β-Lapachone, an antimicrobial agent, was reduced by Trypanosoma cruzi epimastigotes to a semiquinone radical. It markedly increased the generation of superoxide anion and hydrogen peroxide in intact cells. Using NADH as electron donor, β-lapachone also increased significantly the rate of H2O2 generation in epimastigote homogenates. Michelle Lamont and her collaborators examine the experiences and responses of racialized minorities in confronting racism in the United States, Brazil and Israel. Among their findings, they find that while U.S. blacks often confront offenders, black Brazilians respond by ignoring or humouring them. They often do not understand such situations as racial because of Brazilian narratives that extensive miscegenation has diminished racism and that class may be the major culprit. The book’s comparative approach permits them to leverage theoretical claims about race by examining it beyond the U.S. black–white experience. The authors interpret micro subjective experiences as responses to a macrocomparative context filtered through cultural repertoires, groupness and institutions. My main critique for Brazil is that conclusions are based on a highly selective sample of the educated elite in Rio de Janeiro.  相似文献

This study reports on infection by Trypanoxyuris (Trypanoxyuris) minutus among Alouatta guariba clamitans through biometry on adults and eggs. A total of 58 adult helminths were recovered from the small segment of the animal's large intestine. This study is the first report of this parasite among A. guariba clamitans in Rio de Janeiro.  相似文献

Tumor microenvironment has a high concentration of inorganic phosphate (Pi), which is actually a marker for tumor progression. Regarding Pi another class of transporter has been recently studied, an H+-dependent Pi transporter, that is stimulated at acidic pH in Caco2BBE human intestinal cells. In this study, we characterized the H+-dependent Pi transport in breast cancer cell (MDA-MB-231) and around the cancer tissue. MDA-MB-231 cell line presented higher levels of H+-dependent Pi transport as compared to other breast cell lines, such as MCF-10A, MCF-7 and T47-D. The Pi transport was linear as a function of time and exhibited a Michaelis-Menten kinetic of Km = 1.387 ± 0.1674 mM Pi and Vmax = 198.6 ± 10.23 Pi × h?1 × mg protein?1 hence reflecting a low affinity Pi transport. H+-dependent Pi uptake was higher at acidic pH. FCCP, Bafilomycin A1 and SCH28080, which deregulate the intracellular levels of protons, inhibited the H+-dependent Pi transport. No effect on pHi was observed in the absence of inorganic phosphate. PAA, an H+-dependent Pi transport inhibitor, reduced the Pi transport activity, cell proliferation, adhesion, and migration. Arsenate, a structural analog of Pi, inhibited the Pi transport. At high Pi conditions, the H+-dependent Pi transport was five-fold higher than the Na+-dependent Pi transport, thus reflecting a low affinity Pi transport. The occurrence of an H+-dependent Pi transporter in tumor cells may endow them with an alternative path for Pi uptake in situations in which Na+-dependent Pi transport is saturated within the tumor microenvironment, thus regulating the energetically expensive tumor processes.  相似文献

Sporotrichosis associated with exposure to domestic cats is hyperendemic in Rio de Janeiro, Brazil. A review of the clinical records at our institute revealed four patients with clinical signs of dacryocystitis and a positive conjunctival culture for Sporothrix who were diagnosed with Sporothrix dacryocystitis. Three patients were children (< 13 years of age) and one patient was an adult. Two patients reported contact with a cat that had sporotrichosis. Dacryocystitis was associated with nodular, ulcerated lesions on the face of one patient and with granulomatous conjunctivitis in two patients; however, this condition manifested as an isolated disease in another patient. All of the patients were cured of the fungal infections, but three patients had chronic dacryocystitis and one patient developed a cutaneous fistula. Sporotrichosis is usually a benign disease, but may cause severe complications when the eye and the adnexa are affected. Physicians, especially ophthalmologists in endemic areas, should be aware of the ophthalmological manifestations and complications of sporotrichosis.  相似文献

Glioblastoma (GBM) is the most malignant primary brain tumor, with an average survival rate of 15 months. GBM is highly refractory to therapy, and such unresponsiveness is due, primarily, but not exclusively, to the glioma stem-like cells (GSCs). This subpopulation express stem-like cell markers and is responsible for the heterogeneity of GBM, generating multiple differentiated cell phenotypes. However, how GBMs maintain the balance between stem and non-stem populations is still poorly understood. We investigated the GBM ability to interconvert between stem and non-stem states through the evaluation of the expression of specific stem cell markers as well as cell communication proteins. We evaluated the molecular and phenotypic characteristics of GSCs derived from differentiated GBM cell lines by comparing their stem-like cell properties and expression of connexins. We showed that non-GSCs as well as GSCs can undergo successive cycles of gain and loss of stem properties, demonstrating a bidirectional cellular plasticity model that is accompanied by changes on connexins expression. Our findings indicate that the interconversion between non-GSCs and GSCs can be modulated by extracellular factors culminating on differential expression of stem-like cell markers and cell-cell communication proteins. Ultimately, we observed that stem markers are mostly expressed on GBMs rather than on low-grade astrocytomas, suggesting that the presence of GSCs is a feature of high-grade gliomas. Together, our data demonstrate the utmost importance of the understanding of stem cell plasticity properties in a way to a step closer to new strategic approaches to potentially eliminate GSCs and, hopefully, prevent tumor recurrence.  相似文献

In crustaceans, successful reproductive processes, such as the transition from juvenile to adult, exhibit important morphological changes that can be detected by analyzing relative growth. This study describes the relative growth of body structures in Nematopalaemon schmitti and its secondary sexual characteristics, and also estimates the morphological sexual maturity of this species in a region influenced by upwelling. The carapace length (CL), second pleuron length (PlL), cheliped carpus length (CaL), cheliped propodus length (PrL) and the length of appendix masculina (AML) of the shrimp were measured. The relationships that best demonstrated the changes in allometric coefficient between demographic categories were AML vs. CL for males, and PlL vs. CL for females. The estimated CL for morphological sexual maturity in males was 8.51 mm and 9.30 in females. Our results showed the appendix masculine and the second pleuron were secondary sexual characteristics that play roles in reaching the morphological sexual maturity necessary for reproductive success and to assure the life cycle of this species.  相似文献

Boat-based surveys were conducted from August 2002 to July 2003 to study the activity patterns, spatial pattern of area use, and group characteristics of Sotalia guianensis in Sepetiba Bay, southeast Brazil. Predetermined routes covered the entrance and interior of the bay, resulting in 210 dolphin sightings during 3,300 km total effort. Data on activity were collected using scan group sampling with instantaneous recording after 5 min of observation. Dolphins were sighted more frequently in the entrance of the bay, where water is deeper, and salinity and transparency are higher, than in the interior of the bay, where the environment is more influenced by freshwater inputs. Foraging and feeding were the most frequent activities, and occurred predominantly between 0600 and 1000. Foraging and feeding peaked during ebbing, low, and flooding tides, while socializing predominated at high tide. Mean group size was larger in the interior of the bay and when seabirds were present. Large aggregations containing >100 individuals of Sotalia guianensis seen year-round indicate that Sepetiba Bay is an important area for this species in coastal Brazil.  相似文献

Darwin and Wallace, in the mid‐nineteenth century, were the first to document examples of natural selection acting on island dwellers. A century later a pattern of morphological differences among organisms on islands was coined the ‘island rule’, which states that on islands species with small individuals tend toward gigantism and large individuals tend toward dwarfism. Selective pressures such as limited resources and increased intraspecific competition modulate the size of organisms in these environments. Of the several works that have tested vertebrates for adherence to the island rule only two have addressed amphibians. This work is the third record of body size variation of island amphibian populations, and the first for the Southern Hemisphere. The islands investigated were once continuous with mainland, and now are isolated as a result of sea level fluctuations that took place in the Pleistocene and Holocene. This study compared morphometric variation in populations of Rhinella ornata (Bufonidae) occurring on three islands of the Costa Verde to populations on five continental areas in Rio de Janeiro, Brazil. We measured 18 morphometric variables of 177 individuals. There was a shift toward smaller body size (dwarfism) in two of the three island populations studied. We attribute this general pattern to geographic factors, verifying the expression of the island rule in tropical frogs populations (insular dwarfism) operating inversely in relation to those of temperate environments (island gigantism).  相似文献

Esophageal cancer is the sixth most common cause of cancer-related death worldwide. Current chemotherapy regimens include a combination of 5-fluorouracil (5-FU) and cisplatin, but more efficient therapy strategies are needed to increase 5-year survival. Alterations in the signaling pathway of the tumor suppressor gene Rb-1, which encodes a phosphoprotein (pRB) that negatively regulates the G1/S transition of the cell cycle, are present in 70% of all tumors, but its role in esophageal cancer is still unclear. Most of these are alterations leading to up-regulation of the activity of cyclin-dependent kinases (CDKs) to phosphorylate pRB, which suggests that keeping the wild type pRB phosphorylated might be advantageous. Besides proliferation, pRB also regulates apoptosis induced by tumor necrosis factor-alpha (TNF-α) and DNA-damage. We investigated the status of phosphorylation of pRB along esophageal tumorigenesis stages, as well as whether hyperphosphorylation of pRB could suppress apoptosis induced by cisplatin, 5-FU, or TNF-α in esophageal cancer cells. pRB phosphorylation increased progressively from normal esophageal tissue to metaplasia and adenocarcinoma, suggesting that pRB phosphorylation increases along esophageal tumor stages. When RB-1 was knocked down or CDK inhibitors reduced the levels of phosphorylated pRB, opposite apoptotic effects were observed, depending on the combination of drugs tested: whereas TNF-α- and cisplatin-induced apoptosis increased, 5-FU-induced apoptosis decreased. Taken together, these data suggest that pRB plays a role in esophageal adenocarcinoma and that, depending on the type of anti-cancer treatment, combining CDK inhibitors and chemotherapy has the potential to increase the sensitivity of esophageal cancer cells to cell death.  相似文献

Cutaneous leishmaniasis (CL) in the state of Rio de Janeiro is sporadic and can be characterised as a peridomestic transmission that occurs in modified natural environments. The aim of this work was to study the fauna and ecological characteristics of sandflies in an environmentally protected area (the State Park of Serra da Tiririca) within the remnants of the Atlantic Forest in the municipalities of Niterói and Maricá and their possible relationship with leishmaniasis. Captures were performed using light traps during the night once a month for one year in both sylvatic environments and areas surrounding homes near the park. A total of 1,037 sandflies were captured, belonging to nine genera and 12 species: Evandromyia tupynambai (34.1%), Migonemyia migonei (20.6%), Brumptomyia cunhai (13.8%), Micropygomyia schreiberi (9.7%), Psathyromyia lanei (6.5%), Brumptomyia nitzulescui (5.7%), Evandromyia edwardsi (5.4%), Nyssomyia intermedia (2.8%), Evandromyia cortelezzii (0.6%), Pintomyia bianchigalatiae (0.5%), Lutzomyia longipalpis (0.2%) and Sciopemyia microps (0.1%). Both Mg. migonei and Ny. intermedia may be acting as vectors of CL in this area.  相似文献

A new species of Pseudascarophis (Nematoda: Cystidicolidae) found in the stomach of Kyphosus sectatrix (Linnaeus) (Kyphosidae), off Rio de Janeiro, Brazil, is described. The new species can be differentiated from the other congeners by the presence of lateral alae, distinct but inconspicuous cephalic papillae at the anterior end, three pairs of precloacal and one pair of adcloacal papillae in males, egg morphology and morphometry of glandular oesophagus and spicules. Pseudascarophis tropica is transferred to Ascarophis as Ascarophis tropica (Solov''eva) comb. n. due to its ambiguous diagnosis.  相似文献

We investigated long-term trends in brown macroalgal assemblages inhabiting shallow subtidal rocky bottoms under the influence of thermal effluent discharge from the Brazilian nuclear power plant (BNPP). Three operational periods based on the units of the BNPP were analysed: T0 = pre-operational, between the years 1980 and 1983; T1 = operational period of unit 1, between 1988 and 1999; and T2 = operational period of units 1 and 2, between 2005 and 2012. Using generalized linear mixed models (GLMMs), we found significant declines in the numbers of genera and species over time. More than half of the species of brown macroalgae disappeared during T2. In addition, the numbers of macroalgal genera and species were inversely related to the local surface seawater temperatures. Multivariate analyses revealed a clear separation between the years of T2 and those of T0, indicating long-term changes in the community assemblages. Among the most common species in the area, the frequencies of Canistrocarpus cervicornis, Dictyopteris delicatula, Hincksia mitchelliae, Sargassum spp. and Sphacelaria tribuloides decreased during T2, while Padina gymnospora maintained rather high yearly frequencies during T2 (>40%). Our data suggest that seawater temperatures consistently higher than 30 °C together with peaks higher than 35 °C may have triggered the decline in the brown algae on rocky bottoms under the influence of the BNPP discharge. These results from southern Brazil are consistent with studies from other locations that ascribe changes in seaweed diversity to increasing seawater temperatures, highlighting the sensitivity of brown macroalgae to thermal stress and demonstrating their effectiveness as an ecological indicator for monitoring the effects of nuclear power plant effluents on coastal environments.  相似文献

The structure of the zooplankton community in an estuary adjacent to the Admiral álvaro Alberto Nuclear Power Plant at Angra dos Reis, state of Rio de Janeiro, Brazil was studied from 2001 through 2005. At that time, the power plant had been operating for 20 years. The results were compared with a previous study in 1991–1993. The zooplankton was sampled 4 times a year, in vertical hauls using a 150 μm net, at two fixed points near the plant’s intake and discharge sites. Temperature, salinity, and chlorophyll a were measured. The water of Ribeira Bay is warm, with salinities typical of Coastal Water and more saline waters. Zooplankton density at the discharge site (Mean: 245,434 ind m−3, SD: 335,358 ind m−3) was higher than that at the intake site (Mean: 84,634 ind m−3, SD: 101,409 ind m−3). A total of 121 mesozooplankton taxa were recorded from 2001 to 2005. Copepoda constituted the most common taxon and comprised more than 57% of the total zooplankton, followed by cladocers and gastropod larvae. A seasonal zooplankton cycle was observed only during 2004; in other years, the plankton varied only between years. Overall mesozooplankton abundance at the discharge site was similar to levels reported from the inner zone of this estuary in 1991–1993. Surface temperature was the important factor structuring the zooplankton community at the discharge site. No effect on the mesozooplankton by passage through the condensers could be discerned, and no permanent negative influence on the plankton populations could be detected. Guest editors: U. M. An unusual species of Leptophlebiidae is described based on males, females, and nymphs from Rio de Janeiro State, Brazil. As a consequence of its distinct characteristics on all stages, a new genus is established. The new genus can be distinguished from other South American Leptophlebiid genera mostly by: Adults: vein MA of fore wings asymmetrical; hind wings with costal projection well developed, Sc ending at cross vein near costal projection; tarsal claws dissimilar, one blunt other acute; projections of styliger plate forming two well developed lobes with rounded apex, ventrally obstructing view of the penes; penes fused on basal half, each lobe with a ventral furrow and a long and slender spine directed anteriorly. Nymphs: Head prognathous, wider than labrum; labrum with prominent median emargination, with three subtle crenulations; body flattened; hind wings pads present; tarsal claws with over 20 denticles, subapical denticle much larger than remaining denticles; gills long and narrow, present on abdominal segments I-VII; posterolateral projections present on abdominal segments VIII-IX. Phylogenetic analyses conducted based on a previously published data matrix that included other South America leptophlebiid genera placed Poranga nessimiani gen. nov. et sp. nov. as sister to Bessierus + Perissophlebioides. In most analyses this clade was recovered within the Farrodes complex. Nymphs of the new taxa are particularly similar to Bessierus, whereas adults share the very acute costal projection on hind wings seen in non-dipterous members of the Farrodes complex.  相似文献

In the present work the metabolism of poly(A)+ RNA was investigated in cells of Tetrahymena pyriformis derived either from stationary cultures or from starved suspensions that were initiating growth. Under these circumstances the organisms derived from stationary cultures synthesize ribosomal and poly(A)+ RNA and form polysomes. In the presence of actinomycin D (actD) the observed expansion of the polysomal population is arrested. Pre-starved cells, on the other hand, start making polysomes in the virtual absence of ribosomal and poly(A)+ RNA synthesis soon after being transferred to peptone medium. In this case polysome formation is only partially sensitive to actD. These results have been interpreted as indicating that, in the beginning of growth, cells derived from stationary cultures are dependent on RNA synthesis for polysome formation, whereas pre-starved cells use pre-synthesized RNA for the same purpose.  相似文献

Esophageal squamous cell carcinoma (ESCC) is a frequent and lethal neoplasia. As recent advances in targeted therapy have not improved ESCC prognosis, characterization of molecular alterations associated to this tumor is of foremost relevance. In this study, we analyze, for the first time, the complete genomic profile of ESCC by RNA-seq. TP53 was the most frequently mutated gene in the investigation and validation sets (78.6% and 67.4%, respectively). Differential expression analysis between tumor and nontumor adjacent mucosa showed 6698 differentially expressed genes, most of which were overexpressed (74%). Enrichment analysis identified overrepresentation of Wnt pathway, with overexpressed activators and underexpressed inactivators, suggesting activation of canonical and noncanonical Wnt signaling pathways. Higher WNT7B expression was associated with poor prognosis. Twenty-one gene fusions were identified in 50% of tumors, none of which involving the same genes in different patients; 71% of fusions involved syntenic genes. Comparisons with TCGA data showed co-amplification of seven gene pairs involved in fusions in the present study (~33%), suggesting that these rearrangements might have been driven by chromoanagenesis. In conclusion, genomic alterations in ESCC are highly heterogeneous, impacting negatively in target therapy development.  相似文献
