Summary Fibroblast cultures from 49 possible Hunter disease carriers were collected. These cultures were analysed for the incorporation of 35S-sulphate into acid mucopolysaccharides in the presence and the absence of fructose 1-phosphate. For 10 of these women more than one abnormal result was observed, when two or three cultures from each individual were tested. For six additional women only one abnormal result was found, when three cultures for each of these females were analysed. The implication that just one abnormal result indicates carriership stems from the observation that 24 out of 25 obligate carriers have been confirmed by this criterion (Tønnesen et al. 1983). By mean of the same criterion we have thus established carriership for 16 possible carriers. From genetic inference three additional carriers were found among the females showing normal results in the fibroblast cultures. As a test of the reliability of the method, analyses of the tested informative female offspring of Hunter carriers showed 20 of 38 informative females to be carriers.  相似文献

Polyandry is a widespread mating strategy, found in a broad number of taxa. Among amphibians, polyandry, and multiple paternity as its direct consequence, is quite common in salamanders, especially within Ambystomatidae and Plethodontidae. In the suborder Salamadroidea the existence of two different types of spermatheca allows several kinds of polyandry strategies to appear. We used multilocus microsatellite genotyping to investigate the presence of polyandry and its effects on the paternity in a previously unstudied species with a terrestrial habit, Salamandrina perspicillata. We collected gravid females in their natural habitat and analysed the paternity of the offspring by using the software COLONY and GERUD. We found that all the analysed clutches had been fertilized by 2–4 males and that in every clutch one male had sired most of the offspring. Our results confirmed that polyandry is an important component of the mating system of this species, suggesting that females are able to recognize the sperm of the male that will provide a genetic benefit for their offspring. We found evidence of female cryptic choice based on males' genetic dissimilarity: (1) males who sire most of the offspring of a given female tend to be genetically different from their sexual partner; (2) a same male, when mated with two females, sired a proportion of the offspring inversely correlated with his genetic similarity to the female; (3) genetic dissimilarity between mating partners is positively correlated with offspring heterozygosity. According to the genetic compatibility model, we hypothesized that in the observed non resource‐based mating system the indirect benefit for the offspring should reflect interactions between paternal and maternal genomes rather than the inheritance of the so‐called ‘good genes’. This study suggests a polygynandrous mating system for the study species and provides the first report in a salamandrid species in natural condition that reproductive success of males is correlated with genetic dissimilarity between mates. Moreover, we found evidence of an offspring benefit (higher heterozygosity) derived from the most genetically dissimilar father.  相似文献

The last recorded presence of the Eurasian otter (Lutra lutra) in the Netherlands dates from 1989 and concerned a dead individual. In 2002 a reintroduction programme was started, and between June 2002 and April 2008 a total of 30 individuals (10 males and 20 females) were released into a lowland peat marsh in the north of the Netherlands. Noninvasive genetic monitoring based on the genetic profiles obtained from DNA extracted from otter faeces (spraints) was chosen for the post-release monitoring of the population. To this end, the founding individuals were genotyped before release and spraints were collected in the release area each winter from 2002 to 2008. From June 2002 to April 2008 we analysed the genetic profile of 1,265 spraints on the basis of 7–15 microsatellite loci, 582 of which (46%) were successfully assigned to either released or newly identified genotypes. We identified 54 offspring (23 females and 31 males): the females started to reproduce after 2 years and the males after 4 years. The mating and reproductive success among males was strongly skewed, with a few dominant males fathering two-thirds of the offspring, but the females had a more even distribution. The effective population size (Ne) was only about 30% of the observed density (N), mainly because of the large variance in reproductive success among males. Most juvenile males dispersed to surrounding areas on maturity, whereas juvenile females stayed inside the area next to the mother’s territory. The main cause of mortality was traffic accidents. Males had a higher mortality rate (22 out of 41 males (54%) vs. 9 out of 43 females (21%)). During winter 2007/08 we identified 47 individuals, 41 of which originated from mating within the release area. This study demonstrates that noninvasive molecular methods can be used efficiently in post-release monitoring studies of elusive species to reveal a comprehensive picture of the state of the population.  相似文献

Drosophila melanogaster samples were collected from a large population in two habitats: farmyards and distilleries. Samples were taken from two villages in each habitat. Three isofemale lines were established from all four samples and full-sib crosses were set in each isofemale line. Activities of four enzymes (ADH, alpha GPDH, IDH and 6PGDH) were measured in the offspring of each cross on starch gel after electrophoresis. Broad sense heritabilities and additive genetic variances were estimated in all four samples. Most of the activity variation was observed within the isofemale lines. The isofemale lines tended to be more different in the distilleries than in the farmyards. There was no significant difference in the average activities between the two habitats for any of the enzymes investigated. The additive genetic variance of the enzyme activities did not exhibit a consistent habitat pattern. In the farmyard habitat, we detected a higher activity variation in Tiszafüred than in the other village. Strong correlation was observed among the activities of the enzymes investigated. Correlation coefficients indicated higher level of correlation in the samples collected in Tiszafüred than in those originating from Tiszaszolos. The heritability values were rather high and they had a considerable variation both between the habitats and across the enzymes.  相似文献

Age at onset of motor symptoms was collected on 611 persons affected with Huntington disease (HD) among 3,201 persons "at risk" in 108 kindreds. Life-table estimates correcting for truncated intervals of observation (censoring) produced a median age at onset 5 years older than the observed mean. Risk estimates of HD onset for persons at risk, as calculated by life-table methods, were significantly higher for older ages than were estimates based on the observed distribution of onsets. Age-specific incidence was found to be highest at age 35-64 years, a considerably older age interval than suggested by previous estimates. The offspring of affected males had significantly younger onset than did offspring of affected females, and a trend suggesting and excess of paternal descent among juvenile-onset cases was present. Life-table analysis is contrasted with analyses of (a) the observed distribution of age at onset and (b) remote cohorts age 63 or older at the time of data collection. The implications for risk prediction, genetic counseling, and genetic analysis of HD are discussed.  相似文献

Multiple mating is common in many species, but it is unclear whether multiple paternity enhances offspring genetic diversity or fitness. We conducted a survey on wild house mice (Mus musculus musculus), and we found that in 73 pregnant females, 29% of litters had multiple sires, which is remarkably similar to the 23–26% found in feral populations of Mus musculus domesticus in the USA and Australia, respectively. The question is: How has selection maintained multiple mating in these subspecies since the evolutionary divergence, ca. 2800–6000 years ago? We found no evidence that multiple paternity enhanced females’ litter size, contrary to the fertility assurance or genetic benefits hypotheses. Multiple paternity was associated with reduced mean and variance in offspring body mass, which suggests that females allocate fewer resources or that there is increased intrauterine conflict in multiple-versus single-sired litters. We found increased allelic diversity (though not heterozygosity) in multiple-sired litters, as predicted by the genetic diversity hypothesis. Finally, we found that the dams’ heterozygosity was correlated with the mean heterozygosity of their offspring in single-and multiple-sired litters, suggesting that outbred, heterozygous females were more likely to avoid inbreeding than inbred, homozygous females. Future studies are needed to examine how increased genetic diversity of litters and smaller mean (and variance) offspring body mass associated with multiple paternity affect offspring fitness.  相似文献

油茶地蜂Andrena camellia Wu 是江西、湖南等地油茶花期主要传粉昆虫之一。2006–2007年在江西宜春对油茶地蜂除成虫外各虫态的形态特征、生活史及幼虫和成虫的行为等生物学特性进行了观察研究。结果表明: 该蜂在江西宜春1年发生1代。成虫10月中旬开始出巢活动, 雄蜂早于雌蜂3 d左右出现,完成交尾后数日死亡,雄蜂出巢后寿命18 d左右,雌蜂约38 d。卵期约8 d。幼虫期24 d左右,幼虫取食完花粉球后长成老熟幼虫进入滞育期,滞育期约241 d。翌年9月中旬开始化蛹,蛹期约30 d。出土后的雌蜂当天就可以交配,交配后的雌蜂在羽化地点附近选址筑巢,采集油茶的花粉和花蜜于虫室中制成球形蜂粮并在上面产卵,每粒花粉球上产卵一枚。  相似文献

To clarify the genetic relationship between Swamp and River buffaloes, the restriction fragment length polymorphisms (RFLPs) of nuclear genomic ribosomal DNA (rDNA) and cytoplasmic mitochondrial DNA (mtDNA) were analysed. Blood or liver samples from 73 Swamp and three River buffaloes were collected in East and South-east Asian countries. DNA samples from cattle, goats and sheep were used for comparisons. The analysis of rDNA allowed water buffaloes, cattle, goats and sheep to be characterized by four distinct repeat-types. However, swamp and river buffaloes showed the same repeat-type. Divergence of water buffalo and cattle is considered to have occurred approximately four to six million years ago. The RFLPs for mtDNA divided water buffaloes into three haplotypes, swamp-1, swamp-2 and river types. Swamp-1 accounted for 91% of all swamp buffaloes while swamp-2 was observed only in water buffaloes from Thailand (9%). All river buffaloes were of the same haplotype. No differences were observed between swamp and river buffaloes at the rDNA level. In contrast, a few distinct differences between them were found at the mtDNA level. Therefore, mtDNA polymorphisms provide an adequate means for classifying water buffaloes into either swamp or river buffaloes.  相似文献

The sex-allocation behaviour of the solitary ectoparasitoid Dinarmus basalis (Hymenoptera: Pteromalidae) was investigated by examining the female's response to two proximal factors: the host-patch characteristics and the conspecific female density in the patch. The offspring sex-ratio of single females presented with unparasitised hosts was female biased (approximately three daughters to one son in the progeny) whatever the host density tested. The sex-ratio in the presence of hosts parasitised by a conspecific 48 h beforehand was male biased. The proportion of male offspring also increased when the number of conspecific females exploiting the patch changed from a single female to a group of females, whatever the group size. No sexual differences in mortality as a result of larval competition on superparasitised hosts were observed. The offspring sex-ratios observed in response to the host-patch characteristics were the result of manipulation of the egg fertilisation by the females during the oviposition phase. Thus, the variability of the sex-ratio in response to an increase in the number of conspecific females in the same patch represented a reduction in egg fertilisation. These results are analysed with regard to the predictions of Local Mate Competition theory and the females' host discrimination ability.  相似文献

典型的猕猴(Macaca multta)社群为多雄多雌型,雌性留群并形成母系单元,雄性多在亚成年或成年期离开出生群,而群内成年雄性多为外部迁入个体。雄性的迁出被认为可降低近亲繁殖或提升繁殖成功。然而,诸多野外数据显示,少数本群出生雄性个体会居留于出生群一年或数年。尽管驱动雄性离群的因素较复杂(如社会关系、近亲回避、繁殖成功),但繁殖成功的差异可能是驱动雄性离群的主导因素。为探讨居留于出生群是否影响雄性的繁殖成功,于2010年3月至2014年1月,在太行山猕猴国家级自然保护区王屋山地区,以一群野生太行山猕猴为研究对象,采用非损伤取样法并结合分子生物学方法,分析了群内出生和迁入成年雄性个体的繁殖成功(以子代数量评估)。研究发现:(1)群内51只子代个体中有36只个体可以匹配到其遗传学父亲;(2)4个迁入雄性繁衍了34个子代,仅1只群内出生雄性ZM繁衍了2只子代,但群内出生的雄性BB未匹配到子代。本研究提示,迁入雄性较本群出生雄性的繁殖成功较高,即迁移有助于繁殖成功的提升。  相似文献

The design and implementation of assisted reproductive technology to improve genetic diversity and augment captive populations is an important but rarely applied research field in reptiles. Using the corn snake (Elaphe gutatta) as a model, the Henry Doorly Zoo recently produced offspring born as a result of artificial insemination using both fresh, diluted semen, and diluted semen stored at refrigeration for 3 days. Semen was collected noninvasively from sexually mature male corn snakes using a gentle massaging technique, extended in medium then inseminated into the oviducts of adult females. Using molecular genetic techniques to confirm or refute the success of the insemination using primers developed for the black rat snake, Elaphe obsolete, all possible parents and offspring genotypes were evaluated. Biochemical genetic polymorphism in cod from several fjords in Troms, northern Norway was analysed. Gene frequencies at several polymorphic loci ( Hb, Pgi, Ldh, Pgm, Gpd and Idh ) are given. Significant variation was found both within and between fjords, and even between samples from the same locality sampled in different years, indicating a mosaic structure of the cod population in the area studied. The results of the haemoglobin variation are compared to similar results obtained more than 20 years ago in the same and adjacent areas. The biochemical genetic variation is discussed in relation to stability of gene frequencies, isolation mechanisms and migration patterns.  相似文献

Microsatellite analysis was applied to scale samples of Atlantic salmon collected up to 60 years ago. Samples from the 1930s, from a now endangered Danish population, were compared with recent samples (1989), to test if the present population consists of descendants from the original one. Allele frequencies had changed over time, but individuals from the two samples caught about 60 years apart clustered together when compared with the closest neighbouring population and another reference population. However, fewer alleles were detected in the recent sample from the endangered population, most likely due to a population bottleneck or sampling artefacts.  相似文献

Promiscuous systems where both males and females mate several times with different individuals are widespread among mammalian species. As a consequence, females obtain sperm from more than one male and paternity is decided by sperm competition. In theory, females might gain ‘genetic benefits’ for their offspring from this mechanism. In a mating experiment we now demonstrate in the promiscuous rodent Galea musteloides that females which were paired with four males, and became pregnant, weaned more surviving offspring than females which were paired with a single male. Litter sizes did not differ between the two groups. The data support the hypothesis that promiscuous females copulate with several males to induce sperm competition and/or to enforce cryptic female choice and thereby increase the viability of their offspring.  相似文献

Since flowering often varies among years in wind-pollinated woody species, the genetic composition of pollen pools accepted by seed parents can differ between years. The interannual heterogeneity of pollen flow may be important for maintaining genetic diversity within populations because it can increase genetic variation within populations and the effective sizes of the populations. In this study we examined heterogeneity, using paternity analysis and analysis of molecular variance, in the genetic composition of pollen pools among different reproductive years for six Quercus salicina seed parents in an 11.56-ha plot in a temperate old-growth evergreen broadleaved forest. The genotypes at seven microsatellite loci were determined for 111 adult trees and 777 offspring of the six seed parents in 2-5 reproductive years. Genetic differentiation of pollen pools among different reproductive years for each seed parent was significant over all seed parents and for each of four seed parents that were analysed for more than 2 years, but not for either of the other two seed parents (analysed for 2 years). For both the pollen pools originating from inside the plot and those originating from outside it, genetic differentiation among different reproductive years for each seed parent was significant over all seed parents. However, among-year genetic differentiation in the pollen pools originating from within the plot was detected for all four of the seed parents that were analysed for more than 2 years, but for only one of the four in the pools originating from outside the plot. Genetic diversity (estimated as allelic richness and gene diversity) was higher for pollen pools over all reproductive years than for pollen pools in single years. These results indicate that the year-to-year genetic variation of pollen pools increases genetic diversity in offspring and is strongly affected by the variation in pollen parents within the plot because of their high pollination contributions. The high year-to-year variation in pollen parents within the plot and overall supports the hypothesis that the offspring produced across years represent a larger genetic neighbourhood.  相似文献

为了研究我国札如病毒(Sapovirus,SV)的流行情况及变异特点,收集了2006年2月至2007年1月安徽省、福建省、海南省、河北省、河南省等9个地区5岁以下腹泻患儿的粪便标本共1 110份,应用逆转录-聚合酶链反应(RT-PCR)对札如病毒进行检测,结果表明,札如病毒阳性率为0.9%。对检测到的全部10株札如病毒进一步进行系统进化分析,表明这10株札如病毒属于三个基因型:GI/1、GI/3和GII/3。  相似文献

The Australian lizard Egernia stokesii lives in spatially and temporally stable groups of up to 17 individuals. We have recently shown that these groups are comprised of breeding partners, their offspring and, in some cases, highly related adults, providing the first genetic evidence of a family structure in any lizard species. Here we investigated the mating system of E. stokesii using data from up to eight polymorphic microsatellite loci and tested the hypothesis that breeding partners are monogamous both within and between mating seasons. Among 16 laboratory-born litters from field collected gravid females from two sites in South Australia, 75% had a single male parent and no male contributed to more than one litter, indicating a high level of genetic monogamy within a season. Additional analyses of field caught individuals, captured between 1994 and 1998, enabled assignment of parentage for 70 juveniles and subadults. These data showed that most young (88.6%) had both parents from within the same group and that high proportions of males (88.9%) and females (63.6%) have multiple cohorts of offspring only with the same partner. Our results suggest that monogamy both within and between seasons is a common mating strategy of E. stokesii and that breeding partners maintain stable associations together and with multiple cohorts of their offspring over periods of up to at least 5 years.  相似文献

Understanding and monitoring the population status of endangered species is vital for developing appropriate management interventions. We used noninvasive genetic analyses to obtain ecological and genetic data on the last remaining Far Eastern leopard population in the world. During seven winters from 2000–2001 to 2007–2008, we collected feces, hair, and saliva from most of the leopard habitat. Of the 239 leopard samples collected during the study period, 155 were successfully genotyped at 13 microsatellite loci and 37 individuals (18 males and 19 females) were identified. Population size estimates based on the Capwire model were 28 (95 % CI 19–38) in 2002–03 and 26 (95 % CI 13–33) in 2007–2008. The leopard population had a low level of genetic diversity (expected and observed heterozygosity = 0.43; average number of alleles per locus = 2.62), and effective population size was estimated to be low (N e = 7–16) by two genetic-based methods. We observed little improvement in the genetic diversity during the study period and did find an indication of allele loss compared with individuals from the mid-1990s, suggesting that the remaining population will continue to suffer loss of genetic diversity. Given the small population size and the low genetic diversity, with little expectation of replenishment of the genetic variation by natural immigration, successful expansion of available habitat and development of a second population based on captive individuals may be crucial for persistence of this leopard subspecies in the wild.  相似文献

The bluntnose sixgill shark (Hexanchus griseus) is a widely distributed species found in tropical and temperate waters of every ocean, yet we know relatively little about their basic biology including their life history and population structure. From 2003–2007, we collected over 300 biopsy samples from sixgills during research operations in Puget Sound, WA, USA. Genotypic data using ten polymorphic microsatellites were used to describe sixgill genetic diversity, relatedness and mating pattern. Diversity within sixgills was found to be moderate with an average observed heterozygosity of 0.45, an average expected heterozygosity of 0.61, an average of 12 alleles per locus, and an average allelic richness of eight within microsatellite loci. Our data suggests all of the sampled individuals come from one intermixing population, and we found no historical evidence of significant population bottlenecks. Many of the sharks were sampled using longline techniques with several sharks captured at the same time and place. Similarly, multiple sharks were sampled on several occasions during research events at the Seattle Aquarium. The proportion of individuals that were full- or half-siblings was high among sharks sampled at the same time and place (range 0.65–0.87). Analysis of the genetic relationship between one large female washed ashore and 71 of her near-term pups suggested a polyandrous mating system with as many as nine males contributing to her offspring. This study is the first to investigate genetic diversity, relatedness and paternity within sixgill sharks and sheds light on important conservation implications for this little known shark population.  相似文献
