Aphid transmission studies of two soybean mosaic virus isolates have shown that both isolates are transmitted by Myzus persicae. Only one of the isolates is transmitted by Rhopalosiphum maidis. The R. madis non-transmissible isolate could be transmitted from plants co-infected with the R. maidis transmissible isolate; aphid acquisition factor did not seem to mediate this transmission. The two isolates could be differentiated by enzyme-linked immunosorbent assay experiments, but peptide mapping experiments revealed few differences between the isolates.  相似文献

A disease of chickpea in India, characterised by chlorosis, severe stunting and phloem browning, was shown to be caused by a geminivirus. This virus was transmitted by the leafhopper Orosius orientalis from chickpea to chickpea and several other plant species. A method for purification of this virus was devised and a polyclonal antiserum produced. The majority of the purified particles were geminate. The size of the coat protein was shown to be 32 kD and the nucleic acid was shown to be circular ssDNA of 2900 nucleotides. By immunosorbent electron microscopy this virus was shown to be unrelated to the leafhopper-transmitted geminiviruses known to infect dicotyledons such as beet curly top, bean summer death and tobacco yellow dwarf viruses. On the basis of particle morphology, leafhopper transmission, host range and serology this virus was considered to be a new, hitherto undescribed, geminivirus and was named chickpea chlorotic dwarf virus.  相似文献

Dry, fresh uredospores ofPuccinia recondita that have been killed by infra-red radiation showed no striking ultrastructural differences from dry, fresh, viable uredospores. Numerous spherical and elipsoidal mitochondria with distinct and deep cristae, ribosomes, endoplasmic reticulum and convoluted plasmalemma were well shown by both. When moistened and incubated, killed uredospores lost ultrastructural organization, whereas moistened, viable, fresh uredospores imbibed moisture, became more spherical and germination commenced. A more or less centrally located nucleus was seen with the double membrane invaginated at some points. The advance of the germ tube was initially by enzymic degradation and concluded by mechanical disruption of the degraded pore plug. The mitochondria and the endoplasmic reticulum increased in number and the stored lipid bodies were gradually depleted as germination progressed. Features known as the ‘foamy cytoplasm’ and ‘folded membranes’ were seen in the germinating uredospores only. It was suggested that the ‘foamy cytoplasm’ could be functionally similar to the glyoxisome because of the close association of the former to lipid bodies. The ‘folded membranes’ may be accumulated endoplasmic reticulum being transported to the site of wall formation.  相似文献

Tobacco bushy top disease is caused by a complex of the viruses tobacco bushy top virus (TBTV, a member of the genus Umbravirus) and tobacco vein distorting virus (TVDV, a member of the genus Polerovirus), which acts as a helper virus encapsidating the TBTV genomic RNA. RNA from purified virions is separated as five bands. The two largest (6.0 and 4.2 kb) were shown by Northern blot analysis to be the genomic RNAs of TVDV and TBTV, respectively. A band of about 3 kb was cloned and sequenced and shown to be the RNA of a previously undescribed virus with two open reading frames (ORFs), the second of which is an RNA‐dependent RNA polymerase (RdRp) and is probably expressed by readthrough of the ORF1a stop codon. BLAST and phylogenetic analyses of the RdRp show that it is related to two RNAs previously reported in association with the poleroviruses Beet western yellows virus and Carrot red leaf virus. These three RNAs appear to represent species of a new genus of plant viruses dependent upon a helper polerovirus for their transmission.  相似文献

The uredospores ofM. vogesiaca are not smooth, as stated in literature, but finely verrucose; size and density of the verrucae are rather variable. In contrast, the uredospores ofM. whitei are finely echinulate.

The extent of contamination of wheat seedlots by brome mosaic virus (BMV), and the possible transmission of the virus through seed, were investigated by radial immunodiffusion (RID), enzyme linked immunosorbent assay (ELISA), and enzyme-assisted immunoelectroblotting (IEB) tests. BMV was purified from seed washings, and from plants grown from contaminated seed. All three serological tests proved of value in detecting contaminated seeds, while ELISA and IEB were especially useful in detecting virus in infected plants grown from such seed. Rhopalosiphum padi aphids were shown to increase the incidence of BMV infectLon in seedling batches containing a few seed-infected plants. The implications of these findings for wheat breeding schemes are discussed.  相似文献

Oilseed rape (Brassica napus L. ssp. oleifera) was studied as a potential overwintering host for the sugar-beet yellowing viruses, beet yellows virus (BYV) and beet mild yellowing virus (BMYV), and their principal vector, Myzus persicae. In spring 1982, plants infected with a virus which reacted positively in enzyme-linked immunosorbent assay (ELISA) with BMYV antibody globulin were found in oilseed-rape crops; none of the plants contained virus which reacted with BYV antibody globulin. This virus was subsequently identified as beet western yellows virus (BWYV). No leaf symptoms could be consistently associated with infection of oilseed rape, but the virus was reliably detected by sampling any leaf on an infected oilseed-rape plant. Some isolates from oilseed rape did infect sugar beet in glasshouse tests, but the proportions of inoculated plants which became infected were low. Apparently there is therefore little danger of much direct transmission of BWYV by M. persicae from oilseed rape to sugar beet in spring. BWYV was introduced to and spread within oilseed-rape crops in autumn by M. persicae, and autumn-sown oilseed rape proved to be a potentially important overwintering host for M. persicae. In a survey of 80 autumn-sown crops of oilseed rape in East Anglia, northern England and Scotland in spring 1983, 78 were shown to be extensively infected with BWYV. Experimental plots of oilseed rape with 100% BWYV-infection yielded approximately 13.4% less oil than plots with 18% virus infection, the result of a decrease in both seed yield and oil content.  相似文献

Nicotiana glauca is a naturalised introduced species widely distributed in Australia. A survey of stands in the central zone of its recorded distribution has shown that a type of tobacco mosaic virus which characteristically induces a bright yellow mosaic symptom (TMV-Y) is commonly associated with N. glauca throughout this zone. Surveys of a natural community of N. glauca less than 5 yr old, and in an experimental block, showed that TMV-Y could spread at a rate, and in a pattern, which could not be explained by contact transmission alone, and which indicated that an airborne vector was probably involved in transmission. These results together have led to the conclusion that the common association of TMV-Y with N. glauca is due to spread of the virus by an unknown vector(s). Seed transmission of TMV-Y in N. glauca has not been detected. Molecular hybridisation analysis has shown that all but one of the TMV-Y isolates studied were related to TMV-U2, and that the isolate which was unrelated to U2, was partially related to TMV-U1, TMV-U5, and tomato mosaic virus. Selected isolates which showed a close relationship to TMV-U2, also showed a close relationship to TMV-U5, a distant relationship to TMV-U1, and tomato mosaic, and very little or no relationship with three other tobamoviruses (sunn-hemp mosaic virus, odontoglossum ringspot virus, and cucumber virus 4). These results show that the Australian TMV-Y isolates are similar to the TMV isolated from N. glauca in California.  相似文献

The interaction between beet western yellows luteovirus (BWYV) and the aphid species Brevicoryne brassicae was investigated using virus transmission and serological detection experiments. This species failed to transmit a BWYV isolate from infected to healthy oilseed rape plants, although virus was readily detected by double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) in single B. brassicae adults. When virus-carrying adults were tested by ELISA after different inoculation access periods, the number of virus-positive individuals decreased after 5 days, whereas with the efficient vector Myzus persicae, virus-positive individuals were found even after 10 days. This confirms the inability of B. brassicae to transmit BWYV, even though it may acquire the virus. It is suggested that B. brassicae, as compared with the efficient vector M. persicae, may serve as an experimental model for studying the mechanisms of the luteovirus-vector specificity  相似文献

A disease characterised by severely stunted plants with small dark green leaves was found in groundnut (Arachis hypogaea) in sandy soils in Punjab State, India. The disease occurred in patches in the field and reappeared in the same positions in succeeding groundnut crops. Plants infected early did not produce mature pods. Seeds sown in soil collected from infected fields produced plants with typical disease symptoms. Phaseolus vulgaris cv. Local and Chenopodium quinoa were found to be good diagnostic hosts. The disease was shown to be caused by a rod-shaped virus c. 24 nm in diameter with predominant particle lengths of c. 249 and 184 nm when stained in uranyl acetate. The virus, named Indian peanut clump virus (IPCV), resembled peanut clump virus (PCV) reported from W. Africa in symptomatology on groundnuts, particle morphology and soil-borne nature. However, it is not serologically related to two W. African PCV isolates tested, or to tobacco rattle (PRN and CAM strains) or pea early browning virus (Dutch isolate) in microprecipitin, enzyme linked immunosorbent assay and immunosorbent electron microscopy tests.  相似文献

Malic enzyme was found in both bean rust and cat stem rust uredospores. In bean rust uredospores it was shown to catalyze the formation of pyruvic acid from l-malic acid and to synthesize malic acid from pyruvic acid and CO2. The malic enzyme from bean rust uredospores was specific for NADP and dependent on manganous ions for activity. The specific activity of the bean rust malic enzyme in crude extracts of ungerminated uredospores was approximately 6 times greater than that found in crude extracts obtained from germinated uredospores. The malic enzyme was also found in extracts obtained from healthy and rust-infected bean leaves. The specific activity of the enzyme was approximately 2 to 5 times greater in partially purified extracts obtained from the infected bean tissue at 6 days after inoculation. The specific activity of the malic enzyme in crude extracts obtained from oat stem rust uredospores was 2 times greater than the specific activity of this enzyme in crude extracts obtained from bean rust uredospores. Phosphoenolpyruvate carboxylase activity could not be demonstrated in crude extracts obtained from the ungerminated uredospores of the bean rust fungus.  相似文献

The influence of temperature and light on prepenetration development of single and mixed isolates of Puccinia graminis avenae and Puccinia coronata avenae was studied on 0–2% water agar and on leaves of three oat cultivars and on three non-cultivated species of Avena. Germination of uredospores of P. graminis avenae and P. coronata avenae occurred best at 10–30oC and at 20oC respectively. The optimum temperature for germ-tube growth and for appressorial formation was 20oC for both rusts. An inverse relationship was observed between light intensity and prepenetration development with maximal germination of uredospores, germ-tube growth and appressorial formation occurring in darkness. Under optimum conditions maximum percentage germination and appressorium formation of both rusts was attained within 4 and 12 h after inoculation respectively. The proportion of germinated uredospores of crown rust which gave rise to appressoria was about twice that observed for stem rust. No significant differences were observed in prepenetration development between the single and mixed race inocula of the two rusts. Although germination of uredospores was significantly greater on water agar than on oat leaves, there were no significant differences in prepenetration development of the rusts on the various oat cultivars and species examined. Consequently, the data failed to indicate the presence of resistance mechanisms operating during the prepenetration phase of the infection process on the cultivars and species examined.  相似文献

Horsegram yellow mosaic disease was shown to be caused by a geminivirus; horsegram yellow mosaic virus (HYMV). The virus could not be transmitted by mechanical sap inoculation. Leaf dip and purified virus preparations showed geminate virus particles, measuring 15-18 * 30 nm. An antiserum for HYMV was produced and in enzyme-linked immunosorbent assay (ELISA) and immunosorbent electron microscopy (ISEM) tests HYMV was detected in leaf extracts of fieldinfected bambara groundnut, french bean, groundnut, limabean, mungbean, pigeonpea and soybean showing yellow mosaic symptoms. Bemisia tabaci fed on purified HYMV through a parafilm membrane transmitted the virus to all the hosts listed above but not to Ageratum conyzoides, okra, cassava, cowpea, Croton bonplandianus, Lab-lab purpureus, Malvastrum coromandalianum and tomato. No reaction was obtained in ELISA and ISEM tests between HYMV antibodies and extracts of plants diseased by whitefly-transmitted agents in India such as A. conyzoides yellow mosaic, okra yellow vein mosaic, C. bonplandianus, yellow vein mosaic, M. coromandalianum yellow vein mosaic, tomato leaf curl and cassava mosaic. HYMV was also not found to be related serologically to bean golden mosaic, virus.  相似文献

Several Agropyron species were tested for new sources of resistance to barley yellow dwarf virus (Bydv ) and wheat streak mosaic virus (WSMV). With BYDV strain PAV, 11 of the 17 Agropyron species showed no virus transmission when plants were given access feed by viruliferous Rhopalosiphum padi. Similar trials with BYDV strain RMV (vectored by R. maidis) indicated that all plants, except susceptible control plants, remained virus free. Virus status was confirmed by enzyme-linked immunosorbent assays. When plants were mechanically inoculated with WSMV, 11 Agropyron species failed to express symptoms, while five other species showed a segregating response or had some accessions segregating and some resistant. Test results suggest that resistance to BYDV and WSMV in Agropyron species does not appear to be correlated with any specific genome of Agropyron species although most of the Agropyron species containing S genome were resistant to BYDV and WSMV.  相似文献

Ectocarpus-like marine brown algae are frequently parasitized by polyhedric DNA viruses. Infected hosts have been studied in unialgal and axenic cultures, and the present state of knowledge is summarized in regard to stage-specific virus expression, discharge and survival time of virus particles, infection mechanism, association with host's nuclear genome, passage of the virus genome through mitosis and meiosis of the host, suppression of symptoms and spontaneous recovery of infected plants, host specificity and intergeneric transmission, vitality of infected plants, pandemic occurrence of virus infections, molecular data on Ectocarpus and Feldmannia viruses, and algal DNA-viruses as potential vectors for gene transfer. A scheme for the nomenclature of brown algal viruses is proposed.  相似文献

Verticillium psalliotae isolates Taiw and Thai C are effective parasites of the soybean rust fungus. Cell-free culture filtrates of these fungi, prepared after growth on autoclaved uredospores, contained β-1,3-glucanase, chitinase and protease activities and caused degradations, when rust spores were treated with them for 24 or 72 h. During these lytic processes carbohydrates, amino compounds and N-acetylhexosamine were released. The carbohydrate fraction was composed of mannitol, arabitol, trehalose, glucose and unidentified substances showing low Rf-values during thin layer chromatography. The amino compounds consisted of 10 amino acids (leucine and/or isoleucine, phenylalanine, glutamic acid, valine, arginine, asparagine, glutamine, serine, aspartic acid, histidine) and 5—7 substances which could not be identified. Verticillium lecanii isolate Konz is a weak parasite of soybean rust. During growth on uredospores the fungus produced culture filtrates without chitinase activity and with low total activities of β-1,3-glucanase and protease. Compared with V. psalliotae, culture filtrates of V. lecanii exerted lower lytic activities on soybean rust uredospores. The results are consistent with the aspect that the rapid growth of V. psalliotae on soybean rust fungus is primarily based on the secretion of lytic enzymes which make nutrients available to the mycoparasite.  相似文献

Two isolates of the mycoparasite Verticillium psalliotae grew rapidly in liquid cultures on autoclaved uredospores of the soybean rust fungus (Phakopsora pachyrhizi Syd.) as sole carbon source and secreted β-l,3-glucanase, chitinase, and protease activities into the medium. One isolate of Verticillium lecanii grew slowly, failed to produce measurable chitinase activity and secreted lower specific activities of β-l,3-glucanase and protease, compared with V. psalliotae. The tested isolates of V. psalliotae and V. lecanii produced comparable levels of lipolytic activity. Amylolytic activity was secreted by V. lecanii but not by V. psalliotae. The isolates of V. psalliotae and V. lecanii used in our experiments differed clearly in protein and protease pattern, determined by electrophoresis on polyacrylamide gels. The results indicate that the rapid growth of V. psalliotae on autoclaved uredospores in liquid culture and on uredosori is probably based primarily on nutrients made available to the mycoparasite by activities of β-1,3-glucanases, chitinases and proteases.  相似文献

In 1983, cereal plants showing symptoms of barley yellow dwarf virus (BYDV), collected from 15 localities in Sweden, were tested for BYDV using enzyme-linked immunosorbent assay (ELISA). Antisera against two Swedish isolates of BYDV were used, a mild isolate (27/77) transmitted specifically by Sitobion avenae and a severe one (39/78) transmitted mainly by Rhopalosiphum padi. No virus was detected in 57 of 607 plants of oats and barley tested. Of the 550 plants in which virus was detected, 366 were infected with viruses similar to isolate 27/77, 116 with viruses similar to 39/78 and the remaining 68 reacted strongly with both antisera. When tested, the latter isolates were shown to be mixtures. Thirty-nine selected samples were also tested with antisera against the USA isolates RPV, RMV, MAV and PAV, and for transmission by S. avenae and R. padi. Twenty-six of these samples were transmitted specifically by S. avenae, one was transmitted only by R. padi and the remaining 12 samples were shown to be infected with a mixture of an S. avenae-specific isolate and one transmitted mainly by R. padi. Antisera against PAV and MAV each detected all isolates tested and the results were very similar to those with the antisera to the 39/78 and 27/77 isolates, respectively. None of the field isolates reacted with antisera against RMV or RPV. It was concluded that 1983 was an epidemic year for BYDV in Sweden and that isolates specifically transmitted by S. avenae predominated. Symptoms of infection by these isolates on oat plants ranged from mild to severe.  相似文献

The development of infection structures by the directly infecting soybean rust fungus of different artificial membranes was followed by light and scanning electron microscopy. On water agar uredospores developed germ tubes without appressoria. On dialysis membranes more than 80% of the uredospores formed appressoria. With low frequencies (1–7%) also primary hyphae and/or penetration hyphae were present. When cellulose nitrate membrane filters with pore diameters ≤ 0.2 μm were used, uredospores germinated but showed a strongly reduced appressoria formation. Membranes with pores ≥ 0.1 μm allowed a development of infection structures similar to that on dialysis membranes. In experiments with paraffin oil incorporated into collodion membranes more than 90% of the uredospores formed appressoria, about 50% of the appressoria developed hyphae. Ungerminated spores and germ tubes always contained 2 nuclei. In fully developed appressoria 4 nuclei were present. Compared with stomata entering rust fungi appressoria formation by Phakopsora pachyrhizi occurred more frequently and seemed to be less dependent on specific stimuli. Moreover, in most cases only few of the appressoria formed penetration or primary hyphae. The induction of these structures seemed to be dependent on further unknown stimuli.  相似文献

Walls of uredospores, infection structures, intercellular hyphae and haustoria of the soybean rust fungus (Phakopsora pachyrhizi) were studied by electron microscopy using gold-labeled wheat germ lectin (WGL) and Concanavalin A (ConA) as cytochemical probes. Receptors for WGL (probably chitin) were detected in all fungal walls included in this study. WGL-binding occurred throughout the entire walls (uredospores, appressorial cone, penetration hyphae, haustorial mother cells) or only to the inner wall layers (germ tubes, appressoria, intercellular hyphae).  相似文献
