Histochemical studies of the microcirculatory system were reviewed with regard to the organ specificity of the structural organization and fine distribution of the lymphatic capillary network. The lymphatics and blood vessels are characterized by an enzyme-histochemical method using 5'-nucleotidase (5'-Nase), alkaline phosphatase (ALPase) and/or diaminopeptidase (DAPase) staining in addition to an immunohistochemical method. The 5'-Nase-positive lymphatic vessels can be distinguished histochemically from arterial and venous vessels based on ALPase and DAPase activity, respectively. The specificity and localization of the enzyme reactions were confirmed by comparative histochemical studies of the same specimen with light microscopy and scanning or transmission electron microscopy. These histochemical methods are discussed in relation to their ability to demonstrate the organ specificity of vascular networks under normal and pathological conditions.  相似文献

Synopsis The cholinergic and adrenergic innervation of human minor sublingual buccal and labial salivary glands has been studied with histochemical techniques for localizing acetylcholinesterase and catecholamines. A rich cholinergic innervation was observed around the acini, blood vessels and some ducts of the three glands.The adrenrgic innervation, however, was virtually absent from the parenchyma although present around the blood vessels, in marked contrast to the dense parenchymal adrenergic innervation observed in the human parotid and submandibular glands. These results suggest that the autonomic nervous mechanism which regulates salivary secretion is more elaborate in the major than in the minor salivary glands.  相似文献

The adrenergic innervation of the lymph vascular wall was studied by means of the Falck fluorescence histochemical tecnique and electron microscopy with Tranzer and Richards' histochemical tecnique. The lymph vessels wall, compared with that of blood vessels, shows very few adrenergic nerve fibers located in the adventitia outside the smooth muscle cells. The possible role of the nervous system in the motor control of the lymph vessels is discussed.  相似文献

Almond glycopeptidase is an enzyme which cleaves specifically beta-aspartylglucosylamine linkages in glycoproteins with asialo-carbohydrate moieties. With this enzyme, it was possible to demonstrate the localization of asparagine-linked oligosaccharides in glycoproteins of human placenta and umbilical cord tissues. In these tissues, the oligosaccharides were shown to react positively for a series of histochemical procedures for neutral complex carbohydrates such as periodic acid-Schiff (PAS), peroxidase-labelled Ricinus communis agglutinin-I-diaminobenzidine (PO-RCA-DAB) and concanavalin A-peroxidase-diaminobenzidine (Con A-PO-DAB). The asparagine-linked carbohydrates were localized in the placental villi, blood vessels and perivascular tissues and the umbilical cord blood vessels and matrix. The results of previous biochemical analyses performed upon the same tissues (Takahashi et al., 1981) have corroborated the results of the histochemical studies. The present results appear to substantiate the usefulness of almond glycopeptidase for the histochemical demonstration of the particular oligosaccharides of glycoproteins in tissues in general.  相似文献

We studied histochemical and ultrastructural characteristics of the gastric submucosal blood vessels in portal hypertensive (PHT) rats. PHT was induced by two-stage ligation of the portal vein. Control rats were sham operated (SO). On the fifth day after surgery portal vein blood pressure was measured and rats were killed under nembutal anesthaesia. Gastric specimens were obtained for histological, histochemical and ultrastructural assessment. PHT rats showed thickening and increased cellularity of submucosal vessels including increase in number and size of endothelial cells. All these mesenchymal cells were vimentin-positive. Thickening of vascular wall in submucosal vessels was also observed ultrastructurally together with prominent elaboration of luminal surface of endothelial cells. These changes were not observed in SO rats. All changes in PHT rats reflect vasculopathic involvement of gastric wall in portal hypertension.  相似文献

In our experiments we observed the relationship of the blood vessels to the small, intensely fluorescent cells (SIF cells) in the lower mesenteric ganglion of the cat. We injected the solution of Evan's blue into the ganglia and processed them with the fluorescent histochemical method by Falck and Hillarp. We observed that the SIF cells are placed in the ganglia closely to the blood vessels or closely round them. When observing lager groups of SIF cells placed at the edge of the ganglia a dense network of the blood vessels was observed among these cells.  相似文献

Synopsis The nature of the cholinesterase activity present on the wall of blood vessels in amphibian brain has been studied histochemically. It is concluded that the enzyme involved is acetylcholinesterase rather than butyrylcholinesterase, which more frequently occurs in the blood vessels of the brain of other vertebrates. The histochemical results are in agreement with most biochemical data concerning substrate specificity and inhibitor response for both acetylcholinesterase and butyrylcholinesterase. The two main hypotheses put forward by others in order to explain cholinesterase activity in brain vessels are discussed briefly.  相似文献

Brain capillaries have structural and functional characteristics that constitute a regulatory interface, or “barrier,” between the blood and the brain. We have investigated the role of the neural tissue environment in the differentiation of the endothelial barrier, by transplanting embryonic brain fragments to the coelomic cavity, where they were vascularized by nonneural vessels, and fragments of embryonic mesoderm to the brain, where they were vascularized by neural vessels. A major problem in this approach is that when embryonic tissues are transplanted to an ectopic site, their own blood vessels survive and form a part of the new vascular system. This has made the results of previous experiments difficult to interpret. We overcame this problem by transplanting fragments of tissue that had not yet been vascularized from very young quail embryos to host chick embryos. These grafts did not contain vascular channels that could form part of a new vascular system. Furthermore, the distinctive quail nuclear morphology allowed us to demonstrate that the grafted tissue was, in fact, vascularized by the host vessels. Abdominal vessels vascularizing grafted neural tissue formed structural, functional, and histochemical features of the blood-brain barrier. In contrast, brain vessels vascularizing grafted mesodermal tissue were devoid of barrier characteristics. These results indicate that endothelial blood-brain barrier characteristics develop in response to some aspect of the neural environment.  相似文献

Acetylcholinesterase activity was detected in the carotid labyrinth of amphibians by both biochemical and histochemical methods. The histochemical tests showed enzyme activity in surfaces of afferent and efferent nerve terminals in contact with type I cells, but none within the type I cells. Acetylcholinesterase activity occurred on some, though not all, nerve fibers in the extra-cellular spaces. These fibers might be parasympathetic cholinergic fibers innervating blood vessels.  相似文献

The histochemical activity of 4 lysosomal enzymes, acid phosphatase (AP), nonspecific esterase (NE), aryl sulphatase (AS), and beta-glucuronidase (BG), was compared in following structures of the human placenta: syncytiotrophoblast, villous stromal cells, fetal capillaries and larger blood vessels, cells of basal plate, macrophages, and Hofbauer cells. In spite of a general similarity in distribution of the investigated enzymes, differences concerning particular structures were found. Thus positively stained granules in endothelia of capillary vessels were revealed only in the reaction for BG, although contours of capillaries were also outlined by the diffuse reaction product for AS. The muscular layer of larger vessels reacted strongly for AS and weakly for NE with the remaining reactions being negative. In syncytiotrophoblast, BG appeared much less active than the other 3 enzymes. The possible significance of the BG positive granules in endothelial of capillaries and of the occasional divergence in distribution of the classical lysosomal markers (AP and BG) is discussed.  相似文献

With histological and histochemical methods the innervation of the mammary gland of the cow was studies. The skin of the udder has a weak innervation in contrast to the test. In the district of the basement of the gland body nerve stems enter in the teat. In the skin of the teat only isolated free nerve terminals are found. Intraepithelial nerve terminals are missed. In the middle layer of the teat round the blood vessels bundles of nerve fibers and many organs of nerve terminals were shown. The organs of nerve terminals are typical of the teat and present mechanical receptors. It is discussed the functional significance of the afferent and efferent innervation of the mammary gland of the cow.  相似文献

Morphologic and histochemical characteristics of selected portions of normal arteries from two species known to differ in susceptibility to vascular disease were examined. Arteries were classified as predominantly elastic, muscular or complex. Species differences in the structural organization of the abdominal aortic segment were observed. Arterial mucopoly-saccharides were stained more intensely in the tunica intima and media of chicken vessels than within those of the rat, and tended to be most concentrated in proximity of the internal elastic membrane. Histochemical procedures for the demonstration of enzymatic activity revealed inter-and intraspecies variations in vascular metabolism. Pronounced differences in reaction intensity for hydroxybutyrate dehydrogenase and malic enzyme, affecting chicken and rat coronary arteries, were noted. In contrast, theses vessels displayed only minimal activity for acid phosphatase. Marked endothelial deposition of alkaline phosphatase reaction products in the arteries of the chicken was demonstrated, while this enzyme's activity in the vessels of the rat was restricted to the tunica adventitia. The implications of these structural and histochemical factors with regard to vascular susceptibility to disease were discussed.  相似文献

Summary The distribution and density of the noradrenergic and acetylcholinesterase-positive nerve fibres were histochemically studied in different uterine regions of prepubertal and cycling rats in dioestrus and oestrus. Besides the rich and double innervation of blood vessels, both types of nerve fibre were found in the myometrium and cervical musculature. The non-vascular noradrenergic network looked denser at the tubal end of the horns and at the cervix, whereas the acetylcholinesterase-positive innervation was poor at the tubal end, increasing toward the cervix. Contrasting with the middle third of the uterine horn, at the tubal end, the myometrial longitudinal layer was much more innervated than the circular one, especially by the noradrenergic nerve fibres. The prepubertal rats presented an adult pattern of uterine autonomic innervation. In the cycling animals, this innervation was nearly the same during oestrus and dioestrus regarding both the density of nerve fibres and intensity of the histochemical reactions.  相似文献

Enzymatic activity of cells, antigenic cellular markers and extracellular matrix of the hyperplastic intima of the aorta and carotid arteries was investigated in non-specific aorto-arteritis by immunomorphological and histochemical techniques. The cells of subendothelial layer of thickened arterial intima contained smooth muscle cell myosin, gave positive reactions to myosin ATP-ase and revealed high activity of thiamine pyrophosphatase. Fibronectin and type IV and V collagen were located in close proximity to these cells. The data obtained make it possible to consider these cells as modified smooth muscle cells. Type III collagen was the prevalent type of extracellular matrix of the thickened intima. A great number of blood vessels of the capillary and precapillary types have been found to penetrate into the intima from the adventitia. A possible role of pericytes surrounding newly formed capillaries as the precursors of subendothelial cell population in the hyperplastic intima is discussed.  相似文献

The reactions of capacitance blood vessels of the abdominal cavity and pelvis were studied in vagotomized dogs during cytotoxic heart injury. The same reactions were investigated in recipient dogs during cross circulation with cardiac damage in the donors. 本文首次用5′-Nase-ALP双重组织化学方法观察了20例胃癌组织和癌旁胃壁组织内淋巴管的细微分布。在光镜下毛细淋巴管和淋巴管5′-Nase染色强阳性,管壁显示明显的棕色或深棕色,而毛细血管和血管的ALP反应显示强阳性,管壁呈明显的蓝色。据此可将淋巴管、血管区别开来、本研究发现胃癌组织内有较多的淋巴管、毛细淋巴管以及较多的棕色实性条状组织,这些条状物可能是新生的毛细淋巴管。上述结果为研究癌组织内淋巴管的分布及胃癌淋巴道转移,提供了可靠的实验方法。  相似文献

Visualising vascular endothelial cell function in individual blood microvessels allows elucidation of molecular interactions at the vascular wall, the first barrier between blood-borne therapeutic agent and its target. Functional analysis in situ requires sub-micrometer spatial resolution and tagged molecules generating contrast in living blood vessels. Light microscopy fulfills these requirements, particularly if fluorescent tags deliver the contrast. However, vascular arborisations in living organs defy morpho-functional analysis, filling tissues with closely meshed three-dimensional networks which are inaccessible to optical imaging. We protocol here successful morpho-functional analysis of microvascular processing in a living organ, the human placental cotyledon. Fluorescence-tagged tracer was positionally fixed by snap-freezing, frozen sections were cut, freeze-dried and heat-fixed. A brief histochemical procedure then labelled all vascular elements in the sections, providing fluorescence contrast in two colour channels. Mosaic monochromatic images acquired in both channels delivered high-resolution maps of centimeter-wide tissue areas. Quantitative analysis of the images’ greyscale histograms defined objectifiable, reproducible thresholds, used to reduce the images to colour-coded wide-area functional maps tracking placental vascular processing of the tagged molecules. Rapid positional fixing of tracer with reduction of images to maps was combined with ultrastructural tracking to elucidate vascular processing at scales of nanometres and seconds.  相似文献

No diabetic angiolopathy was found in the retinas, kidneys and skeletal muscles of protodiabetic and overtly diabetic sand rats. The terminal blood vessels were investigated using histological, enzyme histochemical, immunofluorescence microscopic, autoradiographic and electron microscopic methods. There seems to be little or no connection in sand rats between the diabetic metabolism syndrome and the metabolic process leading to angiolopathy.  相似文献

NADPH-diaphorase (NADPH-D) activity and immunoreactivity for neural and endothelial nitric oxide synthase (nNOS and eNOS, respectively) were used to investigate nitric oxide (NO) regulation of penile vasculature. Both the histochemical and immunohistochemical techniques for NOS showed that all smooth muscles regions of the penis (dorsal penile artery and vein, deep penile vessels, and cavernosal muscles) were richly innervated. The endothelium of penile arteries, deep dorsal penile vein, and select veins in the crura and shaft were also stained for NADPH-D and eNOS. However, the endothelium of cavernous sinuses was unstained by both techniques. Fewer fibers were seen in the glans penis, those present being associated with small blood vessels and large nerve bundles near the trabecular walls. All penile neurons in the pelvic plexus, located by retrograde transport of a dye placed in the corpora cavernosa penis, were stained by the NADPH-D method. Essentially similar results were obtained with an antibody to nNOS. These data suggest that penile parasympathetic neurons comprise a uniform population, as all seem capable of forming nitric oxide. However, in contrast to the endothelium of penile vessels, the endothelium lining the cavernosal spaces may not be capable of nitric oxide synthesis.  相似文献

In the experiment performed on cats, using histological, histochemical and impregnative techniques it has been stated that choose of the small intestine area, suitable for transplantation should be based on counting straight vessels ramified in it. In order to ensure viability of the small intestine fragment the method of its preliminary treatment is recommended, based on training blood vessels. For estimating viability of the transplant it is possible to ascertain SDG activity in neurons of the intramural nervous ganglia and estimate amount of secreting gobletlike cells in mucous membrane of the small intestine loop.  相似文献
