarly, baclofen (the GABA-B agonist) provided no protection, either alone or together with anions or divalent cations. These results indicate that the independent but interacting recognition sites of GABA, BZ, anions, and divalent cations, previously detected in the membrane-bound state, are retained in the solubilized state.  相似文献

Two tobacco lines with (Nicotiana tabacum cv. Gatersleben, WT) or without (transformant LNR-H) nitrate reductase in roots were chosen as model systems to re-evaluate the role of root nitrate reduction for survival of anoxia. In this first paper, the two hydroponically grown lines were compared with respect to their root morphology, root respiration and the root content of inorganic cations, anions, and metabolites. Leaf transpiration in relation to root morphology was also determined. In comparison to WT roots containing NR, the NR-free LNR-H transformants had slightly shorter and thicker roots with a lower root surface area per g leaf FW. Consistent with that, LNR-H leaves had lower transpiration rates than WT. LNR-H-roots also showed consistently higher respiration and higher contents of ATP, starch and hexose monophosphates than WT roots. Concentrations of free sugars were only slightly higher in LNR-H roots. Total soluble protein content was identical in both lines, whereas amino acids were higher in LNR-H. Contents of major inorganic cations and anions were also almost identical in both lines. We conclude that WT versus LNR-H plants are a suitable tool to re-evaluate the role of nitrate reduction in flooding tolerance.  相似文献

The herbivory activity of the bordered patch larvae (Chlosyne lacinia, Lepidoptera) on leaves of a Brazilian population of Tithonia diversifolia and the antifeedant potential of its leaf rinse extract were investigated. The caterpillars fed only on the adaxial face, where the density of glandular trichomes is very low, and avoided the abaxial face, which contains high levels of trichomes. Deterrent activity against the larvae was observed in leaf discs treated with leaf rinse extract at concentrations of 1-5% of fresh leaf weight. High-performance liquid chromatography (HPLC) analysis indicated that sesquiterpene lactones are the main constituents of the glandular trichomes. Dichloromethane rinse extracts of the leaves and inflorescences were chemically investigated, and 16 compounds were isolated and identified: 14 sesquiterpene lactones, a flavonoid and a diterpenoid. In this study, five sesquiterpene lactones are described for the first time in the genus, including two lactones, one of which has an unusual seco-guaianolide skeleton. Our findings indicate that the caterpillars avoid the sesquiterpene-lactone-rich glandular trichomes, and provide evidence for the antifeedant activity of the dichloromethane leaf rinse extract. In addition, a study of the seasonal variation of the main constituents from the leaf surface throughout a year demonstrated that a very low qualitative but a very high quantitative variation occurs. The highest level of the main metabolite tagitinin C was observed between September and October and the lowest was from March to June, the later corresponding to the period of highest infestation by the larvae.  相似文献

Nitric oxide (NO), a small diffusible, ubiquitous bioactive molecule, acts as prooxidant as well as antioxidant, and also regulates remarkable spectrum of plant cellular mechanisms. The present work was undertaken to investigate the role of nitric oxide donor sodium nitroprusside (SNP) and/or calcium chloride (CaCl(2)) in the tolerance of excised mustard leaves to salt stress. After 24h, salt stressed leaves treated with SNP and/or CaCl(2), showed an improvement in the activities of carbonic anhydrase (CA) and nitrate reductase (NR), and leaf chlorophyll (Chl) content, leaf relative water content (LRWC) and leaf ion concentration as compared with the leaves treated with NaCl only. Salinity stress caused a significant increase in H(2)O(2) content and membrane damage which is witnessed by enhanced levels of thiobarbituric acid reactive substances (TBARS) and electrolyte leakage. By contrast, such increases were blocked by the application of 0.2mM SNP and 10mM CaCl(2) to salt stressed leaves. Application of SNP and/or CaCl(2) alleviated NaCl stress by enhancing the activities of antioxidative enzymes viz. superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX) and glutathione reductase (GR) and by enhancing proline (Pro) and glycinebetaine (GB) accumulation with a concomitant decrease in H(2)O(2) content, TBARS and electrolyte leakage, which is manifested in the tolerance of plants to salinity stress. Moreover, application of SNP with CaCl(2) was more effective to reduce the detrimental effects of NaCl stress on excised mustard leaves. In addition to this, ameliorating effect of SNP was not effective in presence of NO scavenger cPTIO [2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide]. To put all these in a nut shell, the results advocate that SNP in association with CaCl(2) plays a role in enhancing the tolerance of plants to salt stress by improving antioxidative defence system, osmolyte accumulation and ionic homeostasis.  相似文献

Capillary zone electrophoresis (CZE) and capillary isotachophoresis (CITP) were applied for the determination of peptide purity degree and counter-ion content in lecirelin, the synthetic analogue of luteinizing hormone-releasing hormone (LHRH). CZE analyses were carried out in acidic background electrolyte (100 mM H3PO4, 50 mM Tris, pH 2.25) in bare fused silica capillary using UV-absorption detection at 206 nm. CITP analyses were performed in the electrophoretic analyzer with column coupling, equipped with contactless conductivity detectors both in preseparation capillary and in analytical capillary, and with UV-absorption detector (220 and 254 nm) in analytical capillary. Determinations of peptide purity were carried out in cationic mode with leading electrolyte (LE), 10 mM KOH/AcOH, pH 4.5, and terminating electrolyte (TE), 10 mM beta-alanine (BALA)/AcOH, pH 4.4. Degree of peptide purity determined by both CZE and CITP was in the range 60.1-80.9% for crude preparations of lecirelin and in the range 96.4-99.9% for HPLC purified batches. Concentrations of contaminating counter-ions, the anions of trifluoromethanesulfonic acid (TFMSA), trifluoroacetic acid (TFA) and acetic acid (AcOH), were determined by CITP analyses in anionic mode with LE 10 mM HCl/His, pH 6.0, and TE 10 mM 2-(N-morpholino)-ethanesulfonic acid (MES), pH 4.0, by the calibration curve method. Mass percentages of the counterion contents in the analyzed lecirelin batches varied from zero to ca. 9% (TFMSA), 3% (TFA) and 11% (AcOH), respectively.  相似文献

A micellar electrokinetic capillary chromatographic method to separate prednisolone, prednisolone acetate, naphazoline, Zn-bacitracin, sulfacetamide and phenylefrine is described. The separation was carried out by using a fused-silica capillary (57 cmx75 micrometer I.D.) at 25 degrees C and 30 kV, using a 5 mM phosphate-5 mM borate buffer adjusted to pH 8.2, 50 mM sodium dodecyl sulfate (SDS) and 10% methanol-water (v/v) as background electrolyte. Under these conditions, the run time was 8 min and the limits of quantification were about 1.0 mg/l for every component. The method was applied to pharmaceutical preparations and the results provided recoveries close to 100% and the method gave good results when compared with a reference multivariate calibration spectrophotometric method.  相似文献

Many plants are exposed to prolonged episodes of anthropogenic acid precipitation with pH values of 4 or less, but there is little evidence of widespread direct damage to the plant cells. Acids appear to permeate leaf cuticle via charged pores, which act as a fixed buffer that delays but does not stop acid movement. We investigated the effect of cations on the movement of protons through astomatous isolated leaf cuticles of pear (Pyrus communis L.) and rough lemon (Citrus limon [L.] Burm. fils cv Ponderosa). Chloride salt solutions of Na, K, Ca, Cd, Mg, Gd, or Y in a diffusion apparatus were applied to the morphological inner surface of the cuticle, while the outer surface faced a large volume of pH 3 or 4 sulfuric acid. Effective permeability was calculated from the change in the pH of the inner solution as measured with a pH microelectrode. Monovalent cations caused either no change (pear) or promotion (rough lemon) of proton movement. Divalent cations reduced proton movement in a concentration-dependent manner (both species), whereas trivalent cations (rough lemon only) caused the effective permeability to decrease to near zero. Inhibition by 10 mM CaCl2 was reversed with water. The effects of these cations on the permeability of cuticles to protons was used to elucidate mechanisms by which cations can protect leaves from acid precipitation in nature.  相似文献

Carbonic anhydrase (CA, carbonate hydro-lyase EC 4.2.1.1) from lysed red blood cells of the channel catfish (Ictalurus punctatus) was studied to determine physico-chemical kinetic properties and relative sensitivity to 56 chemicals of significance in water quality research. In general, enzyme activity was strongly reduced by heavy metal inorganic cations; less strongly by organometallic cations, some anions, and certain pesticides; and weakly inhibited by light metal cations and organometallic and organic compounds. Spectrophotometric transmittance and reflectance curves were recorded for wavelengths from 0.45 (in some cases 0.34) to 2.7 micrometers for faces and backs of leaves and for stacked leaves of several plant species. Measurements were made at different angles of illumination. Leaf spectrophotometric curves were compared with curves for leaf extracts, potato tuber tissue, glass beads in water, and frozen leaves to demonstrate the physical bases for the leaf curves. Leaves were infiltrated with liquids of different refractive indices for further comparison of spectrophotometric curves. Goniophotometric reflectance curves were recorded, giving visible reflectance and degree of polarization as functions of viewing angle for two different angles of illumination.  相似文献

Studies were conducted to compare the modulation of β-nicotinamide adenine dinucleotide (NADH): nitrate reductase (NR; EC 1.6.6.1) and sucrose-phosphate synthase (SPS: EC 2.4.1.14) with respect to regulation by the inorganic anions, phosphate (P1), sulfate and tungstate. Following inactivation of both enzymes in vivo by transferring spinach plants ( Spinacia oleracea L. cv. Bloomsdale) to a darkened growth chamber, spontaneous reactivation occurred in vitro when desalted leaf extracts were preincubated at 25°C prior to assay. All three inorganic anions inhibited SPS activation in vitro and also reduced the light activation of SPS in situ when they were fed to excised leaves via the transpiration stream. As expected, feeding tungstate to excised leaves prevented the light-dependent increase in extractable NR activity. However, in contrast to SPS, the light activation of NR in situ was relatively unaffected by Pi, and sulfate, and in vitro, both anions stimulated (rather than inhibited) the reactivation of NR. Part of the stimulation by Pi and sulfate was the result of increased ionic strength, and stimulation could also be demonstrated with other inorganic and organic salts. In the presence of high ionic strength (0.1 to 0.2 M KCl) the rate of NR activation in vitro was relatively constant when the pH of the preincubation medium was varied from pH 6.5 to 8.0, whereas in the absence of added salt the rate of activation was nearly zero at pH 6.5 but increased progressively as pH was raised. The stimulation by salts could be reversed, in part, by glycerol and ethylene glycol suggesting that hydrophobic interactions might play some role in the activation of NR.  相似文献
