Two yellow and two red pigmented strains of Thermus were monitored for changes in fatty acid content and composition with reference to growth phase at the optimum temperature. Fatty acid content per mg of dry weight increased as the cultures aged. In addition the quantities of iso C 15:0, iso C 17:0 and iso C 16:0 increased in yellow pigmented strains, but in red pigmented strains, an increase was seen in iso C 15:0, but C 16:0 and iso C 16:0 levels decreased. Thus the fatty acid composition of these organisms varies with growth phase, and shows also strain specific variability.  相似文献

嗜冷菌及耐冷菌是冷适应酶及生物活性物质的重要资源。本研究从内陆土壤筛得33株冷适应细菌,包括6株革兰氏阳性菌与27株革兰氏阴性菌。通过细胞膜脂肪酸分析表明,革兰氏阳性菌的膜脂肪酸主要为分支脂肪酸,推测分支结构是阳性菌膜脂的主要适冷机制。革兰氏阴性菌呈现了不饱和、分支、短链等多样的膜脂适冷调节方式。根据脂肪酸组分的多样性,选择并鉴定了17株嗜冷及耐冷菌分布在11个属中,细胞膜脂肪酸组成的变化规律与细菌16SrRNA的进化分布高度一致。研究还表明同为不饱和脂肪酸为主的革兰氏阴性菌呈现了不同的适冷机理。相关研究不仅阐述了冷适应细菌的细胞膜脂肪酸的适应机制,而且为相关适冷酶源的开发利用提供了宝贵的资源。  相似文献

Wild, pigmented strains of Serratia marcescens and their non-pigmented mutants were compared on the basis of fatty acid profiles and lipid content. Classic biochemical tests show only minor differences, as well as fatty acid ratio C18:C16. The total amount of lipid synthesized and the saturated/unsaturated fatty acids ratio disclose a sharp total lipid reduction and a high percentage of unsaturated fatty acids in the pigmented strains, placing them in separated clusters compared with the nonpigmented mutants. It is hypothesized that the synthesis of the polyacetate required for the completion of the prodigiosin molecule may result in waste of methyl groups and thus affect the total amount of lipids.  相似文献

The correlation of bacterial lipid composition with antibiotic resistance was investigated with particular emphasis on those organisms in which resistance may be related to membrane or envelope structure or function, as in resistance to tetracyclines and polymyxin. Chloroform-methanol-extractable lipids, phosphatidyl ethanolamine fractions, and both fatty acids of these lipid fractions and total fatty acids were studied by using thin-layer chromatography, gas chromatography, and infrared spectroscopy. Consistent quantitative differences were found between the fatty acid compositions of sensitive and resistant strains. Most notable was the fact that, in gram-negative organisms, resistant strains showed decreases in cyclopropane acids as compared with sensitive strains. These changes were found to be inherent in the strains and not due to growth stage or culture age. No significant qualitative differences were noted. In contrast, no such variation in fatty acid content was observed in penicillin-sensitive and resistant strains of gram-positive cocci. As significant alterations of fatty acid composition were noted in gram-negative strains resistant to antibiotics, we suggest that resistance is correlated to membrane or envelope lipid composition.  相似文献

Fatty acid composition of total lipids, neutral lipids and phospholipids of strains of Aedes aegypti were determined. The fatty acid composition of the strains differed quantitatively with regard to the relative percentage of commonly occurring fatty acids. Gas-liquid chromatography of fatty acid methyl esters showed 18:1 (oleic or elaidic) to be the predominant fatty acid. The fatty acid was identified as oleic by argentation thin-layer chromatography. A modified colorimetric method was used to determine tissue-free fatty acids. The lipids were predominantly triacylglycerol with lesser amounts of free fatty acids and decreasing amount of sterol ester, sterol, monoacylglycerol, diacylglycerol and hydrocarbons. The data show considerable lipid differences between the Caribbean strains (Les Cayes, Haiti, and San Juan, Puerto Rico) and the Jakarta (Indonesia) strain. The Shimba Hills (Kenya) strain was more similar to Jakarta than to the Caribbean strains. The results obtained with the different strains are discussed in relation to the established oral susceptibility to Dengue 1 and Dengue 2, yellow fever, and genetic analysis by isoenzyme studies.  相似文献

Summary From a metabolic point of view, sodium propionate and sodium caprylate in sub-fungistatic concentrations, manifest some interesting effects upon the metabolism, fatty acid synthesis, and pigmentogenesis ofTrichophyton rubrum in its earlier growth phases.Propionate at the higher concentration and caprylate inhibit growth as measured by mycelial dry weight.The higher concentration of propionate suppressed pigment production but a yellow pigment, apparently differing from others previously described, was observed consistently in cultures with the lower concentration of the salt.Caprylate in the concentration tested resulted in a culture in which fatty acid concentration was significantly decreased, however, there appears to be no correlation between fatty acid content depression and mycelial dry weight.Data on fatty acid composition suggest that propionate, an odd-numbered carbon chain fatty acid, diverts synthesis of even-numbered carbon chain fatty acids to the odd-numbered ones.  相似文献

To determine differences in microbial community structures, fatty acids from two strains of the green microalga Apatococcus constipatus were isolated and identified by instrumental means. The main fatty acids found were 16:0 and 14:0. These predominant acids represented more than 53% of the total fatty acid (content in both algal isolates. In addition, saturated fatty acids were present in much greater quantity than unsaturated ones. Differences between the strains in the composition of other, modified fatty acids were also evident. The occurrence of fatty acid biomarkers characteristic of certain taxonomic groups confirmed the presence of Gram-positive and Gram-negative bacteria, and fungi. Those observed variations were undoubtedly due to distinct community structures of symbiotic microorganisms living in close associations with the alga. The results presented here indicate that different isolates of the same alga might exhibit different microbial community structures.  相似文献

A comparative analysis of fatty acid composition of lipid components from skeletal muscle sarcolemma and sarcoplasmic reticulum membranes of rabbits and crayfishes Astacus fluviatilis and A. leptodactylus has been made by means of gas-liquid chromatography of methyl esters of fatty acids. There are slight differences between external and internal membranes in fatty acids composition of lipids in the same animal. Considerable differences were found, however, when lipids in corresponding membranes of different species were compared. There are more unsaturated fatty acids in the crayfish than in the rabbit membranes. The most expressive differences in fatty acid composition between the two animals concern the content of linoleic acid type. Polyunsaturated acids in the crayfish are mostly of a 3 type and those in the rabbit of a 6 type. During biochemical evolution the following changes seem to take place, a decrease in the amount of unsaturated fatty acids as well as of polyunsaturated ones of a 3 type and an increase of those of the 6 type. These changes very probably reflect the transition from an aquatic to a terrestrial habitat.  相似文献

The action of reactive oxygen species, i.e. hydrogen peroxide, cupric sulphate and ascorbic acid, added at different concentrations to culture media, has been studied in two strains of Deinococcus radiodurans (red-pigmented parental and colourless mutant strains) in relation to their defense antioxidant systems. While the pigmented bacteria were more resistant to elevated concentrations of the different oxidants, the colourless bacteria were more sensitive and their sensitivity was dose-dependent. Reactive oxygen species induced oxidative damage, particularly to the polyethylenic fatty acids, which were more abundant in the mutant strain. Similarly, a significant increase in lipid peroxide levels was observed, whatever the chemical added during the growth of the mutant bacteria. The parental strain required high concentrations of oxidants to shorten its survival. Vitamins A and E, carotenoids and enzymes, largely present in the parental strain, could be responsible for its higher resistance to the lethal effects of radicals generated within the cells.  相似文献

Thirteen strains of mycobacteria isolated from deer and various species of wild birds were analysed by gas chromatography (GG) for cellular fatty acids and by thin-layer chromatography (TLG) for polar lipids. These strains were compared to reference strains of Mycobacterium avium, M. para tuberculosis and M. mal-moense. All the examined strains exhibited a generally similar fatty acid pattern characterized by relatively large amounts of hexadenca-noate (16:0), octadecenoate (18:1), octadecanoate (18:0) and 10-me-thyl-octadecanoate (tuberculostearic acid, 10-Me-18:0). Several additional acids were also generally present but in smaller amounts. By means of small but distinct differences in fatty acid composition, the wild animal isolates could be distinguished from both M. paratuber-culosis and M. malmoense but not from M. avium. The TLG polar lipid patterns on the other hand separated the wild animal isolates into 2 distinct groups of complex and simple polar lipid composition which corresponded to the morphologically smooth and rough types, respectively. The complex patterns of the smooth strains were comparable to those of the M. avium serovars whereas both the rough wild animal isolates and all the M. paratuber-culosis strains showed a simple pattern of polar lipids. Both fatty acid profiles and TLG polar lipid patterns support allocation of the wild animal isolates to the MAIS complex. Moreover, the 2 chemical techniques, particularly the GC procedure, are very useful for a more rapid and precise identification of the slow-growing wild animal mycobacterial isolates which have hitherto been characterized on basis of vague criteria.  相似文献

Total lipid fatty acid composition was investigated in brain hemispheres of male Spontaneously Hypertensive Rats (SHR), compared with normotensive Wistar Kyoto rats (WKY) used as controls. Both strains were suckled by adoptive Wistar mothers, and then fed a standard diet after weaning. No difference was observed between the two hemispheres of WKY killed either at 10 or 30 days. In SHR killed at 10 days, the two hemispheres showed differences, SHR left hemispheres exhibiting greater fatty acid composition changes than those of WKY, phenomenon that toned down at 30 days. Hence, SHR pups showed a different total lipid fatty acid composition of their brain hemispheres when compared with their WKY controls, though the two strains received the same diet. Genetically programmed hypertension might be, directly or not, involved in these changes.  相似文献

The aim of the present study was to examine the effect of acute streptozotocin diabetes on long chain fatty acid content and composition in different lipid classes of particular muscle types in the rat. Two days after streptozotocin administration, rats were anesthetised, and the white and red sections of the gastrocnemius, the soleus and the blood were taken. Lipids were extracted with chloroform/methanol and separated into different fractions (phospholipids, free fatty acids, di- and triacylglycerols) by means of thin layer chromatography. Fatty acids of each fraction were identified and quantified by means of gas-liquid chromatography. The diabetes resulted in elevation of the concentration of blood glucose (over four-fold) and the plasma free fatty acid (over two-fold). Total free fatty acid content in the muscles of diabetic rats increased by 26% in the white, 24% in the red gastrocnemius and 21% in the soleus. There were also changes in the composition of that fraction in each muscle. Diacylglycerol fatty acid content was elevated in both parts of the gastrocnemius (the white part by 15%, the red part by 44%) and remained stable in the soleus of the diabetic rats. The content of triacylglycerol fatty acids was elevated only in the red gastrocnemius in the diabetic group (by 112%), but changes in fatty acid composition in this fraction occurred in each muscle. The content of phospholipid fatty acids was elevated in the white gastrocnemius (by 13%) and remained stable in other muscles. There were only minor changes in phospholipid fatty acid composition in the diabetic rats. We concluded that acute insulin deficiency changes fatty acid content and composition in skeletal muscle lipids. The changes depend both on lipid fraction and muscle type.  相似文献

The rotational dynamics of spin-labelled fatty acids of different chainlengths (9, 10, 12, 14, 16 and 18 C-atoms) and different positions of labelling (5-C, 6-C and 7-C) have been studied in dimyristoylphosphatidylcholine bilayers using EPR spectroscopy. The segmental flexibility at a given label position is found to vary considerably with the length of the lipid chain, when this is less than that of the dimyristoylphosphatidylcholine host lipid. For both the charged and protonated forms of labelled fatty acids with chainlengths of 9, 10, and 12 C-atoms, the spectral anisotropy decreases steadily with decreasing chainlength in fluid phase bilayers. The differences become especially pronounced at the 7-C position of caprylic acid and the 6-C position of nonanoic acid, where the label is located close to the terminal methyl end of the chain. An unusually high degree of motional freedom is found for both these spin-labels, even in gel phase bilayers. There is relatively little effect of chainlength of the labelled fatty acid when this is longer or comparable to that of the host lipid (i.e., for fatty acid chainlengths of 18, 16 and 14 C-atoms), except if the label position is close to the terminal methyl end of the chain. The implications for the heterogeneous lipid chain composition in biological membranes are discussed.  相似文献

In order to correlate the lipid composition of the membrane of Lactobacillus acidophilus CRL 640 with the freeze-thaw behaviour of the cultures grown at different temperatures, fatty acid methyl esters (FAMEs) from extracts grown at 25, 30, 37 and 40 degrees C were obtained and compared. Cultures grown at 25 degrees C (M25) exhibited more resistance to the freeze-thaw process probably because of an increase in C18:2 and C16:0 fatty acids. This culture also exhibited a lesser amount of phospholipids as shown by the sugar: phosphorus ratio. In all cases, the presence of the uncommon 10-hydroxyoctadecanoic acid was determined. From the extracts of the M25 and M37 cultures, diacylphosphatidylglycerol, cardiolipin, diglycosyldiglycerides, triglycosyldiglycerides and neutral lipids were isolated and identified. The structural elucidation was carried out by FAMEs and sugar analysis and by mass spectrometry using fast atom bombardment ionization. The changes in lipid composition due to different growth temperatures could be indicative of the resistance of the bacteria to freeze-thaw processes.  相似文献

Lipid A is the hydrophobic anchor of lipopolysaccharide (LPS) in the outer membrane of Gram-negative bacteria. Lipid A of all Rhizobiaceae is acylated with a long fatty acid chain, 27-hydroxyoctacosanoic acid. Biosynthesis of this long acyl substitution requires a special acyl carrier protein, AcpXL, which serves as a donor of C28 (omega-1)-hydroxylated fatty acid for acylation of rhizobial lipid A (Brozek, K.A., Carlson, R.W., and Raetz, C. R. (1996) J. Biol. Chem. 271, 32126-32136). To determine the biological function of the C28 acylation of lipid A, we constructed an acpXL mutant of Sinorhizobium meliloti strain 1021. Gas-liquid chromatography and mass spectrometry analysis of the fatty acid composition showed that the acpXL mutation indeed blocked C28 acylation of lipid A. SDS-PAGE analysis of acpXL mutant LPS revealed only a fast migrating band, rough LPS, whereas the parental strain 1021 manifested both rough and smooth LPS. Regardless of this, the LPS of parental and mutant strains had a similar sugar composition and exposed the same antigenic epitopes, implying that different electrophoretic profiles might account for different aggregation properties of LPS molecules with and without a long acyl chain. The acpXL mutant of strain 1021 displayed sensitivity to deoxycholate, delayed nodulation of Medicago sativa, and a reduced competitive ability. However, nodules elicited by this mutant on roots of M. sativa and Medicago truncatula had a normal morphology and fixed nitrogen. Thus, the C28 fatty acid moiety of lipid A is not crucial, but it is beneficial for establishing an effective symbiosis with host plants. acpXL lies upstream from a cluster of five genes, including msbB (lpxXL), which might be also involved in biosynthesis and transfer of the C28 fatty acid to the lipid A precursor.  相似文献

Cells of four reference strains (Scott A, LO 28, CNL 895807 and ATCC 19115) and of five recent food isolates (A00M011, A00M018, A00M087, A00M092 and A00M123) of Listeria monocytogenes were grown until late exponential phase in Brain Heart Broth at two different temperatures (37 °C and 4 °C). Our results show that significant differences exist between the cellular lipid fatty acid profile of reference and recent food isolates. Like the reference strains, and in keeping with previous reports on the cellular lipid fatty acid profile of L. monocytogenes, the recent food isolates were characterised by the presence of ai15:0, i15:0 and ai17:0. In addition, the fatty acid ai13:0 was observed in all of the recent food isolates grown at 4 °C, whereas only two reference strains, Scott A and LO 28, showed ai13:0 in their cellular lipid fatty acid profile at 4 °C. When grown at 4 °C, the recent food isolates showed a mean aiC15/aiC17 ratio of 66, while reference strains were characterised by significantly lower ratios, ranging between 4.3 (ATCC 19115) and 28.9 (Scott A). These results showed that all of the recent food isolates, Scott A and LO28 strains use chain length and anteiso-branching (ai15:0) as their major response to cold temperature adaptation. However, the cold adaptation response of reference strains CNL 895807 and ATCC 19115 appears to be different.  相似文献

Inducible pigmentation changes were observed in pigmented strains of Brevibacterium sp. M27 and B. flavum treated with N-methyl-N'-nitro-N-nitrosoguanidine. The highest frequency of induction was reached already at a survival of 30-40% with the maximal yield of 6-10%. As compared with the initial yellow colour, three new pigmentation types, viz. white, pink and orange, were observed. The yellow pigmented parent strains are most resistant to the lethal effects of UV radiation. By selecting pigmented mutants of all types on media containing antibiotics it was possible to obtain strains that were resistant either to tetracycline or to streptomycin. Auxotrophic pigmented mutants were also isolated. In multiple mutant strains of Brevibacterium sp. M27 a number of strainsexhibited a changed L-lysine production. In some strains the production was variable, whereasother strains did not produce L-lysine at all and stains with a limited production of other amino acids were also detected.  相似文献

Thermoacidophilic and halotolerant microorganisms from the Antarctic continent were studied for their lipid modulation under stress growth conditions. Temperature-induced changes in complex lipids and fatty acids of four strains belonging to the genus Alicyclobacillus involved the relative proportions of different polar lipids and the synthesis of ω-cyclohexyl-acyl chains, which were favoured by high temperatures. Studies were carried out on the lipid composition of four strains of extremely halotolerant bacteria belonging to the genus Micrococcus grown at different salt concentrations from 0 up to 4.5 M NaCl. The main lipids found were two unidentified glycolipids and two phospholipids: 1,2 diacylglycero-3-phosphoryl-glycerol (PG) and cardiolipin (DPG). Among the strains analysed, the lipids of the Micrococcus strain Erebus were shown to be strongly influenced by salt concentrations, in that DPG and one glycolipid were absent at a low salt molarity while, under these conditions, PG was the main lipid found. The predominant fatty acids in all halotolerant strains were of the anteiso type; growth under increasing salinity gave rise to an increase in long chain fatty acids and of straight chain fatty acids, while a decrease in iso fatty acids occurred. The lipid composition of fluorescent vibrios, V. eltor and nonagglutinating vibrios has been studied. In the fraction of polar lipids phosphatidylethanolamine, phosphatidylinositol and cardiolipin and in the fraction of neutral lipids monoglycerides, free fatty acids, diglycerides, triglycerides, sterol esters have been identified. The fatty acid composition of some classes of neutral lipids have been determined. Both similarity and differences between the strains under study in their lipid and fatty acid composition have been established.  相似文献
