The conversion of both parental- and progeny-nascent open circular M13 RF DNA into covalently closed RF I is drastically reduced in an E. coli mutant deficient in the 5′ → 3′ exonuclease associated with DNA polymerase I. The nascent progeny RF DNA also contains a significant proportion of fragments of smaller than unit length.  相似文献

Results are presented of an investigation on electric and magnetic fields leaking from inductive (magnetic) heaters that are used for thermal processing of high-power electron tubes and lasers in an industrial plant. Measurements of electric and magnetic fields were done using both commercially available and laboratory-developed instrumentation. Isotropic H-field sensors were developed to allow quantitative evaluation of high-intensity magnetic fields. Ten induction heaters with nominal A.C. power ranging from 2.5 kW to 15 kW and operating at frequencies between 300 kHz and 790 kHz were surveyed. Electric field strengths up to 8 kV/m and magnetic field strengths up to 20 A/m were measured.  相似文献

Molecules with single-stranded tails (rolling circles) were isolated as replicating intermediates in G4 progeny single-stranded DNA synthesis. Lysates from infected cells harvested late in infection during single-stranded DNA synthesis were not deproteinised but analysed directly in caesium chloride and propidium diiodide gradients. The gradient fractionated them on the basis of tail length. If the lysates were first deproteinised however, the tailed replicative intermediates banded as a peak at a density just greater than that of replicative form II DNA (RFII) and did not spread down the gradient. The origin of synthesis of the viral strand tail was mapped by electron microscopy as 55 to 60% away from the single EcoRI cleavage site. Termination molecules finishing a round of viral strand DNA synthesis have been identified as molecules consisting of a closed single-stranded DNA circle attached by a very small region to the parent double-stranded DNA circle.  相似文献

In a preceding paper (Schröder and Kaerner, 1972) a rolling circle mechanism has been described for the replication of bacteriophage φX174 replicative form. Replication involved nicking and elongation of the viral (positive) strand component of the RF molecule resulting in the displacement of a single-strand tail of increasing length. The synthesis of the new complementary (negative) strand on the single-strand tails appears to be initiated with considerable delay and converts the tail into double-stranded DNA. Before the new negative strand is completed the replicative intermediates split into (I) a complete RF molecule containing the “old” negative and the new positive strand, and (II) a linear, partially double-stranded “tail” consisting of the complete old positive strand and a fragment of the new negative strand.The present study is concerned with the fate during RF replication of these fragments of the rolling circles. Those RFII molecules containing the old negative strands appear to go into further replication rounds repeatedly. Some of the tails were found in the infected cells in their original linear form. “Gapped” RFII molecules, which have been described earlier by Schekman and co-workers (Schekman &; Ray, 1971; Schekman et al., 1971), are supposed to originate from the tails of rolling circle intermediates by circularization of their positive strand components. Evidence is provided by our experiments that even late during RF replication these gaps are present only in the negative strands of RFII. Appropriate chase experiments indicated that the tails finally are converted to RFI molecules. Progeny RFI molecules could not be observed to start new replication rounds under our conditions although we cannot exclude that this might happen to some minor extent.The results presented suggest that the master templates for RF replication are the first negative strands to be formed, rather than the parental positive strands.  相似文献

The break in the complementary DNA strand of early G4 replicative form II DNA (RFII) and in the viral DNA strand of late RFII DNA was located using two single cleavage restriction enzymes (EcoRI and PstI) and by limited nick translation of the break using DNA polymerase I and 32P-labelled deoxyribonucleotides followed by digestion with the restriction enzymes HaeIII and HindII. The break in the complementary DNA strand was unique and in HaeIII Z5 close to the EcoRI cleavage site whereas the break in the viral DNA strand was on the other side of the molecule in HaeIII Z2 approxiately 50% away from the EcoRI cleavage site. Distribution of a short 3H pulse in early G4 replicating intermediates that were synthesising both DNA strands at the same time showed that synthesis of the strands started on opposite sides of the molecule and proceeded in opposite convergent directions, suggesting that initiation of synthesis of the two strands was independent and not unified in a single growing fork.  相似文献

Electron microscope partial denaturation maps of two viral DNAs, simian virus 40 and φX174 replicative form, have been obtained. A simple computer program has been developed to predict denaturation maps from any given DNA sequence, based on the percentage of A · T base-pairs along the molecule. Maps constructed from the SV40 DNA and φX174 replicative form DNA base sequence show a good correlation with the experimental maps. The results show that the regions of a DNA molecule that denature first are, in fact, those regions with the highest content of adenine and thymine base-pairs.  相似文献

Five distinct DNA replicating intermediates have been separated from lysates of bacteriophage G4-infected cells pulse-labelled during the period of replicative form synthesis using propidium diiodide/caesium chloride gradients. These are a partially single-stranded theta structure that is labelled in both the viral and complementary DNA strands; partially single-stranded circles, some with an unfinished viral DNA strand (25%) and some with an unfinished complementary DNA strand (75%); replicative form II(RFII) and replicative form I(RFI) DNA labelled only in the complementary DNA strand. To explain the pulse-label data a model is proposed in which G4 replicative form replication takes place by a displacement mechanism in which synthesis of the new viral DNA strand displaces the old viral DNA strand as a single-stranded DNA loop (D-loop) and when the displacement reaches half way round the molecule (the origin of synthesis of the G4 viral and complementary DNA strands are on opposite sides of the genome, Martin &; Godson 1977) synthesis of the complementary DNA strand starts, but in the opposite direction. Strand separation of the parent helix runs ahead of DNA synthesis, releasing two partially single-stranded circles from the replicating structure which then complete their replication as free single-stranded DNA circles. No evidence was found to support a rolling circle displacement mechanism of G4 replicative form synthesis.  相似文献

RF-amides (RFa), a superfamily of evolutionary-conserved neuropeptides, are expressed in both invertebrates and vertebrates. While some endocrine functions have been attributed to these peptides in lower vertebrates and few mammalian models, not much is known about their actions in primates. Therefore, the present study was designed to examine the effects of peripheral administration of two recently cloned human RFa peptides, 26RFa and 43RFa, on testosterone and prolactin secretion in the adult male adult male rhesus monkey (Macaca mulatta). For control purposes, a scrambled sequence of 26RFa (Sc-26RFa) and normal saline (1ml) were injected. Three different doses of 26RFa and 43RFa (19-nmol, 38-nmol and 76-nmol) and a single dose (38-nmol) of Sc-26RFa were tested. A set of four chair-restraint habituated monkeys was used. Comparison of post-treatment T levels with respective pre levels showed that none of the doses of both 26RFa and 43RFa changed T release. Similarly, Sc-26RFa and saline administration also did not affect T levels. In contrast, all doses of 26RFa and 43RFa significantly (P<0.05) stimulated prolactin secretion. 43RFa dose dependently increased prolactin secretion while dose dependency was not observed for 26RFa. Saline and Sc-26RFa injection had no effect on prolactin concentrations. Thus, present study demonstrated that peripheral administration of 26RFa and 43RFa, in the doses tested, have no effect on T secretion, suggesting possible selective lack of their neuroendocrine role in controlling hypothalamic-pituitary-gonadal axis in the adult male primates. The prominent stimulation of prolactin suggests a neuroendocrine role of RFa peptides in regulation of prolactin release in primates.  相似文献

We discuss initial magnetic field strength measurements made around radiofrequency (RF) induction heaters. These measurements were made with a monitor developed for the National Institute for Occupational Safety and Health (NIOSH) by the National Bureau of Standards (NBS). The monitor has a dynamic range of .01 to 10,000 A2/m2, a frequency range of 300 kHz to 100 MHz, an isotropic response (+/- .3 dB) with three mutally orthogonal loop antennas that have the ability to measure and display each of three orthogonal magnetic field components, a high probe burnout protection level of 20,000 A2/m2, and an accuracy of +/- 1.0 dB at 13 calibration frequencies. The portable survey monitor was used to measure the magnetic field strengths in the vicinity of 16 RF induction heaters. Typically these induction heaters are operated continuously for several hours. The maximum field strengths (without duty factor correction) ranged from 15 to 4,500 A2/m2 and were measured 5 to 51 cm from the loop applicators of the induction heaters. At locations commonly occupied by workers (ie, approximately 30 to 76 cm from heaters), the fields ranged from .01 to 300 A2/m2 (without duty factor correction).  相似文献

Translation of genetic information encoded in messenger RNAs into polypeptide sequences is carried out by ribosomes in all organisms. When a full protein is synthesized, a stop codon positioned in the ribosomal A site signals termination of translation and protein release. Translation termination depends on class I release factors. Recently, atomic-resolution crystal structures were determined for bacterial 70S ribosome termination complexes bound with release factors RF1 or RF2. In combination with recent biochemical studies, the structures resolve long-standing questions about translation termination. They bring insights into the mechanisms of recognition of all three stop codons, peptidyl-tRNA hydrolysis, and coordination of stop-codon recognition with peptidyl-tRNA hydrolysis. In this review, the structural aspects of these mechanisms are discussed.  相似文献

In situ radiofrequency (RF) exposure of the different RF sources is characterized in Reading, United Kingdom, and an extrapolation method to estimate worst-case long-term evolution (LTE) exposure is proposed. All electric field levels satisfy the International Commission on Non-Ionizing Radiation Protection (ICNIRP) reference levels with a maximal total electric field value of 4.5 V/m. The total values are dominated by frequency modulation (FM). Exposure levels for LTE of 0.2 V/m on average and 0.5 V/m maximally are obtained. Contributions of LTE to the total exposure are limited to 0.4% on average. Exposure ratios from 0.8% (LTE) to 12.5% (FM) are obtained. An extrapolation method is proposed and validated to assess the worst-case LTE exposure. For this method, the reference signal (RS) and secondary synchronization signal (S-SYNC) are measured and extrapolated to the worst-case value using an extrapolation factor. The influence of the traffic load and output power of the base station on in situ RS and S-SYNC signals are lower than 1 dB for all power and traffic load settings, showing that these signals can be used for the extrapolation method. The maximal extrapolated field value for LTE exposure equals 1.9 V/m, which is 32 times below the ICNIRP reference levels for electric fields.  相似文献

By using a new mild procedure for detecting DNA crosslinks it has been shown that adriamycin and daunomycin are able to form interstrand DNA crosslinks in HeLa cells. This effect seems to be preceded by transformation of the parent antibiotics in the cell to active forms. In addition, interstrand DNA crosslinks formed by adriamycin and daunomycin were found to be temperature- and alkali-labile.  相似文献

To obtain more detailed information relative to the potential usefulness of using radio frequency (RF) energy in treating hypothermia, anesthetized rhesus monkeys were used in a rewarming study that compared a conventional method (heating pad) with an RF induction coil system. Rectal temperature (Tre) of each subject was monitored, and enzyme and isoenzyme levels were determined from blood samples collected before, during, and up to 48 h after hypothermia in order to assess the effects of each rewarming method. The previously observed postprocedure rise in serum enzymes (most visible at 24 h) was again seen, with no statistically significant difference in the time course of serum enzyme levels between the two treatments for comparable durations of hypothermia. To test the limits of the ability of the RF induction coil system, successively more severe hypothermia was induced in the subjects to the point of cardiovascular collapse (Tre less than 20 degrees C); RF energy was successful in resuscitating the profoundly hypothermic subjects without discernible harmful effects.  相似文献

During translation in Escherichia coli, the ribosome rescue factor YaeJ and the alternative ribosome rescue factor (ArfA, previously called YhdL) can release stalled ribosomes from mRNA. Here, I used a reconstituted cell-free protein synthesis system to examine YaeJ- and ArfA-dependent recycling of stalled ribosomes, in which mRNA lacks in-frame stop codons. It is shown that YaeJ alone could recycle the ribosome but that ArfA required the presence of release factor 2 (RF2). Furthermore, I show that RF2 binds to stalled ribosomes only in the presence of ArfA, demonstrating that ArfA recruits RF2 into the A site of the ribosome to facilitate peptidyl-tRNA hydrolysis. It is also demonstrated that the efficiency of the ArfA-dependent process decreases rapidly with an increase in mRNA length downstream of the A site of the ribosome whereas YaeJ function is maintained on mRNA with sufficient length. From the results, I discuss differences of in vivo roles of these two systems in addition to the well-known tmRNA-dependent trans-translation system.  相似文献

The aim of this study was to examine experimentally effects of low level, modulated microwaves on human central nervous system function utilizing the phenomenon of visual masking. Ten healthy volunteers, four males and six females, were exposed to electromagnetic field (450 MHz, 0.16 mW/cm2) with 7 Hz modulation frequency. Two photo series (visual stimuli) of unfamiliar, young male faces were presented to the subjects, one picture after another. All the photos were frontal views of unfamiliar faces, which could be recognized only by their unique combinations of features. The task was to identify the pictures from a group of six photos and to decide which order they were presented in. The phenomenon of visual masking is revealed as anamorphosis in subject's perception of two instantaneous visual stimuli presented within a short time interval. When both stimuli were to be recognized correctly and put in the right order, there was a statistically significant difference (P < 0.05) between the identification of the stimulus with microwave electromagnetic field and sham exposure. Recognition of both stimuli in a pair was better under the sham exposure conditions but the actual difference was only 5%. It was concluded that early stages of visual information processing are overwhelmingly robust and routine (and adaptively significant) activities, so that the low level 7 Hz modulated electromagnetic field effects exerted upon it are extremely weak.  相似文献

Some macaques infected with SRV-2 developed SAIDS and RF, a Kaposi's sarcoma (KS)-like tumor. We investigated the immunophenotypic markers of this SAIDS-associated retroperitoneal fibromatosis (RF). RF tumor is characterized by proliferation of spindle cells accompanying inflammatory cell infiltrates, fibroblasts, and endothelial cells. RF spindle cells in tumor tissues revealed several immunobiologic characteristics similar to vascular smooth-muscle cells or myofibroblasts based on positive immunoreactivity of smooth-muscle α-actin and desmin. The majority of cultured RF spindle cells also expressed specific markers for vascular smooth muscle. These results suggest that the RF spindle cells are derived from vascular smooth-muscle cells. Furthermore, RF tissues and cells were persistently infected with SRV-2, which may play an important role in viral etiology of AIDS-associated neoplasm in this macaque model.  相似文献
