Influence of termolabile enterotoxin (LT-enterotoxin) of Enterobacter cloacae on functional activity of mice peritoneal macrophages was studied and following combinations were used: LT-enterotoxin-producing E. cloacae, its isogenic pair--LT-enterotoxin non-producing E. cloacae, supernatantof broth culture containing LT-enterotoxin, and physiological salt solution (in control group). Data showing decrease in phagocytic and lysosomal activity, disorder in functions of hexosemonophosphate shunt enzymes in peritoneal phagocytes were obtained.  相似文献

The influence of low-frequency ultrasound on the chemotactic, ingestive and digestive activity of peritoneal macrophages in rats was studied. The intraoperative treatment of the peritoneum with ultrasound enhanced chemotactic activity 3.3-fold in comparison with that in the control animals. The digestive function of peritoneal macrophages considerably increased, the stimulation of their ingestive capacity also occurred. The activation of the phagocytic function of macrophages was observed within 7 days after a single sonar treatment. The authors believe that the stimulation of the macrophage system is probably one of the mechanisms of the sanative action of ultrasound which is used at present in purulent surgery.  相似文献

In the present experiment we investigate the effects of diazepam on macrophage activity and serum corticosterone levels in mice. Adult mice were treated with diazepam (1.5 mg/kg/day - group E) or with control solution (group C1) for 7 days; some animals were only handled, receiving no treatment (group C2). Oral onco-BCG was used for peritoneal macrophage activation. Diazepam treatment: 1-decreased macrophage spreading and phagocytosis; 2-decreased the concentrations of H2O2 spontaneously but not phorbol myristate-acetate-induced release. In relation to mice of group C1, diazepam treatment increased the serum levels of corticosterone. No differences were detected between data of groups C1 and C2 both for macrophage activity and serum corticosterone levels. The present data were explained on the basis of a synergistically action for diazepam through peripheral type binding sites (PBR) present in both adrenals and macrophages, stimulating adrenal glucocorticoid production and altering the macrophage cytokine network.  相似文献

Observations made on white mice showed that doses of alcohol had a stimulating effect on the indices of macrophage acitivity (pagocytic number and phagocytic index), while longer administration of 40% ethyl alcohol solution (in doses of 0,1 ml for 21--30 days) suppressed the functional activity of macrophages. In vitro experiments in which macrophages were subjected to the action of ethyl alcohol showed that alcohol, depending on its concentration, had a direct effect on the cells of the microphage system.  相似文献

Macrophages, in general, are critical effectors of body's immune system. Chemical inhibition of phagocytic activity of such macrophages as Kupffer cells has been extensively studied. We have earlier shown that methyl palmitate (MP) inhibits the activation of Kupffer cells. To evaluate the potential of MP to inhibit the activation of other macrophages, we treated rat peritoneal macrophages with varying concentrations of MP. Its treatment led to a dose-dependent inhibition of phagocytic activity, which was found to be 34%, 47%, and 66% at 0.25, 0.50, and 1.0 mM MP, respectively, as measured by latex bead uptake. When MP-treated peritoneal macrophages were stimulated with lipopolysaccharide (LPS), the nitric oxide (.NO) release was inhibited at 6 h, while cyclooxygenase-2 expression decreased after 24 h. The treatment with MP increased the release of interleukin (IL)-10 in the LPS-treated cells at 6 h, while IL-6 and tumor necrosis factor-alpha were significantly increased both at 6 and 24 h. Our data suggest that MP inhibits phagocytic activity and .NO production similar to that observed in isolated Kupffer cells. Therefore, inhibition of phagocytosis by MP may be a general phenomenon, and it could be used as an inhibitor of macrophage function.  相似文献

目的 研究双歧杆菌四联活菌片(商品名:思连康)对小鼠腹腔巨噬细胞吞噬鸡红细胞吞噬率及吞噬指数的影响。方法 将SPF小鼠30只随机分成三组,每组10只,Ⅰ组灌胃生理盐水,Ⅱ组灌胃婴儿双歧杆菌菌悬液,Ⅲ组灌胃双歧杆菌四联活菌片菌悬液,每天给药0.5 mL,菌液浓度为1.0×108 CFU/mL,连续给药10 d后小鼠腹腔注入2%鸡红细胞悬液1 mL(红细胞数量为2×108个/mL),30 min后处死,取小鼠腹腔洗液,观察并记录吞噬鸡红细胞的巨噬细胞数及被吞噬的鸡红细胞数,计算吞噬率及吞噬指数。结果 与Ⅰ组相比,Ⅱ组和Ⅲ组小鼠腹腔巨噬细胞吞噬鸡红细胞的吞噬率和吞噬指数均显著升高(Ps<0.05),其中Ⅲ组高于Ⅱ组(P<0.05)。结论 双歧杆菌四联活菌片及其婴儿双歧杆菌通过提高小鼠腹腔巨噬细胞的吞噬率和吞噬指数提高机体的免疫力。  相似文献

A study of the mouse Balb/c peritoneal macrophages has been made after oral antiinflammatory drugs administration on animals which previously had an inflammation with carrageenan. The results show an enhancement of the number of the macrophages, of their enzymatic activities and of their proteins' content. The effect of bilirubin on the phagocytic activity of mouse peritoneal macrophages and on the expression of Fc receptors and receptors for SRBC was studied. Intraperitoneally administered bilirubin influenced the expression of Fc receptors for IgM, IgG2B, IgA and IgE, whereas the expression of other receptors as well as the phagocytic activity of peritoneal macrophages remained unchanged. The possible mechanism of the effect of bilirubin on Fc receptors is discussed.  相似文献

Streptomycin inhibited the phagocytic activity of mouse peritoneal macrophages forHistoplasma capsulatum. The inhibitory effect was demonstrable following both in vitro and in vivo administration of drug. The observations from examination by direct smear were confirmed by culturing for viable phagocytized organisms. A simple and reproducible technique for the counting of viable phagocytized organisms was developed. Forty-eight hours in vitro treatment of macrophage cultures with 10 to 200 µg/ml of streptomycin produced a graded inhibition of phagocytic activity, minimal at 10 µg/ml and maximal at 200 µg/ml of streptomycin. The parenteral administration of streptomycin significantly reduced phagocytic activity of mouse peritoneal macrophages forH. capsulatum. Mice were treated daily with the subcutaneous injections of 5, 2.5 or 1 mg streptomycin or saline. At 7, 14, 21 and 28 days post-treatment phagocytic activity of macrophages obtained from these mice was tested. There was a progressive, dose-dependent decrease in the phagocytic activity of macrophages derived from streptomycin-treated mice.  相似文献

Hypothermia affects various components of the immune system, leading to impaired immune resistance. To examine the in vitro effect of low temperature on the ultrastructure and phagocytic function of rat peritoneal macrophages, cells were incubated at 4, 10, 24, and 37 degrees C for 60 min. Subsequently, their ultrastructure and capacity to engulf latex particles and generate superoxide anions were evaluated. The results showed a close inverse relationship between incubation temperature and ultrastructural changes, i.e., the lower the temperature, the higher the number of altered cells. In addition, at lower temperatures the number of cells capable of phagocytosis was reduced; the cells engulfed fewer particles per cell and generated less superoxide anions. These findings may be relevant for explaining the increased susceptibility to bacterial infections under hypothermic conditions.  相似文献

Mice treated up to 31 weeks previously with intraperitoneal BCG yielded peritoneal macrophages with decreased phagocytosis of starch granules, latex beads, graphite dust and formalinized Listeria monocytogenes, with or without opsonin, compared to macrophages from untreated mice. These assays were selected to allow quantitative determinations of the rate or extent of particle uptake under nonrate-limiting conditions. Phagocytosis could be depressed to a similar degree by the prior addition of either starch granules or BCG to normal adherent peritoneal cell cultures in vitro. However, with these two particles, two different mechanisms of inhibition of subsequent phagocytosis appeared to be at work. Inhibition of phagocytosis by prior exposure to large amounts of starch granules appeared to consist largely of mechanical interference, as if through the preemption of intracellular space. In contrast, inhibition by prior uptake of BCG occurred with very small amounts of BCG and appeared gradually with time after uptake of BCG. The ability of a “macrophage activating agent” to inhibit selected functions of parasitized cells may help to explain some of the discordant results obtained by others studying phagocytosis by “activated” macrophages. Such agents may simultaneously enhance some macrophage functions and depress others.  相似文献

Because of the great importance of phagocytosis as a key process in host defence, the influence of HIV-infection on the phagocytic activity of monocytes/macrophages (M0/MAC) and granulocytes was investigated. Therefore, blood samples from the peripheral blood of 70 HIV-infected individuals were incubated with fluorescein isothiocyanate (FITC) labeled Escherichia coli. The uptake of the bacteria was monitored by flow cytometer analysis. A strong and significant increase in the relative number of phagocytic granulocytes was observed ranging from 12.8% in an uninfected control collective to over 30% in AIDS patients. This effect was obtained for all patients and independent of the stage of disease. For monocytes, only marginal changes were found in their phagocytic function. These data suggest that the high susceptibility of HIV patients for secondary infections is not linked to a loss of phagocytic ability of monocytes/macrophages and/or granulocytes.  相似文献

The influence of genotype on interferon effect on phagocytic activity of unstimulated mouse peritoneal macrophages (MPM) was studied in in vitro experiments. Treatment of MPM from BALB/c and ICR mice with mouse fibroblast interferon (MuIFN-beta) enhanced the ingestion of non-opsonized Escherichia coli. This effect was dose-dependent and neutralized by anti-interferon globulin. MPM from C57B1/6 mice were not stimulated by the same treatment. Treatment of MPM with pH 2-sensitive immune interferon (MuIFN-gamma) depressed the ingestion independently on the genotype of mice.  相似文献

A study was made of all the different stages of the phagocytic function in peritoneal macrophages from male guinea pigs [3 (SD 1) months old] before, immediately after, and 24 h after being subjected to stress from physical activity (swimming until exhaustion). The early (10 min) and late (40 min) adherence to tissue substrates, chemotaxis, attachment and phagocytosis of Candida albicans, ingestion of inert particles (latex beads), and basal oxidative metabolism [measured by nitroblue tetrazolium (NBT) reduction] were significantly stimulated by the physical activity. After 24 h, late adherence, attachment capacities, and basal oxidative metabolism returned to basal values, whereas early adherence, chemotaxis, phagocytosis of cells and inert particles, and microbicidal capacity (production of superoxide anion measured by NBT reduction in presence of ingested material) remained significantly increased. The stress produced by physical activity, reflected in increased serum corticosterone values, led to a global stimulation of the phagocytic function.  相似文献
