The effect of Ni supply on growth, N metabolism and leaf urease activity of six plant species (rye, wheat, soybean, rape, zucchini and sunflower) grown on urea-based nutrient solutions was investigated. Activity of urease, which is activated by Ni, was hardly detectable in plants of all six species grown without supplementary Ni. As a consequence Ni-deprived plants accumulated considerable amounts of urea, showed a reduced dry matter production and reduced total N concentrations. The lack of urease activation in low-Ni plants made them metabolically N deficient, which is illustrated by the chlorotic appearance of these plants. The soluble amino acid N concentration was reduced by inadequate Ni supply. The amide concentrations (glutamine, asparagine) were considerably affected in all species. The same applied to the concentrations of the urea cycle intermediates arginine, ornithine, and citrulline. These results stress the necessity of Ni for urease activation and thus for the growth of plants on urea-based media.  相似文献

Nickel is considered to be an essential micronutrient in plants because of its role in the metalloenzyme urease. In order to characterize the metabolic consequences of Ni deprivation, the significance of Ni supply for growth and N metabolism of rice plants grown with either NH4NO3 or urea as sole N source was evaluated. Growth of plants receiving NH4NO3 was not affected by the Ni status, and neither were the activities of arginase and glutamine synthetase. However, urease activity was not detectable in leaves of low-Ni plants, which in conjunction with arginase action, led to the accumulation of urea in plants grown with NH4NO3. Amino acid contents and mineral nutrient status (except Ni) were not affected by the Ni treatment.Urea-grown Ni-deprived plants showed reduced growth and accumulated large amounts of urea owing to the lack of urease activity. These plants were further characterized by low amino acid contents indicating impaired usage of the N supplied. They also exhibited reduced levels of the urea precursor arginine, which is merely attributed to the overall N economy in these plant. When urea-grown plants were supplied with 0.5 mmol m-3 Ni in the nutrient solution, the dry weight and the amino acid N contents were increased at the expense of the urea contents, indicating efficient use of urea N in Ni-supplemented plants.A critical Ni concentration in the shoot regarding dry matter production of NH4NO3-grown plants could not be deduced, while 25 g Ni kg-1 DW is certainly inadequate for urea-grown plants. The significance of nickel (Ni), which is essential for ureaseactivity, for growth and nitrogen (N) metabolism ofBrassicanapusgrown in nutrient solution with either NH4NO3or urea assole N source was investigated. Although Ni contents were below25 µg kg-1d. wt, growth of plants relying on NH4NO3wasnot affected by the Ni status. However, supplementing the growthmedium with 0.04 µMNi enhanced dry matter production ofurea-grown plants significantly. Urease activity was significantlyreduced in leaves and roots of plants grown without supplementaryNi irrespective of N source. Plants grown with urea withoutadditional Ni accumulated large amounts of urea and had loweramino acid contents indicating impaired usage of the N supplied,while those grown with NH4NO3under Ni-deprived conditions accumulatedendogenous urea in their older leaves. It is suggested thatNi may not be strictly essential for plants receiving mineralN, or that the critical level is well below 25 µg kg-1d.wt. Background and aims

Urea is the major nitrogen (N) form supplied as fertilizer in agriculture. However, urease, a nickel-dependent enzyme, allows plants to use external or internally generated urea as a nitrogen source. Since a urease inhibitor is frequently applied in conjunction with urea fertilizer, the N-metabolism of plants may be affected. The aim of this study was to determine physiological and molecular effects of nickel deficiency and a urease inhibitor on urea uptake and assimilation in oilseed rape.

Methods

Plants were grown on hydroponic solution with urea as the sole N source under three treatments: plants treated with nickel (+Ni) as a control, without nickel (?Ni) and with nickel and phenylphosphorodiamidate (+Ni+PPD). Urea transport and assimilation were investigated.

Results

The results show that Ni-deficiency or PPD supply led to reduced growth and reduced 15N-uptake from urea. This effect was more pronounced in PPD-treated plants, which accumulated high amounts of urea and ammonium. Thus, Ni-deficiency or addition of PPD, limit the availability of N and decreased shoot and root amino acid content. The up-regulation of BnDUR3 in roots indicated that this gene is a component of the stress response to nitrogen-deficiency. A general decline of glutamine synthetase (GS) activity and activation of glutamate dehydrogenase (GDH) and increases in its expression level were observed in control plants. At the same time, in (?N) or (+Ni+PPD) treated plants, no increases in GS or GDH activities and expression level were found.

Conclusions

Overall results showed that plants require Ni as a nutrient (while most widely used nutrient solutions are devoid of Ni), whether they are grown with or without a urea supply, and that urease inhibitors may have deleterious effects at least in hydroponic grown oilseed rape.  相似文献

Cowpeas grown in nutrient solutions, from which Ni had been removed by a ligand exchange technique, accumulated urea in most tissues. Urea levels were highest (up to 3.1 percent dry weight) in necrotic leaf tips. Urea accumulation in Ni-deficient cowpea tissues amounted to about 1 percent of the total N. The accumulation of urea was presumably associated with the catabolism of N compounds in older tissues and the redistribution of N catabolites within the plant during the reproductive growth. The exclusion of N salts from the nutrient media at a late stage of growth, either with or without added Ni, led to a general amelioration of urea accumulation and a lower level of the related amino acid, arginine, in root and stem tissue. Plant leaves that contained toxic levels of urea and displayed necrotic symptoms had tissue Ni levels ranging from less than 0.01 to 0.15 μg Ni per gram dry weight. Nickel concentrations in tissue from plants not treated with Ni, were initially very low, but increased as the cowpeas matured. Apparently, there was a source of Ni contamination in the Ni-deficient growth media which provided a source of Ni for uptake by the plants during growth. Ureide levels were low and unaffected by Ni deprivation. No evidence for free purines or uric acid accumulation in plant tissues could be found. It is hypothesized that Ni (and urease) participates in the normal N metabolism of these plants during the reproductive phase of growth.  相似文献

In order to evaluate whether Co can functionally replace Ni in urease, as suggested by Watanabe et al. (Soil Sci. Plant Nutr. (1994) 40, 545–548), zucchini and soybean plants were grown in purified nutrient solutions containing nitrate and ammonium as sole N source supplemented either with 0.05 mmol m-3 Ni or Co or with no supplementation (control). In addition, isolated soybean cotyledons were incubated axenically with either 1 mmol m-3 Ni or Co supplements. Plant growth was not affected by the Ni and Co additions, but urease activity was only detected in Ni-supplemented tissue, and in no case did Co raise the urease activity level. Only Ni-deprived plants (–Ni – Co and –Ni + Co) accumulated appreciable amounts of urea in the leaves. These results strongly suggest, that Co does not replace Ni with respect to functional urease.  相似文献

The detrimental effects of excessive Ni on plant growth have been well known for many years. More recent evidence indicates that Ni is required in small amounts for normal plant growth and development. Ni is an essential component of urease in plants and microorganisms. A deficiency of Ni in plants is reported to result in necrotic lesions in leaves in response to toxic accumulations of urea. Urease plays an essential role in mobilization of nitrogenous compounds in plants, a process that is especially important during seed germination and fruit formation when protein reserves are degraded into amino acids. Arginine, an abundant amino acid in plants, when degraded produces urea as a product and urease is needed for urea utilization. Theories of urea formation during allantoin degradation in Glycine max have been recently refuted. In G. max ureides apparently are metabolized via an amidohydrolase reaction with subsequent degradation of ureidoglycine, yielding glyoxylate, NH+4 and CO2. No evidence is available for the formation of urea in this pathway. Nitrogen-fixing symbionts, such as Rhizobium and Bradyrhizobium, contain two known Ni enzymes: urease and hydrogenase. Optimum growth of nodulated legumes and actinorhizal plants may depend on an adequate supply of Ni to meet the requirements of the Ni-requiring enzymes in host plants and endophytes. The seeds of severely Ni-deficient Hordeum are completely inviable, thus providing conclusive evidence for the essentiality of Ni for this species. The evidence indicates that Ni must be added to the list of micronutrient elements generally required by plants.  相似文献

The influence of urease activity on N distribution and losses after foliar urea application was investigated using wild-type and transgenic potato (Solanum tuberosum cv Désirée) plants in which urease activity was down-regulated. A good correlation between urease activity and (15)N urea metabolism (NH(3) accumulation) was found. The general accumulation of ammonium in leaves treated with urea indicated that urease activity is not rate limiting, at least initially, for the assimilation of urea N by the plant. It is surprising that there was no effect of urease activity on either N losses or (15)N distribution in the plants after foliar urea application. Experiments with wild-type plants in the field using foliar-applied (15)N urea demonstrated an initial rapid export of N from urea-treated leaves to the tubers within 48 h, followed by a more gradual redistribution during the subsequent days. Only 10% to 18% of urea N applied was lost (presumably because of NH(3) volatilization) in contrast to far greater losses reported in several other studies. The pattern of urease activity in the canopy was investigated during plant development. The activity per unit protein increased up to 10-fold with leaf and plant age, suggesting a correlation with increased N recycling in senescing tissues. Whereas several reports have claimed that plant urease is inducible by urea, no evidence for urease induction could be found in potato.  相似文献

To study the effect of root-zone pH on characteristic responses of NH4+ -fed plants, soybeans (Glycine max?L.? Merr. cv. Ransom) were grown in flowing solution culture for 21 d on four sources of N (1.0 mol m-3 NO3-, 0.67 mol m-3 NO3- plus 0.33 mol m-3 NH4+, 0.33 mol m-3 NO3- plus 0.67 mol m-3 NH4+, and 1.0 mol m-3 NH4+) with nutrient solutions maintained at pH 6.0, 5.5, 5.0, and 4.5. Amino acid concentration increased in plants grown with NH4+ as the sole source of N at all pH levels. Total amino acid concentration in the roots of NH4+ -fed plants was 8 to 10 times higher than in NO3(-)-fed plants, with asparagine accounting for more than 70% of the total in the roots of these plants. The concentration of soluble carbohydrates in the leaves of NH4+ -fed plants was greater than that of NO3(-)-fed plants, but was lower in roots of NH4+ -fed plants, regardless of pH. Starch concentration was only slightly affected by N source or root-zone pH. At all levels of pH tested, organic acid concentration in leaves was much lower when NH4+ was the sole N source than when all or part of the N was supplied as NO3-. Plants grown with mixed NO3- plus NH4+ N sources were generally intermediate between NO3(-)- and NH4+ -fed plants. Thus, changes in tissue composition characteristic of NH4+ nutrition when root-zone pH was maintained at 4.5 and growth was reduced, still occurred when pH was maintained at 5.0 or above, where growth was not affected. The changes were slightly greater at pH 4.5 than at higher pH levels.  相似文献

Background and aims

The importance of seed Ni reserves for plant growth and N metabolism is poorly understood. This study investigated the effects of both seed Ni and externally supplied Ni on the impact of foliarly-applied urea and N-nutritional status of soybean.

Methods

Soybean seeds were produced by growing plants in nutrient solutions containing different Ni levels, and their urease activities were measured. Plants were then grown from these seeds with or without external Ni. After treating half of the plants with foliar urea, the urea damage symptoms, elongation rates and chlorophyll concentrations were followed over one week. Biomass and mineral concentrations of different plant parts were determined.

Results

Nickel supply at increasing rates improved seed yield by up to 25 %. Seeds with Ni concentrations varying between 0.04–8.32 mg.kg?1 were obtained. Depending on the Ni concentration, the seed urease activities differed up to 100-fold. Leaf damage due to foliar urea spray was significantly alleviated by higher seed Ni as well as external Ni supply. Higher Ni also promoted shoot elongation and improved chlorophyll concentrations. Nickel was 10-times more concentrated in the youngest part than in older leaves. In the absence of foliar urea, Ni enhanced the N concentration of the growing part of the shoot by up to 30 %.

Conclusion

A better utilization of foliarly-applied urea-N is achieved in soybean when adequate Ni is supplied to plants by seed reserves and/or externally. High seed Ni levels are also required for preventing foliar urea damage and improving N remobilization.  相似文献

Gene expression and protein accumulation patterns of nitrogen-responsive lipoxygenase (LOX-NR), as a representative vegetative storage protein, were investigated in nonnodulated soybeans (Glycine max [L.] Merr. cv Wye). The form of available nitrogen (supplied as NH4NO3, NH4+, NO3-, or urea) influenced the mRNA level and the amount of LOX protein, indicating that preferential accumulation of LOX may occur. Soybeans were grown with 0, 2, 5, and 16 mM total nitrogen to determine the extent to which LOX accumulation responded to soil nitrogen levels. Analysis of both mRNA and protein levels was conducted in shoot tips, stems, pod walls, and leaves over the entire life cycle of the plant. A general correlation between increasing available nitrogen level and LOX level was seen in the shoot tip and other organs throughout the soybean life cycle. However, appreciable amounts of LOX-NR mRNA and protein accumulated even when plants were grown under conditions of nitrogen deficiency. The results indicate that LOX may play an important role as a temporary storage site for amino acids in the developing shoot tip. The expression patterns of LOX-NR in plants grown under nitrogen deficiency suggest that these proteins, although responsive to nitrogen status, may not function solely as temporary storage pools for amino acids.  相似文献

The application of urease inhibitors in conjunction with urea fertilizers as a means of reducing N loss due to ammonia volatilization requires an in-depth study of the physiological effects of these inhibitors on plants. The aim of this study was to determine how the urease inhibitor N-(n-butyl) thiophosphoric triamide (NBPT) affects N metabolism in pea and spinach. Plants were cultivated in pure hydroponic culture with urea as the sole N source. After 2 weeks of growth for pea, and 3 weeks for spinach, half of the plants received NBPT in their nutrient solution. Urease activity, urea and ammonium content, free amino acid composition and soluble protein were determined in leaves and roots at days 0, 1, 2, 4, 7 and 9, and the NBPT content in these tissues was determined 48 h after inhibitor application. The results suggest that the effects of NBPT on spinach and pea urease activity differ, with pea being most affected by this treatment, and that the NBPT absorbed by the plant caused a clear inhibition of the urease activity in pea leaf and roots. The high urea concentration observed in leaves was associated with the development of necrotic leaf margins, and was further evidence of NBPT inhibition in these plants. A decrease in the ammonium content in roots, where N assimilation mainly takes place, was also observed. Consequently, total amino acid contents were drastically reduced upon NBPT treatment, indicating a strong alteration of the N metabolism. Furthermore, the amino acid profile showed that amidic amino acids were major components of the reduced pool of amino acids. In contrast, NBPT was absorbed to a much lesser degree by spinach plants than pea plants (35% less) and did not produce a clear inhibition of urease activity in this species.  相似文献

BACKGROUND AND AIMS: Tea (Camellia sinensis) is considered to be acid tolerant and prefers ammonium nutrition, but the interaction between root zone acidity and N form is not properly understood. The present study was performed to characterize their interaction with respect to growth and mineral nutrition. METHODS: Tea plants were hydroponically cultured with NH4+, NO3- and NH(4+) + NO3-, at pH 4.0, 5.0 and 6.0, which were maintained by pH stat systems. KEY RESULTS: Plants supplied with NO3- showed yellowish leaves resembling nitrogen deficiency and grew much slower than those receiving NH4+ or NH(4+) + NO3- irrespective of root-zone pH. Absorption of NH4+ was 2- to 3.4-fold faster than NO3- when supplied separately, and 6- to 16-fold faster when supplied simultaneously. Nitrate-grown plants had significantly reduced glutamine synthetase activity, and lower concentrations of total N, free amino acids and glucose in the roots, but higher concentrations of cations and carboxylates (mainly oxalate) than those grown with NH4+ or NH(4+) + NO3-. Biomass production was largest at pH 5.0 regardless of N form, and was drastically reduced by a combination of high root-zone pH and NO3-. Low root-zone pH reduced root growth only in NO(3-)-fed plants. Absorption of N followed a similar pattern as root-zone pH changed, showing highest uptake rates at pH 5.0. The concentrations of total N, free amino acids, sugars and the activity of GS were generally not influenced by pH, whereas the concentrations of cations and carboxylates were generally increased with increasing root-zone pH. CONCLUSIONS: Tea plants are well-adapted to NH(4+)-rich environments by exhibiting a high capacity for NH4+ assimilation in their roots, reflected in strongly increased key enzyme activities and improved carbohydrate status. The poor plant growth with NO3- was largely associated with inefficient absorption of this N source. Decreased growth caused by inappropriate external pH corresponded well with the declining absorption of nitrogen.  相似文献

Plant-atmosphere NH(3) exchange was studied in white clover (Trifolium repens L. cv. Seminole) growing in nutrient solution containing 0 (N(2) based), 0.5 (low N) or 4.5 (high N) mM NO(3)(-). The aim was to show whether the NH(3) exchange potential is influenced by the proportion of N(2) fixation relative to NO(3)(-) supply. During the treatment, inhibition of N(2) fixation by NO(3)(-) was followed by in situ determination of total nitrogenase activity (TNA), and stomatal NH(3) compensation points (chi(NH(3))) were calculated on the basis of apoplastic NH4(+) concentration ([NH4(+)]) and pH. Whole-plant NH(3) exchange, transpiration and net CO(2) exchange were continuously recorded with a controlled cuvette system. Although shoot total N concentration increased with the level of mineral N application, tissue and apoplastic [NH4(+)] as well as chi(NH(3)) were equal in the three treatments. In NH(3)-free air, net NH(3) emission rates of <1 nmol m(-2) s(-1) were observed in both high-N and N(2)-based plants. When plants were supplied with air containing 40 nmol mol(-1) NH(3), the resulting net NH(3) uptake was higher in plants which acquired N exclusively from symbiotic N(2) fixation, compared to NO(3)(-) grown plants. The results indicate that symbiotic N(2) fixation and mineral N acquisition in white clover are balanced with respect to the NH4(+) pool leading to equal chi(NH(3)) in plants growing with or without NO(3)(-). At atmospheric NH(3) concentrations exceeding chi(NH(3)), the NH(3) uptake rate is controlled by the N demand of the plants.  相似文献

Background and aims

Nickel (Ni) has a critical role in the urea metabolism of plants. This study investigated the impact of seed Ni content along with external Ni supply on the growth, various nitrogen (N) metabolites and N use efficiency (NUE) of soybean plants.

Methods

Soybean plants raised from Ni-poor or Ni-rich seeds were grown in nutrient solution with or without external Ni supply and fed with either urea or nitrate as the sole N source. The changes in growth, leaf chlorophyll levels, Ni and N concentrations of different plant parts, tissue accumulation of various N metabolites and N uptake of soybean as well as NUE and its components were examined.

Results

Nickel starvation reduced the shoot biomass of urea-fed plants by 25 % and the leaf chlorophyll levels by up to 35 %, but nitrate-fed plants were unaffected. Visual toxicity symptoms were not observed in urea-fed plants. Under urea supply, Ni-deficient plants had lower levels of total N, protein and free amino acids in various organs. Root uptake of urea was severely depressed in Ni-deprived plants. Availability of Ni did not have any effect on the NUE of nitrate-fed plants, whereas its deficiency reduced the NUE of urea-fed plants by 30 %. The growth and N nutritional status of urea-fed soybean were significantly improved by high seed Ni reserves as well as external Ni supply.

Conclusion

Adequate Ni supply is required for maximizing the growth, root uptake of urea and NUE of urea-fed plants. Seed Ni reserves contribute significantly to the Ni and thus N nutritional status of soybean.  相似文献

 生长在供给NO-3 N、NH+4 N和NH4NO3 N氮源下的荫香(Cinnamomum burmanni)幼树暴露在增高空气NH3浓度下30 d。利用气体交换测定和氮分析研究了植株的光合作用、氮利用和氮在光合过程一些组分中的分配,根据Farquhar-von Caemmerer模式得出相关光合参数。结果表明在增高空气NH3下生长于NO-3 N的植株Rubisco最大羧化速率(Vcmax)和最大光合电子传递速率(Jmax)较正常空气下的高,但生长于NH+4 N和NH4NO3 N的植株则较正常空气下的低。无论生长于何种形式氮下的植株,在空气NH3增高下以单位叶面积为基准的叶氮含量(Na)显著增高(p<0.05)。在增高空气NH3下,生长于NO-3 N下的植株,其类囊体氮量(NT)、Rubisco氮(NR)和结合于光合电子传递链的氮(NE)的含量较正常空气下的增高(p<0.05);而生长于NH+4 N和NH4NO3 N下的植株则较正常空气下的低。表明在空气NH3增高下生长于NO-3 N的植株能有效地利用氮合成光合过程必要的组份,而生长于NH+4 N和NH4NO-3 N的植株氮在NT、NR和NE的分配受到部分限制。在空气NH3增高下生长于NO-3 N和NH4NO3 N的植株,其以单位干重为基准的有机氮量较正常空气下的高,但生长于NH+4 N的植株则较正常空气下的低,此外在空气NH3增高下生长于NO-3 N的植株的可溶性蛋白氮较正常空气下增高,而生长在NH+4 N的植株亦见降低。结果表明空气NH3增高可能有利于NO-3 N下生长的荫香植株利用空气中的氮,促进叶片光合速率提高,而空气NH3增高能抑制NH+4 N或NH4NO3 N下生长的荫香植株光合作用和氮的利用和再分配。  相似文献

To study the direct effects of photosynthesis on allocation of biomass by altering photosynthesis without altering leaf N or nitrate content, phosphoribulokinase (PRK) activity was decreased in transgenic tobacco (Nicotiana tabacum L.) with an inverted tobacco PRK cDNA and plants were grown at different N levels (0.4 and 5 mM NH4NO3). The activation state of PRK increased as the amount of enzyme was decreased genetically at both levels of N. At high N a 94% decrease in PRK activity had only a small effect (20%) on photosynthesis and growth. At low N a 94% decrease in PRK activity had a greater effect on leaf photosynthesis (decreased by up to 50%) and whole-plant photosynthesis (decreased by up to 35%) than at high N. These plants were up to 35% smaller than plants with higher PRK activities because they had less structural dry matter and less starch, which was decreased by 3- to 4-fold, but still accumulated to 24% to 31% of dry weight; young leaves contained more starch than older leaves in older plants. Leaves had a higher ion and water content, and specific leaf area was higher, but allocation between shoot and root was unaltered. The hypothesis that soybean (Glycine max L. [Merrill]) catabolizes ureides to urea to a physiologically significant extent was tested and rejected. Urease-negative (eu3-e1/eu3-e1) plants were supported by fixed N2 or by 2 mM NH4NO3, so that xylem-borne nitrogen contained predominantly ureides (allantoin and allantoic acid) or amide amino acids, respectively. Seed nitrogen yield was equal on either nitrogen regime, although 35-d-old fixing plants accumulated about 6 times more leaf urea. In callus, lack of an active urease reduced growth on either arginine or allantoin as the sole nitrogen source, but the reduction was greater on arginine (73%) than on allantoin (39%). Furthermore, urease-negative cells accumulated 17 times more urea than urease-positive cells on arginine; for allantoin the ratio was 1.8. Urease-negative callus accumulated urea at 3% the rate of seedlings. To test whether urea accumulating in urease-negative seedlings was derived from ureides, seeds were first allowed to imbibe in 1 mM allopurinol, an inhibitor of ureide formation. Seedling ureides were decreased by 90%, but urea levels were unchanged. Thus, ureides are poor precursors of urea, which was confirmed in seedlings that converted no more than 5% of seed-absorbed [14C-ureido]allantoate to [14C]urea, whereas 40 to 70% of [14C-guanido]arginine was recovered as [14C]urea.  相似文献

Certain plant species have been shown to vigorously accumulate some metals from soil, and thus represent promising and effective remediation alternatives. In order to select the optimum forms of nitrogen (N) fertilizers for the arsenic (As) hyperaccumulator, Pteris vittata L., to maximize As extraction, five forms of N were added individually to different treatments to study the effect of N forms on As uptake of the plants under soil culture in a greenhouse. Although shoot As concentration tended to decrease and As translocation from root to shoot was inhibited, overall As accumulation was greater due to higher biomass when N fertilizer was added. Arsenic accumulation in plants with N fertilization was 100-300% more than in the plants without N fertilization. There were obvious differences in plant biomass and As accumulation among the N forms, i.e., NH4HCO3, (NH4)2S04, Ca(NO3)2, KNO3, urea. The total As accumulation in the plants grown in As-supplied soil, under different forms of N fertilizer, decreased as NH4HCO3>(NH4)2S04 > urea > Ca(NO3)2 >KNO3>CK. The plants treated with N and As accumulated up to 5.3-7.97 mg As/pot and removed 3.7-5.5% As from the soils, compared to approximately 2.3% of As removal in the control. NH4+ -N was apparently more effective than other N fertilizers in stimulating As removal when soil was supplied with As at initiation. No significant differences in available As were found among different forms of N fertilizer after phytoremediation. It is concluded that NH4+ -N was the preferable fertilizer for P. vittata to maximize As removal.  相似文献
