We describe 12 microsatellite loci isolated from lake trout (Salvelinus namaycush). The number of alleles at these loci ranged from two to 11 with an average of 5.3 alleles per locus. The expected heterozygosity ranged from 0.29 to 0.76, with an average of 0.68. Accidental (or illegal) introductions of lake trout into watersheds are decimating native trout populations in the northern Rocky Mountains, and these loci will be useful for identifying the source of these introductions and for estimating the number of founding individuals.  相似文献

The objectives of this study were to examine the relationship between thiamine concentrations in unfertilized eggs and yolksac individuals of lake trout (Salvelinus namaycush), along with any associated histopathological changes in the tissues of alevins at the hatching stage. We address these questions in a lake trout population from different spawning grounds of Lake Michigan (North and South), known for compromised survival due to early mortality syndrome (EMS). However, a dichotomous forage base of lake trout spawning stocks, with a dietary thiaminase-rich alewife in the North, and dietary low-thiaminase round goby in the South, provides the basis for the assumption that different diets may lead to differences in severity of EMS between different stocks. Lake trout eggs of 18 females were collected and fertilized individually with the sperm of several males. The eggs, eyed embryos and newly-hatched alevins were sampled to examine thiamine utilization during embryogenesis. Progenies of females with low (< 0.73 nmol/g) and high (> 0.85 nmol/g) levels of thiamine were chosen for histological studies. The obtained results showed that total thiamine levels in the body and yolk of eyed embryos and alevins at hatching were influenced by thiamine levels of unfertilized eggs and it decreased during embryogenesis (to 51% in eyed embryos and 28% in newly-hatched alevins in comparison to unfertilized eggs). The survival of lake trout until hatching stage does not correlate with the thiamine level, however it was affected by collection site and was significantly higher in fish from the South site (Julian's Reef). At the hatching stage, no pathological changes were observed in the brain, olfactory lobe, retina or liver in embryos regardless of thiamine concentrations in unfertilized eggs. It has been concluded that an enhanced thiamine requirement for the fast muscle mass growth near the swim-up stage is responsible for overt and histopathological signs of EMS. Current study confirms earlier findings that lake trout suffering from EMS can be successfully treated by immersion in thiamine solution as late as at the swim-up stage.  相似文献

To learn more about the degree of individual variation in resource use by lake trout Salvelinus namaycush , ultrasonic telemetry was used to study their habitat use in a lake without pelagic schooling fish prey. Individuals spent most of their time within the metalimnion in favourable water temperatures. They also made frequent excursions, however, into lake temperatures exceeding their optimum for physiological performance at all temporal scales considered. Their frequent use of nearshore habitats suggested that feeding in littoral areas may be common. Habitat use was highly variable among individuals, but spatial habitat use by individuals showed remarkable consistency between years. In particular, some lake trout exhibited high site fidelity to shallow, nearshore areas, whereas others used deep areas extensively. This level of between-individual variation indicated niche partitioning by depth and the possibility of alternative foraging strategies.  相似文献

We describe and explain some of the changes in the fish community as reflected by the catches of the commercial fishery, and evaluate the efforts to restore the trout (Salvelinus spp.) in Georgian Bay. The changes in the fish community were caused in part by excessive fishing for lake trout (S. namaycush), lake whitefish (Coregonus clupeaformis) and deepwater ciscoes (Coregonus spp.), and the introduction of new species such as sea lamprey (Petromyzon marinus), alewife (Alosa pseudoharengus), rainbow smelt (Osmerus mordax) and salmonids (Oncorhynchus spp.). The most striking changes were the near extinction of the lake trout, reductions in lake herring (Coregonus artedii), deepwater ciscoes and lake whitefish, and the increase in abundance of smelt and alewife. In an effort to replace the loss of yield from the cold-water habitat, a fast growing and early maturing hybrid trout (splake [S. namaycush × S. fontinalis]) and its backcross to lake trout were selectively bred for stocking in Georgian Bay. Splake did poorly because of low survival immediately after planting. The survival of backcross after planting was better, but adult mortality was still high. Adult survival of backcross improved considerably after restrictions were placed on the commercial fishery in 1984. The first naturally produced backcross larvae were collected in 1985.  相似文献

Selection of habitat to avoid predation may affect the diet of young-of-year (YOY) lake trout (Salvelinus namaycush). YOY lake trout may use inshore habitat to avoid predation; this habitat may be sub-optimal for growth. To test this, YOY lake trout were penned in nearshore and offshore pelagic areas of two arctic lakes. Toolik Lake had a lake trout population, the other lake, S6, did not. YOY lake trout in Toolik Lake lost weight, but those offshore lost less weight. The YOY lake trout in Lake S6 gained weight and those offshore gained more weight. The primary diet item of the YOY lake trout in both lakes during this experiment was the zooplankter Diaptomis probilofensis; it was also one of the most abundant species. However, its density inshore in Lake S6 was similar to inshore and offshore densities in Toolik Lake. The increased availability of alternative zooplankton prey in Lake S6 may account for the growth differential of YOY lake trout in Lake S6 relative to Toolik Lake. Bioenergetic modeling of YOY lake trout suggests that growth similar to that in the offshore of Lake S6 would be necessary for successful recruitment. If the reduced zooplankton availability in Toolik Lake leads to the reduced growth of YOY in the inshore and offshore pelagic areas, then these fish will be more susceptable to winter predation/starvation. Locations of potential spawning areas for lake trout (Salvelinus namaycush) were predicted in Lake Opeongo, Ontario, Canada using information gained via acoustic telemetry and geographic information system (GIS) technologies. From 1998 to 2000, 18 adult lake trout (mean fork length 553 mm) implanted with acoustic transmitters (battery life 2 years) were manually tracked. For evening fall locations within the erosive zone of the lake (determined using an existing sedimentation model), habitat variables (slope, depth, and effective fetch) were summarised using GIS. Sites selected by lake trout during the spawning window were in areas of mean fetch equal to 1.5 km and mean slope of 10.6% (n = 50 fixes). We used GIS to identify areas that matched the mean habitat criteria and thus locate potential spawning areas. This model correctly identified 19 of 21 known spawning sites, as well as additional sites used by spawning females in an earlier telemetry study. Depths of traditional fall netting sites are shallow compared to areas in which telemetered lake trout were found during evenings of the spawning period (means 3.1 vs. 5.1 m, respectively). Through the use of information on spawning habitat selection gained through telemetry and knowledge of the physical characteristics of the lake, we provide an alternative means of identifying potential spawning habitat for lake trout.  相似文献

Identification of sex chromosomes in lake trout (Salvelinus namaycush)   总被引:2,自引:0,他引:2  
In the male trout there is a difference in the quinacrine banding and C-banding patterns between the two homologs of the second largest chromosome pair. This chromosome is the only large submetacentric in the karyotype, making it easy to identify and suggesting that the sex chromosomes have become differentiated since the time of tetraploidization. In males one homolog has a medium-to-large quinacrine bright heterochromatic band on the end of the short arm, while the other lacks it completely. In females both homologs have medium-to-large quinacrine bright heterochromatic bands. Approximately half the progeny from every lake trout cross studied and half the eggs from every lake trout population examined were heteromorphic for a difference in this chromosome band. Results from sexed fish, reciprocal F1 hybrids between brook trout and lake trout, and gynogenetic haploids are all consistent with the interpretation that chromosome 2 is the sex chromosome. These results suggest that the addition of heterochromatin to the X can be the first step in the inhibition of crossing over between the X and Y chromosomes required for sex chromosome differentiation.  相似文献

Synopsis Foraging behaviour of lake charr, Salvelinus namaycush, exposed to 0.5 and 5.0 g l–1 cadmium concentrations for 106–112 d, was examined in a flow-through stream channel using fathead minnows, Pimephales promelas, as prey. Five modes of foraging behaviour were observed and quantified (ending at fixate-orient; ending at approach and/or chase; ending at failed attack; ending at capture and loss; successful consumption). Each mode represented an increasingly progressive end-point from initial sighting of the prey to final consumption. Compared to controls., 0.5 g l–1 Cd-exposed predators captured significantly fewer prey, while 5 g l–1 Cd-exposed predators made fewer attacks. Both Cd-exposed groups consumed fewer prey than controls.  相似文献

Methods for assigning individuals to population of origin are widely used in ecological genetics, resources management, and forensics. Characteristics of genetic data obtained from putative source populations that enhance accuracy of assignment are well established. How non-independence within and among unknown individuals to be classified [i.e., gene correlations within individual (inbreeding) and gene correlations among individuals within group (coancestry)] affect assignment accuracy is poorly understood. We used empirical data for six microsatellite loci and offspring from full-sib crosses of hatchery strains of lake trout (Salvelinus namaycush; Salmonidae) representing known levels of coancestry (mean θ = 0.006 and 0.06) within families to investigate how gene correlations can affect assignment. Additional simulations were conducted to further investigating the influence of allelic diversity (2, 6 or 10 alleles per locus) and inbreeding (F = 0.00, 0.05, and 0.15) on assignment accuracy for cases of low and high inter-population variance in allele frequency (mean F st = 0.01 and 0.1, respectively). Inbreeding had no effect on accuracy of assignments. In contrast, variance in assignment accuracy across replicated simulations, and for each empirical case study increased with increasing coancestry, reflecting non-independence of probabilities of correct assignment among members of kin groups. Empirical estimates of assignment error rates should be interpreted with caution if appreciable levels of coancestry are suspected. Additional emphasis should be placed on sampling designs (spatially and temporally) that define or minimize the potential for sampling related individuals.  相似文献

Swimbladder walls of lake charr, Salvelinus namaycush, from Great Slave Lake (GSL), Northwest Territories, Canada, were unusually thick for the species. The thinnest sections of the GSL bladders (mean = 2.44mm, range = 1.1–4.4mm) were significantly thicker (P = 0.001) than lake charr swimbladders collected from two small Province of Ontario lakes (means = 0.65 and 0.92mm), whose populations were assumed to be representative of the species. Variance in wall thickness was also greater in GSL lake charr than in charr from two small lakes (P < 0.02). Within individuals, some of the GSL bladder walls were markedly irregular in thickness, but whether these anomalies exist in situ or were artifacts of preservation remains uncertain. The bulk of the tissue in the thickest sections of the GSL swimbladders was in the tunica serosa (outer layer). The extent of the modification of the GSL swimbladders is extraordinary for northern fishes in postglacial lakes.  相似文献

The influence of egg predators and physical disturbance on lake trout Salvelinus namaycush egg mortality was investigated in situ in Lake Michigan where recruitment is below detectable levels and egg predator abundance is high. Comparisons were made with Lake Champlain where recruitment is low and egg predator abundance is also low and with Parry Sound (Lake Huron) where recruitment is moderate and egg predators are in low abundance. A multi-density egg seeding method (100 to 5000 eggs m−2) was used to quantify the effect of physical disturbance and egg predation on egg loss. Wind fetch was used as an index of physical disturbance and comparisons across all locations and egg densities suggested that at sites with high wind fetch (>5 km), physical disturbance may be a greater source of egg loss than predation. When analyses were limited to those sites having a wind fetch of <5 km, the percentage of eggs recovered was found to be linearly related to predator density. The strength of this relationship was based largely on egg recovery at 500 and 1000 eggs m−2 because recovery at lower (100, 250 eggs m−2) and very high (5000 eggs m−2) densities was not significantly related to predator density. The reason for this is probably that at low egg densities, crayfish Orconectes spp., the major egg predator at most sites, had difficulty finding and consuming eggs and at high egg densities they became satiated. Egg loss was directly related to wind fetch for Lake Michigan and on average six-fold greater than for Parry Sound suggesting that without corresponding changes in fecundity and age structure, lake trout populations in large lakes like Lake Michigan are inherently less productive than those from enclosed inland waters.  相似文献

Anadromy was documented in 16 lake trout, Salvelinus namaycush, from Canada's central Arctic using capture data and otolith microchemistry. For the first time, estuarine/marine habitat use was described for five individuals using acoustic telemetry. Age-at-first-migration to sea was variable (10–39 years) among individuals and most S. namaycush undertook multiple anadromous migrations within their lifetime. Telemetry data suggested that S. namaycush do not travel far into marine habitats and prefer surface waters (<2 m). These results further our collective understanding of the marine ecology of Arctic S. namaycush.  相似文献

  • 1.1. The effects of storage temperature (2, −20 and −80°C) and duration (3, 9 and 27 days) on plasma metabolites concentrations of lake trout fed three dietary protein levels (20, 40 and 60%) and a single lipid level (20%) for 28 days were investigated.
  • 2.2. Significantly high plasma urea and glucose concentrations were associated with low (20%) and high (60%) dietary protein intake in fish; while, high plasma creatinine concentration seems to characterize insufficient dietary protein and energy consumption.
  • 3.3. Deproteinization of plasma with 5.0% sulfosalicylic acid did not confer better storage stability for any of the plasma metabolites except ammonia which was significantly high in non-deproteinized samples when stored at 2°C over 3, 9 and 27 day periods.
  • 4.4. These studies suggest that non-deproteinized fish plasma can be stored at − 20°C without affecting the baseline concentrations of ammonia, creatinine and glucose for up to 9 days and, urea and total protein for up to 27 days.

The chromosomes of lake trout (Salvelinus namaycush) contain a considerable amount of heterochromatin located at the centromeres and/or telomeres of several chromosomes, including a sex-specific block located distally on the X chromosome. In order to investigate further the repetitive DNAs of lake trout, genomic DNA from a female was size fractionated (<600 bp) with the restriction endonuclease AluI and fragments were cloned into the bacteriophage M13. A total of 42 clones were isolated. Relative copy number of individual inserts within the lake trout genome was estimated by Southern analysis. Twelve clones were determined to be highly repetitive and were chosen for further investigation. Inserts of these clones contained sequences similar to the AluI/RsaI, EcoRI/DraI, DraI/BstEII, and MboI/BglII families reported from Arctic char (Salvelinus alpinus). The chromosomal location of several of these fragments was determined in lake trout by fluorescence in situ hybridization (FISH). Two related AluI/RsaI sequences (Type A, 140 bp, and Type B, 120 bp) showed differential hybridization. Type A hybridized to the centromeres of all metacentric as well as several acrocentric chromosomes. Type B hybridized to the centromeres of most acrocentric chromosomes. A sequence with homology to the EcoRI/DraI family hybridized to the centromeres of several acrocentric chromosomes. Sequences with partial similarity to the DraI/BstEII family hybridized to the major rDNA sites (nucleolar organizer regions, NORs) and several minor telomeric sites. The interstitial and telomeric heterochromatin of lake trout, including that of the X chromosome, appears to comprise sequences belonging to the MboI/BglII family.  相似文献

An 18-day experiment was conducted to investigate the uptake and sublethal toxicity of dietary Ni in adult lake whitefish (LWF, Coregonus clupeaformis) and lake trout (LT, Salvelinus namaycush) fed diets containing 0, 1000 and 10000 microg Ni/g, prepared with and without brine shrimp. The results of this experiment were used to design an experiment of longer duration in which one of the fish species was selected and exposed to lower dietary Ni doses. In the present study feed refusal was observed in LT and LWF fed 10000 microg Ni/g, after three and 4-5 feedings, respectively. LT fed Ni-contaminated diets exhibited different patterns of Ni accumulation than LWF. Increased Ni concentrations in all LWF tissues, except the intestine, were associated with increased doses of Ni. Copper and Zn concentrations in kidney and liver of LWF were altered. Metallothionein concentrations in kidneys of LT fed 1000 microg Ni/g and 10000 microg Ni/g and LWF fed 10000 microg Ni/g and in livers of LWF fed 10000 microg Ni/g (diet without shrimp only) increased significantly. Increased lipid peroxide production in the plasma of LT fed 10000 microg Ni/g was observed. Blood glucose and electrolytes were affected by Ni exposure. Histopathological alterations were observed in kidneys of LWF fed low and high dose diets, livers of whitefish fed high dose diets, and intestines of LWF fed high dose diets and LT fed low and high dose diets. LT fed high dose diets exhibited significant decreases in weight.  相似文献

We coupled dynamic optimization and bioenergetics models to assess the assumption that lake trout (Salvelinus namaycush) depth distribution is structured by temperature, food availability, and predation risk to maximize reproductive mass by autumn spawning. Because the model uses empirical daily thermal-depth profiles recorded in a small boreal shield lake (lake 373 at the Experimental Lakes Area, northwestern Ontario) during 2 years of contrasting thermal stratification patterns, we also assessed how climate-mediated changes in lakes may affect the vertical distribution, growth, and fitness of lake trout, a cold-water top predator. The depths of acoustic-tagged lake trout were recorded concurrently with thermal-depth profiles and were compared to model output, enabling an assessment of model performance in relation to the observed fish behavior and contrasting thermal conditions. The depths and temperatures occupied by simulated fish most closely resembled those of the tagged fish when risk of predation was included in the model, indicating the model may incorporate the most important underlying mechanisms that determine lake trout depth. Annual differences suggest less use of shallow (warm), productive habitats, resulting in markedly less reproductive mass, during the year with the warm stratification pattern. Mass for reproduction may be lower in warmer conditions because of reduced reproductive investment, yet survival may be inadvertently higher because risky surface waters may be avoided more often in warmer, shallower, and metabolically costly conditions. At a minimum our study suggests that lake trout reproductive mass and fitness may be expected to change under the anticipated longer and warmer stratification patterns.  相似文献

Humans have played a significant role in reducing levels of genetic diversity and differentiation of many teleost fishes, leading to homogenization across biological entities. We compare patterns of historical and contemporary genetic structure for three sympatric Great Lake??s lake trout (Salvelinus namaycush) morphs (lean, siscowet, and humper) that differ in patterns of habitat occupancy, susceptibility to overfishing and predation by invasive sea lamprey (Petromyzon marinus). Differential susceptibilities to overfishing and predation were expected to result in different impacts to levels of genetic diversity loss for each morphotype. Genetic data was collected for samples at three points in time: 1948 (pre-collapse), 1959 (collapse) and 1990s (current), corresponding to periods of intensive fishing, mortality due to lamprey and recovery, respectively. The lean morph preferentially targeted by the fishery and recognized as highly preyed upon by sea lamprey was more highly impacted genetically than other morphs, as evidenced by greater loss of genetic diversity first during the period of overfishing, then during the period of high sea lamprey abundance once the fishery collapsed. The siscowet morph also experienced genetic bottlenecks during the period of overfishing (pre-collapse period). Results indicate significant levels of genetic differentiation among morphs historically prior to declines in abundance and also among contemporary populations, suggesting that periods of population decline and resurgence in abundance and distribution did not result in loss of genetic distinctiveness among morphs.  相似文献
