This paper describes the ultrastructure of the seminal vesicle and the isoelectric focusing patterns of its secretion during sexual maturation and after allatectomy in Melanoplus sanguinipes (Fabr.) (Orthoptera : Acrididae). In epithelia from seminal vesicles of newly fledged males, the rough endoplasmic reticulum is well developed, and Golgi complexes are elaborate, which indicates the gland is metabolically active. The cells also contain large glycogen deposits and the lumen microvilli are well differentiated. These ultrastructural features are more dominant in 24-hr-old adults where the cytoplasm is clearly differentiated into basal and apical regions. Basally, the cytoplasm is dominated by rough endoplasmic reticulum, large Golgi complexes, glycogen deposits and numerous mitochondria, while the apical cytoplasm is filled with large secretory and/or lysosomal vesicles. Between days 3 and 7, the ultrastructural features change little other than the rough endoplasmic reticulum cisternae, which become vesicular. Analysis by isoelectric focusing shows that the amount of secretory protein increases with age until day 3, at which time the gland contains its full complement of secretion. In seminal vesicles from allatectomized insects, ultrastructural features of cells and isoelectric focusing patterns of the secretion arc identical to those from normal males. 相似文献

The visual behavior of adult Melanoplus sanguinipesF. (Orthoptera: Acrididae) was investigated by placing individuals in the center of an arena and recording their orientation responses to visual targets at the perimeter of the arena. Targets that reflected more 540- to 570-nm light were approached more frequently; however, when reflectance in the 540 -to 570-nm region was combined with reflectance in the 400- to 520-nm region, orientation responses were reduced significantly. This suggests that spectral discrimination in M. sanguinipesinvolves at least two classes of photoreceptors, which respond to different regions of the wavelength spectrum. In addition, grasshoppers oriented to vertical, but not horizontal, contrasting stripes. However, when vertical stripes were added to targets reflecting 520- to 650-nm light, responses to verticals on these targets were not enhanced relative to verticals presented against a target background of 400- to 520-nm + 520- to 650-nm light. Thus, spectral discrimination and vertical stripe fixation appear to be two distinct visual behaviors, controlled by outputs from two classes of photoreceptors and a single class of photoreceptors, respectively, and may be used in different physiological or ecological contexts. 相似文献

The ejaculatory duct of the migratory grasshopper (Melanoplus sanguinipes [Fabr.]) (Orthoptera : Acrididae) is divisible into 3 regions: upper ejaculatory duct (UED) into whose anterior end the accessory glands and vasa deferentia empty; the funnel characterized by its slit-like lumen; and the lower ejaculatory duct (LED). Anteriorly, the UED has a keyhole-shaped lumen surrounded by a thin intima and highly columnar epithelial cells whose most conspicuous feature is massive aggregations of microtubules. More posteriorly, the UED lumen differentiates into dorsal and ventral chambers, the former having a thick cuticular lining armed with spines. In the hindmost part of the UED, the ventral chamber expands to obliterate the dorsal chamber; its cuticular lining thickens, and conspicuous lateral evaginations develop. The thick cuticle includes 3 distinct layers and on its surface carries numerous spatulate processes. In this region, the epithelial cells develop numerous short microvilli beneath which are many mitochondria. As the funnel is reached, the intima becomes extremely thick, and the epithelial cells lack microvilli and most microtubules. Within the funnel, a new, very distinct form of cuticle appears, which is in “units”, each associated with an epithelial cell and having a rounded epicuticular cap. The new cuticle arises ventrally but rapidly spreads to encircle the entire lumen, at which point the LED is considered to begin. Beneath this new cuticle, the epithelial cells are columnar, have long microvilli, numerous mitochondria in the apical cytoplasm, and rough endoplasmic reticulum basally. Apically, adjacent cells are tightly apposed; however, prominent intercellular channels develop more basally. Neurosecretory cells of only one type (A, sub type A2) are seen in adult Melanoplus. Two groups of about 400 cells each are located dorsally in the pars intercerebralis medialis; four cells are located deep within the protocerebrum. We found no neurosecretory cells in other parts of the central or sympathetic nervous systems. In about 10% of the specimens, there was marked asymmetry in the location of the dorsal cell groups, with both of these groups and their axons located in one lobe of the protocerebrum. The nervi corporis cardiaci 1 cross-over in the corpus cardiacum, with the result that material produced by neurosecretory cells on one side of the brain is transported along axons that undergo two chiasmata to the corpus cardiacum of the same side. Stainable secretory material could be traced clearly from the cerebral cells to the corpus cardiacum, and even into the oesophageal nerves from the hypocerebral ganglion. However, stainable neurosecretory material is never present in the corpus allatum or along any of the nerves to this gland. 相似文献

An oviposition-stimulating protein (OSP) was isolated and purified from the long hyaline tubules of the male accessory gland complex in the migratory grasshopper, Melanoplus sanguinipes. Gel filtration of the native OSP, using Sephadex G-100, indicates its molecular weight to be about 60000Da with the oviposition-stimulating activity while sodium dodecyl sulphate-polyacrylamide gel electrophoresis shows that the OSP comprises two subunits, each with a molecular weight of 30000Da. The purified OSP appears as a single symmetric peak on fast performance liquid chromatography using Mono Q. Isoelectric focusing of the OSP indicates an apparent pI of 5.5. Injection of the OSP induces oviposition in about 70% of ovulated virgin females within 48h. Stimulation of oviposition can be blocked by a polyclonal antibody raised against the OSP 30000Da subunits. Amino acid analysis of the dimer and its subunits shows a comparatively high content of aspartic acid/asparagine (14.8%) as well as leucine (12.2%) and glutamic acid/glutamine (12.0%). The N-terminal 21 amino acid sequence of the OSP shows little similarity to known peptides. Immunoreactivity with the anti-OSP antibody was observed in the viscous secretion, spermatheca, and the egg-pod froth of mated females, confirming transfer of the OSP from male to female during copulation. 相似文献

ABSTRACT. Discharge from the male accessory reproductive gland (ARG) by the male grasshopper, Melanoplus sanguinipes (Fabr.), has been studied by assay of a characterized product, a glycoprotein LHPI, and the rate of formation of spermatophores. LHPI is an exclusive long hyaline gland (a prominent ARG tubule type) product whose discharge is symmetrical from the bilaterally paired glands. LHPI forms 65% of a viscous secretion that is discharged concomitantly with the spermatophores. Though low ARG reserves in 5-day-old males limit both the number of spermatophores formed and LHPI discharged in copulations ≤6 h, this appears to be the result of shorter copulations. The number of spermarophores formed in 1 h was not impaired by general depletion of ARG protein (by repeated copulations) or by selective depletion of long hyaline gland protein (by unilateral and bilateral long hyaline gland removal), though these manipulations reduced LHPI discharge by 22%, 44% and 100%, respectively. However, 56% of spermatophores formed by males with the long hyaline gland bilaterally ablated failed to uncoil properly. These results indicate LHPI and/or other long hyaline gland proteins may act as lubricants. Unlike spermatophore formation and LHPI discharge, which increased steadily up to 90–120 min then levelled off, transfer of radiolabelled male ejaculate to the spermatheca was very variable. In 90 min copulations, only 1% of the total radioactivity (representing c. 5 μg protein) lost from the ARG complex was transferred to the spermatheca. The importance of male-derived protein in vitellogenesis is discussed. 相似文献

The accessory reproductive glands of Melanoplus sanguinipes comprise two bilateral masses of 16 tubules each, distinguishable in sexually mature insects as four white, ten short hyaline, one long hyaline, and a seminal vesicle. Over most of its length, the wall of each tubule consists of a simple glandular epithelium resting on a basal lamina, surrounded by a thin layer of circular muscle. However, near the junction with the ejaculatory duct, the wall of each tubule has a much thickened circular muscle layer and squamous or cuboidal epithelium, the region serving to regulate movement of secretion into the ejaculatory duct. Interdigitation of adjacent epithelial cells is common, and several kinds of specialized junctions occur. In the glandular region, all epithelial cells appear the same and may be flattened, cuboidal, or columnar depending on the tubule type. Except for those of the seminal vesicle, the glandular epithelial cells share ultrastructural features typical of cells engaged in the synthesis of protein for export. Despite these general similarities, in most instances subtle differences occur in the cellular ultrastructure of the epithelia of each tubule and in the appearance of their luminal secretions, suggesting that the tubules are functionally specialized. 相似文献

Three insecticidal acetylchromenes, including encecalin, and precocene II were topically administered to adults of the migratory grasshopper Melanoplus sanguinipes. Metabolites formed and excreted via the frass were identified by gas chromatography-mass spectrometry (GC-MS) and by direct comparison with reference compounds obtained by partial synthesis or from plant sources. Of the 12 metabolites found, six compounds were new natural products. Their tentative structure elucidation is described based on MS analysis. The biological importance of the elucidated chromene metabolism in M. sanguinipes is discussed for the excretion and detoxification of potentially hazardous compounds. 相似文献

Microbial Ecology of the Gut in Laboratory Stocks of the Migratory Grasshopper, Melanoplus sanguinipes (Fab.) (Orthoptera: Acrididae) 总被引:1,自引:1,他引:0 下载免费PDF全文
Mean pH values in pooled samples of foregut, midgut, and hindgut from adult Melanoplus sanguinipes, which had been raised in the laboratory on barley shoots and wheat bran, were 5.15, 6.39, and 5.98, respectively. Homogenates of midgut/hindgut sections and frass (feces) yielded colony counts of bacteria by the spread plate method of 5.7 to 5.9 and 5.3 to 5.5 log10 colonies per mg, respectively; there were no significant differences (P > 0.05) between counts obtained on several media or on media incubated aerobically or anaerobically. There was no evidence of significant populations of protozoa, fungi, or obligately anaerobic bacteria associated with the gut. A total of 168 pure strains of bacteria isolated from the gut sections were characterized and assigned to 11 taxonomic groups, including Enterococcus spp., Serratia liquefaciens, Pseudomonas spp., and Enterobacter spp. Numbers of Enterococcus spp. in the gut were 2 to 3 orders of magnitude higher than those of the other genera. Strains representing only four of the groups were recovered from bran fed to the grasshoppers; the barley shoots, which were raised in sterile soil, appeared virtually sterile. Examination of the gut wall by scanning electron microscopy revealed the presence of epimural bacteria in the foregut and hindgut but not in the midgut. The distribution of epimural cocci and bacilli differed with the gut section examined. Numerous spherical to ovoid structures up to 10 μm in diameter, which were not identified, were associated with the microvillous surface of the midgut epithelium. Acetate was present in gut, hemolymph, and frass, and it was shown that representative isolates of Enterococcus spp. and Enterobacter agglomerans produced acetate when incubated in an aqueous suspension of bran. The egestion time of solid digesta, as measured with methylene blue-stained barley shoots, was 3.0 to 5.7 h. The results show that M. sanguinipes supported extensive indigenous populations of luminal and epimural bacteria in the gut which were composed predominantly of facultatively anaerobic species; the relatively short egestion time, indicating rapid passage of digesta through the gut, was consistent with the microscopic appearance of digesta residues in frass and could account, at least in part, for the absence of a significant population of obligately anaerobic bacteria from the gut. 相似文献

Protein production during sexual maturation or after allatectomy (followed by juvenile hormone replacement therapy) has been examined in the long hyaline glands, short hyaline glands, white glands, and seminal vesicles, which make up the accessory gland complex of male Melanoplus sanguinipes. During maturation, the amount of protein in the long hyaline glands increases about 14-fold, and in each of the other components between 5- and 6-fold. Most protein accumulates between days 3 and 5, and this is reflected in high levels of incorporation of radiolabelled leucine in this period. The components show differential sensitivity to the effects of allatectomy. After this operation, the protein content of, and incorporation of radiolabel into, the long hyaline glands remain near the day 0 level. In the white glands and short hyaline glands, allatectomy also has a marked, though less severe, effect on protein synthesis and accumulation. The seminal vesicles are least affected by allatectomy and continue to accumulate protein (though more slowly) to about 60% of the normal level by day 10. Juvenile hormone compounds applied topically to allatectomized insects on day 2 restored the ability of the gland components to accumulate proteins, though to differing degrees. JHI is the most effective compound, stimulating synthesis and accumulation of protein to near normal levels by day 10, whereas application of JHIII or Stauffer's synthetic JH led to only partial restoration of protein synthesis in the glands. 相似文献

Abstract. Durations of tethered flights by the North American migratory grasshopper Melanoplus sanguinipes Fabricius are bimodally distributed: most individuals either will not fly, or else will fly for many hours. This observation suggests a simple measure (the ‘one‐hour rule’) for distinguishing migrants from non‐migrants. This measure is repeatable (repeatability = 0.6–0.7). Using laboratory‐reared offspring of grasshoppers from an Arizona population of mixed migratory tendency, a breeding experiment was conducted to determine the heritability of migratory tendency and possible correlated responses to selection on migratory behaviour. When migratory tendency is considered as a threshold trait, the heritability of liability is in the range 0.5–0.6. Most families in the breeding experiment had at least some migrants among their offspring; selection on migratory incidence had a correlated effect on the durations of flights by these individuals. The magnitude of thoracic lipid reserves showed a modest correlated response to selection on migratory behaviour. Thoracic and abdominal lipid reserves in identified migrants are reduced by flight, indicating that lipid is mobilized and consumed during flight in this species. 相似文献

The isolated oviduct of the migratory grasshopper, Melanoplus sanguinipes, shows a myogenic, rhythmic pattern of contraction. The pattern of contraction can be modified by treatment with hemolymph or extracts from a variety of tissues from animals of differing age, sex, and mated status. Extracts of oviducts and ventral nerve cord (VNC), as well as hemolymph, from both virgin and mated females were almost always stimulating, though their effect on frequency, amplitude and/or tonus varied. In contrast, whereas extracts of spermatheca and brain from mated females were stimulating, extracts of these tissues from virgin females inhibited contraction. All male material tested (long hyaline tubules (LHT), the male accessory gland complex less the LHT, testes, brain, VNC and hemolymph) stimulated oviduct contraction in a dose-dependent manner, usually enhancing at least two of frequency, amplitude and tonus. However, oviposition-stimulating protein, a purified product of the LHT, provoked only an increase in frequency when applied to the oviduct. Abstract. The North American migratory grasshopper Melanoplus sanguinipes Fabricius (Orthoptera: Acrididae) exhibits heritable variation in predisposition to make long-duration flights, and performance of long-duration flight enhances reproductive output. As a first step in understanding the physiological basis of these phenomena, we examined the mobilization of lipid and carbohydrate reserves during flight and in response to injection of extracts of the corpora cardiaca. Extract of conspecific corpora cardiaca elevates the concentration of haemolymph lipid. Both synthetic locust adipokinetic hormone I (AKH I) and synthetic Locusta migratoria AKH II raise the concentration of lipid in the haemolymph. However, although AKH I is more active than AKH II in locusts, dose-response curves for the two peptides are similar in M.sanguinipes. Neither extract of conspecific corpora cardiaca nor locust AKH I affects haemolymph carbohydrate in this species. Haemolymph carbohydrate and total glycogen reserves are Diminished by tethered flight; in contrast, haemolymph lipid is elevated by flight. Grasshoppers identified as presumptive migrants or non-migrants do not differ significantly in body composition. Total lipid reserves did not decrease measurably after extended flight, even though total reserves of carbohydrate do not appear to be sufficient to maintain the durations of flight performed. 相似文献

Laboratory and field evaluations of imidacloprid against Melanoplus sanguinipes (Orthoptera: Acrididae) on small grains 总被引:1,自引:0,他引:1
The toxicity of imidaloprid to the migratory grasshopper, Melanoplus sanguinipes (F.), was measured in bioassays, greenhouse trials, and field trials. An LD50 of 53 and 86 ppm for the oral/topical applications of imidacloprid confirmed a low toxicity for this chemical when compared with carbofuran as a standard. However, 100% debilitation was observed at concentrations of > or = 1 ppm. Grasshoppers exhibited leg flexing, abdominal quivering, and tremors before becoming motionless and appearing dead. Knockdown was temporary with a high percentage of recovery within 1 h. Efficacy and feeding damage were determined from artificial infestations of M. sanguinipes at the 2nd, 4th, and early tillering growth stages of winter and spring wheat treated with foliar and seed treatments of imidacloprid. All rates of imidacloprid tested resulted in < 45% mortality to 4th instar and adult M. sanguinipes in the greenhouse and field. Although efficacy was low, high rates of debilitation in bioassays suggest that improved control may be gained by combining imidacloprid with insect pathogens or additional chemicals. 相似文献

Abstract The relationship between corpus allatum (CA) regulation of copulatory behaviour and the CA's influence on the development of the accessory reproductive glands was studied in the male grasshopper, Melanoplus sanguinipes (Fabr.). Allatectomy slowed the development of copulatory behaviour; ≤80% of control males 7 days and older mated with females, but allatectomized males did not show comparable levels of copulation until day 19. In a 3 h observation period, the time taken for 50% of males to initiate copulation was c. 60 min for allatectomized males and only c. 10 min for controls. At the age when 50% of males copulated, the accessory glands of allatectomized males were less developed than those of controls; the accessory gland weight was 65%, soluble protein was 72% and reserves of long hyaline protein I were 40% those of controls. Treatment with Juvenile Hormone III on day 2 stimulated accumulation of long hyaline protein I in 9-day-old allatectomized males to the level found in untreated 19-day-old allactec-tomized males, but did not elicit heightened copulation. No positive feedback from the accessory glands appeared necessary to elicit copulation since vasectomy, ablation of the accessory glands or isolation of the terminal abdominal ganglion by ventral nerve cord transection did not inhibit copulatory behaviour in 7-day-old males. Because allatectomy inhibited copulatory behaviour in 7-day-old males in which the accessory glands were removed but not in 19-day-old males without accessory glands, the increased tendency of older allatectomized males to mate with females was not due to release of inhibition caused by continued growth of the accessory reproductive glands. 相似文献
