Microtubules were successfully illustrated in thyrotrophs and thyroidectomy cells of rat pituitary glands. In contrast, microfilaments were mostly seen in the nonglandular follicular cells. Numerous microtubules were observed in the early stages of development of the thyroidectomy cells. In thyroidectomy cells microtubules were located in close proximity to mitochondria, endoplasmic reticula, secretory granules, and membranes of Golgi complexes. Consequently, it is suggested that microtubules may play a role in degranulation or other processes associated with the hypersecretory state.  相似文献

We studied the internalization and intracellular distribution of [3H] GABA in rat anterior pituitary cells. Electron microscopic autoradiography of anterior pituitary fragments or dispersed pituitary cells incubated with [3H] GABA showed that lactotrophs and, to a lesser extent, somatotrophs were the only cells that contained radioactive grains. Grain density analysis performed on dispersed pituitary cells after a pulse-chase experiment (10 min pulse and then change to a medium without radioactive GABA for various periods up to 2 h) revealed that GABA internalized by lactotrophs was distributed in various intracellular membranous organelles. Of the cell compartments examined, plasma membrane, Golgi apparatus, mitochondria and secretory granules had different time-dependent labeling patterns. The highest grain density values were associated with plasma membrane (at the first chase time) and the Golgi apparatus. Mitochondria and secretory granules also showed significant grain density values. A similar pattern of distribution was observed when fragments of prolactin-secreting pituitary adenomas were incubated with [3H] GABA. These results provide morphological data on the cellular specificity and intracellular distribution of GABA in anterior pituitary cells.  相似文献

The ability of muscle fibers to survive the ischemic condition was examined. After 0 to 11 h of ischemia of the rat m. lumbricalis was transplanted to the anterior chamber of the rats eye. Using a morphological and immunohistochemical (monoclonal antibodies to heavy chain of the fast myosin, PAP-method) methods it was revealed that population of surviving muscle fibers reduced significantly beginning from 6 h of ischemia. After 9 h of ischemia there were no muscle fibers in the transplant but one could observe the new formed muscle tubules. It is concluded that up to 6 h the muscle fibers of m. lumbricalis ischemia still preserve the ability to recover.  相似文献

In order to reveal whether thyrotroph cells of the human pituitary are affected by aging, a histologic and immunocytologic study was undertaken on the pituitaries of patients over 80 years of age (10 men, 10 women), dying of various acute illnesses and the findings compared with those of a younger age group (10 men, 10 women), dying of various acute illnesses between the age of 20-56 years. The incidence, distribution, granulation, cytoplasmic vacuolization, as well as immunoreactivity of TSH cells, showed no major differences related to age and sex. Lack of involution of thyrotroph cells in old subjects indicates that the pituitary is capable of producing TSH with advancing age.  相似文献

Summary The characteristic ultrastructure of thyrotrophs of the rat anterior pituitary was observed by immuno-electron microscopy and enzyme cytochemistry with increasing time after thyroidectomy (TX). The rough endoplasmic reticulum (ER) became dilated, the intracisternal granules reacted to serum raised against thyroid stimulating hormone (TSH) around 21 days after TX, and lysosomes and peculiar structures with positive acid phosphatase activity were present. The administration of thyroxine (T4) to the thyroidectomized rats resulted in the reformation of secretory granules, a reduction of dilated cisternae of rough ER and the activation of the lysosomal systems. Morphological features indicating that the TX-cells might be derived from growth hormone (GH) cells or cells other than TSH cells, previously suggested by some researchers, were not recognized in the present study. The amount of serum and pituitary TSH was measured by radioimmunoassay (RIA), and correlated well with the morphological changes. These results indicate that the TX-cells are hypertrophied hyperfunctioning TSH cells that have been affected by the lack of negative feedback of thyroid hormone.  相似文献

The cytogenesis of immunoreactive gonadotropic cells in the fetal rat pituitary was analyzed at the light and electron microscope levels using the indirect peroxidase-labeled antibody method and antisera against ovine FSH (A-oFSH) and ovine LH (A-oLH), and its two subunits (A-oLHβ and A-oLHα). At the light microscope level, the first immunoreactive cells were detected on the seventeenth day postcopulation (dpc) with A-oLHβ. Cells immunochemically stained with A-oLHα and A-oLH were generally observed 24 hr later. At the electron microscope level, the first immunoreactive cells were detected on 16 dpc with A-oLHβ. These first immunoreactive cells were small, but already displayed some small secretory granules (80–120 nm). On 17 dpc, gonadotropic cells were stained with A-oLHβ as well as with A-oLHα and A-oFSH. On 18 dpc, the number and the size of immunoreactive cells began to increase. By 19 dpc, they displayed an important development of ergastoplasmic cisternae and Golgi zone. At term, nevertheless, the ultrastructural features of fetal gonadotropic cells still differed from those of adult gonadotropic cells.  相似文献

E600 resistant non-specific esterase activity or acid phosphatase activity were localized in corticotrophic cells identified by postembedding immunocytochemistry (PAP of protein A-immunogold techniques). The lysosomal system of this cell type consists of dense bodies, of a population of small lysosomes mostly situated at the cell periphery in the vicinity of secretory granules as well as of tubular structures. These latter were located either in the central part of the cytoplasm and probably belonged to the Golgi apparatus or at the cell periphery, partly in the extensions. Small lysosomes occurred to be in continuity with enzyme-containing tubules. In a few structures lysosomal enzyme activity and ACTH immunoreactivity overlapped. Some autophagic vacuoles seemed to contain secretory granule matrix. It is suggested that the concept of crinophagy can be extended to the corticotrophs, though the lysosomal system may be involved in the specific function of this cell type by other mechanisms as well.  相似文献

The prism pattern in the inner enamel of adult rat incisors was studied with the SEM in unfixed tissues that had been sectioned, ground, polished, and etched. Six different types of aberrations in the prism pattern were encountered: 1. Prism lamellae may be shorter than the mesio-lateral width of enamel. 2. Prism lamellae may deviate from a transverse orientation. 3. Prism lamellae may "fuse" or "bifurcate." 4. Prisms of two adjacent lamellae may pursue a common course. 5. Prisms may change direction. 6. Variations exist in the outline of transversely cut prism profiles. Aberrations were observed at any distance from the dentino-enamel junction. These observations were used as a basis for an analysis of the movement of ameloblasts during rat incisor amelogenesis. It was concluded that it is physically possible for the ameloblasts to create the observed aberrations as they move along the path of the prisms. However, the aberrations seem to make it more difficult to understand the factors controlling ameloblast movement. Occasionally crystallite bridges connecting adjacent prisms were observed. A configuration resembling a bifurcating prism is pesented.  相似文献
