As part of program of research into insect cellular immunity, an integrated light and electron microscopic study of the hemocytes of seven members of the Order Dictyoptera has been made. In fresh hemolymph, five cell types, the prohemocytes, plasmatocytes, granular cells, spherule cells and cystocytes, are easilv distinguished. However, in thick Araldite sections and in thin sections in the electron microscope it is sometimes difficult to identify the various cell types. The reasons for this difficulty are discussed. Granules with a microtubular substrcture are found in the plasmatocytes, spherule cells and cystocytes. In the plasmatocytes these granules have a different ultrastructure than those in the spherule cells and cystocytes. The in vitro fragility of these granules in both the spherule cells and cystocytes during coagulation partially explains the previous confusion in distinguishing these two cell types. Evidence is presented which indicates that the plasmatocytes, granular cells and spherule cells represent a developmental series originating from the prohemocytes. Where exactly the cystocytes are derived from is unknown.  相似文献

The morphological features of the hemocytes of the crustacean Ligia exotica are similar to hemocytes of insects and millipedes. Jones system of hemocyte classification is extended to crustacean hemocytes. As in insects, seven classes of hemocytes, identified as prohemocytes, plasmatocytes, granular hemocytes, cystocytes, oenocytoids, spherule cells and adipohemocytes, occur. The prohemocytes can be subdivided into five categories that probably represent the precursor of major cell types. The structural and chemical features of other major cell classes are distinct and support the concept of Jones ('62) that these types might have different lineages and might not be capable of transforming into one another. Some of the prohemocytes, plasmatocytes and granular hemocytes are amoeboid. Cystocytes do not bring about any visible plasma coagulation similar to their counterpart in millipedes. Oneocytoids and adipohemocytes are rare. Plasmatocytes, cystocytes and oenocytoids occur in conglomerates, the significance of which is discussed. The cell types are compared with those of the hemocytes of other crustaceans. It is suggested that the nomenclature based on morphological characters is more suited for crustacean hemocytes than a nomenclature based on behavioural and physiological characters.  相似文献

Summary Hydrocortisone injections into rats on postnatal days 3–9 caused an increase in the number of small granulecontaining cells in the superior cervical ganglia. These cells, corresponding to the small, intensely fluorescent cells, showed an extensive rough endoplasmic reticulum, a large Golgi apparatus and a very large number of granular vesicles. In addition to the granular vesicles, 70–160 nm in diameter, in which the dense core filled most of the vesicle, most cells of the hydrocortisone-injected rats contained also larger granular vesicles, up to 350 nm in diameter, in which the dense core was eccentrically located. A minority of the cells contained only granular vesicles 70–100 nm in diameter, which was the only type seen in the saline-treated control rats.Thirty days after discontinuation of the hydrocortisone treatment, most of the cells with large granular vesicles had disappeared, and only two profiles of such cells were seen on day 40. The other small cells contained only granular vesicles 70–160 nm in diameter, and these cells could not be distinguished from the small granule-containing cells of 40-day-old control rats treated early postnatally with saline.Hydrocortisone treatment, first on days 3–9 and subsequently on days 40–46, caused reappearance of the small granule-containing cells with large granular vesicles up to 350 nm in diameter, the dense core of which was eccentrically located. Electron-mictoscopic observations of the blood cells (Haemocytes) in the legs of the cockroach Blaberus discoidalis reveal three 'types' of cells distinguishable on the basis of fine structure. One type of cell contains numerous membrane-limited opaque bodies which may be lysosomes. A second type contains abundant digestive vacuoles and appears phagocytic. A third type, frequently encountered in rewlymoulted untanned animals but less often observed in mature adults, contains membrane-limited tubule-containing bodies (TCB) filled with rows of 340 A tubules which are quite different from cytoplasmic microtubules. It is suggested that haemocytes containing TCB may be equivalent to the coagulocytes (cystocytes) of light microscopy which actively participate-in-the process of haemolymph coagulation in Orthoptera. No attempt whatsoever is made at classification. Since considerable overlap in fine structure exists between cell types, it seems probable that the electron-images of fixed cells observed in this study represent several morphological expressions of which an individual cell may be capable during its lifetime.  相似文献

We recorded the activity of two types of granular cells in the rostral folia of the paramedial lobe (the projection region of the front legs) of the cerebellar cortex in cats immobilized by administration of ditiline; these cells differed in their receptive fields, the characteristics of their reaction to single stimulation of somatic nerves, and the character of their background activity. The granular cells of the first type were excited only when the nerves of the front legs were stimulated (reacting with 1–3 impulses with a latent period of 8–20 msec) and were inhibited between 20–50 and 70–180 msec after stimulation of the nerves of any leg. The cells of the second type responded with volleys of 3–6 impulses with a latent period of 20–40 msec to stimulation of the nerves of all four legs. Comparison of the reactions of the granular cells and other neurons of the cerebellar cortex showed that the cells of the first type cause excitation of the Purkinje and Golgi cells and the neurons of the molecular layer. The granular cells of the second type have an excitatory effect on the Golgi cells. The differences in the reactions of the two types of granular cells result from the fact that they are selectively innervated by the mossy fibers of different afferent pathways. Comparison with the data in the literature enables us to surmise that the fibers of the cuneocerebellar tract terminate at granular cells of the first type, while the reticular fibers terminate at cells of the second type.Institute of Problems of Information Transmission, Academy of Sciences of the USSR, Moscow. Summary The pineal organ of the killifish, Fundulus heteroclitus, was investigated by electron microscopy under experimental conditions; its general and characteristic features are discussed with respect to the photosensory and secretory function. The strongly convoluted pineal epithelium is usually composed of photoreceptor, ganglion and supporting cells. In addition to the well-differentiated photosensory apparatus, the photoreceptor cell contains presumably immature dense-cored vesicles (140–220 nm in diameter) associated with a well-developed granular endoplasmic reticulum in the perinuclear region and the basal process. These dense-cored vesicles appear rather prominent in fish subjected to darkness. The ganglion cell shows the typical features of a nerve cell; granular endoplasmic reticulum, polysomes, mitochondria and Golgi apparatus are scattered in the electron-lucent cytoplasm around the spherical or oval nucleus. The dendrites of these cells divide into smaller branches and form many sensory synapses with the photoreceptor basal processes. Lipid droplets appear exclusively in the supporting cell, which also contains well-developed granular endoplasmic reticulum and Golgi apparatus. Cytoplasmic protrusions filled with compact dense-cored vesicles (90–220 nm in diameter) are found in dark-adapted fish. The origin of these cytoplasmic protrusions, however, remains unresolved. Thus, the pineal organ of the killifish contains two types of dense-cored vesicles which appear predominantly in darkness. Summary The fine structure of the preterminal nerve fibers of the rabbit myometrial smooth muscle was studied using potassium permanganate fixation or glutaraldehyde fixation with postosmification. The preterminal fibers were mostly formed by 2–10 axons enveloped by Schwann cells. Two kinds of axons and axon terminals were found. (1) Adrenergic axons, which contained many small, granular vesicles (diameter 300–600 Å) and large granular vesicles (diameter 700–1200 Å) which represented ca. 2% of the total count of the vesicles. (2) Nonadrenergic axons, which contained small agranular vesicles (diameter 300–600 Å) and large granular vesicles (diameter 700–1200 Å). Both types of axons formed preterminal varicosities along their course. The real terminal varicosities, representing the anatomical end of the axons, were usually larger than the preterminal ones and showed close contact to the plasma membranes of the smooth muscle cells. Both adrenergic and nonadrenergic terminals were found close to the smooth muscle cells, but a gap of at least 2000 Å was always present between the two cell membranes. The axons and preterminal varicosities of both types of nerves were in intimate contact with each other within the preterminal nerve fiber. Axo-axonal interactions between the two types of axons are possible in the rabbit myometrium. The relative proportion of the nonadrenergic axons from the total was about one fourth.  相似文献

Protective effect of hypothermia during ischemia in neural cell cultures   总被引:5,自引:0,他引:5  
Hypothermia offers protection from the effects of ischemia in small animals. We have recently shown that similar to small animals, hypothermia may also be protective in an astrocytic model of simulated ischemia in cell culture. This study was designed to look at the protective effects of hypothermia in cultures of cerebellar granular (glutamatergic) and cortical (GABAergic) neurons. We used LDH release into the medium as an indicator for neuron damage. Experiments were all done in sister cultures, in groups of six cultures at two temperatures (37 and 32 degrees Celsius). The duration of ischemia was three hours in cerebellar granular neuronal cell cultures and six hours in cortical neurons. LDH release was measured immediately after the insult. Hypothermia protected both granular and cortical neurons. In granular cells, LDH release was 62+/–18 at 32 degrees and 212+/–15 at 37 degrees (p=0.02). Cortical neurons showed LDH release of 15+/–2 at 32 degrees and 32+/–2 at 37 degrees (p=0.005). Our study suggests that similar to astrocytes, the protective effects of hypothermia are evident in neuronal cell cultures from the cerebellum and the cerebral cortex. Cell culture systems should prove useful techniques in understanding mechanisms of hypothermic protection during simulated ischemia in neurons from different sites.  相似文献

Previous studies have suggested the involvement of biogenic amines in insect metamorphic events and post-embryonic development. The effect of azadirachtin (AZ), a natural antifeedant and growth-disrupting compound, on the nutrition, development, and biogenic amine contents of the last instar larvae of the Eastern Death’s Head hawk moth, Acherontia styx, was examined. Single doses of AZ, injected into the haemolymph at day 1 post-ecdysis, inhibited food consumption in a dose-dependent manner (ED50 = 0.65±0.08 μg AZ/g body weight), and was found to be highly effective at producing pupal deformities and inhibiting larval growth (0.1–0.2 μg AZ/g body weight range). Biogenic amine contents, namely octopamine (OA), dopamine (DA) and serotonin (5-HT), in the brain and the haemolymph of 4-day- and 8 day-old larvae were analysed using high-performance liquid chromatography (HPLC) with an electrochemical detector (ECD). A dose-response relationship between AZ and biogenic amine contents in the brain and the haemolymph was also established. Low doses of AZ (0.1–0.2 μg AZ/g body weight) caused a dramatic reduction in OA and 5-HT levels in both the brain and the haemolymph. However, higher doses (0.9–1.2 μg AZ/g body weight) were needed to induce a significant reduction in DA levels. The significance of these findings in relation to the possibility of the involvement of biogenic amines in regulating metamorphic events in insects through mediation of juvenile hormone synthesis and/or release is discussed.  相似文献

Summary The specificity and distribution of carbohydrate moieties, which act as receptors for lectins on the haemocytes of two insect species, Blaberus craniifer and Extatosoma tiaratum, were investigated. Four peroxidase-labelled lectins were utilised as probes: wheat-germ agglutinin, Ricinus communis (120) agglutinin, concanavalin A and Lens culinaris agglutinin, and binding visualised by reaction with DAB/H2O2. The lectin-binding studies indicated that Blaberus and Extatosoma plasmatocytes, and Extatosoma spreading granular cells possess surface N-acetyl-D-glucosamine, D-galactose and mannose moieties; Extatosoma cystocytes have D-galactose and mannose; whilst Blaberus granular cells/cystocytes and Extatosoma fine granular cells have mannose determinants.  相似文献

This paper describes the formation of single polar bundles of pili on Azospirillum brasilensecells, the twitching motility of cell aggregates, and a new type of social behavior—the dispersal of bacterial cells in semiliquid agar associated with the formation of granular inclusions (the so-called Gri+phenotype)—which is an alternative to swarming (the Swa phenotype). The wild-type A. brasilensecells occurring in a semiliquid agar may show either the Swa+Gri, or SwaGri, or SwaGri+phenotype. The formation of single polar flagella (Fla) or polar bundles of pili may reflect two alternative states of A. brasilensecells. 为研究蚧虫血淋巴中血细胞的种类及其结构特征,本文采用荧光显微镜、扫描电镜和透射电镜观察了白蜡绵粉蚧Phenacoccus fraxinus Tang(半翅目:蚧总科:粉蚧科)雌成虫血细胞的显微形态超微结构.结果显示,在荧光显微镜和扫描电镜下观察识别出白蜡绵粉蚧血淋巴中的5种血细胞,即原血胞、浆血胞、粒血胞、类绛色血胞和囊血胞.在透射电镜下,原血胞的细胞核明显,表现出高的细胞质密度;浆血胞最典型的特征是细胞质中有大量的囊泡;粒血胞的细胞质中有发达的粗面内质网和许多玫瑰形的细颗粒;类绛色血胞最典型的特征是有许多结晶,并在细胞质空泡区内分布更多;囊血胞透明性强,具有围核空间,膨大成潴泡状.  相似文献

Two types of salivary alveoli are present in adultArgas (Persicargas) persicus: agranular type I and granular type II alveoli. Type I alveoli consist of a large central cell surrounded by a constrictor cell and peripheral cells with numerous infoldings of the basal membrane similar to epithelia involved in active transport. The basal infoldings form a previously undescribed successively convoluted membranous pattern which may increase the capability of peripheral cells for active transport. Type II alveoli consist of three granular cells (a, b, c) and two agranular (adlumenal and ablumenal interstitial) cells. Golgi bodies and rough endoplasmic reticulum are probably involved in the granule formation. The granules are discharged within 5–10 min after feeding commences, and presumably contain anticoagulant substances and pharmacologically active agents that promote the blood flow of the host during tick feeding. Although the adlumenal cells are not structurally affected by feeding, ablumenal cells develop into transporting epithelia.  相似文献

Summary A method for the production and preparation of an enzyme which degrades yeast cell walls from a species of aRhizoctonia (tentatively identified asR. solani) was established on a commercial scale. The production of crude enzyme powder, having a lytic activity of 100 units/mg, in batches of 80 kg is feasible.The enzyme preparation was evaluated for industrial use. When yeast cells were treated with this enzyme, the digestibility of feed yeast was improved 1.4–2 fold in vitro; the efficiency of a mechanical disintegrator in extracting cellular substances was increased 35–50%; the release of soluble glucans having widely varying degrees of polymerization was induced; the extraction of cellular protein by alkali was facilitated 2–3 fold; an 80% release of cell-bound invertase was induced and 2–3 times more yeast extract could be prepared.Studies on Fungal Enzymes Active in Hydrolysing Yeast Cell Wall (VII)  相似文献

Esterase isozyme polymorphism was documented for digestive juice and haemolymph of the tropical multivoltine silkworm, Bombyx mori L., breed CB5 (GP) and its syngenic lines (CB5Lme-1, CB5Lm-2 and CB5Lm-5) using α- and β-naphthylacetate separately as nonspecific substrates (Ogita, Z., Kasai, T., 1965. Genetico-biochemical analysis of specific esterases in Musca domestica. Jpn. J. Genet. 40, 173–184). Polymorphism existed in the isozyme pattern of α-esterase with two or three bands in digestive juice and three to five bands in haemolymph. No polymorphism was observed in β-esterase isozyme pattern having four bands in digestive juice and two bands in haemolymph. During the course of esterase isozyme studies, the presence of some specific α-esterase bands (Est-1, 4 and 5) in haemolymph and β-esterase bands (Est-1, 2 and 3) in digestive juice were observed. But both α- and β-esterase bands Est-3 and 4 in digestive juice and Est-2 and 3 in haemolymph were found to be nonspecific. Nonspecific β-esterase band (Est-3) in haemolymph of CB5 (GP) and its syngenic lines withstood a temperature up to 80±1°C for 10 min. No thermostable band was observed in the isozyme zymogram of α-esterase in digestive juice and haemolymph or β-esterase in digestive juice. Overall, this study discusses the presence of esterase heterogeneity in the CB5 (GP) genepool, syngenic lines development, occurrence of specific α- and β-esterase bands in digestive juice and haemolymph and thermostable β-esterase band Est-3 in haemolymph in tropical silkworm Bombyx mori L.  相似文献

Summary The innervation of the intestinal wall in the teleosts Myoxocephalus and Pleuronectes was examined electron microscopically. Two classes of axons can be identified. The first, which is in the majority, contains numerous 50–150 nm granular vesicles as well as some 40–50 nm agranular vesicles while the second contains predominantly the 40–50 nm agranular vesicles. Chromate/dichromate staining methods suggest that the first type is aminergic. Both types lie in close association with the perikarya of intrinsic myenteric neurons but only axons containing predominantly agranular vesicles have synaptic membrane specialisations. No axon bundles pass into the longitudinal muscle layer in Myoxocephalus gut and though some do in Pleuronectes, they do not closely approach the smooth muscle cells. Axons containing large granular vesicles lie in intimate contact with the myocytes of the circular muscle layer. Both axon types pass through the submucosa to form a plexus underneath the mucosal epithelium. Varicosities containing agranular or granular vesicles are separated from the epithelial cells by a gap of about 200 nm in which lies a basal lamina.  相似文献

Summary Streptococcus salivarius subsp. thermophilus and Lactobacillus delbrueckii subsp. Bulgaricus were immobilized separately in -carrageenan-locust bean gum gel beads. The beads were prepared by a dispersion process in a two-phase system (water in oil) and two ranges of bead diameter selected by sieving (0.5–1.0 mm and 1.0–2.0 mm). Fermentations with the two strains were conducted in bench bioreactors in a supplemented whey permeate medium. Free and entrapped cells (two ranges of bead diameter and two levels of initial bead cell load) were grown in mixed culture, and carbohydrate utilization, acid production and cell growth or cell release rate measured. Fermentation rates were influenced by bead diameter and initial cell load of the beads. Beads with high initial cell density increased fermentation rates compared to low cell density beads or free cells. Smaller diameter beads (0.5–1.0 mm) showed a stable tendency (not statistically significant p a > 0.05) towards higher cell release rates, lactose utilization, galactose accumulation and lactic acid production than did larger diameter beads (1.0–2.0 mm). Immobilization of S. salivarius subsp. thermophilus and L. delbrueckii subsp. bulgaricus in separate beads did not seem to affect protocooperation during batch fermentation, and allowed for high cell release rates into the medium.  相似文献

Summary Preliminary ultrastructural studies on the effects of 5,6-Dihydroxytryptamine (5,6-DHT) on the anterior byssus retractor muscle (ABRM) of Mytilus show degeneration of 2 types of monoaminergic nerves after 10 days of drug treatment. One type contained large granular vesicles (560–1,680 Å) while the other had small granular vesicles (200–640 Å). These axons may possibly represent serotonergic and dopaminergic nerves, thought to innervate this muscle.Two other types of profiles seemed to be unaffected by the drug. One conforms to cholinergic nerves while the other has a predominance of large opaque vesicles (1,200–2,500 Å). The significance of these findings is discussed in the light of recent observations on the neurotoxic effects of 5,6-DHT on vertebrate and molluscan nerves.The author is grateful to Professor G. Summary The oxygen and carbon dioxide transporting properties of the haemolymph from an amphibious Australian crab,Holthuisana transversa were investigated. Within the temperature range 15 to 35°C increasing temperature markedly decreased oxygen affinity (H=–54 kJ·mol–1). The Bohr effect was small at all temperatures with a mean value of –0.13. Over the temperature range 15–35°C there was a significant increase in the cooperativity of oxygen binding. Changing the concentration of Ca,l-lactate or haemocyanin in the haemolymph could elicit no significant change in either O2 affinity or cooperativity of O2 binding. There was no evidence in support of a specific effect of CO2 on oxygen affinity of either non-dialysed or dialysed haemolymph.The amount of CO2 that could be carried byH. transversa haemolymph was significantly reduced by increased temperature (approx. 14 to 12.5 mmol·l–1 CO2). Comparisons of oxygenated and deoxygenated haemolymph at a fixed pH were unable to demonstrate the presence of a significant Haldane effect. Combining data from oxygenated and deoxygenated haemolymph the buffer value was calculated to be in the range –6.2 to –8.5 mmol·l–1 HCO 3 ·pH unit–1.The insensitivity ofH. transversa haemocyanin function to all modulating influences except temperature is discussed with respect to the ecology of this crab.  相似文献

Summary A cell entrapment process using -carrageenan — locust bean gum gel is presented. Streptococcus thermophilus, Lactobacillus bulgaricus and S. lactis were immobilized in small gel beads (0.5–1.0 mm and 1.0–2.0 mm diameter) and fermentations in bench bioreactors were conducted. Viability of entrapped cells, lactose utilization, lactic acid production and cell release rates were measured during fermentation. The procedure was effective for S. thermophilus, L. bulgaricus and S. lactis, and the viability of these bacteria remained very high throughout entrapment steps and subsequent storage. Bead diameter influenced the fermentation rate: smaller beads (0.5–1.0 mm) permitted an increase in release rates, lactose utilization and acid production by entrapped cells, approximating values attained with free cells.  相似文献
