The structure of the cell wall, the arrangement of the cyst exine, and the origin and distribution of intine vesicles in Azotobacter vinelandii ATCC 12837 were examined by freeze-etching and conventional electron microscopic techniques. In the vegetative organism the cell wall appears to have a woven texture which disappears during cyst formation. The exine is composed of two different types of material: the outer layer is a fibrous, amorphous layer, and the numerous inner layers form the basic hexagonal structures which unite to form the cyst coat. The presence of intine vesicles in the encysting organism was confirmed in frozen-etched cells. The appearance of frozen-etched cells and cysts and the distribution of capsular material indicate that extracellular polysaccharide is an important factor in cyst formation.  相似文献

Summary The formation and nature of the generative cell wall and the detachment mode of the generative cell from the intine in Polystachia pubescens were observed by LM and TEM. Vesicles evenly positioned within the phragmoplast fuse to form a cell plate that divides the microspore into the generative and vegetative cell. This cell plate consists of callose. Before the generative cell leaves the intine, however, the callose is completely resorbed and is not replaced by any other substance. The generative cell becomes detached from the intine by moving towards the centre of the pollen grain. A constriction formed thereby gives the generative cell a bulb-like appearance and leads ultimately to the generative cell being pinched off. Plasma-filled vesicles originating from the generative cell remain between the intine and the plasma membrane of the vegetative cell.  相似文献

Dormant cysts of Azotobacter vinelandii germinated at 30 degrees C in Burk nitrogen-free media containing 1% glucose. Samples taken at intervals and examined by electron microscopy revealed that as germination progressed, vesicle-like and fibrillar structures became visible in the intine region. Lamellae associated with the cell membrane appeared in the central body at 6 h post-initiation of germination. Both electron micrographic and chemical analysis showed that the poly-beta-hydroxybutyrate content of cysts decreased significantly after 4 h of germination. Dormant cysts were resistant to sonic oscillation, but this property was lost during their conversion to metabolically active vegetative cells.  相似文献

Summary Brassica napus pollen development during the formation of the generative cell and sperm cells is analysed with light and electron microscopy. The generative cell is formed as a small lenticular cell attached to the intine, as a result of the unequal first mitosis. After detaching itself from the intine, the generative cell becomes spherical, and its wall morphology changes. Simultaneously, the vegetative nucleus enlarges, becomes euchromatic and forms a large nucleolus. In addition, the cytoplasm of the vegetative cell develops a complex ultrastructure that is characterized by an extensive RER organized in stacks, numerous dictyosomes and Golgi vesicles and a large quantity of lipid bodies. Microbodies, which are present at the mature stage, are not yet formed. The generative cell undergoes an equal division which results in two spindle-shaped sperm cells. This cell division occurs through the concerted action of cell constriction and cell plate formation. The two sperm cells remain enveloped within one continuous vegetative plasma membrane. One sperm cell becomes anchored onto the vegetative nucleus by a long extension enclosed within a deep invagination of the vegetative nucleus. Plastid inheritance appears to be strictly maternal since the sperm cells do not contain plastids; plastids are excluded from the generative cell even in the first mitosis.  相似文献

The present study of the formation of the generative and vegetative cells in wheat has demonstrated some cytological details at the ultrastructural level. The phragmoplast formed in telophase of the first microsporic mitosis extended centrifugally until it connected with the intine of the pollen grain. A new cell wall was then formed to separate the generative and the vegetative cells. By unequal cytokinesis the former is small and the latter large. In early developmental stage of male gametophyte, the organelles in the cytoplasm of the generaVive cell and the vegetative cells are similar, including mitochondria, dictyosomes, rough endoplasmic retieulum, free and clustered ribosomes and plastids, but microtubules were observed only in the early cytokinesis stage. In the further developmental stage of the male gemetophyte, the generative cell gradually detached from the intine of pollen grain and grew inward to the cytoplasm of the vegetation cell. When the generative cell became round and free in the cytoplasm of the vegetative cell, the wall materials between plasma membranes of the cytoplasm of the generative and the vegetative cells disappeared completely, so that it was a naked cell with a double-layer membrane at this time. The heterogeneity between both cells was then very conspiceous. The organelles in the cytoplasm of the generative cell have hardly any changed besides the degeneration of plastids, but in vegetative cytoplasm the mitochondria and plastids increased dramatically both in number and size. The rapid deposition of starch in the plastids of the cytoplasm of the vegetative cell made the most conspicuous feature of the vegetative cell in mature pollen grain. The significance of the presence of a temporary cell wall in generative cell and heterogeneity between generative and vegetative cells are discussed.  相似文献

When young cells of Azotobacter vinelandii are impinged on membrane filters, washed free of carbon substrate, and placed on a mineral salts basal medium, the culture will proceed to encyst although at a slower rate than if n-butanol were supplied as a substrate. The endogenous cysts are depleted in polyβ-hydroxybutyrate and have a narrower intine but show an increased resistance to desiccation and are susceptible to lysis by chelating agents. Membrane-supported cells reveal details of the encystment process such as the formation of a zone within the capsule prior to exine formation and the early deposition of exine structures.  相似文献

In the present study it was demonstrated the existence of a new set of membrane-bounded vesicles in Giardia lamblia. They were found in dividing and non-dividing trophozoites studied by routine transmission electron microscopy, freeze-fracture and Thiéry's technique. Encysting cells were not studied. These vesicles appear different to the previously reported components of the Giardia endomembranous system, such as the endoplasmic reticulum (ER), lysosome-like peripheral vesicles (PV), and the encystation-specific vesicles (ESV) that appear during trophozoite differentiation into cysts. They measure 100-150 nm in diameter, and thus are smaller than the peripheral vesicles, and the encystation-specific vesicles (ESV). They were found in clusters, scattered throughout the cytoplasm, but preferentially located close to the nuclei, axonemes, median bodies, and ER profiles. These internal vesicles are roughly spherical, and their contents present different electron densities and are more electrondense than those of the peripheral vesicles. They appeared to be budding from the outer nuclear membrane envelope. These cytoplasmic vesicles were found only in cells with very good fixation. Only few cells in the same preparation exhibited these vesicles.  相似文献

The formation of cysts by Azotobacter vinelandii involves the synthesis of lipids as major metabolic products. Cells which encyst at low levels in aging glucose cultures undergo the same pattern of lipid synthesis as cells which undergo reasonably synchronous encystment in beta-hydroxybutyrate or n-butanol. The accumulation of poly-beta-hydroxybutyrate (PHB) precedes the synthesis of 5-n-heneicosylresorcinol and 5-n-tricosylresorcinol (AR1), which is then followed in about 6 h by the synthesis of the 5-n-alkylresorcinol galactosides (AR2). In the mature cyst, PHB, AR1, and AR2 account for 8, 5.6, and 4.5%, respectively, of the dry weight. Phospholipid formation levels off 4 h postinduction, which coincides with the final cell division, but fatty acids synthesis continues at a very low level throughout encystment, suggesting some turnover of fatty acid. Distribution studies show that AR1 and AR2 are found in roughly equal amounts in the exine and central body of the cysts, with only trace amounts recovered from the intine. Studies of cysts labeled during encystment with [14C]beta-hydroxybutyrate or during vegetative growth with [14C]glucose suggest that the exine structure is synthesized during encystment, but that the intine is composed largely of vegetative cell components.  相似文献

Knowledge of the fine structural organization, molecular composition and permeability properties of the cell surface of intestinal protozoan cysts is important to understand the biologic basis of their resistance. Recent studies on the biology of the cyst walls of Entamoeba histolytica and Entamoeba invadens have considerably advanced knowledge on the cellular processes involved in the transport and surface deposition of the main cyst wall components. Using transmission electron microscopy, cytochemistry, scanning electron microscopy and freeze-fracture techniques, we have obtained new information. In mature cysts the permeability of Entamoeba cysts is limited to small molecules not by the cyst wall, but by the plasma membrane, as demonstrated with the use of ruthenium red as an electron-dense tracer. Cell walls of E. histolytica cysts are made up of five to seven layers of unordered fibrils 7-8 nm thick. Alcian blue stains a regular mesh of fibrils approximately 4 nm thick, running perpendicularly to the cyst wall. In addition, abundant ionogenic groups are seen in cyst walls treated with cationized ferritin. In the mature cysts of E. histolytica and E. invadens small cytoplasmic vesicles with granular material were in close contact with the plasma membrane, suggesting a process of fusion and deposition of granular material to the cell wall. The plasma membrane of mature cysts is devoid of intramembrane particles when analyzed with the freeze-fracture technique. When viewed with scanning electron microscopy the surface of E. histolytica cysts clearly differs from that of Entamoeba coli and E. invadens.  相似文献

The freshwater dinoflagellate Gloeodinium montanum Klebs (1912) was examined with transmission and scanning electron microscopy. Micrographs of ultrathin sections revealed a series of membrane layers rather than the usual dinoflagellate theca in vegetative cysts and in legates. Swarmers had distinct pellicles but appeared to be devoid of thecal plates and vesicles. The organization of cysts and swarmers appeared remarkably similar. All cell types had typical dinoflagellate nuclei with condensed chromosomes. Chloraplasts had girdle lamellae. One pyrenoid per cell was also present in chloroplasts of vegetative cysts. Starch grains and oil globules were distributed throughout the cytoplasm. Large accumulation bodies and polyvesicular vacuoles were found in aging cysts. Trichocysts and flagellar hairs were absent. Two types of intra-cellular prokaryotic organisms were discovered.  相似文献

The fine structure of Azotobacter vinelandii has been studied by means of electron microscopy of ultrathin sections made of the encysting and germinating cells. The organisms were fixed with KMnO4 and embedded in epoxy resin. On an encystment medium the rod-shaped bacteria begin to assume an almost spherical form and then bark-like exine appears in 1½ to 2 days. The exine thickens and an electron permeable intine forms between it and the shrinking cell body. In 5 days the intine makes up more than half of the cyst volume and begins to show a definite two-layered structure. Meanwhile the peripheral bodies, which may be extensions of the cell membrane of the vegetative cell, disappear as the encystment progresses. The cell wall and membrane of the vegetative cell remain demonstrable as the confining structure of the shrinking central body of the mature cyst. In this central body lipoidal globules appear together with aggregations of nuclear material. Cyst germination begins with an increase in the size of the central body at the expense of the intine. The nuclear aggregations become more diffuse and the lipoidal globules disappear. The exine may be pushed outward and the bark-like fragments separate as the emerging vegetative cell develops. Invagination of the cell wall and membrane may occur at this stage leading to cell division. Empty exines remain as horseshoe-shaped structures.  相似文献

Summary In the mature microspore ofSecale cereale, a set of wall ingrowths deposited as the first (outer) intine layer between exine and the microspore plasma membrane, are revealed by electron microscopy. The wall ingrowths form a girdle in the vicinity of the apertural region at the external pole of microspore which is in contact with the tapetum, so the microspore can be considered as a transfer cell which is polarized. After microspore division the second (inner) intine layer is deposited by the vegetative cell and forms a labyrinth of branched wall ingrowths. As a result, the periphery of a vegetative cell is also irregular and appears as very thin plasmatubules or evaginations delimited by plasma membrane and penetrating the pollen wall.The possible functions of the microspore as a transfer cell and the wall-membrane system of the vegetative cell are discussed.  相似文献

Summary The mature pollen of Larix leptolepis Gord. (Conifer) contains five different cell types, and the plasma membrane of the vegetative cell is continuous and organized. The pollen wall is composed of two morphologically and cytochemically distinct domains: the exine and the intine. In the multilayered exine, the ektexine appears granular and the endexine, lamellar. The intine is thick and bilayered with a microfibrillar structure occupying its inner portion. Cytochemical reactions of the exine and the intine are similar to those found in angiosperms. Pollen wall involvement in the male female recognition system is discussed with respecl to the angiosperms.  相似文献

The composition of ionogenic groups and ion-exchange capacity were studied in the polymeric matrix of cell walls isolated from the pollen grain and tissues of vegetative organs (leaves and stems) of Lilium longiflorum Thunb. The ion-exchange capacity was evaluated at different pH values and ionic strength of 100 mM. In the two-layered pollen wall and the somatic cell walls four types of ionogenic groups were found: amino groups, two carboxyl groups (represented by residues of uronic and hydroxycinnamic acids), and phenolic OH-groups. The groups of all four types are present in the intine, whereas the exine contains one type of anion-exchange and two types of cation-exchange groups. The contents of each type group and their ionization constants were determined. The qualitative and quantitative compositions of structural polymers of the pollen intine and somatic cell walls are significantly different. It is suggested that hydroxycinnamic acids should be involved in cross-linking of polysaccharide chains in both the intine and somatic cell primary walls, and such cross-links play a crucial role in the structural organization and integrity of the pollen grain wall.  相似文献

We have previously reported that precursor-accumulating (PAC) vesicles found exclusively in developing seeds are involved in a transport of seed storage proteins, such as 2S albumin, from the endoplasmic reticulum to protein-storage vacuoles. Here, we constructed chimeric genes that encode fusion proteins consisting of both various lengths of polypeptides derived from pumpkin 2S albumin and a selectable marker enzyme, phosphinothricin acetyltransferase. The chimeric genes were expressed in transgenic Arabidopsis in order to investigate the mechanism of the PAC vesicle formation. A fusion protein expressed by one of the chimeric genes is accumulated as a proprotein-precursor form, and localized in novel vesicles of vegetative cells. The vesicles show distinct features that well much to the PAC vesicles. Despite of the accumulation of the fusion protein, the transgenic Arabidopsis is still sensitive to phosphinothricin. Phosphinothricin acetyltransferase contained in the fusion protein is obviously compartmentalized in the PAC-like vesicles that do not permit the detoxification of this herbicide. These results indicate that the PAC-like vesicle can be induced in vegetative cells by the ectopic expression of the protein that is destined to be compartmentalized into the PAC vesicles.  相似文献

用乙二醇甲基丙烯酸脂(简称GMA)和环氧树脂Epon812包埋的薄切片方法对水稻成熟花药和花粉的结构进行了观察,并对各种结构的性质和细胞中的后含物做了细胞化学的分析.对成熟花药的绒毡层膜及乌氏体的研究采用了分离技术,做了显微和超微观察.证明水稻成熟花药壁和花粉除具一般禾本科植物特征外,还揭示了花药壁表皮上可能有硅质,药壁表皮细胞内含有脂类颗粒,药室内壁具纤维素质的纤维状加厚;发现花粉粒中除了贮存有大量淀粉颗粒外,还含有脂类,成熟花粉中营养核与两个精细胞及两个精细胞间联系紧密;并讨论了薄切片的优越性,绒毡层膜的意义及其上细胞印迹的来源.  相似文献

The monocolpate pollen grain of Narcissus pseudonarcissus L.has two preferred sites for tube emergence, one at each endof the colpus. While the cellulosic microfibnls of the innerlayer of the intine are disposed circumferentially in the centreof the grain, the microfibrils in these terminal sites are shorterand randomly oriented Soon after the beginning of hydration,inclusions of the vegetative cell begin movement, firstly ina rotatory manner, and then in a pattern focused on one or bothgermination sites, where the intine bulges as hydration progresses.These changes are associated with the evolution of the actincytoskeleton. Actin is present in the unactivated grain in theform of fusiform bodies. During hydration these dissociate toform finer fibrils, initially randomly disposed. Then, correlatedwith the change of the pattern of movement in the vegetativecell, the actin fibril system becomes polarized towards thegermination sites, where shorter fibrils accumulate. Callose,absent from the ungerminated grain, is deposited within thecellulosic wall in these locations, forming a shallow dome whicheventually develops into an annulus subtending the inner calloselining of the emerging tube. The transition to cylindrical growthis associated firstly with the development of zonation in thecytoplasm of the vegetative cell, with the tip occupied by apopulation of wall precursor bodies (P-particles) and a denseaggregate of short actin fibrils; and then with the establishmentof the ‘inverse fountain’ pattern of movement characteristicof the apical part of the extending tube. The structural organization of the high affinity receptor for immunoglobulin E has been investigated in plasma membrane vesicles from rat basophilic leukemia cells using lactoperoxidase-catalyzed 125I-iodination to label exposed polypeptide regions. Intact vesicles are predominantly right-side-out in orientation, and lactoperoxidase iodination of these vesicles results in labeling of the alpha subunit of receptor but not the beta and gamma subunits. Lysis of these vesicles to expose the cytoplasmic face of the membrane by two different methods permits labeling of the beta and gamma subunits with no increase in labeling of alpha. The results indicate that both the beta and gamma subunits of the receptor have segments exposed at the cytoplasmic side of the plasma membrane. These studies have also revealed a previously unidentified IgE binding component in the membrane vesicles; its 125I-labeling characteristics and some other properties are described.  相似文献

We studied the response of the pollen grain wall to the passage of materials from the external environment. Pollen grains of Lycopersicon esculentum, a species not previously investigated in similar studies, were used to determine whether the routes of passage through the pollen wall are a characteristic of a given species. We found lanthanum deposits on the cell wall surface, inside microchannels crossing tectum, and in the infratectum. Lanthanum was also seen as a fine precipitate homogenously scattered within the substance of the exine. In the apertural region of the intine, electron-dense deposits filled the tubulations of the intine oncus, whereas the intine matrix was free of precipitate. In the cytoplasm, precipitate was observed inside small vesicles frequently located near or in contact with the plasmalemma. Our findings were similar to those reported in other species, and were not influenced by the fixative or culture medium used to incorporate the tracer element. We noted a relation between the route of passage and the stage of pollen grain maturation. In less mature grains (midbicellular pollen), electron-dense deposits were more abundant than in more mature grains, probably because apertural regions were blocked as routes of passage.  相似文献
