Two hundred nonsuckling beef cows were treated with either 1) a progesterone-releasing intravaginal device (PRID) for 12 days; 2) PRID plus an IM injection of 200 mg progesterone (PRID-P); 3) PRID plus 5-mg IM injection of estradiol valerate (PRID-EV); or 4) PRID-EV-P. Cows were started on treatment on one of the first eight days of the estrous cycle. The number of cows which had P levels above 1 ng/ml one day after PRID removal was 12 to 50% lower in PRID-EV and PRID-EV-P groups than in PRID and PRID-P groups (P < 0.05). The proportion of cows showing estrus by 96 hours after PRID removal was 38, 36, 77, and 88% (P < 0.05) for the PRID, PRID-P, PRID-EV and PRID-EV-P groups, respectively. Thirty-one percent fewer cows treated with PRID on days 5 through 8 of the estrous cycle showed estrus by four days after PRID removal than those treated on days 1 through 4. In addition, 18 to 22% more cows had P levels above 1 ng/ml among cows treated with PRID or PRID-P on days 5 through 8 than among cows treated similarly on days 1 through 4. It was concluded that effective synchronization of estrus is achieved only when estrogen is used in conjunction with PRID in cows treated for twelve days during the first eight days of an estrous cycle.  相似文献

A major cause for reproductive failures in Zebu x Taurine crossbred cows is postpartum anestrus. Crossbred cows were diagnosed to be in postpartum anestrus by palpation per rectum of nonfunctional ovaries. To induce synchronized estrus, the cows were treated by placement of a progesterone releasing intravaginal device (PRID) for various number of days, together with a single administration of prostaglandin F(2) alpha (PGF, 5 mg intravulvosubmucos), estradiol-17beta (E2, 1 mg i.m.) or pregnant mare serum gonadotrophin (PMSG, 500 IU i.m.). Administration of PRID for 7 d, and of E2 and PMSG on Day 6, significantly improved the degree of behavioral estrus manifestation compared to single PRID for 12 d or PRID for 7 d, and of PGF on Day 6. However, the treatment combination of PRID and PMSG alone was associated with higher (P < 0.01) conception rate to 2 fixed time A.I. at induced estrus. The mean interval from treatment to conception was also shortest (P < 0.01) for this group. These results suggest that administration of PRID for 7 d, and of PMSG on Day 6 is highly effective in achieving synchronized behavioural estrus, a near normal CR to fixed time A.I. and a shorter interval from treatment to conception in anestrus Zebu x Taurine crossbred cows under Indian field conditions.  相似文献

Experiments tested whether supplemental antioxidants improved fertility. To test effects of beta-carotene, cows in a hot environment were injected with prostaglandin F2 alpha (PGF2 alpha) and were given 3 injections, i.m., of 800 mg beta-carotene or saline at Days -6 and -3 before the anticipated date of insemination and at insemination (n = 37-41 inseminated cows/group). There was no effect of beta-carotene on the proportion of cows detected in estrus following PGF2 alpha, timing of estrus after PGF2 alpha injection or pregnancy rate in inseminated cows. In a second trial, cows in a temperate climate received intramuscular injections of vitamin E (500 mg) and selenium (50 mg) at 30 d post partum (n = 97) or were untreated controls (n = 89). Treatment did not affect interval from calving to first insemination or the proportion of cows pregnant at first service, but it increased the pregnancy rate at second service (69.8 vs 52.1%; P = 0.07) and reduced services per conception (1.7 vs 2.0; P < 0.05) and interval from calving to conception (84.6 vs 98.1; P < 0.05). Thus, injection of vitamin E and selenium increased fertility in cattle that did not become pregnant at first service.  相似文献

Serum progesterone (P) profiles, following progesterone releasing intravaginal device (PRID) treatment (Experiment I) and use of the PRID for induction of fertile oestrus (Experiments II, III and IV), were studied in five non-cycling, buffalo heifers and 86 post-partum buffalo cows, respectively. The maximum P concentration (2.5 ng/ml) was attained 24 h after insertion of the device. Thereafter, P levels gradually decreased to 1 ng/ml at the time of removal of the device on day 12. In post-partum cows, the retention rate of the PRID was 100%, and more than 80% of the animals exhibited signs of oestrus after PRID removal. However, the conception rate following natural mating (26%) and fixed-time insemination (33%) at 60 and 84 h after PRID removal, was very low. Serum P levels and rectal palpation for active corpora lutea following PRID removal indicated a relatively high incidence of anovulatory oestrus post PRID treatment.  相似文献

A retrospective study was conducted to evaluate the outcome of 223 prostaglandin F(2)alpha (PGF) treatments given to lactating Holstein and Jersey cows over a 44-mo period. The cows were part of a commercial dairy herd. Treated cows were given PGF (25 mg i.m.) because they had not been observed in estrus or because they had been diagnosed as having a luteal cyst or pyometra. Outcome of treatments was assessed for frequency of response to treatment (the detection of estrus), time interval to detection and pregnancy outcome of breedings at PGF induced estrus. Of the 223 administrations of PGF, 70% (156 223 ) were given for unobserved estrus, 12.1% (27 223 ) were given for cystic ovarian disease and 15.2% (34 223 ) were given for pyometra. Estrus was detected in 55.3% (120 217 ) of cows over a period of 36 to 130 hours after PGF administration. Interval to estrus and response rates were not influenced by the reason for treatment. Mean time interval to response was 79.1 h. Time interval for response was affected by season (P < 0.01) housing location (P < 0.01) and lactation number (P < 0.05). Insemination was performed following 157 of the treatments. For cows treated for unobserved estrus, insemination was performed in response to estrus or at 80 h if no estrus had been previously seen. Of this group, 39.7% (58 146 ) failed to show estrus and were inseminated at 80 h. Reinsemination was required for 22.7% (20 88 ) of the cows that manifested estrus following insemination at 80 h. Overall conception rate was 41.4% (65 157 ). Cows inseminated in response to estrus had a 53.4% (47 88 ) conception rate, and appointment bred (80 h) cows had a 20.7% (12 58 ) conception rate. Factors significantly influencing conception were response to PGF administration (P < 0.05), season (P < 0.01) and lactation number (P < 0.01). It was concluded that estrus detection efficiency affects the outcome of PGF administration and that conception rates resulting from 80 h breeding when no estrus is observed may be unacceptably low.  相似文献

We studied the effect of recombinant bovine somatotropin (rbST) at the time of estrus on progesterone concentrations and conception rates of repeat-breeding Holstein cows. We used repeat-breeding cows of varied parity (n = 510). All the animals were clinically healthy and had had at least three unsuccessful services before entering the study. After detection of estrus, the cows were randomly assigned to either a treated (n = 201) or a control (n = 309) group. The animals in the treated group were given rbST (500 mg s.c.) at the time of estrus and again 10 d later. Artificial insemination was performed 12 h after the first detection of estrus. In order to evaluate the effect of rbST on luteal function, blood samples were taken from 10 cows in each group every 3 d for 18 d, starting on the day of insemination (Day 0) to determine progesterone concentrations. Conception rates were significantly higher (P < 0.05) in the cows treated with rbST (29.3%) than in the control cows (16.9%). The effects of rbST were maximal in cows with 8 or more previous unsuccessful services and in cows with 2 to 4 calvings. Progesterone concentrations tended to be higher in nonpregnant cows that were treated with rbST than in those that were not treated. The difference between groups was significant (p < 0.05) on Day 18 after insemination. In pregnant cows there were no significant differences in progesterone concentrations between treated and nontreated animals at any time. Treatment with rbST at estrus improved the conception rate of repeat-breeding Holstein cows. This effect was associated with an increase in circulating progesterone concentrations on Day 18.  相似文献

The aim in this study was to compare two estrus synchronization protocols in buffaloes. Animals were divided into two groups: Group A (n=111) received 100 microg GnRH on Day 0, 375 microg PGF(2alpha) on Day 7 and 100 microg GnRH on Day 9 (Ovsynch); Group B (n=117) received an intravaginal drug release device (PRID) containing 1.55 g progesterone and a capsule with 10mg estradiol benzoate for 10 days and were treated with a luteolytic dose of PGF(2alpha) and 1000 IU PMSG at the time of PRID withdrawal. Animals were inseminated twice 18 and 42 h after the second injection of GnRH (Group A) and 60 and 84 h after PGF(2alpha) and PMSG injections (Group B). Progesterone (P(4)) concentrations in milk samples collected 12 and 2 days before treatments were used to determine cyclic and non-cyclic buffaloes, and milk P(4) concentrations 10 days after Artificial insemination (AI) were used as an index of a functional corpus luteum. Cows were palpated per rectum at 40 and 90 days after AI to determine pregnancies. All previously non-cyclic animals in Group B had elevated P(4) (>120 pg/ml milk whey) on Day 10 after AI. Accordingly, a greater (P<0.01) relative percentage of animals with elevated P(4) 10 days after AI were observed in Group B (93.2%) than in Group A (81.1%). However, there was no difference in overall pregnancy rates between the two estrus synchronization protocols (Group A, 36.0%; Group B 28.2%). When only animals with elevated P(4) on Day 10 after AI were considered, pregnancy rate was higher (P<0.05) for animals in Group A (44.4%) than Group B (30.3%). The findings indicated that treatment with PRID can induce ovulation in non-cyclic buffalo cows. However, synchronization of estrus with Ovsynch resulted in a higher pregnancy rate compared with synchronization with PRID, particularly in cyclic buffalo.  相似文献

Ninety-two Friesian cows were used to determine the chemical properties of cervical mucus during normal estrus and estrus induced by progesterone (P4)-releasing intravaginal devices (PRID) and/or prostaglandin F2alpha. The animals were assigned to 4 groups (no treatment, a PRID for 12 days plus injection of 1000 IU PMSG at the removal of PRID, a double i.m. injection of PGF2alpha 11 days apart, or PRID for 7 days plus an im injection of PGF2alpha 24 h before the removal of PRID). A number of cows with normal estrus exhibited three consecutive estrous cycles after delivery. Cows that had not shown estrus for 3 months after delivery had their ovaries palpated twice at 10-day intervals, to determine their ovarian activity. Then PRID and/or PGF2alpha was administered in cows that had a palpable corpus luteum in one of the two palpations (cyclic cows). A double artificial insemination (AI) was performed to the cows of the three induced-estrus groups, while the cows with normal estrus received only one AI. Cervical mucus samples were collected from all cows 5 to 30 min before the first AI. Additionally, samples of cervical mucus were collected from 20 cows during their first estrus after the induced one. The results are summarized as follows: 1) The biochemical properties of cervical mucus in the first three estrus periods after delivery were similar. 2) These properties were similar both in normal estrus after delivery and in the first estrus after an induced one. 3) Glucose and fructose concentrations for normal estrus were similar to those for induced estrus groups. 4) Total protein and cholesterol concentrations were significantly lower (P < 0.001) in normal than in induced estrus, while no difference was found among the induced estrus groups. 5) Pregnancy rates of the cows did not differ significantly among the normal and the induced-estrus groups. 6) The percentages of cows in the induced-estrus groups that produced cervical mucus with total protein and cholesterol concentrations similar to those for the normal estrus groups was very low.  相似文献

In Experiment 1, cows with a history of at least two previous unsuccessful inseminations were allocated to four groups. At the repeated estrus (third or greater service), some of the cows were inseminated according to the a.m.-p.m. rule (Controls, n = 83), or received i.m. 100 mug gonadotropin-releasing hormone (GnRH, n = 32) within 30 sec after insemination. Ovaries of the remaining cows to be treated were palpated during the anticipated ensuing luteal phase to determine the presence of a corpus luteum. Cows found to have luteal tissue received i.m. 25 mg prostaglandin F(2)-alpha (PGF(2)-alpha) and were inseminated after detected estrus or at 72 and 96 h after PGF(2)-alpha in the absence of estrus. Cows given PGF(2)-alpha either received no further treatment (PGF(2)-alpha, n = 40) or were given i.m. 100 mug GnRH (PGF(2)-alpha + GnRH, n = 29) after insemination or at 72 h after PGF(2)-alpha in the absence of estrus. Conception rate of control cows (39%) was similar to that of cows given PGF(2)-alpha (40%) or PGF(2)-alpha + GnRH (43%), but it tended to be lower (P = 0.13) than that of cows given only GnRH at insemination (54%). In Experiment 2, cows with unobserved estrus and diagnosed not pregnant (palpation) were palpated to detect a corpus luteum. Cows with luteal tissue received i.m. PGF(2)-alpha (n = 52) or PGF(2)-alpha + GnRH (n = 45) and were inseminated as described above. Conception rates were similar (39% vs 33%, respectively). In Experiment 3, cows in a large commercial dairy with (n = 93) or without (n = 420) previous reproductive problems were given i.m. 100 mug GnRH after insemination (n = 169) or were left untreated (n = 344) at repeat services (third and fourth services). Treatment with GnRH improved (P < 0.05) conception in normal (47% vs 36%) and abnormal (25% vs 12%) repeat-breeding cows. Treatment with PGF(2)-alpha alone or in conjunction with GnRH failed to further improve conception rates and only delayed intervals to rebreeding when administered during the luteal phase after the repeated estrus. The use of GnRH failed to reduce intervals from treatment to insemination or improve conception in cows with unobserved estrus compared to treatment with PGF(2)-alpha alone.  相似文献

Cows that are diagnosed as anestrus (defined as >3 weeks postpartum, not detected in estrus by 1 week before commencement of seasonal breeding and without a palpable corpus luteum) have a lower probability of being inseminated, a lower conception rate to insemination and hence a lower probability of pregnancy within the breeding period. A current progesterone (P) and estradiol benzoate (EB) treatment for anestrus results in an 85% insemination rate, but only 57% of cows not conceiving to that first insemination are subsequently detected in estrus 14-28 days later. Resynchrony after an initial anestrous treatment has been used to increase the probability that cows not conceiving to first insemination will subsequently be detected in estrus. Anestrous cows (n=971) were initially treated with an intravaginal P-releasing device for 6 days and given 1 mg EB 1 day after device removal (day of EB treatment=Day 0). Cows detected in estrus between Days 0 and 3 were assigned randomly to be treated with reinsertion of a used P-releasing device for 8 days (commencing on Day 15), with 0.5 mg EB at reinsertion and again 1 day after removal (EB-RS), treatment as for EB-RS group but with substitution of 250 microg GnRH for EB at device reinsertion (GnRH-RS), or left as untreated controls (no-RS). Resynchrony treatments increased the proportion of non-pregnant cows detected in estrus from Days 14 to 28 compared to no-RS (79.1, 69.8, and 55.1% for EB-RS, GnRH-RS, and no-RS, respectively; P<0.05). Fewer cows were pregnant by Days 28 and 56 following GnRH-RS than EB-RS and no-RS. The final pregnancy rate was higher following EB-RS treatment than no-RS or GnRH-RS treatment (95.0, 88.3, and 88.6%; P<0.05). In conclusion, EB-RS enhanced reproductive performance of anestrous cows compared to no-RS.  相似文献

Human chorionic gonadotrophin (hCG) plus PGF2 alpha was compared with GnRH plus PGF2 alpha for estrus synchronization of dairy cows. There were 3 treatments: GnRH analog (Buserelin, 12.6 micrograms) plus PGF2 alpha analog (Cloprostenol, 150 micrograms) 6 d later (GnRH + PGF[Day 6]); hCG (2000 IU) plus PGF2 alpha 9 d later (hCG + PGF[Day 9]); and hCG plus PGF2 alpha 6 d later (hCG + PGF[Day 6]). Treatment occurred either Days 55 to 90 or Days 91 to 135 post partum. For responses during the first 10 d after PGF2 alpha administration, estrus synchronization (P = 0.24), efficacy (percentage of treated pregnant; P = 0.20) and conception (percentage of inseminated pregnant; P = 0.23) rates were not different among the 3 treatments. Cows treated between Days 55 and 90 had a higher rate (P < 0.05) of detected estrus during this period (69% for GnRH + PG [Day 6], 70% for hCG + PGF[Day 9] and 72% for hCG + PGF[Day 6]) compared with cows treated between Days 91 and 135 (52% for GnRH + PGF[Day 6], 50% for hCG + PGF[Day 9] and 57% for hCG + PGF[Day 6]). Efficacy of treatment was higher (P < 0.05) in animals treated between Days 55 and 90 (54% for GnRH + PGF[Day 6], 56% for hCG + PGF[Day 9] and 63% for hCG + PGF [Day 6]) compared to animals treated between Days 91 and 135 (36% for GnRH + PGF[Day 6], 35% for hCG + PGF[Day 9] and 47% for hCG + PGF[Day 6]). There were no significant differences in conception between Days 51 and 90 and Days 91 and 135. The interval between parturition-first AI with conception was significantly (P < 0.001) shorter in GnRH + PGF (Day 6; 106 d), hCG + PGF (Day 9; 109 d) and hCG + PGF (Day 6; 103 d) treated cattle than in 106 untreated animals (136 d). Thus, GnRH plus PGF2 alpha or hCG plus PGF2 alpha treatments elicited similar effects in estrus synchronization, treatment efficacy, and conception rate in post-partum dairy cows.  相似文献

Ninety-five cows (79 Boran and 16 Boran-Brahman crossbreeds) and 107 heifers (55 Boran and 52 Boran x Friesian F1 crossbreeds) were used to determine estrus response, estrus response interval and pregnancy rate following synchronization with prostaglandin (PGF(2)alpha), a progesterone-releasing intravaginal device (PRID) and Synchro-mate B (SMB). The proportion of cattle responding to synchronization treatment was 62.5, 43.5 and 57.7% for cows and 85.7, 68.0 and 81.5% for heifers using PGF(2)alpha, PRID and SMB, respectively. The overall mean response was 59 and 81.8% for cows and heifers, respectively. The estrus response of the control animals over a 45-d breeding period was 72.7 and 90% for cows and heifers, respectively. The estrus response interval for cows was 31.8, 22.1 and 18.0 h and it was 51.1, 38.0 and 21.6 h for heifers with PGF(2)alpha, PRID and SMB treatment, respectively. Mean pregnancy rate for cows was 50.0, 34.8, 46.2 and 68.8% and for heifers it was 60.7, 40.0, 55.6 and 77.8% in the PGF(2)alpha, PRID, SMB and control groups, respectively. Based on these findings, it was concluded that both PGF(2)alpha and SMB produce a satisfactory estrus response and pregnancy rate in the cattle studied.  相似文献

The objective was to compare the probability of pregnancy after fixed-time insemination in cows diagnosed as non-pregnant and re-inseminated following the Ovsynch protocol, with or without exogenous progesterone. Cows (n=415) used in this study originated from 25 farms. Upon diagnosis of non-pregnancy between 30 and 60 days after AI, cows were randomly assigned to receive either a progesterone releasing intravaginal device (PRID; n=208) or a placebo intravaginal device (PID; n=207). All cows received GnRH at enrollment (Day 0), PGF(2alpha) concurrent with intravaginal device removal 7 days later, GnRH on Day 9 and fixed-time insemination 16h later (Day 10). Cows observed in estrus prior to Day 7, had the device removed and were inseminated. Ovaries were examined by transrectal palpation at the time of enrollment and the prominent structures were assessed and recorded. Body condition score, lameness status, interval from previous insemination, and times bred at enrollment were recorded. At intravaginal device removal, the occurrence and intensity of vaginitis was determined according to the amount of debris on the device. Overall, the intravaginal device retention rate was 91%. A total of 5.2% of PID-treated cows and 2.9% of PRID-treated cows were detected in estrus within the 7 days treatment period. Pregnancy status was diagnosed between 30 and 56 days after insemination and all cows were followed for a minimum of 150 days after enrollment. Approximately 28% of cows had evidence of mild vaginitis in response to the intravaginal device, whereas 6% of cows had copious debris associated with the intravaginal device at removal. The probability of pregnancy after fixed-time insemination was 43.8% versus 34.9% in PRID-treated versus PID-treated animals. Exogenous progesterone provided through an intravaginal device to non-pregnant cows that had not displayed estrus improved the probability of pregnancy after fixed-time AI.  相似文献

Using two PGF treatments 14 days apart as a way to enhance estrus detection rate following the 2nd treatment is a reproductive management tool that continues to be used on large dairy farms. In one study, in cows with a functional CL and a dominant follicle, treatment with cloprostenol vs. dinoprost resulted in greater peripheral estradiol concentrations. The objective of the present study was to determine if cloprostenol could enhance pregnancy rates of cows in a large dairy herd using a PGF program for 1st artificial insemination (AI). Lactating dairy cows (n = 4549) were randomly assigned to receive two treatments of either 500 μg cloprostenol or 25 mg dinoprost 14 days apart, with the 2nd treatment on the 1st day of the voluntary waiting period (57 DIM). Cows detected in estrus within 5 days after the 2nd treatment were inseminated. There was no effect of treatment on day of estrus detection, with 78% of cows inseminated on Days 3 or 4 following treatment. Cloprostenol increased (P < 0.01) estrus detection rates in 1st parity cows compared to dinoprost, 42.4 vs. 34.0%. In cows inseminated on Days 3 or 4 after treatment, cloprostenol increased (P = 0.05) conception rates compared to dinoprost, 38.3 vs. 34.4%. When treatments and parities were combined, conception rates increased (P < 0.02) with interval after treatment (27.0, 36.4, and 44.5% for Days 1 or 2, Days 3 or 4, and Day 5, respectively). Cloprostenol increased (P = 0.02) overall pregnancy rate compared to dinoprost, 14.4 vs. 12.2%. In summary, cloprostenol increased fertility in 1st parity cows inseminated on Days 3 or 4 following treatment and subsequently enhanced pregnancy rates of 1st parity lactating dairy cows compared to dinoprost. Fertility appeared greater in cows expected to have had a young antral ovarian follicle at treatment.  相似文献

We wished to compare the effect of summer heat stress on pregnancy rate in cows that were inseminated at a set interval associated with a synchronized ovulation vs those inseminated upon routine estrus detection. The study was carried out on a commercial dairy farm in Florida from May to September 1995. Lactating dairy cows were given PGF2 alpha (25 mg i.m.) at 30 + 3 d postpartum and randomly assigned to be inseminated at a set time (Timed group) or when estrus was detected (Control group). Cows in the Timed group were synchronized by sequential administration of Buserelin (8 micrograms i.m.) on Day 0 at 1600 h, PGF2 alpha (25 mg i.m.) on Day 7 at 1600 h and Buserelin (8 micrograms i.m.) on Day 9 at 1600 h. They were inseminated on Day 10 between 0800 and 0900 h (Day 9 + 16 h). Cows in the Control group were given PGF2 alpha at 57 + 3 d postpartum and inseminated when detected in estrus. Estrus detection or insemination rate for control insemination cows was 18.1 +/- 2.5% versus 100% for time inseminated cows (P < 0.01). Mean interval from PGF2 alpha to insemination was shorter for time inseminated cows (3 +/- 2.1 d < 35.5 +/- 1.9 d; P < 0.01). Pregnancy rate was greater for time inseminated cows (13.9 +/- 2.6 > 4.8 +/- 2.5%; P < 0.01) as was overall pregnancy rate by 120 d postpartum (27.0 +/- 3.6 > 16.5 +/- 3.5%; P < 0.05). Number of days open for cows conceiving by 120 d postpartum was less for time inseminated cows (77.6 +/- 3.8 < 90.0 +/- 4.2 d; P < 0.05), as was interval to first service (58.7 +/- 2.1 < 91.0 +/- 1.9 d; P < 0.01). Services per conception were greater for time inseminated cows (1.63 +/- 0.10 > 1.27 +/- 0.11; P < 0.05). The timed insemination program did improve group reproductive performance. However, the timed insemination program will not protect the embryo from temperature-induced embryonic mortality, but management limitations induced by heat stress on estrus detection are eliminated. An economical evaluation of the timed insemination program indicates an increase in net revenue per cow with implementation of timed insemination for first service during the summer months.  相似文献

Two experiments were conducted to determine if progesterone secretion and fertility would be affected by administration of human chorionic gonadotropin (hCG) before or after the first insemination. In Experiment 1, 48 Holstein heifers received 1000 IU of hCG or 1 ml of saline on Days 2, 3, and 4 of an estrous cycle. They were inseminated at the subsequent estrus. In Experiment 2, 110 Jersey and 105 Holstein cows received a single injection of 5000 IU of hCG or 5 ml of saline on Day 3 after estrus. These cows were first inseminated either at the estrus immediately preceding treatment or at the subsequent estrus. In both experiments, blood samples for determination of progesterone were collected thrice weekly for 3 to 4 wk following treatment. In Experiment 1, progesterone concentrations during mid-cycle were higher in hCG-treated heifers than in saline-treated controls. Treatment with hCG resulted in an 11% increase in the first service conception rate (P < 0.48). In Experiment 2, hCG-treated cows displayed higher progesterone secretion during mid-cycle than saline-treated herdmates. The conception rate of cows inseminated prior to hCG-treatment was not affected by treatment, but cows inseminated after treatment had a marginally lower fertility rate. The conception rate of cows receiving a repeat insemination following hCG treatment was higher than for the controls. We conclude that treatment with hCG did not improve the conception rate at the first insemination, but it may be beneficial for cows that require a repeat service.  相似文献

Anestrus is common during the postpartum period in high-producing dairy cows. In a previous investigation, we were able to diagnose persistent follicles of 8 to 12 mm in anestrous cows. This report describes 2 consecutive studies. The objectives of the first were to 1) assess the association of persistent follicles with anestrus; and 2) evaluate 2 therapeutic treatments. In the second study, we compared the effectiveness of the best treatment established in Study 1 with the Ovsynch protocol. For Study 1, anestrous cows were considered to have a persistent follicle if it was possible to observe a single follicular structure > 8 mm in the absence of a corpus luteum or a cyst in 2 ultrasonographic examinations performed at an interval of 7 d. At diagnosis (Day 0), cows were assigned to 1 of 3 treatment groups. Cows in Group GnRH/PGF (n=17) were treated with 100 microg GnRH i.m., and 25 mg PGF2alpha i.m. on Day 14. Cows in Group PRID (n=18) were fitted with a progesterone releasing intravaginal device (PRID, containing 1.55 g of progesterone) for 9 d and were given 100 microg GnRH i.m. at the time of PRID insertion, and 25 mg PGF2alpha i.m. on Day 7. Cows in Group Control (n=18) received no treatment. The animals were inseminated at observed estrus and were monitored weekly by ultrasonography until AI or 5 weeks from diagnosis. Blood samples were also collected on a weekly basis for progesterone determination. The mean size of persistent follicles on Day 0 was 9.4 +/- 0.04 mm. Progesterone levels were < 0.2 ng/mL during the first 35 d in 16 of 18 Control cows. Cows in the PRID group showed a lower persistent follicle rate (16.7% < 70.6% < 88.9%; P < 0.0001; PRID vs GnRH/PGF vs Control, respectively); a higher estrus detection rate (83.3% > 29.4% > 11.1%; P < 0.0001) and a higher pregnancy rate (27.8% > 5.9% > 0%; P = 0.02). For the second study, 145 cows with persistent follicles were randomly assigned to 1 of 2 treatment groups: cows in Group Ovsynch (n=73) were treated with 100 microg GnRH i.m. on Day 0, 25 mg PGF2alpha i.m. on Day 7, and 100 microm GnRH i.m. 32 h later. Cows in this group were inseminated 16 to 20 h after the second GnRH dose (Ovsynch protocol). Cows in Group PRID (n=72) were treated as those in the PRID group of Study 1, and were inseminated 56 h after PRID removal. Cows in the PRID group showed a higher ovulation rate (84.8% > 8.2%: P < 0.0001); a higher pregnancy rate (34.2% > 4.1%; P < 0.0001) and lower follicular persistence rate (22.2% < 63%; P < 0.0001) than those in Ovsynch. Our results indicate that persistent follicles affect cyclic ovarian function in lactating dairy cows. Cows with persistent follicles can be successfully synchronized and time inseminated using progesterone, GnRH and PGF2alpha but show a limited response to treatment with GnRH plus PGF2alpha.  相似文献

Three experiments were conducted to examine the progestogen plus PMSG treatment for its effectiveness in inducing synchronous puberty in prepuberal zebu heifers in three different seasons. In Experiment 1, ten Ongole heifers (age 21 months) were treated with Norgestomet implants for nine days and an intramuscular injection of 400 IU of PMSG two days before implant removal. Ten heifers (age 25 months) were kept as untreated controls. Animals were inseminated 12 h after detection of cyclic estrus (not bred at induced estrus) until all animals conceived. The proportion of treated animals showing estrous, ovulatory, and cyclic activity were 100%, 75% and 25% respectively, while the average age at first conception was significantly less (P < 0.05) than in the control group. In Experiment 2, 18 Ongole heifers (age 22 months) were divided into treatment and control groups. Fixed-time inseminations were done 48 and 72 h after implant removal and 12 h after being detected in heat at other times. Estrus was seen in all while 63% became pregnant (P < 0.05). At the end of the 100-day experiment, the percent pregnant were 33 and 63 in the control and treatment groups, respectively. In the third study, twenty-six Ongole heifers (age 22 months) were assigned to treatment and control groups. Eighty-eight percent of the animals exhibited estrus, 75% ovulated (P < 0.01) and 25% conceived to fixed-time insemination. The pregnancy rate at the end of the experiment was 10 and 56% (P < 0.01) respectively in control and treated groups. Estrous response and fertility were better in the cooler month (February) and the treatment imposed in the hotter month (May) resulted in a significantly higher (P < 0.05) age and body weight at conception.  相似文献

The aim of this study was to compare four methods of estrus resynchronization performed 23 days after timed artificial insemination (TAI) plus estrus observation in Bos indicus cows. Eight hundred fourteen lactating Nelore cows were submitted to TAI and then randomly assigned to one of the five following treatments: R23 (resynchronization without eCG), R23/200 (resynchronization with 200 IU of eCG), R23/300 (resynchronization with 300 IU of eCG), R23/TCR (resynchronization with temporary calf removal [TCR]), and a control group, with estrus observation followed by AI (with no resynchronization). Treatment consisted of a progesterone device plus administration of estradiol benzoate on Day 0; on Day 8, the device was removed and cloprostenol was applied, together with estradiol cypionate. Also on Day 8, either eCG was administered or TCR was performed in the resynchronized groups, except for R23. The females were inseminated 48 hours after device removal or TCR (33 days after the first TAI). The control group was kept under estrus observation from 18 to 23 days after the first TAI and was inseminated 12 hours after detection of estrus. The first pregnancy evaluation was performed using ultrasound examination 31 days after the first TAI. After 30 days of the resynchronization, a second pregnancy evaluation was performed and the animals in the R23/300 and R23/TCR groups achieved the highest conception rates, 76.6% and 74.0%, respectively (P < 0.05). There were no differences between the conception rates of the animals in the R23/200 (63.3%), R23 (61.3%), and control (54.3%) groups (P > 0.05). These results suggest that estrus resynchronization at 23 days after TAI can effectively improve the conception rate of lactating Bos indicus cows in a short time period. Furthermore, resynchronization with 300 IU of eCG or with TCR provided the best results.  相似文献

Two trials involving 85 heifers and 67 cows were conducted to determine the effect of estrous cycle stage at the time of Syncro-Mate-B((R)) (SMB) treatment on interval to estrus following implant removal and on conception rate at the synchronized estrus. In Trial 1, 57 beef and 28 dairy heifers were treated with SMB on each representative day of a 22-d estrous cycle (estrus = Day 0). Beef heifers were artificially inseminated approximately 48 h after implant removal, whereas dairy heifers were inseminated 0 to 12 h after detection of estrus. Inseminations were scored by the inseminator according to their difficulty. Interval to the onset of estrus was not different between heifers treated early ( Day 11) in the cycle (35.2 +/- 7.2 h). Conception rate at the synchronized estrus was slightly higher in early-cycle heifers (22 47 = 47% ) compared to late-cycle heifers (14 38 = 37% , P = 0.2). Heifers that were difficult to inseminate had lower (P < 0.01) conception rates (2 11 = 18% ) at the synchronized estrus than heifers considered normal (21 51 = 41% ) or easier than normal to inseminate (13 23 = 57% ). In Trial 2, of the 131 beef cows synchronized, 67 that were estimated to be either early or late in the estrous cycle by progesterone analysis were utilized. Cows were treated with SMB and inseminated without regard to estrus 48-h after implant removal. Inseminations were scored as in Trial 1. Calves were separated from cows from the time of implant removal to insemination. Conception rate was higher (P < 0.05) in cows treated with SMB early ( Day 11, 16 35 = 46% ). Cows that were difficult to inseminate had a lower (P < 0.01) conception rate (0 8 = 0% ) than cows that were normal (43 94 = 46% ) or easier than normal to inseminate (13 29 = 45% ).  相似文献
