Summary Hydrogen evolution from root nodules has been reported to decrease the efficiency of the nitrogen fixing system. Mutants ofRhizobium meliloti andRhizobium leguminosarum were selected which were deficient in H2-uptake capacity (Hup). The relative efficiency of the nitrogen fixation for both species assessed with C2H2 reduction was 0.66.The hydrogen production was monitored using a simple root incubation method. As such, hydrogen production up to 3.83 and 15.57 ml.day–1.g–1 plant dry weight were recorded forPisum sativum — Rhizobium leguminosarum 4.20 Hup andMedicago sativa — Rhizobium meliloti 1.5 Hup respectively. In a closed container (250 ml), hydrogen concentrations up to 20% (v/v) could be reached in the root phase ofMedicago sativa in a time period of 320 hours.  相似文献

Changes in cellular isoflavone (daidzein and genistein) contents were monitored in root cultures of Albizzia kalkora (Roxb.) Prain after feeding different ratios of NH4^+/NO3^- and treatment with a biotic elicitor (three strains of Rhizobium sp.). The NH4^+/NO3^- ratio appears to be positively correlated with daidzein content in the roots and shows a negative correlation with genistein. Among the three different strains of Rhizobium used, the strain ATCC 15834 caused a 35% increase in daidzein production by infection. In the case of genistein, maximum production (94%) was obtained when cultures were treated on Day 6 by the strains ATCC 15834 and KCTC 1541. The biosynthetic pathway of the two isoflavones apparently reacts differently to the same culture conditions and the same strains of Rhizobium. Therefore, the present data suggest that the production of daidzein and genistein could be modulated by changing the NH4^+/NO3^- ratio and the application of Rhizobium.  相似文献

The sensitivity of Rhizobia isolated from pea, fenugreek, gram, cowpea, urid andRhizobium meliloti, Rhizobium trifolii andRhizobium japonicum was studied in asparagine-mannitol, salt-mannitol and salt media. It was observed that Rhizobia from gram were most resistant.Rhizobium japonicum was found to be resistant up to 50 mg vanillin/100 ml. Vanillin-resistant strains could utilize vanillin as the sole carbon source. In the case of pea, gram and fenugreek vanillin was found to be utilized in the presence of mannitol.  相似文献

A total of 15 rhizobial strains representing Rhizobium meliloti, Rhizobium japonicum, Rhizobium trifolii, Rhizobium leguminosarum, Rhizobium sp. (Sesbania rostrata) and Rhizobium sp. (Hedysarum coronarium), were studied with regard to growth rate under salt stress in defined liquid media. In the presence of inhibitory concentrations of NaCl, enhancement of growth resulting from added glycine betaine was observed for R. meliloti strains and Rhizobium sp. (Hedysarum coronarium) but not for other Rhizobium species. The concentration of glycine betaine required for maximal growth stimulation was very low (1 mM) in comparison with the osmolarity of the medium. The stimulation was shown to be independent of any specific solutes. Other related compounds like proline betaine, carnitine, choline, -butyrobetaine and pipecolate betaine were also effective compounds in restoring the growth rate of cells grown in medium of elevated osmolarity. High rate of glycine betaine uptake was demonstrated in R. meliloti cells grown in media of increased osmotic strength. The intracellular concentration of this solute was found to be 308 mM in 0.3 M NaCl-grown cells and 17 times lower in minimal medium-grown cells. Glycine betaine was used for growth under conditions of low osmolarity but could not serve as sole carbon or nitrogen source in medium of increased osmotic strength. A cultivation system with simultaneous growth of six bacterial cultures in separate bags in dialysis culture was developed. In a medium with no added carbon source (one half concentrated Hoagland solution, water deionized and distilled), cell number ofRhizobium japonicum increased during a 7 day period by a factor of 35, whereas the number ofEnterobacter aerogenes cells decreased to one half. With a concentration of 100 nM succinate as an additional carbon source in the inflow,Rhizobium japonicum 61-A-101 cell number increased by a factor of 50 during an 8 day period, whereas cell number ofEnterobacter cloacae NCTC 10005 only doubled and ofEnterobacter aerogenes NCTC 10006 decreased. At 10 mM concentration of succinate in the inflow, doubling time the twoEnterobacter strains was about 12 h, compared to about 24 h for theRhizobium japonicum strain. Varying the succinate concentration from 10 mM to 100 nM in the inflow,Rhizobium japonicum 61-A-101 surpassed theEnterobacter aerogenes strains in the growth rate between 1 mM and 100 M succinate in the inflowing medium. Three otherRhizobium japonicum strains (fix+ and fix-) did grow with a similar rate as strain 61-A-101 at very low concentrations of substrate. Growth rates for the strains were confirmed by protein data per culture. Growing in competition with twoPseudomonas strains,Rhizobium japonicum RH 31 Marburg (fix-) did overgrow alsoPseudomonas fluorescens, was however outgrown byPseudomonas putida. 为探求促进广金钱草萌发及生长的有效途径,该文使用不同浓度的费氏中华根瘤菌、放射型根瘤菌以及田菁茎瘤固氮根瘤菌菌液分别浸染广金钱草种子,以蒸馏水浸泡种子为空白处理,研究其对广金钱草种子萌发指标(发芽率、发芽势、发芽指数、活力指数)的影响,并在温室培养40、60、80 d后分别测定广金钱草幼苗的生长指标(叶片数、分枝数、株高)及部分生理指标(叶绿素含量、含氮量)的变化。结果表明:(1)在1×107 CFU·mL-1浓度条件下,三种菌液浸种均获得最大萌发指标,其中田菁茎瘤固氮根瘤菌实验组效果最显著,发芽率、发芽势、发芽指数和活力指数高于对照组16.00%、9.33%、9.51和41.34%。(2)除田菁茎瘤固氮根瘤菌实验组外,其余实验组的广金钱草幼苗叶片数、分枝数及株高均低于对照组。(3)放射型根瘤菌实验组和田菁茎瘤固氮根瘤菌实验组幼苗叶绿素含量相比对照组分别增加1.47%和7.47%,含氮量分别增加0.57%和5.17%。综上所述,三种根瘤菌菌液浸种均可在不同程度上提高广金钱草种子的发芽能力和植株生长期的叶绿素和含氮量,其中田菁茎瘤固氮根瘤菌具有最大正向影响,可有效提高种子萌发能力并促进植株生长。该研究可为不同地区广金钱草的种植和栽培提供技术指导,为开发田菁茎瘤固氮根瘤菌作为植物促生长菌剂提供理论支持。  相似文献

[3H]Dihydrozeatin supplied to photoautotrophically growing cell suspension cultures of Chenopodium rubrum was rapidly taken up and metabolized by the cells. The predominant metabolites in extracts of the cells were [3H]dihydrozeatin-O-glucoside and [3H]dihydrozeatin riboside-O-glucoside. Both these compounds could be shown to be compartmented within the vacuole, whereas [3H]dihydrozeatin and [3H]dihydrozeatin riboside, which were both present to a minor extent in cell extracts, were both present to a minor extent in cell extracts, were localized predominantly outside the vacuole. Analysis of the culture medium at the end of the 36-h incubation period showed that there had been an efflux of [3H]dihydrozeatin metabolites out of the cells. Whereas [3H]dihydrozeatin riboside was found to be the major extracellular [3H]dihydrozeatin metabolite, the O-glucosides of neither this compound nor [3H]dihydrozeatin could be detected in the medium. Zusammenfassung Als weiterer Beweis für die Transformierbarkeit von Agrobacterium tumefaciens und Rhizobium leguminosarum werden wechselseitige Transformationsversuche durchgeführt und dabei die Resistenz gegen Streptomycin bzw. gegen ein Sulfonamid (Marbadal) als Selektionsmaterial benutzt. Die ermittelten Transformationsraten liegen in der Größenordnung von 4–7 · 10-6 bzw. 13 bis 16 · 10-6.

Summary As further proof for the transformability of Agrobacterium tumefaciens and Rhizobium leguminosarum reciprocal transformation assays were performed using the resistence against streptomycin resp. against a sulfonamide (Marbadal) as selection marker. The transformations rates are 4–7×10-6 resp. 13–16×10-6.

Summary In pea (Pisum sativum L.), mutants could be induced, modified in the symbiotic interaction withRhizobium leguminosarum. Among 250 M2-families, two nodulation resistant mutants (K5 and K9) were obtained. In mutant K5 the nodulation resistance was monogenic recessive and not Rhizobium strain specific. Out of 220 M2-families one mutant nod3 was found which could form nodules at high nitrate concentrations (15 mM KNO3). This mutant nodulated abundantly with severalRhizobium strains, both in the absence and presence of nitrate. Probably as the result of a pleiotropic effect, its root morphology was also changed. Among 1800 M2-families, five nitrate reductase deficient mutants were obtained and one of them (mutant E1) was used to study the inhibitory effect of nitrate on nodulation and nitrogen fixation.The results of the present investigation show that pea mutants which are modified in their symbiosis withRhizobium leguminosarum, can readily be obtained. The significance of such mutants for fundamental studies of the legume-Rhizobium symbiosis and for applications in plant breeding is discussed.  相似文献

The first step in attachment of Rhizobiaceae cells to plant root hair tips is mediated by a Ca2+-dependent, Ca2+-binding protein, rhicadhesin. The possible role of Ca2+ in synthesis, anchoring and activity of rhicadhesin was investigated. Growth of Rhizobium leguminosarum biovar viciae cells under Ca2+-limitation was found to result in loss of attachment ability. Under these conditions, rhicadhesin could not be usolated from the bacterial cell surface, but was found to be excreted in the growth medium. Divalent ions appeared to be essential for the ability of purified rhicadhesin to inhibit attachment of R. leguminosarum biovar viciae cells to pea root hair tips. Calcium ions were found not to be involved in binding of rhicadhesin to the plant surface, but appeared to be involved in anchoring of the adhesin to the bacterial cell surface. A model for the role of Ca2+ in activity of rhicadhesin is presented.  相似文献

Wheat straw impregnated with a nutrient solution was used to culture Rhizobium leguminosarum. The fermentation was carried out in a periodic pressure, oscillating, solid-state fermenter. At 30 °C and 3 atm, Rhizobium leguminosarum grew to 5.3×1010 c.f.u. g–1 substrate dry matter in about 36 h, while only 1.8×1010 c.f.u. g–1 substrate dry matter was obtained in a conventional static tray fermenter.  相似文献

Summary Phosphate increased nitrogen uptake by lucerne appreciably on a saline soil. Nitrogenous fertiliser or inoculation with an effective strain ofRhizobium meliloti did not increase the yield significantly. In soils where indigenousRhizobium japonicum was absent inoculation increased soybean yields and the additional fixed nitrogen removed by soybeans amounted to 40 to 120 kg ha−1. Summary DNA fragments carrying the recA genes of Rhizobium meliloti and Rhizobium leguminosarum biovar viciae were isolated by complementing a UV-sensitive recA Escherichia coli strain. Sequence analysis revealed that the coding region of the R. meliloti recA gene consists of 1044 by coding for 348 amino acids whereas the coding region of the R. leguminosarum bv. viciae recA gene has 1053 bp specifying 351 amino acids. The R. meliloti and R. leguminosarum bv. viciae recA genes show 84.8% homology at the DNA sequence level and of 90.1% at the amino acid sequence level. recA mutant strains of both Rhizobium species were constructed by inserting a gentamicin resistance cassette into the respective recA gene. The resulting recA mutants exhibited an increased sensitivity to UV irradiation, were impaired in their ability to perform homologous recombination and showed a slightly reduced growth rate when compared with the respective wild-type strains. The Rhizobium recA strains did not have altered symbiotic nitrogen fixation capacity. Rhizobium japonicum CJ1 was capable of growing using formate as the sole source of carbon and energy. During aerobic growth on formate a cytoplasmic NAD+-dependent formate dehydrogenase and ribulose bisphosphate carboxylase activity was demonstrated in cell-free extracts, but hydrogenase enzyme activity could not be detected. Under microaerobic growth conditions either formate or hydrogen metabolism could separately or together support ribulose bisphosphate carboxylase-dependent CO2 fixation. A number of R. japonicum strains defective in hydrogen uptake activity were shown to metabolise formate and induce ribulose bisphosphate carboxylase activity. The trihydroxamate siderophore, hydroxamate K, has been purified from culture filtrates of iron-deficient Rhizobium leguminosarum biovar viciae MNF710. The iron complex has a molecular weight of 828 and an absorption maximum at 443 nm (M=1510). 55Fe complexed to purified hydroxamate K was taken up by MNF710, its hydroxamate-negative mutant MNF7102 and Rhizobium leguminosarum biovar trifolii WU95 via an iron-regulated transport system, but Rhizobium meliloti U45 failed to take up the iron-siderophore complex under any conditions. A similar pattern of iron uptake was observed with ferrioxamine B. MNF710, MNF7102, U45 and WU95 all transported 55Fe-ferrichrome but only the first three strains took up 55Fe-ferrichrome A. All these 55Fe-trihydroxamate uptake systems were ironregulated in MNF710, MNF7102 and WU95. In contrast, uptake of 55Fe-rhodotorulate, a dihydroxamate, was essentially constitutive in all four organisms. Similarly, uptake of 55Fe-citrate and 55Fe-nitrilotriacetic acid was constitutive. None of the strains took up 55Fe complexed with enterobactin or with pyoverdins from Pseudomonas aeruginosa ATCC15692 (PAO1) and Pseudomonas fluorescens ATCC17400.  相似文献

The metabolism of an organophosphorous fungicide, Kitazin P® (S-benzyl O, O-diisopropyl phosphorothioate) by mycelia of Pyricularia oryzae, the rice blast fungus, was studied by using 32P- and 35S-doubled-labeled, 35S-labeled and non-labeled compounds.

The mycelia incorporated the fungicide from the medium, metabolized most of it to water-soluble metabolites and released them into the medium. The main metabolite from Kitazin P was identefied as O, O-diisopropyl hydrogen phosphorothioate by ion-exchange chromatographic separation of double-labeled metabolites. S-Benzyl O-isopropyl hydrogen phosphorothioate, diisopropyl hydrogen phosphate and/or isopropyl dihydrogen phosphate were suspected to be minor metabolites. From the toluene-soluble fraction several metabolites were separated by thin-layer chromatography. The most eminent metabolite among them was found to be the m-hydroxy derivative of Kitazin P by thin-layer chromatographic identification of the labeled metabolite and gas-liquid chromatographic identification of the metabolite and its methylated and acetylated products with respective authentic samples. The o- and p-hydroxy derivatives were not found in the metabolites.

No significant difference in the rate and extent of metabolism was found between susceptible and resistant clones against the fungicide, when its concentration was below the growth-inhibitory concentration to the both clones.  相似文献

The effects of modifying boron (B) and calcium (Ca2+) concentrations on the establishment and development of rhizobial symbiosis in Pisum sativum plants grown under salt stress were investigated. Salinity almost completely inhibited the nodulation of pea plants by Rhizobium leguminosarum bv. viciae 3841. This effect was prevented by addition of Ca2+ during plant growth. The capacity of root exudates derived from salt‐treated plants to induce Rhizobium nod genes was not significantly decreased. However, bacterial adsorption to roots was highly inhibited in plants grown with 75 mM NaCl. Moreover, R. leguminosarum 3841 did not grow in minimal media containing such salt concentration. High Ca2+ levels enhanced both rhizobial growth and adsorption to roots, and increased nodule number in the presence of high salt. Nevertheless, the nodules developed were not functional unless the B concentration was also increased. Because B has a strong effect on infection and cell invasion, these processes were investigated by fluorescence microscopy in pea nodules harbouring a R. leguminosarum strain that expresses green fluorescent protein. Salt‐stressed plants had empty nodules and only those treated with high B and high Ca2+ developed infection threads and exhibited enhanced cell and tissue invasion by Rhizobium. Overall, the results indicate that Ca2+ promotes nodulation and B nodule development leading to an increase of salt tolerance of nodulated legumes.  相似文献

Summary Rhizobium leguminosarum, strain PRE, is unable to use sulphate as the sulphur source. Sulfhydryl compounds must be added to achieve growth.Omission of FeCl3 from the synthetic growth medium resulted in a sharp decrease in growth of this Rhizobium strain as contrasted to other strains of R. leguminosarum. Catechol 1,2-dioxygenase has been purified 46-fold from cells of Rhizobium trifolii TA1 grown on benzoate plus glucose. The dioxygenase had a molecular weight of 107,000 and a sub-unit molecular weight of 59,000. The enzyme had a K m of 2 M for catechol and also cleaved 4-methylcatechol. The dioxygenase contained 2 g atoms of Fe3+ per mole of enzyme which could be removed by treatment with 1,10-phenanthroline, resulting in a complete loss of activity; reactivation of the enzyme occurred specifically with Fe3+.  相似文献

Exopolysaccharides (EPS) of nodulating strains of Rhizobium trifolii and Rhizobium leguminosarum added to red clover seedlings before inoculation reduced the number of nodules. The inhibition of the nodulation was correlated with the amount of EPS. The preparations of EPS from mutants defective in early stages of nodulation (Roa- or Hac-) did not affect the nodulation, whereas EPS from mutants deficient in late stages (post Hac-) exerted an inhibitory effect.Inactive preparation of EPS contained less O-acetyl groups and pyruvic acid residues. Deacetylation and depyruvylation of EPS from R. trifolii Nod+ abolished it inhibitory effect.
