When equimolar ratios of mesyl chloride and methyl 2,6-di-O-mesyl-α-D-glucopyranoside were allowed to react in pyridine and the product resolved by preparative t.l.c., the 2,6-di-, 2,3,6-tri-, 2,4,6-tri-, and 2,3,4,6-tetra-mesyl esters were obtained in (0.5–0.6):1:(4–5):(1-2-1.4) molar ratio. Benzoylation of either the isolated 2,4,6-tri-O-mesyl ester or, more conveniently, the mixture from monomesylation gave the crystalline methyl 3-O-benzoyl-2,4,6-triO-mesyl-α-D-glucopyranoside (8). As both of these trimesyl esters (7 and 8) are unreported, isolation of the benzoate established the 2,4,6-ester arrangement, and the 2,3,6-triester was prepared by standard methods. Treating methyl α-D-glucopyranoside with 3 molar equivalents of mesyl chloride and, subsequently, with 1 molar equivalent of benzoyl chloride, proved a convenient method for preparing the 3-O-benzoyl derivative in moderate yield. Monotosylation of methyl 2,6-di-O mesyl-α-D-glucopyranoside was not so definitive as mesylation, but a molar ratio of 1:2.8 for the 3-O-tosyl:4-O-tosyl product was derived from n.m.r. data. This work, when combined with literature reports, establishes that, in methyl α-D-glucopyranoside, the reactivity toward sulfonylation is 6-OH>2-OH>4-OH>3-OH.  相似文献

Decarboxylative elimination of methyl 2,3-di-O-benzyl-α-D-glucopyranosiduronic acid (1) with N,N-dimethylformamide dineopentyl acetal in N,N-dimethylformamide gave methyl 2,3-di-O-benzyl-4-deoxy-β-L-threo-pent-4-enopyranoside (3). Debenzylation of 3 was effected with sodium in liquid ammonia to give methyl 4-deoxy-β-L-threo-pent-4-enopyranoside (4). Hydrogenation of 3 catalyzed by palladium-on-barium sulfate afforded methyl 2,3-di-O-benzyl-4-deoxy-β-L-threo-pentopyranoside (5), whereas hydrogenation of 3 over palladium-on-carbon gave methyl 4-deoxy-β-L-threo-pentopyranoside (6). An improved preparation of methyl 4,6-O-benzylidene-α-D-glucopyranoside is also described.  相似文献

Methyl 4,6-O-benzylidene-2-deoxy-α- -erythro-hexopyranosid-3-ulose reacted with potassium cyanide under equilibrating conditions to give, initially, methyl 4,6-O-benzylidene-3-C-cyano-2-deoxy-α- -ribo-hexopyranoside (7), which, because it reverted slowly to the thermodynamically stable -arabino isomer, could be crystallised directly from the reaction mixture. The mesylate derived from the kinetic product 7 could be converted by published procedures into methyl 3-acetamido-2,3,6-trideoxy-3-C-methyl-α- -arabino-hexopyranoside, which was transformed into methyl N-acetyl-α- -vancosaminide on inversion of the configuration at C-4. A related approach employing methyl 2,6-dideoxy-4-O-methoxymethyl-α- -erythro-hexopyranosid-3-ulose gave the kinetic cyanohydrin and thence, via the spiro-aziridine 27, methyl 3-acetamido-2,3,6-trideoxy-3-C-methyl-α- -arabino-hexopyranoside, a known precursor of methyl N-acetyl-α- -vancosaminide.  相似文献

Treatment of benzyl 2-acetamido-3-O-benzyl-2,6-dideoxy-4-O-(methylsulfonyl)-α-D-glucopyranoside (1) with sodium azide in hexamethylphosphoric triamide gave the 4-azido-α-D-galacto derivative (2), which was converted into benzyl 2,4-di-acetamido-3-O-benzyl-2,3,6-trideoxy-α-D-galactopyranoside (3) by hydrogenation and subsequent acetylation. Hydrogenolysis of 3 at atmospheric pressure afforded benzyl 2,4-diacetamido-2,4,6-tridcoxy-α-D-galactopyranoside (4), which was acetylated to give the 3-O-acetyl derivative (5). The n.m.r. spectrum of 5 was in agreement with the assigned structure and different from that of benzyl 2,4-di-acetamido-3-O-acetyl-α-D-glucopyranoside (9), which was prepared from the known benzyl 2,4-diacetamido-3-O-benzyl-2,4,6-trideoxy-α-D-glucopyranoside. Catalytic hydrogenolysis of 4 gave 2,4-diacetamido-2,4,6-trideoxy-D-galactose (6).  相似文献

Partial benzylation of methyl 2,3-di-O-benzyl-α-D-galactopyranoside gave methyl 2,3,6-tri-O-benzyl-α-D-galactopyranoside as the major product, whereas the isomeric 2,6-di-O-benzyl ether gave a mixture of products in which the ratio of methyl 2,4,6- to methyl 2,3,6-tri-O-benzyl-α-D-galactopyranoside was ≈4:1. The proportion of unreacted starting-material was low in both cases, whereas after a similar reaction of methyl 2,6-di-O-benzyl-β-D-galactopyranoside more than 50% of the dibenzyl ether was recovered unchanged. In this case also, considerably higher reactivity was exhibited by the hydroxyl group at C-4 than that at C-3. Acid hydrolysis of the methyl glycosides of the tribenzyl ethers afforded crystalline 2,4,6-tri-O-benzyl-α-D-galactose and syrupy 2,3,6-tri-O-benzyl-D-galactose. Structures of intermediates were established by acetylation, examination of their n.m.r. spectra, and conversion into the known 3-O and 4-O-methyl-D-galactose.  相似文献

Addition of methylmagnesium iodide to methyl 2,3,6-trideoxy-3-trifluoro-acetamido-α-l-threo-hexopyranosid-4-ulose (3) gave methyl 2,3,6-trideoxy-4-C-methyl-3-trifluoroacetamido-α-l-lyxo-hexopyranoside (4) and its l-arabino analogue, depending upon the reaction temperature and the solvent. The corresponding 4-O-methyl derivatives were obtained by treatment of 4 and 5 with diazomethane in the presence of boron trifluoride etherate. Treatment of 4 with thionyl chloride, followed by an alkaline work-up, gave methyl, 2,3,4,6-tetradeoxy-4-C-methylene-3-trifluoro-acetamido-α-l-threo-hexopyranoside (8), which was stereoselectively reduced to methyl 2,3,4,6-tetradeoxy-4-C-methyl-3-trifluoroacetamido-α-l-arabino-hexopyranoside. Epoxidation of 8 with 3-chloroperoxybenzoic acid gave the corresponding 4,41-anhydro-4-C-hydroxymethyl-l-lyxo derivative (10), which was also prepared by treatment of 3 with diazomethane. Azidolysis of 10, followed by catalytic hydrogenation and N-trifluoroacetylation, gave methyl 2,3,6-trideoxy-3-trifuloroacetamido-4-C-trifluoroacetamidomethyl-α-l-lyxo-hexopyranoside.  相似文献

The reaction of 2,3-di-O-acetyl-4-O-benzyl-α,β-d-xylopyranosyl bromide (2) with methyl 2,3-di-O-acetyl-β-d-xylopyranoside gave methyl O-(2,3-di-O-acetyl-4-O-benzyl-β-d-xylopyranosyl)-(1→4)-2,3-di-O-acetyl-β-d-xylopyranoside (22). Catalytic hydrogenolysis of 22 exposed HO-4′ which was then condensed with 2. This sequence of reactions was repeated three more times to afford, after complete removal of protecting groups, a homologous series of methyl β-glycosides of (1→4)-β-d-xylo-oligosaccharides. 13C-N.m.r. spectra of the synthetic methyl β-glycosides (di- to hexa-saccharide) are presented together with data for six other, variously substituted, homologous series of (1→4)-d-xylo-oligosaccharides.  相似文献

Derivatives of (S)-2-fluoro- -daunosamine and (S)-2-fluoro- -ristosamine were synthesized, starting ultimately from 2-amino-2-deoxy- -glucose which was converted, according to the literature, into methyl 2-benzamido-4,6-O-benzylidene-2-deoxy-3-O-(methylsulfonyl)-α- -glucopyranoside (2). Treatment of 2 with tetrabutylammonium fluoride gave a 63% yield of (known) methyl 3-benzamido-4,6-O-benzylidene-2,3-dideoxy-2-fluoro-α- -altropyranoside (4), together with a 6% yield of its 2-benzamido-2,3-dideoxy-3-fluoro-α- -gluco isomer. From 4, the corresponding 6-bromo-2,3,6-trideoxyglycoside 4-benzoate (6) was obtained by Hanessian-Hullar reaction. Dehydrobromination of 6, followed by catalytic hydrogenation of the resulting 5-enoside, and subsequent debenzoylation and N-trifluoroacetylation, afforded the fluorodaunosaminide, methyl 2,3,6-trideoxy-2-fluoro-3-trifluoroacetamido-β- -galactopyranoside. Reductive debromination of 6, followed by debenzoylation and N-trifluoroacetylation, gave the fluororistosaminide, methyl 2,3,6-trideoxy-2-fluoro-3-trifluoroacetamido-α- -altropyranoside. The 1H-n.m.r. spectra of the new aminofluoro sugars are discussed with respect to the effects of neighboring amino and acylamido substituents on geminal and vicinal 1H–19F coupling constants, in comparison with the reported effects of oxyge substituents.  相似文献

Methyl β-D-glucopyranoside tetraacetates (1) having a trideuterioacetyl group at O-2 (1a), O-3, (1b), O-4 (1c), and O-6 (1d) were synthesized by unambiguous routes to permit assignment of each individual acetoxyl-group signal in the p.m.r. spectrum of 1. The 6-acetoxyl resonance appears at lower field than signals of the other acetoxyl groups in carbon tetrachloride, chloroform-d, and methyl sulfoxide-d6, but in pyridine-d5 and benzene-d6, the 2-acetoxyl-group signal appears at lower field. The acetoxyl resonances of methyl 2,3,4-tri-O-acetyl-6-O-trityl-β-D-glucopyranoside (2), methyl 2,3,4-tri-O-acetyl-β-D-glucopyranoside (3), methyl 2,3-di-O-acetyl-4,6-O-benzylidene-β-D-glucopyranoside (5), methyl 2,3-di-O-acetyl-β-D-glucopyranoside (6), methyl 2,3,6-tri-O-acetyl-β-D-glucopyranoside (7), and methyl 2,3-di-O-acetyl-6-O-trityl-β-D-glucopyranoside (12) were assigned similarly after synthesis of the 2-(trideuterioacetyl) (2a, 3a, 5a, 6a, 7a, and 12a), 3-(trideuterioacetyl) (2b, 3b, 5b, 6b, 7b, and 12b), 4-(trideuterioacetyl) (2c and 3c), and 6-(trideuterioacetyl) (7c) analogues. In chloroform-d and benzene-d6, the 4-acetoxyl resonance appeared at about 0.3 p.p.m. to higher field than the other acetoxyl-group signals of 2. In chloroform-d and methyl sulfoxide-d6, the 3-acetoxyl resonance is observed at highest field in compounds 1, 3, and 5. In all of these instances, the 4-hydroxyl group is substituted by an acetyl or benzylidene group. When no 4-substituent is present (compounds 6, 7, and 12), the 3-acetoxyl group resonates at lower field than the other acetoxyl groups.  相似文献

Two carrageenans from Iridaea undulosa, isolated by precipitation of the crude polysaccharide at O.70–1.05 M and 1.55–1.65 M KCl concentrations, were studied by methylation analysis. Acid hydrolysis of the methylated derivative of the less soluble carrageenan (molar ratio galactose: 3,6-anhydrogalactose: sulphate 1.00: 0.50: 1.20) yielded major amounts of 2,6-di-O-methylgalactose (51.3 mol %), 4,6-di-O-methylgalactose (25.6%) and 4-O-methylgalactose (51.3mol%), 4,6-di-O-methylgalactose (25.6%) and 4-O-methylgalactose (13.4%). Minor quantities of 3-O-methylgalactose (4.6%) and 6-O-methylgalactose (3.2%) were found together with traces of 2,3,6- and/or 2,4,6-tri-O-methylgalactose, 2-O-methylgalactose and galactose. Oxidative acid hydrolysis produced 3,6-anhydro-2-O-methylgalactonic acid and 3,6-anhydrogalactonic acid in a molar ratio 3.5-4.0:1.0. The methylated derivative of the more soluble carrageenan (molar ratio galactose:3,6-anhydrogalactose:sulphate 1.00:0.04:1.43) gave on acid hydrolysis, 2,3,4,6-tetra-O-methylgalactose (4.6%), 2,3,6-tri-O-methylgalactose (4.2%), 2,4,6-tri-O-methylgalactose (10.7%), 4,6-di-O-methylgalactose (24.1%), 3,6-di-0-methylgalactose (8.0%), 2,3-di-O- methylgalactose (3.4%), 2,4-di-O-methylgalactose (4.6%), 2,6-di-O-methylgalactose (4.2%), 3-O-methylgalactose (19.5%),4-O-methylgalactose (9.6%),6-O-methylgalactose(3.1%),galactose (3.4%)and traces of 2-O-methylgalactose.  相似文献

Reaction of methyl 4′,6′-di-O-mesyl-β-lactoside pentabenzoate (8), synthesised via the 4′,6′-O-benzylidene derivative (6), with sodium azide in hexamethylphosphoric triamide gave three products. In addition to the required 4′,6′-diazidocellobioside (9), an elimination product, methyl 4-O-(6-azido-2,3-di-O-benzoyl-4,6-dideoxy-α-L-threo-hex-4-enopyranosyl)-2,3,6-tri-O-benzoyl-β-D-glucopyranoside (12), and an unexpected product of interglycosidic cleavage, methyl 2,3,6-tri-O-benzoyl-β-D-glucopyranoside (13), were formed. The origin of the latter product is discussed. The diazide 9 was converted into 4′,6′-diacetamido-4′,6′-dideoxycellobiose hexa-acetate (16) by sequential debenzoylation, catalytic reduction, acetylation, and acetolysis.  相似文献

Treatment of methyl 4,6-O-benzylidene-2,3-dideoxy-3-nitro-β-D-erythro-hex-2-enopyranoside (2) with nitrous acid afforded the title 2-nitro sugar (4). The same product was also prepared by heterogeneous reaction of methyl 2-O-acetyl-4,6-O-benzylidene-3-deoxy-3-nitro-β-D-glucopyranoside (1) with sodium nitrite in the presence of a phase-transfer catalyst. Acid hydrolysis of 4 gave methyl 2-deoxy-2-nitro-β-D-glucopyranoside (7). Condensation of methyl 2,6-di-O-benzyl-β-d-galactopyranoside with 2-methyl-(3,4,6-tri-O-acetyl-1,2-dideoxy-α-d-glucopyrano)-[2,1,-d]-2-oxazoline (1) in 1,2-dichloroethane, in the presence of p-toluenesulfonic acid, afforded a trisaccharide derivative which, on deacetylation, gave methyl 3,4-di-O-(2-acetamido-2-deoxy-β-d-glucopyranosyl)-2,6-di-O-benzyl-β-d- glactopyranoside (5). Hydrogenolysis of the benzyl groups of 5 furnished the title trisaccharide (6). A similar condensation of methyl 2,3-di-O-benzyl-β-d-galactopyranoside with 1 produced a partially-protected disacchraide derivative, which, on O-deacetylation followed by hydrogenolysis, gave methyl 6-O-(2-acetamido-2-deoxy-β-d-glucopyranosyl)-β-d-glactopyranoside (10). Condensation of methyl 3-O-(2-acetamido-4,6-O-benzylidene-2-deoxy-β-d-glucopyranosyl)-2,4,6-tri-O-benzyl-β-d- galactopyranoside with 3-O-(2-acetamido-3,4,6-tri-O-acetyl-2-deoxy-β-d-glucopyranosyl)-2,4,6-tri-O-acetyl-α-d-galactopyranosyl bromide in 1:1 benzene-nitromethane in the presence of powdered mercuric cyanide gave a fully-protected tetrasaccharide derivative, which was O-deacetylated and then subjected to catalytic hydrogenation to furnish methyl O-(2-acetamido-2-deoxy-β-d-glucopyranosyl)-(1→3)-O-β-d-galactopyranosyl-(1å3)-O-(2-acetamido-2-deoxy- β-d-glucopyranosyl)-(1å3)-β-d-galactopyranoside (15). Three further compounds, namely, benzyl 2-acetamido-3-O-benzyl-2-deoxy-6-O-(2,3,4,6-tetra-O-benzyl-D-glucopyranosyl)-α-D-glucopyranoside, benzyl 2-acetamido-3-O-benzyl-2-deoxy-6-O-(2,3,4,6-tetra-O-benzyl-D)-mannopyranosyl)-α-D-glucopyranoside, and benzyl 2-acetamido-3-O-benzyl-2-deoxy-4,6-di-O-(2,3,4,6-tetra-O-benzyl-D-mannopyranosyl)-α-D-glucopyranoside, were formed by reaction of the respective glycosyl halide with benzyl 2-acetamido-3-O-benzyl-2-deoxy-α-D-glucopyranoside present as contaminant in 1.  相似文献

Benzoylation of benzyl 2-acetamido-2-deoxy-4,6-O-isopropylidene-α-d-glucopyranoside, benzyl 2-deoxy-2-(dl-3-hydroxytetradecanoylamino)-4,6-O-isopropylidene-α-d-glucopyranoside, and benzyl 2-deoxy-4,6-O-isopropylidene-2-octadecanoylamino-β-d-glucopyranoside, with subsequent hydrolysis of the 4,6-O-isopropylidene group, gave the corresponding 3-O-benzoyl derivatives (4, 5, and 7). Hydrogenation of benzyl 2-acetamido-4,6-di-O-acetyl-2-deoxy-3-O-[d-1-(methoxycarbonyl)ethyl]-α-d-glucopyranoside, followed by chlorination, gave a product that was treated with mercuric actate to yield 2-acetamido-1,4,6-tri-O-acetyl-2-deoxy-3-O-[d-1-(methoxycarbonyl)ethyl]-β-d-glucopyranose (11). Treatment of 11 with ferric chloride afforded the oxazoline derivative, which was condensed with 4, 5, and 7 to give the (1→6)-β-linked disaccharide derivatives 13, 15, and 17. Hydrolysis of the methyl ester group in the compounds derived from 13, 15, and 17 by 4-O-acetylation gave the corresponding free acids, which were coupled with l-alanyl-d-isoglutamine benzyl ester, to yield the dipeptide derivatives 19–21 in excellent yields. Hydrolysis of 19–21, followed by hydrogenation, gave the respective O-(N-acetyl-β-muramoyl-l-alanyl-d-isoglutamine)-(1→6)-2-acylamino-2-deoxy-d-glucoses in good yields. The immunoadjuvant activity of these compounds was examined in guinea-pigs.  相似文献

The 6-O-mesyl, 6-O-tosyl, 6-bromo-6-deoxy, and 6-deoxy-6-iodo derivatives of 1,4-anhydro-DL-allitol were obtained by treatment of the corresponding 1,6-di-substituted derivatives (2, 3, 6, 4) of 2,3,4,5-tetra-O-acetylallitol with hot, methanolic hydrogen chloride. Compounds 2 and 3 were prepared by the acetolysis of the 1,6-di-O-mesyl and 1,6-di-O-tosyl derivatives (8 and 11) of di-O-benzylideneallitol. Iodide displacement on 2 gave 4, and detritylation-bromination of 2,3,4,5-tetra-O-acetyl-1,6-di-O-tritylallitol (5) gave 6. The acetal residues of di-O-benzylideneallitol have been shown to span the secondary carbon atoms.  相似文献

A series of 2-O-benzoyl-4,6-di-O-benzyl-α-d-galactopyranosyl halides carrying either a second benzoyl group (8a, 12a) or a selectively removable, temporary protecting group (8b–d, 12b) at position 3 was synthesized from allyl α-d-galactopyranoside (1). The key intermediate was 1-propenyl 4,6-di-O-benzyl-α-d-galactopyranoside (5), prepared from 1 via the 4,6-O-benzylidene-2,3-di-O-crotyl derivative 2. The successive incorporation of the 2-O-benzoyl group, by selective acylation at low temperature, and of various 3-substituents gave fully substituted 1-propenyl α-d-galactopyranosides 6a–d. These were converted into the glycosyl halides by published methods. An improved preparation of allyl 2,6-di-O-benzyl-(15) and 2,4,6-tri-O-benzyl-(19) α-d-galactopyranoside was achieved. The direct acetonation of 1 to the 3,4-O-isopropylidene derivative 13, followed by benzylation and mild acid hydrolysis, gave 15 in 56% yield. The transient protection of O-3 in 15 was accomplished by the alkylation of the dibutylstannylene derivative 16 with (2-methoxyethoxy)methyl chloride. Successive benzylation and mild acid hydrolysis of the product 17 efficiently furnished 19.  相似文献

The pentasaccharide chain of the Forssman antigen, O-(2-acetamido-2-deoxy-α-d-galactopyranosyl)-(1→3)-O-(2-acetamido-2-deoxy-β-d-galactopyranosyl)-(1→3)-O-α-d- galactopyranosyl-(1→4)-O-β-d-galactopyranosyl-(1→4)-d-glucopyranose (46) was synthesized by a block synthesis in which an α-d-glycoside linkage between two d-galactose residues was formed. The trisaccharide O-(6-O-acetyl-2-azido-3,4-di-O-benzoyl-2-deoxy-α-d-galactopyranosyl)- (1→3)-O-(6-O-acetyl-4-O-benzyl-2-deoxy-2-phthalimido-β-d-galactopyranosyl)-(1→3)-6-O-acetyl-2,4-di-O-benzyl- α-d-galactopyranosyl bromide (40) (this was obtained through acetolysis of O-(6-O-acetyl-2-azido-3,4-di-O-benzoyl-2-deoxy-α-d-galactopyranosyl)- (1→3)-O-(6-O-acetyl-4-O-benzyl-2-deoxy-2-phthalimido-β-d-galactopyranosyl)-(1→3)-1,6-anhydro-2,4-di-O-benzyl-β-d- galactopyranose to the acetyl derivative, followed by reaction with titanium tetrabromide under anhydrous conditions) was condensed with benzyl-4-O-(6-O-benzoyl-2,3-di-O-benzyl-β-d-galactopyranosyl)-2,3,6- tri-O-benzyl-β-d-glucopyranoside were in the presence of silver carbonate and perchlorate. Condensation of 2,4,6-tri-O-acetyl-3-deoxy-3-fluoro-α- -galactopyranosyl bromide (3) with methyl 2,3,4-tri-O-acetyl-β- -galactopyranoside (4) gave a fully acetylated (1→6)-β- -galactobiose fluorinated at the 3′-position which was deacetylated to give the title disaccharide. The corresponding trisaccharide was obtained by reaction of 4 with 2,3,4-tri-O-acetyl-6-O-chloroacetyl-α- -galactopyranosyl bromide (5), dechloroacetylation of the formed methyl O-(2,3,4-tri-O-acetyl-6-O-chloroacetyl-β- -galactopyranosyl)-(1→6)- 2,3,4-tri-O-acetyl-β- -galactopyranoside to give methyl O-(2,3,4-tri-O-acetyl-β- -galactopyranosyl)-(1→6)-2,3,4-tri-O-acetyl-β- -galactopyranoside (14), condensation with 3, and deacetylation. Dechloroacetylation of methyl O-(2,3,4-tri-O-acetyl-6-O-chloroacetyl-β- -galactopyranosyl)-(1→6)-O-(2,3,4-tri-O-acetyl- β- -galactopyranosyl)-(1→6)-2,3,4-tri-O-acetyl-β- -galactopyranoside, obtained by condensation of disaccharide 14 with bromide 5, was accompanied by extensive acetyl migration giving a mixture of products. These were deacetylated to give, crystalline for the first time, the methyl β-glycoside of (1→6)-β- -galactotriose in high yield. The structures of the target compounds were confirmed by 500-MHz, 2D, 1H- and conventional 13C- and 19F-n.m.r. spectroscopy.  相似文献
