Peroxidase activity and localization in the abscission zone of bean leaves were studied histochemically and by gel electrophoresis. Deblading of bean leaves resulted in an increase in peroxidase activity in the abscission zone 2 to 4 days after deblading with highest activity just prior to separation. In debladed plants, the cell division in six to eight layers of cells preceded separation. An ethylene treatment (8 microliters per liter) induced separation of debladed petioles in approximately 24 hours and of intact plants in 36 to 48 hours. Ethylene treatment produced similar results in both debladed and intact plants. In ethylene-treated plants, whether debladed or not, enzyme localization was restricted to only two to three layers of cells with no cell division apparent prior to separation. Infrequent cell divisions were observed after treatment with 2-chloroethylphosphonic acid (1000 micrograms per liter) (Ethephon); however, other changes were similar to those observed with ethylene. Deblading and ethylene treatment resulted in changes in the six peroxidase isozymes observed in the abscission zone. Only four were observed in samples collected 2 centimeters below the abscission zone. Peroxidase bands IV and V increased significantly in debladed and ethylene-treated plants and peroxidase VI decreased only in debladed plants. The changes in peroxidase activity were invariably observed prior to separation in all treatments.  相似文献

The cytokinin levels of butanol and aqueous extracts of proximalportions of Streptocarpus leaves showed very little change duringthe summer and early autumn months, whereas those of the distalportions changed markedly. These changes appear to be closelyrelated to the formation of abscission layers and the subsequentsenescence of the distal part of the leaves. It is postulatedthat the increase in butanol-soluble cytokinins during Januarymay induce abscission, while the decrease in February appearsto be directly related to senescence of the distal portionsof the leaves.  相似文献

The fine structure of the separation zone cells from the basalabscission zone of bean leaves has been described. Examinationof these cells revealed that fracture occurred primarily asa result of the dissolution of the middle lamellar region ofthe walls, leaving intact cells on the two newly exposed fracturesurfaces. The cytoplasm and organelles within these cells appearedquite normal and showed no signs of autolysis or senescence.A comparison of the organelle numbers in these cells with cellsfrom a similar region of a control plant revealed large increasesin the number of Golgi bodies and the amount of rough endoplasmicreticulum. The significance of this finding is discussed inrelation to the secretion of cell wall hydrolysing enzymes whichare known to be produced in these cells.  相似文献

Role of superoxide dismutase isozymes and other antioxidant enzymes was studied in relation to leaf age in sunflower (Helianthus annuus L. cv. ACC 1508) at pre-flowering and grain filling stages. Relative water content (RWC) did not change much in leaves of different age and at the two stages. Protein content declined continuously from the youngest to the oldest leaf, while chlorophyll (Chl) and carotenoids (Car) contents increased down to 7th/9th leaf and declined in subsequent older leaves. Protein, Chl and Car contents were higher at pre-flowering than at seed filling stage. Superoxide dismutase (SOD), its isozymes, and ascorbate peroxidase (APO) and catalase (CAT) activities were highest in the 9th leaf and declined in subsequent older leaves. SOD and APO activities were higher at seed filling, except in oldest senescent (13th, 15th) leaves. Among SOD isozymes, Cu/Zn-SOD and Mn-SOD activities accounted for most of the total SOD, and only marginal activity was observed for Fe-SOD. Peroxidase activity increased from youngest to the oldest leaf at pre-flowering stage and down to 13th leaf at seed filling stage. The abscission of debladed petioles of various ages in Ervatamiaand Coleus has been followed throughout the year. In general,the retention times for the two species were found to be maximalin the summer and minimal in the winter months. In both speciesolder petioles tended to abscise earlier. The occurrences ofan auxin-inhibitable stage-I and an auxin-promotable stage-IIwere clearly established. The sensitivity to auxin in both stagesdeclined with increase in age, particularly in the two-nodeand three-node twigs of Ervatamia and in the four-node twigsof Coleus. The durations of stage-I and stage-II were foundto be characteristically different in different seasons. Ingeneral, stage-I lasted longer both in younger petioles andduring the summer months. Auxin sensitivity in both stages wasgreater in summer than in winter. Intermediate results wereobtained in the other two seasons. In both the species the auxin-promotablestage-II weakened with time, as shown by a smaller promotionwith delayed auxin application.  相似文献

用SDS — 聚丙烯酰胺凝胶电泳的方法,对比分析了老、嫩黄瓜叶片叶绿素蛋白质复合物之间的差异,发现嫩黄瓜叶片中缺少1条属光系统I的CPIb带。从低温荧光发射光谱观察到,嫩黄瓜的光系统I相对高于光系统Ⅱ,而老黄瓜则相反。指出在叶绿体发育过程中首先形成光系统I,以后是光系统Ⅱ。我们还注意到,叶片中的F685/F735比值与叶绿素蛋白质复合物中的单体/寡聚体比值之间呈正相关关系。  相似文献

Treatment of bean pulvinar explants with auxin significantlydelayed abscission. The pattern of protein synthesis in beanexplants that were treated with and without auxin was investigatedby labelling the pulvinar segments with [35S]-methionine andanalyzing the polypeptides by one and two dimensional gel electrophoresis.One dimensional gel electrophoresis of labelled proteins revealedan increased synthesis of 63, 54 and 29 kDa polypeptides anddecreased synthesis of 60, 49, 30 and 23 kDa polypeptides inthe presence of auxin. Further analysis of proteins on two dimensionalgels revealed several differences in polypeptides between controland auxin-treated explants. 干旱条件下冬小麦叶片光化学效率(Fv/Fm)的降低伴随着叶黄素循环组分玉米黄质含量的增加。干旱初期,Fv/Fm 的降低约在暗置过夜后完全恢复。当干旱诱导的玉米黄质含量的增加达最大值时,Fv/Fm 不可逆下降,Fo 上升,表明发生了光破坏。与老叶相比,干旱条件下功能叶具有较高的玉米黄质含量,对光破坏的抗性较强。推测干旱条件下老叶不可逆衰老与其依赖于叶黄素循环的耗散过剩光能能力的下降有关。  相似文献

Malate dehydrogenase isoenzymes from the blades of different aged leaves of the cotton plant have been investigated. The total extractable malate dehydrogenase activity varied widely between leaves of different ages and different locations on the plant. Malate dehydrogenase zymograms developed from the extracts which contained significantly different levels of enzyme activity appear to indicate the presence of different groups of malate dehydrogenase isoenzymes in leaves of different ages. However, under appropriate conditions of polyacrylamide gel electrophoresis, the same number of malate dehydrogenase isoenzymes with the same relative mobilities were detected in all the leaves studied. These findings are discussed in relation to reports that malate dehydrogenase isoenzymes change with plant development or that they have different roles in the plant.  相似文献

Seasonal Changes in the Cytokinin Content of Ginkgo biloba Leaves   总被引:1,自引:0,他引:1  
Young growth-chamber-grown cotton plants were subjected to a series of eight periods of soil water stress, which served as a preconditioning treatment. After preconditioning, water was withheld and changes in the stomatal resistance and leaf water potential were determined and compared with similar well watered control plants. The stomatal response of stress preconditioned plants adjusted such that the diffusion resistance of the lower surface of the leaf did not reach a value greater than 20 s cm?1 until the leaf water potential dropped 14 bars below that required to reach the same resistance on previously unstressed plants. The resistance—leaf water potential relation for the adaxial surface was unaltered by the preconditioning treatment. Adjustment of the osmotic potential of the guard cells on the abaxial surface provides at least a partial explanation of this change in response. The lack of adjustment of stomatal response on the adaxial surface of the leaves was correlated with a lack of adjustment in osmotic potential of guard cells on that surface.  相似文献

A bscission of debladed petioles of Coleus was observed following spray applications of growth retardant chemicals and particularly of Phosfon D to the foliage. Sprays were applied to some branches, which were left intact (inducing branches), or to adjacent branches the leaves of which were later debladed (induced branches). In all experiments two applications of growth retardant chemicals were made, after which the induced branches were debladed. Treatments on induced branches accelerated the petiole abscission relative to the controls. Treatments on inducing branches, instead, decreased abscission speed of debladed petioles. The evidence suggests that phosfon D affects abscission by interfering with the indoleacetic acid mechanism.  相似文献

研究了无核白葡萄(Vitis vinifera L.)采后贮藏过程中离区纤维素酶、果胶酯酶(Pectinesterase,PE)、多聚半乳糖醛酸酶(polygalacturonase,PG)、脂氧合酶(lipoxygenase,LOX)和过氧化物酶(peroxidase,POD)活性的变化与落粒的关系及植物生长调节物质对其的影响。结果表明,葡萄在贮藏过程中,伴随浆果落粒的增加,离区纤维素酶、PG、LOX、POD活性升高,PE活性下降。离区纤维素酶、PG、LOX等酶的活性与葡萄落粒程度之间呈显著正相关。外源ABA和CEPA处理能增强离区纤维素酶、PG、LOX活性,促进落粒;GA3,IAA处理则能抑制离区纤维素酶、PG、LOX活性,减轻落粒。ABA对落粒的促进效应及GA3对纤维素酶活性和落粒的抑制效应尤为明显,表明GA3与ABA比值在葡萄采后落粒过程中起重要的作用。  相似文献

The young and old leaves of Salix babylonica contain at least four cell division-inducing compounds which coeluted with zeatin, zeatin riboside and their glucosylated derivatives. During the course of the growing season quantitative changes in the cytokinin content of the leaves were observed. The cytokinin glucosides increased as the leaves aged. The compounds which co-chromatographed with zeatin and zeatin riboside initially increased until early autumn and then decreased as the leaves senesced. It appears as though the cytokinins transported from the roots are metabolized in the leaves and are converted to their glucosides. Although it has been reported in the literature that Salix root exudate contains very small amounts of cytokinin in late summer and autumn, these compounds increase in the leaves for most of the growing season, suggesting that the leaves may not only obtain cytokinins from the roots but may well be an active site of cytokinin synthesis. It is, however, possible that cytokinins are also transported to the leaves via the phloem, thus accounting for their accumulation in these organs.  相似文献

Pigs are models in human phoniatry. However, features of maturation and ageing have not been considered with regard to the so-called body-cover model in this species. Therefore, the glottis of “young” (2–3 months; n = 6) and “old” (4–7 years; n = 6) minipigs was investigated. Their cranial (CraF) and caudal (CauF) vocal folds were histomorphometrically and stratigraphically analysed with emphasis on their amounts of collagen structures and elastic fibres. A dense subepithelial layer (SEL) was a distinct feature of CraF and CauF of both age groups; it was spread upon the underlying loose, flexible “cover” like a fibro-elastic membrane. The “cover” was characterised by the so-called superficial layer (SL), which was distinctly loose in the “young” minipigs, but had a much denser texture in the “old” minipigs. Here, the SL was dominated by elastic fibres in the CraF, but was of mixed qualities (collagenous and elastic) in the CauF. The structural requirements for the SL’s function as a loose “cover” were thus met only in the “young” animals. A clearly demarcated intermediate layer (IL)—characterised by high amounts of elastic fibres (as in humans)—was only found in the CraF of the “young” animals. In the “old” animals, it had lost its demarcation. In the depth of the CraF of the “old” animals, many thick collagen fibre bundles were detected in a location equivalent to that of the vocal muscle in the CauF. The development of their large diameters was interpreted as part of the maturation process, thereby supporting the hypothesis of their functional importance as a component of the “body.” In the CauF, the amounts of collagen structures increased throughout the entire lamina propria, resulting in a loss of demarcated stratigraphical subdivisions in the “old” minipigs. This situation resembled that described in the vocal fold of geriatric humans.  相似文献
