The relationship between Na+, major cation concentrations and salt tolerance under long-term saline conditions of Medicago arborea and Medicago citrina was studied. Plants were grown in solution culture in 1, 50, 100, or 200 mmol/L NaCl for 30 days in a climate-controlled greenhouse. Stem and petiole growth was the most affected by salt in both species. Leaf growth was inhibited in M. arborea, with increased salt, while only the 200 mmol/L NaCl-treated M. citrina plants were significantly affected. Both species had the highest Na+ concentrations in the shoots, however, the allocation pattern was different; M. arborea showed the highest concentrations in the leaf blades, whereas M. citrina distributed the salt into the petioles. K+/Na+ ratio decreased with salt in both species; however, leaf K+ use efficiency (g leaf DW mg-1 leaf K+) was higher in M. citrina. The difference in Na+ allocation and cation concentrations found in these medic species and their importance is discussed in relation to their response to NaCl salinity.

Long‐term salt effects on plant growth have often been related to direct ion toxicity due to the accumulation of high ion concentrations in plant tissue. This work examines the relative importance of endogenous ABA, as well as Na+ and Cl toxicity, in the inhibition of leaf growth and photosynthesis, in bean plants grown at 1, 25, 50 and 75 m M NaCl until the fruit‐bearing stage. All salt‐treated plants showed very high leaf Cl concentrations, with little difference between plants exposed to 50 or 75 m M NaCl. The 25 and 50 mM salt‐treated plants were able to successfully exclude Na+ from their leaves, and only suffered an initial decline in the rate of leaf growth. Plants exposed to 75 m M NaCl showed an increase in Na+ leaf concentrations with an accompanying decrease in growth and photosynthesis as salt exposure progressed. A high correlation was found between leaf Na+ and leaf growth. Leaf ABA significantly increased with salt supply, and was highly correlated with both leaf Na+ and leaf growth. Our results suggest that in bean plants under long‐term salt stress, leaf ABA may participate in the regulation of leaf growth, and leaf Na+ would be at least partly responsible for increased ABA levels.

This paper describes the physiological effects of abscisic acid (ABA) and 100 mM NaCl on citrus plants. Water potential, leaf abscission, ethylene production, photosynthetic rate, stomatal conductance, and chloride accumulation in roots and leaves were measured in plants of Salustiana scion [Citrus sinensis (L) Osbeck] grafted onto Carrizo citrange (Citrus sinensis [L.] Osbeck × Poncirus trifoliata [L.] Raf) rootstock. Plants under salt stress accumulated high amounts of chloride, increased ethylene production, and induced leaf abscission. Stomatal conductance and photosynthetic rates rapidly dropped after salinization. The addition of 10 mM ABA to the nutrient solution 10 days before the exposure to salt stress reduced ethylene release and leaf abscission. These effects were probably due to a decrease in the accumulation of toxic Cl- ions in leaves. In non-salinized plants, ABA reduced stomatal conductance and CO2 assimilation, whereas in salinized plants the treatment slightly increased these two parameters. The results suggest a protective role for ABA in citrus under salinity.

Tolerance of salt stress in potato (Solanum tuberosum L.) increased when the plants were pre-exposed to low concentrations of salt (salt acclimation). This acclimation was accompanied by increased levels of abscisic acid (ABA) in the shoot. To further study the role of roots and shoots in this acclimation process, reciprocal grafts were made between a salt-tolerant (9506) and salt-sensitive ABA(−) mutant and its ABA(+) normal sibling potato genotype. The grafted plants were acclimated with 75 or 100 mM NaCl for 3 weeks and then exposed to 150–180 mM NaCl, depending on the salt tolerance of the rootstock. After 2 weeks of exposure to the salt stress, the acclimated and unacclimated plants were compared for physiologic and morphologic parameters. The response to the salt stress was strongly influenced by the rootstock. The salt-tolerant 9506 rootstock increased the salt tolerance of scions of both the ABA-deficient mutant and its ABA(+) sibling. This salt tolerance induced by the rootstock was primarily modulated by salt acclimation and manifested in the scion via increased plant water content, stem diameter, dry matter accumulation, stomatal conductivity, and osmotic potential, and is associated with a reduction in leaf necrosis. There was also a pronounced scion effect on the rootstock. Using 9506 as a scion significantly increased root fresh and dry weights, stem diameter, and root water content of ABA(−) mutant rootstocks. Specific evidence was found of the role of exogenous ABA in the enhancement of water status in grafted plants under salt stress beyond that of grafting alone. This was verified by more positive stomatal conductivity and upward water flow in ABA-treated grafted and nongrafted plants and the absence of upward water flow in nontreated grafted plants through NMR imaging. Grafting using either salt-tolerant scions or rootstocks with inherently high ABA levels may positively modify subsequent responses of the plant under salt stress.

The short-term growth response to salt of the developing barley leaf
Recent results concerning the short-term growth response to salinity of the developing barley leaf are reviewed. Plants were grown hydroponically and the growth response of leaf 3 was studied between 10 min and 5 d following addition of 100 mM NaCl to the root medium. The aim of the experiments was to relate changes in variables that are likely to affect cell elongation to changes in leaf growth. Changes in hormone content (ABA, cytokinins), water and solute relationships (osmolality, turgor, water potential, solute concentrations), gene expression (water channel), cuticle deposition, membrane potential, and transpiration were followed, while leaf elongation velocity was monitored. Leaf elongation decreased close to zero within seconds following addition of NaCl. Between 20 and 30 min after exposure to salt, elongation velocity recovered rather abruptly, to about 46% of the pre-stress level, and remained at the reduced rate for the following 5 d, when it reached about 70% of the level in non-stressed plants. Biophysical and physiological analyses led to three major conclusions. (i) The immediate reduction and sudden recovery in elongation velocity is due to changes in the water potential gradient between leaf xylem and peripheral elongating cells. Changes in transpiration, ABA and cytokinin content, water channel expression, and plasma membrane potential are involved in this response. (ii) Significant solute accumulation, which aids growth recovery, is detectable from 1 h onwards; growing and non-growing leaf regions and mesophyll and epidermis differ in their solute response. (iii) Cuticular wax density is not affected by short-term exposure to salt; transpirational changes are due to stomatal control.

Ginseng (Panax ginseng) is a typical perennial shade plant. Aim of this study was to investigate the effects of exogenous hormones on photosynthesis of P. ginseng. At different growth stages, the aerial parts of P. ginseng plants were cut at the stem base and they were inserted into the nutrient solutions containing different exogenous hormones. Then the leaf photosynthesis and water absorbing capacity (absorbing water mass) of the excised plants were measured. The results showed that exogenous abscisic acid (ABA) decreased significantly net photosynthetic rate (P N), stomatal conductance, transpiration rate, and absorbed water mass of excised P. ginseng at all growth stages, while both cytokinin (CTK) and indole-3-acetic acid (IAA) enhanced those parameters. Comparing different growth stages, ABA caused more severe inhibition of leaf photosynthesis at the early growth stage, while CTK and IAA showed significant enhancement of leaf photosynthesis at later growth stage. ABA reduced highly intercellular CO2 concentration of P. ginseng at the flowering and green fruit stages, but it had only a small effect at red fruit early and red fruit stages. During the early growth stage, the inhibitory effect of ABA on leaf P N might be caused mainly due to the stomatal limitation. However, the reason for this reduction was complex at the later growth stage and it included stomatal and other factors.

The influence of short-term salinity (day 1–day 2: 50 mol m–3 NaCl, day 3–day 7: 100 mol m–3 NaCl in the nutrient solution) on leaf gas exchange characteristics were studied in two fig clones (Ficus carica L.), whose root mass had been varied in relation to the leaf area. The stomatal conductance was diminished by NaCl in the first week of treatment. NaCl slightly reduced the calculated intercellular partial pressure of CO2. The net photosynthetic rate of plants with many roots was stimulated by NaCl on some days of the first week of treatment, whereas the net assimilation rate of the plants with few roots remained unaltered or decreased by NaCl. Only the assimilation of the salt-treated plants of one clone for some days during the first week of treatment seemed to be influenced by stomatal conductance. Nonstomatal factors were primarily responsible for the changes in CO2 uptake in response to salt and/or root treatment. The water use efficiency increased during several days of the first week of NaCl treatment. Decreased stomatal conductance, increased water use efficiency and stimualtion of the net CO2 assimilation rate appear to enhance salt tolerance during the first few days of salinity. ei]H Lambers

Exposing plants to low VPD reduces leaf capacity to maintain adequate water status thereafter. To find the impact of VPD on functioning of stomata, stomatal morphology and leaf anatomy, fava bean plants were grown at low (L, 0.23 kPa) or moderate (M, 1.17 kPa) VPDs and some plants that developed their leaves at moderate VPD were then transferred for 4 days to low VPD (M→L). Part of the M→L‐plants were sprayed with ABA (abscisic acid) during exposure to L. L‐plants showed bigger stomata, larger pore area, thinner leaves and less spongy cells compared with M‐plants. Stomatal morphology (except aperture) and leaf anatomy of the M→L‐plants were almost similar to the M‐plants, while their transpiration rate and stomatal conductance were identical to that of L‐plants. The stomatal response to ABA was lost in L‐plants, but also after 1‐day exposure of M‐plants to low VPD. The level of foliar ABA sharply decreased within 1‐day exposure to L, while the level of ABA‐GE (ABA‐glucose ester) was not affected. Spraying ABA during the exposure to L prevented loss of stomatal closing response thereafter. The effect of low VPD was largely depending on exposure time: the stomatal responsiveness to ABA was lost after 1‐day exposure to low VPD, while the responsiveness to desiccation was gradually lost during 4‐day exposure to low VPD. Leaf anatomical and stomatal morphological alterations due to low VPD were not the main cause of loss of stomatal closure response to closing stimuli.

Bean plants, Phaseolus vulgaris L. cv. Contender, were grown in the spring and summer seasons to study the relationship between xylem Na+/Cl-, transpiration rate, and salt tolerance. Eight-day-old seedlings were transplanted to 50% modified Hoagland solution with 1 mM NaCl. Four days after transfer, one of two treatments was applied: a control of 1 mM NaCl or a treatment of 25 mM NaCl every two days to reach a final treatment concentration of 75 mM NaCl. Plants were sampled on the fourth day after the final salt concentration was reached, eight days after the salinisation treatment began. Relative growth rate was 2.6-fold greater in summer than in spring. However, while no differences were found between treatments in spring, summer salt-treated plants had growth rates that were 31% lower than those of controls. In summer, CO2 assimilation, stomatal conductance, and transpiration rate of salinised plants declined with respect to controls. Leaf Na+ and trifoliolate leaf Cl- were higher in salt-treated plants in summer, although root Na+ was significantly higher in spring. Moreover, in summer salinity inhibited Ca2+ and K+ uptake and changed its distribution. Summer salt-treated plants had an average of 17-fold higher xylem Na+ during the daily cycle, while xylem Cl-, only in the afternoon, showed higher values (1.5-fold) compared to spring-grown plants. Our results suggest that the faster growth response to salt in summer-grown bean was at least partly due to an increase in xylem Na+ independent of the transpiration rate and possibly related to an increase in xylem Na+ influx or/and Na+ recirculation.

Longan species (Dimocarpus longan Lour.) exhibit a high agronomic potential in many subtropical regions worldwide; however, little is known about its responses to abiotic stress conditions. Drought and salinity are the most environmental factors inducing negative effects on plant growth and development. In order to elucidate the responses of longan to drought and salinity, seedlings were grown under conditions of drought and salt stresses. Drought was imposed by suspending water supply leading to progressive soil dehydration, and salinity was induced using two concentrations of NaCl, 100 and 150 mM in water solution, for 64 days. Data showed that salt concentrations increased foliar abscisic acid (ABA) and only 150 mM NaCl reduced indole-3-acetic acid (IAA) and increased proline levels. NaCl treatments also increased Na+ and Cl? content in plant organs proportionally to salt concentration. Drought increased leaf ABA but did not change IAA concentrations, and also increased proline synthesis. In addition, drought and salt stresses reduced the photosynthesis performance; however, only drought decreased leaf growth and relative leaf water content. Overall, data indicate that under severe salt stress, high ABA accumulation was accompanied by a reduction of IAA levels; however, drought strongly increased ABA but did not change IAA concentrations. Moreover, drought and high salinity similarly increased (or maintained) ion levels and proline synthesis. Data also suggest that ABA accumulation may mitigate the impact of salt stress through inducing stomatal closure and delaying water loss, but did not mediate the effects of long-term drought conditions probably because leaves reached a strong dehydration and the role of ABA at this stage was not effective to detain leaf injuries.

Salinization is one of the most important causes of crop productivity reduction in many areas of the world. Mechanisms that control leaf growth and shoot development under the osmotic phase of salinity are still obscure, and opinions differ regarding the Abscisic acid (ABA) role in regulation of biomass allocation under salt stress. ABA concentration in roots and leaves was analyzed in a genotype of processing tomato under two increasing levels of salinity stress for five weeks: 100 mM NaCl (S10) and 150 mM NaCl (S15), to study the effect of ABA changes on leaf gas exchange and dry matter partitioning of this crop under salinity conditions. In S15, salinization decreased dry matter by 78% and induced significant increases of Na+ and Cl in both leaves and roots. Dry matter allocated in different parts of plant was significantly different in salt-stressed treatments, as salinization increased root/shoot ratio 2-fold in S15 and 3-fold in S15 compared to the control. Total leaf water potential (Ψw) decreased from an average value of approximately −1.0 MPa, measured on control plants and S10, to −1.17 MPa in S15. In S15, photosynthesis was reduced by 23% and stomatal conductance decreased by 61%. Moreover, salinity induced ABA accumulation both in tomato leaves and roots of the more stressed treatment (S15), where ABA level was higher in roots than in leaves (550 and 312 ng g−1 fresh weight, respectively). Our results suggest that the dynamics of ABA and ion accumulation in tomato leaves significantly affected both growth and gas exchange-related parameters in tomato. In particular, ABA appeared to be involved in the tomato salinity response and could play an important role in dry matter partitioning between roots and shoots of tomato plants subjected to salt stress.

Abstract. Maize seedlings ( Zea mays L. John Innes F1 hybrid) were grown in a greenhouse in l-m-long tubes of soil. When the plants were well established, water was withheld from half of the tubes. Control plants were watered every day during the 20-d experimental period. The soil drying treatment resulted in a substantial restriction of stomatal conductance and a limitation in shoot growth, even though there was no detectable difference in the water relations of watered and unwatered plants. From day 7 of the soil drying treatment, xylem ABA concentrations (measured using the sap exuded from detopped plants) were substantially increased in unwatered plants compared to values recorded with sap from plants watered every day. Measurements of water potential through the profile of unwatered soil suggest that xylem ABA concentrations reflects the extent of soil drying. Leaf ABA content was a much less sensitive indicator of the effect of soil drying and during the whole of experimental period there was no significant difference between ABA concentration in leaves of well watered and unwatered plants. In a second set of experiments, ABA was fed to part of the roots of potted maize plants to manipulate xylem ABA concentration. These manipulations suggested that the increases in ABA concentration in xylem sap, which resulted from soil drying, were adequate to explain the observed variation in stomatal conductance and might also explain the restriction in leaf growth rate. These results are discussed in the light of recent work which suggests that stomatal responses to soil drying are partly attributable to an as-yet unidentified inhibitor of stomatal opening.

Nitraria retusa and Atriplex halimus (xero-halophytes) plants were grown in the range 0–800 mM NaCl while Medicago arborea (glycophyte) in 0–300 mM NaCl. Plants were harvested after 120 days of salt-treatment. The present study was designed to study the effect of salinity on root, stem and leaf anatomy, water relationship, and plant growth in greenhouse conditions. Salinity induced anatomical changes in the roots, stems and leaves. The cuticle and epidermis of N. retusa and A. halimus stems were unaffected by salinity. However, root anatomical parameters (root cross section area, cortex thickness and stele to root area ratio), and stem anatomical parameters (stem cross section area and cortex area) were promoted at 100–200 mM NaCl. Indicating that low to moderate salinity had a stimulating effect on root and stem growth of these xero-halophytic species. At higher salinities, root and stem structures were altered significantly, and their percentages of reduction were higher in A. halimus than in N. retusa whereas, in M. arborea, they were strongly altered as salinity rose. NaCl (100–300 mM) reduced leaf water content by 21.2–56.2% and specific leaf area by 51–88.1%, while increased leaf anatomical parameters in M. arborea (e.g. increased thickness of upper and lower epidermis, palisade and spongy mesophyll, entire lamina, and increased palisade to spongy mesophyll ratio). Similar results were evidenced in A. halimus leaves with salinity exceeding 100 mM NaCl. Leaves of N. retusa were thinner in salt-stressed plants while epidermis thickness and water content was unaffected by salinity. The size of xylem vessel was unchanged under salinity in the leaf’s main vein of the three species while we have increased number in M. arborea leaf main vein in the range of 200–300 mM NaCl. A longer distance between leaf vascular bundle, a reduced size and increased number of xylem vessel especially in stem than in root vascular system was evidenced in M. arborea treated plants and only at (400–800 mM) in the xero-halophytic species. The effects of NaCl toxicity on leaf, stem and root ultrastructure are discussed in relation to the degree of salt resistance of these three species. Our results suggest that both N. retusa and A. halimus show high tolerance to salinity while M. arborea was considered as a salt tolerant species.

The effects of salinity (400 mM NaCl) on growth, biomass partitioning, photosynthesis, and leaf ultrastructure were studied in hydroponically grown plants of Aeluropus littoralis (Willd) Parl. NaCl produced a significant inhibition of the main growth parameters and a reduction in leaf gas exchange (e.g. decreased rates of photosynthesis and stomatal conductance). However, NaCl salinity affected neither the composition of photosynthesis pigments nor leaf water content. The reduction in leaf gas exchange seemed to correlate with a decrease in mesophyll thickness as well as a severe disorganisation of chloroplast structure, with misshapen chloroplasts and dilated thylakoid membranes. Conspicuously, mesophyll chloroplasts were more sensitive to salt treatment than those of bundle sheath cells. The effects of NaCl toxicity on leaf structure and ultrastructure and the associated physiological implications are discussed in relation to the degree of salt resistance of A. littoralis.

A two-chamber-system was used to study whole-plant gas exchange responses of Spartina alterniflora to long-term and transient salinity treatments over the range of 5 to 40 ppt NaCl. Lower photosynthetic rates, leaf water vapor conductances, belowground respiration rates, and higher aboveground respiration rates in plants adapted to 40 ppt NaCl were observed. Area-specific leaf weight increased with salinity, although the salt content of leaf tissues did not. A reduced rate of gross photosynthesis and higher aboveground respiration rate in 40-ppt NaCl plants significantly lowered the net whole-plant CO2 gain below that of 5-ppt NaCl plants, while the net CO2 gain of 25-ppt NaCl plants was intermediate. Within 6 hr of increasing the salinity of 5- and 25-ppt NaCl plants by 20 and 15 ppt NaCl, S. alterniflora responded by reducing leaf water vapor conductance, which in turn reduced the photosynthetic rate. This response was reversed by returning the plants to their original salinity, which indicates that S. alterniflora adjusts water loss and gas exchange in response to transient salinity stress by regulating stomatal aperture. On the other hand, decreasing salinity of the growth media of plants cultured at 25 and 40 ppt NaCl had little or no effect on gas exchange characteristics. This suggests that S. alterniflora adapts to constant salinity through fixed, salinity-dependent structural modifications, such as stomatal density.

Plants of Tagetes erecta L. (marigold) cultivated in vitro in ventilated containers exhibited greater control of leaf water loss and increased survival in the field than plants cultivated in sealed containers. Increased field survival of plants cultivated in ventilated containers was attributed to higher levels of endogenous abscisic acid (ABA). Therefore, ABA was supplied exogenously to plants in sealed or ventilated containers by adding ABA (10(-6), 10(-5), 10(-4) M) to the in vitro culture media in order to evaluate control of leaf water loss, growth and field survival. The addition of 10(-4) M ABA to the culture media in sealed containers produced plants that had similar control of leaf water loss and were morphologically similar to plants cultivated in ventilated containers without the addition of ABA. Field survival of 10(-4) M ABA plants (75%) was increased compared to plants cultivated in sealed containers without ABA (31%), with survival being closer to that of plants cultivated in ventilated containers (90-100%). Plants cultivated with 10(-4) M ABA (sealed and ventilated) also exhibited increased plant vigour and leaf area in the field compared to plants cultivated without ABA. The results suggest that the limited field survival and growth of plants cultured in vitro are related to the limited ABA concentrations they accumulate while in vitro. Consequently, conditions that increase the endogenous ABA concentrations of in vitro plants (like ventilation or ABA addition to the medium) would improve the control of leaf water loss, field survival and plant vigour.
