In Phormidium laminosum cells, nitrogen starvation caused a decrease in the intracellular levels of all amino acids, except glutamate, and an increase in the total level of the analyzed organic acids. The addition of nitrate or ammonium to N-starved cells resulted in substantial increases in the pool size of most amino acids. Upon addition of ammonium the total level of organic acids diminished, whereas it increased upon addition of nitrate, after a transient decay during the first minutes. Nitrogen resupply stimulated amino acid synthesis, the effect being faster and higher when ammonium was assimilated. The data indicate that nitrate and ammonium assimilation induced an enhancement of carbon flow through the glycolytic and the tricarboxylic-acid pathways to amino acid biosynthesis, with a concurrent decrease in the carbohydrate reserves. Theoretical plant growth models postulate an important role for growth substrates such as sugars and amino acids. To test this experimentally, spinach plants were grown under controlled conditions and with nitrogen added daily, following different exponential addition schemes. Plants were harvested during exponential growth. Free amino acid levels or free sugar levels were only weakly correlated with growth and biomass partitioning. Factor analysis showed however that the product of free sugar concentration and amino acid concentration yielded a parameter adequately reflecting the plant's nutritional state.It is concluded that growth and biomass partitioning under limiting N conditions cannot be modelled solely based on N substrate levels.  相似文献

This study was done to evaluate the effects of two sublethal doses of gossypol (4 and 20 mg/kg of BW, every other day) on some amino and fatty acid concentrations in male rabbit seminal plasma. Rabbits were chosen as an experimental animal owing to the fact that they are excellent model for reproductive toxicological effects. The experiment lasted 16 weeks and included two periods: a treatment period (first 8 weeks) where the animals were given the tested product, and a recovery period (second 8 weeks) where all drugs were withdrawn. Results showed that total amino acids (TAA), total essential amino acids (EAA), total non-essential amino acids (non-EAA) and EAA/non-EAA ratio were decreased in a dose-dependent manner during gossypol treatment. The deleterious effect on TAA concentrations was mainly due to the reduction in total EAA. However, these concentrations regained their normal values after gossypol cessation. Basic, acidic, neutral amino acids and basic/acidic amino acids ratio decreased in a dose-dependent manner by gossypol treatment. Additionally, gossypol administration caused decreases in total unsaturated fatty acids (USFA) and increases in total saturated fatty acids (SFA) and the SFA/USFA ratio in a dose-dependent manner. During the recovery period, total SFA and USFA showed significant reduction and significant increase, respectively, after gossypol withdrawal. In conclusion, gossypol administration affected rabbit seminal plasma concentrations of amino and fatty acids in a dose-dependant manner. Gossypol reduced TAA, total EAA and total non-EAA. Additionally, gossypol caused decreases in total USFA and increases in total SFA. These deleterious effects were associated with poor-quality semen observed in our previous studies.  相似文献

Increasing sea surface temperatures (SST) and blooms of lipid‐poor, filamentous cyanobacteria can change mesozooplankton metabolism and foraging strategies in marine systems. Lipid shortage and imbalanced diet may challenge the build‐up of energy pools of lipids and proteins, and access to essential fatty acids (FAs) and amino acids (AAs) by copepods. The impact of cyanobacterial blooms on individual energy pools was assessed for key species temperate Temora longicornis and boreal Pseudo‐/Paracalanus spp. that dominated field mesozooplankton communities isolated by seasonal stratification in the central Baltic Sea during the hot and the cold summer. We looked at (a) total lipid and protein levels, (b) FA trophic markers and AA composition, and (c) compound‐specific stable carbon isotopes (δ13C) in bulk mesozooplankton and in a subset of parameters in particulate organic matter. Despite lipid‐poor cyanobacterial blooms, the key species were largely able to cover both energy pools, yet a tendency of lipid reduction was observed in surface animals. Omni‐ and carnivory feeding modes, FA trophic makers, and δ13C patterns in essential compounds emphasized that cyanobacterial FAs and AAs have been incorporated into mesozooplankton mainly via feeding on mixo‐ and heterotrophic (dino‐) flagellates and detrital complexes during summer. Foraging for essential highly unsaturated FAs from (dino‐) flagellates may have caused night migration of Pseudo‐/Paracalanus spp. from the deep subhalocline waters into the upper waters. Only in the hot summer (SST>19.0°C) was T. longicornis submerged in the colder subthermocline water (~4°C). Thus, the continuous warming trend and simultaneous feeding can eventually lead to competition on the preferred diet by key copepod species below the thermocline in stratified systems. A comparison of δ13C patterns of essential AAs in surface mesozooplankton across sub‐basins of low and high cyanobacterial biomasses revealed the potential of δ13C‐AA isoscapes for studies of commercial fish feeding trails across the Baltic Sea food webs.  相似文献

Sterols, fatty acids and free amino acids of Helvella crispa and H. monachella were investigated. They contained traces of ergosterol and a high amount of ergosta 5.22-dien-3β-ol. Linoleic acid and l-Dopa are the most abundant fatty acid and free amino acid, respectively. 为了探讨不同生境盐地碱蓬对低氮生境的适应机制,测定了盐渍环境下(200 mmol/L Na Cl)不同浓度硝态氮(0.3、5 mmol/L NO~-_3-N)预处理两种生境盐地碱蓬经氮饥饿后的NO~-_3含量、硝酸还原酶(NR)活性、光合特性及生长状况。结果表明,0.3和5 mmol/L NO~-_3-N处理以及进行氮饥饿时,潮间带生境盐地碱蓬叶片NO~-_3含量均高于内陆生境盐地碱蓬。与内陆生境盐地碱蓬相比,氮饥饿后,潮间带生境盐地碱蓬叶绿素含量、NR活性和光合放氧速率下降幅度均小于内陆生境盐地碱蓬,在0.3mmol/L NO~-_3-N预处理进行氮饥饿时趋势更加明显。0.3 mmol/L NO~-_3-N预处理后氮饥饿对潮间带生境盐地碱蓬根冠比没有影响,却降低内陆生境盐地碱蓬根冠比。上述结果表明,低氮条件下潮间带生境盐地碱蓬具有较高的NO~-_3储存能力,在环境持续氮素缺乏时具有较高的NO~-_3-N再利用能力,能更好地维持氮代谢以及光合性能。说明潮间带生境盐地碱蓬能更好地适应低氮生境。  相似文献

NineZoogloea strains including the type strain ofZ. ramigera (IAM 12136=ATCC 19544=N.C. Dondero 106) and newly isolated strains were investigated for isoprenoid quinone composition and whole-cell fatty acid profiles. Seven of the tested strains, having phenotypic properties typical ofZoogloea, were characterized by their production of both ubiquinone-8 and rhodoquinone-8 as major quinones, whereas the remaining two strains,Z. ramigera IAM 12669 (=K. Crabtree I-16-M) and IAM 12670 (=P.R. Dugan 115), formed ubiquinone-10 and ubiquinone-8, respectively, as the sole quinone. All rhodoquinone-producing strains contained palmitoleic acid and 3-hydroxy-decanoic acid as the major components of nonpolar and hydroxylated fatty acids, respectively. Marked differences were noted in the fatty acid composition between the strains with and without rhodoquinones. The chemotaxonomic data suggested that the rhodoquinonelacking strains should be excluded from the genusZoogloea. Since there have been no reliable taxonomic tools forZoogloea, rhodoquinone analysis may provide a new criterion of great promise for identifyingZoogloea strains.  相似文献

To investigate the full potential of hydrolases for the removal of two amine-protecting groups, 15 different, commercially available lipases, acylases, proteases and esterases were studied for the hydrolyses of N-acetyl and N-formyl protecting groups. In addition to the well-known acylases from porcine kidney and Aspergillus melleus, this screening revealed that porcine liver esterase and the lipases from Rhizomucor miehei and Pseudomonas stutzeri are also catalysts for the hydrolysis of N-acetylalanine. The activity of lipases in this reaction was unexpected, since lipases are commonly believed not to hydrolyse amides. In addition, from these 15 enzymes, three were found to be active in the hydrolysis of N-formylalanine, i.e. porcine liver esterase and the two acylases. This is the first example where esterase is employed to deprotect N-formyl amides.  相似文献

A brown blotch bacterium,Pseudomonas tolaasii strain PT814, expresses a high degree of cross-protection against generalized stress imposed by physical/chemical treatment, H2O2, UV, high temperature, ethanol and NaCl during the interaction withPleurotus ostreatus. Stress resistance was also noted in the bacterium in vitro under limited carbon and nitrogen sources. In addition, changes in cell morphology from a “metabolically active” rod to an “energy-saving” spherical shape were detected during starvation and the interaction. All the changes under stress were reversible. A homologue ofrpoS (σ S), a regulator that controls such physiological status during starvation in other bacteria, was identified inP. tolaasii strain PT814. Data suggest that the bacterium is able to withstand a complex stress environment for its survival through changes in its metabolic pattern.  相似文献

A continuous culture ofIsochrysis aff.galbana clone T.iso, used to feedPecten maximus larvae at IFREMER (Brest, France), was carried out in a chemostat at its optimum temperature for growth (26 °C). Changes in pigments, lipid class (neutral, glyco- and phospholipids) and degree of fatty acid unsaturation were studied at three different growth rates (0.33, 0.5, 1 d–1). As predicted by chemostat theory, a slow growth rate produced higher cell numbers and higher biomass per unit volume. These cells were low in chlorophylla and carotenoids, but rich in neutral lipids. In contrast, cultures with a fast growth rate yielded lower cell concentrations, buth higher chlorophylla, carotenoid and membrane lipid contents per cell. Changes in polyunsaturated fatty acid distribution were related to differences in algal growth rates. Neutral lipids contained mainly saturated and monounsaturated fatty acids (C18:19) at low growth rates whereas they were enriched in polyunsaturated fatty acids, especially C22:63, at high growth rates. Therefore, it is suggested that the growth rate in continuous cultures be controlled so as to adjust the relative proportions of polyunsaturated fatty acids in lipid classes of the diet meant for larval nutrition.Author for correspondence  相似文献

为探究熊蜂Bombus spp.蜂王体内氨基酸和脂肪酸对其不同繁殖状态的影响,本研究分别利用高效液相色谱方法和气质联用方法测定成群蜂王和未成群蜂王体内氨基酸和脂肪酸的含量并进行比较分析。研究结果表明:熊蜂蜂王体内分别包含16种氨基酸和17种脂肪酸。熊蜂蜂王体内含量最多6种的水解氨基酸为谷氨酸、丙氨酸、亮氨酸、丝氨酸、天冬氨酸和苏氨酸,含量最多的3种脂肪酸为油酸、亚油酸和α-亚油酸。成群与未成群阶段熊蜂蜂王体内13种氨基酸含量存在显著差异(P<0.05),且除丙氨酸外,成群蜂王体内其余12种氨基酸含量均显著低于未成群蜂王。成群与未成群阶段熊蜂蜂王体内有6种脂肪酸含量存在显著差异(P<0.05),其中未成群蜂王体内辛酸、癸酸、月桂酸的含量较高,而成群蜂王体内亚油酸、顺-11-二十烯酸、二十三酸的含量较高。本研究结果表明,熊蜂成群和未成群蜂王体内氨基酸和脂肪酸含量存在显著差异(P<0.05),蛋白质和脂类代谢异常可能是导致熊蜂蜂王成群率低的一个主要原因。  相似文献

Plasma amino acid (AA) levels of carp, Cyprinus carpio L., 1758, were analysed after various periods of starvation as well as after 12 days of refeeding. The levels were compared to control groups, which had been previously fasted for 24 h. A positive correlation between dietary and plasma essential amino acid (EAA) concentrations was observed in all of the control groups.
The effect of starvation on the dynamics of AA concentration was different according to the period of starvation. Fasting already produced a decrease of total α-AA levels at 2 days, and these low levels were maintained until 5 and 8 days. These short periods of fasting affected the levels of EAA (especially branched-chain AA) more than those of non-essential amino acids (NEAA). The only AA that increased was Ala, which rose at 5 days of starvation, surpassing the levels of the control group. These high levels were maintained until 19 days of starvation.
A different situation was observed at 19 days, when an important increase of total α-AA levels was produced, the branched-chain AA being the most notable among EAA and Glu/Gln among NEAA. Later, at 50 days of starvation, total α-AA, EAA (except Leu and Ile) and NEAA decreased.
After 50 days of starvation, 12 days of refeeding did not modify the levels of EAA and NEAA, and their concentrations were lower than those of the control group, which presented an increase of total AA at this time. The differences observed on the changes of individual AA levels are discussed.  相似文献

Morganella morganii produced CO when cultured in a medium containing casamino acids or peptone as the sole carbon source. Although the production of CO was distinctly enhanced by the addition of hemin to the medium, the amounts of CO produced in the absence of hemin were nearly proportional to the amounts of peptone added to the culture media. Examination of 20 amino acids for their ability to produce CO by resting cells revealed that phenylalanine, tyrosine, histidine and tryptophan were the sources of CO. Oxygen and hemin were necessary for CO production from the amino acids except tryptophan which produced CO in the absence of hemin. When cells were incubated for 4 h at 30° C in the mixture containing 40 mol tyrosine and 1 mol hemin, about 15 mol CO was produced; the activity of CO production was about 1.2 mol CO/h · mg cell nitrogen. Phenylpyruvic acid, p-hydroxyphenylpyruvic acid and imidazolepyruvic acid also produced CO in the presence of hemin, while indolepyruvic acid produced CO regardless of the presence or absence of hemin. The production of CO by the 2-oxo acids proceeded spontaneously and did not require the presence of M. morganii cells.  相似文献

The polar lipid fatty acids, lipopolysaccharide hydroxy-fatty acids, and respiratory quinones of Geobacter metallireducens str. GS-15, Geobacter sulfurreducens str. PCA, and Geobacter bemidjiensis str. Bem are reported. Also, the lipids of G. metallireducens were compared when grown with Fe3+ or nitrate as electron acceptors and G. sulfurreducens with Fe3+ or fumarate. In all experiments, the most abundant polar lipid fatty acids were 14:0, i15:0, 16:1ω7c, 16:1ω5c, and 16:0; lipopolysaccharide hydroxy-fatty acids were dominated by 3oh16:0, 3oh14:0, 9oh16:0, and 10oh16:0; and menaquinone-8 was the most abundant respiratory quinone. 黄土高原不同土壤微生物量碳、氮与氮素矿化势的差异   总被引:2,自引:0,他引:2  
以采自于黄土高原差异较大的25个农田石灰性耕层土壤为供试土样,对黄土高原主要类型土壤中微生物量碳(Bc)、微生物量氮(BN)和氮素矿化势(NO)的差异性进行了比较研究.结果表明,Bc、BN和NO在不同类型土壤间存在显著差异,由关中平原至陕北风沙区,BC、Bn和NO总体呈现下降趋势,其中以土垫旱耕人为土最高,简育干润均腐土最低,黄土正常新成土和干润砂质新成土居中:土垫旱耕人为土、简育干润均腐土、黄土正常新成土和干润砂质新成土等各土类平均BC分别为305.2μg·g-1,108.4μg·g-1,161.7μg·g-1和125.4μg·g-1,BN分别为43.8μg·g-1,20.3μg·g-1,26.0μg·g-1和30.6μg·g-1,NO分别为223μ·g-1,75μg·g-1,163μg·g-1和193μg·g-1.土壤氮素矿化速率(k)则以简育干润均腐土最大,干润砂质新成土最低,土垫旱耕人为土和黄土正常新成土居中:土垫旱耕人为土、简育干润均腐土、黄土正常新成土和干润砂质新成土的k分别为0.039w-1,0.044w-1,0.031w-1和0.019w-1.不同类型土壤BC、BN与NO的差异,主要与土壤形成过程、输入土壤的植物同化产物和土壤有机质的差异等有关,从较大尺度进一步证明了在黄土高原,土壤有机质是影响BC、BN的主要因子.研究结果对分析黄土高原土壤生产力形成过程具有一定参考价值.  相似文献

Ergosterol and cholesterol supplementation resulted in a significant increase (1·5-fold) in the sterol content while phospholipid remained unaffected inMicrosporum gypseum. The levels of phosphatidylethanolamine and phosphatidylcholine increased in ergosterol supplemented cells. However, a decrease in phosphatidylcholine and an increase in phosphatidylethanolamine was observed in cholesterol grown cells. The ratio of unsaturated to saturated fatty acids decreased on ergosterol/cholesterol supplementation. The uptake of amino acids (lysine, glycine and aspartic acid) decreased in sterol supplemented cells. Studies with fluorescent probe l-anilinonaphthalene-8-sulfonate showed structural changes in membrane organisation as evident by increased number of binding sites in such cells.  相似文献

林窗尺度对侧柏人工林土壤微生物生物量碳氮的短期影响   总被引:2,自引:0,他引:2  
为了明晰林窗尺度对侧柏人工林土壤微生物生物量碳氮的短期影响,以徐州市50年生侧柏人工林为研究对象,探讨了开设林窗2a后3种林窗尺度(半径分别为4m,S林窗;8m,M林窗;12m,L林窗)和位置(林窗内、林缘和林窗外部)对土壤微生物生物量碳氮(MBC,Microbial Biomass Carbon;MBN,Microbial Biomass Nitrogen)的影响。结果表明:(1)与对照样地相比,L林窗显著提高了春季(207.1mg/kg)和夏季(169.5mg/kg)林窗外部的土壤MBN含量;M林窗显著提高了春季林窗内部(2959.3mg/kg)和林缘(3008.8mg/kg)位置土壤MBC含量和林缘位置(207.7mg/kg)土壤MBN含量,且显著提高了夏季林窗内部(144.4mg/kg)土壤MBN含量;S林窗显著降低了春季林窗外部和林缘位置土壤MBC(分别为2159.2mg/kg和1955.1mg/kg)和MBN(分别为153.1mg/kg和131.3mg/kg)含量。(2)土壤MBC含量与土壤全碳和土壤可溶性有机碳(DOC)含量呈极显著(P0.01)正相关,与土壤温度呈极显著负相关(在3—27℃之间);土壤MBN含量与土壤含水量和DOC含量呈极显著正相关,与土壤全碳呈显著正相关;土壤MBC和MBN含量与凋落物量没有显著相关关系。本研究中,相对于S和L林窗,M林窗对土壤微生物生物量的提高作用较为明显,可促进侧柏人工林土壤碳氮循环过程,在徐州侧柏人工林中开设M林窗有利于提高土壤肥力和林木生长。  相似文献
