Malpighian tubules of Namib Desert tenebrionid beetles of the genus Onymacris are strongly stimulated by homogenates of the corpora cardiaca. The corpora cardiaca of other arid-adapted tenebrionids also contain diuretic material. Biogenic amines, which could be released during the preparation of corpora cardiaca extracts, do not stimulate fluid secretion in tubules of Onymacris rugatipennis. The diuretic factor in corpora cardiaca extracts is stable to boiling and to incubation with pronase. HPLC separation of the corpora cardiaca of O. rugatipennis gives a single region with diuretic activity in both secretory and electrical bioassays. Diuretic activity can not be detected in the haemolymph of Onymacris, and injection of corpora cardiaca extracts into the beetles does not cause diuresis. Simultaneous injection of corpora cardiaca and the dye amaranth shows that the most of the dye transported by the Malpighian tubules moves anteriorly into the midgut, indicating fluid recycling by this route. The most likely function for this “diuretic hormone” is clearance of metabolic wastes from the haemolymph.  相似文献

Summary Malpighian tubules of a desert tenebrionid beetle,Onymacris plana, have been studied as isolated preparations. Under control conditions tubules of female beetles secreted fluid at an average rate of 3.3 nl/min, but this rate was increased 20–25 times by a diuretic hormone (DH).Homogenates of the brain, corpora cardiaca (CC) and prothoracic ganglion induced striking increases in tubule secretion rates, which sometimes exceeded 100 nl/min. The increased rates were sustained for 3 h without renewal of the medium. Diuretic activity was also present in the other thoracic ganglia. High K treatment caused release of DH from the CC only.Exogenous cyclic AMP (1 mM) stimulated the isolated tubules ofO. plana, but to a lesser extent than the DH. The cationic composition of the secreted fluid resembled that of most other insect tubules, with high K and low Na concentrations. Stimulation with DH doubled the Na concentration.The DH was not inactivated by the tubules themselves, but was destroyed by contact with the haemolymph. An inactivation mechanism is vital in the apparently contradictory situation of a desert beetle possessing a diuretic hormone. ABSTRACT Dose-response curves are presented for the diuretic activity in aqueous extracts of brain, retrocerebral complex, and ventral nerve cord ganglia from Acheta domesticus . Diuretic activity is highest in extracts of brain and corpora cardiaca. In comparison with such extracts, those of the suboesophageal ganglia and thoracic ganglia I-III produce truncated responses, whilst abdominal ganglia 1–4 show evidence of an inhibition of the diuretic response at high doses. ED50 values, obtained from Hill plots, are similar for extracts of brain, corpora cardiaca, corpora allata, and abdominal ganglia, but are 3–4 times higher for extracts of suboesophageal and thoracic ganglia.
Separation of aqueous extracts of corpora cardiaca by reversed-phase HPLC yields a number of fractions which stimulate fluid secretion in isolated tubules. Diuretic activity in these fractions is destroyed by treatment with Pronase E, and on this basis is identified as peptidic. In general, diuretic activity is found in the same RP-HPLC fractions prepared from aqueous extracts of brain, suboesophageal ganglia, thoracic ganglia I-III, and abdominal ganglia 1–4.  相似文献

The diuresis which follows the pupal-adult ecdysis of Pieris brassicae is hormonally controlled. Use of the isolated Malpighian tubules as a bioassay shows the presence of substantial diuretic activity in homogenates of the brain and corpus cardiacum-corpus allatum complex. The hormone is probably produced in the brain and released from a storage site in the corpora cardiaca. The tubules of the butterfly are maximally responsive to the diuretic hormone at the time of eclosion.  相似文献

We have isolated a peptide from brains and corpora cardiaca of Locusta migratoria which is immunologically related to the diuretic hormone of Manduca sexta. We determined its structure as a 46 amino acid linear peptide with 43-50% identity to the M. sexta hormone. Moreover, we showed that the new peptide functions as a diuretic hormone in L. migratoria, stimulating urine production by Malpighian tubules and elevating levels of cAMP in tubules.  相似文献

In the adult males of the Colorado potato beetle, Leptinotarsa decemlineata, corpora cardiaca extract injected in vivo gives rise to an increase in glucose and a decrease in alanine concentration of the haemolymph. The regulation of proline synthesis and glucose release in the fat body of the Colorado potato beetle was investigated further in vitro. Proline regulates its own synthesis by a feedback inhibition. Moreover, a factor present in extracts from the corpora cardiaca stimulates synthesis in the fat body in vitro. This effect was demonstrated with corpora cardiaca extracts from beetles, locusts and cokroaches. Also, synthetic adipokinetic hormone stimulates proline synthesis in the fat body of the Colorado beetle. In addition, a release of glucose from the fat body of the beetle could be evoked by the addition of locust and beetle corpora cardiaca extracts or synthetic adipokinetic hormone. The physiological significance of both effects is discussed.  相似文献

The activity of the substance(s) which are contained in the cephalic endocrine organs of the locust which induce egg diapause in Bombyx mori was examined by implantation and injection of saline extracts of these organs. Extracts from the median and lateral neurosecretory parts of the locust brain were not effective in inducing egg diapause. Extracts of the corpora cardiaca, corpora allata, and suboesophageal ganglion of the locust induced diapause eggs in Bombyx pharate adults from which the suboesophageal ganglion had been removed. The first two extracts could induce egg diapause even in isolated abdomens of pharate adults of Bombyx. In the locust corpora cardiaca, the activity was present only in the glandular lobe and not in the nervous region. This activity decreased when the nervi corporis cardiaci I and II and of nervi corporis allati I were cut. Allatectomy also brought about a decrease in the activity in the glandular lobe which could not be restored by the injection of juvenile hormone. The activity in the corpora allata was enhanced slightly by the disconnection though not significantly.From these results, it is assumed that the corpora cardiaca, corpora allata and suboesophageal ganglion of the locust contain and active principle(s) capable of inducing egg diapause in Bombyx mori. The nervous connections between the brain, corpora cardiaca, and corpora allata are essential for the accumulation of the active substance(s) in the glandular lobes of the corpora cardiaca.  相似文献

A diuretic peptide (Acheta-DP) has been isolated from extracts of whole heads of the house cricket, Acheta domesticus. The native peptide increases both cyclic AMP production and the rate of fluid secretion by isolated Malpighian tubules in vitro to an extent comparable with those responses obtained with supra-maximal amounts of crude extracts of corpora cardiaca. The primary structure of Acheta-DP was established as a 46-residue amidated peptide: TGAQSLSIVAPLDVLRQRLMNELNRRRMRELQGSRIQQNRQLLTSI-NH2. Acheta-DP has 41% sequence identity with a diuretic peptide isolated from Manduca sexta, providing direct evidence for the presence of a family of diuretic peptides in insects.  相似文献

We have developed a histological technique that allows all of the cells in the malpighian tubules of Aedes taeniorhynchus (Diptera : Culicidae) and a few other mosquitoes to be counted and identified to cell type. We used this technique to examine the number and distribution of primary and stellate cells. Tubules, taken from the 4th instar larvae, pupae, adult males, and adult females, were examined. The mosquitoes were reared as larvae in either 10% or 100% seawater. In all cases examined, the total number of cells composing a tubule was somewhat variable, but no correlation between cell number and developmental stage, environmental salinity, or adult sex was observed. Under all conditions examined, stellate cells compose a fairly constant 16–18% of the total cell population of the tubule. Stellate cells were absent from the tubules in a region near the point of attachment to the midgut. Similar distribution ratios of primary and stellate cells were observed in the tubules of Aedes aegypti, Culex tansalis, and Culiseta inornata. The significance of these findings to a previously proposed tole for stèllate cells in ion transport is discussed.  相似文献

The insect arginine vasopressin-like (AVPL) peptide is of special interest because of its potential function in the regulation of diuresis. Genome sequences of the red flour beetle Tribolium castaneum yielded the genes encoding AVPL and AVPL receptor, whereas the homologous sequences are absent in the genomes of the fruitfly, malaria mosquito, silkworm, and honeybee, although a recent genome sequence of the jewel wasp revealed an AVPL sequence. The Tribolium receptor for the AVPL, the first such receptor identified in any insect, was expressed in a reporter system, and showed a strong response (EC(50)=1.5 nM) to AVPL F1, the monomeric form having an intramolecular disulfide bond. In addition to identifying the AVPL receptor, we have demonstrated that it has in vivo diuretic activity, but that it has no direct effect on Malpighian tubules. However, when the central nervous system plus corpora cardiaca and corpora allata are incubated along with the peptide and Malpighian tubules, the latter are stimulated by the AVPL peptide, suggesting it acts indirectly. Summing up all the results from this study, we conclude that AVPL functions as a monomer in Tribolium, indirectly stimulating the Malpighian tubules through the central nervous system including the endocrine organs corpora cardiaca and corpora allata. RNA interference in the late larval stages successfully suppressed mRNA levels of avpl and avpl receptor, but with no mortality or abnormal phenotype, implying that the AVPL signaling pathway may have been near-dispensable in the early lineage of holometabolous insects.  相似文献

Changes in total haemocyte counts in relation to the endocrine glands have been studied in Halys dentata. Unfixed total haemocyte counts were taken after extirpation and implantation of three endocrine structures. It is concluded that haemocyte count decreases after the extirpation of the corpora allata in the early life span of adults, while an abrupt increase was noted after the 7th day. When both the corpora allata and corpora cardiaca are extirpated, the count was significantly decreased throughout the life span of adults. However, when only the corpora cardiaca were removed there was no significant variation up to the 7th day, but later on the count gradually decreased. The implantation of brain, corpora cardiaca and allata is followed by a significant rise in the haemocyte count throughout the life span. Total haemocyte count also fluctuated after the implantation of corpora allata and brain, separately. After implantation of corpora cardiaca of one-day-old adult insects there was no variation in the total haemocyte count while the corpora cardiaca of 6-day-old adults when implanted in one-day-old adults cause a significant rise in the total haemocyte count. Therefore, it is concluded that the corpora cardiaca influences the total haemocyte count after a critical period.  相似文献

Serotonin stimulates secretion by Malpighian tubules (MT) of a number of insects, and functions as a diuretic hormone in Rhodnius prolixus and in larval Aedes aegypti. Serotonin is here shown to be a potent stimulant of secretion by MT of the house cricket, Acheta domesticus, with an apparent EC50 of 9.4 nmol L−1, although its diuretic activity is just 25% of the maximum achievable with either the native CRF-related peptide, Achdo-DH, or a crude extract of the corpora cardiaca. In this respect, the diuretic activity of serotonin is similar to that of the cricket kinin Achdo-KI, and when tested together their actions are not additive, which suggests they target the same transport process. Consistent with this suggestion, the activity of serotonin is chloride-dependent and is associated with a non-selective stimulation of NaCl and KCl transport. In common with Achdo-KI, serotonin has no effect on cAMP production by isolated MT, and both act synergistically with exogenous 8bromo-cAMP in stimulating fluid secretion, most likely by promoting the release of Ca2+ from intracellular stores. A number of serotonin agonists and antagonists were tested to determine the pharmacological profile of receptors on cricket MT. The results are consistent with the diuretic activity of serotonin being mediated through a 5-HT2-like receptor.  相似文献

The insect diuretic hormone (DH) binds to their receptor in malpighian tubules, and stimulates water secretion and cAMP synthesis. Complementary DNA encoding a diuretic hormone receptor was cloned from the malpighian tubules of Bombyx mori. The cloned cDNA encodes a protein consisting of 391 amino acid residues with the seven transmembrane domains. The receptor protein is homologous with that of other insects, and is structurally related to G-protein coupled receptors such as corticotropin relating factor (CRF), secretin, and vasoactive intestinal peptide.  相似文献

Extracts of the corpora cardiaca of the stick insect Carausius morosus elevate the haemolymph carbohydrate concentration in adult and 6th-instar larvae which are ligated behind the first pair of legs, but not in non-ligated (intact) insects. The increase in haemolymph sugars is due to trehalose elevation, is time dependent (with a maximal effect about 90–120 min after injection), and is dose dependent (needing 0.005 gland equivalents for a significant effect and a tenfold higher dose for a maximal response). The hyperglycaemic factor is localised entirely in the corpora cardiaca and appears to be specific to stick insects; corpora cardiaca extracts of two lepidopteran species (Acherontia atropos and Aglais urticae) and of Locusta migratoria have no effect, whereas corpora cardiaca extracts of the stick insects Cuniculina impigra and Sipyloidea sipylus have similar activity to those from C. morosus. This specificity is also shown when S. sipylus is used as the recipient. Synthetic adipokinetic hormone and red pigment concentrating hormone possess no hyperglycaemic activity in the stick-insect system. Two peaks of hyperglycaemic activity were obtained after column chromatography of corpora cardiaca extract of C. morosus on Sephadex G-25 and Sephadex LH-20. The factor seems to act via activation of fat-body glycogen phosphorylase, which, although 60% active in the control insects, is significantly increased to approx. 85% upon corpora cardiaca injection. However, the activation is demonstrated in ligated and intact insects. No significant decrease in the glycogen level of the fat body is observed after corpora cardiaca injection.  相似文献

Summary The amounts of adipokinetic and diuretic hormone in the separate storage and glandular lobes of the locust corpora cardiaca during the imaginal moult and up to the onset of sexual maturation have been measured. The levels of the hormones are high prior to the imaginal moult, fall at emergence and increase during the somatic growth period. The effects of surgical interference with the neuroendocrine system upon the hormonal content of the corpora cardiaca have been investigated. Cautery of the brain neurosecretory cells or allatectomy in mature locusts has no effect on the content of adipokinetic hormone. Diuretic hormone is absent from the storage lobes of locusts deprived of their cerebral neurosecretory cells but normal levels are present in the corpora cardiaca of allatectomised animals. Severance of the nervus corporis cardiacum I and II reduces the level of diuretic hormone in the storage lobes of the corpora cardiaca but is without effect on the levels of adipokinetic hormone in the glandular lobes. Low infections of Mermis nigrescens reduce the flight performance of adult male Locusta migratoria, and bring about changes in the levels of blood carbohydrate and protein but not lipid. The quantities of all three metabolites in the fat body are reduced, as is the amount of tissue present. Lipid content only, is markedly decreased in the flight muscle, but the water content is increased.

Blood volume and malpighian tubule activity are unaffected by mermithid infection, but the rates of lipid mobilisation in response to injections of extracts of corpora cardiaca or flight are greatly reduced compared with those in uninfected locusts. Additional carbohydrate does not correct flight performance in parasitised locusts as it does in starved animals. Poor flight performance is not due to an effect of the worm on the host endocrine system.  相似文献

ABSTRACT. Diuretic hormone is a peptide neurohormone of as yet unknown chemical structure. Attempts to characterize the locust hormone have concentrated on the storage lobes of the corpora cardiaca as a source of material. Bioassay data now indicates that the occurrence of diuretic activity is more widespread in the locust CNS. Dose-response curves indicate that the levels of the diuretic factor(s) in the brain, sub-oesophageal and thoracic ganglia are remarkably similar to those found in the corpora cardiaca; activity was also detected in the abdominal ganglia (1–5) and terminal abdominal ganglion. Reverse-phase HPLC has confirmed that the diuretic material found in the various parts of the CNS is chemically similar to at least one of the diuretic factors separated from the corpora cardiaca. The significance of the widespread distribution of this peptide hormone is considered.
Recent evidence of other workers has indicated the possible structural similarity between vertebrate vasopressin and the diuretic factor in the locust sub-oesophageal ganglion. Chromatographic criteria have been used to demonstrate that the diuretic hormones isolated in our work are not vasopressin-like molecules, but are chemically distinct entities.  相似文献

This paper reports the effects of different diuretic factors on the Malpighian tubules of beetles. Calcitonin (CT)-like peptides from silkmoth and mosquito increase fluid secretion in a dose-dependent manner in the tubules of Tenebrio molitor, but the cockroach CT-like peptide, Dippu-DH(31), has no effect. Thapsigargin induces a small but significant increase in tubule secretion rates. The interactions between different factors in mealworm tubules were explored by testing CT-like peptides, thapsigargin and the mealworm CRF-related diuretic factor Tenmo-DH(37) in various combinations, but no synergistic effects were observed. C-terminal fragments of the CRF-related diuretic peptides Locmi-DH(46) and Dippu-DH(46) fail to increase fluid secretion in mealworm tubules, unlike their corresponding whole peptides. Cross-reactivity of factors between beetle species was investigated using the scarabaeid Onthophagus gazella. Tenmo-DH(37) increases fluid secretion in isolated tubules of O. gazella in a dose-dependent manner, revealing a high degree of cross-reactivity in this distantly related beetle species. However, homogenates of O. gazella brains inhibited fluid secretion in mealworm tubules.  相似文献

In the Malpighian tubules of Acheta, the distal and middle segments are functionally and morphologically quite distinct (Spring and Kim, Mol Comp Physiol 12:130–145, 1993). Furthermore, they respond quite differently to corpora cardiaca (CC) homogenates, dibutyryl cAMP, and A23187 (Kim and Spring, J Insect Physiol 38:373–381, 1992). In this study we compared secretion by these two regions in response to Acheta and Romalea CC extracts, synthetic Manduca sexta diuretic peptide (Mas-DP1), and the family of synthetic myotropic peptides, the achetakinins, isolated from Acheta. Both Acheta and Romalea CC extracts had opposite effects on the two regions: mid-tubule secretion increased 3-fold whereas secretion by the distal segment declined 75–80%. Mas-DP1 increased secretion by the mid-tubule more than 3-fold and had no effect on the distal segment. All of the achetakinins decreased secretion by the distal tubule, with achetakinin 1 being least effective (55% inhibition) and achetakinin 5 being most effective (75% inhibition). Achetakinins 1 and 2 increased midtubule secretion by 3.7- and 3.3-fold, respectively, whereas the others had no effect on this region. Regarding HPLC fractions of CC extracts, in general the more hydrophilic fractions inhibited secretion by both distal and mid-tubules. The more hydrophobic fractions were nearly uniformly stimulatory when applied to the mid-tubule, and either inhibited secretion or had no effect on the distal region. Hindgut stimulating neurohormone (HSN) was synthesized and stored by cultured brains of the cockroach, Leucophaea maderae. The presence of either HSN or cultured corpora cardiaca in the medium caused the brains to release their accumulated HSN. The corpora cardiaca were able to sequester HSN from the medium when the concentration was above a threshold level. Thus, the corpus cardiacum may provide an homeostatic mechanism for maintaining physiological levels of neurohormone.  相似文献
