海栖热袍菌(Thermotoga maritima)是嗜极端高温的厌氧细菌,其产生的葡萄糖异构酶由于其出色的耐热性有着潜在的工业应用价值.由于海栖热袍菌苛刻的培养条件导致其葡萄糖异构酶产量较低.通过PCR方法克隆编码T. maritima MSB8葡萄糖异构酶基因xylA,构建重组质粒pHsh-xylA,转入Escherichia coli JM109,通过热激诱导表达.通过热处理和离子交换层析纯化两步得到电泳纯的酶制品,纯化倍数和回收率分别为8.02和49.02.对酶学性质研究表明,该重组酶为金属离子激活性酶,Mg2 ,Co2 对相对酶活有很强的激活作用,其最适pH为7.0,最适反应温度为95℃,且在pH 6~8之间有着较好的稳定性,在95℃下半衰期长达5 h以上.以葡萄糖为底物时的表观Km和Vmax分别为105 mmol/L和45.2 mol/min·mg.  相似文献

嗜热链霉菌变株(Streptomyces diastaticus No.7mutant strain)M1033—9菌的胞外葡萄糖异构酶作用最适温度为80—85℃,最适pH为8—9,pH大于7.8时有较强的热稳定性,Co2+和Mg2+对酶有激活作用,Co2+对酶的热稳定性有保护作用。5000L发酵罐经板框过滤的清发酵液含胞外酶885u/ml,占总酶活94%,可直接作酶源吸附于大孔阴离子交换树脂上制成固定化葡萄糖异构酶(酶活13000u/g(干))工艺简便。年产2000吨42型果葡糖浆规模下每公斤干固定化酶可生产果葡糖浆4 59吨。M1033—9是产胞外葡萄糖异构酶的优良菌种。  相似文献

氧化铝为载体的固定化葡萄糖异构酶某些性质的研究   总被引:2,自引:0,他引:2  
本文报道了以廉价的大孔氧化铝作为葡萄糖异构酶的固定化载体。该固定化酶的热稳定性是60—70℃,如高于70℃就开始失活。最适Ph和温度分别为7.2—8.0和70℃。Gu2+、Fe2+、Zn2+、Mn2+和Ba2+等金属离子对它的酶活有抑制作用,Co2+和Mg2+金属离子却有明显的激活作用。该固定化酶酶活是6000—7000μ/g。通过柱运转,它的半衰期为40天以上,转化率是葡萄糖异构为果糖理论值的80一85%。  相似文献

以树状黄杆菌(Flavobacteriumaraborescens)NRRL11022为出发菌株,用紫外线对其进行诱变,经筛选得到一株葡萄糖异构酶的高产菌株U-616,其酶活力提高31%。经保存三年和多次传代复测,其产酶能力保持稳定。其生长和产酶需较高的溶氧水平,最适产酶温度为30℃,最适产酶pH为7.0-7.5,铁离子对其生长和产酶无明显的影响。所产葡萄糖异构酶的最适温度为60-80℃,最适pH为7.5-8.5,Co2+和Mg2+对酶有激活作用,对金属离子耐受性较强,对Ca2+不敏感,热稳定性较好。树状黄杆菌变株U-616是一株产胞内葡萄糖异构酶的优良菌株。  相似文献

从野外采集并筛选到一株能降解黄原胶的菌株,并对菌种进行了对照鉴定。此外,对菌株的发酵参数、黄原胶降解酶的性质(最适反应温度、pH值,金属离子的影响、底物专一性、动力学研究等)作了初步的研究。结果显示,该酶的最适催化反应温度和pH分别为30℃~40℃和5.0~7.0;能够专一性降解黄原胶;测试大多数金属离子对酶活力没有明显影响,但Ca^2 能很大程度地缓解EDTA对酶活的抑制作用。  相似文献

【目的】嗜热链球菌(Streptococcus thermophilus)作为酸奶、奶酪等发酵乳制品的常用发酵剂,在乳品工业中应用广泛。大多数嗜热链球菌为半乳糖阴性(Gal-)菌株,不能代谢半乳糖而将其排出到胞外,导致发酵乳中半乳糖含量增加。因此可通过化学诱变的方法处理嗜热链球菌,使其可代谢半乳糖,开发一种低半乳糖含量的发酵乳。【方法】利用亚硝基胍(nitrosoguanidine, NTG)对嗜热链球菌IMAU80846进行诱变处理,测定野生株嗜热链球菌IMAU80846与突变株β-半乳糖苷酶(β-galactosidase,β-Gal)、半乳糖激酶(galactokinase, GalK)、丙酮酸激酶(pyruvate kinase, PK)和葡萄糖激酶(glucokinase, GK)活性,分析编码这些酶的氨基酸序列,得到一株可代谢半乳糖的嗜热链球菌IMAU80846Y,同时对突变株进行全基因组测序,并检测野生株和突变株发酵乳中乳酸、乳糖、半乳糖和葡萄糖的含量,最后将野生株和突变株分别与德氏乳杆菌保加利亚亚种IMAU20450进行复配发酵,测定两组发酵乳在发酵与贮藏期间的发酵特性,制备一款低半乳糖含量的发酵乳。【结果】突变株嗜热链球菌IMAU80846Y的β-Gal、GalK活性高于野生株,PK和GK活性低于野生株,氨基酸序列与全基因组测序结果表明突变株与糖代谢有关基因发生突变,HPLC检测结果表明突变株发酵乳中乳糖、半乳糖的含量均低于野生株,而乳酸、葡萄糖的含量高于野生株。发酵特性的分析结果表明,与野生株复配组相比,突变株复配组发酵乳具有良好的滴定酸度、活菌数、黏度以及持水力。【结论】嗜热链球菌IMAU80846经NTG诱变后,菌株代谢半乳糖的能力发生了变化,利用该突变株可生产一款低半乳糖含量的发酵乳。  相似文献

从贵州、云南、海南、安徽、四川、辽宁等地采集碱性土样,分离筛选到1株能稳定的产生碱性纤维素酶的嗜碱性芽孢杆菌AH-8。研究表明,该菌株最适产酶温度为37℃,最适发酵时间为36 h;采用均匀设计法对其发酵培养基进行优化,优化培养基配方(%):淀粉3.0,胰蛋白胨1.5,牛肉膏1.5,葡萄糖0.3,KH2PO40.1,初始pH 10.0。在优化培养基条件下,其产酶量提高了120%。碱性纤维素酶最适反应温度为60℃;最适反应pH 10.0;0.01%Co2 对酶活力有一定激活作用。  相似文献

从2株芽胞杆菌中通过筛选获得了一株产α-葡萄糖苷酶活力较高的嗜热脂肪芽孢杆菌U2,以嗜热芽孢杆菌U2为菌种,优化发酵培养基后,在温度45℃、初始pH6.8、转速200r/min和10%接种量条件下,发酵20h,菌株U2产酶水平可达到2.62U/mL,比出发菌提高了4倍。  相似文献

一株嗜热子囊菌产生的碱性耐热过氧化氢酶及其应用潜力   总被引:11,自引:0,他引:11  
研究了一株嗜热子囊菌产过氧化氢酶的摇瓶发酵条件,并对其在纺织工业中的应用潜力进行了评价。以20 g/L糊精和1%(V/V)乙醇为混合碳源时,过氧化氢酶酶活达到1594 u/Ml,比以糊精和乙醇单独为碳源时过氧化氢酶的活力之和还高23%。改变培养基的初始Ph、提高发酵液中的溶氧水平及添加外源过氧化氢,过氧化氢酶的产量进一步提高到2762 u/Ml,比优化前提高了5.8倍。将嗜热子囊菌的过氧化氢酶同来源于牛肝、黑曲霉的过氧化氢酶进行了热(70℃, 80℃, 90℃)、碱(Ph 9.0, Ph 10.0, Ph 11.0)稳定性的比较。结果显示,产自嗜热子囊菌的过氧化氢酶对高温和强碱性的耐受性能明显优于其它来源的酶,在纺织染整工艺中具有良好的应用潜力。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
