Cell surface glycoproteins apparently influence cell interactions, morphogenesis and the course of cellular differentiation. We have therefore investigated the biosynthesis of glycoproteins in 8-cell mouse embryos by using 1.0 μg tunicamycin/ml, a specific inhibitor of glycosylation of N-glycosidically linked glycoproteins. The antibiotic had little effect on the incorporation of leucine into polypeptides, but the incorporation of glucosamine and mannose was inhibited by about 60% with a marked reduction in their incorporation into the majority of the glycopeptides as analysed on polyacrylamide gels. The binding of concanavalin A (conA) and peanut lectin to the embryonic cell surface was also markedly diminished by tunicamycin. However, the binding of peanut lectin to isolated blastomeres displayed a polar distribution with predominant binding to the outer apical surface in all cases, despite a marked reduction in microvilli. Hence tunicamycin has no substantial effect on the molecular distribution of at least some cell surface antigens. Analysis of iodinated cell surface components showed that two components of mol, wt <68 000 and >165 000 were inhibited by tunicamycin. Whereas embryos in the control group underwent compaction and blastulation, those in the experimental group remained uncompacted, although cleavage continued to about the 32-cell stage. However, some embryos initially underwent partial compaction but later decompacted in the presence of tunicamycin; numerous adherens and possibly a few gap junctions were also detected between blastomeres. We suggest that a number of cell surface antigens including N-glycosidically linked glycoproteins may be engaged sequentially during compaction.  相似文献

During preimplantation mouse embryo development expression of Cdx2 is induced in outer cells, which are the trophectoderm (TE) precursors. The mechanism of Cdx2 upregulation in these cells remains unclear. However, it has been suggested that the cell position and polarization may play a crucial role in this process. In order to elucidate the role of these two parameters in the formation of TE we analyzed the expression pattern of Cdx2 in the embryos in which either the position of cells and the time of polarization or only the position of cells was experimentally disrupted. Such embryos developed from the blastomeres that were isolated from 8-cell embryos either before or after the compaction, i.e. before or after the cell polarization took place. We found that in the embryos developed from polar blastomeres originated from the 8-cell compacted embryo, the experimentally imposed outer position was not sufficient to induce the Cdx2 in these blastomeres which in the intact embryo would form the inner cells. However, when the polarization at the 8-cell stage was disrupted, the embryos developed from such an unpolarized blastomeres showed the increased number of cells expressing Cdx2. We found that in such experimentally obtained embryos the polarization was delayed until the 16-cell stage. These results suggest that the main factor responsible for upregulation of Cdx2 expression in outer blastomeres, i.e. TE precursors, is their polarity.  相似文献

Summary Blastomeres isolated from 8-and 16-cell embryos (that is 1/8 and 1/16) show a smooth surface at their point of contact with other blastomeres and a microvillous free surface. Microvilli reappear completely on the smooth surface of 52% of 1/8 embryos and partially on 88% of 1/16 embryos if cultured in vitro for 6 h. When 2-to 8-cell embryos are aggregated to 8-cell embryos and forced apart after 1–3 h, the contact surface of the 8-cell embryos has become smooth. Fixed 8-cell embryos are also able to induce complete disappearance of microvilli on the contact surface of a living 8-cell embryo. Embryos having more than 8 cells do not induce complete disappearance of microvilli on the contact surface of 8-cell embryos. Aggregates of late morulae do not show complete disappearance of microvilli at their contact surfaces but rather a loosening of their peripheral blastomeres.Our results show that isolated 1/8 and 1/16 embryos tend to recover from regionalization, that the process of aggregation of embryos having 8 cells or less is similar to compaction and that embryos having more than 8 cells seem to aggregate by cell sorting. The processes of compaction, adhesion and reassortment are briefly discussed. We submit that blastomere regionalization, which depends on cell to cell contact, may be the spatial basis of embryonic regulation and of the inside-outside normal differentiation of early mouse embryos.  相似文献

The preimplantation development of the mouse embryo leads to the divergence of the first two cell lineages, the inner cell mass and the trophectoderm. The formation of a microvillus pole during compaction at the eight-cell stage and its asymmetric inheritance during mitosis are key events in the emergence of these two cell populations. Ezrin, a member of the ERM protein family, seems to be involved in the formation and stabilization of this apical microvillus pole. To further characterize its function in early development, we mutated the key residue T567, which was reported to be essential for regulation of ezrin function through phosphorylation. Here, we show that expression of ezrin mutants in which the COOH-terminal threonine T567 was replaced by an aspartate (to mimic a phosphorylated residue; T567D) or by an alanine (to avoid phosphorylation; T567A) interferes with E-cadherin function and disrupts the first morphogenetic events of development: compaction and cavitation. The active mutant ezrin-T567D induces the formation of numerous and abnormally long microvilli at the surface of blastomeres. Moreover, it localizes all around the cell cortex and inhibits cell-cell adhesion and cell polarization at the eight-cell stage. During the following stages, only half of the embryos are able to compact and finally to cavitate. In those embryos, the amount of ezrin-T567D decreases in the basolateral areas, while the proportion of adherens junctions increases. The reverse inactive mutant ezrin-T567A is mainly cytoplasmic and does not perturb compaction at the eight-cell stage. However, at the 16-cell stage, it relocalizes at the basolateral cortex, leading to a strong decrease in the surface of adherens junctions, and finally, embryos abort development. Our results show that ezrin is directly involved in the formation of microvilli in the early mouse embryo. Moreover, they indicate that maintenance of ezrin in basolateral areas prevents microvilli breakdown and inhibits the formation of normal cell-cell contacts mediated by E-cadherin, thereby impairing blastomeres polarization and morphogenesis of the blastocyst.  相似文献

Eight-cell mouse embryos undergo a morphogenetically significant process termed compaction during which blastomeres flatten against one another maximizing their areas of cell-cell contact. Data presented here show striking differences (P less than 0.01) in Con A-induced agglutination between groups of uncompacted and compacted embryos suggesting that the compaction process is accompanied by changes in blastomere surface properties.  相似文献

In a normal, intact sea urchin embryo blastomeres are structurally polarized so that all microvilli and cortical "pigment granules" are situated at the apical surfaces facing the hyaline layer and are absent from basolateral surfaces facing adjacent blastomeres and the internal embryonic cavity. To test the roles of intercellular contacts and the hyaline layer in the process of establishing this blastomere polarity, these two factors were experimentally eliminated; sea urchin eggs of four species were denuded of the nascent hyaline layer soon after fertilization and then cultured in calcium-free artificial seawater to prevent subsequent intercellular adhesion and contact. Such free blastomeres divided normally and still developed polarized distributions of microvilli and pigment granules resembling those of the corresponding blastomeres in intact embryos. These results indicate that the process of polarization is intrinsic to individual blastomeres (self-polarization) and that neither intercellular contacts nor adhesion of microvilli to the hyaline layer is necessary. The precise temporal and spatial coincidence of the patterns of polarization and the division cycles further suggests that a mechanistic link is maintained among cell division, blastomere polarization, and probably also a heritable component of the animal-vegetal axis.  相似文献

The process of cell polarization in mouse 8-cell embryos includes the formation of a polar cluster of cytoplasmic endocytotic organelles (endosomes) subjacent to an apical surface pole of microvilli. A similar polar morphology, supplemented by basally localized secondary lysosomes, is evident following division to the 16-cell stage in outside blastomeres, precursors of the trophectodermal lineage. The roles of microfilaments and microtubules in generating and stabilizing endocytotic and surface features of polarity (visualized by horseradish peroxidase incubation and indirect immunofluorescence labeling, respectively) have been evaluated by exposure of 8- and 16-cell embryos and 8-cell couplets to drugs (cytochalasin D, colcemid, nocodazole) that disrupt the cytoskeleton. The generation of endocytotic polarity is dependent upon intact microtubules and microfilaments, but the newly established endocytotic pole in blastomeres from compacted 8-cell embryos appears to be stabilized exclusively by microtubules. Polarized endocytotic organelles at the 16-cell stage are more resistant to drug treatment than at the 8-cell stage (probably due to microfilament interactions) indicating a maturation phase in the polar cell lineage. Microtubules are also responsible for the orientation of endocytotic clusters along the cell's axis of polarity. In contrast, the generation and stability of polarity at the cell surface appears relatively independent of cytoskeletal integrity. The results are discussed in relation to the mechanisms that may control the development and stabilization of polarization during cleavage.  相似文献

Membrane topography and organization of cortical cytoskeletal elements and organelles during early embryogenesis of the mouse have been studied by transmission and scanning electron microscopy with improved cellular preservation. At the four- and early eight-cell stages, blastomeres are round, and scanning electron microscopy shows a uniform distribution of microvilli over the cell surface. At the onset of morphogenesis, a reorganization of the blastomere surface is observed in which microvilli becomes restricted to an apical region and the basal zone of intercellular contact. As the blastomeres spread on each other during compaction, many microvilli remain in the basal region of imminent cell-cell contacts, but few are present where the cells have completed spreading on each other. Microvilli on the surface of these embryos contain linear arrays of microfilaments with lateral cross bridges. Microtubules and mitochondria become localized beneath the apposed cell membranes during compaction. Arrays of cortical microtubules are aligned parallel to regions of apposed membranes. During cytokinesis, microtubules become redistributed in the region of the mitotic spindle, and fewer microvilli are present on most of the cell surface. The cell surface and cortical changes initiated during compaction are the first manifestations of cell polarity in embryogenesis. These and previous findings are interpreted as evidence that cell surface changes associated with trophoblast development appear as early as the eight-cell stage. Our observations suggest that morphogenesis involves the activation of a developmental program which coordinately controls cortical cytoplasmic and cell surface organization.  相似文献

During the process of compaction, mouse 8-cell blastomeres flatten upon each other and polarize along an axis perpendicular to cell contacts. If the process of flattening is prevented, polarization can still occur, but does so in a lower proportion of cells than for control populations, and without the normal contact-directed orientation. We compared contact-directed and noncontact-directed processes to see if they involve common mechanisms. In nonflattened cells, surface polarization was favored in cells with nuclei located close to the cell surface, and the positions of surface poles and of nuclei tended to coincide. We present evidence that microtubules are involved in the development of microvillous poles associated with nuclei. In contrast it is known that polarization of microvilli occurs in the absence of microtubules if blastomeres are allowed to flatten. We conclude that surface polarization of mouse blastomeres can be accomplished by at least two alternative routes. One requires flattening but is independent of microtubules, and another can occur without flattening but involves a microtubule-mediated interaction between the nucleus and the cell cortex. It seems that both these pathways operate in the undisturbed embryo.  相似文献

The effects of phorbol myristate acetate (PMA) and other activators of protein kinase C on the cytoskeletal organization of mouse oocytes and early embryos have been examined. The effects observed depended on the developmental stage on exposure to PMA. PMA had little effect on the cytoskeletal or microvillous organization of unfertilized oocytes. Interphase cells from embryos prior to compaction showed limited disruption and loss of microvilli when exposed to PMA and foci of polymerized actin remained visible in the cytocortex of embryos up to the early 8-cell stage. When compacted late 8-cell embryos were exposed to PMA, most microvilli were lost and little polymerized actin remained in the cytocortex. PMA also caused loss of microtubules from compact 8-cell embryos under some experimental conditions. Intercellular flattening was both prevented and reversed. The relevance of these observations to the rearrangement of cell-cell contacts and cytoskeletal organization seen during compaction at the 8-cell stage is discussed and a possible role for protein kinase C in the generation of cell polarity proposed.  相似文献

Abstract. The organization of the cytoskeleton during early pig embryogenesis was investigated by using fluorescence and electron microscopy. The early morphogenesis of the pig embryo differed from that of the mouse, the standard model of the early mammalian development. In the pig, both compaction and polarization were gradual, and definitive polarization of cell surface microville occurred first shortly before blastocyst formation; the compaction and polarization of the mouse embryo are completed as early as at the 8 cell stage. Furthermore, the pig morula undergoes cycles of compaction and decompaction throughout its development. Distinct changes in the distribution of actin and the actin-associated proteins α-fodrin, vinculin and E-cadherin coincided with these events. In the pig, all these molecules were evenly distributed at all aspects of the blastomeres during early cleavage and then gradually accumulated in regions of intercellular contacts toward the blastocyst stage; microfilaments in trophectoderm cells formed a cortical meshwork associated with apical microvilli and adherent junctions (zonula adherens). In the mouse, the corresponding changes occur earlier, at the 8 cell stage. Microtubules formed a network-like cortical layer beneath the microvilli at the free outer surfaces of pig blastomeres. Cytokeratin bundles were not observed until the early blastocyst, where they characteristically associated with newly formed desmosomes.
In both species a close correlation between morphologically defined developmental stages and the organization of the cytoskeleton: actin and actin-associated proteins are involved in polarization and compaction, whereas the appearance of intermediate filament bundles coincides with the building of the first epithelium, the trophectoderm; it is in the timing of events that a contrast between species is observed.  相似文献

In the eggs of a wide range of animal species, various factors that determine the blastomeres' presumptive fate are known to locate unevenly within the egg. In the embryos of these animals, cleavage occurs not just to increase cell numbers, but also to distribute the factors to the respective blastomeres, resulting in cell specialization at the later stages. In the early cleavage stages, before the establishment of a device such as desmosomes to directly join the blastomeres, some other means is needed to keep the blastomeres together and maintain the relative positions among them. In this study, we found that the embryos of the starfish Astropecten scoparius lack the hyaline layer seen in sea urchin embryos and that blastomeres adhere to the fertilization envelope (FE) via filamentous cellular projections (fixing processes). Electron microscopy revealed the fixing processes to be specialized microvilli formed, after the elevation of the FE, by the elongation of short microvilli that pre-exist in unfertilized eggs. After the first cleavage, the two blastomeres separate from each other and finally attach to the FE. In the subsequent cleavages, the blastomeres undergo repeated cell division without separating from the FE. Between the blastomeres and the FE, only shortened fixing processes were observed. Destruction of the fixing processes caused release of the blastomeres from the FE and disturbance of the relative positions of the blastomeres, resulting in abnormal development of the embryos. These observations suggest that the fixing process is a device to keep the egg placed centrally in the FE up to the first cleavage, and after the first cleavage and beyond to anchor the blastomeres to the FE so that the FE can be used as a scaffold for morphogenesis. Electron microscopy also suggests that the inner layer of the FE, which is derived from the contents of cortical granules, reinforces the adhesion of the fixing processes to the FE. Immuno-electron microscopy, using an antibody against sea urchin hyaline layer, showed that the inner layer of the FE of starfish eggs and the hyaline layer of sea urchin eggs, which are both derived from cortical granules, contain some common elements.  相似文献

At the eight-cell stage, the blastomeres of the preimplantation mouse embryo undergo a dramatic shape change, compaction, which is considered essential to the future segregation of presumptive cell types. This investigation demonstrates that compaction 1) occurs in vivo, 2) is accompanied by the formation of tight and gap junctions and 3) can be reversibly inhibited in vitro by calcium-depleted medium and also by cytochalasin B (CCB). Although microtubules frequently are observed in cortical regions where compaction is proceeding, colcemid and colchicine have no inhibitory effect.Calcium-free medium and CCB dissociate compacted embryos, over 50% of which recover in normal medium in 3 and 0.25 hr, respectively. The Ca2+ threshold for compaction is approximately 0.1–0.2 mM and may be required for normal intercellular adhesions. Since compaction marks the beginning of tight junction formation and provides the necessary cell-to-cell apposition for the development of the zonula occludens at the morula stage, it is considered to be the initial step in blastocyst morphogenesis. In addition, this investigation provides the means to reprogram compaction by reversible inhibition and thereby study theories of cell determination.  相似文献

Summary Early (non-compacted) and late (compacted) 8-cell embryos were observed after few hours of culture in vitro. The former embryos underwent compaction and the latter embryos were found decompacted. Cell counting suggested that decompaction preceded fourth cleavage division of any blastomere and lasted until the blastomeres divided.About one third of mouse morulae, which had about twenty cells, were found non-compacted upon obtaining from females. After few hours of culture in vitro these embryos underwent recompaction and cavitation. Increasing the contributions of mitosis-arrested and cytokinesisarrested cells within the morulae by culture with nocodazole and cytochalasin B respectively, did not delay recompaction.The data show that periods of decompaction and recompaction alternate in preimplantation development.  相似文献

Development of cellular polarity is an important event during early mammalian embryo development and differentiation. Blastomeres of hamster embryos at various stages were examined by scanning electron microscopy (SEM) and immunocytochemical staining. SEM observations revealed that 1- to 7-cell-stage embryos showed a uniform distribution of microvilli throughout the cell surface. Microvillous polarization was initially noted in the blastomeres (10-35%) of 8-cell-stage embryos. The polarized microvilli were observed mostly in the basal region of cell-cell contact and occasionally at the apical, outward-facing surface of the blastomere. Fluorescein-isothiocyanate-conjugated concanavalin A failed to reveal any polarity in the blastomeres regardless of the stages of the embryos. Actin staining showed that microfilaments were present beneath the cell surface, and in addition, areas of cell contact were more heavily stained, indicating a thick microfilament domain. Microtubules were located throughout the cytoplasm and were heavily concentrated near the nucleus during interphase, although they became redistributed in the region of the mitotic spindle during karyokinesis. The position of nucleus changed from the cell center to the apical, outward-facing surface of the cell, and it distanced itself from the basal microvillous pole. It is suggested that the changes in the cell surface and nuclear position are the first manifestations of cell polarity in peri-compacted hamster embryos, which appear as early as the 8-cell stage; furthermore, the outward migration of the nuclei may parallel the redistribution of microtubules in the cytoplasm.  相似文献

During mouse blastocyst formation, a layer of outer cells differentiates in less than 48 h into a functional epithelium (the trophectoderm). Ezrin, an actin-binding structural component of microvilli in epithelial cells, is also involved in signal transduction and ionic pump control. In the mouse embryo, ezrin becomes restricted to the apical cortex of all blastomeres at compaction and of outer cells at later stages. Here we investigated the function of ezrin in living embryos during epithelial differentiation using mutant forms of ezrin tagged with green fluorescent protein (GFP). GFP-tagged wild-type ezrin (Ez/GFPc) behaved like endogenous ezrin and did not interfere with development. Deletion of the last 53 amino acids (Delta53/GFP) changed the localization of ezrin: after compaction, Delta53/GFP remained associated with the apical and basolateral cortex in all blastomeres, and its expression slightly disturbed the cavitation process. Finally, full-length ezrin with GFP inserted at position 234 (Ez/GFPi) was localized all around the cortex throughout development, although it was concentrated at the apical pole after compaction. In embryos expressing Ez/GFPi, the duration of the 16-cell stage was reduced, while the onset of cavitation was delayed. Moreover, cavitation was abnormal, and the blastocoele was small and retracted almost completely several times as if there were major leakages of blastocoelic fluid. Our results suggest that, in addition to its role in microvilli organization, ezrin is involved in the formation of a functional epithelium through a still unknown mechanism.  相似文献

We have examined the synthesis and distribution of the cell adhesion molecule uvomorulin in mouse preimplantation embryos. Uvomorulin can already be detected on the cell surface of unfertilized and fertilized eggs but is not synthesized in these cells. Uvomorulin synthesis starts in late two-cell embryos and seems not to be correlated with the onset of compaction. The first signs of compaction are accompanied by a redistribution of uvomorulin on the surface of blastomeres. During compaction uvomorulin is progressively removed from the apical membrane domains of peripheral blastomeres. In compact morulae uvomorulin is no longer present on the outer surface of the embryo but is localized predominantly in membrane domains involved in cell-cell contacts of adjacent outer blastomeres. On inner blastomeres of compact morulae uvomorulin remains evenly distributed. This uvomorulin distribution once established during compaction is maintained and also found in the blastocyst: on trophectodermal cells uvomorulin localization is very similar to that in adult intestinal epithelial cells while uvomorulin remains evenly distributed on the surface of inner cell mass cells. The possible role of the redistribution of uvomorulin for the generation of trophectoderm and inner cell mass in early mouse embryos is discussed.  相似文献
