Aim  We examined the genetic structure among populations and regions for the springtails Cryptopygus antarcticus antarcticus and Gomphiocephalus hodgsoni (Collembola) to identify potential historical refugia and subsequent colonization routes, and to examine population growth/expansion and relative ages of population divergence.
Location  Antarctic Peninsula for C. a. antarcticus ; Antarctic continent (southern Victoria Land) for G. hodgsoni .
Methods  Samples were collected from 24 and 28 locations across the Antarctic Peninsula and southern Victoria Land regions for C. a. antarcticus and G. hodgsoni , respectively. We used population genetic, demographic and nested clade analyses based on mitochondrial DNA (cytochrome c oxidase subunit I and subunit II).
Results  Both species were found to have population structures compatible with the presence of historical glacial refugia on Pleistocene (2 Ma–present) time-scales, followed by post-glacial expansion generating contemporary geographically isolated populations. However, G. hodgsoni populations were characterized by a fragmented pattern with several 'phylogroups' (likely ancestral haplotypes present in high frequency) retaining strong ancestral linkages among present-day populations. Conversely, C. a. antarcticus had an excess of rare haplotypes with a much reduced volume of ancestral lineages, possibly indicating historical founder/bottleneck events and widespread expansion.
Main conclusions  We infer that these differences reflect distinct evolutionary histories in each locality despite the resident species having similar life-history characteristics. We suggest that this has predominantly been influenced by variation in the success of colonization events as a result of intrinsic historical glaciological differences between the Antarctic Peninsula and continental Antarctic environments.  相似文献

Studies of herbivorous insects have played a major role in understanding how ecological divergence can facilitate genetic differentiation. In contrast to the majority of herbivorous insects, grasshoppers as a group are largely polyphagous. Due to this relative lack of intimate grasshopper-plant associations, grasshopper-plant systems have not played a large part in the study of host-associated genetic differentiation. The oligophagous grasshopper, Hesperotettix viridis (Thomas), is endemic to North America and feeds on composites (Asteraceae) within the tribe Astereae. Previous work has shown both preference and performance differences between H. viridis individuals feeding on either Solidago mollis or Gutierrezia sarothrae. Using 222 AFLP markers, we examined the genetic relationships among 38 H. viridis individuals feeding on these plants both in sympatry and allopatry. Neighbour-joining analysis resulted in two distinct host-associated clades with 71% bootstrap support for host-associated monophyly. Analyses of molecular variation (amova) revealed significant genetic structuring with host plant accounting for 20% of the total genetic variance while locality accounted for 0%. Significant genetic differentiation was detected between S. mollis-feeders and G. sarothrae-feeders even when the two were present at the same locality. These results are consistent with observed differences in preference and performance between H. viridis grasshoppers feeding on either G. sarothrae or S. mollis and indicate that H. viridis is comprised of at least two genetically distinct host plant-associated lineages.  相似文献

We examined the phylogeography of the endemic Antarctic collembolan Gomphiocephalus hodgsoni using allozymes and mitochondrial DNA (mtDNA; COI) to determine if potentially limited dispersal and long-term habitat fragmentation have promoted regional genetic differentiation. Allozyme analyses showed that differentiation among 21 populations within the Ross Dependency was high (FST = 0.55) with two main groups each representing a distinct geographical region: (1) Ross Island and Beaufort Island; and (2) all continental sites. Ross Island populations showed low levels of differentiation (FST = 0.05) and no correlation with geographical distance, suggesting their derivation from a single glacial refuge. By contrast, continental regions revealed moderate levels of differentiation (FST = 0.27) and a strong correlation with geographical distance, indicating a much older history with several refugia likely. Two sympatric allozyme genotypes were found at three continental sites from Taylor Valley and were congruent with two mtDNA haplotypes, implying nonrandom breeding groups. Although haplotype sharing between one Ross Island site (Cape Bird) and one continental site (Granite Harbour) was identified, the clades showed mostly fragmented allopatric distributions. The extensive Pleistocene glaciations, in conjunction with limited dispersal opportunities, appear to have promoted isolation and divergence among the fragmented habitats. Furthermore, the McMurdo Sound appears to be an effective isolating barrier to dispersal. However, we suggest that the unaided dispersal capacity of G. hodgsoni is unlikely to account for the limited mixing of haplotypes across the McMurdo Sound and recent human- or bird-mediated dispersal is highly probable.  相似文献

2004年8月,在可可西里自然保护区的楚玛尔河至五道梁一带观察藏羚羊、藏原羚和藏野驴的行为活动,并收集其粪便,运用粪便显微分析法对其食性进行研究,计算3种动物的营养生态位宽度和营养生态位重叠指数.结果表明:藏羚羊、藏原羚和藏野驴所采食的植物种类基本相似,但在食谱中所占的比例不同,禾本科植物在藏羚羊、藏原羚和藏野驴的食谱中所占的比例分别为58.7%、44.57%和9228%.藏羚羊、藏原羚和藏野驴的营养生态位宽度分别为0.878、0.735和0.695.藏羚羊和藏野驴、藏羚羊和藏原羚、藏野驴和藏原羚的营养生态位重叠值(FT)分别为0.869、0.985和0.785.结合藏羚羊、藏原羚和藏野驴的生态习性,从营养生态位的角度探讨了它们之间竞争与共存的关系.  相似文献

Patterns of genetic variation and covariation can influence the rate and direction of phenotypic evolution. We explored the possibility that the parallel morphological evolution seen in threespine stickleback (Gasterosteus aculeatus) populations colonizing freshwater environments is facilitated by patterns of genetic variation and covariation in the ancestral (marine) population. We estimated the genetic (G) and phenotypic (P) covariance matrices and directions of maximum additive genetic (g(max) ) and phenotypic (p(max) ) covariances of body shape and armour traits. Our results suggest a role for the ancestral G in explaining parallel morphological evolution in freshwater populations. We also found evidence of genetic constraints owing to the lack of variance in the ancestral G. Furthermore, strong genetic covariances and correlations among traits revealed that selective factors responsible for threespine stickleback body shape and armour divergence may be difficult to disentangle. The directions of g(max) and p(max) were correlated, but the correlations were not high enough to imply that phenotypic patterns of trait variation and covariation within populations are very informative of underlying genetic patterns.  相似文献

Information exchange (or signaling) between plants following herbivore damage has recently been shown to affect plant responses to herbivory in relatively simple natural systems. In a large, manipulative field study using three annual plant species (Achyrachaena mollis, Lupinus nanus, and Sinapis arvensis), we tested whether experimental damage to a neighboring conspecific affected a plant's lifetime fitness and interactions with herbivores. By manipulating relatedness between plants, we assessed whether genetic relatedness of neighboring individuals influenced the outcome of having a damaged neighbor. Additionally, in laboratory feeding assays, we assessed whether damage to a neighboring plant specifically affected palatability to a generalist herbivore and, for S. arvensis, a specialist herbivore. Our study suggested a high level of contingency in the outcomes of plant signaling. For example, in the field, damaging a neighbor resulted in greater herbivory to A. mollis, but only when the damaged neighbor was a close relative. Similarly, in laboratory trials, the palatability of S. arvensis to a generalist herbivore increased after the plant was exposed to a damaged neighbor, while palatability to a specialist herbivore decreased. Across all species, damage to a neighbor resulted in decreased lifetime fitness, but only if neighbors were closely related. These results suggest that the outcomes of plant signaling within multi-species neighborhoods may be far more context-specific than has been previously shown. In particular, our study shows that herbivore interactions and signaling between plants are contingent on the genetic relationship between neighboring plants. Many factors affect the outcomes of plant signaling, and studies that clarify these factors will be necessary in order to assess the role of plant information exchange about herbivory in natural systems.  相似文献

Several lines of evidence suggest that patterns of genetic variability in the human angiotensinogen gene (AGT) contribute to phenotypic variability in human hypertension. The A(-6) promoter variant of AGT is associated with higher plasma angiotensinogen levels and increased risk of essential hypertension. The geographic distribution of the A(-6) variant leads to the intriguing hypothesis that the G(-6) promoter variant has been selectively advantageous outside Africa. To test these hypotheses, we investigated the roles of population history and natural selection in shaping patterns of genetic diversity in AGT, by sequencing the entire AGT gene (14400 bp) in 736 chromosomes from Africa, Asia, and Europe. We found that the A(-6) variant is present at higher frequency in African populations than in non-African populations. Neutrality tests found no evidence of a departure from selective neutrality, when whole AGT sequences were compared. However, tests restricted to sites in the vicinity of the A(-6)G polymorphism found evidence of a selective sweep. Sliding-window analyses showed that evidence of the sweep is restricted to sites in tight linkage disequilibrium (LD) with the A(-6)G polymorphism. Further, haplotypes carrying the G(-6) variant showed elevated levels of LD, suggesting that they have risen recently to high frequency. Departures from neutral expectation in some but not all regions of AGT indicate that patterns of diversity in the gene cannot be accounted for solely by population history, which would affect all regions equally. Taken together, patterns of genetic diversity in AGT suggest that natural selection has generally favored the G(-6) variant over the A(-6) variant in non-African populations. Dimorphandra mollis (Leguminosae), known as faveiro and fava d'anta, is a tree that is widely distributed throughout the Brazilian Cerrado (a savanna-like biome). This species is economically valuable and has been extensively exploited because its fruits contain the flavonoid rutin, which is used to produce medications for human circulatory diseases. Knowledge about its genetic diversity is needed to guide decisions about the conservation and rational use of this species in order to maintain its diversity. DNA extraction is an essential step for obtaining good results in a molecular analysis. However, DNA isolation from plants is usually compromised by excessive contamination by secondary metabolites. DNA extraction of D. mollis, mainly from mature leaves, results in a highly viscous mass that is difficult to handle and use in techniques that require pure DNA. We tested four protocols for plant DNA extraction that can be used to minimize problems such as contamination by polysaccharides, which is more pronounced in material from mature leaves. The protocol that produced the best DNA quality initially utilizes a sorbitol buffer to remove mucilaginous polysaccharides. The macerated leaf material is washed with this buffer until there is no visible mucilage in the sample. This protocol is adequate for DNA extraction both from young and mature leaves, and could be useful not only for D. mollis but also for other species that have high levels of polysaccharide contamination during the extraction process.  相似文献

采用接种和不接种(对照)丛枝菌根的方法对翅果油树幼苗在不同生长时期的形态、生物量、矿质元素含量及抗逆性等指标进行比较分析,以探讨丛枝菌根(AM)改善翅果油树在生态系统中的退化及濒危现状的机理.结果显示:(1)摩西球囊霉(Glomus mosseae)和脆无梗囊霉(Acaulospora delicata)与盆栽翅果油树苗木可形成菌根;菌根化苗木的形态及生物量等指标均与对照差异极显著,且与菌根侵染率呈显著正相关(P<0.05).(2)菌根使苗木根系体积增大、总吸收面积增加、根系的活跃吸收面积显著增加,且根系中活跃吸收面积比与磷、钾元素含量呈显著正相关(P<0.05).(3)苗木生长的盛期和末期,菌根化苗木根系过氧化物酶活性显著高于对照,而在生长后期却显著低于对照呈负相关(P<0.05);菌根化苗木根系多酚氧化酶活性显著高于对照(P<0.05),在苗木生长盛期酶活性最强,但接种不同菌种的苗木间差异不显著.研究表明,与不接种相比,接种菌根增加了翅果油树的根系吸收能力,提高了根系酶体系,有利于植物抵抗各种胁迫,对扩大翅果油树植物的分布区有重要意义.  相似文献

We examined levels of genetic variation and genetic structure in the leafy cactus (Pereskia guamacho) in arid and semiarid zones in Venezuela. We surveyed genetic diversity within 17 populations using 19 allozyme loci. Genetic diversity was relatively high at both the species (P(s) = 89%, A(s) = 3.26, AP(s) = 3.53, H(es) = 0.24) and population (P(p) = 63%, A(p) = 1.90, AP(p) = 2.42, H(ep) = 0.20) levels. A significant deficit of heterozygote individuals was detected within populations in the Paraguana Peninsula region (F(IS) = 0.301). Relatively low levels of population differentiation were detected at macrogeographic (G(ST) = 0.112) and regional levels (G(ST) = 0.044 for peninsula region and G(ST) = 0.074 for mainland region), suggesting substantial genetic exchange among populations; however, gene flow in this species seems to be regulated by the distance among populations. Overall, estimates of genetic diversity found in P. guamacho are concordant with the pattern observed for other cacti surveyed, namely high levels of polymorphism and genetic diversity with one common allele and several rare alleles per locus. Differences in gene dispersal systems between this species and other cacti studied were not reflected in the patterns of genetic diversity observed. The concentration of the highest estimates of genetic variation in northwestern Venezuela suggests a potential reservoir of plant genetic diversity within xerophilous ecosystems in northern South America.  相似文献

Genetic and genomic relationships among three taxonomically related species of Leymus, northern European L. arenarius (octoploid, 2n = 56), northern American/Pacific L. mollis (tetraploid, 2n = 28) and central Eurasian L. racemosus (tetraploid, 2n = 28), were examined using molecular and cytogenetic methods. The amplified fragment length polymorphism (AFLP) analysis clearly differentiated Icelandic populations of L. arenarius from Alaskan populations of L. mollis. The former group is more genetically homogeneous than the latter. Leymus arenarius in Iceland has a common gene pool and a relatively recent origin. The Alaskan L. mollis, on the other hand, is probably a glacial survival that has accumulated high level of genetic variation and has differentiated into subspecies. Analysis of the 18S-26S ribosomal genes, by restriction fragment length polymorphism (RFLP) and fluorescence in situ hybridization (FISH), revealed a very close relationship between the octoploid northern European L. arenarius and the tetraploid Eurasian L. racemosus, such that the former could have originated from the latter, probably via interspecific hybridization. Leymus-specific DNA sequences were isolated and used for analyzing genetic relatedness among five Leymus species and four Psathyrostachys species. The RFLP analysis of retrotransposon sequence pLm44 and ribosomal clone pTa71 clearly revealed a close relationship between these two genera, i.e. higher variation was found within genera than between them. The results support the previous notion that Leymus is autopolyploid having all genomes being designated Ns as in Psathyrostachys, but a major taxonomic revision of this group would require analysis of more species.  相似文献

The oxazolidinones are a new class of potent antibiotics that are active against a broad spectrum of Gram-positive bacterial pathogens including those resistant to other antibiotics. These drugs specifically inhibit protein biosynthesis whereas DNA and RNA synthesis are not affected. Although biochemical and genetic studies indicate that oxazolidinones target the ribosomal peptidyltransferase center, other investigations suggest that they interact with different regions of ribosomes. Thus, the exact binding site and mechanism of action have remained elusive. Here, we study, by use of base-specific reagents, the effect of the oxazolidinones on the chemical protection footprinting patterns of the 23S rRNA. We report: (i) reproducible protection of G2607 and G2608 of 23S rRNA by a potent oxazolidinone on a ribosome.tRNA.mRNA complex; (ii) no protections were observed on 70S ribosomes devoid of tRNA and mRNA; (iii) EF-G also weakly protected G2607 and G2608; (iv) mutations at G2608 conferred resistance to the oxazolidinones in Escherichia coli cells; and (v) G2607 and G2608 occur near the exit to the peptide tunnel on the 50S subunit. A mechanism for the pleiotropic action of the oxazolidinones is discussed.  相似文献

利用细胞学、染色体C分带和原位杂交方法,对八倍体小黑麦劲松49与八倍体小滨麦杂种后代选育的稳定遗传材料山农030-1进行了鉴定。染色体观察结果表明,山农030-1根尖染色体数目为42,花粉母细胞减数分裂中期I(PMC MI)染色体构型为2n=21Ⅱ。分别以黑麦、滨麦草基因组DNA为探针进行原位杂交,表明山农030-1含有一对黑麦和小麦的整臂易位染色体,不含有来自滨麦草的遗传物质。染色体C分带结果进一步表明此材料为1RS·1BL易位系。接种鉴定表明,山农030-1对白粉病免疫。Abstract: In the progenies of octoploid Triticale Jinsong49 crossed with octoploid Tritileymus, a stable germplasm line named Shannong030-1 was obtained. Its chromosome number was 42, and 21 bivalents could be observed at PMC MI. FISH analysis by using genomic DNAs from S. cereale and genomic DNAs from L. mollis as probes,respectively, showed that Shannong030-1 contained a pair of translocation chromosome between rye and wheat, and had no genetic materials from L. mollis . C-banding analysis showed that it was translocation lines of 1RS·1BL.Resistance identification showed Shannong030-1 was immune to powdery mildew.  相似文献

Accurate determination of patterns of genetic variation provides a powerful inferential tool for studies of evolution and conservation. For more than 30 years, enzyme electrophoresis was the preferred method for elucidating these patterns. As a result, evolutionary geneticists have acquired considerable understanding of the relationship between patterns of allozyme variation and aspects of evolutionary process. Myriad molecular markers and statistical analyses have since emerged, enabling improved estimates of patterns of genetic diversity. With these advances, there is a need to evaluate results obtained with different markers and analytical methods. We present a comparative study of gene statistic estimates (F(ST), G(ST), F(IS), H(S), and H(T)) calculated from an intersimple sequence repeat (ISSR) and an allozyme data set derived from the same populations using both standard and Bayesian statistical approaches. Significant differences were found between estimates, owing to the effects of marker and analysis type. Most notably, F(ST) estimates for codominant data differ between Bayesian and standard approaches. Levels of statistical significance are greatly affected by methodology and, in some cases, are not associated with similar levels of biological significance. Our results suggest that caution should be used in equating or comparing results obtained using different markers and/or methods of analysis.  相似文献

Population structures of the delicate loach, Niwaella delicata, were inferred from morphology and restriction fragment length polymorphism (RFLP) analysis of part of the mitochondrial DNA (mtDNA) of 25 populations, representing the species range in central Honshu Island. The existence of two types of morphological variation corresponding to regional distributions, the "Pacific slope type" and "Sea of Japan slope type," has been known in N. delicata. Our morphological reexamination of the two types revealed some discrepancies in their distribution pattern. Therefore, we reclassified two new color types corresponded to their distribution areas as "gathered spots type (G type)" and "scattered spots type (S type)," respectively. The present classification of G and S types is closely related to the mtDNA divergence pattern. The current analysis also indicated that each G and S type population was further divided into two genetic groups, corresponding to geographic proximity. In spite of marked morphological differentiation, the genetic diversity between G and S type populations (1.153%) was comparable only to that reported for intraspecific levels in most freshwater fishes. Moreover, in the population of which the color patterns of all fish were characterized to the S type, mtDNA haplotypes corresponding to G and S types were sympatrically detected. This result indicates secondary contact between the two type populations and the possibility that they are not reproductively isolated. DNA damage is a threat to genomic integrity in all living organisms. Plants and green algae are particularly susceptible to DNA damage especially that caused by UV light, due to their light dependency for photosynthesis. For survival of a plant, and other eukaryotic cells, it is essential for an organism to continuously check the integrity of its genetic material and, when damaged, to repair it immediately. Cells therefore utilize a DNA damage response pathway that is responsible for sensing, reacting to and repairing damaged DNA. We have studied the effect of 5-fluorodeoxyuridine, zeocin, caffeine and combinations of these on the cell cycle of the green alga Scenedesmus quadricauda. The cells delayed S phase and underwent a permanent G2 phase block if DNA metabolism was affected prior to S phase; the G2 phase block imposed by zeocin was partially abolished by caffeine. No cell cycle block was observed if the treatment with zeocin occurred in G2 phase and the cells divided normally. CDKA and CDKB kinases regulate mitosis in S. quadricauda; their kinase activities were inhibited by Wee1. CDKA, CDKB protein levels were stabilized in the presence of zeocin. In contrast, the protein level of Wee1 was unaffected by DNA perturbing treatments. Wee1 therefore does not appear to be involved in the DNA damage response in S. quadricauda. Our results imply a specific reaction to DNA damage in S. quadricauda, with no cell cycle arrest, after experiencing DNA damage during G2 phase.  相似文献

Callicarpa x shirasawana is a natural hybrid between C. japonica and C. mollis, and has a morphology that is intermediate between those of the parent species. Characterization of natural Callicarpa populations in the Atsumi Peninsula of central Japan, which all three of the above species inhabit sympatrically, revealed hybrids with various morphologies. Molecular analysis revealed a high occurrence of introgression of the C. japonica genome into that of C. mollis. Moreover, all individuals examined with morphology similar to that of C. mollis had genetic traces of hybridization with C. japonica. Molecular analysis of individual C. mollis and C. japonica from five other areas of Japan showed that introgression of C. japonica into C. mollis occurs widely. Molecular data also strongly suggested that the previously recognized C. x shirasawana individuals with intermediate morphology are not F1 hybrids between C. japonica and C. mollis, but instead are progeny of C. x shirasawana backcrossed with C. japonica. Moreover, it was revealed that individuals with F1-type genotypes are indistinguishable morphologically from pure C. mollis. The results of the present study point to the need for re-evaluation of natural populations of C. mollis and C. x shirasawana.  相似文献

Aim  We tested whether the distribution of three common springtail species ( Gressittacantha terranova , Gomphiocephalus hodgsoni and Friesea grisea ) in Victoria Land (Antarctica) could be modelled as a function of latitude, longitude, altitude and distance from the sea.
Location  Victoria Land, Ross Dependency, Antarctica.
Methods  Generalized linear models were constructed using species presence/absence data relative to geographical features (latitude, longitude, altitude, distance from sea) across the species' entire ranges. Model results were then integrated with the known phylogeography of each species and hypotheses were generated on the role of climate as a major driver of Antarctic springtail distribution.
Results  Based on model selection using Akaike's information criterion, the species' distributions were: hump-shaped relative to longitude and monotonic with altitude for Gressittacantha terranova ; hump-shaped relative to latitude and monotonic with altitude for Gomphiocephalus hodgsoni ; and hump-shaped relative to longitude and monotonic with latitude, altitude and distance from the sea for Friesea grisea .
Main conclusions  No single distributional pattern was shared by the three species. While distributions were partially a response to climatic spatial clines, the patterns observed strongly suggest that past geological events have influenced the observed distributions. Accordingly, present-day spatial patterns are likely to have arisen from the interaction of historical and environmental drivers. Future studies will need to integrate a range of spatial and temporal scales to further quantify their respective roles.  相似文献

We sequenced a part of the mitochondrial cytochrome-b gene from the seven treecreeper species, including 18 subspecies, to reconstruct the phylogeny of the genus Certhia . Species status of all seven species could be affirmed. Certhia discolor , C. himalayana , C . nipalensis , and C . tianquanensis , the species with relatively small distribution ranges in southeast Asia and simple territorial song, are found at the base of all phylogenetic trees, although without good support. A comparatively recent sister species of C. tianquanensis is C. nipalensis , replacing C. discolor as closest relative. Certhia familiaris , C. brachydactyla and C. americana form a derived set of species (again only weak support). The closest relative of C. americana is C. brachydactyla . The C. familiaris subtree is deeply split into two well-defined population groups: a Eurasian group including populations in northern China (Qinling range northward and all Eurasia) and a Sino-Himalayan group (Himalayas and China excluding northern China). In accordance with acoustic characters, the three subspecies hodgsoni , mandellii and khamensis of the Sino-Himalayan group are combined and elevated to species rank: Certhia hodgsoni . Certhia discolor manipurensis is deeply split from nominate discolor as well, and is also promoted to species level. Within C. brachydactyla (western palearctic) and within C. hodgsoni (Himalayas, China) several populations form well-supported separate lineages that diverged quite recently and represent subspecies level. In all other species, molecular–genetic and vocal characters support traditional species delimitation.  相似文献

Neothyonidioside is a triterpene glycoside (TG) isolated from the sea cucumber, Australostichopus mollis, that is potently cytotoxic to S. cerevisiae, but does not permeabilize cellular membranes. We mutagenized S. cerevisiae and isolated a neothionidioside-resistant (neo(R)) strain. Using synthetic genetic array mapping and sequencing, we identified NCP1 as the resistance locus. Quantitative HPLC revealed that neo(R)/ncp1 mutants have reduced ergosterol content. Ergosterol added to growth media reversed toxicity, demonstrating that neothionidioside binds directly to ergosterol, similar to the polyene natamycin. Ergosterol synthesis inhibitors ketoconazole and atorvastatin conferred resistance to neothionidioside in a dose-dependent manner showing that a threshold ergosterol concentration is required for toxicity. A genome-wide screen of deletion mutants against neothionidioside revealed hypersensitivity of many of the component genes in the ESCRT complexes relating to multivesicular body formation. Confocal microscopy of cells stained with a vital dye showed blockage at this step. Thus, we propose neothionidioside may affect membrane curvature and fusion capability in the endosome-vacuole pathway.  相似文献
