Eggs of the turtle Trionyx spiniferus are rigid, calcareous spheres averaging 2.5 cm in diameter. The eggshell is morphologically very similar to avian eggshells. The outer crystalline layer is composed of roughly columnar aggregates, or shell units, of calcium carbonate in the aragonite form. Each shell unit tapers to a somewhat conical tip at its base. Interior to the crystalline layer are two tertiary egg membranes: the outer shell membrane and the inner shell membrane. The outer shell membrane is firmly attached to the inner surface of the shell, and the two membranes are in contact except at the air cell, where the inner shell membrane separates from the outer shell membrane. Both membranes are multi-layered, with the inner shell membrane exhibiting a more fibrous structure than the outer shell membrane. Numerous pores are found in the eggshell, and these generally occur at the intersection of four or more shell units.  相似文献

1. Experiments were carried out to decide whether or not the electromotive properties of dried collodion membranes depend upon their thickness. 2. A number of dried collodion membranes of varying thickness, 3–160 µ, were prepared from collodion preparations of different electrochemical activity. The characteristic concentration potentials across them were measured and the means of these values determined for each thickness. 3. The characteristic concentration potentials across dried collodion membranes are a function of their thickness. The thinnest membranes yield in all cases the lowest concentration potentials; increasingly thicker membranes give increasingly higher potential values, until a constant value is reached which is characteristic of the particular collodion preparation used. With electrochemically active collodion, characteristic concentration potentials approaching the thermodynamically possible maximum are obtained with membranes of only 10 µ thickness, thinner membranes giving appreciably lower values. With two rather inactive commercial collodion preparations the characteristic concentration potential increases from about 30 mv. for membranes 3 µ thick to about 42 mv. for 20 µ membranes; still thicker membranes do not show a significant increase in the potential values. With a highly purified collodion preparation the constant maximum value was found to be about 32 mv., 4 µ thick membranes giving only about 22 mv. 4. These results do not support the homogeneous phase theory as applied to the dried collodion membrane. They are readily compatible with the micellar-structural theory. Several special possible cases of the latter as applied to the dried collodion membrane are discussed.  相似文献

The flat type of dried collodion membrane used by Michaelis and his associates in numerous investigations has been subjected to mensuration in order that the dimensions of these membranes may be placed on record. The membranes had a functioning area of about 30 cm., were approximately 0.1 mm. in thickness and were composed on the average of 87 per cent by volume of collodion and 13 per cent by volume of pores. In reviewing some of the previously reported results of diffusion experiments with non-electrolytes in the light of the calculated values for the total pore area for the same membranes additional evidence was presented to show that a smaller molecule (acetone) probably utilizes a much larger percentage of the total pore area for its diffusion than is available for a larger molecule (glycerol). By using the figures of Fricke and McClendon for the thickness of the membrane of the red blood cell some comparisons were drawn between the dried collodion membrane as a model for certain biological membranes and the red blood cell membrane. In these comparisons emphasis was placed on the exaggerated importance of small electromotive forces and very slight permeabilities when these were associated with membranes of such extreme thinness as the red blood cell membrane.  相似文献

Dried microorganisms are particularly resistant to high hydrostatic pressure effects. However, exposure to high pressures of nitrogen proved to be effective in inactivating dried yeasts. In this study, we tried to elucidate this mechanism on Saccharomyces cerevisiae. High-pressure treatments were performed using different inert gases at 150 MPa and 25 degrees C with holding time values up to 12 months. The influence of cell hydration was also investigated. For fully hydrated cells, pressurized gases had little specific effect: cell inactivation was mainly due to compression effects. However, dried cells were sensitive to high pressure of gases. In this latter case, two inactivation kinetics were observed. For holding time up to 1 h, the inactivation rate increased to 4 log and was linked to a loss of membrane integrity and the presence of damage on the cell wall. In such case cell inactivation would be due to gas sorption and desorption phenomena which would rupture dried cells during a fast pressure release. Gas sorption would occur in cell lipid phases. For longer holding times, the inactivation rate increased more slightly due to compression effects and/or to a slower gas sorption. Water therefore played a key role in cell sensitivity to fast gas pressure release. Two hypotheses were proposed to explain this phenomenon: the rigidity of vitrified dried cells and the presence of glassy solid phases which would favor intracellular gas expansion. Our results showed that dried microorganisms can be ruptured and inactivated by a fast pressure release with gases.  相似文献

Summary Scanning and transmission electron microscopy have been used to study the structure of the hen's shell membranes and their relationship to the shell and to the chorioallantoic membrane.We have confirmed previous observations that the fibres of the outer shell membrane are of larger diameter than those of the inner shell membrane, and that the fibres of the outer shell membrane extend into the mammillary knobs of the shell.The shell membrane fibres are arranged in layers parallel to the surface of the egg and there is no interweaving between the layers. Individual fibres are randomly orientated and may extend for distances of at least 25 m. It is suggested that relative movement between the oviduct and the developing membrane is random in direction and location.Each fibre consists of a core with a covering cortex, but the idea that the core may consist of keratin is criticised. A limiting membrane separating the surface of the albumen from the fibres of the shell membrane is also formed from this cortex. During incubation the chorioallantoic membrane becomes pressed against this inner limiting membrane.No correlation could be found between the positioning of the mammillary knobs and the patterning of the shell membrane fibres. It is suggested that the positioning of the mammillary knobs reflects the pattern of certain secretory cells in the genital tract of the hen.No significant changes in structure of thickness of the shell membrane could be found during incubation. The avian eggshell is a composite biomaterial composed of non-calcifying eggshell membranes and the overlying calcified shell matrix. The calcified shell forms in a uterine fluid where the concentration of different protein species varies between the initial, rapid calcification and terminal phases of eggshell deposition. The role of these avian eggshell matrix proteins during shell formation is poorly understood. The properties of the individual components must be determined in order to gain insight into their function during eggshell mineralization. In this study, we have identified lysozyme as a component of the uterine fluid by microsequencing, and used western blotting, immunofluorescence and colloidal-gold immunocytochemistry to document its localization in the eggshell membranes and the shell matrix. Furthermore, Northern blotting and RT-PCR indicates that there is a gradient to the expression of lysozyme message by different regions of the oviduct, with significant albeit low levels expressed in the isthmus and uterus. Lysozyme protein is abundant in the limiting membrane that circumscribes the egg white and forms the innermost layer of the shell membranes. It is also present in the shell membranes, and in the matrix of the calcified shell. Calcite crystals grown in the presence of purified hen lysozyme exhibited altered crystal morphology. Therefore, in addition to its well-known anti-microbial properties that could add to the protective function of the eggshell during embryonic development, shell matrix lysozyme may also be a structural protein which in soluble form influences calcium carbonate deposition during calcification.  相似文献

In previous studies, we have shown that carrot (Daucus carota L.) somatic embryos acquire complete desiccation tolerance when they are treated with abscisic acid during culture and subsequently dried slowly. With this manipulable system at hand, we have assessed damage associated with desiccation intolerance. Fast drying caused loss of viability, and all K+ and carbohydrates leached from the somatic embryos within 5 min of imbibition. The phospholipid content decreased by about 20%, and the free fatty acid content increased, which was not observed after slow drying. However, the extent of acyl chain unsaturation was unaltered, irrespective of the drying rate. These results indicate that, during rapid drying, irreversible changes occur in the membranes that are associated with extensive leakage and loss of germinability. The status of membranes after 2 h of imbibition was analyzed in a freeze-fracture study and by Fourier transform infrared spectroscopy. Rapidly dried somatic embryos had clusters of intramembraneous particles in their plasma membranes, and the transition temperature of isolated membranes was above room temperature. Membrane proteins were irreversibly aggregated in an extended [beta]-sheet conformation and had a reduced proportion of [alpha]-helical structures. In contrast, the slowly dried somatic embryos had irregularly distributed, but non-clustered, intramembraneous particles, the transition temperature was below room temperature, and the membrane proteins were not aggregated in a [beta]-sheet conformation. We suggest that desiccation sensitivity of rapidly dried carrot somatic embryos is indirectly caused by an irreversible phase separation in the membranes due to de-esterification of phospholipids and accumulation of free fatty acids.  相似文献

This experiment examined the effect of weeks in egg production and type of housing confinement of turkey hens on calcium and soluble collagen levels in egg shell membranes; and discussion was given to their apparent relationship to gas exchange in turkey eggs. 2. The high level of acid-soluble collagen in inner and outer egg shell membranes of aging caged hens compared with the same aged floor-penned hens may have a relationship with the low hatchability generally recognized in caged hens. 3. The levels of calcium found in the outer shell membrane are low and appeared to decrease with the age of the hen. 4. There were no differences over time in levels of total collagen and neutral salt-soluble collagen (newly formed collagen) found in egg shell membranes of turkey hens confined in cages or floor pens. 5. It is suggested that the acid-soluble collagen levels found in inner shell membranes may have a relationship in limiting respiratory gas exchange during latter incubation time, and thus limit embryo survival.  相似文献

Cells use complex biochemical pathways to drive shape changes for polarization and movement. One of these pathways is the self-assembly of actin filaments and myosin motors that together produce the forces and tensions that drive cell shape changes. Whereas the role of actin and myosin motors in cell polarization is clear, the exact mechanism of how the cortex, a thin shell of actin that is underneath the plasma membrane, can drive cell shape changes is still an open question. Here, we address this issue using biomimetic systems: the actin cortex is reconstituted on liposome membranes, in an ‘outside geometry’. The actin shell is either grown from an activator of actin polymerization immobilized at the membrane by a biotin–streptavidin link, or built by simple adsorption of biotinylated actin filaments to the membrane, in the presence or absence of myosin motors. We show that tension in the actin network can be induced either by active actin polymerization on the membrane via the Arp2/3 complex or by myosin II filament pulling activity. Symmetry breaking and spontaneous polarization occur above a critical tension that opens up a crack in the actin shell. We show that this critical tension is reached by growing branched networks, nucleated by the Arp2/3 complex, in a concentration window of capping protein that limits actin filament growth and by a sufficient number of motors that pull on actin filaments. Our study provides the groundwork to understanding the physical mechanisms at work during polarization prior to cell shape modifications.  相似文献

The technique of Abrams and Sollner for the preparation of electropositive dried protamine collodion membranes has been improved. Porous collodion membranes cast on the outside of rotating tubes are treated for 48 hours with a solution of 2 per cent protamine sulfate buffered at pH 11. After being washed thoroughly the membranes are dried in air for several hours, soaked in water for several hours, and removed from the tubes. Further drying in air but without support shrinks the membranes slightly. The resulting membranes are designated "permselective" or "megapermselective" protamine collodion membranes. These membranes regularly give characteristic concentration potentials of –52 to –53 mv. and (in 0.1 M KCl) resistance of 0.5 to 15 ohms per membrane of 50 cm.2 area. This resistance is several orders of magnitude smaller than that of the conventional dyestuff- and alkaloid-impregnated positive membranes. The megapermselective protamine collodion membranes can be kept either dry or in water for prolonged periods without detectable deterioration. They are quite smooth, have a regular shape, and stand considerable handling without breakage. The megapermselective protamine collodion membranes are the electropositive analogues of the electronegative megapermselective collodion membranes described by Carr and Sollner.  相似文献

Cell-shape changes are insured by a thin, dynamic, cortical layer of cytoskeleton underneath the plasma membrane. How this thin cortical structure impacts the mechanical properties of the whole cell is not fully understood. Here, we study the mechanics of liposomes or giant unilamellar vesicles, when a biomimetic actin cortex is grown at the inner layer of the lipid membrane via actin-nucleation-promoting factors. Using a hydrodynamic tube-pulling technique, we show that tube dynamics is clearly affected by the presence of an actin shell anchored to the lipid bilayer. The same force pulls much shorter tubes in the presence of the actin shell compared to bare membranes. However, in both cases, we observe that the dynamics of tube extrusion has two distinct features characteristic of viscoelastic materials: rapid elastic elongation, followed by a slower elongation phase at a constant rate. We interpret the initial elastic regime by an increase of membrane tension due to the loss of lipids into the tube. Tube length is considerably shorter for cortex liposomes at comparable pulling forces, resulting in a higher spring constant. The presence of the actin shell seems to restrict lipid mobility, as is observed in the corral effect in cells. The viscous regime for bare liposomes corresponds to a leakout of the internal liquid at constant membrane tension. The presence of the actin shell leads to a larger friction coefficient. As the tube is pulled from a patchy surface, membrane tension increases locally, leading to a Marangoni flow of lipids. As a conclusion, the presence of an actin shell is revealed by its action that alters membrane mechanics.  相似文献

1. Eggs of Crocodilia and Chelonia, like those of birds, have a pair of egg membranes separating a thick layer of albumen from the calcareous shell. In contrast, eggs of oviparous Lepidosauria have only a single shell membrane, upon which relatively small amounts of calcium carbonate are deposited; and the volume of albumen in eggs is extraordinarily small at the time of oviposition. 2. With the possible exception of certain geckos and some chelonians, eggs of oviparous reptiles seem always to absorb water from the substrate during the course of normal incubation. In so far as the rate of water absorption exceeds the rate of water loss by transpiration from exposed surfaces, the eggs swell during incubation. The term ‘cleidoic’ cannot be used to describe eggs of this type. 3. Embryos of lizards and snakes influence the water potential of extra-embryonic fluids contained within their eggs, thereby maintaining or increasing the gradient in water potential that drives water absorption. 4. Embryos of Crocodilia and Chelonia obtain a substantial portion of the calcium used in ossification of skeletal elements from the inner surfaces of the eggshell. In contrast, embryonic lizards and snakes draw upon extensive reserves of calcium present in the yolk, and obtain little (if any) calcium from the eggshell. 5. All reptilian embryos seem to produce substantial quantities of urea as a detoxification product of protein catabolism. Contrary to expectation, uricotelism may not be common among reptilian embryos, even in those few instances where development takes place within a hard, calcareous egg. 6. In eggs of Crocodilia and Chelonia, respiratory gases seem to pass by diffusion through pores in the calcareous eggshell and through spaces between the fibres of the pair of egg membranes. No pores have been observed in the shell of lepidosaurian eggs, and so gases presumably diffuse between the fibres of the single (multilayered) shell membrane. 7. Metabolism of reptilian embryos is temperature-dependent, as is true for most ectothermic organisms. For each species, there appears to be a particular temperature at which embryonic development proceeds optimally, and departures from this optimum elicit increases in developmental anomalies and/or embryonic mortality. 8. Viviparity has evolved on numerous occasions among species of the Squamata, but seemingly never among Crocodilia or Chelonia. Since the evolution of viviparity entails a progressive reduction in the eggshell, only those organisms whose embryos do not depend upon the eggshell as a source of calcium may have the evolutionary potential to become viviparous. 9. Evolutionary transitions from oviparity to viviparity could have been driven by selection related to (i) thermal benefits to embryos consequent upon retention of eggs within the body of a parent capable of behavioural thermoregulation; (ii) protection of the eggs from nest predators and/or soil microbes; and (iii) more effective exploitation of a seasonal food resource by early emerging young.  相似文献

Questions regarding the structure of the inner and outer shell membranes of the chicken egg were addressed in this study by correlating observations from light microscopy and scanning and transmission electron microscopy. The egg membrane had a limiting membrane, which measured .9 to .15 microns in thickness and appeared to be a continuous and an impervious layer, but the shell membrane did not. Under the SEM, each membrane was seen to be made up of several fibre layers. In the tear preparations viewed under the SEM two layers were observed in the egg membranes and three to five layers in the shell membrane, with an apparent plane of cleavage between each layer. Each fibre was made up of a central core and an outer mantle layers. The central core was perforated by channels which measured .08 to 1.11 microns in diameter and ran longitudinally along the length of the fibre. Between the mantle layer and the fibre core was a gap or cleft measuring between .03 to .07 microns. The diameter of the fibres of the inner layer of the egg membrane ranged between .08 to .64 microns, whereas those of the outer layer of the same membrane ranged from .05 to 1.11 microns. Fibres in the shell membrane ranged from .11 to 4.14 microns diameter.  相似文献

S-nitroso-N-acetylpenicillamine (SNAP) is a pharmacological agent with diverse biological effects that are mainly attributable to its favorable characteristics as a nitric oxide (NO)-evolving agent. It is found that SNAP incorporates readily into dimyristoyl phosphatidylcholine (DMPC) bilayer membranes; and an approximate penetration profile was obtained from the depth dependence of the perturbation that it exerts on spin-labeled lipid chains. The profile of SNAP locates it deep in the hydrophobic core of both fluid- and gel-phase membranes. The spin relaxation enhancement of spin-labeled phospholipids with nitroxide group located at different depths in DMPC membranes was determined for nitric oxide (NO) and molecular oxygen (O2), at close to atomic spatial resolution. The relaxation enhancement, which is proportional to the corresponding vertical membrane profile of the concentration-diffusion product, was measured in the gel and fluid phases of the lipid bilayer. No significant membrane penetration was observed in the gel phase for the two water-dissolved gases. In the fluid phase, the transmembrane profiles of NO and O2 are similar and could be well described by a sigmoidal function with a maximum in the center of the bilayer, but that of NO is less steep and is shifted toward the center of the membrane, relative to that of O2. These differences can be attributed mainly to the difference in hydrophobicity between the two gases and the presence of the donor in the NO experiments. The biological implications of the above results are discussed.  相似文献

Cyanobacteria, descendants of the endosymbiont that gave rise to modern-day chloroplasts, are vital contributors to global biological energy conversion processes. A thorough understanding of the physiology of cyanobacteria requires detailed knowledge of these organisms at the level of cellular architecture and organization. In these prokaryotes, the large membrane protein complexes of the photosynthetic and respiratory electron transport chains function in the intracellular thylakoid membranes. Like plants, the architecture of the thylakoid membranes in cyanobacteria has direct impact on cellular bioenergetics, protein transport, and molecular trafficking. However, whole-cell thylakoid organization in cyanobacteria is not well understood. Here we present, by using electron tomography, an in-depth analysis of the architecture of the thylakoid membranes in a unicellular cyanobacterium, Cyanothece sp. ATCC 51142. Based on the results of three-dimensional tomographic reconstructions of near-entire cells, we determined that the thylakoids in Cyanothece 51142 form a dense and complex network that extends throughout the entire cell. This thylakoid membrane network is formed from the branching and splitting of membranes and encloses a single lumenal space. The entire thylakoid network spirals as a peripheral ring of membranes around the cell, an organization that has not previously been described in a cyanobacterium. Within the thylakoid membrane network are areas of quasi-helical arrangement with similarities to the thylakoid membrane system in chloroplasts. This cyanobacterial thylakoid arrangement is an efficient means of packing a large volume of membranes in the cell while optimizing intracellular transport and trafficking.  相似文献

Summary Decalcified chicken egg shell membrane is a suitable physical substrate for the growth of monolayers of an epithelial line of tissue culture cells, MDCK. The shell membranes with their adherent sheets of cells are easily prepared for cross-section study of monolayer growth with electron microscopy. The theoretical aspects of the problem of sieve-like membranes are developed. The method of preparing the dried collodion membrane is described, and the method of defining the property of a particular membrane is given. It consists of the measurement of the Co P, that is the P.D. between an 0.1 and an 0.01 M KCl solution separated by the membrane. Co P is in the best dried membranes 50 to 53 millvolts, the theoretically possible maximum value being 55 millivolts. Diffusion experiments have been carried out with several arrangements, one of which is, for example, the diffusion of 0.1 M KNO3 against 0.1 M NaCl across the membrane. The amount of K+ diffusing after a certain period was in membranes with a sufficiently high Co P (about 50 millivolts or more) on the average ten times as much as the amount of diffused Cl-. In membranes with a lower Co P the ratio was much smaller, down almost to the proportion of 1:1 which holds for the mobility of these two ions in a free aqueous solution. When higher concentrations were used, e.g. 0.5 M solution, the difference of the rate of diffusion for K+ and Cl- was much smaller even in the best membranes, corresponding to the fact that the P.D. of two KCl solutions whose concentrations are 10:1 is much smaller in higher ranges of concentration than in lower ones. These observations are confirmed by experiments arranged in other ways. It has been shown that, in general, the diffusion of an anion is much slower than the one of a cation across the dried collodion membrane. The ratio of the two diffusion coefficients would be expected to be calculable in connection with the potential difference of such a membrane when interposed between these solutions. The next problem is to show in how far this can be confirmed quantitatively.  相似文献

Incorporation of the fluorescent probe C11-BODIPY581/591 in two dried membrane systems, soy bean phosphatidylcholine liposomes freeze-dried in a carbohydrate/protein matrix and Lactobacillus acidophilus (La-5) freeze-dried in a carbohydrate matrix, was successful and could be visualised by Confocal Laser Scanning Microscopy (CLSM). The C11-BODIPY581/591 probe is a lipid oxidation reporter molecule, which is known to associate with the lipids of biological membranes and exhibit a fluorescence shift from the red range to the green range of the visible spectrum when it is oxidised together with the lipids. The present study is the first to demonstrate that the C11-BODIPY581/591 probe can be used in dried membrane systems, and that a detection of oxidation is possible by CLSM analysis directly on the dried samples.  相似文献

Diphtheria toxin (DT) induces the leakage of dipalmitoylphosphatidic acid (DPPA) membranes but not neutral dipalmitoylphosphatidylcholine (DPPC) membranes. Cholesterol incorporated into liposomes enhances the membrane leakage induced by DT in acidic DPPA membranes but not in neutral DPPC membranes. Membrane leakage was determined by assaying the release of TEMPOcholine, a cationic spin probe from the multilamellar vesicles by using electron spin resonance methods. The effect of DT on membrane leakage is noticeable at 3 micrograms/ml concentrations, and reaches a plateau of about 20% leakage at 20 micrograms/ml. This saturation phenomenon led to the postulation that DT binds to the first shell of DPPA membranes and induces the leakage of TEMPOcholine limited to this layer of DPPA multimellar vesicles.  相似文献
