On the metabolism of N-acetyldopamine in Periplaneta americana 总被引:1,自引:0,他引:1
N-acetyldopamine is rapidly and extensively converted to N-acetyldopamine 3-O-phosphate and N-acetyldopamine 3-O-sulphate in the newly ecdysed cockroach, Periplaneta americana. Dopamine 3-O-sulphate also serves as a naturally occurring precursor of N-acetyldopamine 3-O-sulphate in vivo.Radioisotope experiments revealed that the N-acetyldopamine moiety of these esters is incorporated into the cuticle during sclerotization and that the phosphate and sulphate moieties are not (not, at least, as the intact esters). It is suggested that N-acetyldopamine 3-O-phosphate and N-acetyldopamine 3-O-sulphate may be the forms in which the eventual cuticular sclerotizing agent (N-acetyldopamine?) is transported from the blood into the epidermis by a ‘carrier’ protein. 相似文献

We present the phospholipid fatty acid compositions, determined by GLC,* of four individual tissues (testes, mushroom glands, conglobate glands, and salivary glands) from adult male cockroaches, P. americana. The testes phospholipids contained higher proportions of palmitic acid than did the exocrine tissues (16% vs about 8%). This was also true for palmitoleic (2.3% vs 0.4–1.3%), oleic (49% vs 30–35%), and linolenic acids (7% vs 0.5–2.8%). Testicular extracts were lower in linoleic acid (16%) than were the exocrine gland extracts (at 40–45%). All four tissues also contained low proportions of several long-chain polyunsaturated fatty acids of both omega-3 and omega-6 metabolic families, including C20:3n6, C20:4n6, and C20:5n3. The data suggest that the phospholipid fatty acid profiles of specific tissues differ from each other and from those obtained from whole-insect extracts. The presence of several polyenoics from the n3 and n6 metabolic families is interpreted in the context of complex fatty acid requirements at the tissue level. 相似文献

Selective Depletion of Dopamine, Octopamine, and 5-Hydroxytryptamine in the Nervous Tissue of the Cockroach (Periplaneta americana) 总被引:1,自引:1,他引:0
High-performance liquid chromatography with electrochemical detection was used to measure the concentrations of 3,4-dihydroxyphenylethylamine (dopamine), 5-hydroxytryptamine (5-HT), p-hydroxyphenylethanolamine (octopamine), alpha-methyl-p-tyrosine, and tryptophan in the cerebral ganglia of cockroaches (Periplaneta americana) after peripheral administration of alpha-methyl-p-tyrosine and alpha-methyltryptophan. In addition, the levels of dopamine, 5-HT, octopamine, alpha-methyl-p-tyrosine, and tryptophan were determined after injection of alpha-methyl-p-tyrosine, 6-hydroxydopamine, or 5,7-dihydroxytryptamine directly into the cerebral ganglia by means of microinjection needles. Peripheral administration of alpha-methyl-p-tyrosine (400-1,600 micrograms/insect) caused a reduction in dopamine and 5-HT concentrations in cockroach cerebral ganglia, although the reduction in dopamine concentrations was more pronounced. Peripheral injections of alpha-methyl-p-tyrosine also reduced octopamine levels in the cerebral ganglia. Peripheral injection of alpha-methyltryptophan (400-1,600 micrograms/insect) caused a marked reduction in 5-HT and tryptophan concentrations in cockroach cerebral ganglia without altering dopamine or octopamine concentrations. Central injections of alpha-methyl-p-tyrosine (80 micrograms/insect) reduced dopamine concentrations in the cerebral ganglia. However, neither 6-hydroxydopamine (20 micrograms/insect) nor 5,7-dihydroxytryptamine (20 micrograms/insect) caused reductions in amine levels when applied near or directly into the cerebral ganglia. The results suggest that specific lesions of aminergic neurons in insects by either 6-hydroxydopamine or 5,7-dihydroxytryptamine are impractical. The specific, long-lasting depletion of 5-HT by alpha-methyltryptophan suggests that this chemical may be useful in elucidating the functions of 5-HT in insects. 相似文献

A comparison of the escape behaviors of the cockroaches Blaberus craniifer and Periplaneta americana
A comparison of wind-mediated escape behavior of two species of cockroaches revealed striking differences. In contrast to Periplaneta americana, Blaberus craniifer was unable to escape from a live predator or move away from a synthetically generated puff of wind. Other behavioral differences included degree of preference for areas of low light intensity and propensity to dig and thus bury oneself when disturbed. Anatomical differences, although present, did not seem sufficient to account for the behavioral differences. Also, B. craniifer was able to escape from generated wind puffs but not a live predator when its temperature was raised. These points suggest that the behavioral differences in escape behavior can be attributed to differences in the physiological state of the two nervous systems. 相似文献

The concentrations of tryptophan, 5-hydroxytryptophan, 5-hydroxytryptamine, N-acetyl 5-hydroxytryptamine and N-acetyl dopamine were determined in the cerebral ganglia, haemolymph and Malpighian tubules of the cockroach Periplaneta americana, using high performance liquid chromatography with electrochemical detection. Injected 5-hydroxytryptamine was rapidly removed from the haemolymph with a concomitant elevation of circulating N-acetyl 5-hydroxytryptamine and little accumulation of 5-hydroxytryptamine in the cerebral ganglia. N-acetyl 5-hydroxytryptamine and N-acetyl dopamine were also rapidly removed from the haemolymph. Incubation of haemolymph from 5-hydroxytryptamine-injected insects and glucosidase or phosphatase, indicated that most of the injected 5-hydroxytryptamine had been converted to a sugar conjugate of N-acetyl 5-hydroxytryptamine. Whole haemolymph did not catabolize 5-hydroxytryptamine or N-acetyl 5-hydroxytryptamine whereas Malpighian tubules N-acetylated both 5-hydroxytryptamine and dopamine and metabolized N-acetyl 5-hydroxytryptamine. Injection of p-chlorophenylalanine (200 and 500 μg/g) had no effect on 5-hydroxytryptamine concentrations in the cockroach cerebral ganglia. 相似文献

Summary The ocelli of Periplaneta americana were studied by light and electron microscopy. The view that the ocellus of the cockroach represents a degenerated structure can no longer be supported. All organelles necessary for function are present.The club-shaped retinular cells lie homogeneously distributed in the cupule of the ocellus. Rhabdoms are seen as sickle-, y-, x-or star-shaped structures with up to six cells in formation. Cells were found which had formed two rhabdomeres.The mass of cell organelles lies in the cytoplasm between the cell nucleus and the rhabdom. Smooth endoplasmic reticulum is wound into a spindle formation of considerable size at the origin of the axon in some cells. A cylindrical body in which 10–40 microtubules are packed, as yet unknown in insect retinular cells, is described.The receptory area of the ocellus terminates in a tapetum which contains granules, soluble in alcohol. Previously, we purified Periplaneta lectin from the hemolymph of adult Periplaneta americana (American cockroach) (Kubo and Natori Eur. J. Biochem. 168, 75–82, 1987). Immunoblotting analysis using antibody against Periplaneta lectin showed that the cockroach hemolymph contains another lectin that cross reacted immunologically with Periplaneta lectin. We have purified this new lectin (regenectin) to homogeneity. Affinity purified antibody against regenectin cross reacted with Periplaneta lectin. Thus, Periplaneta lectin and this new lectin were found to be different lectins sharing common antigenicity. After leg amputation, this new lectin was found to appear transiently at a specific stage of regeneration as revealed by immunoblotting, suggesting that it plays a role in the regeneration process. 相似文献

Changes in the frequency of cardiac pulsations have been monitored in the decapitated body of adult P. americana before and 5 h after the injections of [Arg(7)]-corazonin and CCAP, using newly invented touch-free, noninvasive optocardiographic methods. Relatively large dosages of these peptides (10(-6) M concentrations in the body) had no effect on the rate of the heartbeat beyond the Ringer control limits. It has been concluded, therefore, that Corazonin and CCAP, which are currently cited in the literature as "the most potent cardiostimulating peptides" in insects, have no effect on the physiological regulation of cardiac functions in the living body. 相似文献

Poly(ADP-ribosylation) is a post-translational modification of nuclear proteins typical of most eukaryotic cells. This process participates in DNA replication and repair and is mainly regulated by two enzymes, poly(ADP-ribose) polymerase, which is responsible for the synthesis of polymers of ADP-ribose, and poly(ADP-ribose) glycohydrolase, which performs polymer degradation. The aim of this work was to investigate in the cockroach Periplaneta americana L. (Blattaria: Blattidae) the behaviour of poly(ADP-ribosylation). In particular, we addressed: (i) the possible modulation of poly(ADP-ribosylation) during the embryonic development; (ii) the expression of poly(ADP-ribose) polymerase and glycohydrolase in different tissues; and (iii) the role of poly(ADP-ribosylation) during spermatogenesis. In this work we demonstrated that: (i) as revealed by specific biochemical assays, active poly(ADP-ribose) polymerase and glycohydrolase are present exclusively in P. americana embryos at early stages of development; (ii) an activity carrying out poly(ADP-ribose) synthesis was found in extracts from testes; and (iii) the synthesis of poly(ADP-ribose) occurs preferentially in differentiating spermatids/spermatozoa. Collectively, our results indicate that the poly(ADP-ribosylation) process in P. americana, which is a hemimetabolous insect, displays catalytical and structural features similar to those described in the holometabolous insects and in mammalian cells.
