Natural, 2R,4R',8R'-alpha-tocopherol (vitamin E), labelled selectively with 13C in the methyl group at position 5, was incorporated into unilamellar vesicles of egg phosphatidylcholine. The vesicles are impermeable to the shift reagent Pr3+ and, in the presence of this reagent, separate 13C resonances due to labelled alpha-tocopherol in the outer and inner monolayers could be observed with relative intensities, 2:1. Subsequent addition of the relaxation reagent Gd2+ causes broadening and greatly shortened spin-lattice relaxation times for the resonance due to alpha-tocopherol in the outer monolayer only. These data confirm that alpha-tocopherol is located in both halves of the bilayers with its more hydrophilic chroman moiety very near the lipid-water interface, and indicate that the methyl group at position 5 of the alpha-tocopherol in the inner monolayer must be at least 40 A from the aqueous interface of the outer monolayer.  相似文献

With the aid of paramagentic praseodymium ions the resonances at 60 MHz of the inward and outward facing choline methyl protons of sonicated egg yolk phosphatidylcholine vesicles were resolved. The subsequent addition of 2,2,6,6,-tetramethylpiperidine-N-oxyl (TEMPO) to the vesicle suspension broadened the inner and outer resonances equally. TEMPO easily penetrates the egg yolk phosphatidylcholine vesicles and has free access to the entire lipid volume above the gel to liquid crystalline transition temperature. The electron spin resonance (ESR) spectrum of TEMPO in the egg yolk phosphatidylcholine suspension exhibits features indicating that TEMPO dissolves principally in the hydrocarbon portion of the egg yolk phosphatidylcholine bilayer. The egg yolk phosphatidylcholine methylene chain proton resonances are also broadened by TEMPO notably to a greater extent than the choline methyl resonances. These data indicate that TEMPO should be more sensitive to the melting behavior of the fatty acyl chains of phospholipid suspensions than to the polar head groups.  相似文献

1. Shifts in the 1H and 31P-nmr signals originating from the outer and inner phosphorylcholine head-groups and from the lipid acyl chains are observed when phosphatidylcholine vesicles are treated with increasing extravesicular concentrations of the lanthanides Eu3+, Pr3+, Yb3+, and Dy3+. 2. The addition of KNCS to increase the binding of the lanthanide ions to the outer head-groups is used to demonstrate that the intravesicular group shifts are not caused by bulk susceptibility effects. 3. The magnitude and direction of the observed shifts in the 1H-nmr spectrum are shown to be consistent with (a) pseudocontact interaction of the paramagnetic lanthanide ions with the outer phospholipid head-groups, (b) current views of the conformation of the phosphatidylcholine head-group in the presence of lanthanides, and (c) a conservation of magnetic field within the vesicles due to their spherical nature. 4. Variation of the shifts with temperature are compared for egg phosphatidylcholine and dipalmitoyl phosphatidylcholine. The temperature variation in shifts is also used to study phase transitions in each monolayer and phase separations in mixed lipid systems.  相似文献

NMR, calorimetric, and optical spectroscopic studies have been performed on a trifluoromethyl-substituted styryl molecular probe bound to vesicles and multilamellar suspensions formed from dimyristoylphosphatidylcholine (DMPC). In the fluorine NMR spectrum at 35 degrees C there are two partially resolved resonances, but these collapse to an apparently single resonance at temperatures above 60 degrees C. However, a line-shape analysis is not consistent with exchange between two sites on an NMR time scale, and the two resonances are assumed to be due to probe sites in the inner and outer leaflets of the vesicles. Two fluorescence lifetimes, each associated with one of these sites, characterize the decay curves for the molecular probe bound to DMPC vesicles. The shift reagent Eu(FOD)3 and several nitroxide spin labels covalently bound to lipophilic structures strongly attenuate the lower frequency component of the fluorine NMR spectrum and also shift the other resonance to higher frequencies. The effect of two spin labels on the probe fluorine T2 relaxation time has been used to estimate the distance between the spin label unpaired electron and the trifluoromethyl group. The location of the spin label site in the membrane was determined from the effect of the unpaired electron on the lipid 13C linewidths. A model for the location of the probe in the bilayer was developed from the above information and refined using molecular mechanics calculations on a probe-DMPC lipid complex. The long axis of the probe parallels the bilayer normal; the styryl-group portion of the optical chromophore is located slightly below the glycerol backbone, and the remainder of the chromophore extends well into the hydrophobic region of the bilayer. Therefore, the optical properties of the probe should not be significantly influenced by alterations of the membrane surface charge density. Parameters derived from DSC studies in the gel-to-lipid crystal phase transition of DMPC are extremely sensitive to the probe. Even at 0.0001 mol fraction of probe, the transition is substantially broadened, and the delta H for the transition has increased, just as one predicts for the formation of a tight complex described above.  相似文献

We discuss the thermodynamic behavior of a bilayer composed of two coupled leaves and derive the Gibbs Phase Rule for such a system. A simple phenomenological model of such a system is considered in which the state of the bilayer is specified by the relative number of ordering lipids in the outer leaf, and in the inner leaf. Two cases are treated. In the first, both inner and outer leaves could undergo phase separation when uncoupled from one another. The bilayer can exist in four different phases, and can exhibit three-phase coexistence. In the second case, an outer layer which can undergo phase separation by itself is coupled to an inner leaf which cannot. We find that when the coupling is weak, the bilayer can exist in only two phases, one in which the outer layer is rich in ordering lipids and the inner leaf is somewhat richer in them than when uncoupled, and another in which the outer layer is poor in ordering lipids and the inner leaf is poorer in them than when uncoupled. Increasing the coupling increases the effect on the inner leaf composition due to small changes in those of the outer leaf. For sufficiently large coupling, a phase transition occurs and the bilayer exhibits four phases as in the first case considered. Our results are in accord with several observations made recently.  相似文献

The optical responses of metal nanoparticles induced by subtle variations in geometry, especially by the rounding of the edges and corners, have generated great interest at present due to the requirement of fabricating refined structures of metal nanoparticles and theoretical simulations of the real particles. We study the effect of both inner and outer edge rounding on the optical properties of gold nanobox and gold nanobox dimer with small interparticle distances by using the discrete dipole approximation method. The shift of extinction peaks, the electric field distribution, and the variation of refractive index sensitivities by changing the curvature of the inner and outer edges of gold nanobox are investigated. We demonstrate that the optical properties of nanobox are more sensitive to the outer edge rounding than the inner edge rounding. By edge rounding of two very close gold nanoboxes, the blue shift of the dipolar and the quadrupolar plasmonic resonances of nanobox dimer are shown. Comparing with the inner edge rounding of nanobox dimer, we find that rounding of the outer edges causes the larger shift of the quadrupolar mode and approximate shift of the dipole mode.  相似文献

Effects of paramagnetic shift reagents on the 13C NMR spectra obtained from single-walled vesicle dispersions of egg phosphatidylcholine enriched with 13C in the N-methyl carbons are investigated. Spectra obtained at 25.1 MHz show that, at Yb3+ to phospholipid molar ratios as low as 0.06, complete resolution of the N-methyl carbon resonances is obtained from molecules on the inner and outer faces of the vesicle bilayer. No precipitation of the vesicles is caused by Yb3+ at these concentrations nor is appreciable line broadening observed. Other paramagnetic shift reagents frequently used in proton NMR investigations of phosphatidylcholine vesicles do not give complete separation of the N-methyl 13C signals from the two bilayer surfaces. K3Fe(CN)b,Eu3+, and Pr3+ cause precipitation of the phosphatidylcholine vesicles at concentrations, which give only incomplete resolution of these signals. T1 measurements of the resonances separated by Yb3+ indicate that the choline groups on the inner bilayer surface are less mobile than are the same groups in the outer surface. Gated proton decoupling measurements, which show that the nuclear Overhauser effect is 2.8 +/- 0.1, indicate that the dominant mode of relaxation is dipolar interaction.  相似文献

The interaction of diacylglycerols, primarily 1,2-dilauroyl-sn-glycerol (1,2-DLG), with egg phosphatidylcholine (PC) bilayers was studied by NMR spectroscopy and other physical techniques. In the low proportions used (less than or equal to 20 mol % with respect to total lipid), 1,2-DLG formed bilayers with PC with no hexagonal phase separation, as assessed by light, polarizing and electron microscopy, and 31P and 13C NMR spectroscopy. The 13C-carbonyl chemical shift of 90% [13C]carbonyl 1,2-DLG was monitored in small unilamellar vesicles as a function of relative DLG content (1.5-20%) and temperature (10-55 degrees C). The chemically inequivalent sn-1 and sn-2 carbonyls gave a single, narrow resonance in vesicles, in contrast to neat 1,2-DLG and 1,2-DLG in organic solvents, whose spectra showed two well-separated carbonyl resonances. The chemical shift of 1,2-DLG in PC shows that the carbonyl groups are proximal to the aqueous interface, necessitating orientation of the DLG molecule along the normal to the bilayer. Both carbonyl groups are H-bonded to H2O, but the secondary ester (sn-2) carbonyl is relatively more hydrated than the primary ester (sn-1) carbonyl. The 13C-carbonyl chemical shift data further suggest that the interfacial conformation resembles that of crystalline and liquid crystalline lamellar 1,2-dilauroyl-sn-glycero-3-phosphatidylethanolamine and certain PCs, in which the glycerol backbone is perpendicular to the bilayer plane. This conformation is different from that of crystalline 1,2-dilauroyl-sn-glycerol, in which the glycerol backbone is parallel to the bilayer plane. Between 1.5 and 8% DLG in vesicles, the chemical shift of the 1,2-DLG carbonyl at a given temperature was constant. However, above 8% DLG the chemical shift at each temperature increased with increasing DLG concentration, suggesting increased hydration at higher DLG content. At low temperatures 13C NMR spectra of vesicles with the highest proportions of 1,2-DLG studied (15 and 20%) showed two DLG carbonyl resonances, which most likely represent 1,2-DLG on outer and inner leaflets of the vesicle bilayer. The two peaks collapsed into a single resonance by 38 degrees C, at which temperature the two environments equilibrate with a rate constant of approximately 60 s-1 (t1/2 approximately 10 ms). Thus, transbilayer movement of DLG is extremely fast compared with phospholipids. In vesicles the 1,3-isomer of DLG exhibited a narrow carbonyl peak slightly downfield from that of 1,2-DLG. Acyl chain migration from 1,2-DLG to 1,3-DLG was monitored directly in the vesicle by time-dependent NMR measurements.  相似文献

The surface plasmon resonances of silver nanoshell particles are studied by Green’s function. The nanoshell system of plasmon resonances results from the coupling of the inner and outer shell surface plasmon. The shift of the nanoshell plasmon resonances wavelength is plotted against with different dielectric environments, several different dielectric cores, the ratio of the inner and outer radius, and also its assemblies. The results show that a red- and blue-shifted localized surface plasmon can be tuned over an extended wavelength range by varying dielectric environments, the dielectric constants and the radius of nanoshell core respectively. In addition, the separation distances, the distribution of electrical field intensity, the incident directions and its polarizations are also investigated. The study is useful to broaden the application scopes of Raman spectroscopy and nano-optics.  相似文献

The helix-coil transition of the octanucleotide self-complementary duplex dG-dG-dA-dA-dT-dT-dC-dC has been monitored at the Watson-Crick protons, the base and sugar nonexchangeable protons and the backbone phosphates by high-resolution nuclear magnetic resonance (NMR) spectroscopy. The melting transition of the octanucleotide monitored by ultraviolet absorbance spectroscopy is characterized by the thermodynamic parameters delta H degree = -216.7 kJ/mol and delta S degree (25 degrees C) = -0.632 KJ mol-1 K-1 in 0.1 M NaCl, 10 mM phosphate solution. Correlation of the transition midpoint values monitored by the ultraviolet absorbance studies at strand concentrations below 0.2 mM and by NMR studies at 5.3 mM suggest that both methods are monitoring the octanucleotide duplex-to-strand transition. The NMR spectra of the Watson-Crick ring NH protons of the octanucleotide duplex have been followed as a function of temperature. The resonance from the terminal dG.dC base pairs broadens out at room temperature while the resonances from the other base pairs broaden simultaneously with the onset of the melting transition. The nonexchangeable base and sugar H-1' protons are resolved in the duplex and strand states and shift as average peaks through the melting transition. The experimental shifts on duplex formation have been compared with calculated values based on ring-current and atomic diamagnetic anisotropy contributions for a B-DNA base-pair-overlap geometry in solution. Several nonexchangeable proton resonances broaden in the fast-exchange region during the duplex-to-strand transition and the excess widths yield a duplex dissociation rate constant for the octanucleotide of 1.9 x 10(3) s-1 at 32 degrees C (fraction of duplex = 0.86) in 0.1 M NaCl, 10 mM phosphate buffer. The 31P resonances of the seven internucleotide phosphates are distributed over 0.6 ppm in the duplex state, shift downfield during the duplex-to-strand transition and undergo additional downfield shifts during the stacked-to-unstacked strand transition with increasing temperature.  相似文献

Spherical phospholipid bilayers, vesicles, were prepared by using the layer-by-layer double emulsion technique, which allows individual layers to be formed asymmetrically. Phases of the layers were adjusted by selecting the lipid tail group. The head group composition of the vesicle outer layer varied 0–100 % of phosphatidylcholine (PC) by 10 % under the condition that the diameter of the vesicle was kept constant. On the outer layer of the vesicle, the phospholipase D (PLD) reacted to convert PC to phosphatidic acid. The reaction induced a curvature change of the vesicles, which eventually led them to rupture. Response time from the PLD injection to the rupture was measured against the different compositions of the outer layer at each phase (solid and liquid) using the fluorescence intensity change of pH-sensitive dye encapsulated in the vesicles. From this measurement, the rupture caused by the PLD reaction was analyzed with respect to the phase asymmetry of the layers and the composition of the outer layer. These results were interpreted with the lipid density and stability of the layers. It was observed that the solid phase of the outer layer had a variance in response time according to the phase of the inner layer, whereas the liquid phase did not. Additionally, the response of the solid phase of the outer layer at the liquid phase of the inner layer was faster than at the solid phase of the inner layer as a result of its stability.  相似文献

Phosphorus-31 NMR studies of solutions of small L-alpha-dipalmitoyl phosphatidylcholine bilayer vesicles containing sodium dimethyl phosphate uniformly distributed between the continuous external and the intravesicular aqueous spaces, with the paramagnetic shift reagent Pr3+ present only in the external space, are reported. These studies give the distribution both of dipalmitoyl phosphatidylcholine in the vesicle inner and outer monolayers and of dimethyl phosphate in the aqueous spaces. With the third necessary parameter obtained from the vesicle sedimentation coefficient, the very different packing parameters of dipalmitoyl phosphatidylcholine in inner and outer monolayers can be determined. The vesicle outer radius is 109 A. Although the total bilayer thickness is virtually identical to that of planar bilayers, the outer monolayer is thicker (20 A) and the inner monolayer thinner (15 A). The area per head group at the inner surface, 68 A2, is like the planar value, but the tails are much more folded, so as to decrease the radial lengths and increase the tangential spreat (to 94A2). The reverse is true in the outer layer: the surface per head group is 76 A2, tapering to 51 A2 in the tail region, so that outer layer tails are relatively extended. The difference is equivalent to a shift of about two 2g1 kinks from outer to inner layers; the uneven packing certainly affects fluidity, and may have important biological consequences.  相似文献

Arrhenius plots of various enzyme and transport systems associated with the liver mitochondrial inner membranes of ground squirrels exhibit changes in slope at temperatures of 20-25 degrees C in nonhibernating but not in hibernating animals. It has been proposed that the Arrhenius breaks observed in nonhibernating animals are the result of a gel to liquid-crystalline phase transition of the mitochondrial membrane lipids, which also occurs at 20-25 degrees C, and that the absence of such breaks in hibernating animals is due to a major depression of this lipid phase transition to temperatures below 4 degrees C. In order to test this hypothesis, we have examined the thermotropic phase behavior of liver inner mitochondrial membranes from hibernating and nonhibernating Richardson's ground squirrels, Spermophilus richardsonii, by differential scanning calorimetry and by 19F nuclear magnetic resonance and fluorescence polarization spectroscopy. Each of these techniques indicates that no lipid phase transition occurs in the membranes of either hibernating or nonhibernating ground squirrels within the physiological temperature range of this animal (4-37 degrees C). Moreover, differential scanning calorimetric measurements indicate that only a small depression of the lipid gel to liquid-crystalline phase transition, which is centered at about -5 degrees C in nonhibernating animals and at about -9 degrees C in hibernators, occurs. We thus conclude that the Arrhenius plot breaks observed in some membrane-associated enzymatic and transport activities of nonhibernating animals are not the result of a lipid phase transition and that a major shift in the gel to liquid-crystalline lipid phase transition temperature is not responsible for seasonal changes in the thermal behavior of these inner mitochondrial membrane proteins.  相似文献

Several synthetic amphiphiles with pyridinium ylide head-groups have been synthesized. The indicator groups are useful for microacidity measurement on inner and outer vesicular surfaces. Since the amphiphiles are water-insoluble, they constitute fully integrated parts of the vesicle membranes, Interfacial pH deviations on charged vesicle surfaces as compared to neutral surfaces are therefore much larger than with more water-soluble indicators.  相似文献

Binding of 2,3-diphosphoglycerate to monophosphoglycerate mutase, of which it is an obligatory cofactor, causes changes in the resonance positions of the 31P nuclear magnetic resonance spectra of both phosphate groups. It has previously been shown that these resonances shift when other glycolytic enzymes, such as phosphoglycerate kinase, are added to form the 2,3-diphosphoglycerate . monophosphoglycerate mutase . phosphoglycerate kinase complex. In view of this association, we have examined the set of glycolytic enzymes from aldolase to pyruvate kinase and found evidence of direct communication between all of these enzymes. A multi-enzyme complex of 1--2 . 10(6) daltons has been separated from broken cell ghosts by Biogel column filtration and evidence has been presented to show that this complex exhibits aldolase, glyceraldehyde 3-phosphate dehydrogenase and phosphoglycerate kinase activity. The glycolytic multi-enzyme complex interacts with the outer face of inside-out vesicles prepared from human red cells and the interaction is suppressed by application of 10(-6) M ouabain to the inner face of these vesicles. These studies show that the conformation of the enzymes comprising the megadalton complex are responsive to the application of ouabain to the outer red cell membrane surface.  相似文献

We report here on a series of studies aimed at characterization of the structural and dynamical properties of the synthetic lipid diphytanoyl phosphatidylcholine, in multilamellar dispersions and vesicle suspensions. The lipid exhibits no detectable gel to liquid crystalline phase transition over a large temperature range (-120 degrees C to +120 degrees C). Examination of proton nuclear magnetic resonance (NMR) free induction decays obtained from multilayer dispersions of diphytanoyl phosphatidylcholine provided an estimate of the methylene proton order parameter. The estimated magnitude of 0.21 is comparable to those determined for other phospholipids. Sonication of aqueous dispersions of diphytanoyl phosphatidylcholine led to formation of bilayer vesicles as determined by the measurement of the outer/inner choline methyl proton resonances, vesicle sizes in electron micrographs, and comparison of proton NMR linewidths between multilayer and sonicated dispersions. Ultracentrifugation studies of diphytanoyl phosphatidylcholine vesicles in H2O and 2H2O media yielded a value of 1.013 +/- 0.026 ml/g for the partial specific volume of this lipid. We have measured spin lattice relaxation rates for the methyl and methylenemethyne protons of the hydrocarbon chains of diphytanoyl phosphatidylcholine in bilayer vesicles over a range of temperatures and at two NMR frequencies (100 and 220 MHz). The observed relaxation rates for the methylene protons in this system were approximately twice those previously reported for dipalmitoyl phosphatidylcholine at comparable temperatures and resonance frequencies, whereas the relaxation rates measured for the methyl protons were greater than those of the straight chain lipid by an order of magnitude. Measurement of the spin lattice relaxation rates of the hydrocarbon protons of the diphytanoyl phosphatidylcholine in a 10 mol% mixture of the branched-chain lipid in a deuterated host lipid, diperdeuteropalmitoyl phosphatidylcholine, showed a discontinuity in the temperature dependence of the proton NMR longitudinal relaxation rates of the branched-chain lipid in the region of the gel to liquid crystalline phase transition temperature of the deuterated dipalmitoyl phosphatidylcholine host lipid. This result may be taken as evidence of lateral phase separation of a liquid cyrstalline phase enriched in diphytanoyl phosphatidylcholine from a gel phase enriched in diperdeuteropalmitoyl phosphatidylcholine at temperatures below the phase transition temperature of deuterated host lipid. This conclusion is supported by the observation of an abrupt change in the hydrocarbon methylene linewidth (at 100 MHz) of 10 mol% diphytanoyl phosphatidylcholine in diperdeuteropalmitoyl phosphatidylcholine over the temperature range where lateral phase separation is taking place according to differential thermograms.  相似文献

The process of biological membrane fusion can be analysed by topological methods. Mathematical analysis of the fusion process of vesicles indicated two significant facts: the formation of an inner, transient structure (hexagonal phase - H(II)) and a translocation of some lipids within the membrane. This shift had a vector character and only occurred from the outer to the inner layer. Model membrane composed of phosphatidylcholine (PC), phosphatidylethanolamine (PE) and phosphatidylserine (PS) was studied. (31)P- and (1)H-NMR methods were used to describe the process of fusion. (31)P-NMR spectra of multilamellar vesicles (MLV) were taken at various temperatures and concentrations of Ca(2+) ions (natural fusiogenic agent). A (31)P-NMR spectrum with the characteristic shape of the H(II) phase was obtained for the molar Ca(2+)/PS ratio of 2.0. During the study, (1)H-NMR and (31)P-NMR spectra for small unilamellar vesicle (SUV), which were dependent on time (concentration of Pr(3+) ions was constant), were also recorded. The presence of the paramagnetic Pr(3+) ions permits observation of separate signals from the hydrophilic part of the inner and outer lipid bilayers. The obtained results suggest that in the process of fusion translocation of phospholipid molecules takes place from the outer to the inner layer of the vesicle and size of the vesicles increase. The NMR study has showed that the intermediate state of the fusion process caused by Ca(2+) ions is the H(II) phase. The experimental results obtained are in agreement with the topological model as well.  相似文献

The orientation of melittin in lecithin membranes was investigated by means of 13C-NMR spectroscopy. Phospholipase-free melittin was labeled with 13C-methyl groups at the -amino side chains of lysine 7, 21, and 23. From the pH dependence of the corresponding 13C resonances, pK values of the lysine residues were derived that were different for free and membrane-bound melittin. The shift reagent Pr(NO3)3 induced shifts in the 13C resonance position of all three lysines when melittin and the shift reagent were added to a lecithin vesicle suspension, whereas Pr3+ ions included in the inner volume of the vesicles did not affect the 13C resonances of melittin bound to the outer vesicle membrane. 19F-labeled phospholipids, L-α-bis-(ω-fluoro palmitoyl)phosphatidylcholine, L-α-bis-(12,12-difluoro stearoyl)phosphatidylcholine and L-α-bis-(6,6-difluoro palmitoyl)phosphatidylcholine, were incorporated in phospholipid vesicles by sonication of aqueous lipid emulsions. Vesicles were prepared both from the pure fluorine substituted phospholipids as well as from lipid mixtures obtained by combining the fluorine substituted lipids with the synthetic phospholipids, L-α-dilauroyl-, L-α-dimyristoyl-, L-α-dipalmitoyl- and L-α-distearoyl-phosphatidylcholine. Characterization by gel permeation chromatography showed that stable unilamellar vesicles with diameters of ≌200 Å could be obtained with a minimum or absence of multilamellar material. The vesicles give rise to two 19F resonances in most cases as observed previously [K.J. Longmuir and F.W. Dahlquist, Proc. Natl. Acad. Sci. U.S.A., 73 (1976) 2716]. The chemical shift differences undergo systematic changes that confirm the interpretations that the dual 19F resonances arise from the inner and outer halves of the vesicle bilayer. The shift separation increases systematically as the fluorine label is positioned closer to the phospholipid headgroup and decreases systematically with increasing temperature. Both observations agree with what is currently known about phospholipid vesicle structure. Anomalous results are obtained with DSPC as host vesicle since only a single resonance of inbedded fluorinated phospholipids is found.  相似文献

We report here on a series of studies aimed at characterization of the structural and dynamical properties of the synthetic lipid diphytanoyl phosphatidylcholine, in multilamellar dispersions and vesicle suspensions.This lipid exhibits no detectable gel to liquid crystalline phase transition over a large temperature range (?120°C to +120°C).Examination of proton nuclear magnetic resonance (NMR) free induction decays obtained from multilayer dispersions of diphytanoyl phosphatidylcholine provided an estimate of the methylene proton order parameter. The estimated magnitude of 0.21 is comparable to those determined for other phospholipids.Sonication of aqueous dispersions of diphytanoyl phosphatidylcholine led to formation of bilayer vesicles as determined by the measurement of the outer/inner choline methyl proton resonances, vesicle sizes in electron micrographs, and comparison of proton NMR linewidths between multilayer and sonicated dispersions. Ultracentrifugation studies of diphytanoyl phosphatidylcholine vesicles in H2O and 2H2O media yielded a value of 1.013 ± 0.026 ml/g for the partial specific volume of this lipid.We have measured spin lattice relaxation rates for the methyl and methylenemethyne protons of the hydrocarbon chains of diphytanoyl phosphatidylcholine in bilayer vesicles over a range of temperatures and at two NMR frequencies (100 and 220 MHz). The observed relaxation rates for the methylene protons in this system were approximately twice those previously reported for dipalmitoyl phosphatidylcholine at comparable temperatures and resonance frequencies, whereas the relaxation rates measured for the methyl protons were greater than those of the straight chain lipid by an order of magnitude.Measurement of the spin lattice relaxation rates of the hydrocarbon protons of the diphytanoyl phosphatidylcholine in a 10 mol% mixture of the branched-chain lipid in a deuterated host lipid, diperdeuteropalmitoyl phosphatidylcholine, showed a discontinuity in the temperature dependence of the proton NMR longitudinal relaxation rates of the branched-chain lipid in the region of the gel to liquid crystalline phase transition temperature of the deuterated dipalmitoyl phosphatidylcholine host lipid. This result may be taken as evidence of lateral phase separation of a liquid cyrstalline phase enriched in diphytanoyl phosphatidylcholine from a gel phase enriched in diperdeuteropalmitoyl phosphatidylcholine at temperatures below the phase transition temperature of deuterated host lipid. This conclusion is supported by the observation of an abrupt change in the hydrocarbon methylene linewidth (at 100 MHz) of 10 mol% diphytanoyl phosphatidylcholine in diperdeuteropalmitoyl phosphatidylcholine over the temperature range where lateral phase separation is taking place according to differential thermograms.  相似文献
