Summary Siter chromatid exchanges (SCE) were analyzed in the cord and postnatal blood of controlled groups of low and high birth weight infants to detect possible associations between abnormal birth weight and SCE frequency. Structural chromosome aberration rates had previously been evaluated for all infants, and possible correlations between aberration and SCE rates were sought.No correlation was found between neonatal or postnatal SCE frequency and birthweight, nor was there evidence of association of chromosome aberration rates with SCE frequency. In all groups of infants, however, mean postnatal SCE frequencies were significantly lower than mean neonatal SCE frequencies.  相似文献

Sister chromatid exchange (SCE), percentage of first, second, third mitoses, blastic transformation index and mitotic index in patients with myelodysplastic syndrome (MDS) (3 with refractory anaemia, 2 with refractory anaemia with sideroblasts, 1 with refractory anaemia with excess of blasts, 4 with refractory anaemia with excess of blasts in transformation) and in 15 healthy volunteers were estimated. Three types of lymphocytes cultures were set up: first with phytohemaglutinin (PHA), second with PHA and bromodeoxyuridine (BRdU), third with BRdU. In healthy persons the SCE frequency was negatively correlated to proliferating rate index, but in MDS such correlation was not found. The lymphocytes cell cycle duration based on percentage of mitoses was longer in MDS patients than in controls. The results of our studies show the disturbances of lymphocytes during cell cycle division resulting in higher SCE frequency and lower proliferating rate compared to controls.  相似文献

Sister chromatid exchange (SCE) frequencies were studied in differentially stained chromosomes from lymphocytes of 17 patients with viral disease. The mean SCE score for the patients was 8.7 +/- 2.9 standard deviations. SCE scores were significantly elevated in the patients compared with the controls (p less than 0.01); however, variability in SCE means was observed in the patients. SCE elevations were also present in long term cultured Epstein Barr virus positive human B lymphocytes.  相似文献

Aberrant meiotic chromosome configurations in an experimental population of rye lines are known to result from spontaneous U-type exchanges during meiotic prophase. Both sister chromatid and non-sister chromatid exchanges occur and this study is concerned with the relative frequencies of sister and non-sister exchanges. The anaphase I observations reveal a marked excess of non-sister U-type exchange configurations and it is argued that this reflects an original excess of prophase non-sister U-type exchanges. This conclusion is discussed with special reference to the origin of meiotic U-type exchanges and their relation to regular crossover exchanges.  相似文献

Sister chromatid exchange (SCE) studies have been performed in a variety of animals, both in vitro and in vivo, as well as in plants. To date, no such studies have been performed in any member of the gerbil sub-family (Gerbillinae). A new sister chromatid differential staining method was used with mice in vivo and has now been applied to the Mongolian gerbil, Meriones unguiculatus. With some modifications, this in vivo method was found to be highly reproducible in gerbils, and had consistently produced many metaphase cells with clear sister chromatid differentiation. The method involves subcutaneous implantation of a 50 mg slow-release BrdU pellet, the fluorochrome Hoechst 33258, phosphate buffering at pH 6.8, and incandescent light exposure.  相似文献

The rate of sister chromatid exchange induced by several anti-herpes agents was measured to assess their potential mutagenicity. The agents--5-iodo-deoxyuridine (IDU), 5-trifluoromethyl-deoxyuridine (TFT), and [E]-5-(2-bromovinyl)-deoxyuridine (BVDU)--were incubated at various concentrations with human lymphocytes and fibroblasts, and that rate of sister chromatid exchanges was measured. In lymphocytes and fibroblasts BVDU and IDU did not induce exchange except at concentrations of 50 mg/l, while TFT increased the rate of exchange at a concentration of 0.5 mg/l. The rate of sister chromatid exchange is a sensitive index of chromosomal damage, and these findings provide information on the safety of some of the antiherpes agents tested. TFT increased the rate of exchange at a concentration that coincides with its minimal antiviral concentration, but BVDU did not induce exchange at therapeutic concentrations.  相似文献

The structures of rDNA spacers from several species have been characterized and virtually all have internally repeated sequences. Different numbers of these internal repeats are responsible for most spacer length variation. Because unequal recombination between these internal repeats will cause new length variation, while unequal exchange between rDNA copies will homogenize the variants, we modeled the interaction of these two processes. Two models were used to simulate both types of unequal exchange at the sister chromatid level. Both models indicate that a narrow range of relative recombination frequencies is required to produce levels of variability comparable to those published. One model puts a lower limit on the number of internal repeats, and the other puts both a lower and upper limit on the number of repeats. The model with both maximum and minimum constraints produces a distribution closer to actual spacer distributions. These results imply that small changes in recombination rates can generate the differences in numbers of length variants observed in different species.  相似文献

Summary Sister chromatid exchange (SCE) has been studied in the fibroblasts of five Bloom's syndrome patients, one heterozygote, and two normal individuals. The high frequency of SCE already known in the lymphocytes of Bloom's syndrome was also found in the fibroblasts of all five patients. However, populations with low and high frequency of SCE were not found. In addition, chromosome aberrations appeared with a lower frequency.The cell cycle duration in the Bloom's fibroblasts appeared to be similar to that in the normal cell line, and the difference in the growth pattern appeared to be due to the variation in the mitotic index. The cell cycle lasted about 24 h in at least four of the Bloom's lines studied during the present experiments.  相似文献

Sister chromatid exchange (SCE) was measured in peripheral lymphocytes of 90 workers from 14 hospital pathology departments in Israel who were occupationally exposed to formaldehyde (FA) and of 52 unexposed workers from the administrative section of the same hospitals. The mean exposure period to FA was 15.4 years (range 1-39). The results of SCEs are expressed in two variables: (a) mean number of SCEs per chromosome and (b) proportion of high frequency cells (cells with more than eight SCEs). A high correlation was found between these two variables. The adjusted means of both SCEs variables were significantly higher among the exposed compared with that of the unexposed group (P<0.01). Adjustment was made for age, sex, smoking habits, education workers and origin. Evaluation of the influence of years of exposure on the frequency of SCEs showed that the two variables of SCEs were higher among those who were exposed to FA for 15 or more than among those with less than 15 years of exposure. Concerning levels of exposure, both variables of SCEs were the same in the low and in the high levels of exposure sub-groups. However, among the smokers, both variables of SCEs were higher in the high exposure sub-group than in the low exposure sub-group.Our finding of a significant increase of SCEs frequency in peripheral lymphocytes in pathology staff indicates a potential cytogenetic hazard due to FA exposure. We conclude that our data indicate that FA is mutagenic to humans.  相似文献

Summary Sister chromatid exchange (SCE) rates were determined in human peripheral blood B and T lymphocyte populations highly purified by immunologic methods. The purified populations were supplemented with -irradiated unseparated autologous mononuclear cells to restore helper-functions and stimulated with pokeweed mitogen (PWM) and phytohemagglutinin (PHA), respectively. Measured at the different peaks of proliferation after identical bromodeoxyuridine (BrdU) incubation times, T lymphocytes showed significantly higher SCE frequencies than B lymphocytes. In both populations, different proliferation kinetics and a different minimal BrdU concentration for sister chromatid differentiation (SCD) were observed.  相似文献

Summary Sister chromatid exchange (SCE) frequencies were studied in differentially stained lymphocytes from 47 patients with malignant lymphoma. Thirteen patients were untreated when studied. The mean SCE frequency [±standard error (SE)] for these patients was 12.7±0.9 per mitosis. The mean score for 40 controls was 6.1±0.3. SCE mean scores were significantly higher in the untreated patients than in the controls (P<0.001). Seven patients were treated with radiotherapy alone. They demonstrated a mean SCE frequency (8.8±0.8) significantly lower (P<0.01) than that found in untreated patients. Eleven patients received cyclophosphamide within 4 weeks prior to study. They demonstrated a mean SCE frequency (14.3±1.3) significantly higher (P<0.05) than that found in patients who had received regimens that did not contain cyclophosphamide in the prior 4 weeks (11.1±1.3) or who had been off drugs for at least 8 weeks (10.1±0.8). Our data suggest that untreated patients with malignant lymphoma have elevated SCE frequencies, which may be further increased by certain chemotherapeutic agents.  相似文献

Summary Human lymphocytes from 18 healthy adults were cultured in minimal essential medium without folic acid. The addition of 4mM cytidine or 1 mM guanosine to cultures 24h prior to harvest produced a statistically significant increase in hot point 3p14 breaks. An excess of 3mM adenosine or thymidine had no such effect on the hot point. The mechanism of the effects of nucleosides on hot points was discussed.  相似文献

The sister chromatid exchange (SCE) frequency and cell-cycle specific metaphase patterns were investigated in PHA-stimulated lymphocytes in vitro after in vivo exposure of 15 patients to diagnostic ultrasound. No significant differences were found in SCE rates before and after ultrasound exposure. The evaluation of cell-cycle specific metaphase patterns, differentiating cells which passed one, two, or more cell cycles, gave no evidence for a cell-growth inhibitory effect. Since SCE is regarded as a highly sensitive method for the detection of mutagenic effects, our data confirm previous reports on the genetic safety of diagnostic ultrasound.  相似文献
