The biotransformation of [2-14C](±)9, 10-dihydrojasmonic acid (DJA) was studied in excised shoots of 6-day-old barley seedlings after 72 h. From the ethyl acetate extract, some minor metabolites were isolated and purified by DEAE-Sephadex A-25 chromatography, thin-layer chromatography (TLC), C18-cartridges, and high-performance liquid chromatography (HPLC). The structural identification of these metabolites was performed by gas chromatography-mass spectrometry (GC-MS), circular dichroism (CD), and amino acid analysis, and the following amino acid conjugates were found:N-[(?)9,10-dihydrojasmonoyl]valine,N-[(?)9,10-dihydrojasmonoyl]isoleucine,N-[9,10-dihydrojasmonoyl]leucine,N-[11-hydroxy-9,10-dihydrojasmonoyl]valine,N-[11-hydroxy-9,10-dihydrojasmonoyl]isoleucine,N-[12-hydroxy-9,10-dihydrojasmonoyl]isoleucine; and the cucurbic acid-related compoundsN-{[3-hydroxy-2(4-hydroxypentyl)-cyclopent-1-yl]-acetyl}isoleucine andN-{[3-hydroxy-2(5-hydroxypentyl)-cyclopent-1-yl]-acetyl}isoleucine. The results suggest conjugation with isoleucine and valine, as well as preferential hydroxylation at position C-11 or hydrogenation at position C-6, as being important steps in the metabolism of (±)DJA in barley shoots.  相似文献

Water stress in the leaves was induced by gradual decreasing of substrate moisture in five-day cycles.The hydration level of the leaves was characterized by their water potential (ψw), osmotic potential (ψs), pressure potential ψp and water saturation deficit(ΔW sat ).The activities of Photosystems 1 and 2 were determined polarographically with Pt/Ag(AgOH) electrode as changes in oxygen concentration in chloroplast suspensions. The shape of light curves of Hill reaction was not influenced by leaf (ψw), hence both quantum efficiency and dark phase of this process were affected in a similar manner by water deficit. The activities of both photosystems measured at saturating photon flux density declined with the lowering of leaf (ψw) (in the range from -5 to -14 x 105 Pa) and the decrease in activity of Photosystem 2 was more rapid than that of Photosystem 1. The ratio of activities of Photosystems 1 and 2 was mildly enhanced by a lowering of (ψw), but it decreased with increasing age. The lowering of ψwinduced lowering in the chlorophyll a/b ratio thus concealing the usual ontogenetic course of this ratio.  相似文献

The syntheses of two potential cAMP affinity lables, 1,N 6-(3-iodopropyleno)adenosine 3′,5′-cyclic-monophosphate and 2′-O-(2-iodo-3-hydroxypropyl) adenosine 3′,5′-cyclic-monophosphate, by a two-step chemical procedure are described. TheN 6- and 2′-O-allyl intermediates were prepared selectively by alkylation of cAMP in organic and alkaline aqueous solutions, respectively. Treatment of theN 6-allyl derivative withN-iodosuccinimide resulted in iodine addition to the double bond and cyclization to theN 1 position of the purine ring. The iodohydrin analog was synthesized by reaction of 2′-O-allyl-cAMP with potassium iodide and thallium trichloride in acetate buffered solution. The products were isolated by column chromatography and characterized by thin-layer chromatography, elemental analysis, and ultraviolet,13C, and1H NMR spectroscopy. The cAMP analogs were found to react with lysine and cysteine. Both cAMP derivatives were tested for their reaction with the low-K m cAMP phosphodiesterase of human platelets. The ribose-substituted analog functioned as a competitive inhibitor (K I =0.72 μM) and caused a time-dependent irreversible inactivation of the phosphodiesterase. In contrast, the purine-substituted derivative acted neither as a reversible competitive inhibitor nor as an irreversible inactivator of the enzyme. These results indicate the specificity of these potential cAMP analogs in their interaction with the phosphodiesterase.  相似文献

Most reproductive traits have low heritability and are greatly affected by environmental factors. Teat number and litter size are traits related to the reproduction ability of pigs. To identify quantitative trait loci (QTLs) for teat number traits, a genome-wide association study (GWAS) was conducted using an F2 intercross between Landrace and Korean native pigs. Genotype analysis was performed using the porcine SNP 60 K beadchip. The GWAS was performed using a mixed-effects model and linear regression approach. When a genome-wide threshold was determined using the Bonferroni method (P = 1.61 × 10?6), 38 single nucleotide polymorphism (SNP) markers in pig chromosome 7 (SSC7) were significantly associated with three teat number traits (total teat number, left teat number, and right teat number). Among these, SNPs in 5 genes (HDDC3, LOC100156276, LOC100155863, ANPEP, SCAMP2) were selected for further study based primarily on their statistical significance. A significant association was detected in SCAMP2 g.25280 G>A for total teat number (P = 2.0 × 10?12), HDDC3 g.1319 G>A SNP for left teat number (P = 2.3 × 10?7), and SCAMP2 g.14198 G>A for right teat number (P = 4.7 × 10?12). These results provide valuable information about the selective breeding for desirable teat numbers in pigs.  相似文献

The Central Andes in northern Chile contains a large number of closed basins whose central depression is occupied by saline lakes and salt crusts (salars). One of these basins is Salar de Llamara (850 m a.s.l.), where large domed structures of seemingly evaporitic origin forming domes can be found. In this work, we performed a detailed microbial characterization of these domes. Mineralogical studies revealed gypsum (CaSO4) as a major component. Microbial communities associated to these structures were analysed by 454 16S rDNA amplicon sequencing and compared between winter and summer seasons. Bacteroidetes Proteobacteria and Planctomycetes remained as the main phylogenetic groups, an increased diversity was found in winter. Comparison of the upper air-exposed part and the lower water-submerged part of the domes in both seasons showed little variation in the upper zone, showing a predominance of Chromatiales (Gammaproteobacteria), Rhodospirillales (Alphaproteobacteria), and Sphingobacteriales (Bacteroidetes). However, the submerged part showed marked differences between seasons, being dominated by Proteobacteria (Alpha and Gamma) and Verrucomicrobia in summer, but with more diverse phyla found in winter. Even though not abundant by sequence, Cyanobacteria were visually identified by scanning electron microscopy (SEM), which also revealed the presence of diatoms. Photosynthetic pigments were detected by high-performance liquid chromatography, being more diverse on the upper photosynthetic layer. Finally, the system was compared with other endoevaporite, mats microbialite and Stromatolites microbial ecosystems, showing higher similitude with evaporitic ecosystems from Atacama and Guerrero Negro. This environment is of special interest for extremophile studies because microbial life develops associated to minerals in the driest desert all over the world. Nevertheless, it is endangered by mining activity associated to copper and lithium extraction; thus, its environmental protection preservation is strongly encouraged.  相似文献

The effects of mineral nutrient were examined on in vitro growth of Gerbera hybrida (G. jamesonii?×?G. viridifolia), specifically Gerbera hybrida cv. Pasadena. Four types of experiments were conducted to quantify the effects of mineral nutrients on four in vitro growth responses (quality, shoot number, leaf number, and shoot height) of gerbera and included groups of mineral nutrients (macros/mesos, micros, and Fe), individual salts (CuSO4·5H2O, MnSO4·4H2O, ZnSO4·7H2O, and Fe/EDTA), and the specific ions NO3 ?, NH4 +, and K+. Experiments included mixture-amount designs that are essential for separating the effects of proportion and concentration. Highly significant effects were observed in all experiments, but the mineral nutrients with the largest effects varied among the four growth responses. For example, leaf number was strongly affected by the macronutrient group in one experiment and by NH4 + and K+, which were in the macronutrient group, in the NO3 ?/NH4 +/K+ ion-specific experiment, whereas quality was strongly affected by the micronutrients ZnSO4 and Fe/EDTA. Because mineral nutrient effects varied significantly with the response measured, defining an appropriate formulation requires a clear definition of “optimal” growth.  相似文献

Adult myogenesis responsible for the maintenance and repair of muscle tissue is mainly under the control of myogenic regulatory factors (MRFs) and a few other genes. Transthyretin gene (TTR), codes for a carrier protein for thyroxin (T4) and retinol binding protein bound with retinol in blood plasma, plays a critical role during the early stages of myogenesis. Herein, we investigated the relationship of TTR with other muscle-specific genes and report their expression in muscle satellite cells (MSCs), and increased messenger RNA (mRNA) and protein expression of TTR during MSCs differentiation. Silencing of TTR resulted in decreased myotube formation and decreased expression of myosin light chain (MYL2), myosin heavy chain 3 (MYH3), matrix gla protein (MGP), and voltage-dependent L type calcium channel (Cav1.1) genes. Increased mRNA expression observed in TTR and other myogenic genes with the addition of T4 decreased significantly following TTR knockdown, indicating the critical role of TTR in T4 transportation. Similarly, decreased expression of MGP and Cav1.1 following TTR knockdown signifies the dual role of TTR in controlling muscle myogenesis via regulation of T4 and calcium channel. Our computational and experimental evidences indicate that TTR has a relationship with MRFs and may act on calcium channel and related genes.  相似文献

The effects of salt stress on nitrogenase (N2-ase) activity, growth and nitrogen content ofVicia faba (L.),Medicago sativa (L.) Merrill,Glycine max andVigna sinensis (L.) were investigated. Four levels of salinity were applied and salt treatments were imposed on inoculated and N-fertilized plants.M. sativa tolerated mild levels of salinity but higher salt concentrations depressed N2-ase activity of this species. The other three legumes were considerably affected by salt treatments, and N2-ase activity was significantly reduced by salinity. Vicia faba, carrying elongate nodules, could restore a partial N2-ase activity upon recovery from salt stress whereasG. max andV. sinesis, both with spherical nodules, could not regain significant activity when salinity was removed. Salt stress retarded growth of both inoculated and N-fertilized plants. The nitrogen content of both treatments was also affected by salinity and the effect was more severe for inoculated than N-fertilized plants.  相似文献

In the present study, we synthesized silver and gold nanoparticles with a particle size of 10–20 nm, using Zingiber officinale root extract as a reducing and capping agent. Chloroauric acid (HAuCl4) and silver nitrate (AgNO3) were mixed with Z. officinale root extract for the production of silver (AgNPs) and gold nanoparticles (AuNPs). The surface plasmon absorbance spectra of AgNPs and AuNPs were observed at 436–531 nm, respectively. Optimum nanoparticle production was achieved at pH 8 and 9, 1 mM metal ion, a reaction temperature 50 °C and reaction time of 150–180 min for AgNPs and AuNPs, respectively. An energy-dispersive X-ray spectroscopy (SEM–EDS) study provides proof for the purity of AgNPs and AuNPs. Transmission electron microscopy images show the diameter of well-dispersed AgNPs (10–20 nm) and AuNPs (5–20 nm). The nanocrystalline phase of Ag and Au with FCC crystal structures have been confirmed by X-ray diffraction analysis. Fourier transform infrared spectroscopy analysis shows the respective peaks for the potential biomolecules in the ginger rhizome extract, which are responsible for the reduction in metal ions and synthesized AgNPs and AuNPs. In addition, the synthesized AgNPs showed a moderate antibacterial activity against bacterial food pathogens.  相似文献

The surface anionic groups of untreated or dimethyl sulfoxide (DMSO)-treatedHerpetomonas samuelpessoai cells were analyzed by cell electrophoresis, ultrastructural cytochemistry, and identification of sialic acids using thin-layer chromatography. Differentiation ofH. samuelpessoai induced by DMSO treatment caused a significant increase in the net negative surface charge. In flagellates exposed to DMSO, more cationized ferritin, colloidal iron hydroxide, and sendai virus particles bound to the cell surface. Treatment of both untreated and DMSO-treated flagellates with neuraminidase decreased markedly the EPM of cells to the cathodic pole. These findings suggest that sialic acid residues are the major anionogenic groups exposed on the surface ofH. samuelpessoai. Thin-layer chromatography showed thatN-acetyl andN,O-diacylneuraminic acids, in equal proportions, were present inH. samuelpessoai. However,N-acetylneuraminic acid predominates in DMSO-treated cells.  相似文献

Symbiotic relationships between N2-fixing prokaryotes and their autotrophic hosts are essential in nitrogen (N)-limited ecosystems, yet the importance of this association in pristine boreal peatlands, which store 25 % of the world’s soil (C), has been overlooked. External inputs of N to bogs are predominantly atmospheric, and given that regions of boreal Canada anchor some of the lowest rates found globally (~1 kg N ha?1 year?1), biomass production is thought to be limited primarily by N. Despite historically low N deposition, we show that boreal bogs have accumulated approximately 12–25 times more N than can be explained by atmospheric inputs. Here we demonstrate high rates of biological N2-fixation in prokaryotes associated with Sphagnum mosses that can fully account for the missing input of N needed to sustain high rates of C sequestration. Additionally, N amendment experiments in the field did not increase Sphagnum production, indicating that mosses are not limited by N. Lastly, by examining the composition and abundance of N2-fixing prokaryotes by quantifying gene expression of 16S rRNA and nitrogenase-encoding nifH, we show that rates of N2-fixation are driven by the substantial contribution from methanotrophs, and not from cyanobacteria. We conclude biological N2-fixation drives high sequestration of C in pristine peatlands, and may play an important role in moderating fluxes of methane, one of the most important greenhouse gases produced in peatlands. Understanding the mechanistic controls on biological N2-fixation is crucial for assessing the fate of peatland carbon stocks under scenarios of climate change and enhanced anthropogenic N deposition.  相似文献

A simple model for membrane fusion mediated by vial spike glycoproteins is presented. The viral proteins are considered to be allosteric proteins that undergo concerted conformational transitions when they bind the ligand. The ligand in this case is H+. The effect of the conformational transition is to bring membranes together and induce their fusion. An equation is derived for the dependence of fusion rates on ligand concentration, for a given dissociation constant (K d), equilibrium constant for the conformational change (L), and number of cooperating subunits (n). Curves generated by this equation provide a reasonable fit to data on the rates of fusion of Vesicular Stomatitis virus with cells for a pK d of 6.3,L=1000 andn=6.  相似文献

Intercropping, a traditional and worldwide cropping practice, has been considered as a paradigm of sustainable agriculture based on complementary mechanisms among different crop species. Soybean (Glycine max) is widely relay intercropped about 60 days before maize (Zea mays) harvest in Southwest China. However, shade caused by tall maize plants may be a limiting factor for soybean growth at a seedling stage. In field research, we studied the ecophysiological responses of two widely cultivated soybean varieties [Gongqiudou494-1 (GQD) and Gongxuan 1 (GX)] to maize shading in a relay intercropping system (RI) compared with monocropped soybean plants (M). Our results showed that soybean seedlings intercropped with maize exhibited significantly downregulated net photosynthetic rate (P N) (?38.3%), transpiration rate (?42.7%), and stomatal conductance (?55.4%) due to low available light. The insignificant changes in intercellular CO2 concentration and the maximum efficiency of PSII photochemistry suggested that the maize shading-induced depressions in P N were probably caused by the deficiency of energy for carbon assimilation. The significantly increased total chlorophyll (Chl) content (+27.4%) and Chl b content (+52.2%), with lowered Chl a/b ratios (?20.5%) indicated soybean plants adjusted their light-harvesting efficiency under maize shading condition. Biomass and leaf area index (LAI) of seedlings under RI decreased significantly (?78.7 and ?71%, respectively) in comparison with M. Correlation analysis indicated the relative reduction in biomass accumulation was caused by the decline in LAI rather than P N, it affected negatively the final yields of soybean (32.8%). Cultivar-specific responses to maize shading were observed in respects of LAI, biomass, and grain yield. It indicated that GX might be a better cultivar for relay intercropping with maize in Southwest China.  相似文献

In a survey of endophytic fungal biodiversity, an antimicrobial endophytic isolate zjwcf069 was obtained from twigs of Forsythia viridissima, Zhejiang Province, Southeast China. Zjwcf069 was then identified as Pezicula sp. through combination of morphological and phylogenetic analysis based on ITS-rDNA. Zjwcf069 here represented the first endophytic fungus in Pezicula isolated from host F. viridissima. From the fermentation broth, four compounds were obtained through silica gel column chromatography and Sephadex LH-20 under the guide of bioassay. Their structures were elucidated by spectroscopic analysis as mellein (1), ramulosin (2), butanedioic acid (3), and 4-methoxy-1(3H)-isobenzofuranone (4). Compound 4 here stood for the very first time as natural product from microbes. In vitro antifungal assay showed that compound 1 displayed growth inhibition against 9 plant pathogenic fungi, especially Botrytis cinerea and Fulvia fulva with EC50 values below 50 μg/mL. Endophytic fungi in medicinal plants were good resources for bioactive secondary metabolites.  相似文献

The aim of this study was to evaluate genetic variability in the Turkish Arab horse population using pedigree information. This study is the first detailed pedigree analysis of the breed in Turkey. Pedigree data were collected from the National Studbook. The pedigree data for 23 668 horses, born between 1904 and 2014, were used in the analysis. From this data set, a reference population (RP) of 14 838 animals symbolising the last generation was defined. Demographic parameters, the inbreeding level (F), the average relatedness (AR), the effective population size (Ne), the effective number of founders (fe), the effective number of ancestors (fa) and the number of founder genome equivalents (fg) were calculated for the population. The average generation interval for the RP was 12.2±4.6 years, whereas the calculated pedigree completeness levels were 98.2%, 96.6% and 95.0% for the first, second and third known generations. The mean equivalent generations (t), the average complete generations and the mean maximum generations for the RP were 7.8, 5.4 and 12.2, respectively, whereas the meanFand AR were 4.6% and 9.5% for the RP. The rate of inbred animals was 94.2% for the RP, whereas the number of founders, the number of ancestors and thefe,faandfgwere 342, 223, 40, 22 and 9.6 for the RP. The large differences observed betweenfe, and the number of founders demonstrates that genetic diversity decreased between the founder and the RP. Contribution of the 14 most influential founder to the RP was 50.0%, whereas just eight ancestral horses can account for 50% of the genetic variability.Neestimated via an individual increase in inbreeding per generation (N¯e), and paired increase in coancestry(N¯eC), were 74.4±3.9 and 73.5±0.58, respectively. The inbreeding increases with the pedigree knowledge. In addition, the decrease in inbreeding in last years is more noticeable.  相似文献
