Most of the research concerning magnetic antinociception was focused on brief exposure less than 1 h. The main purpose of the present study was to determine the effect of extremely low frequency (ELF) magnetic field (MF) repeated exposures on rats in inducing antinociception and to find the effective analgesic "time window." Meanwhile this investigation was to examine the role of central beta-endorphin, substance P, and 5-HT in magnetic analgesia. We found tail flick latencies (TFLs) increased significantly after the rats were exposed to 55.6 Hz, 8.1 mT magnetic field for 4 days, 6 h each day. The analgesic effects seemed to decrease gradually when the rats were exposed daily for another 10 days. Their levels of TFLs decreased within 1 day when the rats were removed after a 4-day exposure. The concentrations of hypothalamus beta-endorphin, substance P, and brainstem serotonin (5-HT) were increased significantly on Day 4. However, no differences were found when rats were exposed for another 10 days, and there were no significant increases when rats were removed after the fourth day of exposure and tested for nociception on Days 5 and 7 with no changes in the biochemical markers at 7 days. These results suggest that the ELF magnetic field has analgesic effect, but only on Days 3 and 4. The effect may be associated with increases in endogenous beta-endorphin, substance P, and 5-HT stimulated by the 55.6 Hz, 8.1 mT magnetic field.  相似文献

The previous observation with an electron microscope showed that extremely low frequency (ELF) pulsed magnetic field (PMF) (with the maximum intensity of 0. 6-2. 0 T, gradient of 10-100 T. M-1, pulse width of 20-200 ms and frequency of 0. 16-1. 34 Hz) inhibited the growth of S-180 sarcoma in mice and enhanced the ability of immune cell's dissolving sarcoma cells. In this study, the DNA contents of nuclei were assayed by using Faulgen Staining method. With an electron microscope and cell stereoscopy technology it was observed that magnetic field affected the sarcoma cell's metabolism, lowered its malignancy, and restrained its rapid and heteromorphic growth. The magnetic field enhanced the cellular immune ability and the reaction of lymphocytes and plasma. Since ELF pulsed magnetic fields can inhibit the growth of sarcomas and enhance the cellular immune ability, it is possible to use it as a new method to treat cancer.  相似文献

The effects of low intensity, low frequency magnetic fields (MFs) on catalytic activity of the calcium dependent protease calpain was determined following the enzyme activation both in "in vitro" and "in vivo" conditions. We have observed that a 0.3 mT MF induces a significant increase in the requirement of the protease for this metal ion. This change is detectable at low [Ca(2+)] and disappears when the level of Ca(2+) is raised to saturating amounts. The observed effects are not due to transient MF(-) induced conformational changes occurring in calpain, but to direct effects of the MF on Ca(2+) ions, which become less available for the binding sites present in calpain. Altogether, these results indicate that exposure to low intensity, low frequency MFs alters the intracellular Ca(2+) "availability," thereby modifying the related cell response.  相似文献

Exposure to extremely low frequency (ELF) magnetic fields has been shown to attenuate endogenous opioid peptide mediated antinociception or “analgaesia” in the terrestrial pulmonate snail, Cepaea nemoralis. Here we examine the roles of light in determining this effect and address the mechanisms associated with mediating the effects of the ELF magnetic fields in both the presence and absence of light. Specifically, we consider whether the magnetic field effects involve an indirect induced electric current mechanism or a direct effect such as a parametric resonance mechanism (PRM). We exposed snails in both the presence and absence of light at three different frequencies (30, 60, and 120 Hz) with static field values (BDC) and ELF magnetic field amplitude (peak) and direction (BAC) set according to the predictions of the PRM for Ca2+. Analgaesia was induced in snails by injecting them with an enkephalinase inhibitor, which augments endogenous opioid (enkephalin) activity. We found that the magnetic field exposure reduced this opioid-induced analgaesia significantly more if the exposure occurred in the presence rather than the absence of light. However, the percentage reduction in analgaesia in both the presence and absence of light was not dependent on the ELF frequency. This finding suggests that in both the presence and the absence of light the effect of the ELF magnetic field was mediated by a direct magnetic field detection mechanism such as the PRM rather than an induced current mechanism. The aim of this study was to evaluate the influence of extremely low frequency magnetic field (ELF MF) on the reactivity of the central dopamine D(1) receptor in rats with dopamine neurons chemically damaged by 6-hydroxydopamine (6-OHDA), an animal model of human's Parkinson's disease. The experiment was carried out on male Wistar rats. On day 3 of postnatal life, a lasting and selective chemical damage of the central dopamine system was induced in the rats by infusion of 6-OHDA HBr (133.4 microg intracerebroventricular, base form) given bilaterally into lateral ventricles of the brain. Control animals received similar treatments injecting only vehicle. At 2 months of age, both 6-OHDA treated and control rats were randomly divided into two groups. Rats from the first group were exposed to 10 Hz sinusoidal, 1.8-3.8 mT magnetic field one hour daily for 14 days. Rats of the second group were sham exposed, with the applicator solenoid turned off. On the day after the final exposure the evaluations were made of the rat's spontaneous irritability, oral activity, and catalepsy. The MF exposed rat with chemically induced dopamine neurons damage exhibited a reduction of irritability and oral activity when stimulated with SKF 38393 (the agonist of central dopamine D(1) receptor) and some increase of catalepsy after administration of SCH 23390(the antagonist of central dopamine D(1) receptor). These results indicate that ELF MF reduce the reactivity of central dopamine D(1) receptors in rats.  相似文献

The effects of intraperitoneal administration of gamma-hydroxybutyric acid (GHB) on biogenic amine levels in hemispheres, hypothalamus, midbrain, and medulla-pons, and on tryptophan in serum and brain, were studied. One hour after GHB administration (50 and 100 mg/kg) significant increases of dopamine concentration were observed in the hemispheres with both doses and in the hypothalamus with the higher dose, but a significant decrease of noradrenaline in the hypothalamus. No significant changes of serotonin metabolism were observed. These results indicate that low doses of GHB selectively affect the catecholaminergic neuronal activity.  相似文献

In this study, the genotoxic and cytotoxic potential of extremely low frequency magnetic fields (ELF-MF) was investigated in Wistar rat tibial bone marrow cells, using the chromosomal aberration (CA) and micronucleus (MN) test systems. In addition to these test systems, we also investigated the mitotic index (MI), and the ratio of polychromatic erythrocytes (PCEs) to normochromatic erythrocytes (NCEs). Wistar rats were exposed to acute (1 day for 4 h) and long-term (4 h/day for 45 days) to a horizontal 50 Hz, 1 mT uniform magnetic field generated by a Helmholtz coil system. Mitomycin C (MMC, 2 mg/kg BW) was used as positive control. Results obtained by chromosome analysis do not show any statistically significant differences between the negative control and both acute and long-term ELF-MF exposed samples. When comparing the group mean CA of long-term exposure with the negative control and acute exposure, the group mean of the long-term exposed group was higher, but this was not statistically significant. However, the mean micronucleus frequency of the longer-term exposed group was considerably higher than the negative control and acutely exposed groups. This difference was statistically significant (p < 0.01). The results of the MI in bone marrow showed that the averages of both A-MF and L-MF groups significantly decreased when compared to those in the negative control (p < 0.001 and p < 0.01, respectively). No significant differences were found between the group mean MI of A-MF exposure with L-MF. We found that the average of PCEs/NCEs ratios of A-MF exposed group was significantly lower than the negative control and L-MF exposed groups (p < 0.001 and p < 0.01, respectively). In addition, the group mean of the PCEs/NCEs ratios of L-MF was significantly lower than negative control (p < 0.01). We also found that the MMC treated group showed higher the number of CA and the frequency of MN formation when compared to those in all other each groups (p-values of all each groups <0.01) and also MMC treated group showed lower MI and the PCEs/NCEs ratios when compared to those in all other each groups (p-values of all groups <0.01). These observations indicate the in vivo suspectibility of mammals to the genotoxicity potential of ELF-MF.  相似文献

To explore whether the age at cancer diagnosis was associated with residential exposure to magnetic field, we compared average ages at diagnosis for cases of leukemia, brain tumor, or female breast cancer with elevated exposure (magnetic flux density >or= 0.2 microT, or residential distance from major power lines 100 m from major power lines). Comparing with brain tumor cases with background magnetic field exposure (n = 506), brain tumor cases with elevated exposure (n = 71) were 6 years older on average at diagnosis (P = 0.01). The difference was greater for males (45.2 vs. 52.1 years, P = 0.01) than for females (44.3 vs. 48.2 years, P = 0.27). No such phenomena at a significant level was observed for leukemia, female breast cancer, or a random sample of general population. We noted an association between magnetic field exposure and a greater mean age at diagnosis for brain tumors. Whether or not these phenomena suggest a delayed occurrence of brain tumors following a higher than background residential magnetic field exposure deserves further investigation.  相似文献

目的:探讨极低频磁场对大鼠急性和慢性关节炎模型的影响。方法:大鼠足内皮下注射角叉菜胶建立急性关节炎模型,磁场分别暴露6h和8h,注射弗氏完全佐剂建立慢性关节炎模型,磁场暴露7d,每天6h,观察炎症肿胀和炎性细胞因子的改变情况。结果:急性和慢性关节炎大鼠经过不同时间的磁场暴露后,踝关节和足垫肿胀均有不同程度的减轻,急性模型暴露6h和慢性模型血清和关节液的细胞因子均无改变,急性模型暴露8h血清IL-6,关节浸液IL-6和TNF-α均比对照组降低,其余指标无变化,但是有下降的趋势。结论:极低频磁场对急性和慢性关节炎肿胀有抑制作用,每天暴露8h可以部分抑制细胞因子的产生,但其详细机制还有待于进一步研究。  相似文献

目的:探讨极低频正弦磁场对痛阈的影响以及脑内氨基酸神经递质在磁场镇痛中的作用。方法:20只SD大鼠暴露于55.6Hz、8.1mT正弦磁场中并利用辐射热甩尾法检测痛阈,同时用高效液相色谱检测暴露第11天和第14天大脑皮层和延髓谷氨酸和γ-氨基丁酸(GABA)含量。结果:与对照组相比大鼠磁场暴露第10天和第11天痛阈明显提高(P0.05)。第11天大脑皮层和延髓的γ-氨基丁酸水平有明显改变(P0.05)。结论:极低频正弦磁场有镇痛作用,调节γ-氨基丁酸可能是其作用机制之一。  相似文献

Extremely low frequency (ELF) magnetic fields (MF) are omnipresent in our modern daily environment, but their effects on humans are still not clearly established. The aim of this study was to determine the effect of a 50 Hz, 1,000 microT MF centered at the level of the head on human index finger micro-displacements. Twenty-four men recruited among the personnel of the French company, Electricité de France (EDF), completed the experiment. Their postural and kinetic tremors were recorded under four "field-on" and four "field-off" conditions, each tested during a real and a sham sequence. Eight postural and four kinetic tremor characteristics were calculated on recorded time series and were used for statistical analysis. No effect of the MF was found for kinetic tremor. Concerning postural tremor, the proportion of oscillations at low frequencies (between 2 and 4 Hz) was higher during the real than during the sham exposure sequence (P<.05). It suggests that MF could have a subtle delayed effect on human behavior, which is clearly not pathological. These results should be taken into account for the establishment of new exposure limits.  相似文献

The combined effects of diabetes and a 50 Hz, 5 mT RMS flux density sinusoidal magnetic field for 8 h a day, for 21 consecutive days on the permeation of Evans-blue dye through the blood-brain barrier were studied in male Wistar albino rats. Our results suggest that magnetic field has no effect on the blood-brain barrier permeability in normoglycemic animals, but that diabetic rats are vulnerable to magnetic fields.  相似文献

The induction of sister chromatid exchanges (SCEs) was evaluated in the cultured mouse m5S cells after exposure to extremely low frequency magnetic field (ELFMF; 5, 50 and 400 mT). Exposure to 5 mT and 50 mT ELFMF led to a very small increase in the frequency of SCEs, but no significant difference was observed between exposed and unexposed control cells. The cells exposed to 400 mT ELFMF exhibited a significant elevation of the SCE frequencies. There was no significant difference between data from treatments with mitomycin-C (MMC) alone and from combined treatments of MMC plus ELFMF (400 mT) at any MMC concentrations from 4 to 40 nM. These results suggest that exposure to highest-density ELFMF of 400 mT may induce DNA damage, resulting in an elevation of the SCE frequencies. We suppose that there may be a threshold for the elevation of the SCE frequencies, that is at least over the magnetic density of 50 mT.  相似文献

Previous experiments with mice have shown that a repeated 1 h daily exposure to an ambient magnetic field shielded environment induces analgesia (anti-nociception). This shielding reduces ambient static and extremely low frequency magnetic fields (ELF-MF) by approximately 100 times for frequencies below 120 Hz. To determine the threshold of ELF-MF amplitude that would attenuate or abolish this effect, 30 and 120 Hz magnetic fields were introduced into the shielded environment at peak amplitudes of 25, 50, 100 and 500 nT. At 30 Hz, peak amplitudes of 50, 100, and 500 nT attenuated this effect in proportion to the amplitude magnitude. At 120 Hz, significant attenuation was observed at all amplitudes. Exposures at 10, 60, 100, and 240 Hz with peak amplitudes of 500, 300, 500, and 300 nT, respectively, also attenuated the induced analgesia. No exposure abolished this effect except perhaps at 120 Hz, 500 nT. If the peak amplitude frequency product was kept constant at 6000 nT-Hz for frequencies of 12.5, 25, 50, and 100 Hz, the extent of attenuation was constant, indicating that the detection mechanism is dependent on the nT-Hz product. A plot of effect versus the induced current metric nT-Hz suggests a threshold of ELF-MF detection in mice at or below 1000 nT-Hz.  相似文献
