剪切力对脑微血管内皮细胞骨架蛋白的影响   总被引:7,自引:2,他引:5  
利用自行研制的细胞流动小室对大鼠脑微血管内皮细胞在剪切力作用下细胞骨架蛋白的结构改变进行了初步研究,结果提示脑微血管内皮细胞在剪切力作用下,细胞形态学发生明显改变,细胞间隙增大、皱缩、脱落,细胞骨架蛋白的结构也有类似的变化,骨架蛋白沿流动方向重新排列,微丝中F-Actin的数量增加、变粗。这些改变的直接后果是内皮细胞通透性的增加。该工作为进一步开展剪切力对微血管内皮细胞功能、代谢等方面的影响提供了实验数据  相似文献

利用内皮细胞流动小室方法,对大鼠脑微血管内皮细胞的剪切力作用下细胞内粘附分子-1(ICAM-1,intercellular adhesion molecule-1)的表达进行了研究。图像分析结果提示,脑微血管内皮细胞在剪切力作用下ICAM-1的表达呈特异上调,且存在着时间依赖性,与一定范围内的剪切力强度无关,用对细胞施加剪切力作用后提取上清液孵育内皮细胞的方法证明:剪切力对鼠脑微血管内皮细胞ICAM-1表达的影响,是直接作用于内皮细胞引起的细胞内的直接反应,而不是剪切力导致细胞先释放细胞因子,释放的细胞因子再引起ICAM-1变化的间接反应。该工作为进一步开展剪切力对微血管内皮细胞信号转导机制的影响提供了实验数据。  相似文献

利用膜片钳及内皮细胞流动小室方法对大鼠脑微血管内皮细胞在剪切力作用下内皮细胞膜K 通道的开放进行了初步研究 ,结果提示脑微血管内皮细胞膜上存在剪切力敏感的K 通道 ,剪切力作用后 ,内皮细胞膜上K 电流明显增大 ,此电流有明显的短暂延迟现象 ,也可以被胞外施加的TEA抑制 ,符合IKv特征。流动剪切力可以通过影响内皮细胞膜上的K 通道的开放引起穿细胞的离子通透性的增加 ,进而引起细胞内Ca2 的变化。在K 、Ca2 等离子浓度改变的诱导下可以促使G -Actin装配为F -Actin。同时诱导内皮细胞内钙库调节机制的激活 ,这些变化都可以进一步引起细胞信号转导机制的激活。该工作为进一步开展剪切力对微血管内皮细胞信号转导机制的影响提供了实验数据。  相似文献

白细胞介素(IL-1)及阿片肽作为神经调质参与了神经细胞兴奋性毒性作用,以大鼠大脑皮层神经细胞为研究对象,探讨了IL-1、阿片肽和c-fos、c-jun表达产物之间的关系,结果表明,IL-1β能诱导大脑皮层神经细胞c-fos、c-jun mRNA瞬时短暂表达,15min增高,30min达高峰,c-fos mRNA 2h回至基线水平,c-jun m RNA 8h回至基线水平;联合应用c-fos,c-  相似文献

豚鼠气道及肺组织原癌基因表达与哮喘的相关研究   总被引:4,自引:0,他引:4  
目的:为研究原癌基因在哮喘发病中的作用。方法:以卵白蛋白致敏豚鼠建立哮喘模型,用Dot-blot、North-ern-blot分子杂交及免疫组化技术,分别观察正常及哮喘发作后豚鼠气道上皮及肺组织中原癌基因c-fos、c-myc的表达水平。结果:正常豚鼠气道及肺组织中c-fos及c-mycmRNA无或很少表达,哮喘发作后豚鼠气道上皮及肺组织中c-fos和c-mycmRNA表达明显增强,30min达高峰,发作后4h降至正常水平。地塞米松对c-fos及c-mycmR-NA有部分抑制作用。结论:原癌基因c-fos及c-myc在哮喘发病过程中可能起一定作用。  相似文献

为了探讨癫痫发病机理与原癌基因的关系,将马桑内酯注入侧脑室,于大鼠癫痫发作后不同时间点取材海马,用Northern印迹杂交和Western印迹分析技术对海马c-fosmRNA和Fos蛋白表达的动力学进行了定量研究,激光扫描仪定量分析表明:(1)c-fosmRNA在0.5h表达最高,依次渐减,至4h几无表达;(2)Fos蛋白在1h开始表达,2h最多,以后渐少。以上结果表明:c-fosmRNA和Fos蛋白的表达随癫痫的发生发展出现规律性变化,c-fosmRNA是快速,一过性表达,Fos蛋白的表达滞后且延长。提示:c-fos在癫痫发病的基因机制中发挥作用,本文对此进行了讨论。本实验为深入研究癫痫发病的分子机制奠定了基础。  相似文献

一氧化氮合酶(NOS)活性与一氧化氮(NO)合成及血管功能关系密切,为了研究脑血管内皮细胞NOS表达及其调节作用,本文采用脑微血管内皮细胞培养技术和组织化学方法,观察了内皮细胞NOS的表达。未加刺激物对照组的脑微血管内皮细胞染色淡,阳性反应物主要聚集在细胞核周围胞浆中。加入肿瘤坏死因子-α(tumornecrosisfactor-α,TNF-α)或酯多糖(lipopolysaccharide,LPS)5min后,NOS染色开始增加,1h达顶峰。以后呈下降趋势。LPS比TNF-α作用时间较长,NOS染色较深  相似文献

为探讨八肽胆囊收缩素(CCk-8)和阿片肽相互作用的分子机理,利用抗体免疫沉淀技术研究了CCK-8与NDAP(k阿片受体激动剂)对大鼠脑(去皮层和小脑)和脊髓背柱组织Fos蛋白的影响。结果表明,0.1μmol/LCCK-8可显著刺激脑和脊髓组织中Fos蛋白增加(分别是对照组的3.8倍和3.6倍)。相同浓度的NDAP对Fos蛋白的生成亦有一定的诱导作用,分别是对照组的2.7倍和2.6倍。CCK-8和NDAP共同处理组织,Fos蛋白生成水平相似(脑)或高于(脊髓)CCK~-8单独诱导的水平。结果表明,CCK-8和NDAP均可直接诱导大鼠脑和脊髓组织c-fos的表达,它们对c-fos表达的相互作用在脑和脊髓中呈现不同的模式。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
