BACKGROUND: A comparative analysis by using static headspace (HS) and steam distillation (SD) GC-MS of the volatile and the semi-volatile secondary metabolites from leaves of cultivated Majorana syriaca. METHODS: The essential oils endogenous to cultivated thyme were isolated and identified by HS-GC-MS technology and compared to those from SD-GC-MS. RESULTS: The HS-GC-MS results showed that the Palestinian cultivated thyme is rich in monoterpene hydrocarbons and phenolic monoterpenes such as alpha-phellandrene, alpha-pinene, beta-myrcene, m-cymene, p-cymene, gamma-terpinene, thymol and carvacrol. In all the samples gamma-terpinene, p-cymene, thymol and carvacrol were the most abundant compounds. CONCLUSIONS: HS and SD-GC-MS have proved that most of the cultivated thyme samples examined has thymol isomer as the major phenolic constituent.  相似文献

The water-distilled essential oil the leaves of Coridothymus capitatus were analyzed by GC/MS and also analyzed by direct thermal desorption GC/MS. Comparison was made between two analyses techniques. The essential oil consisted mainly of monoterpenes 98.9%, while oxygenated hydrocarbons were identified as 55.6 % and non-oxygenated hydrocarbons as 43.6%. As major components were found carvacrol (35.6%), p-cymene (21.0%), thymol (18.6%), gamma-terpinene (12.3%), alpha-terpinene (3.2%), beta-myrcene (3.0%) and alpha-thujene (1.3%) by hydrodistillation and by the GC/MS method. The direct thermal desorption GC/MS analysis also showed the same major components, namely carvacrol (51.6%), thymol (21.7%), p-cymene (9.7%) gamma-terpinene (8.2%), alpha-terpinene (1.64%). As a part of an investigation of natural antioxidants from Dalmatian aromatic plants, in this paper we report a study of the antioxidant activity related to the chemical composition of savory free volatile compounds. Twenty-one compounds were identified in the essential oil without fractionation, representing 97.4% of the total oil. The major compound was phenolic monoterpene thymol (45.2%). Other important compounds were monoterpenic hydrocarbons p-cymene (6.4%) a nd gamma-terpinene (5.9%)and oxygen-containing compounds carvacrol methyl ether (5.8%), thymol methyl ether (5.1%), carvacrol (5.3%), geraniol (5.0%) and borneol (3.9%). The evaluation of antioxidant power was performed in vitro by the beta-carotene bleaching and thiobarbituric acid (TBA) methods. As determined with both methods, the total savory essential oil as well as different fractions or pure constituents containing hydroxyl group exhibited relatively strong antioxidant effect. The hydrocarbons, when isolated as CH fraction, showed the poorest effectiveness in spite the fact that this fraction contained gamma-terpinene, alpha-terpinene, p-cymene and terpinolene which previously were identified as potential antioxidants.  相似文献

Small-scale GC headspace analyses combined with SPME sampling of individual oil glands have been used to measure the variation in volatile content and composition in and within different oregano plants as affected by age, season and developmental state. The main monoterpenes found were p-cymene, carvacrol and their precursor gamma-terpinene. The early season preponderance of p-cymene over carvacrol was reversed as the season progressed and this pattern could also be seen at any time within the plant, from the latest leaves to be produced (low in cymene) to the earliest (high in cymene). Seedlings from the same mother plant developed this pattern at different rates. Within individual leaves the pattern was not observed, even within the youngest developing leaves. However it was found that the oil composition of individual glands within a single leaf varied considerably, most notably in respect of the production of carvacrol and its isomer thymol.  相似文献

The leafy parts of thyme and its essential oil have been used in foods for its flavour, aroma and preservation for many years. In the present study the genotoxic potential of major compounds of thyme oil, i.e. thymol, carvacrol, and gamma-terpinene and of the methanolic extracts of thyme, were investigated in human lymphocytes by single-cell gel electrophoresis. Also, the effects of these substances on the induction of DNA damage by 2-amino-3-methylimidazo[4,5-f]-quinoline (IQ) and mitomycin C (MMC) were evaluated. No increase in DNA strand breakage was observed at thymol and gamma-terpinene concentrations below 0.1 mM, but at the higher concentration of 0.2 mM significant increases in DNA damage were seen. Thymol and gamma-terpinene significantly reduced the DNA strand breakage induced by IQ and MMC at the lower concentrations studied. Carvacrol, which is an isomer of thymol, seemed to protect lymphocytes from the genotoxic effects of IQ and MMC at non-toxic concentrations below 0.05 mM, but at the higher concentration of 0.1 mM carvacrol itself induced DNA damage. Also the constituents of the n-hexane and ethyl acetate fractions prepared from the concentrated aqueous methanolic extracts of Thymus spicata protected lymphocytes against IQ- and MMC-induced DNA damage in a concentration-dependent manner.  相似文献

The antimicrobial activity, essential oil composition and micromorphology of trichomes of Satureja laxiflora C. Koch, a native plant from Iran, were studied. The essential oil was obtained from the aerial parts at the flowering stage by hydrodistillation, and analyzed by GC and GC/MS. Thirty-three compounds representing 99.1% of the total oil were characterized. The major compounds were thymol (63.9%) and gamma-terpinene (11.9%) followed by carvacrol (4.8%), p-cymene (3.9%), geraniol (3.2%) and geranyl acetate (3.1%). Furthermore, the essential oil and its three main components were tested against two bacteria and three fungi. The result of the bioassays has been shown that the oil possesses potent antimicrobial property. Chemical studies confirmed that a major portion of this antimicrobial activity is due to thymol present in the oil. Micromorphological analysis by SEM of both vegetative and reproductive organs revealed the presence of abundant sessile capitate and sparse short-stalked glandular trichomes along with retrorse eglandular hairs, giving useful diagnostic characters for identification of this medicinal plant.  相似文献

The chemical composition of essential oil isolated by hydrodistillation from the aerial parts of Origanum acutidens was analyzed by GC-MS. Carvacrol (87.0%), p-cymene (2.0%), linalool acetate (1.7%), borneol (1.6%) and beta-caryophyllene (1.3%) were found to be as main constituents. Antifungal, phytotoxic and insecticidal activities of the oil and its aromatic monoterpene constituents, carvacrol, p-cymene and thymol were also determined. The antifungal assays showed that O. acutidens oil, carvacrol and thymol completely inhibited mycelial growth of 17 phytopathogenic fungi and their antifungal effects were higher than commercial fungicide, benomyl. However, p-cymene possessed lower antifungal activity. The oil, carvacrol and thymol completely inhibited the seed germination and seedling growth of Amaranthus retroflexus, Chenopodium album and Rumex crispus and also showed a potent phytotoxic effect against these plants. However, p-cymene did not show any phytotoxic effect. Furthermore, O. acutidens oil showed 68.3% and 36.7% mortality against Sitophilus granarius and Tribolium confusum adults, respectively. The findings of the present study suggest that antifungal and herbicidal properties of the oil can be attributed to its major component, carvacrol, and these agents have a potential to be used as fungicide, herbicide as well as insecticide.  相似文献

The waterdistilled essential oil from dried aerial parts of one of the Algerian thymes, Thymus fontanesii Boiss. & Reut. grown in Setif province, Algeria, was analyzed by GC/MS. Zataria multiflora Boiss. is an aromatic shrub belonging to the Lamiaceae family. Its aerial parts are used in the traditional medicine and in the pharmaceutical and food industries. The terpenoid and genetic profiles of 18 accessions of Z. multiflora, collected in different locations in Iran, have been analyzed by GC/FID and GC/MS or by AFLP (amplified fragment length polymorphism) analyses, respectively. Altogether, 56 compounds were identified in the essential oils, with the major constituents being thymol (6.0-54.9%), carvacrol (0.7-50.6%), linalool (1.2-46.8%), and p-cymene (1.6-14.8%). On the basis of the essential-oil composition, the 18 accessions were divided into four groups. The first group was characterized by a high content of carvacrol, thymol, and linalool, the second group was dominated by carvacrol, thymol, and p-cymene, the third group was characterized by a high concentration of thymol and a low content of carvacrol and p-cymene, and the forth group contained linalool and carvacrol as the main components. The AFLP results revealed that the average genetic similarity (GS) between the accessions was 0.61, ranging from 0.40 to 0.77. The UPGMA (unweighted pair-group method with arithmetic mean) cluster analysis divided all accessions into five groups at a similarity level of 0.60. The two clusters generated, the first based on the essential-oil compositions and the second on the AFLP data, showed a different pattern of relationships among the accessions. The knowledge of the Z. multiflora chemotype diversity, illustrated in this study, will allow an improvement of the homogeneity of the plant material for the production of different types of essential oils, depending on the demands of the pharmaceutical and food industries for specific uses.  相似文献

The chemical composition of the volatile metabolites of Crithmum maritimum harvested from several geographic localities along the Mediterranean coasts was studied by GC and GC-MSD. The major oil constituents were found to be dillapiole, gamma-terpinene, sabinene, limonene and beta-phellandrene. The Western populations were richer in dillapiole, whereas the Southern collections were characterized by increased amounts of thymol methyl ether and gamma-terpinene. The Italian chemical profiles differentiated by the significant contributions of carvacrol methyl ether and isoterpinolene. The essential oils were also investigated for their insecticidal activity and their repellency against Pheidole pallidula (Nylander) ants and found to possess significant activity.  相似文献

Environmental factors affecting essential oils variability in Thymus piperella L. are studied. The relationship between the bioclimatics index and edaphic factors with three chemotypes is determined through the application of multivariate analysis (CANOCO) and other statistical processes to both ecological and the essential oil data set components. Chemotype p-cymene γ-terpinene carvacrol shows a positive relationship with the aridity index (Ia) and Altitude. The p-cymene thymol chemotype is related to factors affecting water balance in both the soils and plants. The influence of climatic factors over p-cymene carvacrol is less evident. The compound p-cymene, common to the three chemical groups and the most abundant of all, presents positive correlation with the organic matter in the soil.  相似文献

The fresh leaves and brine of leaves of Thymbra spicata var. spicata (KARAKIZ) were analyzed by hydrodistillation, headspace and GC/MS techniques. The main components were determined as carvacrol, p-cymene, beta-myrcene, gamma-terpinene, a-terpinene and trans-caryophyllene. The essential oil and the main compounds, carvacrol and trans-caryophyllene, have been tested against E. coli, S. epidermidis, B. subtilis, S. aureus, S. typhimurium, K. pneumoniae, P aeruginosa, E. faecalis and C. albicans. While the essential oil and carvacrol showed strong activity against all microorganisms, except P. aeruginosa, trans-caryophyllene showed activity only against C. albicans. The essential oil and carvacrol also showed strong antimycobacterial activity.  相似文献

Bakhtiarian savory (Satureja bachtiarica Bunge) is an endemic herb growing wild in Iran with interesting pharmacological and biological properties. In this research, the variability of essential oil content and phytochemical compositions among seven populations of this species was studied in both natural and field conditions. There was a wide interpopulation variability in phytochemical variation of studied populations under both natural and field conditions, indicating the existence of different chemotypes. Six chemical compositions showed high value in studied plants so that carvacrol and thymol were known as major compounds and confirmed by principal component analysis. Ilam population had the highest thymol in both field (77.10 %) and natural (42.32 %) conditions and was known as a thymol chemotype. Beside, Yazd and Shahrekord populations, respectively, showed the highest carvacrol percentages in both field (84.83 and 63.81 %) and natural (72.50 and 58.05 %) conditions and were known as carvacrol chemotypes. In other populations, carvacrol and thymol were the main components and showed partly equal percentage and these populations were known as carvacrol/thymol chemotypes. According to Pearson correlation analysis, carvacrol was negatively correlated with p-cymene, γ-terpinene and thymol (r = ?0.61, ?0.70 and ?0.98, respectively), indicating very high reverse correlation between carvacrol and thymol. In conclusion, the high value of the essential oil compositions in studied S. bachtiarica populations could provide useful information for conservation and selection of cross parents in breeding programs to develop improved cultivars.  相似文献

Thymol and carvacrol can bind to major and minor grooves of B-DNA. The aim of this study was to examine the interaction of calf thymus DNA with thymol and carvacrol in aqueous solution and physiological pH with thymol/DNA and carvacrol/DNA (phosphate) molar ratios of 1/20, 1/10, 1/5, and 1/1. Fourier transform infrared and UV-visible difference spectroscopy were used to determine the thymol and carvacrol binding mode, binding constant, sequence selectivity, DNA secondary structure, and structural variations of thymol/DNA and carvacrol/DNA complexes in aqueous solution. Spectroscopic evidence showed that the thymol and carvacrol interaction occurred mainly through H-bonding of the thymol and carvacrol OH group to the guanine N7, cytosine N3, and backbone phosphate group with overall binding constant of K(thymol-DNA) = 2.43 x 10(3) M(-1), K(carvacrol-DNA) = 1.55 x 10(3) M(-1). In thymol and carvacrol-DNA complexes, DNA remains in the B-family structure.  相似文献

利用水蒸汽蒸馏方法研究了生长于中国西北部不同采收时期百里香花,叶,茎和全草芳香油含量和化学成分的变化,结果表明:不同采收时期对花和叶芳香油含量却很大,而对茎中芳香油含量的影响较小,最佳采收时期在盛花期,此时不仅芳香油含量高,而且油中百里香酚和香酚的含量也高。气-质色谱图上出现了53中化合物,鉴定了其中的41中,占总成分的94.419%,芳香油中以百里香酚,香芹酚,对聚伞花素和松油烯为主,它们的含量在整个生长时期从43.953%,增加47.810%。  相似文献

The composition of essential oil isolated from Thymus algeriensis growing wild in Libya was analyzed by GC and GC-MS. The essential oil was characterized with thymol (38.50%) as the major component. The oil was screened for antioxidant activity using DPPH assay, and compared to thymol and carvacrol. Antioxidant activity was high, with the IC50 of 0.299 mg/ml, compared to 0.403 and 0.105 mg/ml for thymol and carvacrol, and 0.0717 mg/ml for BHA. In addition, antimicrobial activity was tested against eight bacteria and eight fungi. T. algeriensis oil showed inhibitory activity against tested bacteria at 0.001–0.05 mg/ml, while bactericidal activity (MBC) was achieved at 0.0025–0.05 mg/ml. For antifungal activity MICs ranged 0.0005–0.025 mg/ml and MFC 0.001–0.05 mg/ml. High antimicrobial activity against the fungi in particular suggests that the essential oil of Thymus algeriensis could have a useful practical application.  相似文献

The aerial parts of Gontscharovia popovii (B. Fedtsch. and Gontsch.) Boriss. were collected at full flowering stage. The essential oil was isolated by hydrodistillation and analyzed by a combination of capillary GC and GC-MS. Thirty-one components were identified with the main constituent being carvacrol (71.9%), followed by linalool (5.5%), p-cymene (4.5%) and gamma-terpinene (4.4%). The in vitro antimicrobial activity of the essential oil of G. popovii was studied against seven Gram-positive and Gram-negative bacteria (Bacillus subtilis, Enterococcus faecalis, Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Pseudomonas aeruginosa and Klebsiella pneumoniae) and three fungi (Candida albicans, Saccharomyces cerevisiae and Aspergillus niger). The results of the bioassays showed that the oil exhibited strong antimicrobial activity against all the tested fungi and bacteria except for the resistant bacterium Pseudomonas aeruginosa.  相似文献

The essential oils of oregano and thyme are active against a number of food-borne pathogens, such as Escherichia coli O157:H7. Carvacrol is one of the major antibacterial components of these oils, and p-cymene is thought to be its precursor in the plant. The effects of carvacrol and p-cymene on protein synthesis in E. coli O157:H7 ATCC 43895 cells were investigated. Bacteria were grown overnight in Mueller-Hinton broth with a sublethal concentration of carvacrol or p-cymene, and their protein compositions were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and confirmed by Western blotting. The presence of 1 mM carvacrol during overnight incubation caused E. coli O157:H7 to produce significant amounts of heat shock protein 60 (HSP60) (GroEL) (P < 0.05) and inhibited the synthesis of flagellin highly significantly (P < 0.001), causing cells to be aflagellate and therefore nonmotile. The amounts of HSP70 (DnaK) were not significantly affected. p-Cymene at 1 mM or 10 mM did not induce HSP60 or HSP70 in significant amounts and did not have a significant effect on flagellar synthesis. Neither carvacrol (0.3, 0.5, 0.8, or 1 mM) nor p-cymene (0.3, 0.5, or 0.8 mM) treatment of cells in the mid-exponential growth phase induced significant amounts of HSP60 or HSP70 within 3 h, although numerical increases of HSP60 were observed. Motility decreased with increasing concentrations of both compounds, but existing flagella were not shed. This study is the first to demonstrate that essential oil components induce HSP60 in bacteria and that overnight incubation with carvacrol prevents the development of flagella in E. coli O157:H7.  相似文献

The aim of the present study is to assess the possible protective effects of thymol and carvacrol against cisplatin (CP)‐induced nephrotoxicity. A single dose of CP {6 mg/kg, intraperitoneally (i.p.)} injected to male rats revealed significant increases in serum urea, creatinine, and tumor necrosis factor alpha levels. It also increased kidney contents of malondialdehyde and caspase‐3 activity with significant reduction in serum albumin, kidney content of reduced glutathione as well as catalase, and superoxide dismutase activity as compared to that of the control group. In contrast, administration of thymol {20 mg/kg, orally (p.o.)} and/or carvacrol (15 mg/kg, p.o.) for 14 days before CP injection and for 7 days after CP administration restored the kidney function and examined oxidative stress parameters. In conclusion, thymol was more effective nephroprotective than carvacrol. Moreover, a combination of thymol and carvacrol had a synergistic nephroprotective effect that might be attributed to antioxidant, anti‐inflammatory, and antiapoptotic activities.  相似文献
