Groups of mice from genetically defined inbred strains were infected genitally with a pathogenic human strain of Chlamydia trachomatis and their subsequent fertility was compared. The CBA, C3H (H-2o) and C3H/He-mg (H-2k) mice were less fertile than control mice, at least up to 6 months after infection. In contrast, fertility was not impaired in BALB/c mice or in congenic BALB/K mice, which had the H-2k haplotype. Reduced fertility was paralleled by the extent of histological oviductal inflammation in mice of each strain. No salpingitis was seen 21 days after infection in the BALB strains, but lesions were apparent in CBA and C3H strains up to about 70 days after inoculation and these sometimes developed into hydrosalpinges. These results indicate that susceptibility to chlamydial salpingitis and subsequent infertility is under genetic control. This control was not simply associated with the major H-2 gene complex, as mouse strains of the same haplotype (H-2k) differed in susceptibility. The fertility of BALB/c (H-2d) and BALB/K (H-2k) strains was no different from that of controls, and congenic C3H mice of differing H-2 haplotypes (H-2k and H-2o) showed reduced fertility. Although all the infected F1 (BALB/K x C3H/He-mg) mice produced litters at the same rate as untreated controls, the litters were considerably smaller. This was due to the occurrence of unilateral pregnancies in the mice inoculated under the ovarian bursae and possibly also to early fetal death in mice inoculated directly in the uterus. These findings emphasize the importance of early diagnosis and treatment of infection of the lower genital tract of women.  相似文献

Suspected superfetation was investigated in a Glasgow hybrid stock of mice. The male was removed either (i) a few days before parturition, or (ii) immediately after mating and on 23 and 25 occasions, respectively, a second litter was born. Members of the anomalous litters were inbred for 10 generations, but the incidence of supernumerary litters did not increase beyond 2-5%. The anterior part of over 500 reproductive tracts, at various stages of pregnancy and after parturition, were serially sectioned but a second set of embryos was not found. The second gestation was of normal length and superfetation was not therefore considered to be the cause of the anomalous litters. In two females, one non-pregnant and one pregnant, spermatozoa were found in the uterus and oviducts 8 days after mating and in distended uterine glands 15 days after mating respectively. It is concluded that the anomalous litters were derived from the fertilization of eggs ovulated at the post-partum oestrus by spermatozoa which had been retained in the female tract for at least 23 days.  相似文献

The distribution of larval Aspiculuris tetraptera was studied in 4-week-old male and female CFLP mice. Whereas on days 10–12 the larvae were entirely confined to the anterior third of the colon, by day 14 larvae could be found throughout the colon. After day 17 the larvae were again restricted to the anterior colon. This change in distribution was co-incident with a loss of a large proportion of the worm burden, which occurred more consistently in female than in male mice.The degree of acquired immunity stimulated by various immunizing regimens was assessed by the survival of a challenge infection in experimental and control mice. It was found that a high level of immunity was achieved by exposure to a 19-day primary infection, a 36-day low-level infection and also by three 6-day infections, in each of which the larvae were removed by piperazine treatment immediately after the crypt phase.  相似文献

Various influences of the host altered the establishment and reproduction of mouse-adapted Nippostrongylus brasiliensis. Establishment and egg production were greater in mature male than in female mice. Worm fecundity increased similarly in both sexes of mice prior to 41 days of host age. Male mice at 62 days of age or older harbored more N. brasiliensis than did females, but the number of nematodes was similar in younger hosts of both sexes. Crowding of male mice significantly increased the establishment of helminths, but not the production of eggs by N. brasiliensis. Pregnancy of the host significantly increased both establishment of and reproduction by N. brasiliensis. Ligation of the bile duct of the mouse reduced both establishment and fecundity of the nematode after subcutaneous or surgical infection with larvae and adults, respectively.  相似文献

Trichinella spiralis infection elicits a vigorous IgE response and pronounced intestinal and splenic mastocytosis in mice. Since IgE both activates mast cells (MC) and promotes their survival in culture, we examined its role in MC responses and parasite elimination in T. spiralis-infected mice. During primary infection, wild-type but not IgE-deficient (IgE(-/-)) BALB/c mice mounted a strong IgE response peaking 14 days into infection. The splenic mastocytosis observed in BALB/c mice following infection with T. spiralis was significantly diminished in IgE(-/-) mice while eosinophil responses were not diminished in either the blood or jejunum. Similar levels of peripheral blood eosinophilia and jejunal mastocytosis occurred in wild-type and IgE-deficient animals. Despite the normal MC response in the small intestine, serum levels of mouse MC protease-1 also were lower in parasite-infected IgE(-/-) animals and these animals were slower to eliminate the adult worms from the small intestine. The number of T. spiralis larvae present in the skeletal muscle of IgE(-/-) mice 28 days after primary infection was about twice that in BALB/c controls, and the fraction of larvae that was necrotic was reduced in the IgE-deficient animals. An intense deposition of IgE in and around the muscle larvae was observed in wild-type but not in IgE null mice. We conclude that IgE promotes parasite expulsion from the gut following T. spiralis infection and participates in the response to larval stages of the parasite. Furthermore, our observations support a role for IgE in the regulation of MC homeostasis in vivo.  相似文献

Vertical transmission of Neospora caninum was evaluated in BALB/c mice using an N. caninum-specific polymerase chain reaction (PCR) assay as a means of detecting parasite transmission to offspring. BALB/c mice were infected with the NC-1 isolate of N. caninum during pregnancy (days 8-15 gestation). Transmission of parasite, detected by PCR, was determined in 2- to 23-day-old offspring. When dams were infected on days 13-15 of gestation, transfer of parasites was detected in only a proportion of the litter. Infection between days 8 and 12 of gestation resulted in a high frequency of parasite transmission; every offspring from all litters was infected. The tissue locations of parasites in pups of different ages were determined. In young pups (2- to 4-days-old), the predominant sites of infection were the lungs and the brain. In older pups (7- and 23-days-old) the predominant site of infection was the brain. This study shows that PCR may be useful for evaluation of candidate vaccines against horizontal N. caninum infection, vertical transmission, or both.  相似文献

The mode of larval migration (visceral larva migrans) in Toxocara canis infection was compared for BALB/c, C57BL/6, C3H/He, DBA/2, NC and BALB/c nude mice following oral infection with 400 eggs. The mean recovery of larvae from the liver on day 2 post infection (PI) was not different in terms of the strain, age or sex of the mice. The number of larvae recovered from the liver decreased in all strains on days 6, 12 and 21 PI, but the mean for BALB/c and (NC X BALB/c) F1 mice was significantly higher than that for C567BL/6, NC and BALB/c nude mice, unless the total number of larvae in the carcasses on day 21 PI was the same among those strains including athymic nude mice. The mean recovery of larvae from the liver on day 6 PI increased with age in both NC and BALB/c mice, although no sex difference was observed. From these results, it is emphasized that the age and strain of animals should be properly selected for animal experimentation with T. canis infection.  相似文献

Five strains of male and female mice were infected with salivary gland-passaged murine cytomegalovirus (SG-MCMV) intraperitoneally. Although none of them became hyperglycemic, 13-30% of BALB/c male and female, ICR/Slc male and female and C57BL/6J male mice had abnormal values in glucose tolerance tests (GTT) 14 to 30 day post infection. Serum insulin levels in BALB/c male mice were low and infective virus was detected in the pancreas during the acute phase of infection. Histopathology showed mild edematous changes with inflammatory cell infiltrations in the pancreas. Viral antigen was located in acinar cells and occasionally in beta cells by an immunofluorescence test.  相似文献

The incidence and duration of transmission of infection with ectromelia virus strain NIH-79 was tested in innately resistant (C57BL/6) and innately susceptible (BALB/c) inbred mice. Transmission by C57BL/6 index mice occurred through 3 weeks and by BALB/c index mice through 4 weeks, although the duration of infection in individual index mice was often shorter. Soiled caging that previously housed infected mice was inconsistently infectious. Transmission was high in cages where infected mice died and were cannibalized by cagemates, but was low to moderate in cages where there was no cannibalism. Infected mice that were bred 6 weeks after they were infected, delivered virus-free progeny and did not transmit infection to their non-immune breeding partners. Sentinel mice housed in the room with experimentally infected mice were seronegative for antibody to ectromelia virus and to other murine viruses. These results support the view that infection with NIH-79 virus is typically short-lived. They also indicate that breeding of recovered mice can save valuable colonies that have been exposed to ectromelia virus.  相似文献

The implantation and development of intravenously injected Trichinella spiralis newborn larvae were examined in different strains of inbred mice by determining muscle larvae burden. This was compared to the numbers of muscle larvae that established after a natural infection during which a quantitative assessment of intestinal newborn larvae production was made. In most inbred strains of mice, newborn larvae do not all successfully implant in muscle. Mice of the DBA/1 strain are the most resistant to successful implantation, and C3H mice are the most permissive. This pattern is evident in the strains studied whether newborn larvae are injected intravenously or are produced by intestinal adults. Thus, after a natural infection, 100% of intestinally produced newborn larvae implanted in C3H mice, whereas in NFR 68% and DBA/1 mice 62% successfully matured in muscle. Immunity to newborn larvae could be demonstrated as early as 10 days after exposure to this stage of the life cycle. This immunity was protective against a complete challenge infection given 9 days after newborn larvae had been injected intravenously. Protection against newborn larvae was identical in male and female mice or in mice from 1 to 9 months of age. We conclude that there are two mechanisms by which mice impair newborn larvae establishment or development in muscle. The first appears to be nonimmunological (non-specific resistance), and the second is immunological. Genetically determined variation in strain-specific expression is apparent with both mechanisms. In strains displaying high intrinsic "resistance" (DBA/1), this process is likely to account for most of the 38% reduction in newborn larvae establishment in a primary infection. However, immunity against newborn larvae develops quickly enough to have a significant effect on migratory larvae in primary infections where adults persist in the intestine (e.g., the B10 congenic mice), or when high adult worm burdens delay adult worm rejection. Muscle larvae burden, therefore, reflects systemic nonspecific resistance to newborn larvae as well as immunological processes that occur in the intestine and systemically.  相似文献

Research was undertaken to answer basic questions on susceptibility, clinical response and transmission of ectromelia virus in selected strains of inbred mice. C57BL/6J and AKR/J were found to be markedly more resistant to a virulent strain of ectromelia virus (isolated during the 1979-80 outbreak at the National Institutes of Health) than C57LJ, BALB/cByJ, DBA/2J, A.By/SNJ and C3H/HeJ when infected by footpad inoculation. In C57BL/6J and AKR/J the LD50 was about 7 logs higher than the ID50. With one exception, C57LJ, the LD50 and ID50 titers in the other strains were about equal. In C57LJ the LD50 titer was intermediate. Following intragastric inoculation, virus was isolated from feces of C57BL/6J mice for as long as 46 days and up to 29 days from BALB/cByJ mice. Transmission to cage mates from intragastrically infected C57BL/6J and BALB/cByJ occurred up to 36 and 30 days respectively after infection. Virus was isolated from the spleen in 2 of 5 BALB/cByJ mice and 1 of 7 C57BL/6J mice tested 95 days after gastric inoculation. Following footpad inoculation, BALB/cByJ mice consistently transmitted virus to cage mates before death at 10-12 days. C57BL/6J mice transmitted between days 8 and 17, but not beyond. Virus was maintained in C57BL/6J mice by exposure to infected cage mates for seven passages, which was the most attempted. Clinical signs in infected C57BL/6J mice were usually subtle or inapparent.  相似文献

The parasitic nematode Haemonchus contortus was cultured in vitro to the adult male and the egg-laying female. Gastric contents from 2 cannulated calves and 2 sheep as well as extracts of stomach mucosa from both hosts were added to a mixture of API-1 culture medium and Fildes' reagent (a pepsin digest of defibrinated bovine blood) adjusted to pH 6.4 for the first week and pH 6.8 thereafter. The gas phase was 85% N2:5% O2:10% CO2. Best results were obtained when API-1 culture medium plus Fildes' reagent was supplemented with ovine gastric contents. Adult males up to 10 mm were obtained, and they produced and stored sperm at about 28 days in culture. Spermatozoa were never seen in the uteri of females grown in vitro. After 36 days in culture, female worms up to 16 mm were obtained which produced and layed eggs. Some of the eggs underwent division up to the 8-cell stage but did not develop further. No live larvae were observed at a stage earlier than the fourth. Presumably, this was because no matings were observed, although large clumps of worms did occur in the culture medium and mating might have occurred. The growth of adults from young to mature phases of this stage included: for the female, oogenesis and laying of eggs; for the male, development of paired sclerotized spicules, and spermatogenesis with the production of spermatozoa.  相似文献

杨树粒肩天牛在福建省2a发生1代;老熟幼虫在第2年4月下旬至5月初开始化蛹,成虫出现期始于5月底6月初,6月中下旬到7月上旬为盛发期,9月上旬只见个别成虫;产卵期始于6月中旬,6月下旬至7月中旬为产卵高峰期;幼虫孵化期为6月下旬到8月中旬,7月上中旬为为盛发期;幼虫不越冬。杨树粒肩天牛成虫为补充营养所取食的树种较集中,主要是构树与桑树。杨树粒肩天牛幼虫对杨树危害严重,但成虫却不喜欢取食杨树,用杨树饲养的粒肩天牛成虫寿命很短,仅3-19d,不产卵;而以桑树为补充营养的雌成虫的平均寿命为55d、雄26.5d;以构树为补充营养的雌成虫的平均寿命为78d、雄45.5d,补充桑树与构树的粒肩天牛成虫均可正常产卵。雌雄成虫一生可多次交尾。雌虫有多次产卵现象,单雌每天产卵数量1-12粒不等。  相似文献

ABSTRACT. Amblyospora californica is a polymorphic, eukaryotic microsporidian. Three types of sporogony producing three types of spores occur in male larvae and female adults of its mosquito host, Culex tarsalis , and an alternate copepod host, Acanthocyclops vernalis. Development of A. californica in male larvae includes merogony and sporogony. Karyogamy and meiosis was observed in sporogony in male larvae but not in the female adult or in the copepod. Chromosomal evidence showed that sporogony included two consecutive meiotic divisions and a subsequent mitosis forming an octosporont, ultimately containing eight haploid, uninucleate mature spores. In this species, the haploid number of chromosomes is nine. Macrosporoblasts and macrospores, containing 1, 2 or more nuclei, can be seen in infected male larvae. The stage of sporogony in which cytokinesis was arrested seems to determine the number of nuclei. Those with only one nucleus, we believe are due to failed nuclear division at meiosis. Although A. californica displayed a process of karyogamy and meiosis similar to that of the species from Cx. salinarius , they may not be the same species because of the difference in their chromosome numbers.  相似文献

The objective of this study was to investigate, using the Heligmosomoides polygyrus (= Nematospiroides dubius)-mouse model, whether live post-infection trichostrongylid larvae recovered from the intestinal wall of donor animals and placed subcutaneously would serve as vaccine protecting against oral challenge by third-stage (infective) larvae (L3). Experiments were conducted to determine the effect of number and age of post-infective larvae as well as age and sex of host on vaccination. Vaccinated BALB/cByJ mice were challenged with 30 L3 and total adult worm burdens compared between vaccinated groups and sham-treated controls (greater than 90% infection rates). All mice subcutaneously vaccinated with either five or 10 larvae harbored significantly fewer challenge parasites in their intestines than did sham-treated controls (P less than 0.001). Both young and mature mice were significantly protected against challenge by the subcutaneous larval vaccine. Adult female mice had significantly (P less than 0.05) fewer parasites than adult male mice. The age of the larvae (indicated as the days between infection and harvesting of the larvae) was important in that day-4 or day-6 larvae (L4) were significantly more protective (P less than 0.001) than day-2 (L3) or day-8 larvae (L5-preadult). Reduction in worm burden for young vaccinated animals ranged from 31 to 39% (P less than 0.001) and for mature animals from 88 to 100% (P less than 0.001). Five pairs of female Swiss-Webster mice were caged with 5 males (2 females/1 male). Eight females were inoculated orally with 2.6 x 10(5) Caryospora bigenetica oocysts either 3 days before mating, 3 days after mating (PMD), 9 PMD, or 16 PMD. The remaining 2 females were inoculated orally with Hanks' balanced salt solution and served as controls. One female from each cage delivered naturally at full term and the second female delivered by cesarean section on postmating day 18. The number of offspring per litter ranged from 7 to 12. One female produced a litter of 3 stillborn and 5 liveborn offspring. Seven of 8 female mice exhibited swollen muzzles and footpads 8 days after inoculation. Caryospora bigenetica was identified in tissues of muzzle, tongue, footpad, uterus, and placenta at necropsy. This is the first report of C. bigenetica in uterus and placenta. Clinical signs and tissue infections were not observed in control mice or in any offspring of the 10 female mice. This study presents evidence that C. bigenetica is not transmitted transplacentally.  相似文献

The mermithid parasite Heleidomermis magnapapula was maintained in larvae of the midge Culicoides variipennis for 20 months in enamel pans containing nutrient-rich water and polyester pads as a substrate. Inseminated female mermithids were introduced to the pad surface when the host was in the late second or early third-instar. Host larvae were harvested from the pans 9 days after exposure and held in tap water for nematode emergence. Preparasite yield was positively correlated with female nematode size and averaged 1,267 preparasites/female. Male and female nematodes emerged an average of 12.2 and 13.4 days after host exposure, respectively. Supplemental host food (Panagrellus) during the final days of parasitism did not alter time of emergence. Parasites emerging singly were 64% females, whereas superparasitized hosts yielded males (up to nine/host). Nematode carryover into the adult midge normally occurred at a level of 0.5-2.5%. Parasite load (nematodes/ parasitized individual) in midge adults was lower than that of larvae from the same cohort, and adult midges were more likely to harbor female parasites. Exposure of fourth-instar host larvae resulted in higher levels of adult parasitism (up to 17%).  相似文献

Eumycetoma due to Madurella mycetomatis is a major mycological health problem in endemic areas. We infected BALB/c mice (male or female) with various amounts of M. mycetomatis mycelium, containing sterilized soil as a natural adjuvant or Freund's incomplete adjuvant. Mice differed with respect to age and immune status. Intraperitoneal, subcutaneous and intravenous inoculation was explored and survival was monitored. Mice were killed at various intervals after inoculation, checked for the presence of the characteristic black grains, and organs were cultured for M. mycetomatis. Infected organs were subjected to histopathological examination. Immunocompetent male mice were as susceptible as immunocompromised female mice, but showed higher mortality rates. In conclusion, a reproducible mouse model of intraperitoneal M. mycetomatis infection with characteristic black grains in immunocompetent adult or young female mice was developed. Although this experimental model does not simulate macroscopic features of the subcutaneous M. mycetomatis infection in humans, the histopathological characteristics of the lesions and the development of black grains are clearly representative for the human infection. This model will enable further studies on the pathogenesis as well as prevention and treatment of the fungal infection.  相似文献
