An acyltransferase was detected in young leaves of pedunculate oak (Quercus robur) that catalyzed the formation of 1,2,3,4,6-penta-O-galloyl-beta-D-glucose, the common precursor of gallotannins and the related ellagitannins. This enzyme depended on beta-glucogallin (1-O-galloyl-beta-D-glucose) as acyl donor; 1,2,3,6-tetra-O-galloyl-beta-D-glucose was specifically required as acceptor molecule, whereas no reaction occurred with the 1,2,4,6-isomer of this substrate. The partially purified enzyme (Mr 260,000) was stable between pH 5.0 and 6.5; highest activities were observed at pH 6.3 and 40 degrees C. Km values of 2.3 and 1.0 mM, respectively, were determined for the substrates beta-glucogallin and tetragalloylglucose. In accordance with stoichiometric studies, the systematic name "beta-glucogallin: 1,2,3,6-tetra-O-galloylglucose 4-O-galloyltransferase" is proposed for this new enzyme.  相似文献

Pathways to complex gallotannins have been elucidated by enzyme studies, indicating that beta-glucogallin is required as principal acyl donor. Evidence for the in vitro oxidation of pentagalloylglucose, the pivotal metabolite in this sequence, to ellagitannins, is presented. Immunohistochemical studies with antibodies raised against pentagalloylglucose and the galloyltransferase catalyzing the formation of this ester revealed that leaf mesophyll cell walls were a typical site of origin and deposition of hydrolyzable tannins. Seasonal changes of these compounds were studied with extracts from cell walls and intracellular space of oak leaves.  相似文献

Leaves of staghorn sumac (Rhus typhina) contain several galloyltransferases that catalyze the beta-glucogallin dependent transformation of 1,2,3,4,6-pentagalloylglucose to gallotannins. Among these, an enzyme has been isolated that preferentially acylates the 3-position of the hexagalloylglucose, 3-O-digalloyl-1,2,3,4,6-tetra-O-galloylglucose, to afford the corresponding heptagalloylglucose being characterized by a 3-O-meta-trigalloyl side-chain. The enzyme, for which a M(r) of ca. 260,000 was determined, was purified to apparent homogeneity. SDS-PAGE suggested an alpha4beta4-conformation of the native enzyme. It had a pH-optimum and an isolelectric point at pH 5.6, was most stable at pH 4.0-4.3, and displayed excellent heat-stability and in particular an extreme cold-tolerance. We propose the systematic name "beta-glucogallin: hexagalloylglucose 3-O-galloyltransferase" for this new enzyme.  相似文献

Shoot cultures of Tamarix tetrandra on Linsmaier–Skoog (LS) agar medium with 30 g l−1 sucrose, 2.13 mg l−1 indoleacetic acid and 2.25 mg l−1 benzyl adenine produced ellagitannins found in intact plants of the Tamaricaceae. This was demonstrated by the isolation of 14 monomeric–tetrameric ellagitannins from the aq. Me2CO extract of the cultured tissues. This is the first report on the production of ellagitannin tetramers by plant tissue culture. The effects of light and certain medium constituents on tissue growth and ellagitannin production were examined. The contents of representative tannins of different types [i.e., tellimagrandin II (monomer), hirtellin A (linear GOG-type dimer), hirtellin B (hellinoyl-type dimer), hirtellin C (macrocyclic-type dimer), and hirtellin T1 (linear GOG-type trimer)] in the resultant tissues in response to these factors were estimated by HPLC, and the optimal condition for production of these tannins were established. Shoots cultured on LS hormone-free medium promoted root development, and regenerated plants could adapt to ordinary soil and climate. Acclimatized and intact T. tetrandra plants that were collected in November and May, respectively, demonstrated seasonal differences in individual ellagitannin contents. HPLC comparison of individual ellagitannin contents in different plant materials (i.e., leaves, stems, and roots) of intact T. tetrandra plants is also reported. The results are discussed with respect to cellular deposition and biosynthetic relationship of tannins.  相似文献

The contents of 13 hydrolysable tannins in the leaves of white birch (Betula pubescens L.) trees were analysed at twelve stages throughout the growing season. All individual galloylglucoses, from 1-O-galloyl-beta-D-glucopyranose to 1,2,3,4,6-penta-O-galloyl-beta-D-glucopyranose, accumulated in young leaves, while ellagitannins showed significantly variable seasonal trends. The major ellagitannin during the whole growing season was pedunculagin while 2,3-(S)-HHDP-glucopyranose. the end product of the proposed ellagitannin pathway, accumulated in mature leaves. Relationships between the characteristics of seasonal variation in the contents of individual ellagitannins and their chemical structures were used to unravel the biogenesis of ellagitannins in birch leaves. Evidence of degradation of ellagitannins through hydrolysis during leaf growth and development is presented and implications for herbivory are discussed.  相似文献

First evidence for the in vitro synthesis of a dimeric ellagitannin has been obtained with cell-free extracts from the weed Tellima grandiflora (fringe cups, Saxifragaceae). Partially purified enzyme preparations from leaves of this plant catalyzed the oxidation of 1,2,3,4,6-pentagalloyl-beta-D-glucose to the monomeric ellagitannin, tellimagrandin II, followed by oxidative coupling of two units of this intermediate to yield a dimeric derivative. Chemical degradation, MALDI-TOF mass spectrometry, 1H and 13C nuclear magnetic resonance, and CD spectroscopy were employed to identify this enzyme reaction product as cornusiin E which is characterized by a (S)-valoneoyl bridge between glucose-positions 2, 4' and 6'. This result was supported by comparison with data obtained for cornusiin E that had been isolated from leaves of intact T. grandiflora plants. No indication for the earlier proposed existence of rugosin D (an isomer with a 1,4',6'-bound valoneoyl unit) in T. grandiflora has been obtained in this investigation.  相似文献

Two flavonoids and three ellagitannins, squarrosanins A, B, and C, were isolated from the leaves of Melaleuca squarrosa. The flavonoids were characterized structurally as kaempferol-3-O-(2″-O-galloyl)-glucuronide and herbacetin-3-O-glucuronide, while the ellagitannins were characterized as monomeric and dimeric C-glucosidic ellagitannins by application of spectroscopic and chemical methods. The antioxidant effect of the polyphenolic constituents of the M. squarrosa leaves was also examined in vitro, and C-glucosidic tannins including oligomers were shown to be more effective radical scavengers against 1,1-diphenyl-2-picrylhydrazyl (DPPH) than flavonoids and ordinary ellagitannins.  相似文献

Chemical study of Cunonia macrophylla, a New Caledonian Cunoniaceae, based on bioactive effects of a crude methanol extract of the leaves, detected bioactive tannins for the first time in this plant family. These ellagitannins have been identified as ellagic acid-4-O-beta-D-xylopyranoside (6), mallorepanin (3), mallotinic acid (1) along with corilagin (2), chebulagic acid (4), ellagic acid (5) and gallic acid (7) and have been shown to possess antimicrobial activity and to inhibit xanthine oxidase. Antimicrobial effects on bacterial human pathogens (Staphylococcus aureus, Corynebacterium accolans) and on a plant pathogen (Erwinia carotovora) as well as on a human pathogenic yeast (Candida albicans) were investigated. Activity is reported here for the first time for compounds 1, 3, 4 and 6. The inhibitory effects of all molecules against xanthine oxidase in relation to their structure was evaluated and compared. Compound 6 presented the best activity and seems to be of considerable interest for further studies.  相似文献

Four hydrolyzable tannins, nobotanins Q, R, S, and T, were isolated from the aqueous acetone extract of the dried leaves of Monochaetum multiflorum (Melastomataceae), a plant indigenous to Colombia. Their dimeric and tetrameric structures were elucidated by spectral and chemical methods. Eight known hydrolyzable tannin monomers and eight ellagitannin oligomers characteristic of melastomataceous plants were also characterized as tannin constituents of the plant.  相似文献

Three classes of chemically defined tannins, gallotannins, ellagitannins and condensed tannins were examined for their inhibitory activities against purified poly (ADP-ribose) glycohydrolase. Ellagitannins showed higher inhibitory activities than gallotannins. In contrast, condensed tannins, which consist of an epicathechin gallate (ECG) oligomer without a glucose core were not appreciably inhibitory. Kinetic analysis revealed that the inhibition of ellagitannins was competitive with respect to the substrate poly(ADP-ribose), whereas gallotannins exhibited mixed-type inhibition. These results suggest that conjugation with glucose of hexahydroxy-diphenoyl (HHDP) group, which is a unique component of ellagitannins, potentiated the inhibitory activity, and that the structure of ellagitannins may have a functional domain which competes with poly(ADP-ribose) on the poly(ADP-ribose) glycohydrolase molecule.  相似文献

麻栎(Quercus acutissima Carr.)是优良的能源和用材树种,在我国分布广泛并已有悠久栽培利用的历史,研究其种子和苗木性状地理变异规律对选择优良种源和适生区十分必要.研究收集了13个省(区)36个种源的麻栎种子,在测定麻栎种子性状的基础上,在安徽省滁州市红琊山林场进行了苗期试验和苗木性状测定,并采用趋势面分析方法对这些性状在经纬2维方向的地理变异模式进行了剖析.结果表明:1)麻栎种源间种子长度、宽度、百粒重和营养内含物存在极显著差异.种子百粒重、长度和宽度总体表现双向渐变趋势,随经度增高而增大,随纬度增高而减小,主要受到经度的控制;以西南到东北为中间地带,可溶性糖含量向东南表现先下降后上升的趋势,向西北则相反;淀粉含量从西北到东南呈逐渐减小的趋势.2)麻栎种源苗高、地径、生物量、热值和木材化学组分存在极显著差异.苗高、地径和生物量均呈双向渐变,经正向变异且变化幅度较大,纬负向变异且变化幅度较小,经度影响大于纬度;热值拟合回归方程不显著,方程无意义.木质素含量北部大于南部,但北部以西北部最高,南部以东南部最高.3)研究显示,麻栎种子和苗木性状多数存在显著的地理变异模式,这也是麻栎在长期进化过程中为适应复杂多变的环境而产生与之相适应的遗传变异结果.  相似文献

ent-Eudesmane sesquiterpenoids, 8,11-dihydroxy-2,4-cycloeudesmane, 11-hydroxy-2,4-cycloeudesman-8-one and 2,4-cyclo-7(11)-eudesmen-8-one, were isolated from the wood of Platycarya strobilacea, which has been used as an aromatic tree since at least the 18th century. On charring the wood, 2,4-cyclo-7(11)-eudesmen-8-one was detected in the smoke. In the charred wood, the concentrations of ellagitannins, such as galloyl pedunculagin, dramatically decreased, whereas concentrations of pentagalloyl glucose, and other gallotannins were relatively stable. In addition, two other compounds, the 6′-O-m- and p-digalloyl oak lactone precursor and the 3-O-methylellagic acid 4′-O-(4″-O-galloyl)-xylopyranoside, were isolated from the charred wood along with m- and p-digallic acid.  相似文献

A strong selection for acorn characteristics is expected to have evolved in the mutualistic relationship between the European jay (Garrulus glandarius) and the oak (Quercus spp.). Bossema's pioneer work suggested that jays do not select acorns randomly, but rather they preferentially select some size and species. Preference for some seeds over others may have implications on plant community dynamics by conferring advantages (or disadvantages) on the selected (avoided) seed characteristics. In this paper we test to what extent jays select acorns by species and/or by size and the relation between these two traits in Mediterranean oak species. The experiments consist of a set of field tests in which acorns from four different coexisting Mediterranean oak species (Quercus ilex, Quercus faginea, Quercus suber, and Quercus coccifera) were placed in artificial feeders accessible to wild jays. The acorns were previously measured to control individual acorn characteristics. Using video-recording techniques, we followed jay activity and the fate of each acorn (sequence of acorn selection and method of transport). Q. ilex acorns were preferred over other acorns, and Q. coccifera acorns were avoided when other acorns were available. Preference for Q. faginea and Q. suber acorns was intermediate, that is, they were preferred over Q. coccifera acorns but not over Q. ilex acorns. Large acorns were also preferred although acorn species selection was stronger than size selection. Jays selected species and size both by visual means and by using acorn area as an indicator of size. Acorns wider than 17–19 mm were carried in the bill because of throat limitation. Our results confirm Bossema's study on temperate oaks and extend it to Mediterranean oak species, revealing implications on mixed oak forest dynamics.  相似文献

Continuing studies on the total synthesis of ellagitannin plant metabolites have led to the preparation of the dimeric antitumor compound, coriariin A, as well as designed structural analogues. In related investigations, the synthesis of a 2,4-hexahydroxydiphenoyl (HHDP)-bearing glucopyranose structure has been achieved. This species is related to the geraniin family of ellagitannins, and its subsequent chemistry is suggestive of a mechanistic rationale for the observation that the HHDP units within (3,6-bridged)2,4-HHDP-containing ellagitannins invariably are oxidized further in vivo. Companion studies designed to assay the immunomodulatory properties of coriariin A and analogues have led to the thesis that tumor necrosis factor alpha (TNFalpha) serves as a mediator of this ellagitannin's tumor remissive activity. Furthermore, certain tannins and tannin analogues appear to act in an immunosuppressive capacity with peripheral blood monocytes that were exposed to the bacterially derived septic shock inducing agent lipid A.  相似文献

Current enzyme studies on the biosynthesis of gallotannins with cell-free extracts from leaves of staghorn sumac (Rhus typhina L.) revealed the existence of two new beta-glucogallin-dependent galloyltransferases (EC 2.3.1.-) that preferentially catalyzed the acylation of hexa- and heptagalloylglucoses. One enzyme was most active with the hexagalloylglucose, 3-O-digalloyl-1,2,4,6-tetra-O-galloylglucose, to form the corresponding heptagalloylglucose, 3-O-trigalloyl-1,2,4,6-tetra-O-galloylglucose. This polyester, in turn, was the preferred substrate for a second enzyme that catalyzed its conversion to higher substituted derivatives. This latter enzyme also displayed considerable affinity towards 2-O-digalloyl-1,3,4,6-tetra-O-galloylglucose which was acylated to various hepta- and octagalloylglucoses. These recent findings, together with data from earlier reported related enzymes, allowed the presentation of a scheme that summarizes the major transitions in the biogenetic routes from 1,2,3,4,6-pentagalloylglucose to complex gallotannins.  相似文献

There is evidence that secondary metabolism may modulate plant interactions and is modified by different biotic stress agents, such as herbivores or pathogens. However, it is poorly understood whether secondary metabolism is altered during competition among plants. The intraspecific and interspecific coexistence of some Mediterranean potted seedlings, namely Rosmarinus officinalis, Pinus halepensis, Cistus albidus and Quercus coccifera was investigated through their terpene accumulation within leaves (except for Q. coccifera, a non-storing species) and terpene emissions (for all species). Competition had both positive and negative effects for both terpene emissions and content, depending on the species a seedling coexisted with. For R. officinalis, terpene concentrations (1.8-cineole and camphor) and terpene emissions (camphene, camphor and overall monoterpenes) were lower when the neighbour species was P. halepensis. For C. albidus, no changes were observed in its content, while the overall sesquiterpene emissions (70% of total emissions) were reduced in all competition conditions, except in intraspecific competition. In the case of P. halepensis, the highest terpene content occurred when it grew with C. albidus, and in intraspecific competition, while its emissions were reduced under these conditions. Only emissions of Q. coccifera showed no significant changes in the different competition treatments.  相似文献

The individual and interactive effects of physicochemical parameters on ellagitannin acyl hydrolase activity and ellagic acid production by Aspergillus oryzae using ellagitannins from acorn fringe of oak as substrate were studied. Ellagitannins concentration, incubation time were identified as important physicochemical parameters influencing the enzyme synthesis and the production accumulation, and the substrate concentration with initial pH was determined to has an interactive effect on the enzyme synthesis, while ellagitannins concentration and initial pH with incubation time were found to have interactions on the production accumulation. Furthermore, the parameters were optimized by quadratic programming. Under optimum condition, the fermentation run lasted 84 h with 4 g L−1 ellagitannins concentration, yielding 17.7% ellagic acid. However, the maximum enzyme activity was obtained in 96 h with 5 g L−1 substrate concentration. The research demonstrated a possible way to develop an efficient approach for recovery of higher added-value product (ellagic acid) from forestry byproduct (acorn fringe of oak).  相似文献

Tyramine, an important plant intermediate, was found to be a substrate for two proteins, a copper amine oxidase and a peroxidase from Euphorbia characias latex. The oxidation of tyramine took place by two different mechanisms: oxidative deamination to p-hydroxyphenylacetaldehyde by the amine oxidase and formation of di-tyramine by the peroxidase. The di-tyramine was further oxidized at the two amino groups by the amino oxidase, whereas p-hydroxyphenylacetaldehyde was transformed to di-p-hydroxyphenylacetaldehyde by the peroxidase. Data obtained in this study indicate a new interesting scenario in the metabolism of tyramine.  相似文献
