The enantioselective pharmacokinetics of TJ0711 hydrochloride were studied in rats given different doses of rac‐TJ0711 hydrochloride via intravenous and oral routes. R‐ and S‐TJ0711 hydrochloride were both rapidly absorbed, and the average AUC0‐∞ of R‐TJ0711 hydrochloride was greater than that of S‐TJ0711 hydrochloride after intragastric administration, with an R/S AUC ratio 1.11 and 1.35 for 30 and 50 mg/kg dose group, respectively. In contrast, the average AUC0‐∞ of R‐TJ0711 hydrochloride was smaller than that of S‐TJ0711 hydrochloride after intravenous injection, with an R/S AUC ratio 0.57 and 0.73 for 10 and 20 mg/kg dose group, respectively. R‐TJ0711 hydrochloride plasma half‐lives were shorter than those of S‐TJ0711 hydrochloride for all groups. AUC0‐4h and Cmax between the two enantiomers were significantly different after oral administration of 50 mg/kg dose of the racemate, while no significant differences between the two enantiomers were found for all the pharmacokinetic parameters of the 30 mg/kg dose group. Significant differences between the two enantiomers were detected for nearly all the pharmacokinetic parameters after intravenous administration, except for the VZ of 20 mg/kg dose group. This study suggests that dose and route of administration will influence the enantioselectivity in the pharmacokinetics of TJ0711 hydrochloride in rats. The blood glucose response following the intravenous injection of butyric acid, 2.5 mM/kg, was studied in normal and carbon tetrachloride poisoned sheep. In normal sheep there was a rapid increase in blood glucose. Carbon tetrachloride greatly reduced the glucose response. In animals pretreated with nicotinic acid, 50 mg/kg, on the day before carbon tetrachloride administration, the glucose response was not altered. When a larger dose of nicotinic acid, 100 mg/kg, was given simultaneously with carbon tetrachloride, the glucose response was reduced more than after only carbon tetrachloride. Nicotinic acid alone at the dose of 100 mg/kg reduced the glucose rise somewhat more than carbon tetrachloride. There was a good agreement between the glucose rise following butyric acid and the glycogen content of the liver. It thus seemed clear that butyric acid has a glycogenolytic effect when given intravenously. Carbon tetrachloride caused severe necrosis and fatty changes in the liver. These pathological changes were reduced by pretreating the animals with 50 mg/kg of nicotinic acid. The larger dose of nicotinic acid, 100 mg/kg, caused a diffuse degeneration of the liver cells and a complete disappearence of glycogen. All liver injuries were followed by a rise in serum OCT.  相似文献

Blood glucose was significantly decreased by insulin (4 I.U./kg). Glucagon (1 mg/kg) and Cortisol (5 mg/kg) administration produced a significant hyperglycaemia. Insulin administration did not modify liver glycogen levels. Glucagon showed a marked liver glycogen mobilization. Cortisol stimulated liver glycogen deposition. Insulin and Glucagon showed a significant inverse effect on gluconeogenesis from (U-14C)glutamate, decreasing and increasing 14C-glucose formation respectively. Hormonal treatments did not influence the very low levels of incorporation of (U-14C)glutamate into liver and muscle glycogen.  相似文献

(1) Two procedures have been used to change the glutathione concentration in the livers of male rats. The glutathione level is increased to about double that of the controls, 0.5 h after the administration of cysteine (200 mg/kg, i.p.) and to about 25% that of controls, 1 h after the administration of 2-chloroethanol (30 mg/kg, i.p.). (2) The acute LD50 of retrorsine to rats (42 mg/kg) is increased by pretreatment with cysteine to 83 mg/kg and decreased by pre-treatment with chloroethanol to 23 mg/kg. In all three groups, deaths are accompanied by haemorrhagic centrilobular necrosis of the liver. (3) 2 h after the administration of retrorsine to rats (60 mg/kg), the levels of pyrrolic metabolites in the livers of animals pre-dosed with cysteine or chloroethanol are respectively about 60% and 200% those of rats given no pre-treatment. (4) Neither in the normal nor in the pre-treated rats dose retrorsine (60 mg/kg) cause a detectable fall in liver glutathione concentration 0.5-4 h after dosing. By 24 h, the glutathione concentration in the livers of the retrorsine-dosed rats is higher than those of the corresponding controls. There was no significant change in the liver weights of the treated rats relative to the controls. (5) Treatment of rats with retrorsine (60 mg/kg) causes a fall in the liver concentrations of cytochrome P-450, 24 h after dosing. This loss of cytochrome P-450 is increased in rats pre-treated with chloroethanol. The concentrations of cytochrome b5 in the same animals are not significantly reduced.  相似文献

Peroral administration of salvipholin in a dose of 50 mg/kg to intact male rats (the body weight 180-220 g) had a positive effect on the carbohydrate-lipid metabolism in the liver and blood serum of animals. Administration of the same dose to rats with alloxan diabetes induced a significant decrease in the content of glucose, free fat acids, triglycerides and lysophospholipids of the liver and blood serum. The level of these components has sharply increased after subcutaneous alloxan administration in a dose of 150 mg/kg, the content of glycogen and pyruvic acid being normalized and insulin deficiency removed. These changes are closely related to salvipholin-promoted restoration of phospholipid spectra of the blood serum and liver of experimental animals disturbed under conditions of insulin deficiency. The possible mechanism of the salvipholin action is analyzed.  相似文献

Male rats were given a single intragastric dose of 2-acetylaminofluorene, 600 mg/kg body weight, killed at intervals up to 14 days after treatment, and their hepatic tissue examined by electron microscopy. The early cytoplasmic lesion produced in hepatocytes by lower doses, consisting of perinuclear glycogen pooling, peripheral displacement of organelles and pyknosis, was delayed for several days. Among the changes which appeared to be independent of this lesion were disorganization and decrease of the granular endoplasmic reticulum and abnormalities of the bile canaliculi. These changes were similar to those which are seen during chronic exposure to 2-AAF and other hepatocarcinogens and in hepatic cell tumours.  相似文献

1. A single subcutaneous injection of clenbuterol hydrochloride (0.125 mg/kg body wt.) to female Wistar rats produced a rapid increase in muscle cyclic AMP and lactate concentrations and a decrease in muscle glycogen concentrations. These changes are characteristic of muscle beta-adrenoceptor stimulation and were abolished by intraperitoneal injection of propranolol (12.5 mg/kg) 15 min before clenbuterol administration. 2. When this dose of clenbuterol was injected twice daily, the changes in muscle metabolite concentrations which followed its acute administration persisted until day 7 of treatment, and were accompanied by increases in muscle mass, body weight and muscle protein synthesis rate (ks). When the clenbuterol injections were preceded by propranolol injections (12.5 mg/kg administered according to the protocol described above), or if animals were treated with propranolol only, the values of these variables were not significantly different from those of sham-injected controls. 3. In rats fed on a semi-synthetic diet (PW3) supplemented with 2 mg of clenbuterol/kg of diet for 7 days, the muscle mass was greater than that of rats fed on unsupplemented PW3. The increased muscle mass was accompanied by increased muscle lactate and decreased muscle glycogen concentrations. When PW3 was supplemented with 2 mg of clenbuterol/kg and 200 mg of propranolol/kg, the increase in muscle mass remained, but decreased muscle glycogen concentrations and increased muscle lactate concentrations were also observed. 4. These data are consistent with the hypothesis that clenbuterol influences muscle growth via beta-adrenoceptor stimulation.  相似文献

The chronic oral administration of phenformin (20 mg/kg daily during 8 days) to the normal concious dog provokes a decrease in the fasting thiamine levels of plasma and erythrocytes. During intragastric loading tests with thiamine hydrochloride (30 mg/kg), carried out during the chronic treatment with the biguanide, the curve of plasma hyperthiaminemia is increased. The frequent use of some trace elements such as gadolinium and terbium in medicine and modern industries make us worry about their behavior in the organism. In this work, we study the intracellular localization in the liver and in the intestinal mucosa of two rare earths, gadolinium and terbium, after intraperitoneal and intragastric administration. Three methods of observation and microanalysis were used: conventional transmission electron microscopy, secondary ion mass spectrometry, and electron probe microanalysis. After intraperitoneal administration, gadolinium and terbium were detected with phosphorus in lysosomes of hepatocytes and Küppfer cells and in territories near to biliary canalicule. One hour after intragastric administration, gadolinium and terbium were concentrated in lysosomes of the apical part of duodenal enterocytes. No gadolinium or terbium was detected in duodenum 4 days after administration. After intragastric administration, the microanalytical techniques failed to detect gadolinium or terbium in liver whatever the time of sampling. This mechanism of concentration-precipitation in the lysosomes of enterocytes limits the diffusion through the digestive barrier of foreign elements and then permits their elimination with apoptotic cells in the intestinal lumen. Some of these elements may be toxic, and none of them have a recognized physiological function. The intestinal mucosa plays an important role in the protection of the organism against the invasion of foreign elements.  相似文献

Results reveal that bromocriptine at a dose of 6 mg/kg. I.P. in mice caused a significant hyperglycaemic effect which was accompanied by a marked increase in liver glycogen. After adrenalectomy, the effect of bromocriptine on serum glucose level was reduced whereas its effect on liver glycogen content was abolished. In rats, administration of bromocriptine (17.5 mg/kg I.P.) induced a significant rise in serum glucose level which was associated with marked reduction in serum insulin activity and increase in serum corticosterone level with no effect on serum triiodothyronine (T3) or thyroxine (T4) levels. It could be concluded that bromocriptine-induced hyperglycaemia might be attributed at least partially to inhibition of insulin release and stimulation of corticosterone secretion.  相似文献

The effect of various doses of serotonin on the serum gastrin levels and intragastric pH in rats, was studied. After serotonin administration of 10 mg/kg i.p., a significant increase in serum gastrin levels was noted, as well as a strong increase in the intragastric pH. It was also observed that a lower dose, 5 mg/kg i.p., significantly increased serum gastrin levels, while intragastric pH was not affected, remaining at baseline values throughout the study. These results suggest that the increase observed in serum gastrin levels after administration of exogenous serotonin is not mediated by increase in intragastric pH.  相似文献

Acute or chronic intoxication of rats with ethanol (intragastric administration at a dose of 8 g/kg or free-choice drinking of 10% ethanol for 3 months) produced no significant changes in contractile function, glycogen content, glucose uptake and lactate release in isolated hearts. Withdrawal syndrome simulated in rats following a short period of severe intoxication with ethanol at a dose of 4-5 g/kg twice daily has demonstrated a 15 and 28% decrease in peak systolic pressure and tension time index, respectively. In this case glucose uptake and lactate release were 2 times higher. Changes in glycogen level were observed three days after the last ethanol administration. The rats, survived after the abstinence period, revealed areas of perivascular myocardial necrosis. It is concluded that withdrawal syndrome plays an important role in pathogenesis of alcoholic cardiomyopathy.  相似文献

They were separated and their antiulcer activity was studied. Thirty individual peptides with molecular masses of 174–420 amu were isolated from the PHC by HPLC. The PHC was shown to predominantly contain 2-to 4-aa peptides, including PG, GP, and PGP. Experiments on rats demonstrated that, on intragastric administration at a dose of 1 mg/kg, PHC enhances the stability of the gastric mucosa to the action of ulcerogenic factors, such as ethanol and stress, and exhibits a protecting antiulcer effect. Even a lesser dose (0.1 mg/kg), which reduced ulcer area twofold, was effective in the stress model of ulcer formation. The intraperitoneal and intragastric administration of PHC at a dose of 1 mg/kg was found to exhibit a therapeutic effect in the acetate model of ulcer formation.  相似文献

The influence of the synthetic sexual steroid levonorgestrel (LN) on rat liver in various doses and at different structural levels was investigated. A slight reactive hepatosis was found by histological examination after administration of LN in a dose of 10 mg per kg body wt. The same dose caused exclusively distinct lesions of the mitochondria, however, only in centrilobular parenchymal cells, whereas in the periportal hepatocytes only the lipid droplet content appears somewhat elevated. LN decreased the total glutathione content of the liver. The mitochondrial glutathione was decreased more intensively. One mg/kg body wt. of LN decreased the cytochrome P-450 content, but 10 mg/kg body wt. increased ethyl-morphine N-demethylation and 7-ethoxycoumarin O-deethylation activities. Distinct correlations could be shown between the biochemical changes and the ultrastructural findings.  相似文献

The effects of melatonin on several parameters of carbohydrate and lipid metabolism were investigated in exercised and nonexercised rats. Animals were run to exhaustion on a rodent treadmill at 24 m/min and a 12% slope. Exercise resulted in a significant hypoglycemia and increased plasma levels of lactate and beta-hydroxybutyrate, together with a significant reduction of glycogen in muscle and liver. Muscle and liver glycogen content was elevated and plasma free fatty acid decreased in nonexercised animals receiving melatonin (0.5 or 2.0 mg/kg i.p). Melatonin at 2.0 mg/kg reduced plasma lactate and increased lactate concentration in liver. When compared to untreated exercised animals glycemia and muscle and liver glycogen content were significantly higher in melatonin-treated exercised animals, while plasma and liver lactate and plasma beta-hydroxybutyrate were significantly reduced. Our data indicate that melatonin preserves glycogen stores in exercised rats through changes in carbohydrate and lipid utilization.  相似文献

This study was conducted to examine the protective role of crude polysaccharide from brown seaweed Sargassum polycystum against acetaminophen-induced abnormality in blood glucose, serum albumin/globulin ratio, and liver glycogen, lactate, and pyruvate. Liver and renal tissue histology was performed to confirm the efficacy of Sargassum polysaccharide. A toxic dose of acetaminophen (800 mg/kg body weight intraperitoneally) induced severe abnormality in all basic parameters with apparent toxicity in liver (enlargement of hepatocytes, loss of cytoplasmic content with disruption in the hepatic plates and sinusoidal dilation) and renal tissue (glomerular damage with congestion of tubules). The isolated liver cells were stained with acridine orange and examined under fluorescence microscope, which revealed that the acetaminophen induced significant damage. In contrast, the rats pretreated with Sargassum polysaccharide (200 mg/kg body weight) daily for 3 weeks did not show liver and renal tissue with these severe aberrations induced by acetaminophen. Histology results were also consistent with analyzed basic biochemical parameters, which confirmed the effectiveness of the crude polysaccharide against acetaminophen-induced abnormality in rats.  相似文献

The morphological state of the internal organs as well the changes of microsomal oxidation in liver of white rats exposed to the action of sodium sulfate in doses 200 and 600 mg per 100 g of body weight have been studied. The sodium sulfate in dose 600 mg per 100 g of body weight has been found to decrease the cytochrome P450 content, increase its inactivation rate and have an injurious effect on the membranes of hepatocytes. Sodium sulfate administration through the gastroenteric tract causes the destructive and pathological histochemical changes in liver, stomach, duodenum and small intestine. The alternative changes are expressed most of all in duodenum and small intestine.  相似文献

This study was to evaluate the effects of thalidomide on expression of adhesion molecules in liver cirrhosis. The cirrhosis was induced in Wistar rats by intraperitoneal injection of CCl(4), and thalidomide (10 mg/kg/day or 100 mg/kg/day) was given by intragastric administration for 8 weeks. Liver histopathology and immunohistochemistry were significantly improved and the expressions of ICAM-1, VCAM-1, E-selectin, and TNF-alpha mRNA and protein were decreased significantly in rats treated with a high dose of thalidomide. Close positive correlation was observed in the expression of the TNF-alpha mRNA and that of ICAM-1, VCAM-1, and E-selectin mRNA, respectively. These results indicate that thalidomide exerts its effect on the downregulation of adhesion molecules via TNF-alpha signaling pathway to inhibit liver fibrosis.  相似文献

Effects of intravenous cimetidine, ranitidine and intragastric prostaglandin E1 (alprostadil) treatments on the transmucosal potential difference (PD) of the stomach were compared. It was also investigated whether the above-mentioned drugs influenced the decrease of PD which followed both intragastric administration of 30% alcohol or Ca++ solution in 5 Mm final concentration and intravenous administration of pentagastrin. Both cimetidine and ranitidine treatments led to significant (p < 0.05) increase of PD, the effect of ranitidine was dose dependent. Prostaglandin E1 in a dose of 40 micrograms/kg led to significant decrease of PD (< 0.05). Both intragastric administration of prostaglandin E1 in a dose of 40 micrograms/kg and intravenous administration of ranitidine in a dose of 10 mg/kg significantly diminish the effect of Ca++ and alcohol to decrease PD. Neither prostaglandin E1, nor ranitidine pretreatment had any effect on the rapid and highly significant (p < 0.01) decrease of PD following i.v. pentagastrin administration. It is hypothesized that transmucosal PD of the stomach provides information not only on the actual condition of the mucosal barrier but on the electrophysiology of gastric secretion as well.  相似文献
