Effects of Fluctuating Temperatures on Germination   总被引:4,自引:0,他引:4  
Germination responses to alternating daily cycles involvingtransfers between different temperatures over the range 0 toc. 40?C were investigated in four species: Lycopus europaeusL., Clarkia unguiculata Lindl., Apium graveolens L. cv. GiantRed, and Silene dioica (L.) Clairv. Comparisons of the germination of these species revealed distinctivedifferences which could be related to particular features ofthe geographical regions or ecological habitats in which theyoccur naturally. The effects of fluctuating as opposed to constant temperaturesvaried from one species to another. L. europaeus depended almostabsolutely on fluctuating temperatures for germination. Germinationof S. dioica was greatly increased by fluctuating temperatures.Moderate increases occurred over a restricted temperature rangein A. graveolens cv. Giant Red and C. unguiculata was affectedin only minor ways. These results are discussed in relationto previous attempts to explain the mechanisms responsible forthe frequently beneficial effects of fluctuating temperatureson germination.  相似文献

The phenomenon of auto-inhibition of spore germination has beendemonstrated in the slime moulds Dictyostelium purpureum, strainno. 2; Dictyostelium discoideum, strain no. 1; Dictyosteliummucoroides, strain no. 2; and Polysphondy-Iium violeceum, strainno. 6. A water-soluble substance present in spores was shownto cause this self-inhibition. The substance was isolated fromeach species studied. Evidence as presented indicates that thesame substance occurs in each species of Dictyostelium, butthat it differs from that found in Pv6. An attempt was alsomade to relate this inhibitory substance to the phenomenon of'differential inhibition' in Dp2, Dd, Dm2, and Pu6. An explanationbased on evidence for differential sensitivity to a common inhibitorof spore germination in Dictyostelium and an inhibition in otherthan the spore stage in Pu6 is presented. Apparent differentialproduction of a common inhibitor in Dictyostelium is also reported.  相似文献

Sugars in Natural and Artificial Pollen Germination Substrates   总被引:1,自引:0,他引:1  
The stigmatic exudates and pollen grains of five unrelated specieswere tested for sugars. Glucose, fructose, and sucrose werefound in the stigmatic fluid of Yucca aloifolia L. and glucoseand fructose in that of Oenothera adrummondii Hook. In in vitroexperiments with Y. aloifolia pollen, high germination percentageswere obtained in artificial media containing glucose or sucrose.Fructose, which is present in the stigmatic fluid of the Yuccasp. resulted in high in vitro pollen germination only when borateand calcium were added to the medium. Presence of bound sugarsis indicated in the stigmatic secretion of Citrus aurantiumL. and pollen of the single plant tested germinated at a lowpercentage in artificial sugar media. No sugars were detectedin the stigmatic fluids of Hemerocallis fulva L. and Zea maysL. and in these two species in vitro pollen germination in sugarymedia was negligible or absent. The pollen grains of all five species contain sucrose and thoseof Oenothera and Citrus also reducing sugars.  相似文献

Photoinhibition of Seed Germination in Mediterranean Maritime Plants   总被引:4,自引:1,他引:3  
Photoinhibition of seed germination was shown for Allium staticiforme,Brassica tournefortii, Cakile maritima and Otanthus maritimus,all plant species inhabiting sandy coasts of the MediterraneanSea. Germination of A. staticiforme was found to be typicallyMediterranean in regard to its temperature range (>0–20°C), while B. tournefortii germinated optimally at intermediatetemperatures (15–25 °C). Light sensitivity was morepronounced in the latter species and 50 % inhibition of seedgermination was obtained with photon flux densities of approximately0.1 and 0.015 mol m–2 d–1 in A. staticiforme andB. tournefortii, respectively. However, the slopes of the regressionlines of germination plotted against the logarithm of whitelight flux density are similar in both species. From monthlyexperiments performed under fluctuating conditions of temperatureand light, simulating the elimate of Athens throughout the year,optimal germination response peaked in ‘winter’conditions for A. staticiforme, while a bimodal pattern wasobserved for B. tournefortii (maxima at the warm ends of therainy season). In contrast to the other plants, a fifth speciestested, Crithmum maritimum, showed an absolute light requirement,probably related to the rocky habitats of the species; the optimumgermination period coincided with the rainy season. Pot experimentswith seeds of the five species buried at various depths confirmedthat maximum emergence is favoured by shallow depths (0.5–1cm). It is concluded that seed germination in maritime plantsof the Mediterranean rim is mediated by a photoinhibition mechanismwhich can be considered an adaptation strategy against surfaceseedling establishment at the harsh, sandy or shingle, sea coasthabitats. The different germination behaviour of the seeds of two irises,Iris lorteti and I. atropurpurea was found to be due to thedifferent mechanical resistance of the integument, at the micropylarend, to radicle protrusion. A pressure of 135 atm was necessaryin l. lorteti seeds for radicle protrusion, while in I. atropurpurea77 atm was sufficient. In contrast Pancratium maritimum requireda pressure of only 10 atm. The outer integument of seeds ofI. lorteti was found to contain a compound which was toxic tothe germinated embryo but did not act as a germination inhibitor.Extracts of the endosperm also had a slight germination inhibitingeffect. An interaction between this weak inhibitor and the effectof the testa could not be ruled out completely. A test assayfor germination using excised embryos was developed. A methodfor germination of Iris seeds, by cutting off the outer integumentat the micropylar end, was developed and is being exploitedcommercially. Presence of inhibitors of germination in seeds of five umbellifercrops viz. fennel (Foeniculum vulgare Mill.), cumin (Cuminumcyminum L.), carum (Carum copticum Benth. and Hook.), carrot(Daucus carota L.), and coriander (Coriandrum sativum L.) wasdemonstrated. The effect of seed exudates on the pattern ofinhibition of germination of these seeds was also studied. Presence of exudates in general delayed germination althoughthe time of maximum inhibition was different with differentcrop seeds. The inhibitory effect was high at an early stagein the case of carum and carrot and persisted in the latterbut disappeared in the former. The inhibitory effect on cuminand fennel increased up to the seventh day and then began todecrease. Growth of root and hypocotyl of 16-day-old seedlingswas also inhibited by the exudates.  相似文献

We have been successful in building an energy-level model thatdescribes seed germination. We used the autocatalytic reactionrate equation to fit the germination rate for seed germination.The two parameters [A]0 and [F]0 were found by fitting the integratedgermination rate equation to the data. The values of [A]0 and[F]0 obey the Arrhenius equation and give activation energiesfor the second and third stage of the four-compartmental modelof seed germination. The thermodynamics of isothermal seed germinationis proposed and the enthalpy, entropy, and free energy are calculatedfor the transition from state A to state F. The time delay isa function of temperature and it leads to a rate constant thatcan be used to get the activation energy for the total germinationprocess. We believe the model is universal. The achenes of the plant Tithonia rotundifolia show an embryodormancy, requiring a 3-month period of after-ripening to stimulategermination. Mechanical scarification did not enhance imbibitionof the achenes, indicating that the thick bi-layered achenewall does not impede water uptake. The light conditions necessaryfor optimal germination changed with achene age and the achenesbecame less sensitive to temperature. About 30–40% ofrecently harvested achenes germinated in the dark at 25 °C.Following a period of dry storage some of the achenes developeda light requirement and germination increased at 20, 25 and30 °C. Gibberellic acid stimulated the germination of achenestested after 12 weeks dry storage, and could substitute to someextent for the light and temperature requirements of the storedachenes. This study reports the ability of Fusarium to induce orchidseed colouration and germination. The in vitro bioassay germinationtest, using a Fusarium isolate from the protocorm of Cypripediumreginae, was compared with standard chemical procedures of triphenyltetrazolium chloride (TTC) and acid fuchsin (AC) for testingseed viability. With Cypripedium reginae, Cypripedium parviflorumand Platanthera grandiflora, the efficiency of the bioassaywas similar to that of the TTC and AC procedures. However, thebioassay was more appropriate for estimating embryo viabilityafter a prolonged seed pretreatment (more than 2 h) in 10% sodiumhypochlorite, a surface sterilant often used to enhance germinationof terrestrial species. We also obtained in vitro Cypripediumreginae seed germination induction and protocorm formation bythe same Fusarium isolate. This is the first confirmation ofBernard's early reports that orchid fusaria could stimulateseed germination (Bernard N. 1990.Révue Généralede Botanique12 : 108–120). However, the importance ofthe non-mycorrhizal Fusarium fungus in promoting germinationseems to be relatively minor compared to that of specificRhizoctoniaorchid mycorrhizas. Gibberellin A3 (GA3), kinetin and zeatin promoted germinationof conditioned witchweed (Striga asiatica [L.] Kuntze) seedssimilar to dl-strigol and a natural stimulant from corn rootexudates. GA3 promoted germination only with scarified seeds.However, these regulators, as well as dl-strigol and naturalstimulants, inhibited the conditioning of seeds and their subsequentgermination. On the other hand, abscisic acid induced 74% germinationof conditioned seeds but had no effect on seed conditioning.Other chemicals such as sucrose, NaNO3, vitamin B1, L-methionine,nicotinamide, indoleacetic acid and colchicine had no apparenteffect on seed conditioning, while sucrose, L-methionine andinositol induced about 30 % germination of conditioned seeds.Seeds generally required either a longer conditioning time ora higher terminal dl-strigol concentration to eliminate anyinhibition of seed conditioning induced by a compound. dl-Strigol, germination stimulation, parasitic plants, seed conditioning, seed germination, Striga asiatica (L.) The influence of bracteoles on the germination response of asalt marsh annual, Atriplex prostrata, and a salt desert perennial,A. griffithii, was determined. Attached bracteoles did not inhibitgermination of A. prostrata but completely inhibited germinationof A. griffithii seeds. Germination of seeds ofA. griffithiiwas also somewhat inhibited in the presence of detached bracteoles.The osmotic potential of dissolved substances in water-saturatedbracteoles was -4.8 ± 0.05 MPa for A. griffithii and-2.4 ± 0.15 MPa for A. prostrata. Ash content of bracteoleson a dry weight basis was 28.7 ± 0.37% for A. prostrataand 30.0 ± 0.29% for A. griffithii. The presence of relativelyhigh concentrations of dissolved salts in bracteoles may explainthe reduced germination of A. griffithii. Germination of seedsof the perennial A. griffithii may also be inhibited mechanicallyby the presence of persistent bracteoles, whereas the annualA. prostrata has ephemeral bracteoles that may serve to limitgermination in the autumn but are not present during the normalspring germination period. Bracteoles could affect the dispersalof seeds by water since some fruiting structures of A. griffithiiand A. prostrata are able to float in 1.0% NaCl solutions for5 and >30 d, respectively, compared with a maximum of 4 and3 d for seeds without bracteoles. Fruiting structures of A.prostrata floated for longer over a range of salinities thandid those ofA. griffithii ; this may play a significant rolein the dispersal of this salt marsh species. Quantitative data are furnished respecting the seed productionand germination of Limosella aquatica, which is shown to havea large reproduction potential like other species of intermittentlyavailable habitats, despite its small assimila-tory surface.A tentative estimate of the seed output of the rarer diploidspecies L. subtdata suggests that this may be only about one-sixththat of the tetraploid L. aquatica. Features described indicatespecialization primarily to dispersal by water, and submergedcapsules readily dehisce. Germination, which may occur within3 days, is shown to be quasi-simultaneous in bright light underwhich over 90 per cent may be attained but a very small percentagewere found able to germinate in dim light. Experimental evidenceis provided that indicates the presence of an inhibitor in theseeds the amount of which may, it is suggested, vary from seedto seed in a normal manner. As with other ‘mud species’the seedling develops a zone of supporting ‘prop hairs’around the base of the radicle. The markedly intermittent occurrence of the species is attributablemainly to the irregular incidence of the requisite environmentalconditions.  相似文献

Petunia hybrida and Lilium lankongense pollens were germinatedon thin layer chromatography (TLC) plates following chromatographyof extracts from the self-, cross- and unpollinated stigmas,styles and ovaries and the seeds, leaves and pollen of threeinbred Brassica oleracea families. Zones of pollen germinationinhibition on the TLC plates showed that inhibitory compoundswere present in the tissue extracts. The Rf values and numberof these compounds varied with the tissue used, stigma tissuecontaining the largest amounts and the greatest number of inhibitors.In contrast, differences between the inbred lines tested wereslight and quantitative. Pollen from both P. hybrida and L.lankongense gave the same results; that from B. oleracea couldnot be used because of its poor germination. The stimulation of the germination of Agaricus bisporus sporesby mycelium of the same species has been shown to be due toa volatile metabolite, diffusing into the culture medium andinto the atmosphere. A wide range of other fungi has been foundto affect A. bisporus spores in a similar manner. There wasno evidence . that the stimulant was carbon dioxide. A seven-carbonolefin, isolated from air, which had been passed through culturesof A. bisporus mycelium and dried with phosphorus pentoxide,showed germination-stimulating activity but may have been producedby the action of this drying agent on some other metabolitefrom the mycelium. Cell division patterns during germination of spores of Cyatheaaustralis, C. cooperi and Dicksonia antarctica were examinedby light microscopy of sectioned materials and by the scanningelectron microscope. In C. australis and C. cooperi the rhizoidwas traced to a small cell formed by an asymmetric divisionof the spore by a wall parallel to its equatorial plane. Incontrast, the rhizoid was formed by a division of the sporeparallel to its polar axis in D. antarctica. In spores of bothgenera, a second division wall oriented in a plane perpendicularto the first gave rise to the protonemal cell. Certain aspectsof germination described here in spores of Cyathea and Dicksoniaare in conflict with the published accounts of spore germinationin these genera. Aeluropus lagopoides(Linn.) Trin. Ex Thw. (Poaceae) is a perennialgrass distributed from coastal Sindh and Balochistan to salineflats of Punjab, Pakistan. Seeds collected from an inland populationofA. lagopoides located on the University of Karachi campuswere germinated under various levels of salinity (0, 100, 200,300, 400 and 500 m M NaCl) and temperature regimes (10/20, 15/25,20/30 and 25/35 °C) in a 12 h dark/12 h light photoperiod.Highest germination was obtained under non-saline conditions,and an increase in NaCl concentration progressively inhibitedgermination. Inhibition of germination was greater at coolertemperatures (10/20 °C) when no seed germinated above aconcentration of 300 m M NaCl. The germination response at moderatetemperatures (20/30 °C) was optimal, with 30% of seeds germinatingin 500 m M NaCl. The rate of germination decreased as salinityincreased. Germination rate was highest at 20/30 °C andlowest at 10/20 °C. Seeds were transferred from salt solutionsto distilled water after 20 d and those from high salinitiesrecovered quickly at warmer temperatures with an optimal responseat 20/30 °C. Common bean (Phaseolus vulgaris L.) is grown over a wide rangeof environments, including sites with low or high soil temperaturesat sowing time. To describe the temperature responseof seedgermination, 20 bean genotypes were evaluated using a rolledpaper towel system with 11 constant temperatures ranging from12 to 34 °C. Germination response was characterized by fittingcumulative counts using a maximum-likelihood analysis. Rateof germination increased from abase temperature (Tb) typicallynear 8 °C to an optimal development temperature (To) of29 to 34 °C. Tb did not differ among common bean genotypes.Mesoamerican germplasm showed slightlyhigher To than Andeangermplasm, but there was large variation in To within each ofthe two gene pools. The three-horned stock, Matthiola tricuspidata (L.) R. Br. isa widespread annual plant of the Mediterranean sandy shores.Its seeds are dark germinating and negatively photosensitive,in accordance with our previous findings for a number of othermaritime plants. Full germination was obtained at a wide rangeof temperatures (5-25 °C) in the dark. Inhibition of germinationunder light of various spectral qualities could be generallycorrelated, negatively and positively, respectively, with phytochromephotostationary state (ø) and relative cycling rate ofphytochrome (H). The inhibition of germination by white (fluorescent),blue and far-red light, applied either continuously or intermittently,consistently showed a linear dependence upon the logarithm ofthe flux density of the irradiation. The resulting photoinhibitioncurves had parallel slopes and, compared to those of other maritimeplants, they were shifted to higher flux densities, Continuousblue or far-red irradiations, both establishing a similar øvalue (0·26), resulted in statistically similar regressioncurves, thus favouring the hypothesis that phytochrome is thesingle photoreceptor in the photoinhibition of seed germination.Copyright1994, 1999 Academic Press Matthiola tricuspidata (L.) Laboratory and field experiments involving Asplenium ruta-murariaL., Asplenium trichomanes L. subsp. quadrivalens D.E. Meyeremend. Lovis and Asplenium scolopendrium L. [= Phyllitis scolopendrium(L.) Newm.] revealed differences in their temperature requirementsfor germination and in their sexual development on soil. Thegermination responses of A. trichomanes and A. scolopendriumto culture temperatures of 10, 15, 20 and 25 °C were verysimilar and all spores were able to germinate at 10 °C.Spores of A. ruta-muraria differed in that germination was slowerat 15-25 °C and very much delayed, or even inhibited, at10 °C.
