Genetic manipulation of plant volatile emissions is a promising tool to enhance plant defences against herbivores. However, the potential costs associated with the manipulation of specific volatile synthase genes are unknown. Therefore, we investigated the physiological and ecological effects of transforming a maize line with a terpene synthase gene in field and laboratory assays, both above‐ and below ground. The transformation, which resulted in the constitutive emission of (E)‐β‐caryophyllene and α‐humulene, was found to compromise seed germination, plant growth and yield. These physiological costs provide a possible explanation for the inducibility of an (E)‐β‐caryophyllene‐synthase gene in wild and cultivated maize. The overexpression of the terpene synthase gene did not impair plant resistance nor volatile emission. However, constitutive terpenoid emission increased plant apparency to herbivores, including adults and larvae of the above ground pest Spodoptera frugiperda, resulting in an increase in leaf damage. Although terpenoid overproducing lines were also attractive to the specialist root herbivore Diabrotica virgifera virgifera below ground, they did not suffer more root damage in the field, possibly because of the enhanced attraction of entomopathogenic nematodes. Furthermore, fewer adults of the root herbivore Diabrotica undecimpunctata howardii were found to emerge near plants that emitted (E)‐β‐caryophyllene and α‐humulene. Yet, overall, under the given field conditions, the costs of constitutive volatile production overshadowed its benefits. This study highlights the need for a thorough assessment of the physiological and ecological consequences of genetically engineering plant signals in the field to determine the potential of this approach for sustainable pest management strategies.  相似文献

The alarm response of aphids to (E)‐β‐farnesene was examined for 59 species in the subfamilies Aphididae, Callaphididae, Chaitophoridae and Lachnidae, in the field. Forty‐one aphid species responded to (E)‐β‐farnesene at a dosage of 1 ng and/or 10 ng; however, 18 species did not. Myrmecophilous aphids did not react as extensively or strongly as non‐myrmecophilous aphids. The response of aphids to (E)‐β‐farnesene was related to host‐plant species.  相似文献

The western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte, is a destructive pest of maize, Zea mays L. in North America and Europe. Larvae and pupae can be controlled with entomopathogenic nematodes (EPN) of the genus Heterorhabditis. When maize is attacked by WCR, the roots of some maize varieties emit (E)‐β‐caryophyllene that attract EPN to the pest larvae. To use synthetic (E)‐β‐caryophyllene in bioassays for the genetic selection of EPN strains with enhanced chemotactic response to a volatile emitted from the damaged root when attacked by larvae of the WCR, different laboratory bioassays were tested. Three sand assays and one agar assay used synthetic (E)‐β‐caryophyllene as an attractant for H. megidis. In none of the assays, attraction of the nematodes to (E)‐β‐caryophyllene was recorded. Possible reasons why (E)‐β‐caryophyllene was not attracting EPN in the bioassays are discussed.  相似文献

The chemical composition of 45 essential oil samples isolated from the leaves of Polyalthia oliveri harvested in three Ivoirian forests was investigated by GC‐FID (retention indices measured on two columns of different polarities), and by 13C‐NMR, following a method developed in our laboratory. In total, 41 components were identified. The content of the main components varied drastically from sample to sample: (E)‐β‐caryophyllene (1.2 – 50.8%), α‐humulene (0.6 – 47.7%), isoguaiene (0 – 27.9%), alloaromadendrene (0 – 24.7%), germacrene B (0 – 18.3%), δ‐cadinene (0.4 – 19.3%), and β‐selinene (0.2 – 18.5%). The analysis of six oil samples selected in function of their chromatographic profiles is reported in detail. The 45 oil compositions were submitted to hierarchical cluster and principal components analysis, which allowed the distinction of three groups within the oil samples. The compositions of the oils from group I (15 samples) and II (12 samples) were dominated by (E)‐β‐caryophyllene and α‐humulene, respectively. Oil samples of group III (18 samples) needed to be partitioned into four subgroups III.1–III.4 whose compositions were dominated by alloaromadenrene, isoguaiene, germacrene B, and δ‐cadinene, respectively.  相似文献

Attraction of codling moth males to apple volatiles   总被引:4,自引:0,他引:4
The attraction of the codling moth, Cydia pomonella, to apple volatile compounds known to elicit an antennal response was tested both in the field and in a wind tunnel. In the field, (E)‐β‐farnesene captured male moths. The addition of other apple volatiles, including (E,E)‐α‐farnesene, linalool, or (E,E)‐farnesol to (E)‐β‐farnesene did not significantly augment trap catch. Few females were caught in traps which also caught male moths, but female captures were not significantly different from blank traps. In the wind tunnel, males were attracted to (E,E)‐farnesol, but not to (E)‐β‐farnesene. The addition of (E,E)‐α‐farnesene to (E)‐β‐farnesene had a synergistic effect on male attraction. The male behavioural sequence elicited by plant volatiles, including upwind flight behaviour, was indistinguishable from the behaviour elicited by sex pheromone.  相似文献

Parasitoids use herbivore‐induced plant volatiles (HIPVs) to locate their hosts. However, there are few studies in soybean showing the mechanisms involved in the attraction of natural enemies to their hosts and prey. The objective of this study was to evaluate the influence of volatile organic compounds (VOCs) of soybean, Glycine max (L.) Merr. (Fabaceae) (cv. Dowling), that were induced after injury caused by Euschistus heros (Fabricius) (Hemiptera: Pentatomidae), on the searching behavior of the egg parasitoid Telenomus podisi Ashmead (Hymenoptera: Scelionidae). Four HIPVs from soybean, (E,E)‐α‐farnesene, methyl salicylate, (Z)‐3‐hexenyl acetate, and (E)‐2‐octen‐1‐ol, were selected, prepared from standards at various concentrations (10?6 to 10?1 m ), and tested individually and in combinations using a two‐choice olfactometer (type Y). Telenomus podisi displayed a preference only for (E,E)‐α‐farnesene at 10?5 m when tested individually and compared to hexane, but they did not respond to the other compounds tested individually at any concentration or when combinations of these compounds were tested. However, the parasitoids stayed longer in the olfactometer arm with the mixture of (E,E)‐α‐farnesene + methyl salicylate at 10?5 m than in the arm containing hexane. The results suggest that (E,E)‐α‐farnesene and methyl salicylate might help T. podisi to determine the presence of stink bugs on a plant. In addition, bioassays were conducted to compare (E,E)‐α‐farnesene vs. the volatiles emitted by undamaged and E. heros‐damaged plants, to evaluate whether (E,E)‐α‐farnesene was the main cue used by T. podisi or whether other minor compounds from the plants and/or the background might also be used to locate its host. The results suggest that minor volatile compounds from soybean plants or from its surroundings are involved in the host‐searching behavior of T. podisi.  相似文献

We investigated the effect of prohydrojasmon [propyl (1RS,2RS)‐(3‐oxo‐ 2‐pentylcyclopentyl) acetate] (PDJ) treatment of intact corn plants, on their attractiveness to the specialist endoparasitoid, Cotesia kariyai Watanabe (Hymenoptera: Braconidae), and on the performance of the common armyworm, Mythimna separata (Walker) (Lepidoptera: Noctuidae) under laboratory conditions. Attractiveness of C. kariyai to PDJ‐treated plants was studied in a wind tunnel, whereas performance of M. separata larvae was tested in plastic cages. The attractiveness of the treated plants increased with concentrations of PDJ increasing to 2 mm , which was equivalent to the attractiveness of host‐infested plants. PDJ‐treated corn plants emitted 16 volatile compounds (α‐pinene, β‐myrcene, (Z)‐3‐hexenyl acetate, limonene, (E)‐β‐ocimene, linalool, (E)‐4,8‐dimethyl‐1,3,7‐nonatriene, (+)‐cyclosativene, ylangene, (E)‐β‐farnesene, (E, E)‐4,8,12‐trimethyl‐1,3,7,11‐tridecatetraene, α‐bergamotene, γ‐cadinene, δ‐cadinene, α‐muulolene and nerolidol), most of which were observed in the headspace of host‐infested corn plants with some quantitative and qualitative differences. We also tested the effects of PDJ treatment on the performance of M. separata larvae. The survival rates of the larval and pupal stages were significantly lower at 2 mm level of PDJ. A significant decrease in weight at 6th stadium larvae was observed only at 2 mm level of PDJ. In contrast, PDJ treatment at all PDJ concentration levels caused significant reduction in weight of pupal stage as compared to control. These data suggested that PDJ, originally developed as a plant growth regulator, especially to induce coloring of fruits, has the potential to induce direct and indirect defenses in corn plants against common armyworm, M. separata.  相似文献

The European grapevine moth Lobesia botrana Denis & Schiffermüller (Lepidoptera: Tortricidae) is a major pest on grapes worldwide. Attempts to develop control methods for this pest based on grape kairomones demonstrate limited success and studies indicate that a major limiting factor is overlap between synthetic kairomones and background odours in the vineyard. Behaviourally active compounds from non‐host plants may thus represent an effective alternative for monitoring and control methods. Extracts from food plants (i.e. from capsicum, garlic and peppermint, which elicit the so‐called somatosensory sensation) are traditionally used in agriculture for the control of pest insects. Among those plants, Perilla frutescens L. (Lamiales: Lamiaceae), native of Asia, contains compounds activating sensory ion channels in mammals, which are known to be involved in the perception of somatosensory compounds and are expressed in tortricid moth antennae. In the present study, in search of non‐host volatiles with potential application in pest control, essential oil metabolites isolated from P. frutescens are screened for biological activity on the olfactory system of L. botrana. The compounds (S)‐(?)‐perillaldehyde and isoegomaketone, which are released from different P. frutescens varieties, are identified by gas chromatography‐coupled electroantennographic detection. In a dual‐choice oviposition test, females show a preference for a combination of host odours and perillaldehyde, preferring this over a host‐plant odour bouquet alone. In Y‐tube olfactometer assays, virgin males show a higher level of activity in the presence of isoegomaketone, even if not significantly responsive to the compound.  相似文献

1. The aphid alarm pheromone (E)‐β‐farnesene (EBF) is often considered to be used by natural enemies as a prey/host finding kairomone. However, studies show opposing results, some appear to confirm an attraction of aphid natural enemies by EBF whereas others do not provide any evidence for the kairomone function of EBF. 2. To clarify if aphid natural enemies are attracted by the amounts of EBF naturally emitted by aphids, the existing literature was reviewed about EBF attractiveness to aphid natural enemies with consideration of the amounts of EBF used in the studies. 3. Thirty‐one publications that investigated the ability of EBF, aphid cornicle secretion, and attacked aphids, to attract aphid natural enemies were found. Several studies showed an attraction by EBF, but these used much higher amounts of EBF than usually emitted by aphids during a predator attack. Studies investigating EBF amounts similar to what is emitted by aphids are rare and failed to show attraction. Only two studies document an attraction of natural enemies by attacked aphids. 4. As EBF is emitted in very low amounts, not very stable, and only present after an attack, we suggest that aphid‐derived EBF is not a suitable kairomone for most natural enemy species, especially when they are able to use alternative cues. As EBF, amongst other volatiles, is also emitted by herbivore‐induced plants, we propose that natural enemies might use plant‐derived EBF as a synomone to identify aphid‐infested plants via an altered plant volatile bouquet.  相似文献

Farnesyl diphosphate synthase (FPPS) catalyzes the formation of FPP, providing the precursor for the biosynthesis of (E)‐β‐farnesene (EβF) in plants, but it is unknown if FPPS supplies the precursor for the biosynthesis of EβF, the major component of aphid alarm pheromone, though our previous studies support the hypothesis that EβF is synthesized by the aphid itself. Here, we used two cohorts of the green peach aphid Myzus persicae separately, reared on pepper plant and artificial diet to test the correlations among droplet emission, EβF quantity, and FPPS gene expression. It was found that the proportion of aphids emitting cornicle droplets and the quantity of EβF per milligram of aphid were both significantly different between the two cohorts, which were positively correlated with the expression of the two FPPS genes ( MpFPPS1/ 2) in M. persicae. These results were further confirmed by RNAi‐mediated knockdown of MpFPPS1/ 2. Specifically, knockdown of MpFPPS1/ 2 imposed no significant cost on the survival of aphid but remarkably increased the number of offspring per aphid; most importantly, knockdown of MpFPPS1/ 2 significantly reduced the proportion of aphids emitting droplets and the quantity of EβF calculated as per the weight of aphid. Our results suggest that both FPPS genes are involved in the production of EβF in M. persicae and cornicle droplet emission is closely associated with the EβF release in the aphid.  相似文献

Identification of host volatile compounds attractive to codling moth Cydia pomonella, a most important insect of apple, will contribute to the development of safe control techniques. Synthetic apple volatiles in two doses were tested for antennal and behavioural activity in codling moth. Female antennae strongly responded to (Z)3‐hexenol, (Z)3‐hexenyl benzoate, (Z)3‐hexenyl hexanoate, (±)‐linalool and E,Eα‐farnesene. Two other compounds eliciting a strong antennal response were the pear ester, ethyl (E,Z)‐2,4‐decadienoate, and its corresponding aldehyde, E,E‐2,4‐decadienal, which is a component of the larval defence secretion of the European apple sawfly. Attraction of codling moth to compounds eliciting a strong antennal response was tested in a wind tunnel. Male moths were best attracted to a blend of (E,E)‐α‐farnesene, (E)‐beta‐farnesene and ethyl (E,Z)‐2,4‐decadienoate. The aldehyde E,E‐2,4‐decadienal had an antagonistic effect when added to the above mixture.  相似文献

Field studies using the synthetic sex pheromone of Trichophysetis cretacea, a trinary blend of (Z)‐11‐hexadecenyl acetate (Z11‐16:OAc), (Z)‐11‐hexadecenal (Z11‐16:Ald) and (Z)‐11‐hexadecenol (Z11‐16:OH), were performed in Sichuan to determine operational parameters for detection and control, such as dispenser type, blend ratio, dosage, and trap type, height and density. Of three pheromone dispensers tested, grey halo‐butyl isoprene elastomeric septa were significantly more effective than polyvinyl chloride capillary tubing or silicone rubber septa. The ratio of the three components in the blend significantly affected moth catch. In the halo‐butyl isoprene septa, the most effective ratio was 5 : 2 : 1 Z11‐16:OAc:Z11‐16:Ald:Z11‐16:OH. Sticky wing traps caught significantly more moths than water, noctuid moth or cone funnel traps. The most effective height at which wing traps were hung was 20 cm above the jasmine plants. Optimum trap density was 45 traps per hectare. Addition of volatile jasmine compounds did not increase the attractiveness of the sex pheromone. A dosage of 50 μg Z11‐16:OAc per lure was most effective in the autumn weather conditions of Quanwei. These data provide sufficient information to develop effective protocols for using the T. cretacea pheromone to detect and monitor this pest in the jasmine fields.  相似文献

Terpenes are important compounds in plant trophic interactions. A meta‐analysis of GC‐MS data from a diverse range of apple (Malus × domestica) genotypes revealed that apple fruit produces a range of terpene volatiles, with the predominant terpene being the acyclic branched sesquiterpene (E,E)‐α‐farnesene. Four quantitative trait loci (QTLs) for α‐farnesene production in ripe fruit were identified in a segregating ‘Royal Gala’ (RG) × ‘Granny Smith’ (GS) population with one major QTL on linkage group 10 co‐locating with the MdAFS1 (α‐farnesene synthase‐1) gene. Three of the four QTLs were derived from the GS parent, which was consistent with GC‐MS analysis of headspace and solvent‐extracted terpenes showing that cold‐treated GS apples produced higher levels of (E,E)‐α‐farnesene than RG. Transgenic RG fruit downregulated for MdAFS1 expression produced significantly lower levels of (E,E)‐α‐farnesene. To evaluate the role of (E,E)‐α‐farnesene in fungal pathogenesis, MdAFS1 RNA interference transgenic fruit and RG controls were inoculated with three important apple post‐harvest pathogens [Colletotrichum acutatum, Penicillium expansum and Neofabraea alba (synonym Phlyctema vagabunda)]. From results obtained over four seasons, we demonstrate that reduced (E,E)‐α‐farnesene is associated with decreased disease initiation rates of all three pathogens. In each case, the infection rate was significantly reduced 7 days post‐inoculation, although the size of successful lesions was comparable with infections on control fruit. These results indicate that (E,E)‐α‐farnesene production is likely to be an important factor involved in fungal pathogenesis in apple fruit.  相似文献

The distribution of extracellular 1,3‐β‐glucanase secreted by Gaeumannomyces graminis var. tritici (Ggt) was investigated in situ in inoculated wheat roots by immunogold labelling and transmission electron microscopy. Antiserum was prepared by subcutaneously injecting rabbits with purified 1,3‐β‐glucanase secreted by the pathogenic fungus. A specific antibody of 1,3‐β‐glucanase, anti‐GluGgt, was purified and characterized. Double immunodiffusion tests revealed that the antiserum was specific for 1,3‐β‐glucanase of Ggt, but not for 1,3‐β‐glucanase from wheat plants. Native polyacrylamide gel electrophoresis of the purified and crude enzyme extract and immunoblotting showed that the antibody was monospecific for 1,3‐β‐glucanase in fungal extracellular protein populations. After incubation of ultrathin sections of pathogen‐infected wheat roots with anti‐1,3‐β‐glucanase antibody and the secondary antibody, deposition of gold particles occurred over hyphal cells and the host tissue. Hyphal cell walls and septa as well as membranous structures showed regular labelling with gold particles, while few gold particles were detected over the cytoplasm and other organelles such as mitochondria and vacuoles. In host tissues, cell walls in contact with the hyphae usually exhibited a few gold particles, whereas host cytoplasm and cell walls distant from the hyphae were free of labelling. Furthermore, over lignitubers in the infected host cells labelling with gold particles was detected. No gold particles were found over sections of non‐inoculated wheat roots. The results indicate that 1,3‐β‐glucanase secreted by Ggt may be involved in pathogenesis of the take‐all fungus through degradation of callose in postinfectionally formed cell wall appositions, such as lignitubers.  相似文献

The chemical composition of spontaneous volatile emission from Rubus ulmifolius flowers and fruits during different stages of development was evaluated by HS‐SPME‐GC/MS. In total, 155 chemical compounds were identified accounting 84.6 – 99.4% of whole aroma profile of flowers samples and 92.4 – 96.6% for fruit samples. The main constituents were α‐copaene, β‐caryophyllene, germacrene D, (E,E)‐α‐farnesene, 1,7‐octadien‐3‐one,2‐methyl‐6‐methylene, tridecane, (E)‐2‐hexenol acetate, (E)‐3‐hexenol acetate and cyperene. The results give a chemotaxonomic contribution to the characterization of the VOCs emitted from flowers and fruits during their ontogenic development.  相似文献

Understanding host plant volatile – aphid interactions can facilitate the selection of crop border plants as a strategy to reduce plant virus incidence in crops. Crop border plant species with attractive odours could be used to attract aphids into the border crop and away from the main crop. As different cultivars of the same crop can vary in their olfactory attractiveness to aphids, selecting an attractive cultivar as a border crop is important to increase aphid landing rates. This study evaluated olfactory responses of the bird cherry‐oat aphid, Rhopalosiphum padi (L.) (Hemiptera: Aphididae), to three cultivars each of maize [Zea mays L. (Poaceae)], potato [Solanum tuberosum L. (Solanaceae)], and wheat [Triticum aestivum L. (Poaceae)] with the aim of selecting an attractive crop border plant to reduce the incidence of the non‐persistent Potato virus Y [PVY (Potyviridae)] in seed potatoes. Volatiles emitted by the crop cultivars were collected and identified using coupled gas chromatography/mass spectrometry. Quantitative and qualitative differences were found among cultivars. Behavioural responses of alate R. padi to odours of the cultivars and synthetic compounds identified from the plants were determined with a four‐arm olfactometer. Rhopalosiphum padi was attracted to odours emitted from maize cultivar 6Q‐121, but did not respond to odours from the remaining eight crop cultivars. Volatile compounds from maize and wheat cultivars that elicited a behavioural response from R. padi and contributed to differences in plant volatile profiles included (Z)‐3‐hexenyl acetate (attractant) and α‐farnesene, (E)‐2‐hexenal, indole, and (3E,7E)‐4,8,12‐trimethyltrideca‐1,3,7,11‐tetraene (TMTT) (repellents). We conclude that maize cv. 6Q‐121 is potentially suitable as a crop border plant based on the behavioural response of R. padi to the olfactory cues emitted by this cultivar. The findings provide insight into selecting crop cultivars capable of attracting R. padi to crop border plants.  相似文献

Abstract 1 Two codling moth Cydia pomonella kairomonal attractants, ethyl (E,Z)‐2,4‐decadienoate (pear ester) and (E)‐β‐farnesene, were tested in an insecticide‐sprayed apple orchard and an orchard treated for mating disruption with synthetic pheromone (E,E)‐8,10‐dodecadienol (codlemone). Male captures with pear ester were higher in the pheromone‐treated than in the insecticide‐treated orchard, whereas captures with (E)‐β‐farnesene were not different. Subsequent wind tunnel experiments confirmed that pre‐exposure to sex pheromone codlemone increased the behavioural response of codling moth males to pear ester. This supports the idea that male attraction to the plant volatile pear ester and sex pheromone codlemone is mediated through the same sensory channels. 2 Pear ester is a bisexual codling moth attractant and even captures of female moths were significantly increased in the pheromone‐treated orchard. In the laboratory wind tunnel, pheromone pre‐exposure had no effect on female response to pear ester, but significantly more mated than unmated codling moth females flew upwind towards a pear ester source. Differences in mating status in insecticide‐treated vs. pheromone‐treated orchards may thus account for the differences in female trap captures with pear ester. 3 These findings are important with respect to monitoring of codling moth with pear ester in mating disruption orchards. They also emphasize the importance of host plant volatiles in pheromone‐mediated mating disruption, which has been neglected to date.  相似文献

Olfaction is of major importance for survival and reproduction in moths. Males possess highly specific and sensitive olfactory receptor neurones to detect female sex pheromones. However, the capacity of male moths to respond to host‐plant volatiles is relatively neglected and the role that such responses could play in the sensory ecology of moths is still not fully understood. The present study aims to identify host‐plant stimuli for the European grape berry moth Eupoecilia ambiguella Hb. (Tortricidae, Lepidoptera), a major pest of vine in Europe. Headspace volatiles from Vitis vinifera L. cv. Pinot Noir, Vitis vinifera subsp. sylvestris and five other host‐plant species comprising five different families are analyzed by gas chromatography linked to electroantennogram (EAG) recording from male E. ambiguella antennae and by gas chromatography‐mass spectrometry. This procedure identifies 32 EAG‐active compounds, among them the aliphatic compounds 1‐hexanol, (Z)‐3‐hexenol, (Z)‐3‐hexenyl acetate and 1‐octen‐3‐ol; the terpenes limonene, β‐caryophyllene and (E)‐4,8‐dimethyl‐1,3,7‐nonatriene; and the aromatic compounds benzaldehyde and methyl salicylate. Male and female E. ambiguella show similar EAG response amplitudes to individual chemical stimuli and also to mixtures of plant volatiles, as represented by essential oils from ten other plant species. This possibly indicates a common role for plant compounds in the sensory ecology of the two sexes of E. ambiguella.  相似文献

Diversifying agricultural landscapes may mitigate biodiversity declines and improve pest management. Yet landscapes are rarely managed to suppress pests, in part because researchers seldom measure key variables related to pest outbreaks and insecticides that drive management decisions. We used a 13‐year government database to analyse landscape effects on European grapevine moth (Lobesia botrana) outbreaks and insecticides across c. 400 Spanish vineyards. At harvest, we found pest outbreaks increased four‐fold in simplified, vineyard‐dominated landscapes compared to complex landscapes in which vineyards are surrounded by semi‐natural habitats. Similarly, insecticide applications doubled in vineyard‐dominated landscapes but declined in vineyards surrounded by shrubland. Importantly, pest population stochasticity would have masked these large effects if numbers of study sites and years were reduced to typical levels in landscape pest‐control studies. Our results suggest increasing landscape complexity may mitigate pest populations and insecticide applications. Habitat conservation represents an economically and environmentally sound approach for achieving sustainable grape production.  相似文献
