A new species of Phacidiopycnis associated with pome fruits is described. The fungus causes fruit rot on apples during storage and is associated with a twig dieback and canker disease of crabapple trees and dead twigs of pear trees. To characterize the biology of the fungus and compare it with Ph. piri, the type species of the genus, effects of nine media and light on mycelial growth and pycnidial production, mycelial growth in response to temperature and mode of conidial germination in response to nutrient were determined. Apple-juice agar, pear-juice agar, prune-juice agar, potato-dextrose agar (PDA) and malt-extract agar, Czapek-Dox agar and oatmeal agar (OMA) favored mycelial growth. Cornmeal agar (CMA) did not favor mycelial growth. Light effect on pycnidial formation was medium dependent. Abundant pycnidia with mature conidia formed in 14 d old PDA and OMA cultures at 20 C, regardless of light, whereas none or very few pycnidia formed on other media in the dark. Fluorescent light stimulated formation of pycnidia except on CMA. The fungus grew at -3-25 C, with optimum growth at 15-20 C. Conidia germinated either by forming germ tubes or less often by budding. Budding of conidia occurred in 1 and 10% pear-juice solutions but not in 100% pear-juice solution. Six isolates of Ph. washingtonensis from different species of pome fruits had identical ITS sequences. The sizes of the ITS region were the same for both Ph. washingtonensis and Ph. piri, and four polymorphic nucleotide sites were found in the ITS region between Ph. washingtonensis and Ph. piri. The similarity in ITS sequences between these two taxa is confirmatory evidence for the erection of the new species of Phacidiopycnis associated with pome fruits we describe here.  相似文献

A new disease of pods and seeds of mequite caused by Macrophomina phaseolina is described from Northeastern Brazil. Affected pods showed a brown discolouration which often included pycnidia of the fungus. Infected seeds did not germmate.  相似文献

A new hypocrellin A-producing strain, Shiraia sp. SUPER-H168, was isolated from tissues of bamboo, Brachystachyum densiflorum. The morphology of this strain was characterized with a light microscope and a scanning electronic microscope. The mycelia, conidia, pycnidia of fungus were observed. Small pycnidia (10-20 microm in length) full of small conidia appeared on the mycelia, which were first reported in this study. The 18S rDNA region of this strain was amplified and sequenced. Then a neighbor-joining tree of 18S rDNA was constructed. According to the result of analysis, the strain SUPER-H168 was proved to belong to the genus Shiraia. Hypocrellin A was produced by solid-state fermentation, extracted by acetone, isolated by preparative RP-HPLC, and identified by RP-HPLC, ESI-MS and ultraviolet-visible absorbing scanning with diode array detection. The HA production could reach 2.02 mg/g dry solid substrate.  相似文献

A fungal strain, FOM-8108, that was isolated from sea sand was found to produce gentisylquinone and chlorogentisylquinone, inhibitors of neutral sphingomyelinase. The fungus grew well under normal conditions, in the darkness, or on various agar media, but typical morphological characteristics were not observed to determine its taxonomy. Therefore, culture conditions were studied extensively, resulting in formation of a number of pycnidia by the fungus on the media containing seawater or on natural substrates such as hydrangea leaves, gardenia leaves, and rice straw under natural light or near-ultraviolet radiation exposure. Eventually, from the morphological characteristics of pycnidia, conidiogenous cells, and conidia, strain FOM-8108 was considered to belong to the genus Phoma. Culture studies showed seawater in the fermentation medium was necessary for the strain to produce gentisylquinones. Particularly, a full production of chlorogentisylquinone, which has a chloride ion in the structure, was performed in the medium with higher concentration (75%–100%) of seawater. Sphaeropsis pyriputrescens, the causal agent of Sphaeropsis rot of pears and apples, is a recently described species. In this study the effects of culture media, temperature, water potential, pH and light on mycelial growth and pycnidial production of S. pyriputrescens were evaluated. Apple juice agar and pear juice agar were most suitable for mycelial growth of all six isolates tested. Cornmeal agar was not suitable for either mycelial growth or pycnidial production. The fungus grew from -3 to 25 C, with optimum growth at 20 C and no growth at 30 C. The fungus grew at water potential as low as -5.6 MPa on potassium chloride-amended potato-dextrose agar (PDA). Hyphal extension was not observed at -7.3 MPa after 10 d incubation, but growth resumed when the inoculum plugs were placed on PDA. The fungus grew at pH 3.3-6.3 and optimum growth was at pH 3.3-4.2. No mycelial growth was observed at pH above 7.2 after 10 d incubation, but growth resumed when the inoculum plugs were transferred onto PDA. Regardless of medium tested, few pycnidia formed at 20 C in the dark. Pycnidial production was enhanced significantly by fluorescent light, but continuous light appeared to reduce pycnidial production, depending on the medium. Oatmeal agar (OMA) was most suitable for production of pycnidia and conidia. Pycnidia that formed on 3 wk old OMA cultures at 20 C under 12 h light/12 h dark produced abundant conidia, and the technique is recommended for inoculum production.  相似文献

Reproduction by three isolates ofPhoma medicaginis growing on potato dextrose agar was studied. The formation of pycnidia was optimum at 30°C whereas the optimum for the formation of conidia was 20°C. Light consistently increased the numbers of pycnidia and conidia over those formed in darkness and more conidia were produced in light at 23°C than at 30°C. The effects of carbon and nitrogen sources on reproduction were studied using modified Richard's medium as the basal medium. Fourteen carbohydrates, 11 amino acids, 9 inorganic nitrogen sources and urea were evaluated by replacing the carbohydrate or nitrogen in Richard's medium with the test substance. Generally, the monosaccharides and disaccharides were about alike and superior to polysaccharides for the production of pycnidia. The carbon sources were about equally useful for production of conidia, but the polysaccharides were superior to the other two classes of carbon sources when the number of conidia/pycnidium was calculated. Generally, the formation of pycnidia and conidia was favored by nitrate more than by ammonium nitrogen sources. The average number of conidia/pycnidium was greatest, however, when the nitrogen source was NH4NO3. All amino acids tested appeared to be useful nitrogen sources for production of pycnidia but none were especially good for conidia production. L-isoleucine was superior to the other amino acids tested. Of three isolates used, Illinois 23 consistently produced more pycnidia and conidia that did isolates Minnesota 2 and Missouri 5. Usually the significant interactions between isolates and other treatments were due to a greater response by isolate Ill. 23. The histo- and cytopathological effects in resistant (ILC-195) and susceptible (Canitez-87) chickpea cultivars were examined by light, transmission and scanning electron microscopy 3, 5 and 7 days after inoculation (d.a.i) of seedlings with Ascochyta rabiei. The fungus produced typical appressoria that penetrated both cuticle and stomata. The resistant plants had physical barriers and a cuticle layer against fungal penetration 3 d.a.i. The fungus spread intercellularly and subepidermally in the leaves and stems of susceptible plants 3 d.a.i., and was followed 5 d.a.i. by cell plasmolysis, degeneration of organelles and of cellulose, but not lignified, walls. Pycnidia formation occurred between 5 and 7 d.a.i. 7 d.a.i., organelle degeneration, pycnidia formation and symptom severity increased. Tracheidal elements, including lignified elements, were almost intact in both resistant and susceptible cultivars. In the susceptible plants, lignin cell walls were slightly degraded after 7 days. There was less cell degeneration and pycnidia formation in resistant plants. Some electron-dense large bodies and lipid granules were observed within intracellular fungal hyphae in infected cells of resistant plants 7 d.a.i.  相似文献

本文中的多孔菌系指广义非褶菌目中具有孔状子实层体的种类,按照现代分类系统包括担子菌门中多孔菌目、锈革孔菌目、褐褶菌目、糙孢孔目、革菌目、糙孢革菌目和红菇目中孔状子实体的种类,还有伞菌目、阿太菌目、鸡油菌目和木耳目中个别属,如网孔菌属、胶孔菌属、榆孔菌属、牛排菌属和纵隔孔菌属等的种类。基于作者采集的1万余号标本和国内主要标本馆标本的研究,对中国多孔菌的种类进行了系统总结,目前有604种多孔菌发现于中国,多孔菌数量位居世界第一。对中国多孔菌的名称按新近研究成果和最新命名法规(维也纳法规)进行了订正。对121种新拟了汉语学名。  相似文献

Process of infection and histological changes with Ascochyta blight of chickpea caused by A. rabiei (Pass.) Labr. were studied by light microscopy. Germ tubes from conidia of the fungus penetrate the stem tissue at the juncture of two epidermal cells and form subepidermal aggregates until the fourth day. On the sixth day, yellowing and necrotisation of host tissue coincides with formation of mature pycnidia. Fungus causes extensive damage to cellulosic cell walls of parenchymatous cortical and pith tissues in advance of invading hyphae indicating involvement of cell wall degrading enzymes. Lignified tissues, particularly xylem tracheary elements, remain intact.  相似文献

Myrmaeciella caraganae was recollected in and around Vienna, Austria and found to be morphologically different generically from the type species of Myrmaeciella, M. endoleuca. We redescribe M. caraganae in the new genus Stromatonectria. Phylogenetic analyses of LSU sequences place the genus in the Bionectriaceae, Hypocreales. S. caraganae occurs on branches of Caragana spp., Colutea arborescens and Laburnum anagyroides of the Fabaceae. It is characterized by spheroid perithecia partly or entirely immersed in a Hypocrea-like stroma, a Nectria-like centrum and bicellular hyaline ascospores. Conidia of S. caraganae are produced in compound pycnidia that are formed prior to or in association with perithecia. Sporodochia but no pycnidia are formed in culture. We discuss the genus Myrmaeciella and compare S. caraganae with species of the Nectriaceae, including Nectria balansae, N. eustromatica and N. paraguayensis.  相似文献

Blackleg disease, caused by Leptosphaeria maculans, is one of the most important diseases of rapeseed Brassica napus in Iran as in other regions of the world. The samples including canola petals and seeds were collected during 2014–2015 from canola field in North Iran. Isolates characteristics of fungus were assessed based on the colony growth rate and pycnidia in Potato Dextrose Agar. The pycnidia of the fungus were black, globose to subglobose in shape, the single-celled conidia, hyaline and fusiform with diameters of 4–5 × 1.5–2 μm. Most of the isolates were produced pigment in the liquid culture in variable color brown to black. Thirteen isolates were then separated into pathogenicity groups based on the interactions on B. napus differential cultivars. For the direct detection of seed contamination with L. maculans, PCR was developed using specific primers pair (LmacF, LmacR) which can amplify ITS1 and ITS2 along with the 5.8S rRNA region of L. maculans genome. Based on the followed information and sequence analysis, the fungal isolates from these samples were identified as L. maculans. The findings of this research showed that the disease was aggressive and highly distributed from infested seeds to oilseed rape fields.  相似文献

Light and nutrition are the important factors in the production of pycnidia and conidia by cowpea isolates of the Phomopsis state of Diaporthe phaseolorum. The highest number of pycnidia and conidia were produced on plant tissue exposed to cool-white fluorescent light. In semisynthetic media more pycnidia were formed at high glucose concentrations, but they matured more slowly than those formed at lower glucose concentrations. Both the level of conidiation and the percentage of pycnidia that formed conidia were higher at lower glucose concentrations. The best artificial medium for inducing a high number of pycnidia containing abundant conidia was one that contained 0.4% glucose and 0.4% NaNO3. A number of carbon sources could replace glucose in this medium.  相似文献

Tomato roots with brown root rot showed three types of lesion: cortical rot of fine roots, 'corky root', and basal stem rot. The fungi most commonly isocated from diseased roots were: Colletotrichum atramentarium, Chaetomium spp., Cephalosporium spp., Volutella ciliata , and a grey mycelial fungus sometimes producing pycnidia ( Pyrenochaeta sp.). Other fungi isocated less frequently were: Myrothecium roridum, Petriella asymmetrica, Trichoderma airide, Phytophthora spp., Alternaria sp. and Fusarium spp. There were differences between the numbers of each species isocated from the three types of lesion in steamed and in unsteamed soils, and some seasonal variation.
Inoculation experiments with seedlings in vitro showed that Ckaetomium cochliodes and Petriella asymmetrica could infect radicles. On older plants growing in soil Colletotrichum atramentarium was the only effective pathogen. Culture filtrates from C. atramentarium, Chaetomium cochliodes , and P. asymmetrica decreased root growth when tested in vitro. Leachates from loam and a loam-manure mixture decreased the growth of tomato root-tip cultures; the effect of these leachates was altered by autoclaving.  相似文献

Light induces the formation of pycnidia inPhomopsis mali. The induction caused by light can be conserved in darkness. The size and quantity of pycnidia vary with the illuminance. Under low and high illuminances physiologically different pycnidia appear.Phomopsis mali cultivated in darkness in the presence of other microorganisms (fungi or bacteria) can fructify and produces pycnidia similar to those formed under low illuminance and in natural conditions. Our experiments indicate the presence of a pycnogenesis-inducing substance that can exists in different forms and induces the formation of the different pycnidia.  相似文献

Abstract Histological studies were performed on a compatible and an incompatible interaction between chickpea ( Cicer arietinum L.) plants and the fungus Ascochyta rabiei (Pass.) Labr. The time course of infection, development on leaflets and stems of susceptible (ILC 1929) and resistant (ILC 3279) plants was monitored by light or scanning electron microscopy with the aim to compare histological changes as the basis for further work on biochemical changes in this plant-pathogen interaction.
Spores of A. rabiei began to germinate from 12 hpi on and developed a polar germ tube; fungal colonization, secretion of a mucilaginous exudate and appressoria formation (1–3 dpi) were identical on both cultivars. Leaves of susceptible plants were invaded by the fungus directly through the cuticle, the fungus then spread subepidermally followed by a rapid collapse of the leaf tissue (4–6 dpi). Development of leaf spots and fungal pycnidia could be observed 6–8 dpi. The resistant cultivarrapidly responded (24–48 hpi) to fungal infection and cells of the palisade parenchyma exhibited autofluorescence. In later stages of the infection (4–5 dpi) fluorescent areas developed to small necrotic spots all over the leaflet. These necrotic areas, were the result of cell death and a subsequent change in the leaf structure and were characterized by the accumulation of phenolic compounds. Leaves of the resistant cultivar were invaded by the fungus to less than 5%.  相似文献

The loculoascomycete Moristroma quercinum sp. nov. and Moristroma japonicum sp. nov. are described from Northern Europe (Denmark, Lithuania, Sweden) and Japan, respectively. M. quercinum is reported from wood of Quercus robur and Q. petraea, and M. japonicum is reported from wood of Quercus mongolica var. grossoserrata. Ascostromata of both species were found on hard heartwood of attached or shed branches. The two new species differ from the type species of the genus, M. polysporum, by the presence of pycnidia, and by the size of ascostromata, asci and ascospores. Drawings illustrate ascostromata, pycnidia, asci, hamathecium and ascospores of the two new species. A phylogenetic analysis suggests that Moristroma belongs to the Chaetothyriomycetes, rather than to the Dothideomycetes as previously suggested.  相似文献
