Responses of lumbar interneurons located in the most lateral regions of Rexed's laminae IV–VII to stimulation of the medial longitudinal bundle and gigantocellular reticular nucleus of medullary pyramids, red nucleus, and peripheral nerves were investigated in cats anesthetized with pentobarbital. Stimulation of the reticulospinal fibers evoked monosynaptic excitation of many interneurons specialized for transmitting activity of the lateral descending systems, but not of peripheral afferents. Convergence of excitatory influences of all three descending systems (cortico-, rubro-, and reticulospinal) was observed on some cells of this group. In addition, monosynaptic "reticular" E PSPs appeared in interneurons transmitting activity of group Ia muscle fibers and in some interneurons of the flexor reflex afferent system. Stimulation of reticulospinal fibers evoked IPSPs in some neurons of this last group. Neurons not exposed to reticulofugal influences (both specialized neurons and interneurons of segmental reflex arcs) were located chiefly in the dorsal zones of the region studied. Microinjections of aspartic acid and chlorpromazine into the region of the locus coeruleus, which strengthen spontaneous unit activity in that structure, in decerebellate cats anesthetized with chloralose, led to depression of the inhibitory influence of flexor reflex afferents on extensor discharges, but did not change the facilitatory action of these afferents on flexor monosynaptic discharges and had no effect on recurrent inhibition of extensor discharges or reduced it. Microinjection of noradrenalin into this region, which depresses spontaneous unit activity in the locus coeruleus, or of procaine, which blocks action potential generation in neurons, led to potentiation of the inhibitory action of flexor reflex afferents on extensor discharges and to strengthening of recurrent inhibition, but did not affect the facilitatory action of these afferents on flexor discharges. The role of tonic descending influences of the locus coeruleus in the control of spinal inhibition evoked by flexor reflex afferents is discussed.A. A. Synaptic responses of different functional groups of interneurons in segments T10 and T11 to stimulation of the ipsilateral and contralateral medullary reticular formation were investigated in anesthetized cats with only the ipsilateral lateral funiculus remaining intact. Activation of reticulospinal fibers of the lateral funiculus with conduction velocities of 30–100 m/sec was shown to induce short-latency and, in particular, monosynptic EPSPs in all types of cells tested: in interneurons excited by group Ia muscle afferents, in cells activated only by high-threshold cutaneous and muscle afferents (afferents of the flexor reflex), in cells activated mainly by descending systems, and, to a lesser degree, in neurons connected with low-threshold cutaneous afferents. These cell populations are located mainly in the central and lateral parts of Rexed's lamina VII. Most neurons in laminae I–V of the dorsal horn, except six cells located in the superficial layers of the dorsal horn, received no reticulofugal influences. The functional organization of connections of the lateral reticulospinal tract with spinal neurons is discussed and compared with the analogous organization of the medial reticulospinal tract, and also of the "lateral" (cortico- and rubrospinal) descending systems.A. In anesthetized cats in conditions of muscular relaxation we have studied the participation of the interneurons of the lumbar section of the spinal cord in the interaction of the FRA systems of the fore and hind limbs. Using microelectrodes we have made extra- and intracellular recordings of the potentials. It has been shown that from the flexor afferents of the fore limbs both facilitating and inhibitory influences are transmitted. The former are expressed in increased frequency of the background impulse activity of the neurons, in the appearance of evoked responses of the "silent" cells and intensification of the test responses for short time intervals with paired heteronymous stimulation. The inhibitory influences prevail over the facilitating and are manifest in depression of the background activity and evoked segmental responses of the neurons. The maximum inhibition of the segmental responses was noted for intervals of 40–140 msec. The duration of inhibition varied from 100–500 msec and more. Depending on the intensity and duration of the inhibitory influences two groups of interneurons have been isolated. The role of the pre- and postsynaptic mechanisms in the transmission of inhibitory influences from the afferents of the fore limbs on the afferents of the hind limbs is discussed.Pavlov Institute of Physiology, Academy of Sciences of the USSR, Leningrad. Experiments were conducted on anesthetized cats with microelectrode recording to study the synaptic responses that develop in the lumbar motoneurons on stimulation of the afferent fibers of groups II and III in the nerves of the ipsilateral and contralateral forelegs. Stimulation of these afferents evoked predominantly inhibitory postsynaptic potentials (IPSP) in the extensor motoneurons and excitatory postsynaptic potentials (EPSP) in the flexor motoneurons. A basically inhibitory change in the rhythmic background activity developed under the influence of descending impulsation. The duration of the total inhibition of "spontaneous" motoneuron activity corresponded to the duration of the inhibitory influences exerted by the forelimb flexor-reflex afferents (FRA) on the interneurons. The interaction of the descending and segmental PSP resulted in inhibition and facilitation of the segmental responses in the motoneurons. The ultimate result of this interaction was determined by the shifts in the membrane potential of the motoneuron and by the effects created in the interneurons.I. P. Research was performed on spinal cats injected with DOPA and decorticate (decerebrated at level A 13) and spinal cats. It was found that formation (activation) of the spinal locomotor generator is accompanied by heightened excitability in the extensor and the reverse trend in the flexor motoneurons, by an increase in the efficacy of recurrent and reciprocal Ia inhibition of -motoneurons, and by a weakening of the influence of Ib afferents and extensor reflex afferents on these same motoneurons. The likely functional role of these changes in tuning of the spinal segmental apparatus in the generation of locomotor rhythm is discussed.A. A. We carried out intracellular recording from neurons of Clarke's column of the spinal cord of a cat. It is demonstrated that relay neurons of the dorsal spino-cerebellar tract — which are activated by proprioceptive fibers from one muscle only — are not subject to corticofugal postsynaptic control, in contrast to neurons on which excitory and inhibitory influences converge from various groups of afferents. The corticofugal effects in such neurons coincide in direction with effects evoked by flexor reflex afferents. Properties are described of neurons that are situated in the vicinity of Clarke's column but are not relay neurons of the DSCT. The hypothesis is expressed that these neurons are identical to the "border cells" — short-axon interneurons on which axonic collaterals of relay neurons terminate.A. A. Studies on immobilized decerebrate (at intracollicular level) cats in which the scratch generator had been set up following bicuculline application to the upper cervical segments of the spinal cord, showed that the state of the segmental apparatus of the lumbosacral section of the spinal cord differs substantially from that seen in the spinal animal. Direct excitability of motoneurons of the "aiming" and "scratching" muscles rises, while recurrent and reciprocal Ia inhibition of motoneurons intensifies and the influence of Ib afferents on motoneurons declines. Afferents of the flexor reflex exert a primarily inhibitory influence on motoneurons of the "aiming" muscles. This influence becomes predominantly excitatory following spinalization, while the inhibitory effects of these afferents on motoneurons of the "scratch" muscles declines. Electrical activity of flexor and extensor alpha-motoneurons of the lumbar segments of cat's spinal cord as recorded intracellularly during electric stimulation of afferents of the contralateral posterior limb. Contralateral postsynaptic potentials (PSP) were shown to be evoked by activation of cutaneous and high-threshold muscle afferents. The high-threshold afferents of various muscle nerves participate to varying degrees in the generation of contralateral PSP. Contralateral inhibitory postsynaptic potentials (IPSP) were recorded in both flexor and extensor motoneurons along with contralateral excitatory postsynaptic potentials (EPSP). There are no fundamental differences in their distribution between flexor and extensor neurons. Inhibitory influences as a rule are predominant in both during the first 20 msec, and EPSP are predominant in the interval between 20 and 100 msec. The balance of excitatory and inhibitory pathway activity was found to be not as stable as that of the homolateral pathways.I. P. Synaptic processes of 119 thoracic spinal interneurons (T10–11) were investigated in anesthetized cats in response to stimulation of the medial and central zones of the gigantocellular nucleus in the medulla and the ventral columns of the spinal cord. Fast (90–130 m/sec) reticulospinal fibers running in the ventral column were found to produce monosynaptic or disynaptic excitation of interneurons of Rexed's layers VII–VIII, which are connected monosynaptically with group I muscle afferents, and interneurons excited both by group I muscle afferents and low-threshold cutaneous afferents. In most neurons of layer IV, connected monosynaptically with low-threshold cutaneous afferents, and in neurons of layers VII–VIII excited by afferents of the flexor reflex no marked postsynaptic processes were observed during stimulation of the reticular formation. Excitatory, inhibitory, and mixed PS Ps during activation of reticulospinal fibers were found in 14 neurons, high-threshold afferents in which evoked predominantly polysynaptic IPSPs. Seventeen neurons activated monosynaptically by reticulospinal fibers and not responding to stimulation of segmental afferents were found in the medial part of the ventral horn (layers VII–VIII).A. Effects of stimulation of flexor reflex afferents were studied in decerebrate immobilized cats. Stimulation of ipsilateral afferents evoked late long-lasting discharges in the nerves to the flexors, whereas stimulation of contralateral afferents led to similar discharges in nerves to both extensors and flexors. Compared with spinal animals, early segmental reflexes in thalamic cats were tonically depressed. Similar tonic inhibition of segmental reflexes took place in spinal animals after injection of dopa. Segmental reflexes were clearly modulated during late or rhythmic discharges. The possible central mechanisms of these changes in the segmental reflexes are discussed on the basis of data in the literature.A. A. Microelectrode discharges of potentials have been realized from segmentary interneurons of the dorsal horn and intermediate nucleus of the spinal cord in cat at the L6–L7 level by electrical stimulation of the sensorimotor region of the brain cortex. It has been established that corticifugal influences on segmentary interneurons of the system of the flexor reflex and on neurons activated by high threshold muscle afferents (groups Ib, II, and III), or high threshold cutaneous afferents are predominantly excitatory. Interneurons activated by muscle afferents of group Ia or by the lowest threshold cutaneous fibers are weakly subjected to pyramidal influences. The mean latencies of excitatory postsynaptic potentials (EPSP's) and discharges evoked under the influence of pyramidal volley, for the neurons under study in the system of afferents of the flexor reflex are equal to 11.8±2.6 and 20.1±1.8 msec, respectively; for interneurons, excited only by high threshold muscle afferents, they are equal to 15.5±3.6 and 16.3±2.2 msec, respectively; and for interneurons, excited by high threshold cutaneous fibers they are equal to 11.8±2.6 and 18.3±1.4 msec, respectively. The effects of segmental reflexes on descending intersegmental reflexes to stimulation of forelimb afferents were studied in anesthetized cats by recording postsynaptic responses from single motoneurons. Interaction between these influences was found to be reciprocal in character for groups of neurons with primary connections with afferents of the superficial and deep branches of the peroneal nerve and afferents of the nerve to the gastrocnemius muscle. Excitatory postsynaptic responses arising in groups of motoneurons of the peroneal nerve to stimulation of forelimb afferents underwent profound and prolonged inhibition during conditioning stimulation of afferents in the deep and superficial peroneal nerves. Activation of segmental afferents during conditioning stimulation of the gastrocnemius nerve was accompanied by inhibition of excitatory intersegmental responses and deinhibition of inhibitory responses in motoneurons of the gastrocnemius muscle. Segmental inhibition of intersegmental descending impulse activity appeared in the interneuron system of the segmental reflex centers connecting the descending propriospinal tracts with the motoneurons of these centers.I. The knee jerk was elicited during regular firing of relatively low-threshold motor units of the biceps femoris muscle (during weak voluntary contraction). Besides the reflex response of the rectus femoris muscle, synchronous discharges of motor units of the biceps femoris muscle and activation of new motor units also were observed. Poststimulus histograms and statistical analysis of interspike intervals of motor units of the biceps femoris muscle revealed well-marked excitatory influences synchronous with the reflex response of the rectus femoris. This result can be explained by the presence of excitatory inputs of Ia afferents on motoneurons of the antagonist muscle. In the knee jerk, excitation of motoneurons of the antagonist was followed by later inhibitory influences which evidently correspond to the "silent period" of motoneurons of the agonist muscle during the elicitation of its tendon reflex.Institute for Problems of Information Transmission, Academy of Sciences of the USSR, Moscow. The dynamics of interaction between segmental, propriospinal, and spino-bulbo-spinal components of reflex responses of lumbar flexor centers was investigated during activation of forelimb and hindlimb afferents in cats. The monosynaptic flexor reflex to activation of hindlimb afferents is facilitated up to 300% when it coincides in time with the reflex discharge evoked by activation of forelimb afferents, and it remains increased up to 120–140% for 40–50 msec during the aftereffect of such activation. Polysynaptic flexor reflexes of segmental, propriospinal, and spino-bulbo-spinal origin have both facilitatory and inhibitory effects on each other. Facilitation is observed only while the interacting responses coincide in time, inhibition when they do not coincide. Three type of inhibitory effects with durations of 7–15, 40–150, and 300–500 msec are observed. The possible neuronal mechanisms of interaction between these reflexes and their role in functional relations between the fore- and hindlimbs are discussed.  相似文献

It was shown that excitation of high- and low-threshold superior laryngeal afferents triggers reflexes of varying complexity in a considerable proportion of non-respiratory neurons during experiments on cats anesthetized by Nembutal involving stimulation-induced expiration reflex. Systemic alterations in background firing activity were noted during this reflex in "respiratory" neurons; reflex reaction setting in as a result of low-threshold laryngeal afferent activation was also recorded in 22.4% of this group. Oligo- and polysynaptic excitatory connections were found between low-threshold laryngeal afferents and inspiratory beta neurons, P-cells, and laryngeal muscle motoneurons as opposed to inhibitory connections with inspiratory gammaneurons. This article discusses involvement of the neurons investigated in mechanisms of inspiratory inhibition, closure of the vocal chords, and adaptive decline in breathing rate occurring during expiration reflex.A. Experiments on anesthetized cats with partial transection of the spinal cord showed that reticulo-spinal fibers in the ventral part of the lateral funiculus participate in the inhibition of polysynaptic reflexes evoked by stimulation of the ipsi- and contralateral reticular formation. The reticulo-fugal wave in the ventrolateral funiculus evoked comparatively short (up to 70 msec) IPSPs in some motoneurons of the internal intercostal nerve investigated and at the same time evoked prolonged (up to 500 msec) inhibition of IPSPs caused by activation of high-threshold segmental afferents. This wave also led to the appearance of IPSPs in 14 of 91 (15.5 %) thoracic spinal interneurons studied. The duration of these IPSPs did not exceed 100 msec; meanwhile, segment excitatory responses of 21 of 43 interneurons remained partly suppressed for 120–500 msec. It is concluded that the inhibitory action of the lateral reticulo-spinal system on segmental reflexes is due to several synaptic mechanisms, some of them unconnected with hyperpolarization of spinal neurons. Experiments on cats showed that electrical stimulation of the locus coeruleus leads to diminution of the inhibitory action of flexor reflex afferents (FRA) on the extensor monosynaptic reflex and to a decrease in amplitude of the IPSP evoked by FRA in extensor motoneurons. Injection of microdoses of aspartic acid and chlorpromazine into the locus coeruleus depresses the inhibitory effects of FRA, whereas injections of procaine and noradrenalin potentiate the inhibitory action of FRA. Data are given on the character of the descending influence of the locus coeruleus on different groups of spinal interneurons. It is concluded that depression of the inhibitory action of FRA is effected at the level of the final inhibitory interneurons of the "FRA system."A. A. Repetitive stimulation of the locus coeruleus (up to 150 µA in strength) was accompanied by marked weakening of the inhibitory action of flexor reflex afferents and of the reciprocal inhibitory action on extensor motoneurons. Meanwhile stimulation of this sort had no significant effect on direct inhibition of flexor and extensor motoneurons, on the facilitatory action of flexor reflex afferents and the reciprocal inhibitory action on flexor motoneurons and also on dorsal root potentials. Intravenously injected pyrogallol had a similar action, but its effect was much weaker after spinalization of the animals or blocking of spinal cord conduction by cold. Enhancement of the monosynaptic reflex, which also was observed after injection of pyrogallol, was characterized by different temporal parameters; the intensity of this effect was unaffected both by spinalization and by cold block. These data, and also the results of experiments with partial divisions of the spinal cord, suggest that the effects of stimulation of the locus coeruleus are the result of activity of a descending coerulo-spinal tract, running in the ventral quadrant of the spinal cord.A. Local stimulation in the zone of motor representation of the cat hind limb in the postcruciate cortex (area 4) modulates afferent activity of flexor spindles of the foot. An initial pause, connected with contraction of extrafusal fibers, is observed in this activity. After the muscle has returned to its original length, a sharp rise of discharge frequency develops followed by a return to its initial level. Similar phases, but less marked, are observed in secondary afferents. Stimulation of contralateral and ipsilateral regions of the medial precruciate cortex (area 6) causes selective, intensive, and prolonged facilitation of discharge of type Ia units followed by an after-effect, without involving extrafusal muscle fibers. Since influences of the premotor supplementary cortex on lumbar gamma motoneurons are relatively independent of influences coupled with activation of the alpha system on muscle afferents from the motor cortex, a specific role of area 6 in the regulation of segmental excitability of the gamma system can be postulated.  相似文献
