Rice flour is becoming very attractive as raw material, but there is lack of information about the influence of particle size on its functional properties and starch digestibility. This study evaluates the degree of dependence of the rice flour functional properties, mainly derived from starch behavior, with the particle size distribution. Hydration properties of flours and gels and starch enzymatic hydrolysis of individual fractions were assessed. Particle size heterogeneity on rice flour significantly affected functional properties and starch features, at room temperature and also after gelatinization; and the extent of that effect was grain type dependent. Particle size heterogeneity on rice flour induces different pattern in starch enzymatic hydrolysis, with the long grain having slower hydrolysis as indicated the rate constant (k). No correlation between starch digestibility and hydration properties or the protein content was observed. It seems that in intact granules interactions with other grain components must be taken into account. Overall, particle size fractionation of rice flour might be advisable for selecting specific physico-chemical properties.  相似文献

Waxy maize starch in an aqueous sulfuric acid solution (3.16 M, 14.7% solids) was hydrolyzed for 2–6 days, either isothermally at 40 °C or 4 °C, or at cycled temperatures of 4 and 40 °C (1 day each). The starch hydrolyzates were recovered as precipitates after centrifuging the dispersion (10,000 rpm, 10 min). The yield of starch hydrolyzates depended on the hydrolysis temperature and time, which varied from 6.8% to 78%. The starch hydrolyzed at 40 °C or 4/40 °C exhibited increased crystallinity determined by X-ray diffraction analysis, but melted in broader temperature range (from 60 °C to 110 °C). However, the starch hydrolyzed at 4 °C displayed the crystallinity and melting endotherm similar to those of native starch. The starch hydrolyzates recovered by centrifugation were re-dispersed in water (15% solids), and the dispersion was treated by an ultrasonic treatment (60% amplitude, 3 min). The ultrasonication effectively fragmented the starch hydrolyzates to nanoparticles. The hydrolyzates obtained after 6 days of hydrolysis were more resistant to the ultrasonication than those after 2 or 4 days, regardless of hydrolysis temperatures. The starch nanoparticles could be prepared with high yield (78%) and crystallinity by 4 °C hydrolysis for 6 days followed by ultrasonication. Scanning electron microscopy revealed that the starch nanoparticles had globular shapes with diameters ranging from 50 to 90 nm.  相似文献

The insoluble residues obtained by submitting amylopectin-rich native starch granules from waxy maize to a mild acid hydrolysis consist of polydisperse platelet nanocrystals that have retained the allomorphic type of the parent granules. The present investigation is a detailed characterization of their molecular composition. Two major groups of dextrins were found in the nanocrystals and were isolated. Each group was then structurally characterized using β-amylase and debranching enzymes (isoamylase and pullulanase) in combination with anion-exchange chromatography. The chain lengths of the dextrins in both groups corresponded with the thickness of the crystalline lamellae in the starch granules. Only 62 mol % of the group of smaller dextrins with an average degree of polymerization () 12.2 was linear, whereas the rest consisted of branched dextrins. The group of larger dextrins ( 31.7) apparently only consisted of branched dextrins, several of which were multiply branched molecules. It was shown that many of the branch linkages were resistant to the action of the debranching enzymes. The distribution of branched molecules in the two populations of dextrins suggested that the nanocrystals possessed a regular and principally homogeneous molecular structure.  相似文献

目的研究双歧啤酒的延缓衰老作用。方法将老龄小鼠和黑腹果蝇随机分成纯净水对照组、市售啤酒对照组和双歧啤酒低、中、高三个剂量组,进行果蝇生存实验,检测小鼠血液中超氧化物歧化酶(SOD)、谷光甘肽过氧化物酶(GSH-PX)的活性和丙二醛(MDA)的含量。结果与对照组比较,双歧啤酒高、中剂量组能显著延长果蝇的半数死亡时间、平均寿命和平均最高寿命,提高老龄小鼠血液中SOD、GSH-PX的活性,降低老龄小鼠血液中MDA的含量。结论双歧啤酒具有延缓衰老的作用。  相似文献

The degradation of pea starch granules by acid hydrolysis has been investigated using a range of chemical and structural methods, namely through measuring changes in amylose content by both the iodine binding and concanavalin A precipitation methods, along with small angle X-ray scattering (SAXS), wide angle X-ray diffraction (XRD) and field emission scanning electron microscopy (FE-SEM). The relative crystallinity, intensity of the lamellar peak and the low-q scattering increased during the initial stages of acid hydrolysis, indicating early degradation of the amorphous regions (growth rings and lamellae). In the first 2 days of hydrolysis, there was a rapid decline in amylose content, a concomitant loss of precipitability of amylopectin by concanavalin A, and damage to the surface and internal granular structures was evident. These observations are consistent with both amylose and amylopectin being located on the surface of the granules and attacked simultaneously in the early stages of acid hydrolysis. The results are also consistent with amylose being more concentrated at the core of the granules. More extensive hydrolysis resulted in the simultaneous disruption of amorphous and crystalline regions, which was indicated by a decrease in lamellar peak intensity, decrease in interhelix peak intensity and no further increase in crystallinity. A growth trial with 420 broiler chickens (35 per experimental unit) was conducted in order to evaluate whether bird performance (Days 0–38) is affected by site of starch digestion. Two diets were formulated with the same calculated apparent metabolisable energy, digestible lysine, and digestible starch content. The diets contained starch sources with known amounts of rapidly digestible starch (RDS, starch digested until the posterior jejunum) and slowly digestible starch (SDS, starch digested in the ileum). Diets were either high (H) or low (L) in SDS content. RDS and SDS contents were 298 and 52 g/kg for the H diet and 345 and 7 g/kg for the L diet, respectively. Starch in the H diet was supplied by waxy maize, peas and sorghum and starch in the L diet was supplied by tapioca and common maize. Birds receiving the H diet consumed more feed (P<0.10), grew faster (P<0.01) and had a lower feed conversion ratio (P<0.01) than birds receiving the L diet. The results suggest that broiler chickens perform better on diets containing a minimal amount of slowly digestible starch. Future experiments are necessary to investigate the relation between amount of slowly digestible starch and performance of broiler chickens.  相似文献

In hydrolysis-limited anerobic systems, the key parameters describing degradation are degradability extent (f(d)), and the lumped apparent first order coefficient (k(hyd)). These are often measured in biological methane potential (BMP) tests. Using modern techniques, it should also be possible to estimate these parameters in full-scale systems, especially where inputs are dynamic. In this study, we evaluated f(d) and k(hyd) values and uncertainty based on nonlinear parameter estimation from (i) BMP tests and (ii) effluent gas and solids from two full-scale digesters fed with highly variable feed flows and concentrations (up to 6 kg COD m(-3) day(-1)). The substrate was thermally hydrolyzed activated sludge, and the inoculum for BMP tests was from the full-scale digesters. While identifiability of both parameters in the BMP tests was generally good, only f(d) could be well identified using continuous data. For k(hyd) using continuous data, normally only a lower limit could be found (upper was unbounded). In addition, parameters as estimated on different outputs (VS and gasflow) and two different digesters were consistent, with an f(d) value of 0.45-0.55, and a k(hyd) value of >5 day(-1). Gradual changes in f(d) over the 450 days could be related to upstream changes. f(d) values as estimated in BMP tests were consistent (if conservative) with continuous estimates, with a f(d) in BMP of 0.4-0.5. k(hyd) values were an order of magnitude lower (0.15-0.25 day(-1) vs. >5 day(-1)), and this translated to very poor model performance when BMP-estimated values were used in the continuous model. This means that while BMP testing may be used for project feasibility analysis, values obtained should not be used for dynamic modeling. The parameter confidence regions found were highly nonlinear, especially for continuous systems, indicating that iterative or sampling techniques are required for an estimate of real parameter uncertainty.  相似文献

Correlating the effect of pretreatment on the enzymatic hydrolysis of straw   总被引:4,自引:0,他引:4  
Avicell, Alkali-treated straw cellulose (ATSC), and wheat straw were ball-milled to reduce crystallinity; wheat straw was delignified by hot (120 degrees C) sodium hydroxide solutions of various concentrations. The physically and chemically pretreated cellulosic materials were hydrolyzed by the cellulases of Fusarium oxysporum strain F3. Enzymic hydrolysis data were fitted by the hyperbolic correlation of Holtzapple, which involves two kinetic parameters, the maximum conversion (X(max)), and the enzymic hydrolysis time corresponding to 50% of X(max) (t(1/2)). An empirical correlation between X(max) and cellulose crystallinity, lignin content, and degree of delignification has been found under our experimental conditions. Complete cellulose hydrolysis is shown to be possible at less than 60% crystallinity indices or less than 10% lignin content.  相似文献

为提高胆红素生产效益,采取正交实验法,优化皂化水解pH、温度和时间。结果表明:在pH12.0、105℃、20 min时,胆红素粗制品收率平均为0.866 7 g/L,但纯度仅为24.01%,折合纯品收率为0.208 1 g/L。胆红素的纯度在pH11.0、100℃、10~30 min时,平均纯度为67.91%,但粗制品收率仅为0.402 2 g/L,折合纯品收率为0.273 1g/L。胆红素的折合纯品收率,在pH10~12、100℃、20 min时,平均可达0.366 8 g/L,粗制品收率为0.568 9 g/L、纯度64.12%,此时胆红素的生产效益比传统方法提高了43.9%。  相似文献

The purification and characterization of an extracellular lichenase from the fungus Penicillium occitanis Pol6 were studied. The strain produced the maximum level of extracellular lichenase (45 ± 5 U ml−1) when grown in a medium containing oat flour (2%, w/v) at 30 °C for 7 days. The purified enzyme EGL showed as a single protein band on SDS–PAGE with a molecular mass of 20 kDa. Its N-terminal sequence of 10 amino acid residues was determined as LDNGAPLLNV. The purified enzyme showed an optimum activity at pH 3.0 and 50–60 °C. The half-lives of EGL at 60 °C and 70 °C were 80 min and 21 min, respectively. Substrate specificity studies revealed that the enzyme is a true β-1,3-1,4-d-glucanase. The enzyme hydrolyzed lichenan to yield trisaccharide, and tetrasaccharide as the main products. Under simulated mashing conditions, addition of EGL (20 U/ml) or a commercial β-glucanase (20 U/ml) reduced the filtration time (25% and 21.3%, respectively) and viscosity (10% and 8.18%, respectively). These characteristics indicate that EGL is a good candidate in the malting and brewing industry.  相似文献

The hydrolysis of amylopectin potato starch with Bacillus licheniformis alpha-amylase (Maxamyl) was studied under industrially relevant conditions (i.e. high dry-weight concentrations). The following ranges of process conditions were chosen and investigated by means of an experimental design: pH [5.6-7.6]; calcium addition [0-120 microg/g]; temperature [63-97 degrees C]; dry-weight concentration [3-37% [w/w]]; enzyme dosage [27.6-372.4 microL/kg] and stirring [0-200 rpm]. The rate of hydrolysis was followed as a function of the theoretical dextrose equivalent. The highest rate (at a dextrose equivalent of 10) was observed at high temperature (90 degrees C) and low pH (6). At a higher pH (7.2), the maximum temperature of hydrolysis shifted to a lower value. Also, high levels of calcium resulted in a decrease of the maximum temperature of hydrolysis. The pH, temperature, and the amount of enzyme added showed interactive effects on the observed rate of hydrolysis. No product or substrate inhibition was observed. Stirring did not effect the rate of hydrolysis. The oligosaccharide composition after hydrolysis (at a certain dextrose equivalent) did depend on the reaction temperature. The level of maltopentaose [15-24% [w/w]], a major product of starch hydrolysis by B. licheniformis alpha-amylase, was influenced mostly by temperature.  相似文献

The retrogradation of untreated wild-type starches (potato, maize, and wheat), waxy maize starches, and one pregelatinized, modified amylose-rich starch was investigated continuously using Raman spectroscopy. The method detects conformational changes due to the multi-stage retrogradation, the rate of which differs between the starches. The pregelatinized, modified amylose-rich starch shows all stages of retrogradation in the course of its Raman spectra. In comparison to amylose, the retrogradation of amylopectin is faster at the beginning of the measurements and slower in the later stages. The untreated starches can be ranked in the order of their rate of retrogradation as follows: potato>maize>wheat.  相似文献

Individual cells of the yeast Saccharomyces cerevisiae exhibit a finite replicative lifespan, which is widely believed to be a function of the number of divisions undertaken. As a consequence of ageing, yeast cells undergo constant modifications in terms of physiology, morphology and gene expression. Such characteristics play an important role in the performance of yeast during alcoholic beverage production, influencing sugar uptake, alcohol and flavour production and also the flocculation properties of the yeast strain. However, although yeast fermentation performance is strongly influenced by the condition of the yeast culture employed, until recently cell age has not been considered to be important to the process. In order to ascertain the effect of replicative cell age on fermentation performance, age synchronised populations of a lager strain were prepared using sedimentation through sucrose gradients. Each age fraction was analysed for the ability to utilise fermentable sugars and the capacity to flocculate. In addition cell wall properties associated with flocculation were determined for cells within each age fraction. Aged cells were observed to ferment more efficiently and at a higher rate than mixed aged or virgin cell cultures. Additionally, the flocculation potential and cell surface hydrophobicity of cells was observed to increase in conjunction with cell age. The mechanism of ageing and senescence in brewing yeast is a complex process, however here we demonstrate the impact of yeast cell ageing on fermentation performance.  相似文献

The effect of delignification methods on enzymatic hydrolysis of forest biomass was investigated using softwood and hardwood that were pretreated at an alkaline condition followed by sodium chlorite or ozone delignification. Both delignifications improved enzymatic hydrolysis especially for softwood, while pretreatment alone was found effective for hardwood. High enzymatic conversion was achieved by sodium chlorite delignification when the lignin content was reduced to 15%, which is corresponding to 0.30-0.35 g/g accessible pore volume, and further delignification showed a marginal effect. Sample crystallinity index increased with lignin removal, but it did not show a correlation with the overall carbohydrate conversion of enzymatic hydrolysis.  相似文献

The effects of waxy mutations on starch-granule-bound starch synthases (EC 2.4.1.18) in the developing endosperm of barley (Hordeum vulgare L.) and maize (Zea mays L.) have been investigated. Three granule-bound starch synthases in barley endosperm were identified by use of antibodies to known starch synthases, by reconstitution and assay of individual proteins from sodium dodecyl sulphate-polyacrylamide gels of granule-bound proteins, and by partial purification of proteins released by enzymic digestion of starch. These are proteins of 60, 77 and 90 kDa. Use of antibodies to known starch synthases and partial purification of proteins released by enzymic digestion of starch indicated that there may be at least four granule-bound starch synthases in maize endosperm: proteins of 59, 74, 77 and 83 kDa. Mutations at the waxy loci of both species affected only the 60- (barley) and 59-(maize) kDa isoforms. No evidence was found that other putative isoforms are altered in abundance or activity by the mutations. Wheat and barley contain at least four classes of starch synthases in the endosperm, granule bound starch synthase I (GBSSI) and starch synthases I, II and III (SSI, SSII, SSIII). In this work, SSII in barley is shown to be associated with the starch granule by using antibodies. A cDNA from barley encoding SSII and the genes for SSII from barley and Aegilops tauschii (A. tauschii, the D genome donor to wheat) are characterised. Fluorescent in situ hybridisation (FISH) and PCR were used to localise the wheat SSII gene to the short arm of chromosome 7, showing synteny with the location of the rice SSII gene to the short arm of chromosome 6. Comparison of the genes encoding SSII of A. tauschii, barley and Arabidopsis showed a conserved exon-intron structure although the size of the introns varied considerably. Extending such comparison between the genes encoding starch synthases (GBSSI, SSI, SSII and SSIII) from A. tauschii and Arabidopsis showed that the exon-intron structures are essentially conserved. Separate and distinct genes for the individual starch synthases therefore existed before the separation of monocotyledons and dicotyledons. Electronic Publication  相似文献

Seven types of coffee were prepared by four methods. Three of them – simple coffee infusion, preparation in ibrick and moka pot – are very often used to prepare coffee at home. The fourth one – a single-cup filter is typical for Vietnam. Cookware used for each method was made from glass, aluminium, stainless steel and two types of alloys. Amounts of Al leaching to coffee infusions were determined. On average, the highest amount of Al was in coffee infusions brewed in aluminium single-cup filter, and the lowest in infusions prepared by simple extraction. Other brewing methods in combination with different tool materials resulted in similar Al content. The type of brewing method significantly influences the Al content in final infusion. Aluminium content varies in infusions in relation to the method of choice, especially when using single-cup filter. Despite the fact that coffee is considered to be a poor source of Al for humans, in some cases, Al content in infusions can even reach the values reported for tea infusions.  相似文献

Developmental changes in the starch and sucrose content of grains andthe activities of enzymes of starch synthesis in wheat were studied under waterstress conditions. Water stress caused a marked reduction in the sucrose andstarch content of the grains. Sucrose synthase (SS) and UDP-glucosepyrophosphorylase (UDP-Gppase), showed higher catalytic activity and moreresistance to water stress compared with amyloplastic enzymes. ADP-glucosepyrophosphorylase (ADP-Gppase) activity was reduced to a low level under bothin situ and osmotic stress conditions in which grainsfailed to accumulate dry matter in vivo. Granule-boundstarch synthase (GBSS) also responded rapidly to in situwater stress treatments as did ADP-Gppase. Reduction in GBSS activity at thetime of growth cessation in situ was less than that ofADP-Gppase and the enzyme did not respond to severe osmotic stress. Solublestarch synthase (SSS) was the enzyme most sensitive to water stress in that itresponded earlier, and to a greater extent, than the other enzymes. However,under severe dehydration conditions, leading to cessation of growth, thedeclinein SSS activity was less than that for ADP-Gppase. SSS showed the lowestin vitro activity followed by GBSS. These results suggestthat SSS is the site of response to water stress by which the rate of graingrowth can be affected, whereas growth cessation is due mainly to theinactivation of ADP-Gppase.  相似文献
