Growth performance of the Antarctic bivalve Laternula elliptica was examined both by shell microstructural observation and by applying a fluorescent substance, tetracycline, as a shell growth marker. The shell was composed of two calcareous layers: the thick outer layer was homogeneous or granular in structure and the thin inner layer was nacreous. The architecture of Antarctic L. elliptica was different from that of temperate L. marilina, and the ratio of thickness between the outer and inner layers appeared to be different. The growth rate of the nacreous layer was analyzed to be very low. High correlations were found between the major axis of chondrophore and both shell length and shell dry weight, respectively. It is suggested that the chondrophore is an appropriate growth indicator, and combining the information of growth increments with the fluorescent method may be useful in estimating the bivalve growth performance in the Antarctic sea.  相似文献

Burrowing was investigated in two Antarctic infaunal bivalve molluscs, Laternula elliptica and Yoldia eightsi, representing amongst the least and most active members of the class Bivalvia in the Southern Ocean. Burrowing rate was expressed via the Burrowing Rate Index (BRI=[3wet weight/time to bury]×104), and produced values of 0.1–10.6 for L. elliptica and 8.8–49.8 for Y. eightsi. These compare with values ranging from 3 to 2,000 for N. American bivalves (mean=222, SE=42.6, n=81), and 200 to 2,200 for Hong Kong bivalves (mean=1,140, SE=346, n=6). Values for the Antarctic species are, therefore, low compared to warmer-water bivalves, and the values below 1 for large L. elliptica are the lowest on record by around ×5. There is no compensation of burrowing activity for low temperature in these species. The relative BRI values for L. elliptica and Y. eightsi reflect the differences in their mode of life, with the former being large, sedentary and suspension-feeding, and the latter being smaller, mobile, ploughing through the sediment and feeding on sediment-surface organic matter. Burrowing in L. elliptica is unexpected, because other members of the Laternulidae do not burrow. This ability is most probably a response to the regular disturbance of sediments in Antarctica by ice, and the strong selective advantage to being able to resume a protected position after disturbance. The burrowing cycle in L. elliptica is composed of three main phases: (1) foot extension and sediment penetration; (2) foot dilation to form an anchor; (3) the drawing down of the shell by contraction of the pedal retractor muscles. Burrowing in Y. eightsi also has three phases: (1) foot extension and penetration of the sediment (digging); (2) rocking movements in the upright position; (3) shell anchorage. In excess of burrowing activity, L. elliptica exhibits a unique suite of movements when exposed at the surface. These comprise levering, where the tips of the siphons are pressed against the sediment to lift the shell from the substratum, looping, where the siphons are extended and rotated and, in the process, translocate the whole animal across the sediment, and jetting, where water is ejected forcibly through the siphons while their tips are directed towards the sediment, lifting part or all of the animal clear of the substratum. In the field, following exhumation by icebergs, these activities serve to place the animal in a favourable position for reburial, which is a clear advantage in disturbed polar environments where predatory nemerteans and asteroids are abundant.  相似文献

Lipid peroxidation in Laternula elliptica was assessed by detecting lipid radicals by electronic paramagnetic resonance. The values were compared with data from the temperate mud clam Mya arenaria. Lipid radical content was higher in the Antarctic bivalve than in the temperate mud clam, even within the range of its habitat temperature. The rate of generation of lipid radicals was affected by the iron content in the samples. The iron content in individual samples of digestive glands in L. elliptica ranged from 3 to 6 nmol g−1 fresh weight (fwt) and in M. arenaria from 0.6 to 2.7 nmol g−1 fwt. Arrhenius plots, developed from the rates obtained in the presence of 25 μM iron, showed no significant differences between the activation energy calculated for digestive glands of L. elliptica and M. arenaria. The Fe3+ reduction rate in L. elliptica was higher than in M. arenaria (4.7±0.9 vs. 1.8±0.4 nmol mg−1 protein min−1, respectively). L. elliptica had a higher content of α-tocopherol and β-carotene than M. arenaria. Our data suggest that increased lipid radical content in the membranes of cold-adapted organisms could be related to iron content.  相似文献

Glutathione‐S‐transferases have been identified in all the living species examined so far, yet little is known about their function in marine organisms. In a previous report, the recently identified GST from Antarctic bivalve Laternula elliptica (LeGST) was classified into the rho class GST, but there are several unique features of LeGST that may justify reclassification, which could represent specific shellfish GSTs. Here, we determined the crystal structure of LeGST, which is a shellfish specific class of GST. The structural analysis showed that the relatively open and wide hydrophobic H‐site of the LeGST allows this GST to accommodate various substrates. These results suggest that the H‐site of LeGST may be the result of adaptation to their environments as sedentary organisms. Proteins 2013. © 2012 Wiley Periodicals, Inc.  相似文献

The success of any organism depends not only on niche adaptation but also the ability to survive environmental perturbation from homeostasis, a situation generically described as stress. Although species-specific mechanisms to combat “stress” have been described, the production of heat shock proteins (HSPs), such as HSP70, is universally described across all taxa. Members of the HSP70 gene family comprising the constitutive (HSC70) and inducible (HSP70) members, plus GRP78 (glucose-regulated protein, 78 kDa), a related HSP70 family member, were cloned using degenerate polymerase chain reaction (PCR) from two evolutionary divergent Antarctic marine molluscs (Laternula elliptica and Nacella concinna), a bivalve and a gastropod, respectively. The expression of the HSP70 family members was surveyed via quantitative PCR after an acute 2-h heat shock experiment. Both species demonstrated significant up-regulation of HSP70 gene expression in response to increased temperatures. However, the temperature level at which these responses were induced varied with the species (+6–8°C for L. elliptica and +8–10°C for N. concinna) compared to their natural environmental temperature). L. elliptica also showed tissue-specific expression of the genes under study. Previous work on Antarctic fish has shown that they lack the classical heat shock response, with the inducible form of HSP70 being permanently expressed with an expression not further induced under higher temperature regimes. This study shows that this is not the case for other Antarctic animals, with the two molluscs showing an inducible heat shock response, at a level probably set during their temperate evolutionary past.  相似文献

采用实验生态学方法,在室内水槽条件下研究了金乌贼(Sepia esculenta Hoyle,1885)繁殖过程中社群等级的形成对其行为表型和能量代谢的影响,分析测定了不同优势等级雌雄个体腕部肌肉和性腺组织中己糖激酶(Hexokinase, HK)、丙酮酸激酶(Pyruvate kinase, PK)、乳酸脱氢酶(Lactate dehydrogenase, LDH)、苹果酸脱氢酶(Malate dehydrogenase, MDH)、柠檬酸合酶(Citrate synthetase, CS)活性以及乳酸(Lactic acid, LD)含量。结果显示:(1)金乌贼繁殖期不同优势等级雌雄个体之间行为表型具有显著差异,优势雄性个体游动悬浮、争斗时间显著高于劣势个体,而优势雌性个体静止伏底时间高于劣势个体,游动悬浮时间低于劣势雌性;(2)优势雄性个体在争斗过程中主要通过无氧代谢提供能量,而处于游动悬浮状态时通过有氧代谢提供能量。主要表现在优势雄性个体肌肉中无氧代谢酶(PK、HK、LDH)活性显著高于劣势个体(P0.05),有氧代谢酶(MDH、CS)活性也显著高于劣势个体,雌性个体之间则差异不显著(P0.05);(3)繁殖期雌性个体通过减少运动量来储存能量用于产卵繁殖,主要表现在优势雌性个体肌肉中有氧代谢酶(MDH、CS)活性低于劣势个体,而在性腺中恰恰相反;(4)运动表型与能量代谢之间存在显著相关性,表现在游动悬浮时间与有氧代谢酶(MDH、CS)活性呈显著正相关(P0.001),争斗时间与无氧代谢酶(PK、HK、LDH)及乳酸(LD)含量呈显著正相关(P0.05)。结果表明,社群等级高的雄性个体运动能力强,具有较高的生存适应性。而社群等级高的雌性个体多处于静止状态,以便更好地储存能量用于繁殖。研究结果为金乌贼健康苗种培育以及规模化繁殖技术优化提供了理论依据。  相似文献

 The effects of various compounds bearing an N-OH group such as N-hydroxy-guanidines, amidoximes, and hydroxylamines, on bovine and rat liver arginases was studied. Some of these compounds with an l-α-amino acid function at an appropriate distance from the N-OH group acted as strong competitive liver arginase inhibitors, displaying Ki values between 4 and 150 μM. Two compounds, N ε-hydroxy-l-lysine and N ω-hydroxy-d,l-indospicine, which exhibited Ki values of 4 and 20 μM (at pH 7.4), were the most potent inhibitors of arginase described to date. The distance between the α-amino acid and N-OH functions appeared to be crucial for potent inhibition of arginase, as N δ-hydroxy-l-ornithine, which has one -CH2 group less than N ε-hydroxy-l-lysine, exhibited a 37-fold higher Ki value than N ε-hydroxy-l-lysine. Based on these results, a model for the interaction of N ω-hydroxyamino-l-α-amino acids with the arginase active site is proposed. This model involves the binding of the N-OH group of the inhibitors to the arginase Mn(II) center and suggests that N ε-hydroxy-l-lysine is a good transition state analog of arginase.  相似文献

A malate dehydrogenase (MDH) from Streptomyces avermitilis MA-4680 (SaMDH) has been expressed and purified as a fusion protein. The molecular mass of SaMDH is about 35 kDa determined by SDS-PAGE. The recombinant SaMDH has a maximum activity at pH 8.0. The enzyme shows the optimal temperature around 42°C and displays a half-life (t 1/2) of 160 min at 50°C which is more thermostable than reported MDHs from most bacteria and fungi. The k cat value of SaMDH is about 240-fold of that for malate oxidation. In addition, the k cat/K m ratio shows that SaMDH has about 1,246-fold preference for oxaloacetate (OAA) reduction over l-malate oxidation. The recombinant SaMDH may also use NADPH as a cofactor although it is a highly NAD(H)-specific enzyme. There was no activity detected when malate and NADP+ were used as substrates. Substrate inhibition studies show that SaMDH activity is strongly inhibited by excess OAA with NADH, but is not sensitive to excess l-malate. Enzymatic activity is enhanced by the addition of Na+, NH4 +, Ca2+, Cu2+ and Mg2+ and inhibited by addition of Hg2+ and Zn2+. MDH is widely used in coenzyme regeneration, antigen immunoassays and bioreactors. The enzymatic analysis could provide the important basic knowledge for its utilizations.  相似文献

Summary The presence of empty shells of Laternula elliptica that provide refuge sites for juvenile fishes on the soft-bottom surface of South Bay, Antarctica, has been difficult to explain because this bivalve normally lives deeply buried (>50 cm). L. elliptica individuals unburied and aggregated at the surface by the action of icebergs could be consumed by mobile predators. This paper examines experimentally both burying rates and predation upon unburied L. elliptica in the field at South Bay. Live and intact L. elliptica were hand-collected by diving, and separated in 16 groups of 10 each, and placed on mud-sand bottoms protected from the ice disturbance at a depth of 17 m in South Bay. Eight groups were confined by open wooden frames so as to exclude predators, mainly star-fish and gastropods, the other eight remained as control. Species and number of predators and burying rates were estimated during 23 days. Within the first 24 h, 30% of the bivalves buried themselves, after which, rates fell abruptly. By the end of the day 23, only 60% had successfully buried; of the 40% which remained unburied, 20% were consumed by the following sequence of predators: Odontaster validus, Cryptasterias turqueti, Parborlasia corrugatus, Neobuccinum eatoni and two species of amphipods; all completely consumed one bivalve in 5–7 days. The 20% that remained unburied constituted potential prey though they showed no signs of being attacked by O. validus during the observation period. In addition to serving as prey, L. elliptica provides empty and intact shells used as refuge sites by juvenile fishes.  相似文献

Macrophages, upon phagocytosing endospores of Bacillus anthracis, up-regulate the expression of the immunological isoform of nitric oxide synthase, NOS 2, concomitant with production of nitric oxide (NO•) from metabolism of l-arginine. We have previously demonstrated that macrophages that secrete NO• kill the bacilli of B. anthracis. To circumvent this microbicidal activity of NO•, B. anthracis has evolved pathways that include the enzyme arginase, which metabolizes l-arginine to ornithine and urea. Compounds that inhibit arginase might, therefore, offer a therapeutic approach to controlling B. anthracis infection. 2(S)-Amino-6-boronohexanoic acid (ABH) has been reported to be an inhibitor of mammalian arginase. In this study, we explore the inhibitory effect of ABH against B. anthracis arginase and its potential for future development, as an effective therapeutic agent against microbial infection. We found that ABH is an inhibitor of bacterial arginase in several different endospore strains of B. anthracis. Further, ABH inhibits neither the phagocytosis of these endospores nor the up-regulation of NOS 2 concomitant with secretion of NO•. These findings set the stage to determine how efficacious ABH will be in promoting NO•-mediating killing of B. anthracis.  相似文献

The gene encoding malate dehydrogenase (MDH) was overexpressed in a pflB ldhA double mutant of Escherichia coli, NZN111, for succinic acid production. With MDH overexpression, NZN111/pTrc99A-mdh restored the ability to metabolize glucose anaerobically and 0.55 g/L of succinic acid was produced from 3 g/L of glucose in shake flask culture. When supplied with 10 g/L of sodium bicarbonate (NaHCO3), the succinic acid yield of NZN111/pTrc99A-mdh reached 1.14 mol/mol glucose. Supply of NaHCO3 also improved succinic acid production by the control strain, NZN111/pTrc99A. Measurement of key enzymes activities revealed that phosphoenolpyruvate (PEP) carboxykinase and PEP carboxylase in addition to MDH played important roles. Two-stage culture of NZN111/pTrc99A-mdh was carried out in a 5-L bioreactor and 12.2 g/L of succinic acid were produced from 15.6 g/L of glucose. Fed-batch culture was also performed, and the succinic acid concentration reached 31.9 g/L with a yield of 1.19 mol/mol glucose.  相似文献

The world’s oceans are changing, and dramatic shifts have been documented in the Southern Ocean. The consequences of these shifts to coastal benthic organisms are difficult to predict at present, as ocean warming may increase primary production and food resources for benthic consumers, whilst OA may have negative impacts that differentially affect various species and life stages. A model was developed to investigate how different scenarios of change may influence population size of the Antarctic bivalve Laternula elliptica. The model describes potential implications of both pH and temperature change on survivorship and reproductive output of a population of this bivalve species in McMurdo Sound, Ross Sea. Implications of increases and decreases in mortality rate across different life stages of the population (early, mid and late) were assessed. Additionally, effects on energetic resource partitioning and dictating reproductive potential (RP) were also investigated. Significant declines in RP, due to increased basal metabolic demand, were associated with even relatively small changes in temperature and pH, resulting in populations declining to 25 % of the starting equilibrium density within 60 years. As L. elliptica is a pivotal species to the functionality of the Antarctic coastal benthic ecosystem, wide spread repercussions are expected if populations are impacted as the model predicts. Although further model development is required to explore the ecosystem implications of the population decline described in this paper, this work allows a better understanding of the consequences of change as soon as data on the direction and magnitude of the changes affecting Antarctic seas become available.  相似文献

We document differences in shell damage and shell thickness in a bivalve mollusc (Laternula elliptica) from seven sites around Antarctica with differing exposures to ice movement. These range from 60% of the sea bed impacted by ice per year (Hangar Cove, Antarctic Peninsula) to those protected by virtually permanent sea ice cover (McMurdo Sound). Patterns of shell damage consistent with blunt force trauma were observed in populations where ice scour frequently occurs; damage repair frequencies and the thickness of shells correlated positively with the frequency of iceberg scour at the different sites with the highest repair rates and thicker shells at Hangar Cove (74.2% of animals damaged) compared to the other less impacted sites (less than 10% at McMurdo Sound). Genetic analysis of population structure using Amplified Fragment Length Polymorphisms (AFLPs) revealed no genetic differences between the two sites showing the greatest difference in shell morphology and repair rates. Taken together, our results suggest that L. elliptica exhibits considerable phenotypic plasticity in response to geographic variation in physical disturbance.  相似文献

Total lipid content, lipid classes and fatty acid composition were studied in various tissues of the Antarctic clam Laternula elliptica in an early austral summer. A histological examination of the gonads revealed that most of the clams examined were spawning or ready to spawn. Lipid content was highest in gills (14.9% of tissue dry weight), followed by gonads (10.9%) and digestive glands (9.9%), and averaged 8.2% for the soft tissues. The overall lipid contents were relatively low compared to temperate bivalves at a similar reproductive stage. Lipid class composition in the total lipid of L. elliptica was quite similar to those of most marine bivalves at lower latitudes, being dominated by triacylglycerols (19.3–41.4% of total lipids) and phospholipids (18.9–28.3%) in most of the organs. Large amounts of triacylglycerol deposits in non-reproductive tissues, particularly in siphon and gill, indicate a potential role of lipid as maintenance energy reserve, although the low lipid contents suggest that lipid may not serve as an energy reserve for any food-limited periods. Fatty acid composition in L. elliptica was also typical of marine bivalves with predominance of 16:0 (26%) and 20:5n-3 (18%) acids. Total fatty acids from the soft tissues showed a moderate level of unsaturation (50.6%), and about 35% of the total fatty acids were polyunsaturated. These values were not significantly different from, or even lower than those of marine bivalves in warmer waters. However, the content of 20:5n-3 (18.2% of total fatty acids), which dominated n-3 polyunsaturated fatty acids, was similar to those reported for other marine bivalve species in temperate waters. The fatty acid composition of L. elliptica reflected dietary input of some microalgal species. The nanoflagellates Cryptomonas spp., which were reportedly rich in 16:0, 18:3n-3 and 20:5n-3, predominated in the water column during the present investigation. Accepted: 19 June 1999  相似文献

Crocus sativus L., cultivated since ancient times as the source of saffron, is a triploid plant that can be propagated only via its corms which undergo a period of dormancy. Understanding the processes taking place in the corm is essential to preserve the plant and improve its quality. Color and taste being of prime importance in the quality of the saffron spice, knowledge on polyphenol oxidase (PPO) activity in the plant is of particular interest given the role of the enzyme in fruit and vegetable browning during processing and during the storage of processed food. In this paper, PPO activity was investigated for the first time in extracts obtained from dormant C. sativus L. corms. PPO activity was detectable using l-DOPA, pyrogallol, catechol or p-cresol as substrate, each being oxidized to its corresponding o-quinone; no activity was detectable with l-tyrosine, tyramine or phenol as substrate. Two pH optima, respectively at 4.5 and 6.7, were observed with all substrates and a third one, at 8.5, was found with l-DOPA and p-cresol. Kinetics parameters studied at pH 6.7 indicated the highest catalytic efficiency (in units mg−1 prot mM−1) with pyrogallol: 150, then catechol: 39, l-DOPA: 6.4 and p-cresol: 4.6. The enzymatic activity was inhibited by 50% in the presence of 0.22, 0.35, 0.5 and 0.7 mM kojic acid with, respectively, catechol, pyrogallol, p-cresol and l-DOPA as substrate. When stained for PPO activity, non-denaturing gel electropherograms of extract revealed three distinct bands, indicating the presence of multiple isoenzymes in dormant C. sativus L. corms.  相似文献

Mitochondrial volume density (Vv(mt,f)), cristae surface density (Sv(im,mt)), cristae surface area (Sv(im,f)) and citrate synthase (CS) activity were analysed as indicators of thermal acclimation in foot muscle of the limpet, Nacella concinna, and the clam, Laternula elliptica, collected from 4 locations within the Southern Ocean, South Georgia (54°S, N. concinna only), Signy (60°S), Jubany (L. elliptica only ? 62°S) and Rothera (67°S). Animals were acclimated to 0.0 °C whilst a sub-set of N. concinna (South Georgia, Signy and Rothera) and L. elliptica (Rothera) were acclimated to 3.0 °C. At 0.0 °C N. concinna had higher Vv(mt,f), Sv(im,mt), Sv(im,f) and muscle fibre specific CS activity than L. elliptica which correlated with the more active life style of N. concinna. However, mitochondrial density was very low, 1–2% in both species, suggesting that low temperature compensation of mitochondrial density is not a universal evolutionary response of Antarctic marine ectotherms. Both Sv(im,mt) and Sv(im,f) were reduced by warm acclimation of N. concinna. South Georgia N. concinna maintained muscle fibre specific CS activity after acclimation, in contrast to N. concinna from Rothera and Signy and L. elliptica from Rothera, indicating that they have the physiological plasticity to respond to their warmer, more variable thermal environment.  相似文献

The anatomical and morphological structure of the lichenCaloplaca coralligera (Hue.)Zahlbr. was investigated in connection with mechanisms of colonization and adaptation to the special conditions of maritime Antarctic.Caloplaca coralligera seems to be endemic to the antarctic region. The lichen is unique in its morphology, growing like a crustose lichen with a prothallus, but the very thallus being composed of many vertical, frutescent branches not higher than large isidia. Due to fusions of the distal parts of the branches the morphology is characterized by air-filled cavities between the erect thallus parts where the phycobionts are located only close to the outermost surface. But also anatomically a system of air-filled cavities is developed by a net-like structure of hyphae. This may result in an insulation from temperature exchanges with the rocky substrate. It is speculated that this peculiar thallus structure might be advantageous to a lichen growing on compact substrates by buffering the diurnal temperature extremes which are characteristic for rocks under a strong radiation exchange with the atmospheric surrounding.Dedicated to Prof. DrO. Sporosarcina ureae BS 860, a motile, sporeforming coccus, possesses the enzymes required for a functioning urea (ornithine) cycle. This is only the second known example of urea cycle activity in a prokaryote. Specific activities are reported for ornithine carbamoyltransferase, argininosuccinase, arginase, and urease. Although argininosuccinate synthetase activity could not be detected directly in crude cell extracts, indirect evidence from radiocarbon tracing data for arginine synthesis from the substrate, l-[1-14C]-ornithine, strongly suggest the presence of this or other similar enzyme activity. Furthermore, good growth in defined media containing either 1.0% glutamine, ornithine, or citrulline as sole carbon sources suggests argininosuccinate synthetase activity is necessary for arginine synthesis. The effect of varying pH on arginase and urease activities indicate that these two enzymes may function within the context of the urea cycle to generate ammonia for amino acid synthesis, as well as for raising the pH of the growth micro-environment.  相似文献
