We have previously determined that larval feeding experience with a feeding/oviposition deterrent modified the feeding responses of larvae and oviposition responses of subsequent moths. These behavioural changes were attributed to learning, but the possibility of chemical legacy could not be ruled out. In the present study, we have topically applied a feeding/oviposition deterrent plant extract from Hoodia gordonii (Masson) Sweet ex Decne (Asclepiadaceae) to larvae, pupae, and adults of Trichoplusia ni (Hübner) (Lepidoptera: Noctuidae) to determine whether the feeding response of larvae and oviposition response of subsequent female moths is similarly modified by chemicals applied to the external surface of the insect. Our results indicate that traces of the extract that may be present internally or externally on the larvae do not reduce the feeding deterrent response of larvae. Furthermore, traces of the extract in or on larvae, pupae, or adult moths did not alter oviposition choice of female moths, leading us to discount the role of experience through topical application in this study. The fact that feeding/oviposition choice was only influenced by prior feeding experience of the larvae and not by topical administration suggests that habituation via sensory stimulation through mouthpart chemosensilla is likely a central phenomenon. Continuous exposure of adult moths to the extract over a period of 7 days did not affect the oviposition response of the female moths, ruling out the role of adult experience on host-plant selection in T. ni . To the best of our knowledge, this is the first study to examine the role of experience via topical application of chemicals onto all life stages of the insect except the egg. Chemical legacy may not be playing a role in influencing the oviposition choices of female T. ni moths.  相似文献

Angiotensin converting enzyme (ACE) is a zinc metallopeptidase capable of cleaving dipeptide or dipeptideamide moieties at the C-terminal end of peptides. ACE is present in the hemolymph and reproductive tissues of insects. The presence of ACE in the hemolymph and its broad substrate specificity suggests an important role in processing of bioactive peptides. This study reports the effects of ACE inhibitors on larval growth in the cotton leafworm Spodoptera littoralis. Feeding ACE inhibitors ad lib decreased the growth rate, inhibited ACE activity in the larval hemolymph, and down-regulated trypsin activity in the larval gut. These results indicate that S. littoralis ACE may influence trypsin biosynthesis in the larval gut by interacting with a trypsin-modulating oostatic factor (TMOF). Injecting third instar larvae with a combination of Aea-TMOF and the ACE inhibitor captopril, down-regulated trypsin biosynthesis in the larval gut indicating that an Aea-TMOF gut receptor analogue could be present. Injecting captopril and enalapril into newly molted fifth instar larvae stopped larval feeding and decreased weight gain. Together, these results indicate that ACE inhibitors are efficacious in stunting larval growth and ACE plays an important role in larval growth and development.  相似文献

Angiotensin I-converting enzyme (ACE) is a dipeptidyl carboxypeptidase that removes C-terminal dipeptides from relatively short oligopeptides, usually smaller than 15 amino acids. In mammals, the enzyme has several important roles in the metabolism of vasoactive peptides, but its physiological role in insects is not fully understood. We now report the properties of an ACE in a lepidopteran species (the tomato moth, Lacanobia oleracea) and suggest new physiological roles for the enzyme in this insect. ACE activity increases four-fold during the last stadium and in early pupae, a rise which, in its timing, is similar to what has been observed previously in the transition of larva to pupa in Drosophila melanogaster. This suggests that the increase in ACE activity might be of general importance for peptide metabolism during metamorphosis in holometabolous insects. High levels of ACE activity were found in the haemolymph of sixth stadium larvae and adult insects, and in the reproductive tissues of both male and female adults. Almost all of the ACE activity in the reproductive tissues was found in the accessory glands of the male and the spermatheca and bursa copulatrix of the female. The decline in accessory gland ACE in mated males and the concomitant rise in ACE activity in the spermatheca and bursa copulatrix of the female suggested the transfer of ACE from the male to the female during copulation. Using several convenient peptides as substrates, we have shown that the spermatophore/bursa copulatrix taken from mated female insects possess an aminopeptidase, a carboxypeptidase and a dipeptidase, in addition to high levels of ACE. These peptidases might be involved in the breakdown of proteins to peptides and eventually to amino acids in the spermatophore. Evidence for such a proteolytic pathway and its role in providing substrates for the TCA cycle has been obtained previously in a study of reproduction in Bombyx mori.  相似文献

Vascular angiotensin converting enzyme (ACE) may be important as a potential site of action for the antihypertensive effect of ACE inhibitors. ACE activity, estimated by hydrolysis of [3H] HipGlyGly, was similar in the aorta, mesenteric and carotid arteries of SHR. ACE activity in veins was not as consistent and was significantly lower in the jugular veins and vena cava of SHR than in the arteries. Nevertheless, ACE activity in all the blood vessels examined, although less than lung ACE activity, was higher than ACE activity found in other tissues such as brain, heart and kidney. Equieffective antihypertensive doses of captopril (10 mg/kg p.o.) and MK-421 (1.0 mg/kg p.o.) dramatically inhibited ACE activity in all the arteries and veins examined. Maximal ACE inhibition occured within 15 minutes after the oral administration of captopril. In contrast, maximal ACE inhibition was slower in onset and of longer duration after MK-421 than after captopril for all the vessels. Thus, relatively high ACE activity can be measured in both arteries and veins from the spontaneously hypertensive rat (SHR) and ACE was dramatically inhibited after antihypertensive oral doses of captopril or MK-421. Furthermore, vascular ACE inhibition can be used to compare the onset and duration of activity of ACE inhibitors; MK-421 has a longer onset and duration in SHR than captopril based on inhibition of ACE in blood vessels.  相似文献

Abstract  In order to explore the role of ecdysteroids in development and reproduction of ixodid ticks, we studied the quantitative variation of ecdysteroids in the hemolymph, synganglion, ovary and whole body of the female ixodid ticks, Dermacentor niveus and Haemaphysalis longicornis , before and after engorgement and oviposition by HPLC and RIA. The ecdysteroid content in eggs of these ticks was determined by HPLC. The results indicated that before engorgement the quantitative variation of ecdysteroids in the whole female body was not significant, but their levels increased rapidly after engorgement. The ecdysteroid titer in hemolymph was peaked on the 5th day after engorgement, which was one day prior to oviposition. It may be regarded as a singnal of oviposition. In the synganglion the peak of ecdysteroid level occurred also on the 5th day after engorgement. This is coincident with the secretory activity of neurosecretory cells of synganglion. From the 3rd day after engorgement until oviposition the ecdysteroid level in the ovary increased rapidly. Ecdysteroids were detected in eggs of H. longicornis too. They stem from ovary and accumulated with the process of embryonic development.  相似文献

为阐明蜕皮激素在硬蜱发育和生殖中的作用,用高效液相色谱法(HPLC)和放射免疫测定法测定了银盾革蜱和长角血蜱中饱血和产卵前后雌虫整体、血淋巴、合神经节及卵巢中蜕皮激素含量的变化.此外,还用HPLC测定了长角血蜱的卵和幼虫中蜕皮激素的含量.结果表明:在饱血前雌虫整体中蜕皮激素含量的变化不大,饱血后迅速增加.血淋巴中的蜕皮激素在饱血后第5天(即产卵前1天)达到高峰,可做为产卵的信号.合神经节中蜕皮激素含量的高峰在饱血后第5天,与血淋巴中高峰出现的时间一致,这与神经分泌细胞的分泌活性变化相吻合.从饱血后第3天起,卵巢中蜕皮激素含量增加很快,直到产卵时止,在长角血蜱的卵中也测到α-蜕皮素,来源于卵巢,随胚胎发育进程其含量逐渐增加.卵产出后第4天到第12天,蜕皮激素的含量增加十几倍.  相似文献

【目的】本文旨在明确营养状况不同造成的梨小食心虫Grapholitha molesta(Busck)雌、雄蛹重量差异对其羽化的成虫产卵量、产卵期、寿命及下一代(F1)幼虫发育的影响。【方法】室内条件下,通过不同的饲养方法,获得个体重量不同的梨小食心虫雌、雄蛹,待其羽化交配后,记录其产卵量、产卵时间和成虫寿命;卵孵化前后,分别测量卵和初孵幼虫大小,计算卵孵化率,统计幼虫发育历期。【结果】雌蛹重量对梨小食心虫的成虫产卵量影响显著,其重量与产卵量呈正相关(y=15.505x-59.292);同一条件下,雌蛹与雄蛹重量也呈正相关(y=0.823x-0.538)。同时,雌蛹重量对成虫产卵期影响也较大,蛹重大的个体羽化的雌虫比蛹重小的个体羽化的雌虫产卵高峰期提前1 d;较重、中等和较轻蛹羽化出的雌虫个体每天产卵量高于10粒/雌的时间分别为9~10,7和5~6 d;产卵量高于5粒/雌的时间分别为12~13,9和6~7 d。而雄蛹重量对产卵量、雄成虫寿命影响没有明显影响。较轻的蛹羽化的雌成虫寿命比较重蛹羽化的雌成虫短2~3 d;而雄蛹重量对其羽化的雄成虫寿命影响没有明显规律。雌、雄蛹重量对其羽化成虫的卵孵化率、卵和初孵幼虫的大小影响均不显著,对F_1幼虫发育历期影响也不显著。【结论】梨小食心虫雌蛹重对羽化成虫的产卵量和产卵期等影响显著,田间防治时应注意在不同条件下完成发育的个体,尤其是雌虫,由于营养差异引起的个体大小对随后种群增长的影响。  相似文献

Some di-peptides have been proven to exert an antihypertensive effect in mild-hypertensive subjects. The aim of this study was to clarify whether combined administration of an ACE inhibitor, captopril, with an antihypertensive di-peptide Val-Tyr (VY) would alter their potent antihypertensive effects in spontaneously hypertensive rats (SHRs). Single oral administration of captopril (2.5 mg/kg), VY (25 mg/kg), or captopril (2.5 mg/kg)+VY (25 mg/kg) to 18-week-old male SHRs was performed. Systolic blood pressure (SBP) was measured up to 9 h, and plasma captopril concentrations were determined. A transport study of captopril and/or VY across living rat jejunum from SHRs was also performed to evaluate the kinetics of absorption. Combined administration of captopril with VY failed to lower the BP during the 9-h experiment. A transport study of captopril or VY revealed that VY inhibited captopril transport, and vice versa, in a competitive manner and exhibited an approximately 1/3-fold lower Ki value for captopril compared with that for VY; indicating that both compounds compete for the same membrane transport pathway. A 50% decrease in plasma captopril levels by combined administration with VY supported that the attenuation of the BP lowering effect was due to inhibition of captopril uptake by VY. Consequently, our findings suggest that subjects treated with ACE inhibitors for hypertension should avoid combined-intake with antihypertensive foods that are rich in small peptides due to the competitive inhibition of drug uptake by these peptides.  相似文献

Angiotensin-converting enzyme (ACE) plays a major role in the regulation of blood pressure. A diagnostic assay to measure angiotensin-converting enzyme (ACE) activity was transformed into an enzyme inhibition assay and optimised, which led to a more sensitive and less expensive assay. By this spectrophotometric method, ACE inhibition is measured using the substrate furanacryloyl-Phe-Gly-Gly and as ACE source rabbit lung acetone extract. The optimised as well as the original ACE inhibition assay were used to verify the ACE inhibitory activity of captopril. The ACE inhibition assay was further validated by enalapril, its active derivative enalaprilat and the ACE-inhibitory peptide Ala-Leu-Pro-Met-His-Ile-Arg, corresponding to a tryptic fragment of bovine beta-lactoglobulin. Sigmoid curves could be fit adequately to the data points representing ACE inhibition in function of inhibitor concentration. IC(50) values for these compounds corresponded well with literature data. Furthermore, pea and whey protein hydrolysates obtained by digestion with trypsin showed ACE inhibitory activity in the ACE inhibition assay. Hence, this optimised assay is suitable to screen for ACE inhibitory peptides derived from food proteins with a possible antihypertensive effect in vivo.  相似文献

A strong and constitutive angiotensin converting enzyme- or ACE-like activity was demonstrated in the hemolymph of the adult grey fleshfly Neobellieria bullata. In a competition assay, the N. bullata trypsin modulating oostatic factor (Neb-TMOF) was confirmed to be an in vitro substrate for this circulating Neb-ACE. Oral uptake of captopril, a selective and specific inhibitor of ACE, resulted in a complete phenotypic knockout of circulating ACE activity. When compared with control animals, captopril-fed female flies showed an increase in the liver meal-induced trypsin peak in the midgut and elevated levels of protein meal-induced yolk polypeptides in the hemolymph. The latter effect was not due to a slower vitellogenin uptake by the ovaries, because oocyte growth was not affected by the captopril treatment. The apparent synergism between the demonstrated ACE functionality and the previously reported effects of the oostatic peptide Neb-TMOF are discussed in the context of our recent finding that Neb-TMOF represents a prime candidate for being the first known in vivo substrate for circulating insect ACE. Female adults of the bagworm moth, Eumeta variegata, are completely wingless; by contrast, the male adults have functional wings. Sex-specific differences in the development of wing discs appear to arise during the 8th (penultimate) larval instar. We have previously found that the wing discs of female E. variegata terminate development and disappear during the prepupal period, whereas the wing discs of males continue to develop fully into adult wings. We have investigated the effects of ecdysteroid (20-hydroxyecdysone, 20E) when cultured with larval wing discs, which are normally attached to the larval integument of both male and female larvae. Male wing discs cultured with 20E undergo a remarkable transformation: the discs undergo apolysis and then differentiation. Female wing discs cultured with 20E also undergo apolysis; however, the disc cells enter apoptosis. We have observed condensed chromatin, fragmented nuclei, and secondary lysosomes in the epithelial cells of these female discs. This report establishes that the reduction of female wing discs arises through apoptotic events triggered by ecdysteroid in vitro.  相似文献

While studying the effect of peroral captopril injections on the activity of renin-angiotensin system (RAS), the activity of angiotensin-converting enzyme (ACE) and angiotensin II content from anterior and mediobasal hypothalamus, medulla and adenohypophysis of intact rats has been established to decrease. Captopril administration decreases ACE activity which increases after hydrocortisone injection in rat medulla and striatum. Captopril results in no potentiation of hormonal effect in hypothalamus and in adenohypophysis where ACE activity decreases following hydrocortisone injection. A decrease in the RAS activity of brain structures and adenohypophysis induced by captopril administration to rats is accompanied by the inhibition of the activity in the pituitary-adrenal system. A decrease in ACTH level and in 11-hydroxycorticosteroids of the rat blood plasma has been determined after single captopril injection in the dosage of 10 and 50 mg/kg of body weight. The lizard beetle Doubledaya bucculenta (Coleoptera: Erotylidae: Languriinae) female has evident asymmetric head morphology. The females excavate small holes in host bamboo internodes for the deposition of eggs. To understand the asymmetry and allometry of male and female adults and larvae of D. bucculenta, mandibular length, genal and head width, and elytral length were measured, and the oviposition preference for different-sized internodes of the bamboo Pleioblastus simonii and the relationship between internode size and emerging adult size were examined. Larval mandibles exhibited no clear asymmetry pattern, and genae showed fluctuating asymmetry in length. Adult male mandibles showed left-directional asymmetry, but genae showed fluctuating asymmetry. Adult female mandibles and genae exhibited marked left-directional asymmetry. The degree of asymmetry of mandibles and genae remained constant regardless of body size. Large females tended to choose large-diameter internodes of P. simonii and to lay eggs successfully, whereas small females tended to choose intermediate-diameter internodes, but to fail in oviposition, suggesting that small females pay a high cost on oviposition. There was a positive correlation between internode size and emerging adult size. Marked directional asymmetry of female mandibles and genae are discussed in relation to greater frequency of cutting bamboo fibers compared with adult males, and the traits of bamboo internode.  相似文献

In this research the presence of angiotensin converting enzyme (ACE) in larvae of the lepidopteran Spodoptera littoralis was evaluated. Making use of the substrate Abz-FRK-(Dnp)P-OH and the specific inhibitor captopril at 10 microM, ACE activity was determined in a fluorescence assay for intact larvae, hemolymph, head, midgut and dorsal tissue. In dorsal tissue and hemolymph, ACE activity was highest. These data are consistent with a possible role for ACE in contractions of the dorsal vessel and metabolism of circulating peptide hormones in the hemolymph. After the presence of ACE was confirmed, a sequential procedure of anion exchange and size exclusion chromatography was applied to purify ACE from whole wandering larvae (last stage). With this procedure, three different ACE pools were collected that cleaved the fluorogenic substrate Abz-FRK-(Dnp)P-OH. Activity could be inhibited by a final concentration of 2.5 microM captopril. In addition, two out of three samples eluted at different salt concentration and thus ACE 1, 2 and 3 represent at least two different ACE isoforms. These data reveal that ACE is present in S. littoralis and that at least two out of three isolated ACE forms are truly isoforms.  相似文献

Among various tannins tested, Areca II-5-C, a fraction isolated from seeds of Areca catechu L., showed the most potent angiotensin-converting enzyme (ACE) inhibitory activity in vitro. Its antihypertensive activity was therefore investigated in normotensive and spontaneous hypertensive rats (SHR) after both oral and intravenous (i.v.) administration. The activity was compared with that of captopril (D-3-mercapto-2-methylpropanoyl-L-proline), a potent ACE inhibitor. Oral administration of Areca II-5-C to SHR produced a lasting, dose-related antihypertensive effect, and the responses obtained with doses of 100 and 200 mg/kg were comparable to those of captopril at doses of 30 and 100 mg/kg. Intravenous administration of Areca II-5-C to SHR produced a rapid and marked reduction in blood pressure at doses of 10 and 15 mg/kg. The maximum antihypertensive effect of Areca II-5-C in SHR, at an i.v. dose of 15 mg/kg, was about 5 times as large as that of captopril at the same dose. Although the vasopressor response to norepinephrine and vasodepressor responses to bradykinin and acetylcholine were not appreciably changed by i.v. treatment with Areca II-5-C at a dose of 5 mg/kg, it did produce dose-related inhibition of the pressor responses to angiotensin I and II. It is suggested that Areca II-5-C has favorable properties as a hypotensive drug through its ability to inhibit the pressor responses to both angiotensin I and II.  相似文献

Captopril is an inhibitor of angiotensin converting enzyme (ACE) used against arterial hypertension in mammals. The aim of this study was to evaluate captopril in the mealworm Tenebio molitor (Coleoptera: Tenebrionidae). The drug was diluted in acetone and administered topically (10 microg/3microl) on newly emerged adult females. The biochemical composition (proteins, lipids, carbohydrates, nucleic acids) and the ecdysteroid amounts of ovaries were determined at day 4 following treatment corresponding to the end of the vitellogenesis process. Biochemical analyses revealed that captopril applied in vivo reduced both total protein, lipid and carbohydrate amounts of ovaries, respectively. In addition, measurement of ecdysteroids by an enzyme-immunoassay indicated a significant decrease in ovarian ecdysteroid amounts. As a consequence, the reduction of reproductive capacity is possibly related to ecdysteroid production. Further experiments are needed to determine the action mechanism of captopril and the functions of ACE in insects.  相似文献

Abstract:  Ecdysteroid is one of the two most important insect hormones, which controls growth and developmental activities of insect. In silkworm, Bombyx mori L., basically ecdysteroid induces molting and metamorphosis in larvae and exogenous administration hastens the silk synthetic activity and cocoon spinning process but it strictly depends on the time of administration. In the present study, we administered phytogenous ecdysteroid to the 48-h-old 5th instar silkworm, B. mori at a single dose of 2  μ g per larva to study its effects on the larval growth, cocoon and silk variables. The possible role of ecdysteroid in altering the developmental sequence of silkworm and thereby its effect on cocoon and silk production at molecular level have been tried to get elucidated. The genomic DNA was isolated from the posterior silk gland on day 5 and 7 of the 5th instar from the ecdysteroid treated and the control larvae and was randomly amplified with arbitrary primers. The result presented notable variation in the amplified product suggesting the participation of ecdysteroid in regulating the silk gene. The feeding period of treated larvae was unaffected while the cocoon characters exhibited considerable improvement. The filament traits also were improved notably in the treated larvae. The participation of ecdysteroid in the silk biosynthetic process with its physiological and molecular implications was discussed.  相似文献

The unblocked hexapeptidic Trypsin Modulating Oostatic Factor of the fleshfly, an inhibitor of both trypsin and ecdysone biosynthesis, resists very well proteolytic breakdown by enzymes present in the lumen of the gut of previtellogenic fleshflies. However, when incubated in hemolymph of adult flies, females and males, its half-life time is a mere 0.5 min. In hemolymph of last instar larvae, this value increases to about 1.5 min. Whereas PMSF, a potent inhibitor of serine proteases has no effect, captopril and lisinopril, both known to be specific inhibitors of mammalian angiotensin I converting enzyme (ACE), effectively inhibit TMOF breakdown in fly hemolymph. Digestion of Neb-TMOF by recombinant Drosophila AnCE on itself results in identical degradation products as with total hemolymph. In both cases ESI-Qq-oa-Tof mass spectrometry demonstrated the appearance of peptide fragments with the sequences NPTN, LH and NP. These observations not only confirm the reported presence of circulating ACE-like activity in flies but also strongly suggest that in flies this hemolymph ACE-like activity might be involved in the regulation of the oostatic activity as exerted by Neb-TMOF.  相似文献
