An in vitro embryo culture protocol was developed for Scirpus acutus Muhl. A maximum of 85.6% of germination was obtained when isolated embryos were cultured in vitro, a result similar to those reported in the literature with traditional dormancy breaking treatments. In vitro seedling development was optimal in half-strength Murashige and Skoog (1962) liquid medium. An average of 3–4 shoots were produced from the initial seedlings. Clusters of plantlets were successfully acclimatized and transferred to soil. These results corroborate the findings of previous studies stating that seed dormancy in Scirpus is caused by the seed/fruit coats. In vitro embryo culture thus allows for the production of Scirpus acutus Muhl. seedlings that can be transferred to natural or artificial wetlands.  相似文献

Conclusion Anthropological legal concepts of the so-called primitive societies originally induced by prevailing Western philosophical ideas, political thought and jurisprudence require a serious review. The current emphasis by scholars in the anthropology of law on dispute processes in Africa is fulfilling an important need. Given the constantly transforming and unstable nature of modern African political systems, a clearer understanding of native concepts and fundamental value systems is essential. For instance, concepts such as ownership, property rights in land, legal obligation, individual or collective rights must be thoroughly understood within the framework of history and state transformation to be able to help to mediate and settle major inter and intra ethnic disputes in such areas as land tenure, marriage and succession.It is apparent that among the Dagaaba, the obligation to obey the law of the modern state is seriously complicated by the existing native concept of legal obligation which requires the evaluation of laws. To the extent that coercive sanctions are imposed, obedience may be achieved, but the values embodied in the laws may not be internalized. Thus, the laws of the modern state may simply constitute ordar (plural of ordir), that is, coercive commands with no necessary utility or rationality behind them. Such ordar would then not constitute a solid enough basis for dispute resolution when the modern legal system falters in a political crisis.The importance of continuing legal research in fundamental normative systems, concepts and values of tradition society can be seen from our discussion of legal obligation and the mental requirements for culpable conduct among the Dagaaba. These concepts help explain not only some of the institutional structures of the Dagaaba, but also the Dagaaba vision of their own native reality and the legal choices made. Without such studies, one would have only a partial view of African societies. Such research should employ original language techniques such that the full implications of concepts such as collective rights, individuality, and similar group-based legal rights and duties can be properly analyzed. With particular reference to the Dagaaba, further research should address the full implications of the yir, yiiluu and balu in terms of ownership, use of or access to property, and succession to various types of property. It should also examine the effects of customary, statutory and Christian marriage on the group concepts and the implications for succession to property. Since the Dagaaba appear to have been a male dominated society, it will be important in future work to focus attention on women's rights within the framework of these groups. Melanin is the major factor that determines skin color and protects from ultraviolet radiation. In present study we evaluated the anti‐melanogenesis effect of acetazolamide (ACZ) using four different approaches: enzyme kinetic, in vitro, in vivo and in silico. ACZ demonstrated significant inhibitory activity (IC50 7.895 ± 0.24 μm ) against tyrosinase as compared to the standard drug kojic acid (IC50 16.84 ± 0.64 μm ) and kinetic analyses showed that ACZ is a non‐competitive inhibitor without cytotoxic effect. In in vitro experiments, A375 human melanoma cells were treated with 20 or 40 μm of ACZ with or without 50 μm of l ‐DOPA. Western blot results showed that ACZ significantly (< 0.05) decreased the expression level of tyrosinase at 40 μm . Zebrafish embryos were treated with 10, 20 or 40 μm of ACZ and of positive control kojic acid. At 72 h of treatment with ACZ and kojic acid, ACZ significantly (< 0.001) decreased the embryos pigmentation to 40.8% of untreated embryos at the dose of 40 μm of ACZ while kojic acid decreased only 25.0% of pigmentation at the same dose of kojic acid. In silico docking were performed against tyrosinase using PyRx tool. Docking studies suggested that His244, Asn260 and His85 are the major interacting residues in the binding site of the protein. In conclusion, our results suggest that ACZ is a good candidate for the inhibition of melanin and it could be used as a lead for developing the drugs for hyperpigmentary disorders and skin whitening.  相似文献

Somatic embryogenesis was obtained through anther culture of Citrus reticulata, cv. Mandarino Tardivo di Ciaculli. The work was carried out to evaluate the response of somatic embryos inside a sodium alginate coating to different storage periods, and to the effects of the germicide PPM (1 ml l−1) and the fungicide Thiophanate-methyl (100 mg l−1). The effect of these alone or in combination, added to the artificial endosperm on the performance of the encapsulated somatic embryos was tested. After 45 days of culturing, sprouting, root development and conversion were recorded. The encapsulated somatic embryos can be conveniently stored at 4°C, but not for a period longer than 60 days. The presence of Thiophanate-methyl in the artificial endosperm resulted in high levels of sprouting with promising frequencies of root development and conversion in to plantlets under invitro and ex vitro conditions. The concentration of PPM used, alone or in combination with the fungicide, showed some depressing effects in this regard. The prelimnary results obtained with the application of Thiophanate-methyl in a nutrient enriched alginate matrix are promising and warrant further experimentation in attempts to find a solution for the problems faced with ex vitro use of encapsulated somatic embryos.  相似文献

The philosophical or metaphysical architecture of Darwin's theory of evolution by natural selection is analyzed and diflussed. It is argued that natural selection was for Darwin a paradigmatic case of a natural law of change — an exemplar of what Ghiselin (1969) has called selective retention laws. These selective retention laws lie at the basis of Darwin's revolutionary world view. In this essay special attention is paid to the consequences for Darwin's concept of species of his selective retention laws. Although Darwin himself explicity supported a variety of nominalism, implicit in the theory of natural selection is a solution to the dispute between nominalism and realism. It is argued that, although implicit, this view plays a very important role in Darwin's theory of natural selection as the means for the origin of species. It is in the context of these selective retention laws and their philosophical implications that Darwin's method is appraised in the light of recent criticisms, and the conclusion drawn that he successfully treated some philosophical problems by approaching them through natural history. Following this an outline of natural selection theory is presented in which all these philosophical issues are highlighted.  相似文献

Summary Bactris major and Desmoncus orthacanthos are native palms from the Yucatan Peninsula which could be used as substitutes for rattan. When their seeds were germinated in vivo and in vitro they proved to be highly recalcitrant. Therefore, the culture of isolated embryos was studied as an alternative means of producing planting material for nurseries. It was found that the in vitro germination of the isolated embryos was gradually reduced by storage, falling to zero by 5 wk. However, isolated embryos from freshly collected seeds germinated at ∼100% frequency. The presence of the endosperm, whether still attached to the embryos or separated from them but in direct contact with the nutrient medium, greatly reduced germination in both species. High concentrations of abscisic acid (ABA, 100 μM) only slightly diminished it, suggesting a different cause for the observed endosperm-induced inhibition. This embryo rescue method permits the production of sufficient plants for in vitro micropropagation and the establishment of experimental plots to evaluate the full potential of these materials.  相似文献

There is the general philosophical question concerning the relationship between physics, which is often taken to be our fundamental and all-encompassing science, on one hand and the special sciences, such as biology and psychology, each of which deals with phenomena in some specially restricted domain, on the other. This paper deals with a narrower question: Are there laws in the special sciences, laws like those we find, or expect to find, in basic physics? Three arguments that are intended to show that there are no such laws are presented and examined. The paper ends with brief remarks concerning the implications of these arguments for explanation and causation in the special sciences.  相似文献

Developmental biologists distinguish between mosaic embryos, in which the removal of a cell or group of cells results in a defective adult, andregulative embryos, in which the adult appears normal in spite of such removal. I suggest that the mosaic/regulative distinction is best viewed by contrastingwithin-cell signals(i.e., a cell can develop autonomously, perhaps on the basis of instructions derived from the mother) againstbetween-cell signals (i.e., development, and the origin of form and shape, is based on intercellular communication). This distinction is not rigid; the same embryo can make use of both within-cell and between-cell signals. During evolution, signalling between cells is likely to have become advantageous as organisms increased in size. However, the fact that an embryo displays regulative behaviour may be an automatic consequence of the way it develops rather than an evolved adaptation.  相似文献

Polyploids are pervasive in plants and have large impacts on crop breeding, but natural polyploids are rare in animals. Mouse diploid embryos can be induced to become tetraploid by blastomere fusion at the 2-cell stage and tetraploid embryos can develop to the blastocyst stage in vitro. However, there is little information regarding mouse octaploid embryonic development and precise mechanisms contributing to octaploid embryonic developmental limitations are unknown. To investigate the genetic and epigenetic mechanisms underlying octaploid embryonic development, we generated mouse octaploid embryos and evaluated the in vitro/in vivo developmental potential. Here we show that octaploid embryos can develop to the blastocyst stage in vitro, but all fetus impaired immediately after implantation. Our results indicate that cell lineage specification of octaploid embryo was disorganized. Furthermore, these octaploid embryos showed increased apoptosis as well as alterations in epigenetic modifications when compared with diploid embryos. Thus, our cumulative data provide cues for why mouse octaploid embryonic development is limited and its failed postimplantation development.  相似文献

Embryonic stem cells (ESCs) represent a useful tool for cell therapy studies, however the use of embryos for their derivation give rise to ethical, religious and legal problems when applied to the human. During the last years parthenogenesis has been proposed as an alternative source to obtain ESCs. Based on the fact that parthenotes avoid many concerns surrounding the “ad hoc” in vitro production and following destruction of viable human embryos. Unfortunately many aspects related to parthenogenetic cell biology are not fully understood and still need to be elucidated. In this review we describe advantages and limits of these cells. We discuss their typical ESC morphology and high telomerase activity, which disappears after differentiation. We examine the pluripotency signature that they share with bi-parental ESCs. We review their high differentiation plasticity that allow for the derivation of several mature cell type populations when we expose these cells to adequate conditions. On the other hand, in-depth analysis demonstrated chromosome mal-segregation and altered mechanisms controlling centriole arrangement and mitotic spindle formation in these cells. We hypothesize their monoparental origin as one of the possible cause of these anomalies and suggest a great caution if a therapeutic use is considered.  相似文献

Synopsis We tested the reactions of free embryos of the amphidromous goby, Rhinogobius brunneus, to light under both artificial and ambient conditions along streams in which their downstream migration occurs. The embryos showed a positive phototaxis to 500 1ux light but a negative response to light of more than 5000 lux. They were able to swim at 1.54 cm sec–1 t in still water and showed positive rheotaxis, but their swimming ability was not sufficient to allow active movement in rapids. Ambient natural light conditions varied among locations in relation to local topographical features. The variation in the diel periodicity of their migration could be explained by the interaction between behavioral reactions of embryos and environmental factors along river courses.  相似文献

Linnaeus's artificial and natural arrangements of plants are examined using a Spearman rank coefficient (which is explained) on his presentations of his own and others' arrangements in the Classes plantarum and elsewhere. There is little alteration in his successive artificial arrangements. In contrast, between 1751 and 1764 his natural arrangements changed considerably, partly in the sequences of genera within orders but mostly by rearrangement of the orders. Comparison with Cesalpino's and Ray's natural arrangements, using the longest-recognized natural groups as signposts, suggests that Linnaeus in his latest natural arrangement (1764) approximated more closely to Ray's. Examination of Linnaeus's successive treatments of certain groups (palms, Zingiberaceae, Hydrocharis-Stratiotes-Vallisneria) and of Giseke's exposition of Linnaeus's lectures on natural groups (1792) shows that Linnaeus was much influenced by habitus and vegetative characters as well as those of the fructification. He recognized orders consisting of a chain of genera linked successively by overall affinity and without any single diagnostic character. Where possible, he preferred characters of the fructification and his ‘secret’ consulting of the habitus is explained as secondary to such characters. It is suggested that in his latest arrangement he approximated more to a scala naturae, as he probably did in zoology about the same time. Within his artificial arrangements Linnaeus kept to sequences of genera as natural as possible. He realized that some groups in his natural arrangements were still artificial, and his aphorism that all genera and species are natural, classes and orders part natural and part artificial, refers to his and others' practice until the natural system could be completed. It is not a statement of the essential natures of these ranks. Linnaeus's distinction in practice between natural and artificial arrangements was less clear-cut than Sachs believed. Linnaeus's rejection of the ancient tree/herb division was empirical, not a reasoned repudiation of an a priori grouping. The tree/herb division could be upheld in his day as obviously natural, not merely accepted on authority.  相似文献

Background  

The cytochrome P450 aromatase (CYP19), catalyses the aromatisation of androgens to estrogens, a key mechanism in vertebrate reproductive physiology. A current evolutionary hypothesis suggests that CYP19 gene arose at the origin of vertebrates, given that it has not been found outside this clade. The human CYP19 gene is located in one of the proposed MHC-paralogon regions (HSA15q). At present it is unclear whether this genomic location is ancestral (which would suggest an invertebrate origin for CYP19) or derived (genomic location with no evolutionary meaning). The distinction between these possibilities should help to clarify the timing of the CYP19 emergence and which taxa should be investigated.  相似文献

为探明华重楼离体胚培养及植株再生的基本体系,该文以华重楼离体幼胚为试验材料,以MS培养基为基本培养基,研究不同光照、不同浓度梯度组合的植物生长调节剂对华重楼离体幼胚萌发、成苗的影响。结果表明:培养到60 d时,暗培养条件下华重楼离体幼胚的生长率和萌发率分别比光培养条件下高45.25%、19.17%,故暗培养比光培养更有利于华重楼离体幼胚生长发育。当GA3浓度相同时,离体幼胚萌发所需时间随IAA浓度增加而延长。不同浓度的GA3都可促进离体幼胚萌发,促进作用由强到弱依次为5 mg·L-1 GA3>1 mg·L-1 GA3> 10 mg·L-1 GA3。在黑暗条件下,优选得到最适华重楼离体幼胚生长发育配方为1/2 MS+30 g·L-1 蔗糖+7 g·L-1琼脂+0.5 g·L-1 活性炭+5 mg·L-1GA3+1 mg·L-1 IAA。此配方可诱导华重楼离体幼胚在2个月左右萌发,萌发率达50%需80 d左右。在华重楼成熟胚出苗培养时发现,高浓度的植物生长调节剂会抑制华重楼成熟胚生长发育,高浓度GA3甚至会导致华重楼成熟胚死亡。优选得到最适华重楼成熟胚出苗配方为MS+30 g·L-1 蔗糖+6.2 mg·L-1 硼酸+7 g·L-1 琼脂+0.7 g·L-1 活性炭+0.5 mg·L-1 2,4-D+1.5 mg·L-1 IAA + 1.5 mg·L-1 ZT+5mg·L-1 GA3,诱导成熟胚出苗需43 d左右,75 d左右可形成真叶。  相似文献

Natural selection can act against maladaptive hybridization between co‐occurring divergent populations leading to evolution of reproductive isolation among them. A critical unanswered question about this process that provides a basis for the theory of speciation by reinforcement, is whether natural selection can cause hybridization rates to evolve to zero. Here, we investigated this issue in two sibling mosquitoes species, Aedes mariae and Aedes zammitii, that show postmating reproductive isolation (F1 males sterile) and partial premating isolation (different height of mating swarms) that could be reinforced by natural selection against hybridization. In 1986, we created an artificial sympatric area between the two species and sampled about 20,000 individuals over the following 25 years. Between 1986 and 2011, the composition of mating swarms and the hybridization rate between the two species were investigated across time in the sympatric area. Our results showed that A. mariae and A. zammitii have not completed reproductive isolation since their first contact in the artificial sympatric area. We have discussed the relative role of factors such as time of contact, gene flow, strength of natural selection, and biological mechanisms causing prezygotic isolation to explain the observed results.  相似文献

In 2005, a 7‐ha artificial watershed (Chicken Creek) was built on a post mined landscape in Lusatia, Germany from sandy substrates of Pleistocene origin, commonly used in reclamation. The watershed was developed to investigate the initial phase of soil and ecosystem development under natural conditions. At this early stage, mineral nitrogen in young sandy soils is primarily limited and nitrogen fixing legumes become key components of natural succession. Local abundant pioneering legumes Lotus corniculatus and Trifolium arvense and one pioneer grass species Calamagrostis epigeios were investigated 5 years after watershed construction. In this study, we investigated the influence of spatial root and nodule distribution of these species on soil nitrogen accumulation. Soil, including roots, was sampled from field monoliths covered with the aforementioned plant species. Root systems of both legumes were mainly restricted to the upper 20 cm of soil, whereas roots of C. epigeios also developed strongly at greater depths. A positive relationship was found, with higher plant densities associated with higher root densities which were associated with higher nodule densities for legumes and which were all associated with significantly higher soil nitrogen content relative to non‐vegetated areas. This research provides rare information on the role root systems of pioneer legumes play in soil nitrogen input in the early stage of soil and ecosystem development during revegetation by natural succession.  相似文献

Naturally spawned egg strands of perch, Perca fluviatilis L. were sampled from exposed fir trees submerged in Lake Plußsee, Holstein, Northern Germany from April to May 1989. These were incubated in situ and in the laboratory to estimate the viability, development and abnormalities of the embryos. From the in siru incubated eggs 91.6% embryos were viable, the nonviable portion comprising 1.1 % unfertilized eggs, 5.1 % dead and 2.2 % with abnormalities. As compared to the eggs incubated in the laboratory with 84.1 % viable embryos, the nonviable portion consisting 1.1 % unfertilized eggs, 11.3 % dead and 3.5 % with abnormalities. Incubation rate of P. fluviatilis embryos both in situ and laboratory, ranged from 200-210 ATUs (10°C in situ and 8°C in the laboratory) with incubation period of 20-27 days from fertilization to 50 % hatching.  相似文献

Summary Biological nitrogen fixation is the most important process in which some prokaryotic organisms fix N2 into ammonium. From an agricultural standpoint, biological nitrogen fixation (BNF) is critical because industrial production of nitrogen fertilizers seldom meets agricultural demands. To increase the BNF is one of the main challenges for the future. There are different possibilities for extending biological nitrogen fixation to the economically important plants. One of the possibilities is to create new artificial systems between diazotrophic bacteria and different higher plants. This is the main topic of the present review article which discusses the establishment of new associative and/or symbiotic systems, via introduction of diazotrophic bacteria into the roots by different methods; and incorporation of nitrogen-fixing bacteria in the entire plant by in vitro methods, through the establishment of intracellular endosymbioses via induced uptake of bacteria by plant protoplasts (endocytobiosis), and establishment of intercellular associations by forced introduction of bacteria into the plant tissues (exocytobiosis). The common characteristic of the methods to create artificial plant-microbe systems for atmospheric nitrogen fixation is the use of in vitro plant systems: cells, tissues and organ cultures. The review pays particular attention to new bacterial inoculation procedures for introduction of the diazotrophic bacteria inside the plant tissues.  相似文献
