Secretory granules of chromaffin cells from the adrenal medulla store catecholamines and a variety of peptides that are secreted in the extracellular medium during exocytosis. Among these fragments, several natural peptides displaying antimicrobial activities at the micromolar range have been isolated and characterized. We have shown that these peptides, derived from the natural processing of chromogranins (CGs), proenkephalin-A (PEA) and free ubiquitin (Ub), are released into the circulation and display antibacterial and antifungal activities. In this review we focus on three naturally secreted antimicrobial peptides corresponding to CGA1–76 (vasostatin-I), the bisphosphorylated form of PEA209–237 (enkelytin) and Ub. In addition, the antimicrobial properties of the synthetic active domains of vasostatin-I (CGA47–66 or chromofungin) and Ub (Ub65–76 or ubifungin) are reported.  相似文献

In landfills, deposited waste material is usually faced with strictly anoxic conditions. This means that the design of new biodegradable polymers must take into consideration that degradation should be possible especially in the absence of molecular oxygen. Poly-beta-hydroxybutyrate is depolymerized by the anaerobic fermenting bacterium Ilyobacter delafieldii through an extracellular hydrolase. Monomers are degraded inside the cells through classical beta-oxidation. Polyalkanoates containing odd-numbered or branched-chain acid monomers should he degraded in an analogous manner; in most cases the final mineralization of these residues requires special pathways. A comparison of the chemistry of natural polymer biodegradation leads to the conclusion that synthetic biodegradable polymers should be designed in the future to contain linkages which can be cleaved by extracellular hydrolytic enzymes. Recent findings on aerobic and anaerobic bacterial degradation of synthetic polyethers suggest that natural evolution of new depolymerizing enzymes, perhaps from existing hydrolases, could be possible in a reasonable amount of time, provided that the monomers are likely energy sources for a broad variety of microbes.  相似文献

Evolution is based on natural selection of existing biological phenotypic traits. Natural selection can only eliminate traits. It cannot create new ones, requiring a theory to explain the origin of new genetic information. The theory of pangenesis was a major attempt to explain the source of new genetic information required to produce phenotypic variety. This theory, advocated by Darwin as the main source of genetic variety, has now been empirically disproved. It is currently a theory mainly of interest to science historians.  相似文献

Six kinds of glycolipid of monogalactosyl and digalactosyl diglyceride were separated and purified from the wheat and the natural latex of chilling-tolerant and chilling-sensitive variety by two kind solvent systems and twice one-dimensional thin-layer chromatography. Besides, the compositions of fatty acid of hydrophobic side chain of six kinds of glycolipid were compared. The indices of unsaturation of fatty acid of hydrophobic side chains in the glycolipids of wheat flour, in the glycolipids of natural latex of chilling-tolerant, variety and in the digalactosyl diglycerides, are much bigger than those, respectively, in the glycolipids of natural latexes, in the glycolipids of chilling-sensitive variety and in the monogalactosyl diglycerides.  相似文献

One new species of Aspidistra, i.e. A. nutans, and one new variety, i.e. A. tillichiana var. latifolia, recently discovered in Vietnam are described and illustrated. In addition, the rank of species is proposed for A. formosa widely distributed in northern Vietnam but originally described as a variety of A. fungilliformis. Aspidistra subrotata is recorded as a new species for the flora of Laos. Data on ecology, phenology, natural variation, distribution and expected conservation status are provided for these plants.  相似文献

Myxotrichum ochraceum var.frigidum, a new variety ofMyxotrichum, is described and illustrated from the material from Japanese soil. It is morphologically characterized by dull green to black ascomata with a yellow-orange centrum, peridium consisting of a fragile hyphal network, peridial hyphae with internal coiled branches and short spine-like appendages, pale yellow fusiform striate ascospores, and the absence of an anamorph. The new variety primarily differs from the type variety in the absence of elongate appendages. A key to all accepted species of the genus is revised.  相似文献

Numerous studies using a variety of cell/acceptor combinations have demonstrated differences in cholesterol efflux among cells. These studies also show that different acceptors, ranging from simple molecules like cyclodextrins to serum, stimulate efflux through a variety of mechanisms. By combining early observations with data derived from recent studies, it is now possible to formulate a model for cell cholesterol efflux which proposes that an array of different mechanisms, including aqueous diffusion, lipid-free apolipoprotein membrane microsolubilization, and SR-BI-mediated cholesterol exchange contribute to cholesterol flux. In this model the relative importance of each mechanism would be determined both by the cell type and the nature of the extracellular cholesterol acceptor.  相似文献

Antibodies, proteins produced in animals that bind with high specificity and affinity to a seemingly limitless variety of biomolecules, have an essential role in clinical medicine and basic biological research. Methods to produce antibody fragments on the surface of bacterial viruses were surveyed for their ability to replace animal-dependent antibodies. The use of filamentous phage to display antibody fragments derived from semisynthetic antibody genes was found to produce proteins that bind to antigens with a variety, specificity, and affinity similar to those produced in animal systems.  相似文献

The discovery of drugs used to combat infectious diseases is in the process of constant change to address the ever-worsening problem of antibiotic resistance in pathogens and a lack of recent success in discovering new antibacterial drugs. In the past 2 decades, research in both academia and industry has made use of molecular biology, genetics, and comparative genomics, which has led to the development of key technologies for the discovery of novel antibacterial agents. Genome-scale efforts have led to the identification of numerous molecular targets. Chemical diversity from synthetic combinatorial libraries and natural products is being used to screen for new molecules. A wide variety of approaches are being used in the search for novel antibiotics, and these can be categorized as being either biochemically focused or cell based. The over-riding goal of all methods in use today is to discover new chemical matter with novel mechanisms of action against drug-resistant pathogens.  相似文献

The discovery and development of new drugs targeting voltage-gated ion channels are important for treating a variety of medical conditions and diseases. Ion channels are molecular nanostructures expressed ubiquitously throughout the whole body, and are involved in many basic physiological processes. Over the years, natural products have proven useful in the pharmacological assessment of ion channel structure and function, while also contributing to the identification of lead molecules for drug development. Essential oils are complex chemical mixtures isolated from plants which may possess a large spectrum of biological activities most of them of clinical interest. Among their bioactive constituents, terpenes are small to medium-sized components and belong to different chemical groups. Various reports have drawn our attention to the fact that terpenes are novel compounds targeting voltage-gated ion channels. The purpose of this review is to provide a focused discussion on the molecular interaction between monoterpenes and phenylpropenes with voltage-gated ion channels in different biological scenarios.  相似文献

Inhibition of Hsp90: a new strategy for inhibiting protein kinases   总被引:10,自引:0,他引:10  
The 90-kDa heat shock protein (Hsp90) is a ubiquitous, evolutionarily highly conserved, molecular chaperone in the eukaryotic cytosol. Hsp90, together with a number of other chaperones, promotes the conformational maturation of a large variety of protein kinases. Inhibition of Hsp90 function results in the collapse of the metastable conformation of most of these kinases and leads to their proteolytic elimination by the proteasome. Numerous natural and synthetic Hsp90 inhibitors have been developed in recent years. Some of these inhibitors are also involved in sensitizing tumor cells to pro-apoptotic insults, hence serve as anti-cancer drugs. Here we review these novel protein kinase inhibitors and their emerging role in various cellular processes, apart from their inhibition of Hsp90 protein function. We focus not only on Hsp90-tumor progression, but also on cytoarchitecture, as the higher levels of cellular organization need constant remodeling, where the role of Hsp90 requires investigation. Our last major aspect deals with protein oxidation, since several Hsp90 inhibitors exert pro-oxidant effects.  相似文献

Tryptophan synthase is a versatile enzyme that catalyzes a wide variety of pyridoxal phosphate dependent reactions that are also catalyzed in model systems. These include beta-replacement, beta-elimination, racemization, and transamination reactions. We now show that the apo-alpha 2 beta 2 complex of tryptophan synthase will bind two unnatural substrates, pyridoxamine phosphate and indole-3-pyruvic acid, and will convert them by a single-turnover, half-transamination reaction to pyridoxal phosphate and L-tryptophan, the natural coenzyme and a natural product, respectively. This enzyme-catalyzed reaction is more rapid and more stereospecific than an analogous model reaction. The pro-S 4'-methylene proton of pyridoxamine phosphate is removed during the reaction, and the product is primarily L-tryptophan. We conclude that pyridoxal phosphate enzymes may be able to catalyze some unnatural reactions involving bound reactants and bound coenzyme since the coenzyme itself has the intrinsic ability to promote a variety of reactions.  相似文献

The author investigated the presence of various carotenoids in Salmo gairdneri Rich. and Salmo trutta morpha fario L. from the trout fish farm (artificial food) and from the river as the natural conditions (natural food).The findings of these investigations indicate that the trout bred in natural conditions are richer in carotenoids, provitamins of vitamin A, than are trout from the trout fish farm. In all probability the variety of food available in natural conditions provides better facilities for the accumulation of carotenoids in the body of trout in their natural habitat.  相似文献

The occurrence of agarolytic microorganisms in a variety of natural environments was investigated using enrichment culture techniques. From samples taken from the IJsselmeer, and adjacent areas in the central part of The Netherlands a new agarolytic flexibacterium was isolated. Enrichments inoculated with samples from other sources were unsuccessful. The yellow-pigmented organism exhibits gliding motility, but spreading was only occasionally detected. Since neither fruiting bodies nor microcysts were observed, the organism was placed into the genus Cytophaga and designated as Cytophaga flevensis sp. nov. The occurrence of this species is probably restricted to the IJsselmeer, a fresh-water lake that was in open contact with the North Sea until 1932, and some other lakes which are in direct contact with the IJsselmeer. The organism utilizes only carbohydrates as a carbon and energy source for growth and is strictly aerobic. When the organism is grown on agar, on its components or on melibiose, an extracellular agarase is produced. This enzyme is highly specific for polysaccharides containing neoagarobiose units and hydrolyzes neoagaro-oligosaccharides with a degree of polymerization of more than four.  相似文献
