Three seedlots of crisp lettuce cv. Saladin were ‘primed’ in 1% tri-potassium orthophosphate (K3PO4) or in water for 6 to 12 h and then dried at room temperature. Two of the seedlots were separated into fractions of different mean weight before ‘priming’. The effects of ‘priming’ and grading on crop uniformity, germination at 35°C and root length in a slant test were evaluated. In general, ‘priming’ increased root length in a slant test but there were no consistent beneficial effects of ‘priming’ on seedling size, head weight or uniformity of head weight. Larger seed produced longer roots in a slant test than small seed and gave larger seedlings but seed size had no effect on head weight or cv. of head weight. ‘Priming’ had no effect on germination at 20°C but significantly increased germination at 35°C. ‘Priming’ in K3PO4 was more effective than in water. It is suggested that the response of seeds to ‘priming’ varies with seedlot and individual seed weight.  相似文献

Using 24 commercial seedlots of the lettuce cvs Ithaca, Pennlake and Saladin, the percentage germination at a range of temperatures from 20 to 35°C, and the forces required to penetrate the pericarp, the endosperm and the embryo were measured. Percentage germination declined with increasing temperature in all seedlots but there were particularly large differences between seedlots of the same cultivar at 30.0 and 32.5°C. The force required to penetrate the pericarp was on average more than twice that required to penetrate the endosperm. It is concluded that in commercial cultivars of crisp lettuce the strength of the pericarp plays a much greater role than that of the endosperm in determining germination. In all three cultivars there were significant positive correlations between seed weight and the force required to penetrate the whole seed. However only in cv. Pennlake were there significant correlations between germination at high temperature and seed penetration forces.  相似文献

The value of the International Seed Testing Association (ISTA) germination test in estimating levels of variation in embryo length between carrot seed lots was examined. For the two groups of seed lots used, the coefficient of variation (c.v.) of embryo length ranged from 16 to 50%. The c.v. of embryo length was negatively correlated with the numbers of normal seedlings counted after 7 days of germination at 20°C, rs= - 0·72 D.F. 12 and - 0·82, D.F. 15 for the two groups of seed lots used. Provided the relationship between these two characters is established for each laboratory, it is likely to be useful in identifying seed lots of potentially low c.v. of embryo length.  相似文献

The emergence from brassica seedlots designated by seed merchants as being of high vigour was measured in sterilised soil and in sterilised soil re-inoculated with Pythium sylvaticum. The mean emergence level of 93·9% in the former compared favourably with the known mean total viability of the seedlots (97·7%). However, P. sylvaticum reduced emergence of 10 out of 16 untreated seedlots. The majority of seedlots gave > 75% emergence but in three cases emergence was reduced by 32·5–40·0%. The effects were not related directly to time taken to emerge, nor were the seedlots most affected those with high levels of infection. A compound seed treatment containing metalaxyl, fenpropimorph and gamma-HCH restored high emergence levels.  相似文献

Predictors of dominance in the male golden hamster (Mesocricetus auratus)   总被引:3,自引:0,他引:3  
The outcome of social interactions between four male hamsters was significantly related to body weight (rs=0·66) and to the size and pigmentation of their lateral flank glands (rs=0·82). Weight was held constant in a second experiment and variations in gland size and pigmentation remained significantly related to the outcome of social encounters (rs=0·77). In a third experiment, castrate hamsters of uniform weight receiving either no hormone, 0·1 mg, 0·50 mg, or 1·00 mg testosterone propionate exhibited flank gland variations significantly related to social rank (rs=0·81). These experiments demonstrate that the state of the flank gland, which is related to endogenous androgen levels, can be used as a predictor of social rank in male hamsters.  相似文献

Crisp lettuce plants cv. Saladin were grown from the time they started flowering, at 20/10°C (16 h day, 8 h night), 25/15°C and 30/20°C in glasshouses on two occasions in 1985. Yields of seed increased from, on average, 15 g to 27 g and then fell to 20 g per plant with progressive increases in temperature. The number of mature florets per plant increased with temperature but the number of seeds per mature floret was lower at 20/10°C and 30/20°C than at 25/15°C. An increase in temperature reduced mean seed weight by up to 45%, seed volume by 15%, cell numerical volume density (Nv) by 27% and the number of cells per seed by 39%. Percentage seed germination reached a maximum early in seed development at the stage when the pappus appeared through the involucral bracts. Differences in percentage germination and vigour of seeds (slope test) from different temperatures were accounted for largely by the effects on mean seed weight. However, when germinated at 30°C seeds produced at 30/20°C germinated more readily than those produced at 25/15°C or 20/10°C. Seed vigour gradually increased with an increase in the length of storage after harvest, reaching a maximum after 260 days. In general, seeds produced at 25/15°C exhibited a greater variation in numbers of seeds per floret, Nv, seed weight, times of seedling emergence, seedling and mature head weight than seeds produced at lower or higher temperatures.  相似文献

Adult female golden hamsters exhibit smaller and less pigmented flank glands than do males. Nevertheless, as has been found in male hamsters, variations in these parameters of the flank glands correlate highly (rs=0·78) with social rank attained in a group of four females. Ovariectomy and subsequent replacement with a graded series of testosterone propionate doses produces a directly related response in the flank gland and in the social rank attained in all-female groups (rs=0·72). Body weight of females also correlates directly with social status (rs=0·74), but, when body weight is held constant, social rank can still be predicted from measures of the flank gland. Oestrous-related fluctuations in aggressive behaviour of females did not alter dominance relationships.  相似文献

The diffusive conductance (Cs) of rice (Oryza sativa cvs Jaya and Bala) leaves was measured during a soil drying cycle from flooding to decreasing soil water potential (φs) in a controlled-environment chamber. Plants were grown continuously under 5 cm submergence up to 69 days after transplanting and thereafter were subjected to gradual soil drying for a period of 17 days in the vegetative growth stage. In both the cultivars, the values of Cs were generally more on adaxial than abaxial leaf surfaces. This response of stomata during the period of soil drying was independent of leaf rolling. Further, the slopes of the curves (Cs, vs φs) also did not differ significantly (P= 0·05). The total Cs, of both cultivars during flooding was almost equal (0·60 cm s-1) but at the end of the soil drying cycle, the values of total Cs, were 0·11 cm s-l at ψs of -1·3 MPa and 0·08 cm s-1 at ψs, of -0·8 MPa in cvs Jaya and Bala, respectively. For total Cs, slopes differed significantly (P = 0·05). A close relationship between total Cs, and ψs, in both cultivars (Cs, = 0·58-0·40 ψs, for cv. Jaya and Cs= 0·46-0·56 ψs, for cv. Bala) indicated that stomata were sensitive to increasing soil water deficit.  相似文献

QTL controlling root and shoot traits of maize seedlings under cold stress   总被引:3,自引:0,他引:3  
The improvement of early vigour is crucial for the adaptation of maize (Zea mays L.) to the climatic conditions of central Europe and the northern Mediterranean, where early sowing is an important strategy for avoiding the effect of summer drought. The objectives of this study were to identify quantitative trait loci (QTL) controlling cold-related traits and to investigate the relationships among them. A set of 168 F2:4 families of the Lo964 × Lo1016 cross was grown in a sand–vermiculite substrate at 15/13°C (day/night) until the one-leaf stage. Twenty QTL were identified for the four shoot and two seed traits examined. Analysis of root weight and digital measurements of the length and diameter of primary and seminal roots led to the identification of 40 QTL. The operating efficiency of photosystem II (PSII) was related to seedling dry weight at both the phenotypic and genetic level (r=0.46, two matching loci, respectively) but was not related to root traits. Cluster analysis and QTL association revealed that the different root traits were largely independently inherited and that root lengths and diameters were mostly negatively correlated. The major QTL for root traits detected in an earlier study in hydroponics were confirmed in this study. The length of the primary lateral roots was negatively associated with the germination index (r=–0.38, two matching loci). Therefore, we found a large number of independently inherited loci suitable for the improvement of early seedling growth through better seed vigour and/or a higher rate of photosynthesis.This paper is dedicated to our friend and colleague Alberto Soldati, who passed away unexpectedly.  相似文献

Young plants of brussels sprout, cv. Cambridge Special, growing in pots in a glasshouse, were sprayed on ten occasions with gibberellic acid (GA) at 0, 25, 100 and 400 p.p.m. and ammonium nitrate (NH4NO3) at 0, 0·0125, 0·025 and 0·05 M concentrations in all combinations. In 24 days both GA and NH4NO3 increased leaf area, leaf number, dry weights of leaf, stem and root, and fresh weights of leaf and stem. GA increased stem height and decreased fresh weight/unit leaf area (leaf thickness), whereas NH4NO3 did not affect stem height and increased leaf thickness. Of the GA treatments, 100 p.p.m. gave the largest plants as judged by fresh weight of the whole plant and leaf area, and of the N treatments 0·05 M NH4NO3 increased growth most. The best treatment combination was 0·05 M NH4NO3 with 100 p.p.m. GA, which gave the greatest fresh and dry weights of the whole plant, leaf area and leaf dry weight as well as increasing leaf thickness. Significant interactions were found between GA and N for dry weight of leaf, fresh and dry weight of root, and leaf thickness but not for leaf area or stem growth (fresh weight, dry weight, height). The combination of levels of GA and N that can be expected to increase leaf area, leaf dry weight and leaf thickness simultaneously, may lie within narrow limits for a particular crop. In a second experiment plants were sprayed with all combinations of GA (0 and 100 p.p.m.) seven times under glass in pots and ten times in the field, and three levels of KH2PO4 on twelve occasions in the field. Sprouts (axillary buds) were harvested in October and February. At the first harvest GA did not affect fresh weight or number, but increased both the total number of sprouts picked (with GA = 80·7; without GA = 69·8 per plant) and the total fresh weight of saleable sprouts (with GA = 2·59; without GA = 2·33 lb/plant). KH2PO4 also increased the weight and number of sprouts at the final harvest and the number of small sprouts at the first. There were interactions between GA and KH2PO4 (P= < 0·001) for both tota weight and number of saleable sprouts.  相似文献

Xylem probe measurements in the roots of intact plants of wheat and barley revealed that the xylem pressure decreased rapidly when the roots were subjected to osmotic stress (NaCl or sucrose). The magnitude of the xylem pressure response and, in turn, that of the radial reflection coefficients (σr) depended on the transpiration rate. Under very low transpiration conditions (darkness and high relative humidity), σr assumed values of the order of about 0·2–0·4. The σr values of excised roots were also found to be rather low, in agreement with data obtained using the root pressure probe of Steudle. For transpiring plants (light intensities at least 10 μmol m?2 s?1; relative humidity 20–40%) the response was nearly 1:1, corresponding to radial reflection coefficients of σr= 1. Further increase of the light intensity to about 400 μmol m?2 s?1 resulted in a slight but significant decrease of the σr values to about 0·8. Similar measurements on maize roots confirmed our previous results (Zhu et al. 1995, Plant, Cell and Environment 18, 906–912) that, in intact transpiring plants at low light intensities of about 10 μmol m?2 s?1 and at relative humidities of 20–40% as well as in excised roots, the xylem pressure response was much less than expected from the external osmotic pressure (σr values 0·3–0·5). In contrast to wheat and barley, very high light intensities (about 700 μmol m?2 s?1) were needed to shift the radial reflection coefficients of maize roots to values of about 0·9. Osmotically induced xylem pressure changes were apparently linked to changes in turgor pressure in the root cortical parenchyma cells, as shown by simultaneous measurements of xylem and cell turgor pressure. In analogy to the σr values of the respective glycophytes, the σc values of the root cortical cells of wheat and barley were close to unity, whereas σc for maize was significantly smaller (about 0·7) under laboratory conditions. When the light intensity was increased up to about 700 μmol m?2 s?1 the cellular reflection coefficient of maize roots increased to about 0·95. In contrast to the σr values, the σc values of the three species investigated remained almost unchanged when the leaves were exposed to darkness and humidified air or when the roots were cut. The transpiration-dependent (species-specific) pattern of the cellular and radial reflection coefficients of the root compartment of the three glycophytes apparently resulted from (flow-dependent) concentration-polarization and sweep-away effects in the roots of intact plants. The data could be explained straightforwardly terms of theoretical considerations outlined previously by Dainty (1985, Acta Horticulturae 171, 21–31). The far-reaching consequences of this finding for root pressure probe measurements on excised roots, for the occurrence of pressure gradients under transpiring conditions, and for the non-linear flow-force relationships in roots found by other investigators are discussed.  相似文献

A series of experiments were carried out to study the effects of different drying regimes on the survival of newly-germinated (radicle emerged 0·5-1·5 mm through the seed coat) cabbage seeds and to establish a treatment for reducing the moisture content of these germinated seeds. A preliminary surface-drying step proved necessary to avoid seed agglutination during the main drying stage. Of the drying regimes compared both viability and vigour were most effectively maintained in air, conditioned to between 20 and 30°C and with 80% r.h., flowing through the seeds at 0·25 m s-1. Under these conditions, the viability and vigour of newly-germinated seed was maintained during drying to an equilibrium moisture content of c. 14%. At this moisture content further radicle growth was prevented and seeds remained free-flowing and separate for sowing or for further treatment.  相似文献

This study describes the size at first sexual maturity, length–weight relationships (LWR) in relation to size at first sexual maturity, and Fulton’s condition factor (KF) of Eutropiichthys vacha in the Ganges River, northwestern Bangladesh. Sampling was done using traditional fishing gear including cast nets, square lift nets and conical traps during January and April, and July to December 2010. For each individual, total length (TL) was measured to the nearest 0.01 cm, and total weight (BW) was determined to the nearest 0.01 g. The gonadosomatic index (GSI) was calculated by the equation, GSI (%) = (Gonad weight in g/BW) × 100. The size at first sexual maturity of males and females was estimated by the relationship between gonadosomatic index and total length. A total of 583 specimens (289 males; 294 females) ranging from 8.30 to 27.00 cm TL and 3.16 to 159.50 g BW were analyzed. Sizes at first sexual maturity for male and female E. vacha were 13.15 and 14.00 cm TL, respectively. The analysis of covariance (ancova ) revealed significant differences in slope and intercept between early and late phases for males (F = 4.532, P < 0.001) and females (F = 21.984, P < 0.001). The KF was not significantly correlated with TL for males (rs = 0.052; P = 0.378), but was highly correlated for females (rs = ?0.165; P = 0.005). This study establishes a strong base for monitoring changes in length at first sexual maturity attributable to high fishing pressures or other reasons within the Ganges and associated river ecosystems.  相似文献

Agricultural biotechnology is very familiar with the properties of nanomaterial and their potential uses. Therefore, the present experiment was conducted to test the beneficial effects of nanosilicon dioxide (nSiO2: size- 12 nm) on the seed germination of tomato (Lycopersicum esculentum Mill. cv Super Strain B). Application of nSiO2 significantly enhanced the characteristics of seed germination. Among the treatments, 8 g L−1 of nSiO2 improved percent seed germination, mean germination time, seed germination index, seed vigour index, seedling fresh weight and dry weight. Therefore, it is very clear that nSiO2 has a significant impact on the seed germination potential. These findings could provide that alternative source for fertilizer that may improve sustainable agriculture.  相似文献

The objective of this study was to determine the effect of freezing on the function in Atlantic salmon Salmo salar spermatozoa. The semen was frozen in Cortland's medium + 1.3M dimethyl sulphoxide + 0.3M glucose + 2% bovine serum albumin (final concentration) in a ratio of 1:3 (semen:cryoprotectant) as the treatment (T) and fresh semen as the control (F). Straws of 0·5 ml of sperm suspension were frozen in 4 cm of N2L. They were thawed in a thermoregulated bath (40° C). After thawing, the percentage of spermatozoa with fragmented DNA [transferase dUTP (deoxyuridine triphosphate) nick‐end labelling (TUNEL)], plasma membrane integrity (SYBR‐14/PI) and mitochondrial membrane potential (ΔΨMMit, JC‐1) were evaluated by flow cytometry and motility was evaluated by optical microscope under stroboscopic light. The fertilization rates of the control and treatment semen were tested at a sperm density of 1·5 × 107 spermatozoa oocyte?1, by observation of the first cleavages after 16 h incubation at 10° C. In the cryopreserved semen (T), the mean ± s.d . DNA fragmentation was 4·8 ± 2·5%; plasma membrane integrity 75·2 ± 6·3%; mitochondrial membrane potential 51·7 ± 3·6%; motility 58·5 ± 5·3%; curved line velocity (VCL) 61·2 ± 17·4 µm s?1; average‐path velocity (VAP) 50·1 ± 17·3 µm s?1; straight‐line velocity (VSL) 59·1 ± 18·4 µm s?1; fertilization rate 81·6 ± 1·9%. There were significant differences in the plasma membrane integrity, mitochondrial membrane potential, motility, fertilization rate, VCL, VAP and VSL compared with the controls (P < 0·05). Also the mitochondrial membrane potential correlated with motility, fertilization rate, VCL and VSL (r = 0·75; r = 0·59; r = 0·77 and r = 0·79, respectively; P < 0·05); and the fertilization rate correlated with VCL and VSL (r = 0·59 and r = 0·55, respectively).  相似文献

Mass‐specific oxygen consumption rate, i.e. standard metabolic rate (Rs) and critical oxygen tension (Pcrit) of red drum Sciaenops ocellatus were measured and scaled over a 2500‐fold range in mass (MF; 0·26–686 g). Rs conformed to well established models (Rs = 3·73·91 MF?0·21; r2 = 0·86) while Pcrit increased over the size range (Pcrit = 3·15 log10MF + 16·19; r2 = 0·44). This relationship may be ecologically advantageous as it would allow smaller S. ocellatus to better utilize hypoxic zones as habitat and refuge from predators.  相似文献

Chimaera carophila (n = 45) and Hydrolagus homonycteris (n = 11), two deep‐sea chimaerids rarely caught in the waters off New Zealand, were collected from research trawl catches and commercial fishery catches around New Zealand at depths between 400 and 1300 m, between 2014 and 2016. Additional preserved specimens of both species (n = 58) from museum collections were analysed for size, sex and maturity. External assessment of male claspers and a combination of internal assessments of female gonad mass and oviducal gland width, were used to determine maturity. For both species, length at first maturity was 0·70–0·82 of their maximum observed chimaera length (LC), with females maturing at a larger size. Length at maturity for C. carophila (LC range: 28·7–103·9 cm) was estimated at 72·5 cm LC for males (n = 163) and 82·5 LC for females (n = 58). In H. homonycteris, length at maturity (length range: 78·6–99·8 cm LC) was estimated at 79·1 cm LC for males (n = 51) and 80·1 cm LC for females (n = 17). Ovarian fecundity was up to 31 for C. carophila and sperm storage was confirmed in the oviducal gland of this species. Both species preyed on benthic invertebrates. Some C. carophila and H. homonycteris inhabit depths beyond most current fisheries, but both species appear to be relatively rare and have reproductive parameters characteristic of low productivity, which may make these species vulnerable to population decline if mortality was to increase in the future.  相似文献

Aims: To clarify the cellular properties of Listeria monocytogenes involved in adhesion to and biofilm formation on polyvinyl chloride, a widely used material in the food manufacturing process. Methods and Results: A significant correlation between the ability of initial adherence to and biofilm formation on PVC was observed for 24 L. monocytogenes strains (Spearman rank‐correlation coefficient, rs = 0·89). The swimming motility assay revealed no relationship between initial adherence and motility of L. monocytogenes. The microbial adhesion to solvent assay revealed an interaction of L. monocytogenes cells with nonpolar solvents, and a significant correlation was also observed between the degree of interaction with nonpolar solvents and initial adherence to PVC (rs = 0·87 and rs = 0·84, between initial adherence and affinities to decane and hexadecane, respectively). Conclusions: Results indicate that cellular hydrophobicity of L. monocytogenes is an important property involved in the initial adherence to and biofilm formation on PVC. Significance and Impact of Study: This study clarified the factors involved in the adherence to and biofilm formation ability of L. monocytogenes strains with PVC.  相似文献

Synechococcus R-2 (PCC 7942) actively accumulated Cl? in the light and dark, under control conditions (BG-11 media: pHo, 7·5; [Na+]o, 18 mol m?3; [Cl?]o, 0·508 molm?3). In BG-11 medium [Cl?], was 17·2±0·848 mol m?3 (light), electrochemical potential of Cl? (ΔμCl?i,o) =+211±2mV; [Cl?]i= 1·24±0·11 mol m?3(dark), ΔμCl?i,o=+133±4mV. Cl? fluxes, but not permeabilities, were much higher in the light: ?Cl?i,o= 4·01±5·4 nmol m?2 s?1, PCl?i,o= 47±5pm s?1 (light); ?Cl?i,o= 0·395±0·071 nmol m?2 s?1, PCl?i,o= 69±14 pm s?1 (dark). Chloride fluxes are inhibited by acid pHo (pHo 5; ?Cl?i,o= 0·14±0·04 nmol m?2 s?1); optimal at pHo 7·5 and not strongly inhibited by alkaline pHo (pHo 10; ?Cl?1i,o= 1·7±0·14 nmol m?2 s?1). A Cl?in/2H+in coporter could not account for the accumulation of Cl? alkaline pHo. Permeability of Cl? is very low, below 100pm s?1 under all conditions used, and appears to be maximal at pHo 7·5 (50–70 pm s?1) and minimal in acid pHo (20pm s?1). DCCD (dicyclohexyl-carbodiimide) inhibited ?Cl?i,o in the light about 75% and [Cl?]i fell to 2·2±0·26 (4) mol m?3. Valinomycin had no effect but monensin severely inhibited Cl? uptake ([Cl?]i= 1·02±0·32 mol m?3; ?Cl?i,o= 0·20±0·1 nmol m?2 s?1). Vanadate (200 mmol m?3) accelerated the Cl? flux (?Cl?i,o= 5·28±0·64 nmol m?2 s?1) but slightly decreased accumulation of Cl? ([Cl?], = 13·9±1·3 mol m?3) in BG-11 medium but had no significant effect in Na+-free media. DCMU (dichlorophenyldimethylurea) did not reduce [Cl?], or ?Cl?i,o to that found in the dark ([Cl?]i= 8·41±0·76 mol m?3; ?Cl?i,o= 2·06±0·36 nmol m?2 s?1). Synechococcus also actively accumulated Cl? in Na+-free media, [Cl?]i was lower but ΔΨi,o hyperpolarized in Na+-free media and so the ΔμCl?i,o was little changed ([Cl?]i= 7·98±0·698 mol m?3; ΔμCl?i,o=+203±3 mV). Net Cl? uptake was stimulated by Na+; Li+ acted as a partial analogue for Na+. Synechococcus has a Na+ activated Cl? transporter which is probably a primary 2Cl?/ATP pump. The Cl? pump is voltage sensitive. ΔμCl?i,o is directly proportional to ΔΨi,o(P»0·01%): ΔμCl?i,o= -1·487 (±0·102) ×ΔΨi,o, r= -0·983, n= 31. The ΔμCl?i,o increased (more positive) as the Δμi,o became more negative. The ΔμCl?i,o has no known function, but might provide a driving force for the uptake of micronutrients.  相似文献

The aim of this study was to investigate post-anthesis N redistribution from the roots to other organs in the wheat plant. Two cultivars of spring wheat (Triticum aestivum L.) Sport and WL were grown to maturity in culture solution with stepwise-decreasing N supply and 15N before sampling. The N concentration of dry weight in the roots decreased from anthesis to full maturity. At maturity the root N concentration was 5.5 mg g−1 dry weight for Sport and 4.9 mg g−1 dry weight for WL, while the main shoot N concentration was 2.1 mg g−1 dry weight for Sport and 2.2 mg g−1 dry weight for WL. The roots were a major sink for N taken up post-anthesis and contained the main proportion of the 15N during the whole post-anthesis period. The N redistributed from the roots contributed 24.3% of the total N amount in the grains at maturity in cv Sport and 8.7% in cv WL. For cv Sport, 46.3% of the root N amount was redistributed to other parts of the plant and for WL 25.4%. The tiller grains were supplied with N from the 15N applied post-anthesis to a higher degree than the main shoot grains. The transport capability was maintained until 12 days after complete yellowness. We concluded that wheat has a great ability to redistribute nitrogen from the root system to the grains.  相似文献
