Three mechanically transmissible viruses were isolated from a diseased Solanum jasminoides plant obtained from India. One is a strain of potato virus Y , which in some potato varieties produces symptoms resembling those caused by potato virus C , but unlike potato virus C it is readily transmitted by Myzus persicae. The second, named tobacco wilt virus, is also transmitted by M. persicae but much less readily, whereas the third, named datura necrosis virus, is not. All three have a wide host range, but neither tobacco wilt nor datura necrosis viruses infects potato plants. All three have long flexuous particles and similar general properties.
Simultaneous infection with datura necrosis virus usually decreases the concentration reached by potato virus Y in tobacco plants but not in Nicotiana glutinosa.  相似文献

Potato virus Y was obtained from field crops of potatoes in many strains which differed widely in virulence and caused diseases in the variety Majestic ranging from severe leaf-drop streak to mild mosaic. The symptoms caused by these strains in seven potato varieties and tobacco are described and compared with those caused by the serologically related potato virus C. No changes were noted in the behaviour of any of the strains over three years, during which they were transmitted to many different plants.
Potato virus C was not transmitted by Myzus persicae , the most efficient vector of other strains of virus Y. Nor was virus C transmitted by eleven other species of aphides, eight of which transmitted virus Y. The efficiency with which different species acted as vectors of virus Y varied greatly, and it is suggested that in some species only occasional individuals can transmit.
Possible mechanisms for the evolution of viruses C and Y are indicated, and the effects of changes in virus, vector, and host on the prevalence of insect-transmitted viruses are discussed.  相似文献

Experiments at Invergowrie, south-east Perthshire, showed that the extent of spread of potato leaf-roll and Y viruses varied from year to year and that virus Y consistently spread more than leaf roll. Most spread of Virus Y occurred before the end of June and of leaf-roll virus before the end of July. Both viruses spread slightly more in late- than in early-planted crops. When plants with leaf roll and already colonized by Myzus persicae were placed in a healthy crop of Majestic potatoes at intervals during the season, the amount of virus spread decreased rapidly with increasing age of the crop. Spread of leaf roll occurred in all of twenty-five 'seed' crops in different districts of eastern Scotland in 1955 but in only twenty out of thirty-six similar crops in 1956. Annual and regional differences in virus spread appear to reflect differences in the time at which migrant aphids reach potato crops in early summer and the rate at which infestation builds up in the crops.  相似文献

Potato virus B , and some other viruses with reactions in potato varieties different from any previously described, are strains of virus X . All produce intracellular inclusions which vary with different hosts and virus strains. Except with virus B, the inclusions are larger and more frequent in potato than in tobacco or tomato. All give systemic infection when inoculated to tobacco, tomato and potato varieties in which they are carried or cause mosaic symptoms; some give systemic infection when inoculated to varieties in which they cause top-necrosis, whereas others give only local lesions.
Potato virus C is a strain of virus Y: in tobacco and a few potato varieties both produce similar symptoms, but in those varieties in which Y causes leaf-drop streak, C causes top-necrosis. C causes systemic infection when inoculated to tobacco and to potato varieties in which it causes mosaic symptoms, but not when inoculated to potato varieties in which it causes top-necrosis. Virus C was not transmitted by M . persicae. Viruses C and Y produce a few small intracellular inclusions in potato and tobacco.
Virus A is not related to Y or X : no inclusions were found in plants infected with A alone.  相似文献

A strain of cucumber mosaic virus isolated from a spinach plant in 1946 was readily transmitted by Myzus persicae until 1955 when it lost this property, although it was still being propagated in conditions in which other strains remained transmissible. M. circumflexus also transmitted other strains but not this one. It was transmitted as readily as other strains by Aphis gossypii and Myzus ascalonicus. M. ascalonicus transmitted less frequently than Aphis gossypii. Transmission of the spinach strain by other aphids did not make it transmissible by Myzus persicae ; nor did propagation in different plant species or several passages through spinach. In 1955 the spinach strain was occasionally transmitted by M. persicae , but the cultures isolated in this way were no more readily transmissible by the aphid than was the bulk culture maintained by manual inoculation of sap, and after a few weeks all cultures ceased to be transmitted by M. persicae.  相似文献

Tobacco viruses transmitted by green peach aphids, Myzus persicae (Sulzer) (Hemiptera: Aphididae), cause severe disease in flue‐cured tobacco, Nicotiana tabacum L. (Solanaceae), in China and throughout the world. Field experiments were conducted in 2016 and 2017 in Longyan City, Fujian Province, China, to determine whether M. persicae and aphid‐transmitted virus diseases are affected by intercropping of oilseed rape, Brassica napus L. (Brassicaceae), in tobacco fields. The results showed that, compared with those in monocultured fields, the densities of M. persicae and winged aphids in intercropped fields significantly decreased in both 2016 and 2017. In particular, the appearance of winged aphids was delayed by ca. 7 days. Moreover, the densities of Aphidius gifuensis Ashmead (Hymenoptera: Aphidiidae), a parasitoid of the aphid, significantly increased in 2016 and 2017. Accordingly, the incidence rates of aphid‐transmitted virus diseases (those caused by the cucumber mosaic virus, potato virus Y, and tobacco etch virus) significantly decreased in the intercropped fields in 2016 and 2017. Tobacco yields and monetary value significantly increased in 2016 (by 10–25 and 14–29%, respectively) and 2017 (by 17–22 and 22–34%, respectively). Consequently, our results suggest that intercropping oilseed rape in tobacco fields is a good approach to regulating and controlling aphids and tobacco mosaic viruses, for example potyvirus, and this intercropping can help control aphid‐transmitted virus diseases in tobacco.  相似文献

The influence of viral disease symptoms on the behaviour of virus vectors has implications for disease epidemiology. Here we show that previously reported preferential colonization of potatoes infected by potato leafroll virus (genus Polerovirus) (luteovirus) (PLRV) by alatae of Myzus persicae, the principal aphid vector of PLRV, is influenced by volatile emissions from PLRV-infected plants. First, in our bioassays both differential immigration and emigration were involved in preferential colonization by aphids of PLRV-infected plants. Second, M. persicae apterae aggregated preferentially, on screening above leaflets of PLRV-infected potatoes as compared with leaflets from uninfected plants, or from plants infected with potato virus X (PVX) or potato virus Y (PVY). Third, the aphids aggregated preferentially on screening over leaflet models treated with volatiles collected from PLRV-infected plants as compared with those collected from uninfected plants. The specific cues eliciting the aphid responses were not determined, but differences between headspace volatiles of infected and uninfected plants suggest possible ones.  相似文献

Enzyme-linked immunosorbent assay (ELISA) was adapted for the efficient detection and assay of potato leafroll virus (PLRV) in aphids. Best results were obtained when aphids were extracted in 0.05 M phosphate buffer, pH 7.0, and the extracts incubated at 37 °C for 1 h before starting the assay. Using batches of 20 green peach aphids (Myzus persicae), about 0.01 ng PLRV/aphid could be detected. The virus could also be detected in single aphids allowed a 1-day acquisition access period on infected potato leaves. The PLRV content of aphids depended on the age of potato source-plants and the position of source leaves on them. It increased with increase in acquisition access period up to 7 days but differed considerably between individual aphids. A maximum of 7 ng PLRV/aphid was recorded but aphids more usually accumulated about 0.2 ng PLRV per day. When aphids were allowed acquisition access periods of 1–3 days, and then caged singly on Physalis floridana seedlings for 3 days, the PLRV content of each aphid, measured subsequently, was not strongly correlated with the infection of P. floridana. The concentration of PLRV in leaf extracts differed only slightly when potato plants were kept at 15, 20, 25 or 30 °C for 1 or 2 wk, but the virus content of aphids kept on leaves at the different temperatures decreased with increase of temperature. PLRV was transmitted readily to P. floridana at all temperatures, but by a slightly smaller proportion of aphids, and after a longer latent period, at 15 °C than at 30 °C. The PLRV content of M. persicae fed on infected potato leaves decreased with increasing time after transfer to turnip (immune to PLRV). The decrease occurred in two phases, the first rapid and the second very slow. In the first phase the decrease was faster, briefer and greater at 25 and 30 °C than at 15 and 20 °C. No evidence was obtained that PLRV multiplies in M. persicae. These results are compatible with a model in which much of the PLRV in aphids during the second phase is in the haemocoele, and transmission is mainly limited by the rate of passage of virus particles from haemolymph to saliva. The potato aphid, Macrosiphum euphorbiae, transmitted PLRV much less efficiently than M. persicae. Its inefficiency as a vector could not be ascribed to failure to acquire or retain PLRV, or to the degradation of virus particles in the aphid. Probably only few PLRV particles pass from the haemolymph to saliva in this species. The virus content of M. euphorbiae collected from PLRV-infected potato plants in the field increased from early June to early July, and then decreased. PLRV was detected both in spring migrants collected from the plants and in summer migrants caught in yellow water-traps. PLRV was also detected in M. persicae collected from infected plants in July and August, and in trapped summer migrants, but their PLRV content was less than that of M. euphorbiae, and in some instances was too small for unequivocal detection.  相似文献

Potato virus Y is transmitted to potato in a nonpersistent manner by many aphid species, some of which do not colonize this crop. The behavior of bird cherry-oat aphid, Rhopalosiphum padi (L.) on potato, Solanum tuberosum L., a plant species that is not colonized by this aphid, was described and compared with that of the potato-colonizing green peach aphid, Myzuspersicae (Sulzer). A higher proportion of winged morph of R. padi than M. persicae left the plant, but aphids that stayed in contact with the plant took the same mean time to initiate the first probe and it lasted the same mean time compared with M. persicae. Electronic penetration graph technique was used to study the probing behavior of the aphids during Potato virus Y (family Potyviridae, genus Potyvirus, PVY) transmission tests. Transmission rate decreased from 29 to 8% when the acquisition time increased from 5 min of continuous probing to 1 h with M. persicae, but it remained low (2 and 1%) with R. padi. Most of the difference in transmission rate between acquisition time with M. persicae and between aphid species was related to the change in the time and behavior taking place between the last cell puncture of the acquisition phase to the first cell puncture of the inoculation phase. Results presented here clearly demonstrated the importance of host plant selection and probing behavior in the transmission of nonpersistent plant viruses. They also stress the need to consider the behavior of the aphid in the design of laboratory tests of virus vector efficacy.  相似文献

Some barley yellow-dwarf (BYD) viruses isolated from cereal crops in Great Britain were transmitted by Rhopalosiphum padi , L. and others were not. Sitobion fragariae (Walker), S. avenae (Fabricius), and Metopolophium dirhodum (Walker) all transmitted viruses of both types, but they usually transmitted those of which Rhopalosiphum was a vector less readily than did R. padi. The transmissibility of a virus by a given aphid species was not affected by transmission with another, less efficient, vector species. Neomyzus circumflexus (Buckt.) and Rhopalosiphum maidis (Fitch) transmitted the few viruses with which they were tested.
A few R. padi acquired virus from infected leaves during 30 min. feeding and inoculated healthy seedlings during 15 min. feeding, but the minimum total time taken to acquire and transmit was 10 hr. and 32 hr. were needed for about half the aphids that were able to acquire and transmit virus to do so. This may indicate the existence of a short latent period of the virus in the vector, although the evidence is not conclusive. The times spent on infected plants influenced the results more than those spent on healthy ones; many transmissions occurred with short feeding times on healthy plants so long as the time spent on infected leaves was long, but the reverse was not true. Nymphs of R. padi that moulted after they left infected plants on which they fed long enough to become infective, infected slightly fewer plants than adults fed for the same times.  相似文献

The numbers of the potato aphids, Macrosiphum euphorbiae and Myzus persicae, were counted on potatoes undersown and not undersown with perennial ryegrass in field experiments during 1980 and 1981. The numbers of aphids on potatoes undersown with grass were found generally to be smaller than the aphid numbers on potatoes not undersown with grass. Tuber yields were not significantly reduced by the undersowing of potatoes. The agronomic feasibility of undersowing potatoes for aphid control is discussed.  相似文献

Removing virus-infected plants from plots of Majestic potatoes at Rothamsted on 2 July 1947 did not reduce the spread of leaf roll but reduced rugose mosaic (potato virus Y) to about one-fifth of that in plots rogued on 21 July or left unrogued. Roguing Arran Pilot potatoes on 16 June or 2 July reduced leaf roll to five-sixths of that in unrogued plots; roguing on 16 June reduced rugose mosaic to about half that in plots rogued on 2 July, and about a quarter of that in unrogued plots. Lifting Arran Pilot potatoes in mid-August reduced virus diseases to about two-thirds.
Roguing flattened the gradient (decrease in percentage plants diseased with increasing distance from the source of infection) with rugose mosaic, but had little effect with leaf roll. Evidently any plants prevented by roguing from contracting virus Y were near the initially infected plants.
In 1948, Majestic and King Edward potatoes at three places were rogued during 22–24 June and tubers were dug during 28–30 July and again at the end of the season. Leaf roll spread more in Majestic than in King Edward, and rugose mosaic spread more in King Edward. Roguing reduced the spread of both by about one-fifth at Rothamsted, but had no effect at Sutton Bonington. At Bretton, in the Derbyshire hills, roguing had no effect on leaf roll, but prevented the spread of rugose mosaic.
The small benefit occasionally achieved by roguing in the ware-growing districts of England does not make the practice economically worth while.  相似文献

The epidemiology of potato virus Y (PVY) in the tobacco crop, Nicotiana tabacum, was examined in the context of the seasonal abundance of aphid vectors, rate of disease progress, and disease gradient from a known virus source. The spring potato crop, Solanum tuberosum, was suspected of being the main source of inoculum; therefore, varying numbers of infected potato plants were used as the inoculum source in different test plots. A 3-wk lag phase was present in all disease progress curves prior to an exponential increase in disease incidence. The relatively low numbers of aphid vectors, primarily transient species, alighting on the crop during the lag phase were responsible for the primary spread of PVY from potato to tobacco. The arrival of large numbers of colonising aphid vectors, Myzus persicae, presumably from the harvested potatoes, coincided with the exponential increase in PVY incidence in tobacco. The initial number of potato plants infected with PVY was positively correlated with the final disease incidence, rate of disease progress, and the magnitude of radial dispersion of PVY into the tobacco. Aphid vector pressure was not a significant variable in the differences in spatial and temporal characteristics of PVY epidemics among test plots.  相似文献

Young potato plants in pots exposed in the open near plots of potatoes for limited periods at intervals during the summer, became infested with large numbers of winged aphids only during warm, calm and dry weather. Although visited by aphids during May and June, when much of the spread of viruses occurred in nearby potato crops, few of the potted plants became infected. Most potted plants became infected in July when alate aphids were leaving neighbouring potato crops. Widely different proportions of the exposed plants became infected in different years; in two of the three years, many more plants were infected with virus Y than with leaf roll virus.  相似文献

In the field, caged potato plants of King Edward and Majestic cultivars infested with the potato aphid Macrosiphum euphorbiae developed top-roll symptoms, the proportion of affected plants increasing with the size and persistence of the aphid population. Yield of tubers from plots in which 90% of the plants had top-roll symptoms was 40% less than that from control plots; yield of saleable ware was even less. Foliage produced after the aphids had been killed was symptomless even when it arose from the axil of an affected leaf. Caged field plants treated with phorate granules to prevent aphid attack did not develop top-roll. Prolonged infestation of Pentland Crown, Majestic and King Edward plants by M. euphorbiae in a glasshouse induced rolling of upper leaves similar to top-roll of field plants. Experimental results suggest that rolling was directly attributable to heavy attack by M. euphorbiae, not to an aphid-transmitted pathogen.  相似文献

Three British strains of potato aucuba mosaic virus (PAMV) were tested for transmissibility by the aphid Myzus persicae. None was aphid transmissible on its own but all three were transmitted in the nonpersistent manner by aphids that had previously been fed on a source of the potyvirus potato virus Y (PVY). Different PVY strains mediated PAMV transmission from Nicotiana clevelandii to Capsicum annuum to different degrees, and different PAMV strains were transmitted at different frequencies when assisted by the same PVY strain. These results are compatible with the idea that subtle differences in the PAMV coat protein and in the PVY helper component are responsible for diffrences in frequencies of transmission of PAMV, without however, excluding the possibility of effects of other undefined factors. Transmission of PAMV was no less frequent when mediated by a PVY strain that was unable to infect C. annuum than when a C. annuum‐infecting PVY strain was used.  相似文献

以CO2浓度为主处理因子,研究了加倍CO2浓度和对照大气CO2浓度条件下,烟蚜、马铃薯Y病毒N株(PVYN)以及二者共同作用下烟草各指标的响应。结果表明,在当前CO2浓度条件下,PVYN、烟蚜及两者联合作用对烟草生物量影响不显著;而在未来高CO2浓度条件下,PVYN、烟蚜及两者联合作用对烟草生物量影响很大。CO2浓度升高后,PVYN和蚜虫二者联合作用显著降低烟草产量,危害加重,高CO2的"肥料"作用被极大地削弱。在有烟蚜、PVYN以及两者共同作用时烟草的化学物质及主要的次生代谢物烟碱的含量对CO2浓度升高的响应也发生一定的变化,表现在:高CO2浓度条件下,蚜虫、蚜虫与PVYN共同作用显著增加了烟草的含氮量;显著减少了烟叶含糖量;PVYN及其与蚜虫共同作用显著升高叶片可溶性蛋白含量;当高CO2浓度下,各处理的烟草烟碱含量均显著下降,而且PVYN感染的烟叶烟碱含量无论在哪一种CO2浓度条件下,都比无毒无虫的对照烟叶烟碱含量升高。结果显示,烟蚜和马铃薯Y病毒N株(PVYN)对烟草的产量、营养物质及防御物质都有影响;CO2浓度升高对烟草的生长有促进作用,增加了烟草的产量,但蚜虫的危害和PVYN感染使烟草产量下降,在高CO2浓度条件下,烟蚜和PVYN共同作用相对于目前CO2浓度对烟草产量的危害加重。  相似文献

Crop borders of soybean (Glycine max), sorghum (Sorghum bicolor), winter wheat (Triticum aestivum) and potato (Solanum tuberosum) were tested as a means of reducing potato virus Y (PVY) incidence in seed potato. Borders of fallow cultivated ground served as controls. Aphid landing rates were monitored weekly in plots using green tile traps, and PVY incidence was assessed by serologically testing tuber progeny from selected rows in each plot. Average weekly aphid landing rates in fallow-bordered and crop-bordered plots were not significantly different in 1992 (29.4 and 25.2 aphids, respectively) or 1993 (7.3 and 6.6 aphids, respectively). However, crop borders significantly reduced PVY incidence. In 1992, fallow-bordered and soybean-bordered plots averaged 47.8% and 35.0% PVY infection, respectively. In 1993, PVY infection averaged across all crop (soybean, sorghum, and wheat) bordered plots was 2.7% compared to 6.8% in fallow-bordered plots. PVY incidence in the centre rows of fallow-bordered and crop-bordered plots was statistically equivalent, while outer rows of crop-bordered plots had significantly less PVY than outer rows of fallow-bordered plots. Crop borders apparently reduced the number of viruliferous aphids landing on the edge of the plot. The choice of crop species used as a border, or treating the border with a systemic insecticide, did not affect aphid landing rates or PVY incidence. In 1995, PVY incidence in the centre 10 row block of potatoes averaged 2.1% across all crop borders (potato and soybean). PVY infection in the four row potato border averaged 5.7%. Crop borders are readily adaptable to current production practices, although the greatest benefits in reducing PVY incidence would occur in average sized, generation 0 (< 0.2 ha), elite seed potato fields.  相似文献

Potato leafroll virus (PLRV) causes one of the most serious aphid-transmitted diseases affecting yield and quality of potatoes, Solanum tuberosum (L.), grown in the United States. The green peach aphid, Myzus persicae (Sulzer), is considered to be by far the most efficient vector of this virus. Even the most strict aphid control strategy may not prevent the spread of PLRV unless measures also are taken to keep virus source plants within and outside the crop at a minimum. Hairy nightshade, Solanum sarrachoides (Sendtner), is one of the preferred weed hosts for green peach aphid. The potential of this weed as an aphid reservoir and virus source and its spread or perpetuation were investigated. With the use of double antibody sandwich enzyme-linked immunosorbent assay, it was confirmed that green peach aphid can transmit PLRV to hairy nightshade and that aphids can become viruliferous after feeding on infected hairy nightshade plants. Transmission from hairy nightshade to potato is 4 times the rate of potato to potato or potato to hairy nightshade. The green peach aphid preferred hairy nightshade over potato plants and reproduced at a higher rate on hairy nightshade than on potato. Therefore, a low level of PLRV-hairy nightshade infection could enhance the disease spread in the field.  相似文献
