Effect of potassium nitrate (KNO3) on the dynamics of neurological disorders and lethality in rats as sequelae of brain ischemia induced by a single-step bilateral common carotid artery occlusion were investigated in Wistar rats. KNO3 at a dose of 5 mg/1000 g administered 60 min prior to the occlusion of the two carotid arteries reliably reduced the severity of neurological disorders and lethality in rats.  相似文献

Summary Studies under growth cabinet conditions investigated the effect of source and concentration of nitrogen and timing of nitrogen application on the growth and nitrogen fixation byLotus pedunculatus cv. Maku andTrifolium repens cv. S184. KNO3, NaNO3 and NH4NO3 were added at transplanting at the following rates: 3.33, 7.78 and 13.33 mg N/plant. KNO3 was added at 3.33 and 7.78 mg N/plant at 0, 6, 12, 18, 24 or 30 days after transplanting.Lotus shoot weight increased with all increasing nitrogen sources but clover only responded to KNO3 and NaNO3. The root weight of both species increased with increasing KNO3 and NH4NO3. The percentage increase in lotus and clover shoot growth was greater than that of root growth when KNO3 was added within a week of transplanting. Increases in growth by both species resulted from added nitrogen except with lotus when NaNO3 was applied where increased nitrogen fixation also contributed to increased growth.Weight and number of effective nodules on both species were increased with 3.33 mg N per plant as KNO3 but nitrogen fixation was not affected. Addition of 13.33 mg N as NaNO3 reduced weight and number of effective nodules in both species and also nitrogen fixation by lotus.KNO3 increased growth and nodulation of both species when applied within one week after transplanting. Nodulated lotus plants responded to KNO3 by increasing growth but not nodulation.KNO3 appeared to affect infection and development of nodules on lotus and may affect the growth of existing nodules on clover.  相似文献

Background

Rumex species are traditionally used for the treatment of neurological disorders including headache, migraine, depression, paralysis etc. Several species have been scientifically validated for antioxidant and anticholinestrase potentials. This study aims to investigate Rumex hastatus D. Don crude methanolic extract, subsequent fractions, saponins and flavonoids for acetylcholinestrase, butyrylcholinestrase inhibition and diverse antioxidant activities to validate its folkloric uses in neurological disorders. Rumex hastatus crude methanolic extract (Rh. Cr), subsequent fractions; n-hexane (Rh. Hex), chloroform (Rh. Chf), ethyl acetate (Rh. EtAc), aqueous fraction (Rh. Aq), crude saponins (Rh. Sp) and flavonoids (Rh. Fl) were investigated against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) at various concentrations (125, 250, 500, 1000 μg/mL) using Ellman’s spectrophotometric analysis. Antioxidant potentials of Rh. Sp and Rh. Fl were evaluated using DPPH, H2O2 and ABTS free radical scavenging assays at 62.5, 125, 250, 500, 1000 μg/mL.

Results

All the test samples showed concentration dependent cholinesterase inhibition and radicals scavenging activity. The AChE inhibition potential of Rh. Sp and Rh. Fl were most prominent i.e., 81.67 ± 0.88 and 91.62 ± 1.67 at highest concentration with IC50 135 and 20 μg/mL respectively. All the subsequent fractions exhibited moderate to high AChE inhibition i.e., Rh. Cr, Rh. Hex, Rh. Chf, Rh. EtAc and Rh. Aq showed IC50 218, 1420, 75, 115 and 1210 μg/mL respectively. Similarly, against BChE various plant extracts i.e., Rh. Sp, Rh. Fl, Rh. Cr, Rh. Hex, Rh. Chf, Rh. EtAc and Rh. Aq resulted IC50 165, 175, 265, 890, 92, 115 and 220 μg/mL respectively. In DPPH free radical scavenging assay, Rh. Sp and Rh. Fl showed comparable results with the positive control i.e., 63.34 ± 0.98 and 76.93 ± 1.13% scavenging at 1 mg/mL concentration (IC50 312 and 104 μg/mL) respectively. The percent ABTS radical scavenging potential exhibited by Rh. Sp and Rh. Fl (1000 μg/mL) were 82.58 ± 0.52 and 88.25 ± 0.67 with IC50 18 and 9 μg/mL respectively. Similarly in H2O2 scavenging assay, the Rh. Sp and Rh. Fl exhibited IC50 175 and 275 μg/mL respectively.

Conclusion

The strong anticholinesterase and antioxidant activities of Rh. Sp, Rh. Fl and various fractions of R. hastatus support the purported ethnomedicinal uses and recommend R. hastatus as a possible remedy for the treatment of AD and neurodegenerative disorders.  相似文献

To probe the effects of renewal regime on the production of polysaccharides, Porphyridium cruentum was cultured semi-continuously in flat plate photobioreactor. Uniform design was used to optimize renewal conditions. Quadratic mathematic models related to productivity, total recovery yield of biomass and polysaccharides were set up to clarify the influence of individual factors and their interactions. According to the mathematic models, the optimal semi-continuous condition for total yield of polysaccharide was NaNO3 3.5 g/L, renewal rate 27%, renewal period 2.91 days. The optimal condition for polysaccharide output rate was NaNO3 0.5 g/L, renewal rate 5%, renewal period 7 days. With the optimal renewal regime, the maximal total recovery yields of polysaccharide achieved at 29.4 g, which was 1.57 times higher than that of batch cultivation. The maximum output rate of polysaccharide was 68.64 mg/L per day, which was 2.02 times higher than previous reported data.  相似文献

We investigated the effect of sodium nitrate on the progress of stress-induced damages in rats of the Krushinskii-Molodkina strain which are genetically prone to audiogenic seizures. NaNO3, at doses of 5 and 50 mg per 100 g of body weight, decreased the severity of movement impairments and the mean sizes of subdural and subarachnoid hemorrhages induced by acoustical stress. Sodium nitrate, at a dose of 0.5 mg per 100 g of body weight, did not significantly influence these changes.  相似文献

Microbial fermentation of citrinin-free Monascus pigments is in favor in the development of food industry. This study investigated the influences of carbon source, nitrogen source, and mineral salts on the cell growth, monascin (MS), and citrinin (CT) production in Monascus M9. A culture medium composition was established for maximizing the production of citrinin-free MS in submerged culture, as follows: 50?g/L Japonica rice powder, 20?g/L NH4NO3, 3?g/L NaNO3, 1.5?g/L KH2PO4, 1?g/L MgSO4?·?7H2O, 0.2?g/L MnSO4. Under these conditions, no CT was detectable by high performance liquid chromatography. The yield of MS reached 14.11?mg/g, improving approximately 30% compared with before optimization.  相似文献

A significant enhancement of spore production by solid-state culture of Beauveria bassiana was achieved via a statistical modeling and optimization approach. A two-level Plackett–Burman design was implemented to screen the significant medium components for spore production. Among the six variables tested, sucrose, NaNO3, and H3BO3 were identified as the most significant factors (confidence levels above 95%) for spore production. The steepest ascent experiments were adopted to determine the optimal region of the medium composition. The developed second-order polynomial model predicted that the maximal spore production was 6.24×109/g with the following optimal culture conditions: sucrose 34.7 g/L, NaNO3 52.8 g/L, and H3BO3 79.6 µg/L. Finally the verification experiments were carried out and the maximal spore production reached 6.221×109/g at the above optimal conditions. The spore production in the verification experiments was very close to that predicted by the model, indicating that the constructed second-order model had superior modeling and optimization ability and can be applied in the optimal spore production of Beauveria bassiana YP9 as a biological control agent.  相似文献

Effects of sodium nitrite on the blood flow rate (V) in microvessels of rat cerebral cortex (5–15 µm in diameter), arterial pressure (AP), plasma osmotic pressure (P osm), and hematological parameters were studied. The mean V and AP in 15 min after administration of NaNO2 (3 mg/100 g mass) decrease by 36% and 45%, while in 30 and 45 min they increase by 14% and 7% and by 20% and 19% as compared with minimal values, respectively. At the end of the experiment, on the background of an elevation of P osm (3%), a decrease of total hemoglobin concentration and of the number of erythrocytes was accompanied by a decrease of the discocyte number from 81 to 35% and a reduction of cell diameters by 7%. It is concluded that NaNO2 produces block of local active mechanisms of the cerebral blood flow regulation. Compensatory responses in this case are provided by passive mechanisms (rheological blood properties) and systemic reactions.  相似文献

Summary The steady-state concentration of M. trichosporium OB3b increased about two-fold in the continuous culture when the feeding medium was supplemented by ferrous sulphate (50mg/L) and citric acid (100mg/L) at a steady state. In batch and continuous cultures, the cell growth was significantly inhibited by excess N-sources (NH4OH, NH4Cl, NH4NO3, HNO3, and NaNO3) and ammonium N-sources were more inhibitory. Both volumetric O2 transfer coefficient and specific O2 uptake rate increased monotonously in an extensive range of air flow rate (0.1–7 vvm) and the methane interfered with the O2 transfer even at very low flow rates (0.01–0.1 vvm).  相似文献

Aspergillus parasiticus NRRL-2999 was inoculated into meat mixtures with curing salts and into yeast extractsucrose (YES) and sucrose-ammonium salts (SAS) broth with and without curing salts to determine if the presence of curing salts significantly affected growth and aflatoxin production by the mold. The effect of individual curing salts or curing salt mixtures on growth and toxin elaboration by the aspergillus was substrate dependent. When YES broth contained 100 ppm of NaNO2, 2% NaCl, or 1 or 2% NaCl plus 200 ppm of NaNO2 or 200 ppm of NaNO3, growth and/or aflatoxin production was depressed. Biosynthesis of aflatoxin B1 was enhanced by presence of 1 and 4% NaCl in YES broth. The SAS broth containing only NaCl or NaCl combined with nitrite or nitrate yielded less aflatoxin than did control broth or no aflatoxin at all. When compared to the control, an increase in growth and amount of aflatoxin occurred in SAS broth which contained 200 ppm of NaNO3. Sausages containing 100 and 200 ppm NaNO2 and no NaCl supported more mold growth and aflatoxin production than did control sausage with 3 % NaCl and 100 ppm of NaNO2. Addition of 2 and 3 % NaCl and no nitrite to sausage resulted in less aflatoxin than in control sausage.  相似文献

Nannochloropsis sp. was grown with different levels of nitrate, phosphate, salinity and temperature with CO2 at 2,800 μl l−1. Increased levels of NaNO3 and KH2PO4 raised protein and polyunsaturated fatty acids (PUFAs) contents but decreased carbohydrate, total lipid and total fatty acids (TFA) contents. Nannochloropsis sp. grew well at salinities from 22 to 49 g l−1, and lowering salinity enhanced TFA and PUFAs contents. TFA contents increased with the increasing temperature but PUFAs contents decreased. The highest eicosapentaenoic acid (EPA, 20:5ω3) content based on the dry mass was above 3% under low N (150 μM NaNO3) or high N (3000 μM NaNO3) condition. Excessive nitrate, low salinity and temperature are thus favorable factors for improving EPA yields in Nannochloropsis sp.  相似文献

Cell growth and lipid production of a marine microalga Nannochloropsis oceanica DUT01 were investigated, and fresh medium replacement with different ratios to promote long term cell growth and lipid accumulation was also tested. The highest lipid content reached 64% in nitrogen deplete f/2 medium containing 37.5 mg/L NaNO3 combined with 1/5 fresh medium replacement, however, the highest lipid titer (0.6 g/L) and lipid productivity (31 mg/L/d) were achieved using BG11 medium containing 1.5 g/L NaNO3, taking advantage of 1/5 fresh medium replacement as well, which corresponded to the maximum biomass production of 1.4 g/L, highlighting the importance of high biomass accumulation for efficient lipid production. When biomass compositions were monitored throughout the culture, decreased protein content was found to be coupled with increased lipid production, whereas relatively stable carbohydrate content was observed. The fatty acids in the lipid of N. oceanica DUT01 comprise over 65% saturated fatty acids and monounsaturated acids (i.e. palmitic acid (C16:0) and oleic acid (C18:1)), suggesting that N. oceanica DUT01 is a promising candidate for biodiesel production. Interestingly, very high content of hexadecadienoic acid (C16:2, about 26–33%) was produced by DUT01, which distinguished this microalga with other microalgae strains reported so far.  相似文献

Synechocystis sp. PCC 6701 has a brilliantly colored pigment, phycobiliprotein containing phycoerythrin. Culture medium was optimized by sequential designs in order to maximize phycobiliprotein production. The observed fresh weights after 6 days were 0.58 g/L in BG-11, 0.83 g/L in medium for Scenedesmus sp. and 0.03∼0.52 g/L in the other tested media. Medium for Scenedesmus sp. was selected to be optimized by fractional factorial design and central composite design since the medium maintained a more stable pH within a desirable range due to higher contents of phosphate. The fractional factorial design had seven factors with two levels: KNO3, NaNO3, NaH2PO4, Na2HPO4, Ca(NO3)2, FeEDTA, and MgSO4. From the result of fractional factorial design, nitrate and phosphate were identified as significant factors. A central composite design was then applied with four variables at five levels each: nitrate, phosphate, pH, and light intensity. Parameters such as fresh weight and phycobiliprotein contents were used to determine the optimum value of the four variables. The proposed optimum media contains 0.88 g/L of nitrate, 0.32 g/L of phosphate under 25 μE·m−2·s−1 of light intensity. The maximum phycobiliprotein contents have been increased over 400%, from 4.9 to 25.9 mg/L after optimization.  相似文献

【目的】以标志链带藻(Desmodesmus insignis)为实验材料,研究不同氮源及其浓度对该藻生长、总脂和淀粉(碳水化合物)含量的影响,为该藻在生物能源方面的应用提供一定的理论依据。【方法】以硝酸钠、碳酸氢铵或尿素为氮源,5个氮浓度(3、6、9、12和18 mmol/L)的BG-11培养基培养标志链带藻,采用干重法测定生物质浓度、重量法测定总脂、苯酚-硫酸法测定、总碳水化合物和淀粉的含量。【结果】标志链带藻在3种氮源下均能很好的生长。最高油脂含量出现在3 mmol/L硝酸钠实验组,达到32.61%(d.w)。当18 mmol/L碳酸氢铵作为氮源时,总碳水化合物与淀粉的含量以及产率都达到最高,分别为56.54%(d.w)和55.33%(d.w)、0.24和0.23 g/(L·d)。以尿素为氮源时,其生物质浓度和各组分含量与其它氮源实验组差别不大,均有利于该藻的生长及各生化组分含量的积累。【结论】以该藻种生产生物能源的成本等综合考虑,以18 mmol/L碳酸氢铵和尿素为氮源培养标志链带藻最优。  相似文献

The production of cellulase by Bacillus subtilis MU S1, a strain isolated from Eravikulam National Park, was optimized using one-factor-at-a-time (OFAT) and statistical methods. Physical parameters like incubation temperature and agitation speed were optimized using OFAT and found to be 40?°C and 150?rpm, respectively, whereas, medium was optimized by statistical tools. Plackett-Burman design (PBD) was employed to screen the significant variables that highly influence cellulase production. The design showed carboxymethyl cellulose (CMC), yeast extract, NaCl, pH, MgSO4 and NaNO3 as the most significant components that affect cellulase production. Among these CMC, yeast extract, NaCl and pH showed positive effect whereas MgSO4 and NaNO3 were found to be significant at their lower levels. The optimum levels of the components that positively affect enzyme production were determined using response surface methodology (RSM) based on central composite design (CCD). Three factors namely CMC, yeast extract and NaCl were studied at five levels whilst pH of the medium was kept constant at 7. The optimal levels of the components were CMC (13.46?g/l), yeast extract (8.38?g/l) and NaCl (6.31?g/l) at pH 7. The maximum cellulase activity in optimized medium was 566.66?U/ml which was close to the predicted activity of 541.05?U/ml. Optimization of physical parameters and medium components showed an overall 3.2-fold increase in activity compared to unoptimized condition (179.06?U/ml).  相似文献

Summary Gibberellic acid (GA3) completely replaced the chilling requirement of intact, dormant seeds of Ruellia humilis. At 30° 100 and 1000 mg/l completely replaced the chilling requirement, while in seeds germinated at 25° 100 mg/l GA3 was ineffective but 1000 mg/l entirely replaced the after-ripening requirement at low temperature.This work was supported by an NIH Biomedical Sciences Support Grant to the University of Kentucky.  相似文献

ABSTRACT

Acute toxicity of sodium nitrite (NaNO2) was assessed in chickens (Gallus gallus domesticus) and domestic mallard ducks (Anas platyrhynchos domestica) by oral gavage and in free-feeding trials with chickens, domestic mallard ducks, pigeons (Columba livia f. domestica), budgerigars (Melopsittacus undulates) and wētā (Family: Rhaphidophoridae). Free-feeding trials involved the presentation of toxic paste and pellet baits containing encapsulated NaNO2 developed for the control of common brushtail possums (Trichosurus vulpecula) and feral pigs (Sus scrofa). The oral gavage LD50 value for NaNO2 in solution was approximately 68.50?mg/kg (95% CI 55.00–80.00?mg/kg) for both chickens and ducks. In feeding trials, six out of 12 chickens consumed toxic paste bait and four of these birds consumed a lethal dose. When chickens consumed toxic paste bait, the LD50 value was approximately 254.6?mg/kg (95% CI 249.1–260.2?mg/kg). Of the other three species of birds presented with toxic baits only one duck consumed a lethal dose of paste bait. There was no evidence of wētā feeding on toxic baits.  相似文献

The intracellular cadmium (Cd) content was measured with early stationary phase cells of a highly Cd-tolerant moderately halophilic bacterium Pseudomonas sp. No. 40 cultivated in 1M and 3M NaCl medium containing 0 to 2500 μg of CdCl2/ml. It was found that the Cd contents were greatly affected by the NaCl concentration of the medium. When the bacterium was cultivated in the 1, 2, 3, and 4M NaCl medium containing 1500 μg of CdCl2/ml, the intracellular Cd content was 25.0, 4.1, 3.1, and 2.0 mg Cd per g of dry cells, respectively. The intracellular Cd content decreased with increases of NaCl concentration of the medium. The fact seems to reflect Cd-tolerance of the bacterium towards the growth in the medium of different NaCl concentration. It is worthwhile to note that the bacterium showed the highest Cd-tolerance (in 3M NaCl) and the lowest Cd content among the bacteria so far known. The bacterial cells grown in the 1M NaNO3 and 1M Na2SO4 medium accumulated 1.8–1.3 times as much Cd2+ as those in the 1M NaCl medium in the presence of 50–200 μg of CdCl2/ml. It would also explain the difference in the Cd toxicity in the medium of NaNO3, Na2SO4, or NaCl.  相似文献

Background and aimSodium nitrite (NaNO2) is an inorganic salt with numerous applications in a variety of industries, as well as in medicine. Nevertheless, exposure to high levels of NaNO2 is toxic for animals and humans. Sodium nitrite intoxication is shown to decrease the activity of major antioxidant defence enzymes which is dependent on the maintenance of specific ion equilibrium. The aim of the present study was to investigate the effect of acute NaNO2 intoxication on the content of the essential metals iron (Fe), calcium (Ca) and zinc (Zn) in mouse spleen.MethodsMature male ICR mice were divided into four groups and subjected to acute NaNO2 exposure by a single intraperitoneal injection of 120 mg/kg body weight. Animals in each group were sacrificed at certain time interval after treatment (1 h, 5 h, 1 day and 2 days). Spleens were excised and processed for atomic absorption spectrometry analysis of Fe, Ca and Zn content.ResultsAt the first hour after treatment, a decrease in Fe and Ca levels was observed. One day following NaNO2 administration, Zn concentration reached its lowest value and Ca levels remained lower, compared to the untreated controls. In contrast, Fe concentration increased on the first and second day after treatment.ConclusionThe results of the present study demonstrate that acute NaNO2 intoxication provokes changes in the endogenous levels of Fe, Ca and Zn in mouse spleen. These findings suggest disruption of the ionic balance and impact on the activity of antioxidant defence enzymes.  相似文献
