Caffeic acid (CA), which is ubiquitously present in plants, is a potent phytotoxin affecting plant growth and physiology. The aim of our study was to investigate whether CA-induced inhibition of adventitious root formation (ARF) in mung bean {Vigna radiata (L.) Wilczek [Phaseolus aureus Roxb.]} involves the induction of conventional stress responses. The effect of CA (0–1000 μM) on ARF in mung bean was determined by measuring the generation of reactive oxygen species (ROS) in terms of malondialdehyde and hydrogen peroxide (H2O2) content, root oxidizability and changes in levels of antioxidant enzymes. Our results show that CA significantly enhanced MDA content, indicating severe lipid peroxidation, and increased H2O2 accumulation and root oxidizability in the lower rooted hypocotylar region (LRHR) of mung bean, thereby inducing oxidative stress and cellular damage. In response to CA, there was a significant upregulation in the activities of scavenging enzymes, such as superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase, catalase and glutathione reductase, in LRHRs of mung bean. Based on these results, we conclude that CA inhibits ARF in mung bean hypocotyls by inducing ROS-generated oxidative stress and upregulating the activities of antioxidant enzymes.  相似文献

Roots of Panax ginseng exposed to various concentrations of Cu (0.0, 5, 10.0, 25.0, and 50.0 μM) accumulated high amounts of Cu in a concentration-dependent and duration-dependent manner. Roots treated with 50 μM Cu resulted in 52% and 89% growth inhibition after 20 and 40 days, respectively. Saponin synthesis was stimulated at a Cu concentration between 5 and 25 μM but decreased at 50 μM Cu. Malondialdehyde content (MDA), lipoxygenase activity (LOX), superoxide ion (O2 •−) accumulation, and H2O2 content at 5 and 10 μM Cu-treated roots were not increased but strongly increased at 50 μM Cu resulting in the oxidation of ascorbate (ASC) and glutathione (GSH) to dehydroascorbate (DHA) and glutathione disulfide (GSSG), respectively indicating a clear oxidative stress. Seven well-resolved bands of superoxide dismutase (SOD) were detected in the gel and an increase in SOD activity seemed to be mainly due to the induction of Fe-SOD 3. Five to 10 μM Cu slightly induced activity of ascorbate peroxidase (APX) and dehydroascorbate reductase (DHAR), guaiacol peroxidase (G-POD) but inhibited monodehydroascorbate reductase (MDHAR) and glutathione reductase (GR) enzyme activities. No changes in catalase (CAT) activity and in activity gel were found up to 25 μM Cu, but both G-POD and CAT activities were inhibited at 50 μM Cu. Glutathione metabolism enzymes such as γ-glutamylcysteine synthetase (γ-GCS), glutathione-S-transferase (GST), and glutathione peroxidase activities (GPx) were activated at 5 and 10 μM Cu but were strongly inhibited at 50 μM Cu due to the Cu accumulation in root tissues. The strong depletion of GSH at 50 μM Cu was associated to the strong induction of γ-glutamyltranspeptidase (γ-GGT) activity. These results indicate that plant could grow under Cu stress (5–25 μM) by modulating the antioxidant defense mechanism for combating Cu induced oxidative stress.  相似文献

Effects of exogenous nickel (Ni: 10 and 200 μM) on growth, mitotic activity, Ni accumulation, H2O2 content and lipid peroxidation as well as the activities of various antioxidative enzymes, such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and glutathione peroxidase (GSH-Px) were investigated in wheat roots. A considerable Ni accumulation in the roots occurred at both the concentrations. Although Ni at 10 μM did not have any significant effect on root growth, it strongly inhibited the root growth at 200 μM. Mitotic activity in the root tips was not significantly affected by exposure of the seedlings to 10 μM Ni; however, it was almost completely inhibited at 200 μM treatment. Ni stress did not result in any significant changes in CAT and APX activities as well as lipid peroxidation. However, H2O2 concentration increased up to 82% over the control in the roots of seedlings exposed to 200 μM Ni. There was a significant decline in both SOD (50%) and GSH-Px (20–30%) activities in the roots when the seedlings were treated with 200 μM Ni. The results indicated that a strong inhibition of wheat root growth caused by Ni stress was not due to enhanced lipid peroxidation, but might be related to the accumulation of H2O2 in root tissue.  相似文献

Senescence is a developmentally regulated and highly ordered sequence of events. Senescence leads to abscission of plant organs and eventually leads to death of a plant or part of it. Present study revealed that Phalaenopsis flower undergo senescence due to over activation of O2 ·−generating xanthine oxidase (XO), which consequently increases the concentrations of O2 ·− leading to enhanced oxidative damage and disturbed cellular redox environment as indicated by increased lipid peroxidation and DHA/AsA + DHA ratio, respectively. While activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), and non-specific peroxidase (POD) were enhanced in sepals and petals of old flower, activities of catalase (CAT) and glutathione reductase (GR) were decreased. Exogenous application of nitric oxide (NO) retarded H2O2-induced senescence of Phalaenopsis flower by downregulating activity of XO and concentrations of O2 ·−, H2O2 and malondialdehyde (MDA, an index of lipid peroxidation). Exogenous application of NO also downregulated SOD activity and upregulated antioxidant enzymes involved in the detoxification of H2O2 (CAT and APX), and in the regulation of redox couples viz, monodehydroascorbate reductase (MDHAR) and GR, together with the modulation in non-protein thiol status and DHA/AsA + DHA ratio.  相似文献

The adverse effects of arsenic (As) toxicity on seedling growth, root and shoot anatomy, chlorophyll and carotenoid contents, root oxidizability (RO), antioxidant enzyme activities, H2O2 content, lipid peroxidation and electrolyte leakage (EL%) in common bean (Phaseolus vulgaris L.) were investigated. The role of exogenous nitric oxide (NO) in amelioration of As-induced inhibitory effect was also evaluated using sodium nitroprusside (100 μM SNP) as NO donor and 2-(4-carboxy-2-phenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (200 μM PTIO) as NO scavenger in different combinations with 50 μM As. As-induced growth inhibition was associated with marked anomalies in anatomical features, reduction in pigment composition, increased RO and severe perturbations in antioxidant enzyme activities. While activity of superoxide dismutase and catalase increased, levels of ascorbate peroxidase, dehydroascorbate reductase and glutathione reductase decreased significantly and guaiacol peroxidase remained normal. The over-accumulation of H2O2 content along with high level of lipid peroxidation and electrolyte leakage indicates As-induced oxidative damage in P. vulgaris seedlings with more pronounced effect on the roots than the shoots. Exogenous addition of NO significantly reversed the As-induced oxidative stress, maintaining H2O2 in a certain level through balanced alterations of antioxidant enzyme activities. The role of NO in the process of amelioration has ultimately been manifested by significant reduction of membrane damage and improvement of growth performance in plants grown on As + SNP media. Onset of oxidative stress was more severe after addition of PTIO, which confirms the protective role of NO against As-induced oxidative damage in P. vulgaris seedlings.  相似文献

Comparative study about the salt-induced oxidative stress and lipid composition has been realised in primary root tissues for two varieties of maize (Zea mays L.) in order to evaluate their responses to salt stress. The root growth, root water content (WC), hydrogen peroxide (H2O2) generation, lipid peroxidation, membrane stability index and the changes in the profile of fatty acids composition were investigated. Salinity impacts in term of root growth, water content, H2O2 generation, lipid peroxidation and membrane destabilisation were more pronounced in primary roots of Aristo than in those of Arper indicating more sensitivity of the first variety. It was confirmed by gas chromatography that the composition of fatty acids in roots of both varieties was constituted mainly by 16:0 and 18:0 as major saturated fatty acids and 18:1ω9, 18:2ω6 and 18:3ω3 as major unsaturated fatty acids. Total lipid extracts from the roots of both varieties showed that the lipid saturation level increased under salt stress, notwithstanding the increased proportion of polyunsaturated fatty acids. The changes in lipid saturation being predominantly due to decreases in oleic acid (18:1ω9) and increases in palmitic acid (16:0). However, Arper root extracts contained a lower proportion of saturated lipids than Aristo. The enhanced proportion of highly polyunsaturated fatty acids especially linolenic and eicosapentaenoic acids was considered to be the characteristic of the relatively salt tolerance in Arper roots.  相似文献

Seedlings of rice (Oryza sativa L.) cv. Pant-12 grown in sand cultures containing 200 and 400 μM NiSO4, showed a decrease in length and fresh weight of roots and shoots. Nickel was readily taken up by rice seedlings and the concentration was higher in roots than shoots. Nickel-treated seedlings showed increased rates of superoxide anion (O2 •− ) production, elevated levels of H2O2 and thiobarbituric acid reactive substances (TBARS) demonstrating enhanced lipid peroxidation, and a decline in protein thiol levels indicative of increased protein oxidation compared to controls. With progressively higher Ni concentrations, non-protein thiol and ascorbate (AsA) increased, whereas the level of low-molecular-weight thiols (such as glutathione and hydroxyl-methyl glutathione), the ratio of these thiols to their corresponding disulphides, and the ratio of AsA to dehydroascorbic acid declined in the seedlings. Among the antioxidant enzymes studied, the activities of all isoforms of superoxide dismutase (Cu-Zn SOD, Mn SOD and Fe SOD), guaiacol peroxidases (GPX) and ascorbate peroxidase (APX) increased in Ni-treated seedlings, while no clear alteration in catalase activity was evident. Activity of the ascorbate-glutathione cycle enzymes monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione reductase (GR)—significantly increased in Ni-treated seedlings. However such increase was apparently insufficient to maintain the intracellular redox balance. Results suggest that Ni induces oxidative stress in rice plants, resulting in enhanced lipid peroxidation and decline in protein thiol levels, and that (hydroxyl-methyl) glutathione and AsA in conjunction with Cu-Zn SOD, GPX and APX are involved in stress response.  相似文献

Hydrogen peroxide (H2O2), an active oxygen species, is widely generated in many biological systems. The present study demonstrates that H2O2 was generated in seedling explants after the primary roots were removed, and it mediates the auxin response prior to adventitious root formation in cucumber (Cucumis sativus L. Ganfeng 8). When compared with the controls, treatment of cucumber seedling explants after primary roots removal with either 20–40 mM H2O2 or 10 μM IAA significantly increased the number of adventitious roots, and treatment with 10–50 mM H2O2 significantly increased the fresh weight of adventitious roots. The effects of H2O2 on promoting the formation and growth of adventitious roots were eliminated by 2 mM ascorbic acid, 100 U CAT or 1 μM DPI, and the effects of IAA were eliminated by 4 mM ascorbic acid, 100 U CAT or 5 μM DPI. Treatment with either 4 mM ascorbic acid or 1–5 μM DPI inhibited the formation and growth of adventitious roots, and these inhibitory effects were partly reversed by exogenous H2O2.Furthermore, a higher concentration of endogenous H2O2 was detected in seedling explants 3 h after the primary roots were removed. However, in 10 μM DPI-treated seedling explants, the concentration of endogenous H2O2 was markedly reduced by DPI. Results obtained suggest that H2O2 may function as a signaling molecule, involved in the formation and development of adventitious roots in cucumber.  相似文献

Rice (Oryza sativa L.) seedlings stressed with CdCl2 (0.5 mM or 50 μM) showed typical Cd toxicity (leaf chlorosis, decrease in chlorophyll content, or increase in H2O2 and malondialdehyde contents). Rice seedlings pretreated with heat shock at 45°C (HS) for 2 or 3 h were protected against subsequent Cd stress. Rice seedlings pretreated with HS had similar Cd concentration in leaves caused by CdCl2 as those non-HS. The content of H2O2 increased in leaves 1 h after HS exposure. However, APX and GR activities were higher in HS-treated leaves than their respective control, and it occurred after 2 h of HS treatment. Pretreatment of rice seedlings with H2O2 under non-HS conditions resulted in an increase in APX, GR, and CAT activities and protected rice seedlings from subsequent Cd stress. HS-induced H2O2 production and protection against subsequent Cd stress can be counteracted by imidazole, an inhibitor of NADPH oxidase complex. Results of the present study suggest that early accumulation of H2O2 during HS signals the increase in APX and GR activities, which in turn prevents rice seedlings from Cd-caused oxidative damage.  相似文献

The present study investigated the effects of aluminum on lipid peroxidation, accumulation of reactive oxygen species and antioxidative defense systems in root tips of wheat (Triticum aestivum L.) seedlings. Exposure to 30 μM Al increased contents of malondialdehyde, H2O2, suproxide radical and Evans blue uptake in both genotypes, with increases being greater in Al-sensitive genotype Yangmai-5 than in Al-tolerant genotype Jian-864. In addition, Al treatment increased the activity of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), glutathione reductase (GR) and glutathione peroxidase (GPX), as well as the contents of ascorbate (AsA) and glutathione (GSH) in both genotypes. The increased activities SOD and POD were greater in Yangmai-5 than in Jian-864, whereas the opposite was true for the activities of CAT, APX, MDHAR, GR and GPX and the contents of AsA and GSH. Consequently, the antioxidant capacity in terms of 2,2-diphenyl-1-picrylhydrazyl (DPPH)-radical scavenging activity and ferric reducing/antioxidant power (FRAP) was greater in Jian-864 than in Yangmai-5.  相似文献

Changes in contents of reactive oxygen species (O2 and H2O2) and non-enzymatic antioxidants, activities of antioxidant enzymes and lipid peroxidation were investigated during senescence of detached cucumber cotyledons dipped in water (control) and 20 mg dm−3 triadimefon (TDM). O2 and H2O2 accumulation and lipid peroxidation were observed during senescence of cucumber cotyledons, which coincided with a drop in the contents of carotenoids (Car) and ascorbic acid (AsA), and the activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), and an increase in the activity of peroxidase (POD). However, TDM could significantly inhibit the accumulation of O2 and H2O2, and lipid peroxidation by preventing the decrease of CAT, APX, Car and AsA and the increase of POD, while TDM had little effect on SOD activity during the senescence. Therefore we can draw a conclusion that TDM protects the membrane system and retards the senescence of detached cucumber cotyledons. During antioxidant screening using 1,1-diphenyl-picrylhydrazyl (DPPH) and a lipid peroxidation assay, a streptomycete strain was found to produce herbimycin A and dihydroherbimycin A as antioxidants in the culture filtrate. These molecules were identified by using spectral analyses, including infrared, ultraviolet, mass spectrum, and nuclear magnetic resonance assays. In the DPPH radical-scavenging assay, dihydroherbimycin A exhibited more potent antioxidant activity (IC50, 1.3 μM) than α-tocopherol (IC50, 2.7 μM) that was used as a reference compound. In the lipid peroxidation assay, both herbimycin A and dihydroherbimycin A demonstrated antioxidant activities of 61% and 72%, respectively, at 100 μg/ml, while α-tocopherol exhibited an activity of 93% at the same concentration. Therefore, dihydroherbimycin A might have the potential to be developed into a new therapeutic agent.  相似文献

In the present study, it was aimed to investigate the influence of exogenous mammalian sex hormones (MSH) (progesterone, β-estradiol and androsterone) on the morphological (root and shoot growth) and biochemical parameters (protein and sugar content, antioxidant enzyme activities, and lipid peroxidation and H2O2 levels) of chickpea (Cicer arietinum L.) plants growing under control conditions. The solutions of hormones prepared at different concentrations (10−4, 10−6, 10−9, 10−12 and 10−15 M) were sprayed once on the leaves of 7-day plants. The plants were harvested on 18 days after the hormone treatment. Although all of the hormones at the tested concentrations significantly increased plant growth, soluble protein and sugar contents, and antioxidant enzyme activities [superoxide dismutase (SOD), peroxidase (POX) and catalase (CAT)], they decreased H2O2 content and lipid peroxidation level when compared with control plants. The activities of SOD, POX and CAT reached to the highest levels at 10−6 M for progesterone, and 10−9 M for β-estradiol and androsterone, which maximum growth, protein and sugar contents were determined. The same concentrations also resulted in the lowest levels for H2O2 content and lipid peroxidation. It can be interpreted that the MSH improve plant growth and development by affecting some biochemical parameters including antioxidative system.  相似文献

The effects of excess copper (Cu) on the accumulation of hydrogen peroxide (H2O2) and antioxidant enzyme activities in roots of the Cu accumulator Elsholtzia haichowensis Sun were investigated. Copper at 100 and 300 μM significantly increased the concentrations of malondialdehyde and H2O2, and the activities of catalase (E.C. 1.11.1.6), ascorbate peroxidase (E.C. 1.11.1.11), guaiacol peroxidase (GPOD, E.C. 1.11.1.7) and superoxide dismutase (SOD, E.C. 1.15.1.1). Isoenzyme pattern and inhibitor studies showed that, among SOD isoforms, only copper–zinc superoxide dismutase (CuZn–SOD) increased. Excess Cu greatly increased the accumulation of superoxide anion (O2 ·−) and H2O2 in E. haichowensis roots. This study also provides the first cytochemical evidence of an accumulation of H2O2 in the root cell walls as a consequence of Cu treatments. Experiments with diphenyleneiodonium as an inhibitor of NADPH oxidase, 1,2-dihydroxybenzene-3,5-disulphonic acid as an O2 ·− scavenger, and N-N-diethyldithiocarbamate as an inhibitor of SOD showed that the source of H2O2 in the cell walls could partially be NADPH oxidase. The enzyme can use cytosolic NADPH to produce O2 ·−, which rapidly dismutates to H2O2 by SOD. Apoplastic GPOD and CuZn–SOD activities were induced in roots of E. haichowensis with 100 μM Cu suggesting that these two antioxidant enzymes may be responsible for H2O2 accumulation in the root apoplast.  相似文献

Cadmium-induced initial changes in the production of reactive oxygen species (ROS) and antioxidant mechanism were investigated in soybean (Glycine max L. cv. Don Mario 4800 RR) leaves. Whole plants (WP) and plants without roots (PWR) were exposed to 0.0, 10.0 and 40.0 μM Cd for 0, 4, 6 and 24 h. Compared to PWR, a higher level of endogenous Cd in WP was associated with a lower oxidative stress measured in terms of lipid peroxidation. Furthermore, O2 •− content decreased in the leaves of Cd-treated WP, whereas it increased in those of Cd-treated PWR. Although O2 •− accumulation in PWR was associated with a decrease in superoxide dismutase (SOD) activity, O2 •− diminution in WP leaves was not related to any increase in SOD activity. H2O2 content increased in the leaves of both Cd-treated WP and PWR, and it was concomitant with a corresponding decline in catalase (CAT) and ascorbate peroxidase (APX) activities. When diphenyl iodonium (DPI), an inhibitor of NADPH oxidase, was added, H2O2 content remained unchanged in Cd-treated WP, suggesting that NADPH oxidase does not participate in the early hours of Cd toxicity. Taken together, our results showed that early ROS evolution and oxidative damage were different in WP and PWR. This suggests that the response in soybean leaves during the early hours of Cd toxicity is probably modulated by the root.  相似文献

In order to investigate the effects of spermidine (Spd) and spermine (Spm) on cadmium stress, the content of chlorophyll, hydrogen peroxide (H2O2), malondialdehyde (MDA), soluble protein and proline, the rate of O2·− generation, and activities of antioxidant enzymes (superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), and glutathione reductase (GR)) in Potamogeton malaianus Miq. were measured. Exogenous application of Spd or Spm significantly enhanced the level of proline, retarded the loss of chlorophyll, enhanced photosynthesis, decreased the rate of O2·− generation and H2O2 content, and prevented Cd-induced lipid peroxidation. Spd and Spm also effectively maintained the balance of antioxidant enzyme activities under Cd stress; however, GR activity was found to increase only slightly in response to polyamines (PAs). The antioxidant systems, which were modified by PAs, were able to moderate the radical-scavenging system and to lessen in this way the oxidative stress. These results suggest that both Spd and Spm can enhance Cd tolerance of P. malaianus.  相似文献

Phaseolus aureus Roxb. was exposed to HgCl2 and Cd(NO3)2 either at the germination stage in concentration 0.5, 5 and 25 μM for 48 and 96 h, or at the seedling stage (5th day of germination) in concentration 0.5, 5 and 20 μM for 6, 24 and 48 h. The germination and the growth of roots (germination stage treatment) were less in Hg than in Cd treatment. The seedlings (seedling stage treatment) were, however, more susceptible to Cd than Hg. Both root and leaf tissues of the plant treated at the germination stage showed enhanced lipid peroxidation and activities of the antioxidative enzymes (catalase, guaiacol peroxidase and ascorbate peroxidase), except the catalase in leaf in 25 μM Cd treatment. At seedling stage the content of malondialdehyde increased significantly only in the leaf tissue, during 6 h exposure. The activities of all the enzymes exhibited an increasing trend in both the tissue of the seedlings, particularly the leaf, at least after 24 and 48 h, except the catalase whose activity declined in response to Cd. Active involvement of the guaiacol and ascorbate peroxidases, rather than catalase, in scavenging cellular H2O2 was indicated. It was concluded that the two metals had little primary damaging effect on membranes.  相似文献

Oxidative stress was induced in 10-day treated with lanthanum and cerium Hydrilla verticillata plants. Low 10 μM concentrations did not exert harmful effects. The plants treated with higher concentrations showed higher H2O2 content and lower chlorophyll and soluble protein contents as compared to control plants. At the same time, malondialdehyde content rose with increasing concentrations of La and Ce. As La and Ce concentrations increased, superoxide dismutase and catalase activities declined progressively, while peroxidase activity increased. Proline content decreased slightly at 10 μM La or Ce and then rose with higher concentrations. The results indicated that La and Ce caused oxidative damage as evidenced by increased lipid peroxidation and decreased chlorophyll and protein levels. The involvement of NO in O2 ·− generation, rootlet development and antioxidant defence were investigated in the adventitious root cultures of mountain ginseng. Treatments of NO producers (SNP, sodium nitroprusside; SNAP, S-nitroso-N-acetylpenicillamine; and sodium nitrite with ascorbic acid), and NO scavenger (PTIO, 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl3-oxide) revealed that NO is involved in the induction of new rootlets. Severe decline in number of new rootlets compared to the control under PTIO treatment indicates that NO acts downstream of auxin action in the process. NO producers (SNP, SNAP and sodium nitrite with ascorbic acid) activated NADPH oxidase activity, resulting in greater O2 ·− generation and higher number of new rootlets in the adventitious root explants. Moreover, treatment of diphenyliodonium chloride, a NADPH oxidase inhibitor, individually or along with SNP, inhibited root growth, NADPH oxidase activity and O2 ·− anion generation. NO supply also enhanced the activities of antioxidant enzymes that are likely to be responsible for reducing H2O2 levels and lipid peroxidation as well as modulation of ascorbate and non-protein thiol concentrations in the adventitious roots. Our results suggest that NO-induced generation of O2 ·− by activating NADPH oxidase activity is related to adventitious root formation in mountain ginseng.  相似文献
