Roles and activities of Aux/IAA proteins in Arabidopsis.   总被引:12,自引:0,他引:12  
Auxin induces various distinct developmental responses, partly by regulating gene expression. The Aux/IAA genes are a large gene family, many of which are induced by auxin. Work on Arabidopsis Aux/IAA genes has begun to reveal that they can regulate development and auxin-induced gene expression. Furthermore, auxin responses require Aux/IAA protein turnover. Finally, recent evidence suggests that Aux/IAA proteins can mediate light responses. Work in the near future should test whether Aux/IAA proteins are antennae that connect auxin and light signals to endogenous developmental responses.  相似文献

通常可通过植物叶片的形态来区分不同植物的种类。叶片由茎顶端分生组织侧翼发育而成,为多种多样大小和形状的扁平结构。叶片的结构看似简单,但调控叶片形态和结构发育的分子机理错综复杂,叶片的发育受植物激素、转录因子、一系列蛋白因子及环境的共同调控。本文回顾了叶片边缘形态和叶脉发育研究的最新进展。在叶边缘形态方面,Aux/IAA生长素响应抑制家族蛋白通过调节生长素浓度最大点的离散分布影响小叶的起始和生长以及叶边缘结构;NAM/CUC转录因子促进叶边缘锯齿的分离以及复叶中小叶的分离和分化,NAM/CUC和Aux/IAA通过不同通路实现对生长素的调控;拟南芥RAX1基因/番茄Potato-leaf基因和拟南芥JAG基因/番茄LYR基因促进叶边缘锯齿发育;RCO调控复叶小叶的发育不通过改变生长素的分布来实现;在番茄中反式小干扰RNA途径中的因子参与叶边缘形态发育;另外,在拟南芥中,mir164A、CUC2、PIN1、DPA4、SVR9-1及SVR9L-1构成复杂的调控网络影响叶边缘锯齿的发育。在叶脉发育方面,PIN1能否正确的定位会影响叶脉发育;AS1和AS2共同参与叶片远近轴极性的分化;另外AXR6、MP、BDL、CVP因子功能的缺失影响叶脉发育;生长素、PIN1、Aux/IAA、MP、ATHB8构成反馈循环调控子叶叶脉的形成。  相似文献

The auxin/indoleacetic acid (Aux/IAA) proteins are negative regulators of the auxin response factors (ARFs) that regulate expression of auxin-responsive genes. The Aux/IAA proteins have four conserved domains. Domain II is responsible for the rapid degradation of these proteins. Degradation of the Aux/IAA proteins, mediated by a SCF(TIR1) E3 ubiquitin protein ligase complex, is critical for auxin-regulated gene expression. Using a steroid-hormone-inducible system, we had previously shown that a protein-stability-enhancing mutation in domain II of IAA1 (iaa1) impaired diverse auxin responses. Inhibition of hypocotyl elongation, leaf expansion, and stem elongation by overexpression of iaa1 suggested that cell enlargement and/or cell division might be affected. We here examined the effects of the domain II mutation on cellular anatomy using light microscopy. Our results show that overexpression of iaa1 in Arabidopsis significantly reduced cell length and cell number and affected cell shape in inflorescences and leaves in a dexamethasone (DEX)-dependent manner. These results suggest that IAA1 might be involved in cell elongation as well as in cell division in the aerial parts of Arabidopsis plants. In addition, the formation of both phloem and xylem in leaves and stems was also impaired in a DEX-dependent manner, indicating a potential involvement of IAA1 in vascular development.  相似文献

Auxin/indole-3-acetic acid (Aux/IAA) proteins function as repressors of auxin response gene expression when auxin concentrations in a cell are low. At elevated auxin concentrations, these repressors are destroyed via the ubiquitin-proteasome pathway, resulting in derepression/activation of auxin response genes. Most Aux/IAA repressors contain four conserved domains, with one of these being an active, portable repression domain (domain I) and a second being an auxin-dependent instability domain (domain II). Here, we have analyzed the effects of amino acid substitutions in the repression domain of selected Aux/IAA proteins. We show that stabilized versions of Aux/IAA proteins with amino acid substitutions in domain I display contrasting phenotypes when expressed in transformed Arabidopsis (Arabidopsis thaliana) plants. An alanine-for-leucine substitution in the LxLxL (where L is leucine and x is another amino acid) repression domain of IAA3, IAA6, or IAA19 confers enhanced auxin response gene expression and "high-auxin" phenotypes when expressed from the 35S or IAA19 promoter (as tested with IAA19) in transformed Arabidopsis plants. In marked contrast, a single alanine-for-leucine substitution in domain I of IAA12 or IAA17 confers repression of auxin response genes and "low-auxin" phenotypes. These results point to intrinsic differences in the repression domain(s) of IAA proteins and suggest that some IAA proteins have stronger or more complex repression domains than others.  相似文献

A semi-dominant mutant suppressor of hy2 (shy2-1D) of Arabidopsis thaliana, originally isolated as a photomorphogenesis mutant, shows altered auxin responses. Recent molecular cloning revealed that the SHY2 gene is identical to the IAA3 gene, a member of the primary auxin-response genes designated the Aux/IAA gene family. Because Aux/IAA proteins are reported to interact with auxin response factors, we investigated the pattern of expression of early auxin genes in the iaa3/shy2-1D mutant. RNA hybridization analysis showed that levels of mRNA accumulation of the early genes were reduced dramatically in the iaa3/shy2-1D mutants, although auxin still enhanced gene expression in the iaa3/shy2-1D mutant. Histochemical analysis using a fusion gene of the auxin responsive domain (AuxRD) and the GUS gene showed no IAA-inducible GUS expression in the root elongation zone of the iaa3/shy2-1D mutant. On the other hand, ectopic GUS expression occurred in the hypocotyl, cotyledon, petiole and root vascular tissues in the absence of auxin. These results suggest that IAA3/SHY2 functions both negatively and positively on early auxin gene expression.  相似文献

Combinatorial interactions of AUXIN RESPONSE FACTORs (ARFs) and auxin/indole acetic acid (Aux/IAA) proteins through their common domains III and IV regulate auxin responses, but insight into the functions of individual proteins is still limited. As a new tool to explore this regulatory network, we generated a gain-of-function ARF genotype by eliminating domains III and IV from the functionally well-characterized ARF MONOPTEROS(MP)/ARF5. This truncated version of MP, termed MPΔ, conferred complementing MP activity, but also displayed a number of semi-dominant traits affecting auxin signaling and organ patterning. In MPΔ, the expression levels of many auxin-inducible genes, as well as rooting properties and vascular tissue abundance, were enhanced. Lateral organs were narrow, pointed and filled with parallel veins. This effect was epistatic over the vascular hypotrophy imposed by certain Aux/IAA mutations. Further, in MPΔ leaves, failure to turn off the procambium-selecting gene PIN1 led to the early establishment of oversized central procambial domains and very limited subsequent lateral growth of the leaf lamina. We conclude that MPΔ can selectively uncouple a single ARF from regulation by Aux/IAA proteins and can be used as a genetic tool to probe auxin pathways and explore leaf development.  相似文献

In Arabidopsis, SHY2 encodes IAA3, a member of the auxin-induced Aux/IAA family. Gain-of-function mutations in SHY2/IAA3 cause enlarged cotyledons, short hypocotyls, and altered auxin-regulated root development. Here we show that the gain-of-function mutation shy2-2 decreases both the induction and repression of auxin-regulated genes, suggesting that SHY2/IAA3 acts as a negative regulator in auxin signaling. shy2-2 affects auxin induction of many previously characterized primary response genes, implying that it might repress primary auxin responses. In addition, shy2-2 also affects expression of multiple auxin-nonresponsive genes. Light regulates expression of SHY2/IAA3, suggesting a possible link between light and auxin response pathways.  相似文献
